Abstract:To overcome the drawbacks of dehydration and low gel strength of the gels of L-arginine-treated konjac glucomannan (KGM), thermally irreversible gel was prepared by L-arginine induced KGM-modified starch mixed sol. The effects of the three modified starches (corn modified starch, acetylated bis-starch adipate, and hydroxypropyl distarch phosphate) on the mixed system were investigated by texture analysis, scanning electron microscopy, Low field NMR, Fourier transform infrared spectroscopy and Differential thermal analysis. The results showed a gradual increase in hardness with increasing concentration of modified starch. Differential thermal results indicated that acetylated bis-starch adipate composited gels had the highest thermal decomposition temperature (309 C). The changes in the textural properties and thermal stability of the three mixed gels were mainly attributed to the enhanced hydrogen bonding between starch/KGM/L-arginine, and changes in the microstructure. When the starch concentration was higher, the denser the gel network structure was, the more difficult it was for water trapped in the gel system to precipitate out. The results indicated that acetylated bisstarch adipate was more suitable to be added to the mixed system to prepare thermally irreversible gels.
Properties of acid gels made from sodium caseinate-maltodextrin conjugates prepared by a wet heating method

Abstract:Covalent attachment of polysaccharides to proteins (conjugation) via the Maillard reaction has been extensively studied. Conjugation can lead to a significant improvement in protein functionality (e.g., solubility, emulsification, and heat stability). Caseins have previously been successfully conjugated with maltodextrin (Md), but the effect on the detailed acid gelation properties has not been examined. We studied the effect of conjugating sodium caseinate (NaCN) with 3 different sized Md samples via the Maillard reaction in aqueous solutions. The Md samples had dextrose equivalents of 4 to 7, 9 to 12, and 20 to 23 for Md40, Md100, and Md200, respectively. The conjugation reaction was performed in mixtures with 5% NaCN and 5% Md, which were heated at 90°C for 10 h. The degree of conjugation was estimated from the reduction in free amino groups as well as color changes. Sodium dodecyl sulfate-PAGE analysis was performed to confirm conjugation by employing staining of both protein and carbohydrate bands. The molar mass of samples was determined by size-exclusion chromatography coupled with multi-angle laser light scattering. After the conjugation reaction, samples were then gelled by the addition of 0.63% (wt/vol) glucono-δ-lactone at 30°C, such that samples reached pH 4.6 after about 13 h. The rheological properties of samples during acidification was monitored by small-strain dynamic oscillatory rheology. The microstructure of acid gels at pH 4.6 was examined by fluorescence microscopy. Conjugation resulted in a loss of 10.8, 8.8, and 11.9% of the available amino groups in the protein for the NaCN-Md40 conjugates (C40), NaCN-Md100 conjugates (C100), and NaCN-Md100 conjugates (C200), respectively. With a decrease in the size of the type of Md, an increase occurred in the molar mass of the resultant conjugate. The weight average molar masses of NaCN-Md samples were 340, 368, and 425 kDa for the conjugates C40, C100, and C200, respectively. Addition of Md to NaCN dispersion resulted in slightly shorter acid gelation times even without the conjugation reaction. The storage modulus (G') of acid gels was significantly lower in conjugated samples compared with the corresponding (unreacted) mixtures of Md and NaCN. The sample with the lowest G' value at pH 4.6 was the C40 conjugate. Fluorescence microscopy showed that gels made by conjugates had slightly larger pores. These results indicate that conjugation of casein modified its acid gelation properties, presumably by the Md polysaccharide moiety hindering aggregation and rearrangements of the casein network.
Hydrolysis by Indigenous Plasmin: Consequences for Enzymatic Cross-Linking and Acid-Induced Gel Formation of Non-Micellar Casein

