Abstract:1. Transmembrane potential (TMP) responses of Purkinje cells (PCs) in isolated turtle cerebellum to externally applied quasi-steady-state electric fields aligned with the dendritic axis were continuously measured using simultaneous intracellular and extracellular recording. TMP was obtained by subtraction of extracellular voltage fields from intracellular potential recorded at the same depth in the cerebellum. 2. The applied field changed the TMP with the polarity and amplitude dependent on the location on the PC membrane. This response at a given location increased linearly with external field up to a threshold level, beyond which active responses appeared. 3. The basic effect on TMP consisted of depolarization in the half of the dendrite towards which the fields were directed, and hyperpolarization in the other half. A pooled TMP depth-profile shows a steady increase in polarization from the middle of the molecular layer towards each end. This profile correlates with that predicted from previously proposed cable models, giving them empirical support for the first time. 4. Active responses were triggered by the field-induced depolarization. Tetrodotoxin (TTX)-sensitive action potentials arose with the primary depolarization in the somatic region. Notched, Ca2+-dependent action potentials arose with primary depolarization in the distal and mid-dendritic regions. 5. A TTX-sensitive voltage plateau was triggered by TMP-depolarization in the proximal region. It in turn activated Na+-spike trains. The frequency of spiking was proportional to the external field. At around 160 spikes/s, the Na+ spikes inactivated, and the TMP level rose to a more depolarized plateau. This latter plateau was also TTX-sensitive. 6. During depolarization of the distal dendritic region, sometimes a Ca2+-dependent plateau was observed. It appears to be associated with a small conductance increase. 7. Field-induced hyperpolarization suppressed local spiking and voltage plateaux, but remote Ca2+ spikes with reduced amplitude appeared in recordings from the proximal region. Similarly, in the distal region, low-amplitude, remote Na+ spikes and a Na+ plateau were observed superimposed on the hyperpolarizing baseline. The Na+ plateau apparently did not contribute to shunting of membrane currents in the distal dendrite. 8. The phase characteristics of the action potentials correlate with the modulation pattern noted in our extracellular study (Chan & Nicholson, 1986). Thus, the extracellular units ("giant spikes") were probably Na+ spikes activated in the soma and spread distally. Physiologically Evoked Neuronal Current MRI in a Bloodless Turtle Brain: Detectable or Not?

Abstract:Contradictory reports regarding the detection of neuronal currents have left the feasibility of neuronal current MRI (ncMRI) an open question. Most previous ncMRI studies in human subjects are suspect due to their inability to separate or eliminate hemodynamic effects. In this study, we used a bloodless turtle brain preparation that eliminates hemodynamic effects, to explore the feasibility of detecting visually-evoked ncMRI signals at 9.4 T. Intact turtle brains, with eyes attached, were dissected from the cranium and placed in artificial cerebral spinal fluid. Light flashes were delivered to the eyes to evoke neuronal activity. Local field potential (LFP) and MRI signals were measured in an interleaved fashion. Robust visually-evoked LFP signals were observed in turtle brains, but no significant signal changes synchronized with neuronal currents were found in the ncMRI images. In this study, detection thresholds of 0.1% and 0.1 degrees were set for MRI magnitude and phase signal changes, respectively. The absence of significant signal changes in the MRI images suggests that visually-evoked ncMRI signals in the turtle brain are below these detectable levels.

Abstract:Isolated turtle brain/eye preparation has recently been used as a bloodless animal model for detecting the magnetic resonance imaging (MRI) signal changes produced by visually evoked neuronal currents. The present work aims to determine whether checkerboard-patterned or full field flash (blank) stimulation should be used in order to achieve stronger neuronal responses in turtle brain/eye preparation. The knowledge gained in this study is essential for optimizing the visual stimulation methods in functional neuroimaging studies using turtle brain/eye preparation. In this study, visually evoked local field potentials (LFPs) were measured and compared in turtle visual cortex and optic tectum elicited by checkerboard and full field flash stimuli with three different inter-stimulus intervals (ISIs=5, 10, and 16s). It was found that the behavior of neuronal adaptation in the cortical and tectal LFP signals for checkerboard stimulation was comparable to flash stimulation. In addition, there was no significant difference in the LFP peak amplitudes (ISI=16s) between these two stimuli. These results indicate that the intensity of neuronal responses to checkerboard is comparable to flash stimulation. These two stimulation methods should be equivalent in functional neuroimaging studies using turtle brain/eye preparation.

