Abstract:A simple and fast (<5 s) method for in situ arsenic speciation on solid surfaces has been developed based on desorption electrospray ionization-tandem mass spectrometry (DESI-MS). Arsenic-polluted environmental samples such as animal feed and plant tissues could be directly monitored by DESI-MS. Each arsenic species in this study, including monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenobetaine (AsB), arsenocholine (AsC), 4-arsanilic acid (p-ASA), 4-hydroxyphenylarsonic acid (4-OH), Nitarsone, Roxarsone and two inorganic arsenic species, arsenate As(v) and arsenite As(iii), could be detected by their typical m/z and collision induced dissociation (CID) behavior respectively. By the characteristic information, mixtures of different arsenic species could be detected without any sample preparation and separation process. This method could give absolute detection limits of the arsenic species at ng/mm(2) to pg/mm(2) level with a best RSD of 5.3% (n = 5). The method could be potentially applied to in situ environmental monitoring of arsenic pollution, especially that caused by arsenic pesticides, animal feed additives, herbicides and wood treatment.

Abstract:The aim of the study was to develop, optimize and validate an analytical method for the simultaneous detection and quantification of As species (As (III), dimethylarsinate (DMA), monomethylarsonate (MMA) and As (V)) from various water matrices (drinking water, surface water and groundwater) using the technique of high performance liquid chromatography coupled with inductively coupled plasma mass spectrometry (HPLC-ICP-MS). The separation of the species was achieved using an Agilent G3154-65002 precolumn, 4.6 mm x 10 mm, and an Agilent G3154-65001 column, 4.6 mm x 150 mm, at ambient temperature. The mobile phase used was 1M NaH2PO4 and 0.1 mM EDTA-2Na solution at a flow rate of 1 mL/min. Linear regression lines were drawn in the range 10-50 µg/L, the coefficients of determination (R2) obtained for each species were higher than 0.99. The quantification limits (LOQ) varied in the range 1.8 µg/L As(V) to 2.0 µg/L As(III), and in the range 3.9 µg/L DMA to 4.1 µg /L MMA for all studied matrices. These limits allow the quantification of As species in drinking water samples, surface water and groundwater at the trace level.

Abstract:It is known that arsenic has different toxicological properties dependent upon both its oxidation state for inorganic compounds, as well as the different toxicity levels exhibited for organic arsenic compounds. The field of arsenic speciation analysis has grown rapidly in the recent years, especially with the utilization of chromatographic analysis. Extraction method as sequential extraction combined to microwave assisted digestion were applied to water and sediments samples. This paper describes the essential background and toxicity of arsenic in environment, and more importantly, some currently used chromatographic applications and sample handling procedures necessary to accurately detect arsenic in its various forms. Experiments and work using ultra high-performance liquid chromatography and inductively coupled plasma optical emission spectrometry for arsenic speciation of environmental samples are presented in this research.

Abstract:As a redox-sensitive and multi-species trace element, the chemical behavior of arsenic in various biological sources was closely related to human health. Direct and sensitive speciation analysis of arsenic from complex matrices is ideally required since it enables obtaining of multidimensional information concerning arsenic concentration, existing forms, redox states and molecular structures without sample pretreatment. Herein, an online speciation analysis method allowing molecular characterization of both inorganic (As(III), As(V)) and organic arsenic species (MMA, DMA, AsB and AsC) at trace level from original bio-matrices (e.g. crayfish, laver and kelp) was developed based on ambient ionization mass spectrometry, namely extraction electrospray ionization-hydride generation-microwave plasma torch-mass spectrometry (EESI-HG-MPT-MS). The novel ambient ionization source of MPT was combined with a hydride generator, resulting in a unique mass spectrum behavior for As. By using a home-made sample introduction device and incorporating HG-MPT, the sensitivity of analytes and matrix tolerance was significantly improved. The detection limits of As(III), As(V), MMA, DMA, AsB and AsC were 0.01, 0.02, 0.08, 0.08, 0.05 and 0.02 μg/kg respectively. Online spike experiment was also conducted, showing the recovery of 94-111%, with an RSD less than 10%. The developed method may play a role in studying the chemical or biological behaviors of arsenic in various environment.