Abstract:Casein is a group of milk proteins with high nutritional value, and the exploitation of its techno-functional potentials has been investigated for decades. In this study, acid casein powder was dissolved in 0.1 mol/L phosphate buffers with different pH, resulting in casein solutions with pH 5.9, 6.6 and 7.3. During preparation and storage (40 °C) of the samples, casein hydrolysis was observed in size exclusion chromatography and gel electrophoresis. The degree of hydrolysis increased with increasing pH, and treatment of casein with commercial plasmin resulted in similar polypeptides, suggesting that the hydrolysis was caused by residual indigenous plasmin present in the acid casein powder. Most polypeptides could be cross-linked by microbial transglutaminase, except for one particular fraction which appeared at constant intensity in the chromatograms. The stiffness of acid-induced gels as determined in small amplitude oscillatory shear rheology decreased with increasing degree of hydrolysis, and was also lower for cross-linked samples when the preceding casein hydrolysis was more pronounced. Enzymatic cross-linking increased the resistance of casein against plasmin-related hydrolysis, presumably because of the resulting lysine modification. However, one particular fraction of polypeptides was released by hydrolysis in spite of cross-linking, suggesting that they did not contain lysine residues that are susceptible for mTGase. The results indicate that plasmin-related hydrolysis should be taken into account for the application of acid casein or sodium caseinate as additive in food design.

Abstract:This study aimed to investigate if differences in β-casein polymorphic structure would affect the acid-induced (glucono-δ-lactone) gelation behaviour of corresponding skim milks. Gels obtained from skim milk containing A2/A2 β-casein had significantly lower elastic modulus, water holding capacity, and gel permeability compared to A1/A1 and A1/A2 gels. Microscopy images also showed a denser microstructure and smaller pore size in acid-induced gels prepared with A1/A1 and A1/A2 milks compared to A2/A2 milk. A number of reasons may account for these differences in gelation, specifically, gels with A1 β-casein contained greater amounts of α-helixes and aggregated β-sheets in their secondary structure compared to A2/A2 gel that was comprised mainly of random coils or polyproline II helixes. In addition, compositional differences such as greater total and micellar calcium and higher levels of total κ-casein existed in A1/A1 and A1/A2 milks compared to A2/A2 milk and may have contributed to the difference in gelation and gel structure. Although the role of β-CN phenotype on skim milk gelation may need further investigation, findings from this study clearly indicate that A2/A2 milk was associated with poor acid gelation properties.

Modulation of acid-induced pea protein gels by gellan gum and glucono-δ-lactone: Rheological and microstructural insights

Abstract:This study investigated the effect of gellan gum (GG) and glucono-δ-lactone (GDL) on the acid-induced gel properties of pea protein isolate (PPI) pretreated with media milling. The inclusion of GG substantially enhanced the gel hardness of PPI gel from 18.69 g to 792.47 g though slightly reduced its water holding capacity (WHC). Rheological analysis showed that GG increased storage modulus (G') and decreased damping factor of gels in the small amplitude oscillatory shear region and transformed its strain thinning behavior into weak strain overshoot behavior in the large amplitude oscillatory shear region. SEM revealed that GG transformed the microstructure of gel from a uniform particle aggregate structure to a chain-like architecture composed of filaments with small protein particles attached. Turbidity and zeta potential analysis showed that GG promoted the transformation of PPI from a soluble polymer system to an insoluble coagulant during acidification. When GG content was relatively high (0.2 %-0.3 %), high GDL content increased the electrostatic interaction between PPI and GG molecules, causing their rapid aggregation into a dense irregular aggregate structure, further enhancing gel strength and WHC. Overall, GG and GDL can offer the opportunity to modulate the microstructure and gel properties of acid-induced PPI gels, presenting potential for diversifying food gel design strategies through PPI-GG hybrid systems.

