Abstract:Despite a substantial body of research to date for the detection of microplastics (MPs) in almost every environmental compartment there remains a lack of standardisation, and methodologies used by different research groups vary widely. Chemical imaging, which provides simultaneous measurement of physical (i.e. spatial) and chemical (i.e. spectroscopic) information, is recognized as a promising tool for MPs analysis. Herein, we first review the state-of-the-art chemical imaging methods, i.e., Fourier-transform infrared spectroscopy (FTIR) and Raman imaging, for the identification and quantification of MPs from different environmental samples. From a technical point of view (e.g., accuracy, speed optimizations and background effects), the limitations and analytical challenges are highlighted from extensive literature data. Finally, we suggest possible strategies and solutions to improve current practices towards an automated routine for MPs analysis.

Abstract:Microplastics are globally recognized as contaminants in freshwater and marine aquatic systems. To date there is no universally accepted protocol for isolation and quantification of microplastics from aqueous media. Various methodologies exist, many of which are time consuming and have the potential to introduce contaminants into samples, thereby obscuring characterization of the environmental microplastic load. Here, we present first steps in the detection of microplastics in liquid samples, based on their fluorescent staining followed by high throughput analysis and quantification using Flow Cytometry. Using controlled laboratory settings nine polymer types [polystyrene (PS); polyethylene (PE); polyethylene terephthalate (PET/PETE); high density polyethylene (HDPE); low density polyethylene (LDPE); polyvinyl chloride (PVC); polypropylene (PP); nylon (PA); polycarbonate (PC)] were tested for identification and quantification in freshwater. All nine plastic types were stained with 10 μg/mL Nile Red in 10% dimethyl sulfoxide with a 10 min incubation time. The lowest spatial detectable limit for plastic particles was 200 nm. Out of the nine polymer types chosen for the study PS, PE, PET, and PC were well-identified; however, results for other plastic types (PVC, PP, PA, LDPE, and HDPE) were masked to certain extent by Nile Red aggregation and precipitation. The methodology presented here permits identification of a range of particle sizes and types. It represents a significant step in the quantification of microplastics by replacing visual data interpretation with a sensitive and automated method.

Micro-flow imaging for in-situ and real-time enumeration and identification of microplastics in water

Abstract:Microplastics (MPs) are emerging contaminants that have recently gained global attention. Current identification and quantification methods are known to be time-consuming, labor-intensive, and lack consensus on protocol standardization. This study explored the potential of micro-flow imaging (MFI) technology for rapid and in-situ identification and enumeration of MPs in water using two (2) MFI-based particle counters. Advantages, limitations, and recommendations for using MFI for MPs analysis were discussed. MPs with diverse physical (i.e., microbeads, fragments, fibers, and films) and surface (i.e., reflectivity, microporosity, color) characteristics were analyzed to understand the detection capabilities and limitations of MFI technology. Results demonstrated that MFI effectively automates most manually obtained particle features, such as size, color, object intensity and shape descriptors. It imparts consistency and reduces the subjective nature of results, thus enabling reliable comparison of the generated data. The particles can be further categorized based on their circularity and aspect ratio providing further insight into the shape and potential erosion of MPs in the environment. Transparent particles, often missed with other techniques such as microscopy, were detected by the MFI technology. The ability to assign particle IDs to MPs was an important advantage of the MFI technology that enabled the further investigation of selected MPs of interest. The limitations of the MFI technology were apparent in samples with high particle concentrations, with reflective MPs, and in the presence of bubbles. The color of the background against which the image was captured also influenced the detection accuracy. Procedural modifications during sample analysis and improvements in image analysis can assist in overcoming these challenges. MFI requires minimal sample preparation and gives real-time imaging data, making it a prime candidate for field monitoring in surface water systems in addition to laboratory analysis. With the potential application of machine learning and similar developments in the future, MFI-based particle counters are well-positioned to meet an important need in in-flow and real-time identification and enumeration of MPs.
A microfluidic chip enables fast analysis of water microplastics by optical spectroscopy

