Abstract:In this study, an environmentally sustainable fluorimetric method for determination of Vonoprazan fumarate (VON) in dosage forms using nanoprobes consisting of nitrogen and sulfur co-doped carbon quantum dots (N, S-CQDs). The N, S-CQDs were prepared through a microwave-assisted method in 30 s. The resulting N, S-CQDs were characterized using transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HRTEM), Fourier transform infrared spectroscopy (FTIR), and X-ray photoelectron spectroscopy (XPS). They exhibit fluorescence emission at 460 nm after excitation at 385 nm with a high quantum yield (60%). The analytical approach for VON determination relies on the quenching effect exerted by VON on the native fluorescence intensity of CQDs. The quenching mechanism was investigated using Stern-Volmer plots. The proposed method demonstrates linearity across a concentration range 10-80 μM (4.6-36.8 μg/mL) with corresponding limits of detection and quantitation calculated as 2.17 μM (0.99 μg/mL) and 6.58 μM (3.02 μg/mL), respectively. The method has been effectively utilized for the determination of VON in the pharmaceutical samples. Statistical comparison with reported RP-HPLC has been performed to verify the accuracy and precision of the developed method. The environmental sustainability of the developed method has been thoroughly examined through various greenness metrics.

Abstract:The suggested study employs a specific technique to ensure the sustainability and ecological friendliness of an analyte evaluation procedure. The utilization of various recently developed methodologies as potential indicators of ecological sustainability and feasibility necessitates attention and consideration. The established analytical methodology employs a novel reagent mixture composed of phthalic dialdehyde and sulfite ion. This reagent is used to create a new fluorescent core that can be monitored using fluorimetric techniques at a wavelength of 385 nm after being excited at 320 nm. Condensation of the amino group of fluvoxamine can occur in a pH-controlled solution containing phthalaldehyde/sulfite. The present investigation presents a new, environmentally sustainable, and accurate fluorometric technique for identifying the medication. A linear relationship was detected for fluvoxamine over the 0.06–1.50 μg mL −1 concentration range. The method's validation and quantum yield determination were assessed following the International Council for Harmonization's (I.C.H.) established guidelines. The results exhibit remarkable precision and accuracy, consistent with the different methodologies published in the literature. Subsequently, an assessment was conducted to evaluate the environmental sustainability of the suggested technique, employing a range of contemporary indicators for determining environmental safety. The WAC concept, which combines ecological and operational elements and makes use of the Green/Red/Blue (RGB 12) scheme, interestingly identifies the current analytical method as white. Finally, BAGI, a novel tool for evaluating the method's applicability, was used in this study and showed that the planned method achieved a high applicability score.

Abstract:In this paper, vilazodone (VLD), a serotonin modulator prescribed for major depressive disorder, was investigated using a rapid, highly sensitive, and eco-friendly spectrofluorometric approach. The native fluorescence of VLD, originating from its indole moiety, exhibited an emission peak at 486 nm upon excitation at 241 nm. However, the presence of a piperazinyl nitrogen atom in the VLD structure, acting as an electron donor, significantly diminished the fluorescence intensity through photoinduced electron transfer (PET) to the indole ring. However, by protonating this nitrogen atom using 0.02 M Teorell–Stenhagen buffer (pH 3.5), inhibition of the PET process effectively blocked electron transfer, restoring the fluorescent properties of the drug. Further, an enhancement in the fluorescence was achieved by employing methanol as the solvent, resulting in a 1.5-fold increase. The combined use of PET blockage and methanol enabled the detection of VLD at levels as low as 0.78 ng mL −1 . Calibration analysis demonstrated linearity within the range 5–400 ng mL −1 , exhibiting a correlation coefficient of 0.9998 and a limit of quantification of 2.37 ng mL −1 . The method obeyed the requirements of International Council on Harmonization (ICH). The proposed approach was applied for the accurate measurement of VLD in pharmaceutical tablets, content uniformity testing based on USP requirements, and determining VLD concentration in spiked human plasma. Moreover, the environmental impact, in addition to practical effectiveness, of the proposed approach was evaluated using different metrics.