Abstract:Multi-elemental speciation analysis in a single run reduces costs, waste and time, and also sample consumption, which is favorable to a large set of instable samples. This work demonstrates mercury and arsenic speciation in a single run using high performance liquid chromatography with inductively coupled plasma mass spectrometric detection (HPLC/ICP-MS). Thiol compounds in mobile phases interfered with the elution of arsenite because of coordination action, which was overcome by gradient elution (elution of As(III) by thiol-free mobile phases at first). Four arsenic forms (As(III), DMA, MMA and As(V)) and three mercury species (Hg(II), MeHg and EtHg) were separated and detected by HPLC/ICP-MS using 4 mM tetrabutylammonium hydroxide (TBAH) (pH 6.0) in the first 1.0 min and 4 mM TBAH + 20 mM L-cysteine (pH 6.0) in the next 9.0 min. The detection limits were obtained at 0.078 for As(III), 0.081 for DMA, 0.078 for MMA and 0.15 mu g L-1 for As(V), and 0.016 for Hg(II), 0.027 for MeHg and 0.032 mu g L-1 for EtHg, and the reproducibilities (50 mu g L-1 for each As-species and 10 mu g L-1 for each Hg-species) were all lower than 3.7%. The proposed method was successfully applied for speciation analyses of arsenic and mercury in five lotus roots, which indicated DMA (3.5-7.9 mu g kg(-1)) as the only arsenic species and MeHg (0.6-1.0 mu g kg(-1)) as the only mercury species. Analyses of two certified reference materials demonstrated good agreement between determined values and certified values. All results prove its advantages including high sensitivity and high analytical efficiency, which are beneficial for routine speciation analyses of arsenic and mercury in a single run in environmental and food fields.

Abstract:The development of analytical techniques suitable for providing structural information on a wide range of elemental species is a growing necessity. For arsenic speciation a variety of mass spectrometric techniques, mainly inductively coupled plasma mass spectrometry (ICP-MS) and electrospray tandem mass spectrometry (ES-MS/MS) coupled on-line with high-performance liquid chromatography (HPLC), are in use. In this paper we report the identification of arsenic species present in samples of marine origin (seaweed extracts) using ES ion trap mass spectrometry (IT) multistage mass spectrometry (MS(n)). Both reversed-phase and anion-exchange HPLC have been coupled on-line to ES-ITMS. Product ion scans with multiple stages of tandem MS (MS(n); n=2-4) were used to acquire diagnostic data for each arsenosugar. The spectra contain structurally characteristic fragment ions for each of the arsenosugars examined. In addition it was observed that upon successive stages of collision-induced dissociation (CID) a common product ion (m/z 237) was formed from all four arsenosugars examined. This product ion has the potential to be used as an indicator for the presence of dimethylated arsenosugars (dimethylarsinoylribosides). The HPLC/ES-ITMS(n) method developed allows the sensitive identification of arsenosugars present in crude seaweed extracts without the need for extended sample preparation. In fact, sample preparation requirements are identical to those typically employed for HPLC/ICP-MS analysis. Additionally, the resulting product ions are structurally diagnostic of the arsenosugars examined, and tandem mass spectra are reproducible and correspond well to those obtained using other low-energy CID techniques. As a result, the HPLC/ES-ITMS(n) approach minimises the potential for arsenic species misidentification and has great potential as a means of overcoming the need for characterised standards.

Abstract:<p>Arsenic (As) is a ubiquitous element in the environment while its toxicity and chemical properties are highly associated with its chemical form. High quality As speciation data in environmental matrices are thus desirable for understanding its biogeochemistry. Here we present a simple and novel head space-single drop microextraction-spectro-pipette (HS-SDME-SP) method for the on-site measurement of As species in natural waters. Arsine was generated and separated from the sample matrix by borohydride-based hydride generation, which was further extracted into a stable, hanging 5 μL microdroplet on the tip of a spectro-pipette. With the spectro-pipette, the microvolume of extractant is easily handled while gas–liquid absorption, redox and chromogenic reactions and <em>in situ</em> absorbance measurement can be carried out in a single step. The speciation analysis of As(III) and As(V) was achieved by utilizing their different pH requirement for reduction and arsine generation in the hydride generation process. Although the proposed method is a simple sensor based assay, it is sensitive, robust and reliable; the limit of detection (3 × S/N) was 0.1 nM, RSDs for 25 nM As(III) were < 2.0% (n = 10) and results were comparable with the commonly used hydride generation atomic fluorescence spectrometry (HG-AFS) method and in good agreement with two certified reference materials (surface freshwater and seawater). The proposed method is easy to operate, the device is light and portable (1.1 kg) and is practical for the measurement of trace inorganic As species in the field.</p>

Abstract:Nanoliter high-performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high-performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high-pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high-performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high-performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C-18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9-1.8 mu g/L were obtained with precisions variable in the range of 1.6-4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87-102%).

