Abstract:Magnesium has mechanical properties similar to those of bone and is being considered as a potential bone substitute. In the present study, two different pore sized scaffolds of the Mg alloy LAE442, coated with magnesium fluoride, were compared. The scaffolds had interconnecting pores of either 400 (p400) or 500 μm (p500). ß-TCP served as control. Ten scaffolds per time group (6, 12, 24, 36 weeks) were implanted in the trochanter major of rabbits. Histological analyses, μCT scans, and SEM/EDX were performed. The scaffolds showed slow volume decreases (week 36 p400: 9.9%; p500: 7.5%), which were accompanied by uncritical gas releases. In contrast, ß-TCP showed accelerated resorption (78.5%) and significantly more new bone inside (18.19 ± 1.47 mm 3 ). Bone fragments grew into p400 (0.17 ± 0.19 mm 3 ) and p500 (0.36 ± 0.26 mm 3 ), reaching the centrally located pores within p500 more frequently. In particular, p400 displayed a more uneven and progressively larger surface area (week 36 p400: 253.22 ± 19.44; p500: 219.19 ± 4.76 mm 2 ). A better osseointegration of p500 was indicated by significantly more trabecular contacts and a 200 μm wide bone matrix being in the process of mineralization and in permanent contact with the scaffold. The number of macrophages and foreign body giant cells were at an acceptable level concerning resorbable biomaterials. In terms of ingrown bone and integrative properties, LAE442 scaffolds could not achieve the results of ß-TCP. In this long-term study, p500 appears to be a biocompatible and more osteoconductive pore size for the Mg alloy LAE442.

Abstract:The magnesium alloy LAE442 emerged as a possible bioresorbable bone substitute over a decade ago. In the present study, using the investment casting process, scaffolds of the Magnesium (Mg) alloy LAE442 with two different and defined pore sizes, which had on average a diameter of 400 μm (p400) and 500 μm (p500), were investigated to evaluate degradation and osseointegration in comparison to a ß‐TCP control group. Open‐pored scaffolds were implanted in both greater trochanter of rabbits. Ten scaffolds per time group (6, 12, 24, and 36 weeks) and type were analyzed by clinical, radiographic and μ‐CT examinations (2D and 3D). None of the scaffolds caused adverse reactions. LAE442 p400 and p500 developed moderate gas accumulation due to the Mg associated in vivo corrosion, which decreased from week 20 for both pore sizes. After 36 weeks, p400 and p500 showed volume decreases of 15.9 and 11.1%, respectively, with homogeneous degradation, whereas ß‐TCP lost 74.6% of its initial volume. Compared to p400, osseointegration for p500 was significantly better at week 2 postsurgery due to more frequent bone‐scaffold contacts, higher number of trabeculae and higher bone volume in the surrounding area. No further significant differences between the two pore sizes became apparent. However, p500 was close to the values of ß‐TCP in terms of bone volume and trabecular number in the scaffold environment, suggesting better osseointegration for the larger pore size.
In vivo investigation of open-pored magnesium scaffolds LAE442 with different coatings in an open wedge defect

Abstract:The magnesium alloy LAE442 showed promising results as a bone substitute in numerous studies in non-weight bearing bone defects. This study aimed to investigate the in vivo behavior of wedge-shaped open-pored LAE442 scaffolds modified with two different coatings (magnesium fluoride (MgF2, group 1)) or magnesium fluoride/calcium phosphate (MgF2/CaP, group 2)) in a partial weight-bearing rabbit tibia defect model. The implantation of the scaffolds was performed as an open wedge corrective osteotomy in the tibia of 40 rabbits and followed for observation periods of 6, 12, 24, and 36 weeks. Radiological and microcomputed tomographic examinations were performed in vivo. X-ray microscopic, histological, histomorphometric, and SEM/EDS analyses were performed at the end of each time period. µCT measurements and X-ray microscopy showed a slight decrease in volume and density of the scaffolds of both coatings. Histologically, endosteal and periosteal callus formation with good bridging and stabilization of the osteotomy gap and ingrowth of bone into the scaffold was seen. The MgF2 coating favored better bridging of the osteotomy gap and more bone-scaffold contacts, especially at later examination time points. Overall, the scaffolds of both coatings met the requirement to withstand the loads after an open wedge corrective osteotomy of the proximal rabbit tibia. However, in addition to the inhomogeneous degradation behavior of individual scaffolds, an accumulation of gas appeared, so the scaffold material should be revised again regarding size dimension and composition.
In vivo corrosion and corrosion protection of magnesium alloy LAE442

