Abstract:The objective of this study was to evaluate the efficacy of propylene glycol (PG) on prevention and treatment of ketosis in dairy cows from 09/2018 to 03/2019. The study consisted of 2 experiments using a total of 126 multiparous dairy cows in a HF crossbred herd. Cows in both experiments were randomly assigned to treatments in a randomized complete block design. In Exp. 1, 96 cows without ketosis disease (BHBA < 1.4 mmol/L) were assigned to 2 groups (48 cows/group), including (1) untreated control group (no PG) and (2) treatment group (oral PG for 3 consecutive days after parturition). In Exp. 2, 30 ketotic cows (BHBA ≥ 1.4 mmol/L) were divided into 3 groups (10 cows/group), including (1) cows provided with glucose + vitamin B12 + dexamethasone (TT1), (2) cows treated with PG for 3 days (TT2), and (3) combination of TT1 and TT2 (TT3). The results of the Exp. 1 showed that the proportion of ketotic cows was lower (P < 0.01) in the PG-treated group (18.75%) than in the control group (47.92%). The PG treatment also reduced (P < 0.01) the blood ketone concentration of cows as compared with the control without PG use (0.89 vs. 1.22 mmol/L). In the Exp. 2, after treatment the reduction of blood ketone concentration was much greater (P 0.05).

Abstract:The purpose of this study was to determine the effect of oral propylene glycol (PG) administration on ketosis resolution and milk yield in cows diagnosed with subclinical ketosis (SCK). Cows from 4 freestall dairy herds (2 in New York and 2 in Wisconsin) were each tested 6 times for SCK from 3 to 16 d in milk on Mon-days, Wednesdays, and Fridays. Subclinical ketosis was defined as a β-hydroxybutyrate (BHBA) concentration of 1.2 to 2.9 m M /L, and clinical ketosis was defined as ≥3.0 m M /L. Cows with SCK were randomized to the treatment group (oral PG) or control group (no PG); treatment cows were drenched with 300 mL of PG once daily from the day they tested 1.2 to 2.9 m M /L until the day they tested <1.2 m M /L. Outcomes evaluated for all farms included time from SCK until BHBA test <1.2 m M /L or until BHBA test ≥3.0 m M /L. Individual milk weights for the first 30 d of lactation were evaluated for the 3 farms monitoring daily milk. Semiparametric proportional hazards models were used to evaluate time to event outcomes; repeated-measures ANOVA was used to assess milk weights. A total of 741 of 1,717 (43.2%) eligible enrolled cows had at least one BHBA test of 1.2 to 2.9 m M /L. Of these, 372 were assigned to the treatment group and 369 to the control group. Based on hazard ratios, PG-treated cows were 1.50 times more likely (95% confidence interval = 1.26 to 1.79) to resolve their SCK and 0.54 times less likely (95% confidence interval = 0.34 to 0.86) to develop clinical ketosis than control cows. Across the 3 herds measuring individual milk weights, treated cows produced 0.23 kg more milk per milking in the first 30 d of lactation than control cows, for a total difference of 0.69 kg/cow per day. After identification of a treatment by herd interaction, stratification by herd showed that treated cows produced more milk per milking on farm A (0.44 kg) and farm B (0.53 kg) in the first 30 d of lactation than control cows, for a total difference of 1.34 and 1.59 kg/d, respectively; milk production did not differ (0.02 kg per milking) between the 2 groups on farm D. These results show the positive effects of oral PG administration in fresh cows with SCK by helping to resolve SCK and preventing clinical ketosis. In addition, oral PG improves milk yield during early lactation in cows diagnosed with SCK.