Abstract:The spectrofluorimetric determination of ZJO777 using salicylic acid as an emission reagent has been investigated by measuring the decrease of fluorescence intensity of salicylic acid due to forming an ion association complex of ZJ0777-salicylic acid.An emission peak of salicylic acid,which is decreased linearly by addition ZJ0777,occurs at 442nm in 1.5% CH3COOH-CH3COOC2H5 solution with excitation at 305nm.The linear range for ZJ0777 is from 0.2 to 10mg/L,and its detection limit is 0.1mg/L.The in? uence of coexisting substance was also inspected,and the result showed that this method has a better selectivity.Therefore,a new fluorescence method,which is high sensitive,simple and rapid,was developed to determinate ZJ0777 at trace amounts.The method could be applied to the determination of ZJ0777 in water sample with satisfactory results.

Abstract:The present work introduces the first spectrofluorimetric approach for the estimation of the novel antiparkinsonian drug safinamide mesylate in pharmaceutical formulations and human plasma. The developed method depends on measuring its native fluorescence in methanol. The measurements were carried out at 299 nm upon excitation at 226 nm. The effects of pH, different solvents, and organized media were thoroughly investigated and optimized. Methanol was found to be the most suitable solvent, and Britton-Robinson buffer (0.02 M) with pH 4 was used. The fluorescence quantum yield of the studied drug was calculated and found to be as high as 44.5 %. The method was validated following the International Council of Harmonization (ICH) validation recommendations. The linearity range was found to be 50.0–2000.0 ng/mL, reaching limits of detection and quantitation of 11.58 and 35.08 ng/mL, respectively. The method's high sensitivity permitted its application to spiked human plasma samples with high percentage recoveries (97.52–101.27%) and low percentage RSD values (<1.24). The designed method was also applied for the estimation of the cited drug in commercial Safinozol® tablets, and the results obtained agreed statistically with those of the comparison method. Furthermore, the greenness and whiteness profiles of the developed method were evaluated for the accomplishment of a complete eco-friendliness profile of the designed methodology.

Abstract:In this work, hydroxyapatite (HAp) obtained from the deorganification of bovine bones using hot NaOH solution was used to synthesize a HAp/epoxy infiltrated composite. Infiltration was carried out by vacuum assisted immersion in hot epoxy resin. The resulting composite was characterized regarding polymer content, morphological aspects and flexural strength. The infiltration method used resulted in thorough infiltration of the HAp but some residual porosity remained. Although the epoxy resin showed good interaction with the HAp, high polymer content was achieved and the flexural strength of the composite was higher than that of the original resin or the HAp, composite strength was lower than that of the human cortical bone.
New Fluorescence Quenching Approach for Determination of Valsartan in Certain Tablets and Spiked Biological Fluids

Abstract:A new, simple, selective, sensitive, fast, economical, and reliable fluorescence quenching method for the quantitation of valsartan was investigated using basic fuchsin act as a fluorometric dye. The method was depended on the detection quenching influence of valsartan on the fluorescence intensity of basic fuchsin and the reaction between valsartan and basic fuchsin in a McIlvaine buffer medium at pH = 6 to yield a new basic fuchsin–valsartan non-fluorescent complex. The excitation and emission of basic fuchsin fluorescence signal were identified at 535 and 728 nm, respectively. A fluorescence quenching value (ΔF) displayed a very good linear relationship (R2 = 0.9992) with valsartan concentration ranging from 0.003 to 3 μg/mL, a detection limit as low as 0.0009 μg/mL with a high precision and accuracy (RSD% <3). Significantly, no interference effect was found due to the presence of other ingredients commonly found in medical formulations. The acquired data were statistically compared with those acquired from reported chromatographic method and were observed to be in excellent agreement at a 95% confidence level; the planned fluorescence quenching procedure was subsequently utilized to detected the concentration of valsartan in spiked biological fluids and commercial medical tablets.