Abstract:The aim of this study was to investigate whether the extruded magnesium alloy LAE442 reacts in vivo with an appropriate host response and to investigate how an additional magnesium fluoride (MgF(2)) coating influences the in vivo corrosion rate. Forty cylinders were machined from extruded LAE442 and 20 of these were coated additionally with MgF(2) and implanted into the medial femur condyle of adult rabbits. Synchrotron-radiation-based X-ray computed micro-tomography (SRmicroCT) was used to quantitatively analyse corrosion non-destructively in vivo and comparisons were made to magnesium degradation rates based on area measurements of the remaining metal on uncalcified sections. Blood concentrations of the alloying elements were measured below toxicological limits. The MgF(2) layer was no longer detected after 4 weeks of implantation by particle-induced gamma emission, and the MgF(2) coating reduced the blood content of alloying elements during the first 6 weeks of implantation with no elevated fluoride concentration in the adjacent bone. Histopathological examinations of liver showed in 9 out of 40 cases minimal infiltrations of heterophil granulocytes of unknown origin (5 LAE442, 4 LAE442+MgF(2)). The kidneys were mainly regular in structure. The synovial tissue showed a granular cell infiltration as a temporary observation in the LAE442+MgF(2) group after 2 weeks. No subcutaneous gas cavities were observed clinically and on postoperative X-rays in all animals. All specimens were scanned by SRmicroCT at 2, 4, 6 and 12 weeks postoperatively before uncalcified sections were performed. All magnesium implants have been observed in direct bone contact and without a fibrous capsule. Localized pitting corrosion occurred in coated and uncoated magnesium implants. This study shows that the extruded magnesium alloy LAE442 provides low corrosion rates and reacts in vivo with an acceptable host response. The in vivo corrosion rate can be further reduced by additional MgF(2) coating.
In vivo comparison of the degradation and osteointegration properties of micro-arc oxidation-coated Mg-Sr and Mg-Ca alloy scaffolds

Abstract:Background: Magnesium (Mg) alloy have biodegradation and mechanical properties that are similar to those of human bone, making it a promising candidate material for inclusion in implantable medical devices. Objective: The osteointegration effect of Mg alloy scaffolds with different corrosion rates were studied and evaluated in large bone defect models. Method: Mg-Sr and Mg-Ca alloy scaffolds with a 20-μm Micro-arc oxidation (MAO) coating were used to repair critical bone defects for subsequent assessment of each alloy's degradation and osteointegration by X-ray, Micro-CT, fluorescence and histological examination. Results: At 12 weeks post-implantation, each defect was found to be effectively reconstructed by either of the Mg alloys based on X-ray and Micro-CT images. The corrosion rate (CR) of each Mg alloy - as calculated based on micro-computed tomography information - demonstrated that the MAO coating could provide effective protection for only 4 weeks post-surgery. From weeks 8 to 12, the CR of the Mg-Ca alloy scaffold increased from 1.34 ± 0.23 mm/y to 1.57 ± 0.16 mm/y. In contrast, the CR of the Mg-Sr alloy scaffold decreased from 0.58 ± 0.14 mm/y to 0.54 ± 0.16 mm/y. However, fluorescence and histological examination revealed more mature, closely and regularly arranged newborn osteocytes at the Mg-Ca scaffold-fracture interface e from weeks 8 to 12 after surgery. Conclusion: The Mg-Sr scaffold was more corrosion resistant and the Mg-Ca scaffold yielded a better overall repair, which indicates that the CR of magnesium alloys matches the rate of new bone formation and is the key to repair bone defects as a bone substitute.

