Abstract:Four dairy cows in a positive energy balance were used to study the effect of propylene glycol supplementation variants on blood indices. The treatments were: without glycol, glycol mixed with the total mixed ration (TMR), glycol delivered as top dressing (TD), and glycol administered as an oral drench (OD). Oral drenching had the positive effect of higher blood insulin concentration 1.5 and 2.5 h after supplementation compared to the TD method ( P ≤ 0.05). Glycol supplemented as TD had the negative effect of higher concentration of non-esterified fatty acids (NEFA) 3.5 h after supplementation ( P ≤ 0.05) with a trend towards a higher mean concentration of this marker ( P ≤ 0.1). A decreasing tendency was recorded for the NEFA concentration 1.5 h after glycol supplementation with TMR or OD ( P ≤ 0.1). Glycol supplemented as OD had the positive effect of lowering the β-hydroxybutyric acid concentration 1.5 and 2.5 h after feeding ( P ≤ 0.05). A downward trend in the mean β-hydroxybutyric acid level after glycol delivery as OD was also observed ( P ≤ 0.1). Glycol supplemented as TD had the negative effect of decreasing dry matter intake of the diet ( P ≤ 0.05). Glycol supplemented as OD had a more glucogenic effect than as part of the TMR. Due to reduction of dry matter intake and the lowest glucogenic effect, using glycol as TD is unjustified. The results confirm that the best method of propylene glycol administration is OD, which can be useful in the prevention of ketosis, however, it should be confirmed in a long-term experiment covering the transition and early lactation periods.

Efficacy of Different Drenching Regimens of Gluconeogenic Precursors during Transition Period on Body Condition Score, Production, Reproductive Performance, Subclinical Ketosis and Economics of Dairy Cows

Abstract:A total of 108 Holstein cows were used to evaluate the effect of drenching propylene glycol (PG) either individually or in combination with glycerol (G) on body condition score (BCS), production, reproductive performance, protection against subclinical ketosis and economic benefit of dairy cows during the transition period. The six groups (n = 18/group) were; Control group, cows received no treatment; PG300, cows were drenched 300 mL PG for 7 days pre-expected day of calving and 21 days post-calving; PG400, cows were drenched 400 mL PG for 7 days pre-and 7 days post-calving; PG500, cows were drenched 500 mL PG for 10 days pre-and 10 days post-calving; GPG150, cows were drenched a mixture of 150 mL G and 150 mL PG for 21 days pre-and 21 days post-calving; GPG250, cows were drenched a mixture of 250 mL G and 250 mL PG for 21 days pre-and 21 days post-calving. BCS at 30 days in milk (DIM) was significantly higher in all treated groups in comparison to the control one and the best values were observed in GPG250, GPG150 and PG500 groups. Lactation length (LL) was significantly (p < 0.001) shorter in GPG250, GPG150 and PG500 groups than control group. There was a significant increment in 305 milk yield (p < 0.001) and average daily milk yield (p < 0.001) in GPG250 and PG500 groups than other groups except for PG300 with the lowest values for control and PG400. Cows in all treatment groups were observed in heat and conceived earlier as well as had shorter open days and calving interval durations (p < 0.001) and low number of services per conception (p = 0.004) compared to control group with better results for PG500 and GPG250 groups. BHB level and percentage of cows suffered from subclinical ketosis at 7 DIM was significantly lower in GPG250, GPG150 and PG500 groups than control group. Cows in treatment groups had a significantly higher glucose level (p = 0.006) compared to control group. Regarding to economics, the control group showed the highest feed costs, insemination costs and total costs per animal. Additionally, control and PG400 groups had the highest cost per kilogram of milk from total and feed costs compared to other cows. PG300, PG500 and GPG250 groups recorded a greater net return and income over feed cost (IOFC). In conclusion, the continuous drenching of propylene glycol either individually or in combination with glycerol regimens for long duration (PG300, PG500, GPG150, GPG250) during the transition period of dairy cows may reduce the incidence of subclinical ketosis and consequently improve milk production, reproduction and economics compared to drenching for short duration (PG400).
A randomized controlled trial to evaluate propylene glycol alone or in combination with dextrose as a treatment for hyperketonemia in dairy cows

