Abstract:WE43, a magnesium alloy containing yttrium and neodymium as main alloying elements, has become a well-established bioresorbable implant material. Implants made of WE43 are often fabricated by powder extrusion and subsequent machining, but for more complex geometries laser powder bed fusion (LPBF) appears to be a promising alternative. However, the extremely high cooling rates and subsequent heat treatment after solidification of the melt pool involved in this process induce a drastic change in microstructure, which governs mechanical properties and degradation behaviour in a way that is still unclear. In this study we investigated the changes in the microstructure of WE43 induced by LPBF in comparison to that of cast WE43. We did this mainly by electron microscopy imaging, and chemical mapping based on energy-dispersive X-ray spectroscopy in conjunction with electron diffraction for the identification of the various phases. We identified different types of microstructure: an equiaxed grain zone in the center of the laser-induced melt pool, and a lamellar zone and a partially melted zone at its border. The lamellar zone presents dendritic lamellae lying on the Mg basal plane and separated by aligned Nd-rich nanometric intermetallic phases. They appear as globular particles made of Mg3Nd and as platelets made of Mg41Nd5 occurring on Mg prismatic planes. Yttrium is found in solid solution and in oxide particles stemming from the powder particles' shell. Due to the heat influence on the lamellar zone during subsequent laser passes, a strong texture developed in the bulk material after substantial grain growth. STATEMENT OF SIGNIFICANCE: Additively manufactured magnesium alloys have the potential of providing a major breakthrough in bone-reconstruction surgery by serving as biodegradable porous scaffold material. This study is the first to report in detail on the microstructure development of the established magnesium alloy WE43 fabricated by the additive manufacturing process of Laser Powder Bed Fusion (LPBF). It presents unique microstructural features which originate from the laser-melting process. An in situ transmission electron microscopy heating experiment further demonstrates the development of two distinct intermetallic phases in additively manufactured WE43 alloys. While one forms already during solidification, the other precipitates due to the ongoing heat treatment during LPBF processing.
Design of magnesium alloys with controllable degradation for biomedical implants: From bulk to surface

Abstract:The combination of high strength, light weight, and natural biodegradability renders magnesium (Mg)-based alloys promising in orthopedic implants and cardiovascular stents. Being metallic materials, Mg and Mg alloys made for scaffolds provide the necessary mechanical support for tissue healing and cell growth in the early stage, while natural degradation and reabsorption by surrounding tissues in the later stage make an unnecessarily follow-up removal surgery. However, uncontrolled degradation may collapse the scaffolds resulting in premature implant failure, and there has been much research in controlling the degradation rates of Mg alloys. This paper reviews recent progress in the design of novel Mg alloys, surface modification and corrosion mechanisms under different conditions, and describes the effects of the structure, composition, and surface conditions on the degradation behavior in vitro and in vivo. Statement of significance: Owing to their unique mechanical properties, biodegradability, biocompatibility, Mg based biomaterials are becoming the most promising substitutes for tissue regeneration for impaired bone, vascular and other tissues because these scaffolds can provide not only ideal space for the growth and differentiation of seeded cells but also enough strength before the formation of normal tissues. The most important is that these scaffolds can be fully degraded after tissue regeneration, which can satisfy the increasing demand for better biomedical devices and functional tissue engineering biomaterials in the world. However, the rapid degradation rate of these scaffolds restricts the wide application in clinic. This paper reviews recent progress on how to control the degrdation rate based on the relevant corrosion mechanisms through the design of porous structure, phase structure, grains, and amorphous structure as well as surface modification, which will be beneficial to the better understanding and functional design of Mg-based scaffolds for wide clinical applications in tissue reconstruction in near futures.
Surface Engineering of Biodegradable Magnesium Alloys for Enhanced Orthopedic Implants.

