Abstract:Machine learning (ML) has transformed neuroimaging research by enabling accurate predictions and feature extraction from large datasets. In this study, we investigate the application of six ML algorithms (Lasso, relevance vector regression, support vector regression, extreme gradient boosting, category boost, and multilayer perceptron) to predict brain age for middle-aged and older adults, which is a crucial area of research in neuroimaging. Despite the plethora of proposed ML models, there is no clear consensus on how to achieve better performance in brain age prediction for this population. Our study stands out by evaluating the impact of both ML algorithms and image modalities on brain age prediction performance using a large cohort of cognitively normal adults aged 44.6 to 82.3 years old (N = 27,842) with six image modalities. We found that the predictive performance of brain age is more reliant on the image modalities used than the ML algorithms employed. Specifically, our study highlights the superior performance of T1-weighted MRI and diffusion-weighted imaging and demonstrates that multi-modality-based brain age prediction significantly enhances performance compared to unimodality. Moreover, we identified Lasso as the most accurate ML algorithm for predicting brain age, achieving the lowest mean absolute error in both single-modality and multi-modality predictions. Additionally, Lasso also ranked highest in a comprehensive evaluation of the relationship between BrainAGE and the five frequently mentioned BrainAGE-related factors. Notably, our study also shows that ensemble learning outperforms Lasso when computational efficiency is not a concern. Overall, our study provides valuable insights into the development of accurate and reliable brain age prediction models for middle-aged and older adults, with significant implications for clinical practice and neuroimaging research. Our findings highlight the importance of image modality selection and emphasize Lasso as a promising ML algorithm for brain age prediction.

Abstract:Background: Alzheimer's disease (AD) is a leading cause of dementia, and it is significantly influenced by the apolipoprotein E4 (APOE4) gene and gender. This study aimed to use machine learning (ML) algorithms to predict brain age and assess AD risk by considering the effects of the APOE4 genotype and gender. Methods: We collected brain volumetric MRI data and medical records from 1100 cognitively unimpaired individuals and 602 patients with AD. We applied three ML regression models-XGBoost, random forest (RF), and linear regression (LR)-to predict brain age. Additionally, we introduced two novel metrics, brain age difference (BAD) and integrated difference (ID), to evaluate the models' performances and analyze the influences of the APOE4 genotype and gender on brain aging. Results: Patients with AD displayed significantly older brain ages compared to their chronological ages, with BADs ranging from 6.5 to 10 years. The RF model outperformed both XGBoost and LR in terms of accuracy, delivering higher ID values and more precise predictions. Comparing the APOE4 carriers with noncarriers, the models showed enhanced ID values and consistent brain age predictions, improving the overall performance. Gender-specific analyses indicated slight enhancements, with the models performing equally well for both genders. Conclusions: This study demonstrates that robust ML models for brain age prediction can play a crucial role in the early detection of AD risk through MRI brain structural imaging. The significant impact of the APOE4 genotype on brain aging and AD risk is also emphasized. These findings highlight the potential of ML models in assessing AD risk and suggest that utilizing AI for AD identification could enable earlier preventative interventions.

Abstract:Biological brain age predicted using machine learning models based on high-resolution imaging data has been suggested as a potential biomarker for neurological and cerebrovascular diseases. In this work, we aimed to develop deep learning models to predict the biological brain age using structural magnetic resonance imaging and angiography datasets from a large database of 2074 adults (21-81 years). Since different imaging modalities can provide complementary information, combining them might allow to identify more complex aging patterns, with angiography data, for instance, showing vascular aging effects complementary to the atrophic brain tissue changes seen in T1-weighted MRI sequences. We used saliency maps to investigate the contribution of cortical, subcortical, and arterial structures to the prediction. Our results show that combining T1-weighted and angiography MR data led to a significantly improved brain age prediction accuracy, with a mean absolute error of 3.85 years comparing the predicted and chronological age. The most predictive brain regions included the lateral sulcus, the fourth ventricle, and the amygdala, while the brain arteries contributing the most to the prediction included the basilar artery, the middle cerebral artery M2 segments, and the left posterior cerebral artery. Our study proposes a framework for brain age prediction using multimodal imaging, which gives accurate predictions and allows identifying the most predictive regions for this task, which can serve as a surrogate for the brain regions that are most affected by aging.