Ruminal and intermediary metabolism of propylene glycol in lactating Holstein cows

Abstract:Four lactating Holstein cows fitted with ruminal cannulas and permanent indwelling catheters in the mesenteric artery, mesenteric vein, hepatic portal vein, and hepatic vein were used in a cross-over design to study the metabolism of propylene glycol (PG). Each cow received 2 treatments: control (no infusion) and infusion of 650 g of PG into the rumen at the time of the morning feeding. Propylene glycol was infused on the day of sampling only. Samples of arterial, portal, and hepatic blood as well as ruminal fluid were obtained at 0.5 h before feeding and at 0.5, 1.5, 2.5, 3.5, 5, 7, 9, and 11 h after feeding. Infusion of PG did not affect ruminal pH or the total concentration of ruminal volatile fatty acids, but did decrease the molar proportion of ruminal acetate. The ruminal concentrations of PG, propanol, and propanal as well as the molar proportion of propionate increased with PG infusion. The plasma concentrations of PG, ethanol, propanol, propanal, glucose, L-lactate, propionate, and insulin increased with PG and the plasma concentrations of acetate and beta-hydroxybutyrate decreased. The net portal flux of PG, propanol, and propanal increased with PG. The hepatic uptake of PG was equivalent to 19% of the intraruminal dose. When cows were dosed with PG, the hepatic extraction of PG was between 0 and 10% depending on the plasma concentration of PG, explaining the slow decrease in arterial PG. The increased net hepatic flux of L-lactate with PG could account for the entire hepatic uptake of PG, which suggests that the primary hepatic pathway for PG is oxidation to l-lactate. The hepatic uptake of propanol increased with PG, but no effects of PG on the net hepatic and net splanchnic flux of glucose were observed. Despite no effect of PG on net portal flux and net hepatic flux of propionate, the net splanchnic flux of propionate increased and the data suggest that propionate produced from hepatic metabolism of propanol is partly released to the blood. The data suggest that PG affects metabolism of the cows by 2 modes of action: 1) increased supply of l-lactate and propionate to gluconeogenesis and 2) insulin resistance of peripheral tissues induced by increased concentrations of PG and propanol as well as a decreased ratio of ketogenic to glucogenic metabolites in arterial blood plasma.

Abstract:We determined the effect of ketosis treatment with propylene glycol (PG) or PG plus l-carnitine and methionine (Metabolase®, Fatro, Bologna, Italy) on the resolution, postpartum health, milk yield, and reproductive performances of dairy cows. Blood from 475 Holstein cows was collected weekly until 4 weeks after calving to measure blood β-hydroxybutyrate (BHBA) concentrations. Cows with blood BHBA concentration ≥1.2 mmol/L were diagnosed with ketosis and were enrolled. One hundred and fifty cows diagnosed with ketosis were randomly assigned to three treatment groups (Day 0): (1) PG (300 g, PO) for 3 days (PG group, n = 50), (2) PG (300 g, PO) plus l-carnitine (1.25 g) plus methionine (5 g, IV) for 3 days (PG + CM group, n = 50), and (3) no treatment (control group, n = 50). On Day 3, blood was collected to evaluate whether the ketosis had resolved. Cows in the PG and PG + CM groups with blood BHBA ≥1.2 mmol/L were retreated for an additional 2 days, and then blood BHBA concentration was evaluated on Days 5 and 10. Blood glucose and haptoglobin concentrations, rumen fill score (RFS), and body condition score (BCS) were measured on Days 0, 3, 5, and 10. Postpartum complications, milk yield during the first 2 months, and reproductive outcomes were evaluated. The probability of resolution from ketosis was higher (P < 0.05) in the PG + CM group than in the control group on Days 3, 5, and 10 (odds ratio: 2.6-6.3). Blood BHBA in the PG + CM group was lower (P < 0.05) than that of the control group on Days 3 and 5, whereas blood glucose in the PG + CM group was higher (P < 0.05) than that of the control group on Days 3 and 5. RFS in the PG and PG + CM groups was higher than that of the control group on Day 10 (P < 0.01), while BCS loss from Day 0-10 in the control group was higher than those of the PG and PG + CM groups (P < 0.05). Milk yields on the 30th and 60th days postpartum were higher in the PG + CM group than the control and PG groups (P < 0.05). Postpartum complications and intervals between calving and first postpartum insemination or pregnancy did not differ among the groups (P > 0.05). In conclusion, treatment of dairy cows with PG plus l-carnitine and methionine improved the chances of resolution of ketosis and increased milk yield, while affecting neither the incidence of postpartum complications nor reproductive performance.
Effects of propylene glycol on in vitro ruminal fermentation, methanogenesis, and microbial community structure