Abstract:Partial replacement of milk proteins with plant proteins is a challenge due to the reported negative effect on physical and sensory properties. Understanding of how the mechanical properties of acidified milk gels can be restored when 30% casein is replaced with soy proteins is therefore explored. Mixtures of sodium caseinate (CAS) and soy proteins (SP) are pre-heated at different conditions (pH, ratio CAS:SP, temperature) prior to acidification with GDL in order to modulate aggregation, structure build-up and final mechanical properties of acidified gels with total protein content of 10% (w/w). Optimal results in mechanical response were obtained when CAS and SP were pre-heated together at a 7:3 protein ratio for 15 min at 95 °C. In addition, heat treatment of CAS-SP mixtures resulted in a shift to higher onset pH and a faster onset gelation as determined by dynamic oscillation rheology. From turbidity measurements a change in aggregation kinetics was observed which can be appointed to the altered aggregation propensity during acidification of the heat induced aggregates. Pre-heating of CAS and SP together resulted in an enhanced increase in aggregate size (as determined with light scattering) compared to unheated mixture or when only SP was heated. By pre-heating mixtures of CAS and SP at the right conditions prior to acidification, the mechanical response of mixed CAS–SP gels can be restored to that comparable with CAS only gels. As a result, 30% CAS can be successfully replaced with SP while maintaining mechanical properties.

Abstract:The present study examined the development of the gel network as well as the final structure obtained from mixed soymilk–cow's milk gels recombined with cream. An understanding of the structural changes occurring as a result of different process manipulations may aid with understanding the reported texture differences in these matrices. Results showed that changing the mode of homogenization, the composition of the fat globule surface changed. When both milk and soymilk were homogenized with the cream, the resulting gel had the highest gel stiffness at the end of fermentation. Confocal microscopy demonstrated that as long as milk was homogenized with cream (regardless of the presence of soymilk during homogenization), the resulting gel showed finer protein aggregates. On the other hand, without homogenization or when soymilk was homogenized with cream before milk addition, a coarser gel structure was noted. The composite gels where both casein micelles and soy proteins aggregated simultaneously appeared homogeneous compared to the gels formed by the gelation of caseins before soy proteins. This work clearly demonstrated that by modulating the timing of casein and soy protein aggregation, as well as the composition of the fat interface, it was possible to modify the gel structure and affect texture properties.

Intragastric restructuring dictates the digestive kinetics of heat-set milk protein gels of contrasting textures

Abstract:The gelation of milk proteins can be achieved by various means, enabling the development of diverse products. In this study, heat-set milk protein gels (15 % protein) of diverse textures were made by pH modulation and two gels were selected for dynamic in vitro gastric digestion: a spoonable soft gel (SG, pH 6.55' G' of ∼100 Pa) and a sliceable firm gel (FG, pH 5.65; G' of ∼7000 Pa). The two gels displayed markedly different structural changes and digestion kinetics during gastric digestion. The SG underwent substantial structural compaction during the first 120 min of gastric digestion into a denser and firmer gastric chyme (26.3 % crude protein, G* of ∼8500 Pa) than the chyme of the FG (15.7 % crude protein, G* of ∼3000 Pa). These contrasting intragastric structural changes of the gels reversed their original textural differences, which led to slower digestion and gastric emptying of proteins from the SG compared with the FG. The different intragastric pH profiles during the digestion of the two gels likely played a key role by modulating the proteolytic activity and specificity (to κ-casein) of pepsin. Preferential early cleavage of κ-casein in SG stimulated coagulation and compaction of solid chyme, whereas rapid hydrolysis of αS- and β-caseins in the FG weakened coagulation. This study provided new insights into controlling the structural development of dairy-based foods during gastric digestion and modulating digestion kinetics.
Dextran modulates physical properties of rennet‐induced milk gels

Abstract:Physical properties of rennet‐induced milk gels as core intermediates of cheese production are mainly affected by milk composition, type and amount of coagulation enzyme, and starter culture activity. We investigated model systems of reconstituted milk and dextran, which triggers effects on milk gels similar to exopolysaccharides from lactic acid bacteria. Furthermore clotting activity (0.02 or 0.04 IMCU mL−1) and milk pH adjusted prior to renneting (6.5 ‐ 5.7) were studied. A lower pH at renneting resulted in an earlier gelation onset, a higher gelation velocity and gel stiffness. The addition of dextran stabilized the gels especially at higher pH, and microstructural analysis revealed larger, more interconnected protein aggregates. However, at pH 5.7, a reverse effect was observed, indicating a destabilization of the casein network. The current study indicates that altering milk pH and addition of polysaccharides gives the potential to change textural properties of cheese by affecting rennet‐induced gelation.This article is protected by copyright. Influence of partially demineralized milk proteins on rheological properties and microstructure of acid gels