Abstract:It has been challenging to analyze inorganic arsenic (iAs) with anion exchange HPLC-Electrospray Ionization-Mass spectrometry (HPLC-ESI-MS), because arsenite (As(III)) is difficult to retain on column and the salts in mobile phase causes ionization suppression of iAs. To address these issues, a method has been developed involving the determination of arsenate (As(V)) with mixed mode HPLC-ESI-MS and the conversion of As(III) to As(V) for total iAs. As(V) was separated from other chemicals on Newcrom B, a bi-modal HPLC column involving anion exchange and reverse phase interaction. The elution employed a two-dimensional gradient, including a formic acid gradient to elute As(V) and a concurrent alcohol gradient to elute organic anions used in sample preparations. As(V) was detected by Selected Ion Recording (SIR) in negative mode at m/z = 141 with a QDa (single quad) detector. As(III) was quantitatively converted to As(V) by mCPBA oxidation and measured for total iAs. By replacing salt with formic acid in elution, the ionization efficiency for As(V) was greatly enhanced in ESI interface. The limit of detection (LOD) for As(V) and As(III) were 0.0263 μM (1.97 ppb) and 0.0398 μM (2.99 ppb), respectively. The linear range was 0.05-1 μM. The method has been used to characterize iAs speciation change in the solution and precipitation in a simulated iron-rich groundwater caused by air exposure.
Feasibility of Identification and Monitoring of Arsenic Species in Soil and Sediment Samples by Coupled High-performance Liquid Chromatography — Inductively Coupled Plasma Mass Spectrometry

Abstract:The determination of four arsenic species (AsIII, dimethylarsinic acid, monomethylarsonic acid and AsV) in soil and sediments following a single microwave extraction procedure was investigated using an on-line system involving high-performance liquid chromatography (HPLC) coupled with inductively coupled plasma mass spectrometry (ICP-MS). Phosphoric acid was used in conjunction with an open focused microwave system to extract arsenic compounds. This system was optimised with respect to acid concentration, microwave power and time in order to obtain the maximum rate of recovery whilst retaining the integrity of arsenic species. Using an anion-exchange column and buffered phosphate solution with methanol added as the mobile phase, good separation and sensitivity were achieved. Under these conditions, recoveries between 60 and 80% of the total As content were obtained, and the detection limits were in the range 1–2 mg kg–1 for all species. The HPLC–ICP-MS system was used for the determination of arsenic species in acid extracts of in-house reference materials (soil and sediment samples from polluted areas). Only arsenate was found in soil but arsenite was the main species found in a polluted river sediment. Three certified reference materials were analysed to determine the concentrations of the four arsenic species, and the sum of the arsenic concentration was compared with the certified total arsenic value for each of the reference materials. This method allows the speciation of arsenic species in soil and sediment samples to be determined at natural levels, and enables their behaviour in the environment to be monitored.

Abstract:A method is presented for arsenic speciation analysis of an oyster sample using ion chromatography coupled with an inductively coupled plasma mass spectrometry (ICP-MS) instrument. A strong anion exchange resin was employed with a step gradient elution of 0.1 mM/0.1 M K 2 SO 4 at pH 10.2. Arsenobetaine and dimethylarsinic acid were determined following extraction based on trypsin enzymolysis with 95-100% extraction efficiency. Limits of detection in the range 0.1-0.3 mg kg m 1 of arsenic were obtained for organic arsenic species. No inorganic arsenic was detected. Validation was performed using TORT-2 as a certified reference material. Although high performance liquid chromatography (HPLC) coupled to ICP-MS is an effective method for speciation analysis it is not always necessary to obtain such a detailed picture. A simple liquid chromatographic separation technique based upon mini-column technology is presented. It was developed to obtain a fast, efficient and reliable separation of inorganic from organic, i.e. assumed toxic from non-toxic, arsenic and selenium species suitable for use as an initial screening method for environmental analysis. Two types of strong anion exchange resin were tested. Excellent separation was obtained for both min-column resins and analysis times were within 7 min. Limits of detection obtained for inorganic arsenic, organic arsenic, selenomethionine, Se IV and Se VI were 1.6, 1.8, 66, 32 and 22 µg kg m 1 , respectively.