Abstract:Microplastics contaminating drinking water is a growing issue that has been the focus of a few recent studies, where a major bottleneck is the time-consuming analysis. In this work, a micro-optofluidic platform is proposed for fast quantification of microplastic particles, the identification of their chemical nature and size, especially in the 1-100 µm size range. Micro-reservoirs ahead of micro-filters are designed to accumulate all trapped solid particles in an ultra-compact area, which enables fast imaging and optical spectroscopy to determine the plastic nature and type. Furthermore, passive size sorting is implemented for splitting the particles according to their size range in different reservoirs. Besides, flow cytometry is used as a reference method for retrieving the size distribution of samples, where chemical nature information is lost. The proof of concept of the micro-optofluidic platform is validated using model samples where standard plastic particles of different size and chemical nature are mixed.

Abstract:Microplastics (MPs) have recently emerged as a new major and ubiquitous environmental pollutant with still undefined, yet potentially high risks for human health and ecosystems. This has prompted growing public concern along with an increasing number of scientific studies. In particular, recent research has highlighted the need for a standardized methodology to monitor microplastics in different media, particularly in water. This study reports on the use of a new particle counter for the quantitative detection of MPs in water samples based on real-time analysis of fluorescence emissions. The instrument was calibrated using two types of plastic particles, i.e., polyvinyl chloride and high-density polyethylene, selected as examples of high- and low-density plastics, respectively. Specific solvents were used to match the different plastic densities. Measurements were also carried out on particles obtained from wastewater samples collected at the inlet and outlet of specific units of a municipal wastewater treatment plant after sieving, filtering, digestion with hydrogen peroxide to remove degradable organic matter and resuspension in the solvents employed during the calibration step. A Fourier transform infrared spectroscopy analysis performed on the same wastewater samples confirmed the presence of MPs, and, in particular, of polyethylene, in some of the samples in which the highest concentrations were measured applying the proposed method. Therefore, the novel particle counter described in this paper could represent a promising method to quantitatively measure MP concentrations in water samples.

Abstract:With the increasing use and release of plastic products, microplastics have rapidly accumulated in ecological environments. When microplastics enter the food chain, they cause serious harm to organisms and humans. Microplastics pollution has become a growing concern worldwide; however, there is still no standardized method for rapidly and accurately detecting microplastics. In this work, we used fluorescence lifetime imaging technology to detect four kinds of Nile red-stained and unstained microplastics, and the unique phasor fingerprints of different microplastics were obtained by phasor analysis. Tracing the corresponding pixels of the “fingerprint” in the fluorescence lifetime image allowed for the quick and intuitive identification of different microplastics and their location distributions in a mixed sample. In our work, compared with staining the four microplastics with a fluorescent dye, using the phasor “fingerprint library” formed by the autofluorescence lifetimes of the microplastics was more easily distinguished than microplastics in the mixed samples. The feasibility of this method was further tested by adding three single substances—SiO2, chitin and decabromodiphenyl ethane (DBDPE), and surface sediments to simulate interferent in the environment, and the results providing potential applications for the identification and analysis of microplastics in complex environments.