Abstract:Innovative clean label processes employed in the manufacture of acid gels are targeted to modify the structure of proteins that contribute to rheological properties. In the present study, CO2-treated milk protein concentrate powder with 80% protein in dry matter (TMPC80) was mixed with nonfat dry milk (NDM) in different ratios for the manufacture of acid gels. Dispersions of NDM and TMPC80 that provided 100, 90, 70, and 40% of protein from NDM were reconstituted to 4.0% (wt/wt) protein and 12.0% (wt/wt) total solids. Dispersions were adjusted to pH 6.5, followed by heat treatment at 90°C for 10 min. Glucono-δ-lactone was added and samples were incubated at 30°C, reaching pH 4.5 ± 0.05 after 4 h of incubation. Glucono-δ-lactone levels were adjusted to compensate for the lower buffering capacity of samples with higher proportions of TMPC80, which is attributable to the depletion of buffering minerals from both the serum and micellar phase during preparation of TMPC80. Sodium dodecyl sulfate-PAGE analysis indicated a higher amount of caseins in the supernatant of unheated suspensions with increasing proportions of CO2-treated TMPC80, attributable to the partial disruption of casein micelles in TMPC80. Heat treatment reduced the level of whey proteins in the supernatant due to the heat-induced association of whey proteins with casein micelles, the extent of which was larger in samples containing more micellar casein (i.e., samples with a lower proportion of TMPC80). Particle size analysis showed only small differences between nonheated and heated dispersions. Gelation pH increased from ˜5.1 to ˜5.3, and the storage modulus of the gels at pH 4.5 increased from ˜300 to ˜420 Pa when the proportion of protein contributed by TMPC80 increased from 0 to 60%. Water-holding capacity also increased and gel porosity decreased with increasing proportion of protein contributed by TMPC80. The observed gel properties were in line with microstructural observations by confocal microscopy, wherein sample gels containing increasing levels of TMPC80 exhibited smaller, well-connected aggregates with uniform, homogeneous pore sizes. We concluded that TMPC80 can be used to partially replace NDM as a protein source to improve rheological and water-holding properties in acid gels. The resultant gels also exhibited decreased buffering, which can improve the productive capacity of yogurt manufacturing plants. Overall, the process can be leveraged to reduce the amount of hydrocolloids added to improve yogurt consistency and water-holding capacity, thus providing a path to meet consumer expectations of clean label products.
Characterization of the gelation and resulting network of a mixed-protein gel derived from sodium caseinate and ovalbumin in the presence of glucono-$δ$-lactone

Abstract:We investigated mixed-protein gels made from sodium caseinate and ovalbumin at different ratios with use of the acidification agent glucono-$\delta$-lactone. Dynamic viscoelastic measurements revealed that increasing the ovalbumin content decreased the mechanical properties of the gel but accelerated onset time of the phase transition. Ultrasound spectroscopy during gelation revealed that the relative velocity gradually decreased, whereas the ultrasonic attenuation increased during the whole acidification process until gelation was complete, although these changes were much smaller than those observed with heat-induced gelation. Confocal laser scanning microscopy along with scanning electron microscopy revealed that although uniform mixing of sodium caseinate and ovalbumin was observed, sodium caseinate is likely to mainly lead formation of the gel network, and the porosity of the resulting gel network depends on the ratio of these two components. The results demonstrate that confocal laser scanning microscopy is a useful tool for analyzing both the networks within mixed-protein gels and the contribution of each protein to the network and gelation.