Abstract:We evaluated the effects of propylene glycol (PG) on in vitro ruminal fermentation, methanogenesis, and microbial community structure. A completely randomized design was conducted in the in vitro incubation, and 4 culture PG dose levels (0, 7.5, 15, and 22.5 μL/g of dry matter) were used in the trial. Based on the fermentation results, the control group (0 μL/g of dry matter, CON) and the second treatment group (15.0 μL/g of dry matter, TRT) were chosen for further analysis to explore the effects of PG on the bacterial and archaeal community structure. The concentrations of propanol, propanal, and succinate increased linearly, whereas the concentration of l-lactate decreased linearly as PG doses increased. The molar proportion of propionate demonstrated a linear increase with increasing PG doses. In contrast with propionate, the molar proportion of acetate and butyrate, and acetate-to-propionate ratio decreased linearly with increasing PG doses. The addition of PG markedly decreased methane production without negative effects on nutrient degradability. In the archaeal level, the relative abundance of Methanobrevibacter tended to decrease, but that of Methanomassiliicoccus significantly increased in TRT group. At the bacterial level, the relative abundance of Bacteroidetes and Prevotella in TRT group was numerically higher than that in CON group. The analysis of the Negativicutes class showed that the relative abundance of Succiniclasticum tended to increase, whereas that of Selenomonas tended to decrease in TRT group. These results demonstrated that PG might be used as an inhibitor to mitigate methane emission. However, the small decrease in methane production will limit the application of PG as a methane inhibitor in production practices. Further research is needed to determine whether use together with other inhibitors may improve the effects of PG on the utilization of reducing equivalents ([H]) and methane production.

Abstract:This study investigates the impact of propylene glycol (PRG) on ketotic cows, focusing on alleviating oxidative stress and enhancing immunity through modulating amino acid and lipid metabolism. Ketosis, a prevalent metabolic disease in dairy cows, negatively affects productivity and health. PRG, known for its gluconeogenic properties, was administered to cows with ketosis daily for three days and compared to an untreated group. Serum samples were taken to measure the biochemical parameters, and metabolomic and lipidomic analyses were performed with ultra-high-performance liquid chromatography–mass spectrometry. The results showed significant reductions in serum non-esterified fatty acids, beta-hydroxybutyrate, and C-reactive protein levels, alongside increased glucose, anti-inflammatory factor interleukin-10, superoxide dismutase, and glutathione peroxidase activities. Metabolomic and lipidomic analyses revealed significant alterations, including increased levels of glucogenic amino acids like glutamate and proline, and decreased levels of ceramide species. A pathway analysis indicated that PRG affects multiple metabolic pathways, including alanine, aspartate, glutamate metabolism, and sphingolipid metabolism. These findings suggest that PRG not only mitigates oxidative stress, but also enhances immune function by restoring metabolic homeostasis. This study provides valuable insights into the biochemical mechanisms underlying PRG's therapeutic effects, offering potential strategies for the effective management and treatment of ketosis in dairy cows.

Effect of Propylene Glycol Supplementation in Cows with Sub-clinical Ketosis

Abstract:The present study was conducted in crossbred dairy cows of Western Rajasthan. Two times sampling were done in each animal pre and post-treatment. The sub-clinical ketosis was diagnosed on the basis of plasma BHBA>1.2 mmol/L and NEFA>0.7 mmol/L during post partum period. Therapeutic trial was conducted using Propylene glycol and choline biocarbonate in sub-clinical ketosis cows. The result revealed increase in the value of glucose, milk yield and decrease NEFA, BHBA levels after propylene glycol supplementation. Whereas, no significant change were recorded in total protein, albumin, PUN, creatinine, AST, ALT and triglyceride. Highly significant increases mean value of glutathione and decrease in malondialdehyde was also observed. Similarly, highly significant decreased GGT value and significant increased cholesterol, calcium level in pre to post-treatment. While NEB is a physiologically normal process, excessive NEB reflects poor adaptation and results in adverse health and production effects after calving.