Abstract:Magnesium (Mg) alloys have been promised for biomedical implants in orthopedic field, however, the fast corrosion rate and mode challenge their clinical application. To push Mg alloys materials into practice, a composite coating with biodegradable and high compatible components to improve anticorrosion property of an Mg alloy (i.e., AZ31) is designed and fabricated. The inner layer is micro-nano structured Mg(OH)2 through hydrothermal treatment. Then stearic acid (SA) is introduced to modify Mg(OH)2 for better reducing the gap below a surface-degradation polymer layer of poly(1,3-trimethylene carbonate). Benefited by the SA modification effect, this sandwiched coating avoids corrosive medium penetration via enhancing the adhesion strength at the interface between outer and inner layers. Both in vitro and in vivo tests indicate that the composite coating modified AZ31 perform a better anticorrosion behavior and biocompatibility compared to bare AZ31. Strikingly, a 1.7-fold improvement in volume of newly formed bone is observed surrounding the composite coating modified implant after 12 week implantation. The sandwiched biocompatible coating strategy paves a hopeful way for future translational application of Mg alloys orthopedic materials in clinics.

Abstract:Additive Manufacturing (AM) exhibits tremendous advantages in producing customized medical implants and has achieved successful clinical applications for non-degradable metals such as titanium, stainless steel and cobalt alloys, particularly in the form of porous scaffolds for bone implants. Biodegradable materials allow the metal to degrade while natural tissue grows thereby reducing implant lifetime within the body to a minimum. The design freedom of AM and especially Laser Powder Bed Fusion (LPBF) overcomes technical difficulties of conventional technologies to manufacture complex structures. Within this work test geometries and porous scaffolds were manufactured via LPBF with pre-alloyed Zinc-Magnesium (Zn-xMg) powder, where x = 0, 1, 2 and 5 wt% to investigate the effect of Mg on manufacturability, microstructure and mechanical properties. Relative material densities of cubic specimens above 99.5% were achieved with all pre-alloyed powders. The addition of Mg reduces the processing window compared to pure Zn. A decreased grain size and an increase in intermetallic precipitates is observed with the addition of Mg. Zn-1Mg samples exhibited 3.2x the maximum ultimate tensile strength (UTS) of pure Zn (i.e., 119 MPa). However, the tensile elongation is only a fifth compared to pure Zn. With increasing Mg content, the amount of Zn+Mg2Zn11 rises and MgZn2 forms. These brittle phases characteristically increase hardness but reduce ductility. Under the chosen processing conditions, all Zn-xMg scaffolds demonstrated good processability. The geometric porosity was designed to be 80.5% with the achieved value of 70.8 % for compression testing. Zn-1Mg scaffold showed the highest compressive strength and Young’s modulus (19.1 MPa and 0.65 GPa respectively). This paper summarizes the technical challenges of manufacturing Zn-xMg alloys via LPBF for tissue engineering applications.

Abstract:Magnesium (Mg) alloys have gained recognition as revolutionary biomaterials, owing to their inherent degradability, favorable biocompatibility and mechanical properties. Additive manufacturing (AM) provides high design flexibility and enables the creation of implants with personalized complex shapes and internal porous structures tailored to individual anatomical and functional needs. Particularly, laser powder bed fusion (LPBF), one prevalent AM technique, utilizes a fine laser beam as heat source and results in tiny molten pool with extremely fast cooling rate, which effectively restricts grain growth, inter-metallic precipitation and macroscopic segregation, thus facilitating the fabrication of high-performance metal parts. This review critically assesses the significance of biodegradable Mg alloys and investigates the feasibility of utilizing LPBF for Mg alloys applications in biomedical field. Detailed discussions on LPBF-processed biomedical Mg alloys parts cover process parameters, microstructure, metallurgical defects, and properties like mechanical performance, corrosion behavior, and biological response in both as-built and post-processed states. Additionally, suggestions for advancing knowledge in LPBF of biodegradable Mg alloys for biomedical applications are highlighted to propel further research and development in this field.

