Abstract:The aim of the study was to develop, optimize and validate an analytical method for the simultaneous detection and quantification of As species (As (III), dimethylarsinate (DMA), monomethylarsonate (MMA) and As (V)) from various water matrices (drinking water, surface water and groundwater) using the technique of high performance liquid chromatography coupled with inductively coupled plasma mass spectrometry (HPLC-ICP-MS). The separation of the species was achieved using an Agilent G3154-65002 precolumn, 4.6 mm x 10 mm, and an Agilent G3154-65001 column, 4.6 mm x 150 mm, at ambient temperature. The mobile phase used was 1M NaH2PO4 and 0.1 mM EDTA-2Na solution at a flow rate of 1 mL/min. Linear regression lines were drawn in the range 10-50 µg/L, the coefficients of determination (R2) obtained for each species were higher than 0.99. The quantification limits (LOQ) varied in the range 1.8 µg/L As(V) to 2.0 µg/L As(III), and in the range 3.9 µg/L DMA to 4.1 µg /L MMA for all studied matrices. These limits allow the quantification of As species in drinking water samples, surface water and groundwater at the trace level.

Abstract:Direct determination of arsenic species in real samples is challenging due to their trace concentration and spectral interferences by coexisting ions. Herein, we proposed an ultrasound-assisted dispersive solid phase microextraction (DSPME) procedure for the analyses of the trace inorganic arsenic. The hydrothermally synthesized cadmium sulfide nanoparticles (CdS NPs) completely adsorbed both arsenic species within 20 s at the initial arsenic concentration of 100 µg L-1. The detection limit (3 S/m) of the proposed method was found to be 0.5 ± 0.2 and 0.8 ± 0.2 ng L-1 for As(III) and As(V), respectively. The accuracy of the method against the systematic and constant errors was confirmed by the analysis of the Standard Reference Material (SRM) (>95% recovery with <5% RSD). The Student's t-test values were found to be less than the critical Student's t value at a 95% confidence level. The method was successfully employed for the determination of arsenic in food samples.

Abstract:Nanoliter high-performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high-performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high-pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high-performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high-performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C-18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9-1.8 mu g/L were obtained with precisions variable in the range of 1.6-4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87-102%).

Abstract:A simple and fast method of low-density solvent based dispersive liquid-liquid microextraction (LDS-DLLME) followed by high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) has been developed for the speciation analysis of organoarsenic and inorganic arsenic in water samples. The low-density solvent (octanol) was given as the organic phase and injected into the aqueous sample (donor phase) with methanol as the disperser. With As (V), As (III), p-APAA, 4-HPAA, ROX and PAA as target species, factors of LDS-DLLME including pH value, anionic carriers, elution conditions and extractant, were studied in detail. Besides, volumes of solvents were further optimised by response surface methodology. Under the optimal conditions, the limits of detection for four phenylarsenics and arsenate were in the range of 0.001–0.039 μg L−1. The relative standard deviations (RSDs) were 3.6–9.4% and the enrichment factors varied from 6.2 to 70.8. The proposed method of LDS-DLLME-HPLC-ICP-MS was satisfactorily applied to the determination of six arsenic compounds in water samples with recoveries of 81.8–111.7% for the spiked lake water samples.

Abstract:The accurate determination of arsenic species in waters is still an environmental challenge because of the rapid redox transformation of As. So, a procedure for on-site separation of As(III), As(V), methylarsonic acid, and dimethylarsinic acid in natural waters has been developed. The procedure involves passing a water sample through tandem cation- and anion-exchange resin columns in the field. Selective retention of arsenic species occurs in resin cartridges followed by selective elution. The eluate is collected and analyzed by inductively coupled plasma mass spectrometry. The standard reference material CASS-5 was diluted 10 times and analyzed directly to validate accuracy of measurements, and SLRS-2 was used to validate on-site separation procedure, respectively. The results agreed well with the certified values of standard reference materials. The proposed procedure was applied to the on-site separation of arsenic species from natural waters.