Abstract:PurposeDespite the efforts that have been devoted to detecting the transient magnetic fields generated by neuronal firing, the conclusion that a functionally relevant signal can be measured with MRI is still controversial. For human studies of neuronal current MRI (nc‐MRI), the blood‐oxygen‐level‐dependent (BOLD) effect remains an irresolvable confound. For tissue studies where hemoglobin is removed, natural sensory stimulation is not possible. This study investigates the feasibility of detecting a physiologically induced nc‐MRI signal in vivo in a BOLD‐free environment.MethodsThe cephalopod mollusc Octopus bimaculoides has vertebrate‐like eyes, large optic lobes (OLs), and blood that does not contain hemoglobin. Visually evoked potentials were measured in the octopus retina and OL by electroretinogram and local field potential. nc‐MRI scans were conducted at 9.4 Tesla to capture these activities.ResultsElectrophysiological recording detected strong responses in the retina and OL in vivo; however, nc‐MRI failed to demonstrate any statistically significant signal change with a detection threshold of 0.2° for phase and 0.2% for magnitude. Experiments in a dissected eye‐OL preparation yielded similar results.ConclusionThese findings in a large hemoglobin‐free nervous system suggest that sensory evoked neuronal magnetic fields are too weak for direct detection with current MRI technology. Abstract:The cerebellum is one of the most connected structures of the central nervous system and receives inputs over an extended frequency range. Nevertheless, the frequency dependence of cerebellar cortical processing remains elusive. In this work, we characterized cerebellar cortex responsiveness to mossy fibers activation at different frequencies and reconstructed the spread of activity in the sagittal and coronal planes of acute mouse cerebellar slices using a high-throughput high-density multielectrode array (HD-MEA). The enhanced spatiotemporal resolution of HD-MEA revealed the frequency dependence and spatial anisotropy of cerebellar activation. Mossy fiber inputs reached the Purkinje cell layer even at the lowest frequencies, but the efficiency of transmission increased at higher frequencies. These properties, which are likely to descend from the topographic organization of local inhibition, intrinsic electroresponsiveness, and short-term synaptic plasticity, are critical elements that have to be taken into consideration to define the computational properties of the cerebellar cortex and its pathological alterations.

Abstract:OBJECTIVE: Transcranial magnetic stimulation (TMS) represents a powerful technique to noninvasively modulate cortical neurophysiology in the brain. However, the relationship between the magnetic fields created by TMS coils and neuronal activation in the cortex is still not well-understood, making predictable cortical activation by TMS difficult to achieve. Our goal in this study was to investigate the relationship between induced electric fields and cortical activation measured by blood flow response. Particularly, we sought to discover the E-field characteristics that lead to cortical activation.APPROACH: Subject-specific finite element models (FEMs) of the head and brain were constructed for each of six subjects using magnetic resonance image scans. Positron emission tomography (PET) measured each subject's cortical response to image-guided robotically-positioned TMS to the primary motor cortex. FEM models that employed the given coil position, orientation, and stimulus intensity in experimental applications of TMS were used to calculate the electric field (E-field) vectors within a region of interest for each subject. TMS-induced E-fields were analyzed to better understand what vector components led to regional cerebral blood flow (CBF) responses recorded by PET.MAIN RESULTS: This study found that decomposing the E-field into orthogonal vector components based on the cortical surface geometry (and hence, cortical neuron directions) led to significant differences between the regions of cortex that were active and nonactive. Specifically, active regions had significantly higher E-field components in the normal inward direction (i.e., parallel to pyramidal neurons in the dendrite-to-axon orientation) and in the tangential direction (i.e., parallel to interneurons) at high gradient. In contrast, nonactive regions had higher E-field vectors in the outward normal direction suggesting inhibitory responses.SIGNIFICANCE: These results provide critical new understanding of the factors by which TMS induces cortical activation necessary for predictive and repeatable use of this noninvasive stimulation modality.

Abstract:Transcranial magnetic stimulation (TMS) delivers highly localized brain stimulations via non-invasive externally applied magnetic fields. This non-invasive, painless technique provides researchers and clinicians with a unique tool capable of stimulating both the central and peripheral nervous systems. However, a complete analysis of the macroscopic electric fields produced by TMS has not yet been performed. In this paper, we addressed the importance of the secondary E-field created by surface charge accumulation during TMS using the boundary element method (BEM). 3D models were developed using simple head geometries in order to test the model and compare it with measured values. The effects of tissue geometry, size and conductivity were also investigated. Finally, a realistically shaped head model was used to assess the effect of multiple surfaces on the total E-field. Secondary E-fields have the greatest impact at areas in close proximity to each tissue layer. Throughout the head, the secondary E-field magnitudes typically range from 20% to 35% of the primary E-field's magnitude. The direction of the secondary E-field was generally in opposition to the primary E-field; however, for some locations, this was not the case (i.e. going from high to low conductivity tissues). These findings show that realistically shaped head geometries are important for accurate modeling of the total E-field.