Abstract:Brain structural morphology varies over the aging trajectory, and the prediction of a person's age using brain morphological features can help the detection of an abnormal aging process. Neuroimaging-based brain age is widely used to quantify an individual's brain health as deviation from a normative brain aging trajectory. Machine learning approaches are expanding the potential for accurate brain age prediction but are challenging due to the great variety of machine learning algorithms. Here, we aimed to compare the performance of the machine learning models used to estimate brain age using brain morphological measures derived from structural magnetic resonance imaging scans. We evaluated 27 machine learning models, applied to three independent datasets from the Human Connectome Project (HCP, n = 1113, age range 22–37), the Cambridge Centre for Ageing and Neuroscience (Cam-CAN, n = 601, age range 18–88), and the Information eXtraction from Images (IXI, n = 567, age range 19–86). Performance was assessed within each sample using cross-validation and an unseen test set. The models achieved mean absolute errors of 2.75–3.12, 7.08–10.50, and 8.04–9.86 years, as well as Pearson's correlation coefficients of 0.11–0.42, 0.64–0.85, and 0.63–0.79 between predicted brain age and chronological age for the HCP, Cam-CAN, and IXI samples, respectively. We found a substantial difference in performance between models trained on the same data type, indicating that the choice of model yields considerable variation in brain-predicted age. Furthermore, in three datasets, regularized linear regression algorithms achieved similar performance to nonlinear and ensemble algorithms. Our results suggest that regularized linear algorithms are as effective as nonlinear and ensemble algorithms for brain age prediction, while significantly reducing computational costs. Our findings can serve as a starting point and quantitative reference for future efforts at improving brain age prediction using machine learning models applied to brain morphometric data.

Abstract:The increasing lifespan and large individual differences in cognitive capability highlight the importance of comprehending the aging process of the brain. Contrary to visible signs of bodily ageing, like greying of hair and loss of muscle mass, the internal changes that occur within our brains remain less apparent until they impair function. Brain age, distinct from chronological age, reflects our brain's health status and may deviate from our actual chronological age. Notably, brain age has been associated with mortality and depression. The brain is plastic and can compensate even for severe structural damage by rewiring. Functional characterization offers insights that structural cannot provide. Contrary to the multitude of studies relying on structural magnetic resonance imaging (MRI), we utilize resting-state functional MRI (rsfMRI). We also address the issue of inclusion of subjects with abnormal brain ageing through outlier removal. In this study, we employ the Least Absolute Shrinkage and Selection Operator (LASSO) to identify the 39 most predictive correlations derived from the rsfMRI data. The data is from a cohort of 176 healthy right-handed volunteers, aged 18-78 years (95/81 male/female, mean age 48, SD 17) collected at the Mind Research Imaging Center at the National Cheng Kung University. We establish a normal reference model by excluding 68 outliers, which achieves a leave-one-out mean absolute error of 2.48 years. By asking which additional features that are needed to predict the chronological age of the outliers with a smaller error, we identify correlations predictive of abnormal aging. These are associated with the Default Mode Network (DMN). Our normal reference model has the lowest prediction error among published models evaluated on adult subjects of almost all ages and is thus a candidate for screening for abnormal brain aging that has not yet manifested in cognitive decline. This study advances our ability to predict brain aging and provides insights into potential biomarkers for assessing brain age, suggesting that the role of DMN in brain aging should be studied further.

Abstract:Introduction: In the last few years, several models trying to calculate the biological brain age have been proposed based on structural magnetic resonance imaging scans (T1-weighted MRIs, T1w) using multivariate methods and machine learning. We developed and validated a convolutional neural network (CNN)-based biological brain age prediction model that uses one T1w MRI preprocessing step when applying the model to external datasets to simplify implementation and increase accessibility in research settings. Our model only requires rigid image registration to the MNI space, which is an advantage compared to previous methods that require more preprocessing steps, such as feature extraction. Methods: We used a multicohort dataset of cognitively healthy individuals (age range = 32.0–95.7 years) comprising 17,296 MRIs for training and evaluation. We compared our model using hold-out (CNN1) and cross-validation (CNN2–4) approaches. To verify generalisability, we used two external datasets with different populations and MRI scan characteristics to evaluate the model. To demonstrate its usability, we included the external dataset's images in the cross-validation training (CNN3). To ensure that our model used only the brain signal on the image, we also predicted brain age using skull-stripped images (CNN4). Results:: The trained models achieved a mean absolute error of 2.99, 2.67, 2.67, and 3.08 years for CNN1–4, respectively. The model's performance in the external dataset was in the typical range of mean absolute error (MAE) found in the literature for testing sets. Adding the external dataset to the training set (CNN3), overall, MAE is unaffected, but individual cohort MAE improves (5.63–2.25 years). Salience maps of predictions reveal that periventricular, temporal, and insular regions are the most important for age prediction. Discussion: We provide indicators for using biological (predicted) brain age as a metric for age correction in neuroimaging studies as an alternative to the traditional chronological age. In conclusion, using different approaches, our CNN-based model showed good performance using one T1w brain MRI preprocessing step. The proposed CNN model is made publicly available for the research community to be easily implemented and used to study ageing and age-related disorders.