Abstract:The toxic and carcinogenic properties of inorganic and organic arsenic species make their determination in natural water vitally important. Determination of individual inorganic and organic arsenic species is critical because the toxicology, mobility, and adsorptivity vary substantially. Several methods for the speciation of arsenic in groundwater, surface-water, and acid mine drainage sample matrices using field and laboratory techniques are presented. The methods provide quantitative determination of arsenite [As(III)], arsenate [As(V)], monomethylarsonate (MMA), dimethylarsinate (DMA), and roxarsone in 2-8 min at detection limits of less than 1 microg arsenic per liter (microg As L(-1)). All the methods use anion exchange chromatography to separate the arsenic species and inductively coupled plasma-mass spectrometry as an arsenic-specific detector. Different methods were needed because some sample matrices did not have all arsenic species present or were incompatible with particular high-performance liquid chromatography (HPLC) mobile phases. The bias and variability of the methods were evaluated using total arsenic, As(III), As(V), DMA, and MMA results from more than 100 surface-water, groundwater, and acid mine drainage samples, and reference materials. Concentrations in test samples were as much as 13,000 microg As L(-1) for As(III) and 3700 microg As L(-1) for As(V). Methylated arsenic species were less than 100 microg As L(-1) and were found only in certain surface-water samples, and roxarsone was not detected in any of the water samples tested. The distribution of inorganic arsenic species in the test samples ranged from 0% to 90% As(III). Laboratory-speciation method variability for As(III), As(V), MMA, and DMA in reagent water at 0.5 microg As L(-1) was 8-13% (n=7). Field-speciation method variability for As(III) and As(V) at 1 microg As L(-1) in reagent water was 3-4% (n=3).

Abstract:The determination of four arsenic species (AsIII, dimethylarsinic acid, monomethylarsonic acid and AsV) in soil and sediments following a single microwave extraction procedure was investigated using an on-line system involving high-performance liquid chromatography (HPLC) coupled with inductively coupled plasma mass spectrometry (ICP-MS). Phosphoric acid was used in conjunction with an open focused microwave system to extract arsenic compounds. This system was optimised with respect to acid concentration, microwave power and time in order to obtain the maximum rate of recovery whilst retaining the integrity of arsenic species. Using an anion-exchange column and buffered phosphate solution with methanol added as the mobile phase, good separation and sensitivity were achieved. Under these conditions, recoveries between 60 and 80% of the total As content were obtained, and the detection limits were in the range 1–2 mg kg–1 for all species. The HPLC–ICP-MS system was used for the determination of arsenic species in acid extracts of in-house reference materials (soil and sediment samples from polluted areas). Only arsenate was found in soil but arsenite was the main species found in a polluted river sediment. Three certified reference materials were analysed to determine the concentrations of the four arsenic species, and the sum of the arsenic concentration was compared with the certified total arsenic value for each of the reference materials. This method allows the speciation of arsenic species in soil and sediment samples to be determined at natural levels, and enables their behaviour in the environment to be monitored.

Abstract:A new method for the speciation analysis of arsenic in food using narrow-bore high-performance liquid-chromatography inductively coupled plasma mass spectrometry (HPLC-ICP-MS) has been developed. Fast separation of arsenite, arsenate, monomethylarsonic acid and dimethylarsinic acid was carried out in 7 min using an anion-exchange narrow-bore Nucleosil 100 SB column and 12 mM ammonium dihydrogen phosphate of pH 5.2 as the mobile phase, at a flow rate of 0.3 mL min(-1). A PFA-ST micronebulizer jointed to a cyclonic spray chamber was used for HPLC-ICP-MS coupling. Compared with standard-bore HPLC-ICP-MS, the new method has provided higher sensitivity, reduced mobile-phase consumption, a lower matrix plasma load and a shorter analysis time. The achieved instrumental limits of detection were in the 0.3 - 0.4 ng As mL(-1) range, and the precision was better than 3%. The arsenic compounds were efficiently (>80%) extracted from various food samples using a 1:5 methanol/water solution, with additional ultrasonic treatment for rice products. The applicability of this method was demonstrated by the analysis of several samples, such as seafood (fish, mussels, shrimps, edible algae) and rice-based products (Jasmine and Arborio rice, spaghetti, flour, crackers), including three certified reference materials.