Abstract:Our objective was to investigate the effect of i.v. dextrose as an adjunct therapy to oral propylene glycol on the resolution of hyperketonemia (HYK; blood β-hydroxybutyrate ≥1.2 mmol/L), disease incidence, and early lactation milk yield. Cows (n = 1,249) between 3 and 16 d in milk (DIM) from 4 New York dairy farms were screened once weekly for HYK for 2 wk. Those with HYK and no previous history of retained placenta, metritis, or HYK were randomly assigned to 1 of 3 treatment groups: 300 mL of oral 100% propylene glycol for 3 d (PG3); 300 mL of oral 100% propylene glycol for 3 d plus 500 mL i.v. 50% dextrose on d 1 (PG3D1); or 300 mL of oral 100% propylene glycol for 3 d plus 500 mL i.v. 50% dextrose on all 3 d (PG3D3). Cows with a blood β-hydroxybutyrate <1.2 mmol/L at initial screening were re-screened the following week and randomly assigned to the above treatment groups if blood β-hydroxybutyrate was ≥1.2 mmol/L. Cows were assessed for post-treatment HYK resolution 1 and 2 wk after initial HYK diagnosis. We collected farm-diagnosed occurrence of adverse events (sold, died, metritis, displaced abomasum, or ketosis) during the first 60 DIM and milk yield data from the first 10 wk of lactation from herd management software. We used mixed-effects multivariable Poisson regression models to assess the risk of post-treatment HYK resolution at 1 and 2 wk following initial HYK diagnosis and adverse event occurrence among treated cows. We used repeated-measures ANOVA to assess differences in average daily milk yield between treatments. The overall HYK incidence was 30.1% (n = 373). Sixty-four percent of cows (n = 237) were assigned to a treatment group in the first week (3 to 9 DIM), and 36% (n = 136) assigned the second week (10 to 16 DIM). The incidence of 1 or more adverse events during the first 60 DIM was 9.4% (n = 35). We found no effect of treatment on risk of post-treatment HYK resolution at wk 1 (PG3 56.9%, PG3D1 45.0%, PG3D3 50.0%) or wk 2 (PG3 60.0%, PG3D1 52.1%, PG3D3 59.5%) following initial diagnosis, or for risk of adverse event occurrence (PG3 7.4%, PG3D1 8.0%, PG3D3 12.6%). Average daily milk yield (mean ± SE) was similar between treatment groups (PG3: 42.7 ± 0.6 kg/d, PG3D1: 42.4 ± 0.6 kg/d, PG3D3: 42.6 ± 0.6 kg/d). The addition of dextrose for 1 or 3 d provided no improvement in resolution of ketosis assessed once weekly, reduction in adverse events during the first 60 d of lactation, or a difference in average daily milk yield during the first 10 wk of lactation.
Effect of propylene glycol dosage during feed restriction on metabolites in blood of prepartum Holstein heifers

Abstract:Different doses of propylene glycol were compared for lowering plasma NEFA concentration during restricted feed intake. Eight Holstein heifers, averaging 90 d prior to calving at initiation of the trial, were in a 4 x 4 Latin square design with 12-d periods. Heifers consumed alfalfa silage on an ad libitum basis during d 1 to 7 of each period. During d 8 to 12, heifers were gradually restricted to 50% of ad libitum intake. Heifers received an oral drench of 0, 296, 592, or 887 ml of propylene glycol once daily at 6 h prior to feeding on d 8 to 12. Propylene glycol linearly increased glucose and insulin and decreased BHBA and NEFA in blood. Quadratic effects of propylene glycol on plasma glucose, BHBA, and NEFA also occurred; response per milliliter of propylene glycol was greatest at the lowest dose. The highest dose of propylene glycol returned blood glucose, insulin, and NEFA concentrations to those prior to feed restriction. Ruminal acetate to propionate ratio decreased as propylene glycol dose was increased, indicating ruminal conversion of propylene glycol to propionate. A dose of 296 ml of propylene glycol was almost as effective as a dose of 887 ml in reducing lipid mobilization during restricted feed intake.
Effect of Propylene Glycol Supplementation in Cows with Sub-clinical Ketosis