Abstract:Leveraging transfer learning, we introduce a deep learning framework trained on vast clinical MRI data to predict brain age with high accuracy across five MRI sequences. Our versatile, open‐source models, effective even with limited data, promote wider application in diverse clinical scenarios and advance brain age research. Estimated age from brain MRI data has emerged as a promising biomarker of neurological health. However, the absence of large, diverse, and clinically representative training datasets, along with the complexity of managing heterogeneous MRI data, presents significant barriers to the development of accurate and generalisable models appropriate for clinical use. Here, we present a deep learning framework trained on routine clinical data (N up to 18,890, age range 18–96 years). We trained five separate models for accurate brain age prediction (all with mean absolute error ≤4.0 years, R2 ≥ .86) across five different MRI sequences (T2‐weighted, T2‐FLAIR, T1‐weighted, diffusion‐weighted, and gradient‐recalled echo T2*‐weighted). Our trained models offer dual functionality. First, they have the potential to be directly employed on clinical data. Second, they can be used as foundation models for further refinement to accommodate a range of other MRI sequences (and therefore a range of clinical scenarios which employ such sequences). This adaptation process, enabled by transfer learning, proved effective in our study across a range of MRI sequences and scan orientations, including those which differed considerably from the original training datasets. Crucially, our findings suggest that this approach remains viable even with limited data availability (as low as N = 25 for fine‐tuning), thus broadening the application of brain age estimation to more diverse clinical contexts and patient populations. By making these models publicly available, we aim to provide the scientific community with a versatile toolkit, promoting further research in brain age prediction and related areas.

Abstract:In the pursuit of studying brain aging, numerous models to predict brain age from T1-weighted MRI have been developed. Recently, many of these models take advantage of deep learning architectures, and although they have shown some success in terms of reported Mean Absolute Error in predicted age, doubts linger regarding their generalization ability due to the large number of trainable parameters in deep models and the limited training data available in medical imaging datasets. Unfortunately, the evaluation details of the proposed models' generalization ability are often overlooked in the literature. In this study, we assess a deep model, SFCN-reg, based on the VGG-16 architecture, and examine whether we can enhance its generalizability and robustness by (1) employing a more than minimal preprocessing pipeline, (2) incorporating more extensive data augmentation during training, and (3) applying regularization to the model. We found that the age prediction mean absolute error in independent out-of-distribution publicly available cohorts is reduced from 5.25 years to 2.96 years (44% improvement) in the Alzheimer's Disease Neuroimaging Initiative (ADNI) dataset and from 4.35 years to 3.40 years (22% improvement) in the Australian Imaging, Biomarker and Lifestyle (AIBL) dataset, closing the generalization gap to less than 0.9 years when better preprocessing is used. Moreover, data augmentation and model regularization reduce the scan-rescan error from 0.86 years to 0.61 years (29% improvement), increasing the model robustness. The final model is also much less sensitive against registration errors, showing almost no bias in comparison with other models when facing incorrectly registered images.

Abstract:The Brain Age Gap (BAG), which refers to the difference between chronological age and predicted neuroimaging age, is proposed as a potential biomarker for age-related brain degeneration. However, existing brain age prediction models usually rely on a single marker and can not discover meaningful hidden information in radiographic images. This study focuses on the application of radiomics, an advanced imaging analysis technique, combined with automated machine learning to predict BAG. Our methods achieve a promising result with a mean absolute error of 1.509 using the Alzheimer's Disease Neuroimaging Initiative dataset. Furthermore, we find that the hippocampus and parahippocampal gyrus play a significant role in predicting age with interpretable method called SHapley Additive exPlanations. Additionally, our investigation of age prediction discrepancies between patients with Alzheimer's disease (AD) and those with mild cognitive impairment (MCI) reveals a notable correlation with clinical cognitive assessment scale scores. This suggests that BAG has the potential to serve as a biomarker to support the diagnosis of AD and MCI. Overall, this study presents valuable insights into the application of neuroimaging models in the diagnosis of neurodegenerative diseases.