Abstract:In recent years, the utilization of flow cytometry for quantitative microplastic analysis has gained prominence. However, the current methods have some drawbacks that need to be improved. The present study aims to enhance the flow cytometry detection protocols for Nile red (NR) stained microplastics, facilitating distinct microplastic and nanoplastic enumeration. By elevating dimethyl sulfoxide (DMSO) concentration to 20%–30% within the solution, NR solubility improved and agglomeration reduced. The analysis of 26 replicates of polystyrene (PS) liquid samples through four distinct dot plots highlighted the superior accuracy of dot plots integrating yellow fluorescence. Through systematic staining of varying NR concentrations across three microplastic liquid samples (polyethylene terephthalate, polyethylene, and polypropylene), the optimal staining concentration was determined to be 15–20 μg/mL. The distributions of agglomerated NR and NR stained PS under two scenarios—dissolved NR and partially agglomerated NR—were compared. Results showed their distinct distributions within the side scatter versus yellow fluorescence dot plot. Counting results from gradient-diluted PS liquid samples revealed a microplastic detection lower limit of 10 4 particles/mL, with an optimal concentration range of 10 5 –10 6 particles/mL. Flow cytometric assessment of PS microspheres spanning 150 nm to 40 μm indicated a 150 nm particle size detection minimum. Our investigation validated the efficacy of NR staining and subsequent flow cytometry analysis across eleven types of microplastics. Separation and concentration of microplastics (1.0–50.0 μm) and nanoplastics (0.2–1.0 μm) were achieved via sequential sieving through 50, 1.0, and 0.2 μm filter membranes. We used a combination of multiple filtration steps and flow cytometry to analyze microplastics and nanoplastics in nine simulated water samples. Our results showed that the combined amount of microplastics (1.0–50.0 μm) and nanoplastics (0.2–1.0 μm) after filtration had a ratio of 0.80–1.19 compared to the total microplastic concentration before filtration. This result confirms the practicality of our approach. By enhancing flow cytometry-based microplastic and nanoplastic detection protocols, our study provides pivotal technical support for research concerning quantitative toxicity assessment of microplastic and nanoplastic pollution.

Abstract:Plastics are vital for society, but their usage has grown exponentially and contributes to the growth of pollution worldwide. The World Health Organization, WHO, already reported that microplastics (MPs) are found everywhere, in waste and fresh water, and in the air and soil. Regarding water effluents, waste-water treatment plants only minimize the problem, trapping only larger size particles. In contrast, smaller ones remain in oxidation ponds or sewage sludges, or are even released to aquifers environment. Classic procedures for MPs detection are still quite laborious, and are usually conducted off-line, involving several steps and expensive equipment. Electrical Impedance Spectroscopy, EIS, is a technique that allows the analysis of a system’s electrical response, yielding helpful information about its domain-dependent on physical-chemical properties. Due to the superficial electronegativity of MPs’ particles, EIS may allow to attain the purpose of the present work: to provide a fast and reliable method to detect/estimate MPs’ concentration in water effluents. Among the most common microplastics are Polyethylene, PE, and Polyvinyl Chloride, PVC. Using the developed setup and experimental data collection methodology, the authors could differentiate between MPs’ suspensions containing the same concentration of the different evaluated MPs, PVC and PE, and assess PVC concentration variation, in the interval between 0.03 to 0.5 g (w/w), with an error, estimated based on the obtained impedance modulus, around or below 3% for the entire stimulus signal frequency range (from 100 Hz to 40 MHz) for the PVC particles.

Abstract:Microplastic (MP) pollution has emerged as a global environmental concern due to its ubiquity and harmful impacts on ecosystems and human health. MP assessment has therefore become increasingly necessary and common in environmental and experimental samples. Microscopy and spectroscopy are widely employed for the physical and chemical characterization of MPs. However, these analytical methods often require time-consuming pretreatments of samples or expensive instrumentation. In this work, we develop a portable and cost-effective polarization holographic imaging system that prominently incorporates deep learning techniques, enabling efficient, high-throughput detection and dynamic analysis of MPs in aqueous environments. The integration enhances the identification and classification of MPs, eliminating the need for extensive sample preparation. The system simultaneously captures holographic interference patterns and polarization states, allowing for multimodal information acquisition to facilitate rapid MP detection. The characteristics of light waves are registered, and birefringence features are leveraged to classify the material composition and structures of MPs. Furthermore, the system automates real-time counting and morphological measurements of various materials, including MP sheets and additional natural substances. This innovative approach significantly improves the dynamic monitoring of MPs and provides valuable information for their effective filtration and management.