Influence of Microbial Transglutaminase on Physicochemical and Cross-Linking Characteristics of Individual Caseins

Abstract:The effects of microbial transglutaminase (MTGase) cross-linking on the physicochemical characteristics of individual caseins were investigated. MTGase was used to modify three major individual caseins, namely, κ-casein (κ-CN), αS-casein (αS-CN) and β-casein (β-CN). The SDS-PAGE analysis revealed that MTGase-induced cross-linking occurred during the reaction and that some components with high molecular weights (>130 kDa) were formed from the individual proteins κ-CN, αS-CN and β-CN. Scanning electron microscopy (SEM) and particle size analysis respectively demonstrated that the κ-CN, αS-CN and β-CN particle diameters and protein microstructures were larger and polymerized after MTGase cross-linking. The polymerized κ-CN (~749.9 nm) was smaller than that of β-CN (~7909.3 nm) and αS-CN (~7909.3 nm). The enzyme kinetics results showed KM values of 3.04 × 10−6, 2.37 × 10−4 and 8.90 × 10−3 M for κ-CN, αS-CN and β-CN, respectively, and, furthermore, kcat values of 5.17 × 10−4, 1.92 × 10−3 and 4.76 × 10−2 1/s, for κ-CN, αS-CN and β-CN, respectively. Our results revealed that the cross-linking of β-CN catalyzed by MTGase was faster than that of αS-CN or κ-CN. Overall, the polymers that formed in the individual caseins in the presence of MTGase presented a higher molecular weight and larger particles.

Abstract:Glutaraldehyde (GA) reacts with amino groups in proteins, forming intermolecular cross-links that, at sufficiently high protein concentration, can transform a protein solution into a gel. Although GA has been used as a cross-linking reagent for decades, neither the cross-linking chemistry nor the microstructure of the resulting protein gel have been clearly established. Here we use small-angle X-ray scattering (SAXS) to characterise the microstructure and structural kinetics of gels formed by cross-linking of pancreatic trypsin inhibitor, myoglobin or intestinal fatty acid-binding protein. By comparing the scattering from gels and dilute solutions, we extract the structure factor and the pair correlation function of the gels. The protein gels are spatially heterogeneous, with dense clusters linked by sparse networks. Within the clusters, adjacent protein molecules are almost in contact, but the protein concentration in the cluster is much lower than in a crystal. At the ∼1 nm SAXS resolution, the native protein structure is unaffected by cross-linking. The cluster radius is in the range 10-50 nm, with the cluster size determined mainly by the availability of lysine amino groups on the protein surface. The development of structure in the gel, on time scales from minutes to hours, appears to obey first-order kinetics. Cross-linking is slower at acidic pH, where the population of amino groups in the reactive deprotonated form is low. These results support the use of cross-linked protein gels in NMR studies of protein dynamics and for modeling NMR relaxation in biological tissue.

Abstract:The objective of this study is to enhance the comprehension of the mechanism of enzymatic gelation in milk by investigating statistically significant changes in milk viscosity and carrying out corresponding electron microscopic studies of the process of structure formation at the enzymatic stage of coagulation using various enzyme preparations. Employing a non-destructive method (Hot-Wire) to monitor viscosity, we were able to confirm a decrease in viscosity during the middle of this stage and identify a previously undescribed peak in viscosity change at the end of the enzymatic stage. By conducting parallel studies utilizing transmission electron microscopy and various methods of specimen preparation, it was possible to discover that a hierarchical transformation process in the protein component of milk started at the end of the enzymatic stage of gelation in milk. This process is triggered by a cooperative conformational transition in casein micelle clusters, which leads to a series of increasingly energy-intensive reactions resulting in the transformation of loosely bound micelle clusters into denser aggregates. The final structure of the milk gel primarily consists of the previously formed individual micelle aggregates. It was noted that no changes in the microstructure of the milk gel were observed during the enzymatic stage of gelation when milk-clotting enzymes (MCEs) of animal, plant, and microbial (GMO) origins were used. Furthermore, investigations into the molecular weight distribution of soluble protein substances in samples generated with different types of MCEs revealed that the enzyme derived from the fungi of Rhizomucor miehei had a greater proteolytic effect on milk proteins compared to other variants of MCEs.