Abstract:BackgroundIt is unknown which metabolites are responsible for propylene glycol (PG)-induced toxicosis, and a better understanding of the underlying mechanisms explaining incidences of abnormal behaviour of dairy cows fed PG is therefore needed.MethodsThe study included three cows of which one developed PG toxicosis. In order to investigate how the metabolism of PG differed in the cow developing toxicosis, proton nuclear magnetic resonance (NMR) spectroscopy was applied on ruminal fluids and blood plasma samples obtained before and after feeding with PG.ResultsPG toxicosis was characterized by dyspnea and ruminal atony upon intake of concentrate containing PG. The oxygen saturation of arterial blood haemoglobin and the oxygen pressure in arterial blood decreased along with the appearance of the clinical symptoms. NMR revealed differences in plasma and ruminal content of several metabolites between the cow responding abnormally to PG and the two control cows.ConclusionIt is concluded that PG-toxicosis is likely caused by pulmonary vasoconstriction, but no unusual metabolites directly related to induction of this condition could be detected in the plasma or the ruminal fluid.

The effect of propylene glycol delivery method on blood metabolites in dairy cows

Abstract:Four dairy cows in a positive energy balance were used to study the effect of propylene glycol supplementation variants on blood indices. The treatments were: without glycol, glycol mixed with the total mixed ration (TMR), glycol delivered as top dressing (TD), and glycol administered as an oral drench (OD). Oral drenching had the positive effect of higher blood insulin concentration 1.5 and 2.5 h after supplementation compared to the TD method ( P ≤ 0.05). Glycol supplemented as TD had the negative effect of higher concentration of non-esterified fatty acids (NEFA) 3.5 h after supplementation ( P ≤ 0.05) with a trend towards a higher mean concentration of this marker ( P ≤ 0.1). A decreasing tendency was recorded for the NEFA concentration 1.5 h after glycol supplementation with TMR or OD ( P ≤ 0.1). Glycol supplemented as OD had the positive effect of lowering the β-hydroxybutyric acid concentration 1.5 and 2.5 h after feeding ( P ≤ 0.05). A downward trend in the mean β-hydroxybutyric acid level after glycol delivery as OD was also observed ( P ≤ 0.1). Glycol supplemented as TD had the negative effect of decreasing dry matter intake of the diet ( P ≤ 0.05). Glycol supplemented as OD had a more glucogenic effect than as part of the TMR. Due to reduction of dry matter intake and the lowest glucogenic effect, using glycol as TD is unjustified. The results confirm that the best method of propylene glycol administration is OD, which can be useful in the prevention of ketosis, however, it should be confirmed in a long-term experiment covering the transition and early lactation periods.

Measured outcomes were development of displaced abomasum (DA) and removal from herd in the first 30 d in milk (DIM), conception to first service, and time to conception within 150 DIM. Cows from 4 freestall dairy herds (2 in New York and 2 in Wisconsin) were each tested 6 times for SCK from 3 to 16 DIM on Mondays, Wednesdays, and Fridays using the Precision Xtra meter (Abbott Laboratories, Abbott Park, IL). Subclinical ketosis was defined as a blood β-hydroxybutyrate concentration of 1.2 to 2.9 mmol/L. Cows with SCK were randomized to treatment group (oral PG) or untreated control group (no PG); treatment cows were orally drenched with 300 mL of PG once daily from the day they tested 1.2 to 2.9 mmol/L until the day they tested <1.2 mmol/L. Mixed effects multivariable Poisson regression was used to assess the effect of PG on DA, removal from herd, and conception to first service; a semiparametric proportional hazards model was used to evaluate the days-to-conception outcome. A total of 741 of 1,717 (43.2%) eligible enrolled cows had at least 1 β-hydroxybutyrate test of 1.2 to 2.9 mmol/L. Of these, 372 were assigned to the PG treatment group and 369 to the control group. Thirty-nine cows (5.3%) developed a DA after testing positive for SCK and 30 cows (4.0%) died or were sold within the first 30 DIM. Based on risk ratios, control cows were 1.6 times more likely [95% confidence interval (CI)=1.3 to 2.0] to develop a DA and 2.1 times more likely (95% CI=1.2 to 3.6) to die or be sold than cows treated with PG. In addition, PG-treated cows were 1.3 times more likely (risk ratio 95% CI=1.1 to 1.5) to conceive at first insemination than control cows in 3 of the herds. No difference was observed in days to conception within 150 DIM between treatment groups (hazard ratio for PG cows=1.1, 95% CI=0.8 to 1.4), with a median time to conception of 100 d (95% CI=93 to 111) and 104 d (95% CI=95 to 114) for PG-treated and control cows, respectively. These results show that intensive detection of SCK, followed by treatment of positive cows with oral PG decreased the risk of developing a DA or leaving the herd within the first 30 DIM and increased the risk of conception to first service.