Abstract:The present study was conducted in crossbred dairy cows of Western Rajasthan. Two times sampling were done in each animal pre and post-treatment. The sub-clinical ketosis was diagnosed on the basis of plasma BHBA>1.2 mmol/L and NEFA>0.7 mmol/L during post partum period. Therapeutic trial was conducted using Propylene glycol and choline biocarbonate in sub-clinical ketosis cows. The result revealed increase in the value of glucose, milk yield and decrease NEFA, BHBA levels after propylene glycol supplementation. Whereas, no significant change were recorded in total protein, albumin, PUN, creatinine, AST, ALT and triglyceride. Highly significant increases mean value of glutathione and decrease in malondialdehyde was also observed. Similarly, highly significant decreased GGT value and significant increased cholesterol, calcium level in pre to post-treatment. While NEB is a physiologically normal process, excessive NEB reflects poor adaptation and results in adverse health and production effects after calving.

Abstract:The objective of this study was to evaluate the efficacy of propylene glycol (PG) on prevention and treatment of ketosis in dairy cows from 09/2018 to 03/2019. The study consisted of 2 experiments using a total of 126 multiparous dairy cows in a HF crossbred herd. Cows in both experiments were randomly assigned to treatments in a randomized complete block design. In Exp. 1, 96 cows without ketosis disease (BHBA < 1.4 mmol/L) were assigned to 2 groups (48 cows/group), including (1) untreated control group (no PG) and (2) treatment group (oral PG for 3 consecutive days after parturition). In Exp. 2, 30 ketotic cows (BHBA ≥ 1.4 mmol/L) were divided into 3 groups (10 cows/group), including (1) cows provided with glucose + vitamin B12 + dexamethasone (TT1), (2) cows treated with PG for 3 days (TT2), and (3) combination of TT1 and TT2 (TT3). The results of the Exp. 1 showed that the proportion of ketotic cows was lower (P < 0.01) in the PG-treated group (18.75%) than in the control group (47.92%). The PG treatment also reduced (P < 0.01) the blood ketone concentration of cows as compared with the control without PG use (0.89 vs. 1.22 mmol/L). In the Exp. 2, after treatment the reduction of blood ketone concentration was much greater (P 0.05).

Abstract:BackgroundOvine pregnancy toxaemia is a common metabolic disorder of ewes due to increased foetal energy requirements in late pregnancy. This pathology is a metabolic condition characterized by hypoglycaemia and hyperketonaemia resulting in the inability of the animal to maintain an adequate energy balance. The response to treatment is effective, if it is started in the early stages of the disease, when irreversible neurological injuries have not yet been established. The aim was to evaluate three therapeutic alternatives to effectively reverse the disease process in its early stages.For this, thirty adult Corriedale ewes, pregnant with a single lamb, were randomly separated in three groups of ten animals each, at day 130 of gestation. From that day onwards, ewes were locked up for forage fasting until glycaemia reached clinical values defining sub-clinical pregnancy toxaemia (1.59 ± 0.24 mmol/L). After fasting, ewes grazed and received a treatment for 4 days: 50 ml i.v. infusions of hypertonic glucose and 20 UI insulin/ewe/day s.c. or 100 ml/sheep/12 h of glycerol together with propylene glycol oral solution or fed with pasture supplemented with two daily intakes 300 g/sheep of cracked corn. Glycaemia and ß-hydroxybutyrate were determined in all the animals from the beginning of fasting until the completion of the treatment.ResultsFasting caused a decline in blood glucose in the 3 groups. This decline continued until fasting was withdrawn and treatment began. Thereafter blood glucose increased in all three groups, although in the group supplemented with glycerol and propylene glycol it started to increase significantly after 12 h. The values of ß-hydroxybutyrate decreased in the 3 groups at the start of treatment, and this decline was more pronounced earlier on and in the group supplemented with glycerol and propylene glycol. We found no significant differences between all experimental groups. No animal showed clinical signs of pregnancy toxaemia throughout the research.ConclusionsThe three treatments administered to sheep affected by sub-clinical pregnancy toxaemia were able to restore normal concentration of glucose and ß-hydroxybutyrate in blood, although per os administration of 100 ml/sheep/12 h of glycerol with propylene glycol, was the most successful treatment, normalizing the aforementioned biochemical parameters in a shorter time.