Abstract:The fabrication, corrosion, mechanical and cell behavior of biodegradable porous Mg-based alloy scaffolds were investigated and are described in this contribution. Using a combination of the casting and computer numerical control methods, a series of porous alloy scaffolds with spherical pore morphology and three different porosity values ranging from 30 to 50%, having pore sizes of about 800 μm were manufactured. Compression tests were used to gauge the porous scaffolds' mechanical characteristics, and contrast findings with those of cancellous bone tissue. Greater compressive strength (72 MPa) was found for porous alloy scaffolds with 30% porosity than for scaffolds with 50% porosity (21 MPa). The scaffolds with 50% and 30% porosity demonstrated the corrosion rate of 4.3 mm/year and 3.5 mm/year, respectively. Additionally, biocompatibility tests showed that high porosity scaffolds (50%) express higher cell viability (97%) and alkaline phosphatase activity. These findings indicate that open-porous Mg alloy scaffolds could be used as biodegradable implants for bone replacement applications.

Abstract:Bone fractures often require internal fixation using plates or screws. Normally, these devices are made of permanent metals like titanium providing necessary strength and biocompatibility. However, they can also cause long-term complications and may require removal. An interesting alternative are biocompatible degradable devices, which provide sufficient initial strength and then degrade gradually. Among other materials, biodegradable magnesium alloys have been developed for craniofacial and orthopaedic applications. Previously, we tested implants made of magnesium hydroxide and RS66, a strong and ductile ZK60-based alloy, with respect to biocompatibility and degradation behaviour. Here, we compare the effects of dissolving magnesium hydroxide and RS66 cylinders on bone regeneration and bone growth in rabbit condyles using microtomographical and histological analysis. Both magnesium hydroxide and RS66 induced a considerable osteoblastic activity leading to distinct but different spatio-temporal patterns of cancellous and periosteal bone growth. Dissolving RS66 implants induced a prominent periosteal bone formation on the medial surface of the original condyle whereas dissolving magnesium hydroxide implants enhance mainly cancellous bone formation. Especially periosteal bone formation was completed after 6 and 8 weeks, respectively. The observed bone promoting functions are in line with previous reports of magnesium stimulating cancellous and periosteal bone growth and possible underlying signalling mechanisms are discussed. Statement of Significance Biodegradable magnesium based implants are promising candidates for use in orthopedic and traumatic surgery. Although these implants are in the scientific focus for a long time, comparatively little is known about the interactions between degrading magnesium and the biological environment. In this work, we investigated the effects of two degrading cylindrical magnesium implants (MgOH 2 and RS66) both on bone regeneration and on bone growth. Both MgOH 2 and RS66 induce remarkable osteoblastic activities, however with different spatio-temporal patterns regarding cancellous and periosteal bone growth. We hypothesize that degradation products do not diffuse directionless away, but are transported by the restored blood flow in specific spatial patterns which is also dependent on the used surgical technique.

Abstract:Laser powder bed fusion (L-PBF) of magnesium (Mg) alloy porous scaffolds is expected to solve the dual challenges from customized structures and biodegradable functions required for repairing bone defects. However, one of the key technical difficulties lies in the poor L-PBF process performance of Mg, contributed by the high susceptibility to oxidation, vaporization, thermal expansion, and powder attachment etc. This work investigated the influence of L-PBF energy input and scanning strategy on the formation quality of porous scaffolds by using WE43 powder, and characterized the microstructure, mechanical properties, biocompatibility, biodegradation and osteogenic effect of the as-built WE43 porous scaffolds. With the customized energy input and scanning strategy, the relative density of struts reached over 99.5%, and the geometrical error between the designed and the fabricated porosity declined to below 10%. Massive secondary phases including intermetallic precipitates and oxides were observed. The compressive strength (4.37-23.49 MPa) and elastic modulus (154.40-873.02 MPa) were comparable to those of cancellous bone. Good biocompatibility was observed by in vitro cell viability and in vivo implantation. The biodegradation of as-built porous scaffolds promoted the osteogenic effect, but the structural integrity devastated after 12 h by the immersion tests in Hank's solution and after 4 weeks by the implantation in rabbits' femur, indicating an excessively rapid degradation rate.