Abstract:Biofilms serve essential ecosystem functions and are used in different technical applications. Studies from stream ecology and waste-water treatment have shown that biofilm functionality depends to a great extent on community structure. Here we present a fast and easy-to-use method for individual cell-based analysis of stream biofilms, based on stain-free flow cytometry and visualization of the high-dimensional data by viSNE. The method allows the combined assessment of community structure, decay of phototrophic organisms and presence of abiotic particles. In laboratory experiments, it allows quantification of cellular decay and detection of survival of larger cells after temperature stress, while in the field it enables detection of community structure changes that correlate with known environmental drivers (flow conditions, dissolved organic carbon, calcium) and detection of microplastic contamination. The method can potentially be applied to other biofilm types, for example, for inferring community structure for environmental and industrial research and monitoring.
A strategy for quantifying microplastic particles in membrane filtration processes using flow cytometry

Abstract:Microplastic (MP) pollution is ubiquitous in the aquatic environment, with significant quantities of MPs originating from municipal wastewater treatment plants. Efforts to evaluate and implement MP removal processes are underway, with membrane technologies often recommended as an "ideal" solution. A key challenge in evaluating these technologies involves efficiently quantifying MP concentrations in samples. Here, flow cytometry (FC) is demonstrated as an effective technique to obtain concentration measurements of plastic microbeads (MBs; 1-5 μm) suspended in water with/without added humic acid. Regardless of solution conditions, MB concentrations were easily quantified via FC. Subsequently, two microfiltration membranes were challenged to these suspensions. As measured via FC, the 0.45 μm membrane demonstrated effective MB rejection (>99%) whereas the 5 μm membrane exhibited a broad range of rejections (40% to >95%) depending on solution conditions and filtration time. Finally, a model was formulated utilizing FC forward light scattering intensity measurements to estimate MB sizes in samples. Using the model, a 33% reduction in median MB size, on average, was noted across the 5 μm membrane when filtering MBs suspended in humic acid solution, affirming a preferential permeation of smaller particles. Overall, this study advances MP quantification techniques towards validating removal processes.

Abstract:FTIR spectral identification is today's gold standard analytical procedure for plastic pollution material characterization. High-throughput FTIR techniques have been advanced for small microplastics (10–500 μm) but less so for large microplastics (500–5 mm) and macroplastics (> 5 mm). These larger plastics are typically analyzed using ATR, which is highly manual and can sometimes destroy particles of interest. Furthermore, spectral libraries are often inadequate due to the limited variety of reference materials and spectral collection modes, resulting from expensive spectral data collection. We advance a new high-throughput technique to remedy these problems using FTIR microplate readers for measuring large particles (> 500 μm). We created a new reference database of over 6000 spectra for transmission, ATR, and reflection spectral collection modes with over 600 plastic, organic, and mineral reference materials relevant to plastic pollution research. We also streamline future analysis in microplate readers by creating a new particle holder for transmission measurements using off-the-shelf parts and fabricating a nonplastic 96-well microplate for storing particles. We determined that particles should be presented to microplate readers as thin as possible due to thick particles causing poor-quality spectra and identifications. We validated the new database using Open Specy and demonstrated that additional transmission and reflection spectra reference data were needed in spectral libraries.

Abstract:Microplastics are of rising health concerns because they have been detected even in remote and pristine environments, from the Artic snow to the Marianne Trench. The occurrence and impact of nanoplastics in ecosystems is almost unknown, in particular due to analytical limitations such as very small sizes that fall below detection limits of current techniques. Here we take advantage of a common interference in analytical flow cytometry to develop a method for the quantification of the number of plastic particles in the 0.6–15 μm size range. Plastic particles are stained with the lipophilic dye Nile-Red then detected by flow cytometry, a method regularly used in biology for rapid quantification of fluorescent cells. We found that sample analysis lasts 90 s, which is hundreds of times faster than the analysis of filter portions by micro-Raman and other spectroscopic techniques. Our method is highly efficient in detecting polyethylene, with staining efficiency higher than 70% and signal linearity with concentration. Staining efficiency up to 96% was observed for polyvinylchloride and for polystyrene.