Abstract:Despite increased efforts in preventing the occurrence of metabolic disorders in transition cows, hyperketonemia remains a frequent early-lactation metabolic disease affecting an average of 40% of cows in herds in the United States. Despite the demonstrated economic effect of this disorder, controlled clinical trials comparing different treatment strategies in affected cows are lacking. The objective of our study was to investigate the effect of treatment with intravenous glucose, oral propylene glycol, or a combination of both on the reduction in blood β-hydroxybutyrate (BHB) concentrations of early-lactation hyperketonemic dairy cows. Multiparous Holstein cows between 3 to 9 d in milk were screened for hyperketonemia using a handheld meter 3 times per week, and enrolled at whole blood BHB concentration ≥1.2 mmol/L to 1 of 4 treatment groups: (1) 500 mL of a 50% dextrose solution i.v. once daily for 3 d (GLU, n = 9), (2) 300 mL of propylene glycol as a drench once daily for 3 d (PG, n = 9), (3) a combination treatment of a 500 mL of 50% dextrose solution i.v. and 300 mL of propylene glycol orally once daily for 3 d (GLU+PG, n = 8), or (4) an untreated control group (CTRL, n = 8). Blood samples were collected immediately before as well as at 1, 2, 4, 8, 12, 24, 36, 48, 60, and 72 h after administration of the first treatment through a jugular catheter and 3 times per week thereafter from coccygeal vessels. Concentrations of BHB were measured in whole blood, and plasma samples were analyzed for glucose, fatty acid (NEFA), insulin, glucagon, and electrolyte concentrations. The EDTA-anticoagulated blood samples were assessed for red blood cell indices, and smears were made for evaluation of red blood cell morphology. Outcomes were analyzed using repeated measures analysis. Overall least squares means (95% CI) of whole blood BHB concentrations between 1 h and d 11 relative to first treatment were 1.11 (0.95 to 1.30), 1.26 (1.07 to 1.47), 0.96 (0.81 to 1.13), and 1.53 (1.30 to 1.80) mmol/L for the GLU, PG, GLU+PG, and CTRL groups, respectively. Treatment with both glucose and propylene glycol led to a greater magnitude and more prolonged decrease in BHB concentrations compared with individual treatments. The NEFA and glucagon concentrations were lower immediately after treatment in GLU and GLU+PG groups compared with CTRL, and treatment with both glucose and propylene glycol was associated with a greater increase in glucose and insulin concentrations immediately after treatment compared with CTRL and GLU treatment alone. Treatments did not lead to differences in plasma mineral concentrations. We conclude that treatments varied in the magnitude of decreasing blood BHB concentrations in hyperketonemic postpartum cows, with the greatest decline after treatment with a combination of intravenous glucose and oral propylene glycol.
Negative energy balance affects perinatal ewe performance, rumen morphology, rumen flora structure, and placental function