Abstract:Magnesium-based alloy WE43 is a state-of-the-art bioresorbable metallic implant material. There is a need for implants with both complex geometries to match the mechanical properties of bone and refined microstructure for controlled resorption. Additive manufacturing (AM) using laser powder bed fusion (LPBF) presents a viable fabrication method for implant applications, as it offers near-net-shape geometrical control, allows for geometry customization based on an individual patient, and fast cooling rates to achieve a refined microstructure. In this study, the laser–alloy interaction is investigated over a range of LPBF-relevant processing conditions to reveal melt-pool dynamics, pore formation, and the microstructure of laser-melted WE43. In situ X-ray imaging reveals distinct laser-induced vapor depression morphology regimes, with minimal pore formation at laser-scan speeds greater than 500 mm/s. Optical and electron microscopy of cross-sectioned laser tracks reveal three distinct microstructural regimes that can be controlled by adjusting laser-scan parameters: columnar, dendritic, and banded microstructures. These regimes are consistent with those predicted by the analytic solidification theory for conduction-mode welding, but not for keyhole-mode tracks. The results provide insight into the fundamental laser–material interactions of the WE43 alloy under AM-processing conditions and are critical for the successful implementation of LPBF-produced WE43 parts in biomedical applications.

Abstract:Porous Magnesium (Mg) is a promising biodegradable scaffold for treating critical-size bone defects, and as an essential element for human metabolism, Mg has shown sufficient biocompatibility. Its elastic moduli and yield strengths are closer to those of cortical bone than common, inert metallic implants, effectively reducing stress concentrations around host tissue as well as stress shielding. More importantly, Mg can degrade and be absorbed in the human body in a safe and controlled manner, thereby reducing the need for second surgeries to remove implants. The development of porous Mg scaffolds via conventional selective laser melting techniques has been limited due to Mg's low boiling point, high vapor pressures, high reactivity, and non-ideal microstructures in additively manufactured parts. Here we present an exciting alternative to conventional additive techniques: 3D weaving with Mg wires that have controlled chemistries and microstructures. The weaving process offers high throughput manufacturing as well as porous architectures that can be optimized for stiffness and porosity with topology optimization. Once woven, we dip-coat the weaves with polylactic acid to enhance their strength and corrosion resistance. Following fabrication, we characterize their mechanical properties, corrosion behavior, and cell compatibilityin vitro, and we use an intramuscular implantation model to evaluate theirin vivocorrosion behavior and tissue response.

Abstract:The development of biodegradable implants is certainly intriguing, and magnesium and its alloys are considered significant among the various biodegradable materials. Nevertheless, the fast degradation, the generation of a significant amount of hydrogen gas, and the escalation in the pH value of the body solution are significant barriers to their use as an implant material. The appropriate approach is able to solve this issue, resulting in a decrease the rate of Mg degradation, which can be accomplished by alloying, surface adjustment, and mechanical treatment. Surface modification is a practical option because it not only improves corrosion resistance but also prepares a treated surface to improve bone regeneration and cell attachment. Metal coatings, ceramic coatings, and permanent polymers were shown to minimize degradation rates, but inflammation and foreign body responses were also suggested. In contrast to permanent materials, the bioabsorbable polymers normally show the desired biocompatibility. In order to improve the performance of drugs, they are generally encapsulated in biodegradable polymers. This study summarized the most recent advancements in manufacturing polymeric coatings on Mg alloys. The related corrosion resistance enhancement strategies and future potentials are discussed. Ultimately, the major challenges and difficulties are presented with aim of the development of polymer-coated Mg-based implant materials.

Abstract:Laser powder bed fusion (L-PBF) of magnesium (Mg) alloy porous scaffolds is expected to solve the dual challenges from customized structures and biodegradable functions required for repairing bone defects. However, one of the key technical difficulties lies in the poor L-PBF process performance of Mg, contributed by the high susceptibility to oxidation, vaporization, thermal expansion, and powder attachment etc. This work investigated the influence of L-PBF energy input and scanning strategy on the formation quality of porous scaffolds by using WE43 powder, and characterized the microstructure, mechanical properties, biocompatibility, biodegradation and osteogenic effect of the as-built WE43 porous scaffolds. With the customized energy input and scanning strategy, the relative density of struts reached over 99.5%, and the geometrical error between the designed and the fabricated porosity declined to below 10%. Massive secondary phases including intermetallic precipitates and oxides were observed. The compressive strength (4.37-23.49 MPa) and elastic modulus (154.40-873.02 MPa) were comparable to those of cancellous bone. Good biocompatibility was observed by <i>in vitro</i> cell viability and <i>in vivo</i> implantation. The biodegradation of as-built porous scaffolds promoted the osteogenic effect, but the structural integrity devastated after 12 h by the immersion tests in Hank's solution and after 4 weeks by the implantation in rabbits' femur, indicating an excessively rapid degradation rate.