Abstract:Protein gel matrices are responsible for the texture of many foods. Therefore an understanding of the chemical reactions and physical processes associated with fracture properties of gels provides a fundamental understanding of select mechanical properties associated with texture. Globular proteins form thermally induced gels that are classified as fine-stranded, mixed or particulate, based on the protein network appearance. The fundamental properties of true shear stress and true shear strain at fracture, used to describe the physical properties of gels, depend on the gel network. Type and amount of mineral salt in whey protein and β-lactoglobulin protein dispersions determines the type of thermally induced gel matrix that forms, and thus its fracture properties. A fine-stranded matrix is formed when protein suspensions contain monovalent cation (Li+, K+, Rb+, Cs+) chlorides, sodium sulfate or sodium phosphate at ionic strengths ≤0.1 mol/dm3. This matrix has a well-defined network structure, and varies in stress and strain at fracture at different salt concentrations. At ionic strengths >0.1 mol/dm3 the matrix becomes mixed. This network appears as a combination of fine strands and spherical aggregates, and has high stress values and minimum strain values at fracture. Higher concentrations of monovalent cation salts cause the formation of particulate gels, which are high in stress and strain at fracture. The salt concentration required to change microstructure depends on the salt's position in the Hofmeister series. The formation of a particulate matrix also occurs when protein suspensions contain low concentrations (10–20 mol/dm3) of divalent cation (Ca2+, Mg2+, Ba2+) chloride salts or di-cationic 1,6-hexanediamine at pH 7.0. The divalent cation effect on β-lactoglobulin gelation is associated with minor changes in tertiary structure involving amide—amide interproton connectivities (determined by 1H NMR) at 40–45°C, increasing hydrophobicity and intermolecular aggregation. The type of matrix formed appears to be related to the dispersed or aggregated state of proteins prior to denaturation. Mixed and particulate matrices result from conditions which favor aggregation at temperatures (25–45°C) which are much lower than the denaturation temperature (~65°C). Therefore, general (e.g. Hofmeister series) and protein-specific factors can affect the dispersibility of proteins and thereby determine the microstructure and fracture properties of globular protein gels.

Abstract:The ligand binding preferences of a series of potentially pentadentate pyridylbis(aminophenol) ligands were explored. In addition to the previously reported ligands 2,2'-(2-methyl-2-(pyridin-2-yl)propane-1,3-diyl)bis(azanediyl)bis(methylene)diphenol (H(2)L(1)) and 6,6'-(2-methyl-2-(pyridin-2-yl)propane-1,3-diyl)bis(azanediyl)bis(methylene)bis(2,4-di-tert-butylphenol) (H(2)L(1-tBu)), four new ligands were synthesized: 6,6'-(2-methyl-2(pyridine-2-yl)propane-1,3-diyl)bis(azanediyl)bis(methylene)bis(2,4-dibromophenol) (H(2)L(1-Br)), 6,6'-(2-methyl-2(pyridine-2-yl)propane-1,3diyl)bis(azanediyl)bis(methylene)bis(2-methoxyphenol) (H(2)L(1-MeO)), 2,2'-(2-methyl-2(pyridine-2-yl)propane-1,3diyl)bis(azanediyl)bis(methylene)bis(4-nitrophenol) (H(2)L(1-NO2)), and 2,2'-(2-phenylpropane-1,3-diyl)bis(azanediyl)bis(methylene)diphenol (H(2)L(2)). These ligands, when combined with copper(II) salts and base, form either tricopper(II) species or monocopper(II) species depending on the nucleophilicity of the phenol groups in the ligands. All copper complexes were characterized by X-ray crystallography, cyclic voltammetry, and spectroscopic methods in solution. The ligands in trimeric complexes [{CuL(1)(CH(3)CN)}(2)Cu](ClO(4))(2) (1), [{CuL(1)Cl}(2)Cu] (1a), and [{CuL(2)(CH(3)CN)}(2)Cu](ClO(4))(2) (1b) and monomeric complex [CuL(1-tBu)(CH(3)OH)] (2) coordinate in a tetradentate mode via the amine N atoms and the phenolato O atoms. The pyridyl groups in 1, 1a, and 2 do not coordinate, but instead are involved in hydrogen bonding. Monomeric complexes [CuL(1-Br)] (3a), [CuL(1-NO2)] (3b), and [CuL(1-MeO)Na(CH(3)OH)(2)]ClO(4) (3c) have their ligands coordinated in a pentadentate mode via the amine N atoms, the phenolato O atoms, and the pyridyl N atom. The differences in tetradentate vs pentadentate coordination preferences of the ligands correlate to the nucleophilicity of the phenolate donor groups, and coincide with the electrochemical trends for these complexes.
Transglutaminase modified type A gelatin gel: The influence of intra-molecular and inter-molecular cross-linking on structure-properties