Abstract:Detection of microplastics in environmental samples requires fast, sensitive and selective analytical techniques, both in terms of the size of the microparticles and their concentration. Single particle inductively coupled plasma mass spectrometry (SP-ICP-MS) allows the detection of plastic particles down to ca. 1 μm and down to concentrations of 100 particles per mL. In SP-ICP-MS, detection of carbon-containing particles is hampered by the presence of other forms of carbon (carbonates, organic matter, microorganisms...). An acidic pre-treatment of river water samples with 10% (v/v) nitric acid for 24 h allowed the reduction of the presence of dissolved carbon to ultrapure water levels and the digestion of potential microorganisms in the samples, recovering polystyrene microparticles up to 80%. Carbon-containing particles were detected in most of the samples analysed from Spanish and French Pyrenean rivers. The presence of microplastics in these samples was confirmed by Raman microscopy and their morphology was defined by electron microscopy combined with energy-dispersive X-ray spectroscopy. The developed SP-ICP-MS method is suitable for the rapid screening of river waters for the presence of microplastics, which can then be analysed by inherently slower but more selective techniques (e.g., Raman microscopy).

Developing and testing a workflow to identify microplastics using near infrared hyperspectral imaging

Abstract:The analysis of microplastics (MP) is time-consuming which limits our capacity to monitor and mitigate plastic pollution. Here, near infrared (1000-2500 nm) hyperspectral imaging (NIR-HSI) offers an advantage over other spectroscopic techniques because it can rapidly image large areas relative to other systems. While NIR-HSI can successfully detect MP, accuracy and limitations of the method have not been fully explored. In addition, lack of open databases and analysis pipelines increases the barrier to use. In this work, we developed a spectral database containing preproduction pellets, consumer products and marine plastic debris, imaged using a Hyspex SWIR-320me imager. A SIMCA model identified four polymer types: polypropylene, polyethylene, polyethylene terephthalate and polystyrene (PP, PE, PET, PS) to identify MP in hyperspectral images. We determined the accuracy of size estimates for PS MP > 1000 μm using fluorescence microscopy and tested the impact of photooxidation on detection of plastics by NIR-HSI (PE, PP, PS, PET) and subsequent prediction by the SIMCA model. The model performed well across all polymers as shown by high specificity, sensitivity, and accuracy for internal cross validation (>88%), and sensitivity >80% for external validation. PS MP < 500 μm Feret diameter were not consistently detected by NIR-HSI when compared with fluorescence microscopy. However, estimates for Feret diameter were consistent for PS MP > 1000 μm. Analysis by NIR-HSI showed no spectral changes and SIMCA showed no decreased precision with increased photooxidation across polymer types. Recall varied across polymer type and photooxidation stage with no clear trends. This study shows that NIR-HSI is a rapid method which can accurately identify MP of the four most relevant polymer types, precluding the need to analyze particles one at a time. NIR-HSI can be a key technology for environmental monitoring of plastic debris where rapid analysis of multiple samples is required.

Abstract:Rapid identification of microparticles in liquid is an important problem in environmental and biomedical applications such as for microplastic detection in water sources and physiological fluids. Existing spectro-scopic techniques are usually slow and not compatible with flow-through systems. Here we analyze single microparticles in the 14 - 20 micrometer range using a combination of two electronic sensors in the same microfluidic system: a microwave capacitive sensor and a resistive pulse sensor. Together, this integrated sen-sor system yields the effective electrical permittivity of the analyte particles. To simplify data analysis, 3D electrode arrangements were used instead of planar electrodes, so that the generated signal is unaffected by the height of the particle in the microfluidic channel. With this platform, we were able to distinguish between polystyrene (PS) and polyethylene (PE) microparticles. We showcase the sensitivity and speed of this tech-nique and discuss the implications for the future application of microwave cytometry technology in the en-vironmental and biomedical fields.