Abstract:Magnesium alloys offer excellent properties with regard to application as degradable implant. For bone implants, it is often desirable to use porous materials. However, the preparation of high‐porosity magnesium implants has been difficult so far. The present study uses melt extracted magnesium fibers as the starting material for the sintering of highly porous magnesium bodies, i.e., from alloys MgY4 (W4) and MgY2Zn1CaMn (WZ21). Single short fibers of these alloys with an equivalent diameter between 100 and 250 µm and a length of 4–8 mm are manufactured by melt extraction. Thermodynamic calculations are used to determine the best conditions for liquid phase sintering of these Mg alloys. As no organic or other substances are needed in the process, it is possible to obtain high‐purity, high‐porosity (up to 75%) bodies with exclusively open porosity. Metallographic studies as well as mechanical and corrosion testing experiments complete this work.

Abstract:Biodegradable magnesium (Mg) alloys exhibit excellent biocompatibility, adequate mechanical properties, and osteogenic effect. They can contribute to complete recovery of damaged tissues without concerns about a second surgery and have achieved clinical applications in orthopedic and cardiovascular fields. Porous scaffolds can provide functions such as bone integration and adjustable mechanical properties, thus widely used for bone repair. Additive manufacturing (AM) offers the advantages of design freedom and high precision, enabling the reliable production of porous scaffolds with customized structures. The combination of biodegradable Mg alloys, porous scaffolds, and AM processes has created tremendous opportunities for the precision treatment of bone defects. This article reviews the current development in the additive manufacturing process and design of Mg alloy biodegradable orthopedic implants, focusing on chemical compositions, structural design, surface treatment, and their effects on mechanical properties, degradation behavior, and biocompatibility. Abstract:Developing Magnesium (Mg) based degradable implants for orthopedic applications is an attractive research area for the past two decades in the biomedical engineering. Mg is well accepted by human system and does not cause any health abnormalities during its degradation in the physiological environment. However, in order to improve its life span by controlling the aggressive degradation, novel Mg alloys are developed and subjected to different treatments to enhance their performance to tailor as promising candidates for implant manufacturing. In this context, recently, a special attention is paid towards using rare earth containing Mg alloys due to their excellent mechanical and corrosion resistance properties. Hence, in the present work, WE43 Mg alloy has been selected and the microstructual modification was carried out by friction stir processing. The role of grain refinement on the degradation behavior of FSPed WE43 Mg alloy was assessed by immersing the samples in simulated body fluids. From the microstructural studies, grain size reduction from 46 ± 4.2 μm to 16.1 ± 5.4 μm was achieved after FSP. The larger intermetallic particles were also observed as dissolved into the solid solution grains and fewer intermetallic particles were remained in the stir zone of FSPed alloy. After immersion studies, the surface of the samples was deposited with mineral phases and were analyzed by X-ray diffraction analysis and scanning electron microscope and found that the grain refinement achieved by FSP has a significant effect on increasing the mineral depositions which helps to control the degradation rate of the samples.

Abstract:The advent of additively manufactured biodegradable porous metals presents a transformative opportunity to meet the criteria of ideal bone substitutes. Precisely tailoring their degradation behavior constitutes a pivotal aspect of this endeavor. In this study, we investigated the effects of topological designs on the degradation profile of laser powder bed fusion (LPBF) Zn scaffolds under dynamic in vitro immersion tests. Specifically, four types of Zn-0.4Mn-0.2Mg scaffolds (beam-based: diamond, face center cubic; surface-based: gyroid, schwarz-P) were designed and fabricated. The degradation mechanism of the scaffolds was comprehensively evaluated using both experimental and simulation methods. The results illuminate the profound impact of structural design on the degradation properties of the Zn alloy scaffolds. The beam-based diamond and face center cubic scaffolds exhibited a degradation rate of 0.08–0.12 mm per year with a relatively uniform degradation mode under dynamic immersion. On the contrary, the surface-based gyroid and Schwarz-P scaffolds demonstrated a notably reduced degradation rate due to lower permeability. This restricted the diffusion of medium ions within the pores, culminating in the accumulation of degradation products and more severe localized degradation. This study underscores the potential of topological design as a compelling strategy for tailoring the degradation profile of additively manufactured biodegradable scaffolds, thereby advancing their suitability as bone substitutes.