Abstract:<p>Arsenic (As) is a ubiquitous element in the environment while its toxicity and chemical properties are highly associated with its chemical form. High quality As speciation data in environmental matrices are thus desirable for understanding its biogeochemistry. Here we present a simple and novel head space-single drop microextraction-spectro-pipette (HS-SDME-SP) method for the on-site measurement of As species in natural waters. Arsine was generated and separated from the sample matrix by borohydride-based hydride generation, which was further extracted into a stable, hanging 5 μL microdroplet on the tip of a spectro-pipette. With the spectro-pipette, the microvolume of extractant is easily handled while gas–liquid absorption, redox and chromogenic reactions and <em>in situ</em> absorbance measurement can be carried out in a single step. The speciation analysis of As(III) and As(V) was achieved by utilizing their different pH requirement for reduction and arsine generation in the hydride generation process. Although the proposed method is a simple sensor based assay, it is sensitive, robust and reliable; the limit of detection (3 × S/N) was 0.1 nM, RSDs for 25 nM As(III) were < 2.0% (n = 10) and results were comparable with the commonly used hydride generation atomic fluorescence spectrometry (HG-AFS) method and in good agreement with two certified reference materials (surface freshwater and seawater). The proposed method is easy to operate, the device is light and portable (1.1 kg) and is practical for the measurement of trace inorganic As species in the field.</p>

Abstract:Arsenic is a well-known carcinogenic, mutagenic and toxic element and occurs in the environment both as inorganic arsenic (iAs) and organoarsenical compounds (oAsCs). Since the toxicity of arsenic compounds depends on their chemical form, the identification and determination of arsenic species are essential. Recently, the European Food Safety Authority, following the European Commission request, published a report on chronic dietary exposure to iAs and recommended the development and validation of analytical methods with adequate sensitivity and refined extraction procedures for this determination. Moreover, the authority called upon new arsenic speciation data for complex food matrices such as seaweeds, grains and grain-based products. Looking at this context, an optimized, sensitive and fast analytical method using high performance liquid chromatography followed by inductively coupled plasma—mass spectrometry (HPLC/ICP-MS) was developed for the determination of iAs (sum of arsenite—AsIII and arsenate—AsV) and the most relevant oAsCs, arsenobetaine, dimethylarsinic acid and monomethylarsonic acid. The method was validated with satisfactory results in terms of linearity, sensitivity, selectivity, precision, recovery, uncertainty, ruggedness and matrix effect, and then successfully applied for the analysis of several matrices, i.e., processed and unprocessed cereal and cereal products, fruits, vegetables, legumes, seaweeds, nuts and seeds. The results obtained indicate that not only seaweed and rice matrices but also many cereals, legumes and plant-based foods for infants and young children contain significant concentrations of iAs and oAsCs. These findings contribute to the data collection necessary to assess the role of these matrices in the total arsenic exposure and if specific maximum limits have to be established.

Abstract:The current paper describes a method for determining As(III) and As(V) concentrations in wastewater samples. The approach is based on the extraction of As(III) from water samples using a non-polar organic solvent in a 10-12 M HCl medium. The extract contains As(III), while the raffinate contains As(V). Distilled water can be used to re-extract As(III) from the organic solvent. ICP-MS was used to analyze As concentrations in the reextract and the raffinate. The average recovery of arsenic by the proposed method is 99.6 %. It was calculated as the sum of As(III) and As(V) concentrations. The collected data demonstrated high repeatability. The standard addition procedure demonstrated the selectivity and accuracy of the employed method. The relative error of recovery of the standard As(III) addition is about 2 %, which is similar to the RSD found under repeatability conditions.
Fast determination of arsenic species and total arsenic in urine by HPLC-ICP-MS: concentration ranges for unexposed german inhabitants and clinical case studies.