Abstract:This study investigated the effects of negative energy balance (NEB) on perinatal ewes, with a focus on changes in growth performance, serum biochemical parameters, rumen fermentation, ruminal bacteria composition, placental phenotype-related indicators, and expression levels of genes related to placental function. Twenty ewes at 130 days of gestation were randomly allocated to either the positive energy balance (PEB) or NEB groups. In the experiment, ewes in the PEB group were fed the same amount as their intake during the pre-feeding baseline period, while ewes in the NEB group were restricted to 70% of their individual baseline feed intake. The experiment was conducted until 42 days postpartum, and five double-lamb ewes per group were selected for slaughter. The results demonstrated that NEB led to a significant decrease in body weight, carcass weight, and the birth and weaning weights of lambs (P < 0.05). Additionally, NEB caused alterations in serum biochemical parameters, such as increased non-esterified fatty acids and β-hydroxybutyrate levels and decreased cholesterol and albumin levels (P < 0.05). Rumen fermentation and epithelial parameters were also affected, with a reduction in the concentrations of acetic acid, butyric acid, total acid and a decrease in the length of the rumen papilla (P < 0.05). Moreover, NEB induced changes in the structure and composition of ruminal bacteria, with significant differences in α-diversity indices and rumen microbial community composition (P < 0.05). Gene expression in rumen papilla and ewe placenta was also affected, impacting genes associated with glucose and amino acid transport, proliferation, apoptosis, and angiogenesis (P < 0.05). These findings screened the key microbiota in the rumen of ewes following NEB and highlighted the critical genes associated with rumen function. Furthermore, this study revealed the impact of NEB on placental function in ewes, providing a foundation for investigating how nutrition in ewes influences reproductive performance. This research demonstrates how nutrition regulates reproductive performance by considering the combined perspectives of rumen microbiota and placental function.

Abstract:Summary Recently, certain batches of heparin have been associated with an acute, rapid onset of serious side-effects indicative of allergictype reactions.These reports generated significant concern regarding the possible presence of a dangerous contaminant within heparin and highlighted the need to re-assess the purity criteria of heparin preparations for clinical use. Given the nature of the array of all possible contaminants, traditional screening tests cannot safely differentiate between contaminated and uncontaminated heparin preparations. Monoand bi-dimensional NMR spectroscopy are powerful techniques that are able to detect and quantify a wide variety of potential sulfated polysaccharide contaminants. As such, these techniques are powerful tools for the analysis and assessment of heparin preparations.
Oral rehydration solution with glycerol to dairy calves: Effects on fluid balance, metabolism, and intestinal microbiota

Abstract:Response to glycerol and glucose as an additive for oral rehydration solution (ORS) was investigated in two trials comprising of total 24 young calves. The aim was to prevent dehydration and energy deficiency and to increase the population of beneficial bacteria in the intestinal microbiota. Calves responded with a modest elevation of glucose and insulin in plasma when fed ORS with glycerol/glucose. The glycerol/glucose mixture concentration in plasma increased markedly following an oral glycerol load. There was no increase in 1,3-propanediol, and the VFA pattern was unchanged in calves receiving ORS containing glycerol/glucose mixture, indicating that glycerol was rapidly absorbed into blood. Numbers of enterobacteria and lactobacilli were not affected by treatment, and glycerol-utilizing Lactobacillus reuteri was detected in calf feces irrespective of treatment and sampling time. In conclusion, glycerol can be used as a component of ORS for young calves without negative impact on calf performance and health.

Abstract:We have earlier reported that patients suffering from acquired immuno-deficiency syndrome (AIDS), systemic lupus erythematosus (SLE) with vasculitis, Wegner granulomatosis and certain types of late stage cancer have interferon inhibitory activity in their serum. The purpose of this study was to identify the factor(s) involved in this interferon inhibitory activity. Twenty patients with advanced AIDS, twenty patients with SLE and vasculitis and twenty normal healthy controls between ages 25-40 years were studied. In contrast to normal, healthy controls, significant interferon inhibitory activity was found in AIDS and SLE patients. This appears to be largely due to: (a) increased soluble circulating interferon alpha/beta receptors, (b) increased prostaglandin E2 levels which inhibits interferon and (c) a interferon inhibitory protein. Further understaging of the nature of interferon inhibitory activity in the patient's sera and development of anti-interferon inhibitory agents would greatly enhance interferons potential as a treatment modality.