Abstract:Summary Recently, certain batches of heparin have been associated with an acute, rapid onset of serious side-effects indicative of allergictype reactions.These reports generated significant concern regarding the possible presence of a dangerous contaminant within heparin and highlighted the need to re-assess the purity criteria of heparin preparations for clinical use. Given the nature of the array of all possible contaminants, traditional screening tests cannot safely differentiate between contaminated and uncontaminated heparin preparations. Monoand bi-dimensional NMR spectroscopy are powerful techniques that are able to detect and quantify a wide variety of potential sulfated polysaccharide contaminants. As such, these techniques are powerful tools for the analysis and assessment of heparin preparations.
Mechanism(S) of interferon inhibitory activity in blood from patients with AIDS and patients with lupus erythematosus with vasculitis.

Abstract:We have earlier reported that patients suffering from acquired immuno-deficiency syndrome (AIDS), systemic lupus erythematosus (SLE) with vasculitis, Wegner granulomatosis and certain types of late stage cancer have interferon inhibitory activity in their serum. The purpose of this study was to identify the factor(s) involved in this interferon inhibitory activity. Twenty patients with advanced AIDS, twenty patients with SLE and vasculitis and twenty normal healthy controls between ages 25-40 years were studied. In contrast to normal, healthy controls, significant interferon inhibitory activity was found in AIDS and SLE patients. This appears to be largely due to: (a) increased soluble circulating interferon alpha/beta receptors, (b) increased prostaglandin E2 levels which inhibits interferon and (c) a interferon inhibitory protein. Further understaging of the nature of interferon inhibitory activity in the patient's sera and development of anti-interferon inhibitory agents would greatly enhance interferons potential as a treatment modality.
Efficacy of Different Drenching Regimens of Gluconeogenic Precursors during Transition Period on Body Condition Score, Production, Reproductive Performance, Subclinical Ketosis and Economics of Dairy Cows

Abstract:A total of 108 Holstein cows were used to evaluate the effect of drenching propylene glycol (PG) either individually or in combination with glycerol (G) on body condition score (BCS), production, reproductive performance, protection against subclinical ketosis and economic benefit of dairy cows during the transition period. The six groups (n = 18/group) were; Control group, cows received no treatment; PG300, cows were drenched 300 mL PG for 7 days pre-expected day of calving and 21 days post-calving; PG400, cows were drenched 400 mL PG for 7 days pre-and 7 days post-calving; PG500, cows were drenched 500 mL PG for 10 days pre-and 10 days post-calving; GPG150, cows were drenched a mixture of 150 mL G and 150 mL PG for 21 days pre-and 21 days post-calving; GPG250, cows were drenched a mixture of 250 mL G and 250 mL PG for 21 days pre-and 21 days post-calving. BCS at 30 days in milk (DIM) was significantly higher in all treated groups in comparison to the control one and the best values were observed in GPG250, GPG150 and PG500 groups. Lactation length (LL) was significantly (p < 0.001) shorter in GPG250, GPG150 and PG500 groups than control group. There was a significant increment in 305 milk yield (p < 0.001) and average daily milk yield (p < 0.001) in GPG250 and PG500 groups than other groups except for PG300 with the lowest values for control and PG400. Cows in all treatment groups were observed in heat and conceived earlier as well as had shorter open days and calving interval durations (p < 0.001) and low number of services per conception (p = 0.004) compared to control group with better results for PG500 and GPG250 groups. BHB level and percentage of cows suffered from subclinical ketosis at 7 DIM was significantly lower in GPG250, GPG150 and PG500 groups than control group. Cows in treatment groups had a significantly higher glucose level (p = 0.006) compared to control group. Regarding to economics, the control group showed the highest feed costs, insemination costs and total costs per animal. Additionally, control and PG400 groups had the highest cost per kilogram of milk from total and feed costs compared to other cows. PG300, PG500 and GPG250 groups recorded a greater net return and income over feed cost (IOFC). In conclusion, the continuous drenching of propylene glycol either individually or in combination with glycerol regimens for long duration (PG300, PG500, GPG150, GPG250) during the transition period of dairy cows may reduce the incidence of subclinical ketosis and consequently improve milk production, reproduction and economics compared to drenching for short duration (PG400).
Correlation between composition of the bacterial community and concentration of volatile fatty acids in the rumen during the transition period and ketosis in dairy cows