Abstract:Brain morphology varies across the ageing trajectory and the prediction of a person's age using brain features can aid the detection of abnormalities in the ageing process. Existing studies on such "brain age prediction" vary widely in terms of their methods and type of data, so at present the most accurate and generalisable methodological approach is unclear. Therefore, we used the UK Biobank data set (N = 10,824, age range 47–73) to compare the performance of the machine learning models support vector regression, relevance vector regression and Gaussian process regression on whole‐brain region‐based or voxel‐based structural magnetic resonance imaging data with or without dimensionality reduction through principal component analysis. Performance was assessed in the validation set through cross‐validation as well as an independent test set. The models achieved mean absolute errors between 3.7 and 4.7 years, with those trained on voxel‐level data with principal component analysis performing best. Overall, we observed little difference in performance between models trained on the same data type, indicating that the type of input data had greater impact on performance than model choice. All code is provided online in the hope that this will aid future research.

Abstract:Structural neuroimaging data have been used to compute an estimate of the biological age of the brain (brain-age) which has been associated with other biologically and behaviorally meaningful measures of brain development and aging. The ongoing research interest in brain-age has highlighted the need for robust and publicly available brain-age models pre-trained on data from large samples of healthy individuals. To address this need we have previously released a developmental brain-age model. Here we expand this work to develop, empirically validate, and disseminate a pre-trained brain-age model to cover most of the human lifespan. To achieve this, we selected the best-performing model after systematically examining the impact of seven site harmonization strategies, age range, and sample size on brain-age prediction in a discovery sample of brain morphometric measures from 35,683 healthy individuals (age range: 5-90 years; 53.59% female). The pre-trained models were tested for cross-dataset generalizability in an independent sample comprising 2101 healthy individuals (age range: 8-80 years; 55.35% female) and for longitudinal consistency in a further sample comprising 377 healthy individuals (age range: 9-25 years; 49.87% female). This empirical examination yielded the following findings: (1) the accuracy of age prediction from morphometry data was higher when no site harmonization was applied; (2) dividing the discovery sample into two age-bins (5-40 and 40-90 years) provided a better balance between model accuracy and explained age variance than other alternatives; (3) model accuracy for brain-age prediction plateaued at a sample size exceeding 1600 participants. These findings have been incorporated into CentileBrain (https://centilebrain.org/#/brainAGE2), an open-science, web-based platform for individualized neuroimaging metrics.

Abstract:Brain age is an imaging-based biomarker with excellent feasibility for characterizing individual brain health and may serve as a single quantitative index for clinical and domain-specific usage. Brain age has been successfully estimated using extensive neuroimaging data from healthy participants with various feature extraction and conventional machine learning (ML) approaches. Recently, several end-to-end deep learning (DL) analytical frameworks have been proposed as alternative approaches to predict individual brain age with higher accuracy. However, the optimal approach to select and assemble appropriate input feature sets for DL analytical frameworks remains to be determined. In the Predictive Analytics Competition 2019, we proposed a hierarchical analytical framework which first used ML algorithms to investigate the potential contribution of different input features for predicting individual brain age. The obtained information then served as a priori knowledge for determining the input feature sets of the final ensemble DL prediction model. Systematic evaluation revealed that ML approaches with multiple concurrent input features, including tissue volume and density, achieved higher prediction accuracy when compared with approaches with a single input feature set [Ridge regression: mean absolute error (MAE) = 4.51 years, R 2 = 0.88; support vector regression, MAE = 4.42 years, R 2 = 0.88]. Based on this evaluation, a final ensemble DL brain age prediction model integrating multiple feature sets was constructed with reasonable computation capacity and achieved higher prediction accuracy when compared with ML approaches in the training dataset (MAE = 3.77 years; R 2 = 0.90). Furthermore, the proposed ensemble DL brain age prediction model also demonstrated sufficient generalizability in the testing dataset (MAE = 3.33 years). In summary, this study provides initial evidence of how-to efficiency for integrating ML and advanced DL approaches into a unified analytical framework for predicting individual brain age with higher accuracy. With the increase in large open multiple-modality neuroimaging datasets, ensemble DL strategies with appropriate input feature sets serve as a candidate approach for predicting individual brain age in the future.