Abstract:Biodegradable magnesium-calcium (Mg–Ca) implants have the ability to gradually dissolve and absorb into the human body after implantation. The similar mechanical properties to bone indicate that Mg–Ca is an ideal implant material to minimize the negative effects of stress shielding. Furthermore, using a biodegradable Mg–Ca implant prevents the need for a secondary removal surgery that commonly occurs with permanent metallic implants. The critical issue that hinders the application of Mg–Ca implants is the poor corrosion resistance to human body fluids. The corrosion process adversely affects bone ingrowth that is critical for recovery. Therefore, sequential laser shock peening (LSP) of a biodegradable Mg–Ca alloy was initiated to create a superior surface topography for improving implant performance. LSP is an innovative treatment to fabricate functional patterns on the surface of an implant. A patterned surface promotes bone ingrowth by providing a rough surface texture. Also, LSP imparts deep compressive residual stresses below the surface, which could potentially slow corrosion rates. Unique surface topographies were fabricated by changing the laser power and peening overlap ratio. The resultant effects on surface topography were investigated. Sequential peening at higher overlap ratios (75%) was found to reduce the tensile pileup region by over 40% as well as compress the overall surface by as much as 35 μm.

Abstract:In recent years, research on magnesium (Mg) alloys had increased significantly for hard tissue replacement and stent application due to their outstanding advantages. Firstly, Mg alloys have mechanical properties similar to bone which avoid stress shielding. Secondly, they are biocompatible essential to the human metabolism as a factor for many enzymes. In addition, main degradation product Mg is an essential trace element for human enzymes. The most important reason is they are perfectly biodegradable in the body fluid. However, extremely high degradation rate, resulting in too rapid loss of mechanical strength in chloride containing environments limits their applications. Engineered artificial biomaterials with appropriate mechanical properties, surface chemistry, and surface topography are in a great demand. As the interaction between the cells and tissues with biomaterials at the tissue–implant interface is a surface phenomenon; surface properties play a major role in determining both the biological response to implants and the material response to the physiological condition. Therefore, the ability to modify the surface properties while preserve the bulk properties is important, and surface modification to form a hard, biocompatible and corrosion resistant modified layer have always been an interesting topic in biomaterials field. In this article, attempts are made to give an overview of the current research and development status of surface modification technologies of Mg alloys for biomedical materials research. Further, the advantages/disadvantages of the different methods and with regard to the most promising method for Mg alloys are discussed. Finally, the scientific challenges are proposed based on own research and the work of other scientists.
Microstructure evolution and ductility improvement of additively manufactured biodegradable zinc–magnesium alloys via annealing

Abstract:Zinc–magnesium (Zn–Mg) alloys, fabricated by laser powder bed fusion (LPBF) additive manufacturing techniques, have emerged as promising candidates for biomedical implants due to their biodegradation capability, superior mechanical strength, and excellent biocompatibility. However, LPBF-fabricated Zn–Mg alloys still face challenges related to extremely low ductility and limited exploration of degradation characteristics. In this study, the impact of Mg incorporation on the printability, degradation properties, microstructure, and mechanical properties of LPBF-fabricated Zn–Mg alloys was primarily investigated. Furthermore, we proposed a viable annealing post-processing route for the first time to tailor the microstructural characteristics of the fabricated Zn–Mg alloy and enhanced its limited ductility. The results demonstrated that by applying a laser power of 80 W and a scanning speed of 600 mm/s, the relative density of LPBF-fabricated Zn–Mg alloy reached 98.62%. Increasing the Mg amount from 1 to 5 wt% refined the grain size while promoting an increase in Mg2Zn11 and MgZn2 phases. Among these compositions, the Zn–1Mg alloy exhibited the greatest degradation rate at 0.126 mm/year. The annealing treatment facilitated the microstructure evolution of the Zn–1Mg alloy, resulting in equiaxed grains, increased average grain size, high-angle grain boundaries, and enrichment of Mg at grain boundaries. After annealing at 300°C for 0.5 h, the tensile strength of Zn– 1Mg alloy decreased from 254.92 to 170.93 MPa, while the elongation significantly increased by a factor of 14.3 from 0.55% to 8.43%. These findings provide valuable insights into an effective post-processing approach for tailoring the microstructure and resultant mechanical properties of LPBF-fabricated Zn and its alloys.