Abstract:The purpose of this study was to determine the effect of oral propylene glycol (PG) administration on ketosis resolution and milk yield in cows diagnosed with subclinical ketosis (SCK). Cows from 4 freestall dairy herds (2 in New York and 2 in Wisconsin) were each tested 6 times for SCK from 3 to 16 d in milk on Mon-days, Wednesdays, and Fridays. Subclinical ketosis was defined as a β-hydroxybutyrate (BHBA) concentration of 1.2 to 2.9 m M /L, and clinical ketosis was defined as ≥3.0 m M /L. Cows with SCK were randomized to the treatment group (oral PG) or control group (no PG); treatment cows were drenched with 300 mL of PG once daily from the day they tested 1.2 to 2.9 m M /L until the day they tested <1.2 m M /L. Outcomes evaluated for all farms included time from SCK until BHBA test <1.2 m M /L or until BHBA test ≥3.0 m M /L. Individual milk weights for the first 30 d of lactation were evaluated for the 3 farms monitoring daily milk. Semiparametric proportional hazards models were used to evaluate time to event outcomes; repeated-measures ANOVA was used to assess milk weights. A total of 741 of 1,717 (43.2%) eligible enrolled cows had at least one BHBA test of 1.2 to 2.9 m M /L. Of these, 372 were assigned to the treatment group and 369 to the control group. Based on hazard ratios, PG-treated cows were 1.50 times more likely (95% confidence interval = 1.26 to 1.79) to resolve their SCK and 0.54 times less likely (95% confidence interval = 0.34 to 0.86) to develop clinical ketosis than control cows. Across the 3 herds measuring individual milk weights, treated cows produced 0.23 kg more milk per milking in the first 30 d of lactation than control cows, for a total difference of 0.69 kg/cow per day. After identification of a treatment by herd interaction, stratification by herd showed that treated cows produced more milk per milking on farm A (0.44 kg) and farm B (0.53 kg) in the first 30 d of lactation than control cows, for a total difference of 1.34 and 1.59 kg/d, respectively; milk production did not differ (0.02 kg per milking) between the 2 groups on farm D. These results show the positive effects of oral PG administration in fresh cows with SCK by helping to resolve SCK and preventing clinical ketosis. In addition, oral PG improves milk yield during early lactation in cows diagnosed with SCK.

Abstract:Ketosis in high‐yielding dairy cows is the most common metabolic pathology during late dryness and early lactation. This work aims to establish the possibility and effectiveness of therapy for ketosis in dairy cows through the intraperitoneal injection of a 20% glucose solution. For the experiment, on days 21‐30 of lactation, two groups of black‐and‐white cows with a β‐hydroxybutyrate level above 1.0 mmol / l were formed. Animals of the first group (n = 10) were injected four times with an interval of 24 hours intraperitoneally with 1000 ml of 20% dextrose solution. Cows of the second group (n = 10) in a similar regimen were injected dextrose intravenously. Blood for research was obtained before the start of injections, after 5, 30 minutes, 1, 2, 3 hours after the first injection of the solution, and one day and seven days after the end of therapy. It was found that a single intravenous injection of dextrose during the first 5 minutes is accompanied by an increase in the concentration of the monosaccharide in the blood by 2.2 times (from 5.1 to 11.4 mmol / L; P <0.001). The level decreases to the initial values within 1 hour, and intraperitoneal infusion does not cause hyperglycemia. A redistribution of protein fractions accompanies therapy of cows with ketosis: a decrease in the albumin‐globulin coefficient by 16.8% (P <0.05) with intraperitoneal administration and by 26.9% (P <0.01) with intravenous administration. The activity of AST after intracavitary injections of dextrose decreased by 36.5% (P <0.05). The loading of erythrocytes with substances of low and medium molecular weight was 19.4% (P <0.05) with the intraperitoneal injection. It was not observed with the intravascular route of administration. The morphological composition of blood after intravenous administration of dextrose was characterized by an increase in the level of leukocytes by 34.8% (P <0.05), a decrease in erythrocytes by 11.9% (P <0.05), hemoglobin by 11.6% (P <0, 05), hematocrit by 12.3% (P <0.05). Therapy via intraperitoneal injection did not cause pronounced changes in these parameters. The intravenous administration of glucose promoted a more rapid elimination of ketone bodies, which decreased by 2 times one day after the end of therapy (P <0.05), but after 7 days, their level began to increase again (by 21.1%). It did not have significant differences with indicators at the time of diagnosis. Intraperitoneal infusions were accompanied by a slight decrease (by 16.3%) in the level of β‐hydroxybutyrate one day after the end of therapy, followed by a pronounced decrease in the concentration of ketones by day 7 (2.3 times; P <0.01). The dynamics of fructosamine indicate that intracavitary injections do not cause glycosylation of plasma proteins, while intravenous infusions increase the values by 37.0 ‐ 39.4% (P <0.001). Thus, intraperitoneal administration of glucose during ketosis in dairy cows has a more favorable effect on the morphobiochemical composition of the blood and provides a more prolonged therapeutic effect.