Abstract:A fast and reliable high-pressure liquid chromatography (HPLC)-inductively coupled plasma-mass spectrometry (ICP-MS) routine method was developed for the determination of inorganic arsenic [As(III) and As(V)], organic monomethylarsonate [MMA(V)], dimethylarsinate [DMA(V)], and arsenobetaine (As-B) in human urine. The complete method validation is described, including internal and external quality assurance. Limits of quantification for the As species are 0.1 microg/L, which is sufficient to determine background concentrations of the arsenic species in human urine. Additionally, total As in all urine samples was determined by conventional ICP-MS. Mean concentrations for 82 non-exposed inhabitants from northern Germany are 12.7, 5.9, 4.0, 0.23, 0.52, and 0.17 microg/L for total As, As-B, DMA(V), As(III), MMA(V), and As(V), respectively. Approximately 15% of the total As was not identified by the anion exchange HPLC-ICP-MS method, and could be other As metabolites in urine. Two case studies underline the need for As speciation, especially when total urinary arsenic concentrations are elevated. In the first case, we investigated the effect of seafood consumption on the concentration of different arsenic species in urine for different persons. A maximum enhancement of total As from 1 up to 2,200 microg/L (2,000 microg/L for As-B) was observed after a normal fish meal. The second case describes the exposure of a 7-year-old child to As(III) by inhalation of calcium arsenite powder. Five hours after exposure, the concentrations in the child's urine for As-B, DMA(V), As(III), MMA(V), and As(V) were < 0.1, 189, 304, 229, and 27 microg/L, respectively, and these concentrations were reduced to normal background values after 4 days.

Abstract:It has been challenging to analyze inorganic arsenic (iAs) with anion exchange HPLC-Electrospray Ionization-Mass spectrometry (HPLC-ESI-MS), because arsenite (As(III)) is difficult to retain on column and the salts in mobile phase causes ionization suppression of iAs. To address these issues, a method has been developed involving the determination of arsenate (As(V)) with mixed mode HPLC-ESI-MS and the conversion of As(III) to As(V) for total iAs. As(V) was separated from other chemicals on Newcrom B, a bi-modal HPLC column involving anion exchange and reverse phase interaction. The elution employed a two-dimensional gradient, including a formic acid gradient to elute As(V) and a concurrent alcohol gradient to elute organic anions used in sample preparations. As(V) was detected by Selected Ion Recording (SIR) in negative mode at m/z = 141 with a QDa (single quad) detector. As(III) was quantitatively converted to As(V) by mCPBA oxidation and measured for total iAs. By replacing salt with formic acid in elution, the ionization efficiency for As(V) was greatly enhanced in ESI interface. The limit of detection (LOD) for As(V) and As(III) were 0.0263 μM (1.97 ppb) and 0.0398 μM (2.99 ppb), respectively. The linear range was 0.05-1 μM. The method has been used to characterize iAs speciation change in the solution and precipitation in a simulated iron-rich groundwater caused by air exposure.

