Abstract:An efficient gas chromatography-mass spectrometry method was developed and validated for determination of sterol oxidation products (SOPs) in edible oils. The sample preparation involved cold saponification, liquid-liquid extraction, solid-phase extraction on a silica gel cartridge, and trimethylsilylation by N-methyl-N-(trimethylsilyl) heptafluorobutyramide with 5% 1-methyl imidazole. The trimethylsilyl ether derivatives of SOPs were separated by gas chromatography with a 30 m DB-5MS capillary column and quantified by a mass spectrometer in selective ion monitoring mode. 5a-Cholestane and 19-hydroxycholesterol were used as dual internal standards. The calibration curves for each compound showed correlation coefficients (R-2) better than 0.98. The detection limits were below 12.9 ng mL(-1) (except for epoxides). The intra- and inter-day determination precisions for diversiform SOPs were <10% in relative standard deviations; the recoveries ranged within 89.72% and 117.42%. The developed approach was successfully applied to study the presence of thirty-four different SOPs present at low levels in camellia, olive, sesame, peanut, rapeseed, rice bran, soybean and corn oils.
Oxidative lipidomics to elucidate the non-volatile derivatives of four typical triglycerides in vegetable oils under simulated frying conditions

Abstract:Vegetable oils with different saturations have varied composition of triglycerides (TGs) and produce different non-volatile derivatives during oxidation. Precise characterization of the non-volatile derivatives of TGs is essential for understanding the degradation of TGs and the production pattern of non-volatile derivatives. Oxidative lipidomics was combined with collision-induced dissociation and electron-activated dissociation to elucidate the precise structures of non-volatile derivatives produced under simulated frying conditions by 1,3-dipalmitoyl-2-oleoylglycerol (POP), triolein (OOO), trilinolein (LLL), and trilinolenin (LnLnLn). The results indicate that the unsaturated fatty acyl chains at the sn-2 position were more susceptible to oxidation compared with those at the sn-1/3 position. Species of non-volatile derivatives included epoxy-, hydroperoxy-, hydroxy-, and oxo-TGs, as well as degradation products. The potential reaction pathways of TGs and their non-volatile derivatives were also proposed. This study elucidated oxidative degradation mechanisms of the four typical TGs and provided a theoretical basis for changes of vegetable oils during frying.
Comprehensive molecular mapping of vegetable oil autoxidation products by NMR-based oxylipidomics

Abstract:Lipid oxidation is one the major causes of food deterioration. Current advancement in mechanistic understanding is limited by the lack of high-throughput methods that can simultaneously quantify a wide range of oxidation product classes, such as lipid hydroperoxides, epoxides, ketones, hydroxides, and aldehydes. Here, we introduce an NMR-based ‘oxylipidomics’ platform by providing the annotation of 42 substructures formed during lipid oxidation in vegetable oils. The annotated substructures accounted for respectively 93, 90 and 70 % of the oxidation products of triolein, trilinolein and trilinolenin. The spectral assignments allowed for quantification of lipid oxidation products in vegetable oil at class level (e.g., epoxides) and substructure level (e.g., trans-epoxides) at the commonly available field strength of 14.1 T (600 MHz). We anticipate that our workflow will enable rapid assessment of health risks, unravelling of precursor-sensory relationships, rational design of antioxidant strategies, and in-depth mechanistic studies into food lipid oxidation.

Abstract:Lipid oxidation of edible oil during frying process is a complex process and involves free radical chain reactions. Soybean oil with high amount of unsaturated fatty acids was used to investigate the impact of frying time on oxidative stability of oil. Electron spin resonance (ESR) spectroscopy technique was applied to identify and quantify the formed radicals, along with the assessment of physicochemical parameters including peroxide value, oxidative stability, fatty acid composition, and volatile profile. Results showed that the amount of formed free radicals determined by ESR in frying oil increased with the prolongation of frying time. The availability of this method was compared with physicochemical properties and the well correlation coefficients were obtained. Besides, main volatile aldehyde compounds produced by β-scission homolytic cleavage of peroxide group in frying oil during thermal oxidation were derived from oxidation of oleic and linoleic acid.