Abstract:In this study, porous magnesium (Mg) scaffolds were investigated with varying strontium (Sr) and constant zinc (Zn) concentrations through the powder metallurgy process. All samples were examined at room temperature to evaluate their microstructure, mechanical andin-vitrodegradation behaviour and biological properties. Results indicated that adding Sr was associated with fine average grain size, increased mechanical strength, and a decreased corrosion rate. All samples show tiny isolated and open interconnected pores (porosities: 18%-30%, pores: 127-279 µm) with a suitable surface roughness of less than 0.5 µm. All the provided samples possess mechanical and hemocompatible properties that closely resemble natural bone. Mg-4Zn-2Sr has the highest hardness (102.61 ± 15.1 HV) and compressive strength (24.80 MPa) than Mg-4Zn-0.5Sr (85 ± 8.5 HV, 22.14 MPa) and Mg-4Zn-1Sr (97.71 ± 11.2 HV, 18.06 MPa). Immersion results revealed that samples in phosphate-buffered saline solutions have excellent degradability properties, which makes them a promising biodegradable material for orthopaedic applications. The scaffold with the highest Sr concentration shows the best optimised mechanical and degradation behaviour out of the three porous scaffolds, with a 2.7% hemolysis rate.
Multi-scale mechanical and morphological characterisation of sintered porous magnesium-based scaffolds for bone regeneration in critical-sized defects

Abstract:Magnesium (Mg) and its alloys are very promising degradable, osteoconductive and osteopromotive materials to be used as regenerative treatment for critical-sized bone defects. Under load-bearing conditions, Mg alloys must display sufficient morphological and mechanical resemblance to the native bone they are meant to replace to provide adequate support and enable initial bone bridging. In this study, unique highly open-porous Mg-based scaffolds were mechanically and morphologically characterised at different scales. In situ X-ray computed tomography (XCT) mechanics, digital volume correlation (DVC), electron microscopy and nanoindentation were combined to assess the influence of material properties on the apparent (macro) mechanics of the scaffold. The results showed that Mg exhibited a higher connected structure (38.4mm-3 and 6.2mm-3 for Mg and trabecular bone (Tb), respectively) and smaller spacing (245µm and 629µm for Mg and Tb, respectively) while keeping an overall appropriate porosity of 55% in the range of trabecular bone (30-80%). This fully connected and highly porous structure promoted lower local strain compared to the trabecular bone structure at material level (i.e. -22067 ± 8409µε and -40120 ± 18364µε at 6% compression for Mg and trabecular bone, respectively) and highly ductile mechanical behaviour at apparent level preventing premature scaffold failure. Furthermore, the Mg scaffolds exceeded the physiological strain of bone tissue generated in daily activities such as walking or running (500-2000µε) by one order of magnitude. The yield stress was also found to be close to trabecular bone (2.06MPa and 6.67MPa for Mg and Tb, respectively). Based on this evidence, the study highlights the overall biomechanical suitability of an innovative Mg-based scaffold design to be used as a treatment for bone critical-sized defects. STATEMENT OF SIGNIFICANCE: Bone regeneration remains a challenging field of research where different materials and solutions are investigated. Among the variety of treatments, biodegradable magnesium-based implants represent a very promising possibility. The novelty of this study is based on the characterisation of innovative magnesium-based implants whose structure and manufacturing have been optimised to enable the preservation of mechanical integrity and resemble bone microarchitecture. It is also based on a multi-scale approach by coupling high-resolution X-ray computed tomography (XCT), with in situ mechanics, digital volume correlation (DVC) as well as nano-indentation and electron-based microscopy imaging to define how degradable porous Mg-based implants fulfil morphological and mechanical requirements to be used as critical bone defects regeneration treatment.
The influence of storage and heat treatment on a magnesium-based implant material: an in vitro and in vivo study