Abstract:Although transcranial magnetic stimulation (TMS) is a popular tool for both basic research and clinical applications, its actions on nerve cells are only partially understood. We have previously predicted, using compartmental modeling, that magnetic stimulation of central nervous system neurons depolarized the soma followed by initiation of an action potential in the initial segment of the axon. The simulations also predict that neurons with low current threshold are more susceptible to magnetic stimulation. Here we tested these theoretical predictions by combining in vitro patch-clamp recordings from rat brain slices with magnetic stimulation and compartmental modeling. In agreement with the modeling, our recordings demonstrate the dependence of magnetic stimulation-triggered action potentials on the type and state of the neuron and its orientation within the magnetic field. Our results suggest that the observed effects of TMS are deeply rooted in the biophysical properties of single neurons in the central nervous system and provide a framework both for interpreting existing TMS data and developing new simulation-based tools and therapies.

Abstract:To understand brain activity relating neurons to circuits to learning and behavior, we explored a bottom-up computational reconstruction of population signals arising from cerebellum granular layer. As a first implementation, using bio-realistic computational models of cerebellum granule cell, in vivo spike train patterns were computed and then translated into functional Magnetic Resonance Imaging, Blood Oxygen-Level Dependent (BOLD) signals. The BOLD response was generated from averaged activity arising from center-surround organization modeled by using excitatory-inhibitory ratios related to experimental data. The averaged responses were converted to BOLD signals using the balloon and modified Windkessel models. Although both models generated BOLD responses corresponding to neural activity, the temporal mismatch was attributed to the response by the delayed compliance parameter in the Windkessel model. The modeling suggests that experimental variability observed in the cerebellar micro-zones could be related to compliance chances, activation patterns and number of neurons. Although detailed neuro-vasculature information was not modeled, the advantage in this methodology is that cerebellar cortex may allow seemingly linear transformations of underlying spiking that could be then used to validate network reconstructions.
Traveling waves in developing cerebellar cortex mediated by asymmetrical Purkinje cell connectivity

Abstract:Correlated network activity is important in the development of many neural circuits. Purkinje cells are among the first neurons to populate the cerebellar cortex, where they sprout exuberant axon collaterals. We used multiple patch-clamp recordings targeted with two-photon microscopy to characterize monosynaptic connections between the Purkinje cells of juvenile mice. We found that Purkinje cell axon collaterals projected asymmetrically in the sagittal plane, directed away from the lobule apex. On the basis of our anatomical and physiological characterization of this connection, we constructed a network model that robustly generated waves of activity that traveled along chains of connected Purkinje cells. Consistent with the model, we observed traveling waves of activity in Purkinje cells in sagittal slices from young mice that require GABA(A) receptor-mediated transmission and intact Purkinje cell axon collaterals. These traveling waves are absent in adult mice, suggesting they have a developmental role in wiring the cerebellar cortical microcircuit.

Abstract:Transcranial direct-current stimulation (tDCS) of the cerebellum is a promising non-invasive neuromodulatory technique being proposed for the treatment of neurological and neuropsychiatric disorders. However, there is a lack of knowledge about how externally applied currents affect neuronal spiking activity in cerebellar circuits in vivo. We investigated how Cb-tDCS affects the firing rate of Purkinje cells (PC) and non-PC in the mouse cerebellar cortex to understand the underlying mechanisms behind the polarity-dependent modulation of neuronal activity induced by tDCS. Mice (n = 9) were prepared for the chronic recording of LFPs to assess the actual electric field gradient imposed by Cb-tDCS in our experimental design. Single-neuron extracellular recording of PCs in awake (n = 24) and anesthetized (n = 27) mice was combined with juxtacellular recordings and subsequent staining of PC with neurobiotin under anesthesia (n = 8) to correlate their neuronal orientation with their response to Cb-tDCS. Finally, a high-density Neuropixels recording system was used to demonstrate the relevance of neuronal orientation during the application of Cb-tDCS in awake mice (n = 6) In this study, we observe that Cb-tDCS induces a heterogeneous polarity-dependent modulation of the firing rate of Purkinje cells (PC) and non-PC in the mouse cerebellar cortex. We demonstrate that the apparently heterogeneous effects of tDCS on PC activity can be explained by taking into account the somatodendritic orientation relative to the electric field. Our findings highlight the need to consider neuronal orientation and morphology to improve tDCS computational models, enhance stimulation protocol reliability, and optimize effects in both basic and clinical applications.