Abstract:Stomatal movements, which regulate gas exchange in plants, involve pronounced changes in the shape and volume of the guard cell. To test whether the changes are regulated by actin filaments, we visualized microfilaments in mature guard cells and examined the effects of actin antagonists on stomatal movements. Immunolocalization on fixed cells and microinjection of fluorescein isothiocyanate-phalloidin into living guard cells of Commelina communis L. showed that cortical microfilaments were radially distributed, fanning out from the stomatal pore site, resembling the known pattern of microtubules. Treatment of epidermal peels with phalloidin prior to stabilizing microfilaments with m-maleimidobenzoyl N-hydroxysuccimimide caused dense packing of radial microfilaments and an accumulation of actin around many organelles. Both stomatal closing induced by abscisic acid and opening under light were inhibited. Treatment of guard cells with cytochalasin D abolished the radial pattern of microfilaments; generated sparse, poorly oriented arrays; and caused partial opening of dark-closed stomata. These results suggest that microfilaments participate in stomatal aperture regulation.

Abstract:Lipid peroxidation, the most aggressive reaction in food, results in the formation of reactive organic compounds that detrimentally impact food sensory qualities and consumers’ health. While controlled lipid peroxidation can enhance flavors and appearance in certain foods, secondary peroxidation products lead to sensory deterioration in a variety of products, such as oils, alcoholic beverages, and meat. This publication reviews the use of modern analytical techniques for detecting and quantifying carbonyl compounds, i.e., secondary lipid peroxidation products. The paper focuses specifically on microextraction-based methods: dispersive liquid-liquid microextraction (DLLME), solid-phase microextraction (SPME), and gas-diffusion microextraction (GDME). These techniques offer efficient and sensitive approaches to extracting and quantifying lipid oxidation products and contribute to the understanding of oxidative deterioration in various food products. The review outlines recent advancements, challenges, and limitations in these microextraction techniques, as well as emphasizes the potential for further innovation and improvement in the field of food analysis.

Abstract:Lipid oxidation is one of the major causes of food spoilage for lipid-rich foods. In particular, oil-in-water emulsions, like mayonnaises and spreads, are prone to oxidation due to the increased interfacial area that facilitates contact between the lipids and hydrophilic pro-oxidants present in the water phase. Polar, amphiphilic lipid species present at the oil/water interface, like the mono- (MAGs) and di-acylglycerols (DAGs), act as oxidation starters that initiate subsequent oxidation reactions of the non-polar lipids in the oil droplets. A comprehensive two-dimensional liquid chromatography (LC×LC) method with evaporative light-scattering detection (ELSD) was set up to study the composition of the complex mixture of oxidized polar and non-polar lipids. The LC×LC-ELSD method employs size exclusion chromatography (SEC) in the 1D (1st dimension) to separate the various lipid species according to size. In the 2D (2nd dimension), normal-phase liquid chromatography (NPLC) is used to separate the fractions according to their degree of oxidation. The coupling of SEC with NPLC yields a good separation of the oxidized triacylglycerols (TAGs) from the large excess of non-oxidized TAGs. In addition, it allows the isolation of non-oxidized DAGs and MAGs that usually interfere with the detection of a variety of oxidized products that have similar polarities. This method facilitates elucidating how lipid composition affects oxidation kinetics in emulsified foods and will aid in the development of more oxidation-stable products.
Release Kinetics Studies of Early-Stage Volatile Secondary Oxidation Products of Rapeseed Oil Emitted during the Deep-Frying Process

Abstract:The research concerns the use of proton transfer reaction mass spectrometer to track real-time emissions of volatile secondary oxidation products released from rapeseed oil as a result of deep-frying of potato cubes. Therefore, it was possible to observe a sudden increase of volatile organic compound (VOC) emissions caused by immersion of the food, accompanied by a sudden release of steam from a potato cube and a decrease of the oil temperature by more than 20 °C. It was possible to identify and monitor the emission of major secondary oxidation products such as saturated and unsaturated aldehydes, namely acrolein, pentanal, 2-hexenal, hexanal, 2-nonenal and 2-decenal. Each of them has an individual release characteristic. Moreover, the impact of different initial frying temperatures on release kinetics was investigated. Subsequently, it was possible to approximate the cumulative emission by a second-degree polynomial (R2 ≥ 0.994). Using the proposed solution made it possible for the first time to observe the impact of the immersion of food in vegetable oil on the early emission of thermal degradation products oil.