Abstract:Background: Magnesium alloys are recommended as a potential material for osteosynthesis. It is known that storage-induced property modifications can occur in materials like aluminum. Thus the aim of this study was to analyze the influence of storage durations of up to 48 weeks on the biomechanical, structural, and degradation properties of the degradable magnesium alloy LAE442. Methods: Extruded implants (n = 104; Ø 2.5 mm × 25 mm) were investigated after storage periods of 0, 12, 24, and 48 weeks in three different sub-studies: (I) immediately after the respective storage duration and after an additional (II) 56 days of in vitro corrosion in simulated body fluid (SFB), and (III) 48 weeks in vivo corrosion in a rabbit model, respectively. In addition, the influence of a T5-heat treatment (206 °C for 15 h in an argon atmosphere) was tested (n = 26; 0 week of storage). Evaluation was performed by three-point bending, scanning electron microscopy, radiography, µ-computed tomography, evaluation of the mean grain size, and contrast analysis of precipitations (such as aluminum or lithium). Results: The heat treatment induced a significant reduction in initial stability, and enhanced the corrosion resistance. In vivo experiments showed a good biocompatibility for all implants. During the storage of up to 48 weeks, no significant changes occurred in the implant properties. Conclusions: LAE442 implants can be safely used after up to 48 weeks of storage.
Biocompatibility evaluation of peo-treated magnesium alloy implants placed in rabbit femur condyle notches and paravertebral muscles

Abstract:Magnesium alloys have been receiving much attention for use in biodegradable metal implants because of their excellent mechanical properties and biocompatibility. However, their rapid breakdown and low bioactivity can cause the implant to lose mechanical integrity before the bone is completely healed. Moreover, hydrogen gas released during degradation can significantly delay the tissue regeneration process. To solve the instability of magnesium alloys, Zn and Ca can be added to improve the mechanical properties and biocompatibility. One other way to improve the mechanical properties of Mg is plasma electrolytic oxidation (PEO), which provides a dense, thick ceramic-like coating on the Mg surface. In this study, high-purity Mg was selected as the control, and Mg-1wt%Zn-0.1wt%Ca alloy and PEO-treated Mg-1wt%Zn-0.1wt%Ca alloy were selected as the test materials; the results of radiographic and histological analyses of their biocompatibility are reported herein. Nineteen New Zealand white rabbits were used in the study. Rod-bars (Ø2.7 × 13.6 mm) were placed on both paravertebral muscles, and cannulated screws (Ø2.7x10mm) were placed on both femur condyle notches. Each animal was implanted in all four sites. X-rays were taken at 0, 2, 4, 8, and 12 weeks, micro-CT, and live-CT were taken at 4, 8, and 12 weeks. At weeks 4, 8, and 12, individuals representing each group were selected and sacrificed to prepare specimens for histopathological examination. The results confirm that in vivo, Mg-1wt%Zn-0.1wt%Ca alloy had higher corrosion resistance than high-purity Mg and safely degraded over time without causing possible side effects (foreign body or inflammatory reactions, etc.). In addition, PEO treatment of Mg-1wt%Zn-0.1wt%Ca alloy had a positive effect on fracture recovery by increasing the bonding area with bone. Our results suggest that PEO treatment of Mg-1wt%Zn-0.1wt%Ca alloy can be a promising biomaterials in the field of various clinical situations such as orthopedic and maxillofacial surgerys.

Abstract:Fracture treatment in children needs new implant materials to overcome disadvantages associated with removal surgery. Magnesium-based implants constitute a biocompatible and bioresorbable alternative. In adults and especially in children, implant safety needs to be evaluated. In children the bone turnover rate is higher and implant material might influence growth capacity, and the long-term effect of accumulated particles or ions is more critical due to the host's prolonged post-surgery lifespan. In this study we aimed to investigate the degradation behavior of ZX00 (Mg-0.45Zn-0.45Ca; in wt.%) in a small and a large animal model to find out whether there is a difference between the two models (i) in degradation rate and (ii) in bone formation and in-growth. Our results 6, 12 and 24 weeks after ZX00 implantation showed no negative effects on bone formation and in-growth, and no adverse effects such as fibrotic or sclerotic encapsulation. The degradation rate did not significantly differ between the two growing-animal models, and both showed slow and homogeneous degradation performance. Our conclusion is that small animal models may be sufficient to investigate degradation rates and provide preliminary evidence on bone formation and in-growth of implant materials in a growing-animal model. Statement of significance: The safety of implant material is of the utmost importance, especially in children, who have enhanced bone turnover, more growth capacity and longer postoperative lifespans. Magnesium (Mg)-based implants have long been of great interest in pediatric orthopedic and trauma surgery, due to their good biocompatibility, biodegradability and biomechanics. In the study documented in this manuscript we investigated Mg-Zn-Ca implant material without rare-earth elements, and compared its outcome in a small and a large growing-animal model. In both models we observed bone formation and in-growth which featured no adverse effects such as fibrotic or sclerotic encapsulation, and slow homogeneous degradation performance of the Mg-based implant material.