Abstract:Gemigliptin-Rosuvastatin single-pill combination is a promising therapeutic tool in the effective control of hyperglycemia and hypercholesterolemia. Organic sensors with high quantum yields have profoundly significant applications in the pharmaceutical industry, such as routine quality control of marketed formulations. Herein, the fluorescence sensor, 2-Morpholino-4,6-dimethyl nicotinonitrile 3 , (λex; 226 nm, λem; 406 nm), was synthesized with a fluorescence quantum yield of 56.86% and fully characterized in our laboratory. This sensor showed high efficiency for the determination of Gemigliptin (GEM) and Rosuvastatin (RSV) traces through their stoichiometric interactions and simultaneously fractionated by selective solvation. The interaction between the stated analytes and sensor 3 was a quenching effect. Various experimental parameters and the turn-off mechanism were addressed. The adopted approach fulfilled the ICH validation criteria and showed linear satisfactory ranges, 0.2–2 and 0.1–1 μg/mL for GEM and RSV, respectively with nano-limits of detection less than 30 ng/mL for both analytes. The synthesized sensor has been successfully applied for GEM and RSV co-assessment in their synthetic polypill with excellent % recoveries of 98.83 ± 0.86 and 100.19 ± 0.64, respectively. No statistically significant difference between the results of the proposed and reported spectrophotometric methods in terms of the F - and t -tests. Ecological and whiteness appraisals of the proposed study were conducted via three novel approaches: the Greenness Index via Spider Diagram, the Analytical Greenness Metric, and the Red–Green–Blue 12 model. The aforementioned metrics proved the superiority of the adopted approach over the previously published one regarding eco-friendliness and sustainability. Our devised fluorimetric turn-off sensing method showed high sensitivity, selectivity, feasibility, and rapidity with minimal cost and environmental burden over other sophisticated techniques, making it reliable in quality control labs.

Abstract:Two simple, valid and green chromatographic based techniques are developed in the present work for first time to simultaneously analyze the recently approved combination of Aspirin (ASP) with the novel gastro-protective agent Vonoprazan (VON). First method is an HPLC-DAD "diode array detection", where separation was successful using C18 (250 × 4.6 mm) column with isocratic elution of phosphate buffer-pH 6.8 and acetonitrile in ratio of 63:37 with detection at 230 nm. Second method is an HPTLC method on HPTLC silica plates using ethyl acetate: ethanol (75%): ammonia (5:5:0.05 v/v) mobile phase followed by densitometric scanning at 230 nm. The methods were applied successfully for analysis of VON and ASP mixture in laboratory-prepared tablets and the methods were validated in regards to linearity, precision, accuracy and selectivity. The proposed methods are assessed for their greenness and whiteness as well using the "Analytical GREEnness Metric Approach", "Complementary Modified Green Analytical Procedure Index" and the new algorithm "RGB 12 model" (Red-Green-Blue) and proved the greenness and the sustainability of the methods in the routine assay of the newly marketed formulation.

Abstract:Darolutamide is an oral nonsteroidal androgen used to delay the process of prostate cancer. A spectrofluorometric method was advanced for quantifying Darolutamide in pharmaceutical formulation and spiked human plasma. The method depends on the fluorescence derivatization of Darolutamide with NBD‐Cl. Darolutamide is an oral nonsteroidal androgen receptor antagonist used to delay the process of prostate cancer to metastatic disease and to increase the quality of life for people with advanced prostate cancer. Here, a second spectrofluorimetric method was advanced for quantifying Darolutamide in pharmaceutical formulation and spiked human plasma. This method depends on the fluorescence derivatization of Darolutamide with 4‐chloro‐7‐nitrobenzo‐2‐oxa‐1,3‐diazole (NBD‐Cl) at 75°C in a (pH 9) of borate buffer to produce a fluorescent derivative that can be detected at 520 nm after excitation at 460 nm. The method has been validated using ICH criteria, and it demonstrated linearity in the range 5–200 ng ml−1. The limit of detection (LOD) and limit of quantitation (LOQ) were 1.15 and 3.84 nm, respectively. The proposed method was applied precisely and accurately for quantifying Darolutamide within the pharmaceutical formulation and spiking human plasma without any interferences. Moreover, the method's sustainability was evaluated and compared with the published method using two greenness assessment tools termed analytical eco‐scale and Analytical GREEnness (AGREE). These findings suggest that the method is more sustainable than the published method.