Surface characterization and cytotoxicity response of biodegradable magnesium alloys

Abstract:Magnesium alloys have raised an immense amount of interest to many researchers because of their evolution as a new kind of third generation materials. Due to their biocompatibility, density, and mechanical properties, magnesium alloys are frequently reported as prospective biodegradable implant materials. Moreover, magnesium alloys experience a natural phenomenon to biodegrade in aqueous solutions due to its corrosion activity, which is excellent for orthopedic and cardiovascular applications. However, a major concern with such alloys is fast and non-uniform corrosion degradation. Controlling the degradation rate in the physiological environment determines the success of biodegradable implants. In this investigation, three different grades of magnesium alloys: AZ31B, AZ91E and ZK60A were studied for their corrosion resistance and biocompatibility. Scanning electron microscopy, energy dispersive spectroscopy, atomic force microscopy and contact angle meter are used to study surface morphology, chemistry, roughness and wettability, respectively. Additionally, the cytotoxicity of the leached metal ions was evaluated by using a tetrazolium based bio-assay, MTS.

Abstract:Synthetic grafting needs improvements to eliminate secondary surgeries for the removal of implants after healing of the defected tissues. Tissue scaffolds are engineered to serve as temporary templates, which support the affected tissue and gradually degrade through the healing period. Beside mechanical function to withstand the anatomic loading conditions, scaffolds should also provide a decent biological function for the diffusion of nutrients and oxygen to the cells, and excretion of the wastes from the cells to promote the new tissue growth and vascularization. Moreover, the degradation byproducts of the scaffolds should be safe to the human body. Development of such multifunctional scaffolds requires selection of the right material, design, and manufacturing method. Mg has been recognized as the prominent biodegradable metal with regards to its mechanical properties matching to that of human bone, degradability in the body fluid, and its ability to stimulate new tissue growth. Scaffolds with intricate porous structures can be designed according to the patient-specific anatomic data using computer aided designs. Additive manufacturing (AM) is the right method to materialize these models rapidly with reasonably acceptable range of dimensional accuracy. Thus, the recent research trend is to develop ideal scaffolds using biodegradable Mg through AM methods. This review compiles and discusses the available literature on the AM of biodegradable Mg parts from the viewpoints of material compositions, process conditions, formation quality, dimensional accuracy, microstructure, biodegradation, and mechanical properties. The current achievements are summarized together, and future research directions are identified to promote clinical applications of biodegradable Mg through the advancement of AM.

Magnesium (Mg)-based materials are a new generation of alloys with the exclusive ability to be biodegradable within the human/animal body. In addition to biodegradability, their inherent biocompatibility and similar-to-bone density make Mg-based alloys good candidates for fabricating surgical bioimplants for use in orthopedic and traumatology treatments. To this end, nowadays additive manufacturing (AM) along with three-dimensional (3D) printing represents a promising manufacturing technique as it allows for the integration of bioimplant design and manufacturing processes specific to given applications. Meanwhile, this technique also faces many new challenges associated with the properties of Mg-based alloys, including high chemical reactivity, potential for combustion, and low vaporization temperature. In this review article, various AM processes to fabricate biomedical implants from Mg-based alloys, along with their metallic microstructure, mechanical properties, biodegradability, biocompatibility, and antibacterial properties, as well as various post-AM treatments were critically reviewed. Also, the challenges and issues involved in AM processes from the perspectives of bioimplant design, properties, and applications were identified; the possibilities and potential scope of the Mg-based scaffolds/implants are discussed and highlighted.