Abstract:In this work, Mg-1Zn-1Ca-0.5Mn with good biocompatibility is selected as the alloy component, and porous magnesium alloy scaffolds with different pore sizes and porosity are prepared by changing the particle size and prepressure of NaCl template in the process of seepage casting, so as to explore their effects on the properties of porous magnesium alloy scaffolds. The effect of passivation layer on the degradation rate and the vitro biological behavior of porous magnesium alloy with different pore size and porosity after surface passivation are studied. The experimental results show that the mechanical properties are inversely proportional to the porosity parameters. The degradation experiment shows that the corrosion rate of porous Mg-1Zn-1Ca-0.5Mn alloy with porosity of 74.34 % decreased to 0.94 mm/year. In vitro degradation experiment found that passivation layer had a significant protective effect on porous magnesium alloy, and the corrosion rate of porous Mg-1Zn-1Ca-0.5Mn alloy with a porosity of 74.34 % after surface passivation is soaked in Hanks' solution for 15 days is only 0.07 mm/year. The passivated porous magnesium alloy shows good cellular activity and biocompatibility, and the passivated porous Mg-1Zn-1Ca-0.5Mn alloy shows better osteogenic differentiation ability in the gene expression test. This study provides more options for the different structures of porous magnesium alloys and provides a feasible method for the surface fluorination coating of porous magnesium alloys. This work discussed the mechanism of the effect of pore structure difference on porous magnesium alloy, which is of great significance for the application of porous magnesium alloy in bone repair.

Abstract:Porous Magnesium (Mg) is a promising biodegradable scaffold for treating critical-size bone defects, and as an essential element for human metabolism, Mg has shown sufficient biocompatibility. Its elastic moduli and yield strengths are closer to those of cortical bone than common, inert metallic implants, effectively reducing stress concentrations around host tissue as well as stress shielding. More importantly, Mg can degrade and be absorbed in the human body in a safe and controlled manner, thereby reducing the need for second surgeries to remove implants. The development of porous Mg scaffolds via conventional selective laser melting techniques has been limited due to Mg's low boiling point, high vapor pressures, high reactivity, and non-ideal microstructures in additively manufactured parts. Here we present an exciting alternative to conventional additive techniques: 3D weaving with Mg wires that have controlled chemistries and microstructures. The weaving process offers high throughput manufacturing as well as porous architectures that can be optimized for stiffness and porosity with topology optimization. Once woven, we dip-coat the weaves with polylactic acid to enhance their strength and corrosion resistance. Following fabrication, we characterize their mechanical properties, corrosion behavior, and cell compatibilityin vitro, and we use an intramuscular implantation model to evaluate theirin vivocorrosion behavior and tissue response.