Abstract:A novel spectrofluorimetric method has been developed for determination of antazoline (ANT) and tetryzoline (TET) in their pharmaceutical formulation. A combined application of synchronous spectrofluorimetry and second derivative mathematical treatment was developed. The proposed method depends on reacting the cited drugs with dansyl chloride (DNS‐Cl) being a suitable derivatizing agent generating highly fluorescent derivatives. A novel spectrofluorimetric method has been developed for determination of antazoline (ANT) and tetryzoline (TET) in their pharmaceutical formulation. A combined application of synchronous spectrofluorimetry and second derivative mathematical treatment was developed. The proposed method depends on reacting the cited drugs with dansyl chloride (DNS‐Cl) being a suitable derivatizing agent generating highly fluorescent derivatives measured at emission wavelengths of 703.0 and 642.0 nm after excitation wavelengths of 350.0 and 320.0 nm for ANT and TET, respectively. The joint use of synchronous spectrofluorimetry with second derivative mathematical treatment is for the first time to be developed and optimized in aid of using fluorescence data manager software generating second derivative peak amplitudes at 556.5 nm for ANT and 516.7 nm for TET. Linear responses have been represented over a wide range of concentration (0.5–12.0 μg/mL for ANT and 0.5–10.0 μg/mL for TET). Additionally, statistical comparison of the developed method with the official ones has been carried out where no significant difference was found. Additionally, greenness profile assessment was accomplished by means of four metric tools. Indeed, the method developed is found to be precise, sensitive, and discriminating to assess the cited drugs for regular analysis.

Abstract:An ultrafast and green method was proposed for the microwave-assisted synthesis of N, S-doped carbon quantum dots (N,S-CDs) in only 90 s. The proposed method used thiourea as a source of sulfur and nitrogen and Dill ( Anethum vulgaris ) as a green carbon source. Characterization of the produced N,S-CDs was performed using several microscopic and spectroscopic methods. They were utilized as a fluorescent nanoprobe for the entacapone (ENT) and clonazepam (CLZ) estimation. After excitation at 310 nm, the developed N,S-CDs revealed an emission band at 380 nm. HR-TEM showed spherical N,S-CDs with a size range of 1–5 nm. The proposed method is the first report on the estimation of each of ENT and CLZ using N,S-CDs and the first one to use Dill as a carbon source for the preparation of N,S-CDs. The prepared N,S-CDs exhibited native fluorescence that was quantitatively quenched by each of ENT and CLZ over the ranges of 1.0-15.0 and 2.0 – 25.0 μM, respectively. The mechanism of N,S-CDs fluorescence quenching was carefully studied. The suggested approach was employed for the determination of ENT and CLZ in their pharmaceutical formulations and human plasma samples with acceptable % RSD and % recoveries (98.50-101.31%). Moreover, the content uniformity testing of the CLZ tablets was efficiently carried out according to the United States Pharmacopoeia recommendations. The excellent greenness and eco-friendliness of the proposed approach were demonstrated using the complementary Green Analytical Procedure Index (ComplexGAPI) and the Analytical GREEnness (AGREE). Validation of the proposed method was performed using ICHQ2 (R1) guidelines.

Abstract:marker that can be used further to investigate differences in the two main aetiological types of diabetes. The parallel of our results for type I diabetes with those for ankylosing spondylitis is striking. In both there are strong associations with particular HLA markers and a significant increase in the proportion of non-secretors. This is further indirect evidence to support the suggestion that an infectious agent is implicated in the initiation of type I diabetes in some genetically predisposed paiients. Determining the secretor state of patients who have various viral infections would be a useful first step in investigating this hypothesis. We suggest that the initial susceptibility to some viral infections might be greater in the non-immune, non-secretor host. The specific immune response to the putative infectious agent, controlled by the class II HLA DR3/4 genes, might contribute to pathogenic sequelae leading to the diabetic state.