Abstract:Deep-fat frying is a popular cooking technique in the world and vegetable oil is widely used as the frying medium in this process. Deep-fat frying produces both desirable and undesirable compounds through various chemical reactions. Undesirable chemical compounds are formed mainly through hydrolysis, oxidation and polymerization reactions. These compounds lower the oxidative stability thereby, the quality of oil and food. Antioxidants are added to improve the oxidative stability of oil during deep-fat frying by lowering the free radical action in frying oil. Even though artificial antioxidants are added to frying oils to lower the effects of undesirable chemical reactions and their products, the efficiency of artificial antioxidants decrease with increasing temperature and may cause adverse health effects to the consumer. Natural extracts of rosemary (Rosmarinus officinalis), sage (Salvinia officinalis), tea (Camelia sinensis), oregano (Origanum vulgare) and barley (Hordeum vulgare) are stable under frying conditions and act as effective antioxidants during deep-fat frying. Despite confirmation by numerous research, the use of natural compounds as antioxidants in deep-fat frying is not popular in the food industry. Hence this review explores the major chemical reactions in vegetable oils during deep-fat frying and the effect of natural compounds and extracts in interrupting these undesirable chemical reactions.

Abstract:One of the commonly used food preparation methods is frying. Fried food is admired by consumers due to its unique taste and texture. Deep frying is a process of dipping food in oil at high temperature, usually 170–190 °C, and it requires a relatively short time. The aim of this study was to analyze the thermo-oxidative changes occurring during the deep frying of products such as potatoes and tofu in cold pressed rapeseed oils and palm olein. Cold pressed rapeseed oil from hulled seeds (RO), cold pressed high oleic rapeseed oil from hulled seeds (HORO), and palm olein (PO) (for purposes of comparison) were used. Characterization of fresh oils (after purchase) and oils after 6, 12, and 18 h of deep frying process of a starch product (potatoes) and a protein product (tofu) was performed. The quality of oils was analyzed by determining peroxide value, acid value, p-anisidine value, content of carotenoid and chlorophyll pigments, polar compounds, smoke point, color (CIE L*a*b*), fatty acids content and profile, calculation of lipid nutritional quality indicators, and oxidative stability index (Rancimat). Cold pressed high oleic rapeseed oil was more stable during deep frying compared to cold pressed rapeseed oil, but much less stable than palm olein. In addition, more thermo-oxidative changes occurred in the tested oils when deep frying the starch product (potatoes) compared to the deep frying of the protein product (tofu).

Abstract:The oxidative degradation of lipids in vegetable oils during thermal processing may present a risk to human health. However, not much is known about the evolution of lipids and their non-volatile derivatives in vegetable oils under different thermal processing conditions. In the present study, a pseudotargeted oxidative lipidomics approach was developed and the evolution of lipids and their non-volatile derivatives in palm oil, rapeseed oil, soybean oil, and flaxseed oil under different thermal processing conditions was investigated. The results showed that thermal processing resulted in the oxidative degradation of TGs in vegetable oils, which generated oxTGs, DGs, and FFAs, as well as TGs with smaller molecular weights. The lower the fatty acid saturation, the more severe the oxidative degradation of vegetable oils and thermal processing at high temperatures should be avoided if possible. From the accumulation of oxTGs concentrations, the hazards during thermal processing at high temperatures were, in descending order, soybean oil, rapeseed oil, flaxseed oil, and palm oil. The non-volatile potential markers were screened in palm oil, rapeseed oil, soybean oil, and flaxseed oil for 1, 7, 5, and 2 markers related to thermal processing time, respectively. The study provided suggestions for the consumption of vegetable oils from multiple perspectives and identified markers for monitored oxidative degradation of vegetable oils.
Investigation into the Reduction of Palm Oil in Foods by Blended Vegetable Oils through Response Surface Methodology and Oxidative Stability Tests