Abstract:A simple and fast method of low-density solvent based dispersive liquid-liquid microextraction (LDS-DLLME) followed by high-performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) has been developed for the speciation analysis of organoarsenic and inorganic arsenic in water samples. The low-density solvent (octanol) was given as the organic phase and injected into the aqueous sample (donor phase) with methanol as the disperser. With As (V), As (III), p-APAA, 4-HPAA, ROX and PAA as target species, factors of LDS-DLLME including pH value, anionic carriers, elution conditions and extractant, were studied in detail. Besides, volumes of solvents were further optimised by response surface methodology. Under the optimal conditions, the limits of detection for four phenylarsenics and arsenate were in the range of 0.001–0.039 μg L−1. The relative standard deviations (RSDs) were 3.6–9.4% and the enrichment factors varied from 6.2 to 70.8. The proposed method of LDS-DLLME-HPLC-ICP-MS was satisfactorily applied to the determination of six arsenic compounds in water samples with recoveries of 81.8–111.7% for the spiked lake water samples.

[Critical assessment of microbiological diagnostics in periodontal diseases with special focus on Porphyromonas gingivalis].

Abstract:Periodontitis is caused by an opportunistic infection with pathogenic microorganisms of the oral biofilm. In this paper, we discuss the usefulness of microbial diagnostics with respect to the differential diagnosis or the treatment approaches of periodontal diseases. Several diagnostic techniques, based on morphological, enzymatic, cultural, genetic or antigenetic properties have been established to analyze the microbial flora. Among the bacterial species some virulent genotypes of P. gingivalis play an important role in the etiology of periodontitis. Expression of fimbriae or different proteases have been identified as potential virulence factors of this gram negative anaerobic rod. To date a characterization of virulence of specific strains or a correlation between expression of different virulence factors and distinct periodontal conditions, however, is missing. Therefore, the importance of a routine identification of P. gingivalis still needs further evaluation.

Abstract:The accurate determination of arsenic species in waters is still an environmental challenge because of the rapid redox transformation of As. So, a procedure for on-site separation of As(III), As(V), methylarsonic acid, and dimethylarsinic acid in natural waters has been developed. The procedure involves passing a water sample through tandem cation- and anion-exchange resin columns in the field. Selective retention of arsenic species occurs in resin cartridges followed by selective elution. The eluate is collected and analyzed by inductively coupled plasma mass spectrometry. The standard reference material CASS-5 was diluted 10 times and analyzed directly to validate accuracy of measurements, and SLRS-2 was used to validate on-site separation procedure, respectively. The results agreed well with the certified values of standard reference materials. The proposed procedure was applied to the on-site separation of arsenic species from natural waters.

Abstract:Three common Chinese edible seaweeds, one brown (Laminaria japonica) and two red (Porphyra crispata and Eucheuma denticulatum), were examined for their total arsenic content. The As species were extracted with yields of 76.4, 69.8 and 25.0%, respectively. Anion-exchange and cation-exchange high-performance liquid chromatography (HPLC) in combination with inductively coupled plasma mass spectrometry (ICP-MS) were used for the separation of the different arsenic species in two of the three seaweed extracts (Laminaria and Porphyra). The main arsenic species in the algal extracts are arseno sugars, although it has been shown that the Laminaria seaweed contains significant amounts of dimethylarsinic acid (DMA). HPLC was coupled with electrospray mass spectrometry (ES-MS) for structural confirmation of the arsenic species. The mass spectrometer settings for the arseno sugars were optimised using standards. The conclusions drawn on the basis of HPLC-ICP-MS were confirmed by the HPLC-ES-MS data. The HPLC-ES-MS method is capable of determining both arseno sugars and DMA in the seaweeds. The unknown compounds seen in the HPLC-ICP-MS chromatogram of Laminaria could not be ascribed to trimethylarsenic oxide or tetramethylarsonium ion.