Abstract:Pemetrexed is a chemotherapeutic medicine, under the trade name Alimta. Malignant pleural mesothelioma patients. Its application in lung cancer has been studied. Here in, a second spectrofluorimetric method was advanced for quantifying of Pemetrexed in pharmaceutical formulation and spiked human plasma. That method depends on fluorescence derivatization of Pemetrexed with 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) at 75 °C in a (pH 9) of borate buffer to produce a fluorescent derivative which can be detected at 520 nm afterwards excitation at 460 nm. The method has been validated using ICH criteria, and it demonstrated linearity in a range of 2–120 ngmL-1. The proposed method was applied precisely and accurately for quantifying Pemetrexed within pharmaceutical formulation and spiking human plasma without any interferences. Moreover, the method's sustainability was evaluated and compared to the published method using two greenness assessment tools termed analytical eco-scale and Analytical GREENness (AGREE). That findings suggest that the method is more sustainable than the published method.

As an analytical chemist, it's always a challenge to accurately and precisely determine a drug along with its metabolites with a single method. Hence, we aim to develop a simple and low-cost assay based on combination of vortex assisted-dispersive liquid-liquid microextraction (VADLLME) with smartphone based digital image colorimetry (DIC) as a promising platform for simple low-cost analysis of ASA and SA. The method comprises of three simple steps viz. (i) extraction and pre-concentration of ASA/SA by VADLLME, (ii) reaction of ASA/SA with Trinder reagent on a filter paper strip to produce blue colour, and (iii) DIC of developed colour with imageJ software. Design of experiment approach comprising of Plackett-Burman design (PBD) and central composite design (CCD) were used for optimization of parameters affecting the efficiency of the proposed method. Under the optimized conditions, the limits of detection (LOD) and the limit of quantitation (LOQ) were found to be 8.88 and 29.0 μg mL−1, respectively. Calibration plot demonstrated good linearity in the range of 50–400 μg mL−1, with coefficients of determination (R2) higher than 0.993 and standard deviation less than 10%. The proposed method has been successfully applied for the estimation of ASA/SA in pharmaceutical formulations (tablets), cosmetic product and human urine samples, with percent recoveries (%R) ranging from 92.14–99.6%. In addition, the green character of the proposed method from sample collection to final determination has been evaluated by using ComplexGAPI index.

Abstract:The cutting-edge combination of fluvoxamine (FVM) and ivermectin (IVM) has been presented as a proposed dosage form for the treatment of COVID-19 infections in early diagnosed patients. The main objective of this work is to develop simple, sensitive, and efficient methods for the synchronous quantification of FVM and IVM without any prior separation. Four green UV-methods were employed for the synchronous quantification, namely: Fourier functions convolution of absorption spectra, FFAS, Fourier functions convolution of derivative spectra of absorption curves, FFDS, Fourier function convolution of ratio spectra of absorption curves, FFRS and the dual-wavelength method, DWM. FFRS and DWM approaches can be able to reconcile the two components' significantly interfering spectrum presented in this commixture. Good linearity was checked in the range of 5–40, and 2.5–25 μg/mL for the FVM, and IVM, respectively. All approaches developed have been recommended in compliance with ICH principles. Furthermore, the approaches' greenness was predestined by "National Environmental Method Index" (NEMI), "Analytical GREEnness metric (AGREE)", the "Analytical Eco-Scale", and the "Green Analytical Procedure Index" (GAPI). In addition, spider diagram was utilized for the assessment of the greenness index of the solvent used. Beside greenness, the sustainability of our methods was investigated using the HEXAGON tool. Continuing the constant pursuit of greenness, drug-drug interactions (DDIs) between FVM & IVM were predicted by insilico tools to ensure the safety of the suggested mixture as a preliminary step before invitro and in vivo studies. Because they were deemed sustainable, affordable, and successful, the suggested UV-methods may be used for routine quality control investigations of the indicated formulations FVM & IVM.