Abstract:We determined the effect of ketosis treatment with propylene glycol (PG) or PG plus l-carnitine and methionine (Metabolase®, Fatro, Bologna, Italy) on the resolution, postpartum health, milk yield, and reproductive performances of dairy cows. Blood from 475 Holstein cows was collected weekly until 4 weeks after calving to measure blood β-hydroxybutyrate (BHBA) concentrations. Cows with blood BHBA concentration ≥1.2 mmol/L were diagnosed with ketosis and were enrolled. One hundred and fifty cows diagnosed with ketosis were randomly assigned to three treatment groups (Day 0): (1) PG (300 g, PO) for 3 days (PG group, n = 50), (2) PG (300 g, PO) plus l-carnitine (1.25 g) plus methionine (5 g, IV) for 3 days (PG + CM group, n = 50), and (3) no treatment (control group, n = 50). On Day 3, blood was collected to evaluate whether the ketosis had resolved. Cows in the PG and PG + CM groups with blood BHBA ≥1.2 mmol/L were retreated for an additional 2 days, and then blood BHBA concentration was evaluated on Days 5 and 10. Blood glucose and haptoglobin concentrations, rumen fill score (RFS), and body condition score (BCS) were measured on Days 0, 3, 5, and 10. Postpartum complications, milk yield during the first 2 months, and reproductive outcomes were evaluated. The probability of resolution from ketosis was higher (P < 0.05) in the PG + CM group than in the control group on Days 3, 5, and 10 (odds ratio: 2.6-6.3). Blood BHBA in the PG + CM group was lower (P < 0.05) than that of the control group on Days 3 and 5, whereas blood glucose in the PG + CM group was higher (P < 0.05) than that of the control group on Days 3 and 5. RFS in the PG and PG + CM groups was higher than that of the control group on Day 10 (P < 0.01), while BCS loss from Day 0-10 in the control group was higher than those of the PG and PG + CM groups (P < 0.05). Milk yields on the 30th and 60th days postpartum were higher in the PG + CM group than the control and PG groups (P < 0.05). Postpartum complications and intervals between calving and first postpartum insemination or pregnancy did not differ among the groups (P > 0.05). In conclusion, treatment of dairy cows with PG plus l-carnitine and methionine improved the chances of resolution of ketosis and increased milk yield, while affecting neither the incidence of postpartum complications nor reproductive performance.
Metabolism, Ketosis Treatment and Milk Production after Using Glycerol in Dairy Cows: A Review

Abstract:The aim of this paper is to review and systematize the current state of knowledge on glycol metabolism in cattle. Glycerol, derived from biodiesel production, must be purified in order to be a useful product for feeding livestock. The use of glycerol in the feeding of ruminants can be justified for several reasons: (i) it is a source of energy in the ration, (ii) it is a glucogenic precursor, and (iii) it may have an effect on milk composition. The high energy value of glycerol provides the opportunity to use this raw material as a partial grain substitute in cattle feed rations. Dietary supplementation of glycerol is associated with increased propionate, butyrate, valerate, and isovalerate concentrations in the rumen. Glycerol can be used at up to 10%-15% of the dietary dry matter (DM) and is well-established as a treatment for ketosis in cows. Glycerol increases plasma glucose and may reduce non-esterified fatty acids and β-hydroxybutyrate levels. The use of glycerol does not have a clear effect on DM intake, milk yield, or milk composition. However, some authors have reported an increase in milk yield after glycerol supplementation associated with decreased milk fat concentration. It is also possible that the concentration in the milk of odd-chain fatty acids and cis-9, trans-11 conjugated linoleic acid may increase after glycerol application.