Abstract:A method is presented for the determination of inorganic arsenic species by hydride generation high resolution inductively coupled plasma mass spectrometry. In the presence of 1.5mg/ml of L-cysteine, arsenic(V) is indirectly determined using the difference between As(III) concentrations determined after 2 and 8min of reaction time, whereas the As(III) concentration is obtained by taking the difference between the concentration of arsenic(III) determined after 8min and the concentration of arsenic(III) converted from arsenic(V) after 8min. 25 fold amounts of arsenic(III) and 100 fold amounts of arsenic(V) hardly affected the determination of 1ng/ml arsenic(V) and 100ng/l arsenic(III), respectively. Interferences from transition metal ions were removed by using a Chelex 100 resin. The detection limits for arsenic(III) and arsenic(V) were 11.8 and 39.7ng/l, respectively. The proposed method for the simultaneous determination of inorganic arsenic species was applied to the determination of As(III) and As(V) in spiked and Japanese standard river water samples. Agreement within statistical uncertainty was obtained in all instances.
Dual-column capillary microextraction (CME) combined with electrothermal vaporization inductively coupled plasma mass spectrometry (ETV-ICP-MS) for the speciation of arsenic in human hair extracts

Abstract:In this work, dual-column capillary microextraction (CME) system consisting of N-(2-aminoethyl)-3-aminopropyltrimethoxysilane (AAPTS)-silica coated capillary (C1) and 3-mercaptopropyl trimethoxysilane (MPTS)-silica coated capillary (C2) was developed for sequential separation/preconcentration of arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid [MMA(V)] and dimethylarsinic acid [DMA(V)] in the extracts of human hair followed by electrothermal vaporization inductively coupled plasma mass spectrometry (ETV-ICP-MS) detection with iridium as permanent modifier. Various experimental parameters affecting the dual-column microextraction of different As species had been investigated in detail. It was found that at pH 9, As(V) and MMA could be quantitatively retained by C1 and only As(III) could be quantitatively retained by C2. With the aid of valve switching, As(V)/MMA(V) retained on C1 and As(III) retained on C2 could be sequentially desorbed by 10 microl of 0.01 mol l(-1) HNO(3) [for As(V)], 0.1 mol l(-1) HNO(3) [for MMA(V)] and 0.2 mol l(-1) HNO(3)-3% thiourea (m/v) [for As(III)], respectively, the eluents were immediately introduced into the Ir-coated graphite tubes for further ETV-ICP-MS detection. With two-step ETV pyrolysis program, Cl(-) in the sample matrix could be in situ removed, and the total As in the human hair extracts or digested solution could be interference-free, determined by ETV-ICP-MS. DMA(V) in the human hair extracts was obtained by subtraction of total As in the human hair extracts from other three As species. Under the optimized conditions, the detection limits (3 sigma) of the method were 3.9 pg ml(-1) for As(III), 2.7 pg ml(-1) for As(V), 2.6 pg ml(-1) for MMA(V) and 124 pg ml(-1) for total As with the relative standard deviations less than 7.0% (C = 0.1 ng ml(-1), n = 7), and the enrichment factor was 286, 262 and 260 for As(III), As(V) and MMA(V), respectively. The developed method was successfully applied for the speciation of arsenic in the extracts of human hair.

Inductively coupled plasma mass spectrometry with dynamic reaction cell (ICP-DRC-MS) and high-performance liquid chromatography hyphenated to ICP-DRC-MS were used for TAs and arsenic species determination, respectively. The DRC with oxygen as a reaction gas was used. Sample preparation, digestion, and extraction were optimized. Microwave assisted digestion and extraction provided good recovery and extraction efficiency. Arsenic species were fully separated in 8 min using 10 mmol L−1 of ammonium dihydrogen phosphate and 10 mmol L−1 of ammonium nitrate. Overlapping of AsB and As(III) of arsenic species in the presence of a high concentration of AsB and trace amounts of As(III) were studied. Detailed validation of analytical procedures proved the reliability of analytical measurements. Both procedures were characterized by short-term and long-term precision: 2.2% (TAs) up to 4.2% (AsB), and 3.6% (TAs) up to 7.2% (DMA), respectively. Limits of detection (LD) were in the range from 0.056 μg L−1 for TAs to 0.15 μg L−1 for As(V). Obtained recoveries were in the range of 85%–116%. Developed methods were applied to freshwater fish samples analysis.