Abstract:Recently, there has been a significant transition in the dietary preferences of consumers toward foods containing health-promoting compounds. In addition, as people's environmental awareness increases, they are increasingly looking for sustainable solutions. Palm oil, an oil used extensively by the food industry, does not fit these criteria. This study investigated the development of a complex oil blend consisting of commonly used vegetable oils such as corn, rapeseed, sunflower, and palm oil. The aim was to find the optimal blended oil and compare this combination with palm oil in terms of its oxidative stability, antioxidant capacity, and the composition of bioactive compounds (i.e., fatty acids, tocopherols, and carotenoids). Palm oil was found to have greater oxidative stability as a result of its increased concentration of saturated fatty acids. The optimal blended oil, which consisted of corn and rapeseed oil at a ratio of 4:3 w/w, inhibited the superior antioxidant activity, showing a ~33% increase in DPPH• inhibition activity. ATR-FTIR spectra further verified the existence of a significant quantity of saturated fatty acids in palm oil and unsaturated fatty acids in the blended oil. Finally, several correlation analyses revealed interesting connections between oil samples and investigated parameters. This work has the potential to establish a basis for the mass production of oil blends that possess high concentrations of antioxidant compounds and reduce the use of palm oil.

Abstract:Background Recent increases in bacterial sexually transmitted infections (STI) and risk behavior have coincided with the introduction of antiretroviral therapy (ART) in homosexual communities of industrialized countries. The reasons for these increases are not fully understood. Goal The goal of this study was to understand the various effects of ART on risk behaviors and STI. Objective The objective of this study was to assess the independent impact of the change in the transmission dynamics of HIV/AIDS as a result of the wide-scale use of ART on a bacterial STI. Study Design We developed a mathematical model of bacterial STI and treated/untreated HIV/AIDS infection for an open homosexual population. At the individual level, we assume that susceptible and healthy HIV-positive individuals do not increase their risk behavior as a result of ART over time. However, individuals with AIDS, who are successfully treated with ART, can resume sexual activity. The impact of the wide-scale use of ART on risky behavior, STI, and HIV/AIDS was evaluated over a wide range of assumptions on treatment use, ART efficacy, and population characteristics. Results Over 10 years, 0% to 55% new bacterial STI could be attributed to the wide-scale use of ART as a result of more modest increases (0–25%) in risky sex occurring at the population level rather than at the individual level. These increases have a negative impact on HIV if coverage is too low. Increasing treatment coverage helps to prevent more HIV infections despite larger increases in risky sex and STI that is predicted to ensue. Conclusion Taking the differential impact of wide-scale use of ART into account helps to interpret recent behavioral and STI trends. Our results have implications for prevention strategies and for the formulation of public health policies. A better understanding of the differential impact of ART on sexual network over time is required.

Abstract:The induction of a type C virus from a strain of human embryonic lung cells (HEL-12) by iododeoxyuridine (IdUrd) was examined at various times during in vitro propagation. IdUrd did not elicit type C virus production immediately following initiation of cultures from frozen primary HEL-12 cells. After overnight treatment with 30 microgram/ml IdUrd the cells expressed viral antigens and produced a type C virus between the 25th and 80th day of in vitro growth. Production of the induced type C virus was transient. Single-cell clones of the parental HEL-12 strain were likewise susceptible to type C virus production by IdUrd. The ability of IdUrd to induce virus terminated with the onset of spontaneous type C virus production from HEL-12 cells at between 80 and 120 days of in vitro growth.

Abstract:One of the main concerns about the use of edible vegetable oils in food industry frying processes is the oxidative degradation due to the high temperatures, presence of oxygen, ultraviolet radiation, and the presence of metal ions. The main compounds formed during oxidative reactions include aldehydes, ketones, alcohols, and carboxylic acids, being some of these electroactive compounds, which can be used with quantification purposes. In this work, cyclic voltammetry and a flow method based on headspace sampling with amperometric detection were performed, followed by principal component and cluster analysis to classify palm olein, soya bean, and sunflower oil samples according to their degradation state. The electrochemical techniques were based on the use of 1-butyl-3-methylimidazolium hexafluorophosphate as conductive media. The amperometric profile information provided a clearer classification than the voltammetric profile. Additionally, the amperometric results were applied to determine iodine value and aldehyde content by means of a partial least square regression. The values obtained were statistically similar than the estimated using 1H nuclear magnetic resonance spectroscopy and HPLC. The combination of electrochemical techniques and chemometric analysis offers valuable information for classification and quantification purposes.