Abstract:In thiopentone-anaesthetized and curarized cats we found that dorsal raphe (DR) stimulation could elicit long latency inhibitory,and in fewer cases,excitatory responses in cerebellar Purkinje cells (PC),and the peripheral inputs elicited by passive forepaw flexion-extension movements may enhance or reduce the response of PCs to DR stimulation.The results suggest that the raphe-cerebellar serotonergic fiber may modulate PCs activity and interact with the peripheral input.

Abstract:The cerebellum integrates motor commands with somatosensory, vestibular, visual and auditory information for motor learning and coordination functions. The deep cerebellar nuclei (DCN) generates the final output by processing inputs from Purkinje cells (PC), mossy and climbing fibers. We investigated the properties of PC connections to DCN cells using optogenetic stimulation in L7-ChR2 mice with in vivo multi electrode extracellular recordings in lobule IV/V of the cerebellar cortex and in the medial nuclei. DCN cells discharged phase locked to local field potentials in the beta, gamma and high frequency bands. We identified two groups of DCN cells with significant differences in action potential waveforms and firing rates, matching previously discriminated in vitro properties of GABAergic and non-GABAergic cells. PCs inhibited the two group of cells gradually (rate coding), however spike times were controlled for only non-GABAergic cells. Our results suggest that PC inputs temporally control the output of cerebellum and the internal DCN circuitry supports this phenomenon since GABAergic cells do not induce a temporal effect through local inhibition.

Prediction of magnetically induced electric fields in biological tissue

Abstract:There are many potential medical applications in which it is desirable to noninvasively induce electric fields. One such application that serves as the backdrop of this work is that of stimulating neurons in the brain. The magnetic fields necessary must be quite high in magnitude, and fluctuate rapidly in time to induce the internal electric fields necessary for stimulation. Attention is focused on the calculation of the induced electric fields commensurate with rapidly changing magnetic fields in biological tissue. The problem is not a true eddy current problem in that the magnetic fields induced do not influence the source fields. Two techniques are introduced for numerically predicting the fields, each employing a different gauge for the potentials used to represent the electric field. The first method employs a current vector potential (analogous to A in classical magnetic field theory where DEL x A = B) and is best suited to two-dimensional (2-D) models. The second represents the electric field as the sum of a vector plus the gradient of a scalar field; because the vector can be determined quickly using Biot Savart (which for circular coils degenerates to an efficient evaluation employing elliptic integrals), the numerical model is a scalar problem even in the most complicated three dimensional geometry. These two models are solved for the case of a circular current carrying coil near a conducting body with sharp corners.

Abstract:The cerebellum is a major motor structure. However, in humans, its efferent topographical organization remains controversial and indirectly inferred from neuroimaging and animal studies. Even central questions such as 'Can we evoke limb movements by stimulating the cerebellar cortex?' have no clear answer. To address this issue, we electrically stimulated the posterior cerebellum of 20 human patients undergoing surgery for tumours located outside this structure (e.g. pineal gland, quadrigeminal plate). Stimulation, delivered at a 60-Hz frequency for 2 s, evoked focal (single-joint) ipsilateral movements. Different regions were associated with the production of head (vermal lobule VI), face/mouth (hemispheric lobule VI) and lower-limb (hemispheric lobules VIIb-IX) responses. Upper-limb representations were more widely distributed. They intermingled with face/mouth representations in the superior posterior cerebellum (hemispheric lobule VI) and lower-limb representations in the inferior posterior cerebellum (hemispheric lobules VIIb-IX). No intra- or inter-limb somatotopy was found in these areas. Functionally, upper-limb (face/mouth movements) and upper limb-lower limb postural coordinations are major elements of our motor repertoire. Representation of these pairs of segments in common regions might favour the production of integrated motor behaviours. The intermediate region of the posterior cerebellum (hemispheric lobule VII and vermal lobules VII-VIII) was mostly silent. Latency results in conjunction with previous electrophysiological evidence in animals suggest that electrically evoked motor responses were not mediated by a cortical route but rather by brainstem structures. The potential role of this descending efferent pathway for fine motor control is discussed.