Abstract:The transition period is a severe challenge to dairy cows. Glucose supply cannot meet demand and body fat is mobilized, potentially leading to negative energy balance (NEB), ketosis, or fatty liver. Propionate produces glucose by gluconeogenesis, which depends heavily on the number and species of microbes. In the present study, we analyzed the rumen microbiome composition of cows in the transition period, cows with ketosis, and nonperinatal cows by terminal restriction fragment length polymorphism (TRFLP) analysis of 16S rRNA genes and quantitative PCR. TRFLP analysis indicated that the quantity of Veillonellaceae organisms was reduced and that of Streptococcaceae organisms was increased in rumen samples from the transition period and ketosis groups, with the number of Lactobacillaceae organisms increased after calving. Quantitative PCR data suggested that the numbers of the main propionate-producing microbes, Megasphaera elsdenii and Selenomonas ruminantium, were decreased, while numbers of the main lactate-producing bacterium, Streptococcus bovis, were increased in the rumen of cows from the transition period and ketosis groups, with the number of Lactobacillus sp. organisms increased after calving. Volatile fatty acid (VFA) and glucose concentrations were decreased, but the lactic acid concentration was increased, in rumen samples from the transition period and ketosis groups. Our results indicate that the VFA concentration is significantly related to the numbers of Selenomonas ruminantium and Megasphaera elsdenii organisms in the rumen.

Abstract:In the study, the effects of propylene glycol (PG) drenched in different doses in dairy cows with positive energy balance were evaluated on serum metabolic parameters. Twenty-four Simmental dairy cows in 60-190 days of lactation period with positive energy balance were included to this study. PG was drenched 3 h after morning feeding, at a doses of 300 mL (8 dairy cows) and 500 mL (8 dairy cows), once a day, for 3 days. Eight dairy cows were included for control group. Blood samples were collected once before PG drenching, daily after drenching of PG, and finally on day 4. Serum biochemical parameters were determined. With regard to energy metabolism, glucose concentrations from serum biochemical parameters significantly increased on day 3 in 500 mL PG group compared to control group, and there were not significant changes in BHBA and NEFA concentrations. Decreased urea, increased chloride concentrations were determined with within reference limits. In dairy cows with positive energy balance, 500 mL PG oral drenching had positive effect on energy balance as determined by serum glucose measurements, might not have negative effects on hepatic and renal function, and may cause serum electrolyte changes within reference limits.

Abstract:Forty-four holstein dairy cows were randomly enrolled in the treatment group (group 1, n=19) or control group (group 2, n=25) at calving. Group 1 received prophylactic propylene glycol treatment (PPGT) (300 ml/cow, beginning at calving, total 3 days). The group 2 remained untreated. All animals were tested on blood and milk beta-hydroxybutyric acid (BHBA) at postpartum week 2 and 4 (PW2 and 4), body condition scores (BCS), average daily milk production (ADMP) and postpartum health disorders were monitored in 90 days in milk (90 DIM). Abstract:1. The tRNA methyltransferase activity in vitro of leaves, cotyledons and roots of 85-day-old tea seedlings was studied. 2. The activity of extracts prepared from tea leaves with Polycar AT (insoluble polyvinylpyrrolidine) had optimum pH7.7 and was greatly influenced by thiol compounds, but only slightly by metal ions and ammonium acetate. 3. The activities of extracts, expressed per mg of protein, were as follows: roots greater than leaves greater than cotyledons. The only methylated base isolated after incubation with these preparations was 1-methyladenine. 4. The results did not support the view of involvement of methylation of nucleic acids in caffeine biosynthesis in tea plants. In contrast, it is suggested that theophylline is synthesized from the specific methylated precursor in nucleic acids, namely 1-methyladenylic acid, via 1-methylxanthine.