Abstract:Abstract To better assess the pathology of neurodegenerative disorders and the efficacy of neuroprotective interventions, it is necessary to develop biomarkers that can accurately capture age-related biological changes in the human brain. Brain serotonin 2A receptors (5-HT2AR) show a particularly profound age-related decline and are also reduced in neurodegenerative disorders, such as Alzheimer’s disease. This study investigates whether the decline in 5-HT2AR binding, measured in vivo using positron emission tomography (PET), can be used as a biomarker for brain aging. Specifically, we aim to (1) predict brain age using 5-HT2AR binding outcomes, (2) compare 5-HT2AR-based predictions of brain age to predictions based on gray matter (GM) volume, as determined with structural magnetic resonance imaging (MRI), and (3) investigate whether combining 5-HT2AR and GM volume data improves prediction. We used PET and MR images from 209 healthy individuals aged between 18 and 85 years (mean = 38, std = 18) and estimated 5-HT2AR binding and GM volume for 14 cortical and subcortical regions. Different machine learning algorithms were applied to predict chronological age based on 5-HT2AR binding, GM volume, and the combined measures. The mean absolute error (MAE) and a cross-validation approach were used for evaluation and model comparison. We find that both the cerebral 5-HT2AR binding (mean MAE = 6.63 years, std = 0.74 years) and GM volume (mean MAE = 6.95 years, std = 0.83 years) predict chronological age accurately. Combining the two measures improves the prediction further (mean MAE = 5.54 years, std = 0.68). In conclusion, 5-HT2AR binding measured using PET might be useful for improving the quantification of a biomarker for brain aging.

Brain age prediction in schizophrenia: does the choice of machine learning algorithm matter?

Abstract:Abstract Background Schizophrenia has been associated with lifelong deviations in the normative trajectories of brain structure. These deviations can be captured using the brain-predicted age difference (brainPAD), which is the difference between the biological age of an individual’s brain, as inferred from neuroimaging data, and their chronological age. Various machine learning algorithms are currently used for this purpose but their comparative performance has yet to be systematically evaluated. Methods Six linear regression algorithms, ordinary least squares (OLS) regression, ridge regression, least absolute shrinkage and selection operator (Lasso) regression, elastic-net regression, linear support vector regression (SVR), and relevance vector regression (RVR), were applied to brain structural data acquired on the same 3T scanner using identical sequences from patients with schizophrenia (n=90) and healthy individuals (n=200). The performance of each algorithm was quantified by the mean absolute error (MAE) and the correlation (R) between predicted brain-age and chronological age. The inter-algorithm similarity in predicted brain-age, brain regional regression weights and brainPAD were compared using correlation analyses and hierarchical clustering. Results In patients with schizophrenia, ridge regression, Lasso regression, elastic-net regression, and RVR performed very similarly and showed a high degree of correlation in predicted brain-age (R>0.94) and brain regional regression weights (R>0.66). By contrast, OLS regression, which was the only algorithm without a penalty term, performed markedly worse and showed a lower similarity with the other algorithms. The mean brainPAD was higher in patients than in healthy individuals but varied by algorithm from 3.8 to 5.2 years although all analyses were performed on the same dataset. Conclusions Linear machine learning algorithms, with the exception of OLS regression, have comparable performance for age prediction on the basis of a combination of cortical and subcortical structural measures. However, algorithm choice introduced variation in brainPAD estimation, and therefore represents an important source of inter-study variability.

Abstract:As an important biomarker of neural aging, the brain age reflects the integrity and health of the human brain. Accurate prediction of brain age could help to understand the underlying mechanism of neural aging. In this study, a cross-stratified ensemble learning algorithm with staking strategy was proposed to obtain brain age and the derived predicted age difference (PAD) using T1-weighted magnetic resonance imaging (MRI) data. The approach was characterized as by implementing two modules: one was three base learners of 3D-DenseNet, 3D-ResNeXt, 3D-Inception-v4; another was 14 secondary learners of liner regressions. To evaluate performance, our method was compared with single base learners, regular ensemble learning algorithms, and state-of-the-art (SOTA) methods. The results demonstrated that our proposed model outperformed others models, with three metrics of mean absolute error (MAE), root mean-squared error (RMSE), and coefficient of determination (R2) of 2.9405 years, 3.9458 years, and 0.9597, respectively. Furthermore, there existed significant differences in PAD among the three groups of normal control (NC), mild cognitive impairment (MCI) and Alzheimer's disease (AD), with an increased trend across NC, MCI, and AD. It was concluded that the proposed algorithm could be effectively used in computing brain aging and PAD, and offering potential for early diagnosis and assessment of normal brain aging and AD.