How Conductivity Boundaries Influence the Electric Field Induced by Transcranial Magnetic Stimulation in in vitro Experiments

Abstract:Background: Although transcranial magnetic stimulation (TMS) has become a valuable method for non-invasive brain stimulation, the cellular basis of TMS activation of neurons is still not fully understood. In vitro preparations have been used to understand the biophysical mechanisms of TMS, but in many cases these studies have encountered substantial difficulties in activating neurons. Objective/hypothesis: The hypothesis of this work is that conductivity boundaries can have large effects on the electric field in commonly used in vitro preparations. Our goal was to analyze the resulting difficulties in in vitro TMS using a simulation study, using a charge-based boundary element model. Methods: We decomposed the total electric field into the sum of the primary electric field, which only depends on coil geometry and current, and the secondary electric field arising from conductivity boundaries, which strongly depends on tissue and chamber geometry. We investigated the effect of the conductivity boundaries on the electric field strength for a variety of in vitro experimental settings to determine the sources of difficulty. Results: We showed that conductivity boundaries can have large effects on the electric field in in vitro preparations. Depending on the geometry of the air-saline and the saline-tissue interfaces, the secondary electric field can significantly enhance, or attenuate the primary electric field, resulting in a much stronger or weaker total electric field inside the tissue; we showed this using a realistic preparation. Submerged chambers are generally much more efficient than interface chambers since the secondary field due to the thin film of saline covering the tissue in the interface chamber opposes the primary field and significantly reduces the total field in the tissue placed in the interface chamber. The relative dimensions of the chamber and the TMS coil critically determine the total field; the popular setup with a large coil and a small chamber is particularly sub-optimal because the secondary field due to the air-chamber boundary opposes the primary field, thereby attenuating the total field. The form factor (length vs width) of the tissue in the direction of the induced field can be important since a relatively narrow tissue enhances the total field at the saline-tissue boundary. Conclusions: Overall, we found that the total electric field in the tissue is higher in submerged chambers, is higher if the chamber size is larger than the coil and if the shorter tissue dimension is in the direction of the electric field. Decomposing the total field into the primary and secondary fields is useful for designing in vitro experiments and interpreting the results.

Abstract:Information technology policy is too important to leave entirely to lawyers. This theme has been at the core of my work in the past decade. It is the reason I have entreated computing professionals, including readers of Communications, to become involved in the policy issues affecting their field. More recently, I have come to realize that information technology policy is also too important to leave entirely to technologists. This thought crystallized for me in June of 1997 when I attended the ACM Digital Library ’97 conference and heard a speaker repeatedly characterize intellectual property rights as “ornery.” Ornery seemed a curious word to use to describe this law. It evinces impatience and implicitly accuses its subject of stubbornness. A mule that won’t budge when you want it to take you back up to the top of the North Rim of the Grand Canyon is acting ornery. I admit that intellectual property rights are complicated, but I don’t think of them as ornery. In the nearly three centuries of its existence, copyright law has evolved an intricate architecture that provides different scopes of protection for different classes of works. Abstract:In continuation of earlier computer simulation studies 15 of the feedback inhibition exercised by the Golgi cells of the cerebellum, an attempt is made at modeling the behavior of Purkinje and basket cells under somewhat more complex functional circumstances. The simulation study is based on a realistic network model (with respect to numerical and metrical parameters, and consisting of more than 3 X 10(4) units) of the cat cerebellar cortex, derived from a recent quantitative histological and stereological analysis 9-12. As an input two identical pairs of foci of incoming mossy fiber activity are applied, separated from one another both spatially and temporally. The stimulation results showed that the assumption of a rigidly preaddressed wiring in the parallel fiber--Purkinje cell synaptic system (i.e. that parallel fibers would systematically select for contact Purkinje cells standing in register) would hardly be realistic because such a solution would offer no advantage whatever over a randomly connected synaptic system. The results would favor the contention that dendritic geometry of the Purkinje cells has a crucial significance in the sense of the concepts developed by W. Rall. The stimulation gives some important hints for the ways in which the specific inhibitory interneuron systems (Golgi and basket cells) add refinement to the operations of the network, particularly in securing integration of the influences of parallel fibers thrown into action by mossy input to spatially and temporally separated foci, especially at the level of Purkinje cells positioned in interfocal areas. Reducing the length of the parallel fibers to one-half of what was suggested by the stereological studies causes a serious breakdown of interfocal neighborhood interactions.