Abstract:In this work, the structure-property differences and mechanisms of gelatin induced by transglutaminase (TGase) covalent cross-linking were investigated. The results showed that higher hardness was obtained with a cross-linking degree of 13.55%, attributing to an active intra-molecular cross-linking and a tight network structure under a certain amount of triple helix-like structure. The inter-molecular cross-linking played a positive role in the transition from random coil to left-handed single helix chains, while triple helix-like structure could not been formed owing to the steric hindrance and hydrogen bond reduction, showing the decrease of hardness and increase of viscosity. Additionally, the ε-(γ-Glu)-Lys isopeptide bond significantly increased the initial degradation temperature. Macromolecular polymers with a cross-linking degree of 37.48% allowed sol state of gelatin gel could be maintained even heating at 100 ℃ for 10 min. This study might provide an innovative reference for the regulation and application of gelatin gel structure-properties.

Abstract:The purpose of this study was to investigate the influence of two-step sequential cross-linking induced by laccase and heat treatment on the conformation and gelling properties of low-salt myofibrillar proteins (MP) modified by konjac glucomannan (KGM) and ultrasound treatments. Adding sugar beet pectin (SBP), irrespective of laccase crosslinking, significantly decreased the fluorescence intensity of low-salt MPs (MP, UMP, UMP-KGM). Laccase crosslinking caused the exposure of hydrophobic and Tyr residues, induced moderate aggregation of MPs, and enhanced rheological properties and gel strength of composite MP gels, especially in the UMP-KGM sample. This result could also be corroborated by the highest WHC, improved textural characteristics, enhanced intermolecular interactions, and increased percentage in t-g-g conformation of S–S bridges in the SBP modified UMP-KGM gel after laccase crosslinking. Therefore, the dual crosslinking could be used to regulate the protein-protein and protein-polysaccharide interactions and improve the gel properties of low-salt meat products.

Abstract:The popularity of fat-free fermented concentrated milk products, such as fresh cheeses and high-protein yogurt, has increased over the recent years, attributed to greater availability and improvements in taste and texture. These improvements have been achieved through modifications and new developments in processing technologies, for example, higher heat treatment intensities and incorporating different membrane filtration technologies. Though numerous processing parameters are discussed in the literature, as well as reasons behind the developments, detailed examinations of how process modifications affect the final textural attributes of these products are lacking. To draw links between processing parameters and texture, we review the literature on fat-free fermented concentrated milk products from the perspective of fermented milk protein-based microgel particles as the basic structural unit. At each main processing step, relationships between process parameters, micro- and macrostructural and sensory (textural) properties are discussed.An overview of particle characteristics that drive structural changes at each processing step is developed in relation to textural characteristics. Using this approach of assessing relationships between structural characteristics of concentrated dispersions of fat-free fermented milk protein-based microgel particles and processing parameters provides a basic context for the selection of optimal parameters to achieve a desired texture.