Abstract:Highlights • Ultra-lean, bioabsorbable Mg–Ca shows excellent biocompatibility in in-vivo studies. • Plasma electrolytic oxidation surface modification leads to improved osseointegration. • Mg–Ca implants exhibit full in-vivo bioabsorption of the degrading implant material. • WE43 implants retain rare-earth elements in their biodegradation products. • Enhanced biodegradation on bone-contacting side may result from osteoclastic activity. Magnesium alloys present a compelling prospect for absorbable implant materials in orthopedic and trauma surgery. This study evaluates an ultra-high purity, lean magnesium–calcium alloy (X0), both with and without plasma electrolytic oxidation (PEO) surface modification, in comparison to a clinically utilized WE43 magnesium alloy. It is shown that the mechanical properties of X0 can be tuned to yield a high-strength material suitable for bone screws (with an ultimate tensile strength of 336 MPa) or a ductile material appropriate for intraoperatively deformable plates (with an elongation at fracture of 24 %). Four plate-screw combinations were implanted onto the pelvic bones of six sheep without osteotomy for 8 weeks. Subsequent analysis utilized histology, micro-computed tomography, and light and electron microscopy. All implants exhibited signs of degradation and hydrogen-gas evolution, with PEO-coated X0 implants demonstrating the least volume loss and the most substantial new-bone formation on the implant surface and surrounding cancellous bone. Furthermore, the osteoconductive properties of the X0 implants, when uncoated, exceeded those of the uncoated WE43 implants, as evidenced by greater new-bone formation on the surface. This osteoconductivity was amplified with PEO surface modification, which mitigated gas evolution and enhanced osseointegration, encouraging bone apposition in the cancellous bone vicinity. These findings thus indicate that PEO-coated X0 implants hold substantial promise as a biocompatible and absorbable implant material. Abstract:Despite the availability of surgical and non‐surgical techniques, the repair of articular cartilage lesions remains a current clinical problem. Especially for the treatment of large osteochondral cartilage defects, the replacement of the subchondral bone plate is a crucial step for optimal cartilage repair. However, no artificial implant material can yet fully restore the properties of the subchondral bone plate. For optimal cartilage tissue repair, mechanical stability for the first two months is essential. Subsequently, a rapid degradation process of the implant material would allow optimal supply of nutrition to the regenerating tissue. To this end, we investigated whether the implantation of an open porous degradable scaffold made of a magnesium alloy (AZ91) could serve as a sufficient temporary replacement of the subchondral bone plate. The results show that this alloy degrades too rapid in vivo to allow sufficient cartilage repair above the scaffold. However, the surrounding cartilage tissue was not negatively affected by the rapid degradation process and new bone formation was observed at the rim of the degrading implant. In conclusion, magnesium scaffolds degrade in vivo but the initial high corrosion rate must be reduced to allow the formation of an appropriate cartilage tissue. Future research will therefore be directed to optimized alloys and additional coating with magnesium fluorides or calcium phosphates like hydroxyapatite. Knorpelregeneration auf subchondralen Knochenersatzimplantaten aus Magnesium Trotz verschiedener operativer und nicht‐operativer Behandlungsmethoden ist die Knorpelläsion immer noch ein ernsthaftes klinisches Problem. Besonders bei großen osteochondralen Defekten ist die Wiederherstellung der subchondralen Knochenplatte eine wichtige Voraussetzung für die Knorpelheilung. Bisher gibt es keine Ersatzmaterialien, die die Funktionen der subchondralen Knochenplatte vollständig ersetzen können. Für eine optimale Knorpelheilung wird eine mechanisch stabile Unterstützung für die ersten 2 Monate und eine anschließende schnelle Degradation des Knochenersatzmaterials gefordert. In dieser Studie haben wir den Einsatz von offenporigen Schwämmen aus der Magnesiumlegierung AZ91 als temporären Knochenersatz untersucht. Wir fanden, dass die Magnesiumschwämme aus AZ91 in vivo zu schnell degradierten, um eine Knorpelheilung zu ermöglichen. Das umgebende Knorpelgewebe war jedoch unbeeinflusst von der schnellen Korrosion und am Implantatrand fanden wir sogar Knochenneubildungen. Zusammenfassend fanden wir, dass Magnesiumschwämme in vivo degradieren, jedoch muss deren initiale Korrosionsrate deutlich gesenkt werden. In zukünftigen Studien wird dies mit Hilfe von weiter optimierten Magnesiumlegierungen und von zusätzlichen Beschichtungen aus Kalziumphosphaten wie z. B. Hyroxylapatit oder Magnesiumflouriden angestrebt.