Abstract:Over the years, Machine Learning models have been successfully employed on neuroimaging data for accurately predicting brain age. Deviations from the healthy brain aging pattern are associated with the accelerated brain aging and brain abnormalities. Hence, efficient and accurate diagnosis techniques are required to elicit accurate brain age estimations. Several contributions have been reported in the past for this purpose, resorting to different data-driven modeling methods. Recently, deep neural networks (also referred to as deep learning ) have become prevalent in manifold neuroimaging studies, including brain age estimation. In this review, we offer a comprehensive analysis of the literature related to the adoption of deep learning for brain age estimation with neuroimaging data. We detail and analyze different deep learning architectures used for this application, pausing at research works published to date quantitatively exploring their application. We also examine different brain age estimation frameworks, comparatively exposing their advantages and weaknesses. Finally, the review concludes with an outlook towards future directions that should be followed by prospective studies. The ultimate goal of this paper is to establish a common and informed reference for newcomers and experienced researchers willing to approach brain age estimation by using deep learning models.

Abstract:Aging is associated with structural brain changes, cognitive decline, and neurodegenerative diseases. Brain age, an imaging biomarker sensitive to deviations from healthy aging, offers insights into structural aging variations and is a potential prognostic biomarker in neurodegenerative conditions. This study introduces BrainAgeNeXt, a novel convolutional neural network inspired by the MedNeXt framework, designed to predict brain age from T1-weighted magnetic resonance imaging (MRI) scans. BrainAgeNeXt was trained and validated on 11,574 MRI scans from 33 private and publicly available datasets of healthy volunteers, aged 5 to 95 years, imaged with 3T and 7T MRI. Performance was compared against three state-of-the-art brain age prediction methods. BrainAgeNeXt achieved a mean absolute error (MAE) of 2.78 years, lower than the compared methods (MAE = 3.55, 3.59, and 4.16 years, respectively). We tested all methods also across different levels of image quality, and BrainAgeNeXt performed well even with motion artifacts and less common 7T MRI data. In three longitudinal multiple sclerosis (MS) cohorts (273 individuals), brain age was, on average, 4.21 years greater than chronological age. Longitudinal analysis indicated that brain age increased by 1.15 years per chronological year in individuals with MS (95% CI = [1.05, 1.26]). Moreover, in early MS, individuals with worsening disability had a higher annual increase in brain age compared to those with stable clinical assessments (1.24 vs. 0.75, p < 0.01). These findings suggest that brain age is a promising prognostic biomarker for MS progression and potentially a valuable endpoint for clinical trials.

Abstract:Measuring differences between an individual's age and biological age with biological information from the brain have the potential to provide biomarkers of clinically relevant neurological syndromes that arise later in human life. To explore the effect of multimodal brain magnetic resonance imaging (MRI) features on the prediction of brain age, we investigated how multimodal brain imaging data improved age prediction from more imaging features of structural or functional MRI data by using partial least squares regression (PLSR) and longevity data sets (age 6–85 years). First, we found that the age-predicted values for each of these ten features ranged from high to low: cortical thickness ( R = 0.866, MAE = 7.904), all seven MRI features ( R = 0.8594, MAE = 8.24), four features in structural MRI ( R = 0.8591, MAE = 8.24), fALFF ( R = 0.853, MAE = 8.1918), gray matter volume ( R = 0.8324, MAE = 8.931), three rs-fMRI feature ( R = 0.7959, MAE = 9.744), mean curvature ( R = 0.7784, MAE = 10.232), ReHo ( R = 0.7833, MAE = 10.122), ALFF ( R = 0.7517, MAE = 10.844), and surface area ( R = 0.719, MAE = 11.33). In addition, the significance of the volume and size of brain MRI data in predicting age was also studied. Second, our results suggest that all multimodal imaging features, except cortical thickness, improve brain-based age prediction. Third, we found that the left hemisphere contributed more to the age prediction, that is, the left hemisphere showed a greater weight in the age prediction than the right hemisphere. Finally, we found a nonlinear relationship between the predicted age and the amount of MRI data. Combined with multimodal and lifespan brain data, our approach provides a new perspective for chronological age prediction and contributes to a better understanding of the relationship between brain disorders and aging.