Novel theoretical database-assisted UHPLC-Q-TOF/MS strategy for profiling and identifying oxidized triglycerides in pharmaceutical excipient soybean oil

Abstract:Pharmaceutical excipient soybean oil is widely used in injections. Its main components, triglycerides, are easily oxidized due to their unsaturated fatty acyls, raising safety concerns. However, it is hard to analyze those oxidized triglycerides due to their diverse compositions and low abundance. In this study, all theoretical oxidized triglycerides were predicted and a database consisting of 329 oxidized triglycerides was constructed. Then, a novel theoretical database-assisted ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF/MS) strategy was developed to finely profile and identify oxidized triglycerides in soybean oil. A total of 106 and 116 oxidized triglycerides were identified and relatively quantified in oxidized soybean oil and long-term stored soybean oil and preparations. It was found that oxidized triglycerides containing carbonyl groups were significantly more prevalent than other forms and oxidized triglycerides with two oxidized fatty acyl chains had the highest relative abundance. Fifteen markers indicating the oxidation of soybean oil were discovered. This strategy could rapidly and directly analyze the oxidized triglycerides and assign their fatty acyl compositions for the first time. This study will improve the quality control of soybean oil and its preparations.

Abstract:The aim of the present study is to determine the repeatability and reproducibility of lipid oxidation analyses performed under the same conditions in an interlaboratory study by 16 participants. Conjugated dienes measured over 9 weeks in sunflower and rapeseed oil at different temperatures were aggregated in the activation energy and statistically analyzed. Accelerated storage tests are frequently used to assess the oxidative stability of foods and related systems due to its reproducibility. Various methods and experimental conditions are used to measure lipid oxidation. Differences between laboratories make it necessary to determine the repeatability and reproducibility of oxidation tests performed under the same conditions. The objective of the present interlaboratory study was to evaluate the outcome of a storage test for two different bulk oils, sunflower oil (SFO) and rapeseed oil (RSO), during a period of 9 weeks at 20°C, 30°C, 40°C, and 60°C. Sixteen laboratories were provided with bottled oils and conducted the storage tests according to a detailed protocol. Lipid oxidation was monitored by the formation of conjugated dienes (CD) and the activation energy (Ea) was determined for comparative purposes and statistically evaluated. An increase in CD formation was observed for both oils when the storage temperature was increased in all laboratories. The Ea,1 ranged from 47.9 to 73.3 kJ mol−1 in RSO and from 27.8 to 62.6 kJ mol−1 in SFO, with average values of 58.2 and 46.8 kJ mol−1, respectively. The reproducibility coefficients were 10.9% and 18.2% for RSO and SFO, respectively.

Abstract:Edible oils are indispensable food components, because they are used for cooking or frying. However, during processing, transport, storage, and consumption, edible oils are susceptible to oxidation, during which various primary and secondary oxidative products are generated. These products may reduce the nutritional value and safety of edible oils and even harm human health. Therefore, analyzing the oxidation of edible oil is essential to ensure the quality and safety of oil. Oxidation is a complex process with various oxidative products, and the content of these products can be evaluated by corresponding indexes. According to the structure and properties of the oxidative products, analytical methods have been employed to quantify these products to analyze the oxidation of oil. Combined with proper chemometric analytical methods, qualitative identification has been performed to discriminate oxidized and nonoxidized oils. Oxidative products are complex and diverse. Thus, proper indexes and analytical methods should be selected depending on specific research objectives. Expanding the mechanism of the correspondence between oxidative products and analytical methods is crucial. The underlying mechanism, conventional indexes, and applications of analytical methods are summarized in this review. The challenges and perspectives for future applications of several methods in determining oxidation are also discussed. This review may serve as a reference in the selection, establishment, and improvement of methods for analyzing the oxidation of edible oil.
