Abstract:We postulated that daily drenching of propylene glycol to cows in early lactation would increase plasma glucose and insulin concentrations and improve fertility in postpartum cows. Thirty-six Holstein cows were assigned to treatment or control groups. Each treatment cow was given 500 ml of propylene glycol by drenching daily from 7 to 42 days of lactation. Blood samples for glucose, insulin, nonesterified fatty acids (NEFA), and plasma urea N were collected at 0, 30, and 90 min postdrenching once weekly during 1-6 weeks. Blood samples were collected for progesterone analysis and cows were palpated three times per week until 11 weeks to assess ovarian status. Propylene glycol did not affect dry matter intake (DMI), milk yield or energy balance in treatment cows. After drenching, propylene glycol increased (P<0.01) plasma glucose and insulin and decreased (P<0.01) NEFA; plasma urea N of the treatment group tended (P=0.07) to be higher than that of the control group through 90 min. Days to first service, days open, and services per conception were not different between groups. Conception rates to first insemination were 33% in the control group and 57% in treated cows, but these were not significantly different. First ovulation of treatment cows occurred earlier than that of control cows (32.3 versus 44.5 days, P=0.06) and the length of the first luteal phase was longer in treated cows (13.1 versus 7.3 days, P<0.05). These data are consistent with the hypothesis that insulin is important for normal ovarian function. During negative energy balance, treatment with propylene glycol, which induced small increases in plasma concentrations of insulin, prevented the short luteal phase characteristic of the first estrous cycle in control cows.

Abstract:We have earlier reported that patients suffering from acquired immuno-deficiency syndrome (AIDS), systemic lupus erythematosus (SLE) with vasculitis, Wegner granulomatosis and certain types of late stage cancer have interferon inhibitory activity in their serum. The purpose of this study was to identify the factor(s) involved in this interferon inhibitory activity. Twenty patients with advanced AIDS, twenty patients with SLE and vasculitis and twenty normal healthy controls between ages 25-40 years were studied. In contrast to normal, healthy controls, significant interferon inhibitory activity was found in AIDS and SLE patients. This appears to be largely due to: (a) increased soluble circulating interferon alpha/beta receptors, (b) increased prostaglandin E2 levels which inhibits interferon and (c) a interferon inhibitory protein. Further understaging of the nature of interferon inhibitory activity in the patient's sera and development of anti-interferon inhibitory agents would greatly enhance interferons potential as a treatment modality.

Abstract:Summary Recently, certain batches of heparin have been associated with an acute, rapid onset of serious side-effects indicative of allergictype reactions.These reports generated significant concern regarding the possible presence of a dangerous contaminant within heparin and highlighted the need to re-assess the purity criteria of heparin preparations for clinical use. Given the nature of the array of all possible contaminants, traditional screening tests cannot safely differentiate between contaminated and uncontaminated heparin preparations. Monoand bi-dimensional NMR spectroscopy are powerful techniques that are able to detect and quantify a wide variety of potential sulfated polysaccharide contaminants. As such, these techniques are powerful tools for the analysis and assessment of heparin preparations.

Abstract:In the study, the effects of propylene glycol (PG) drenched in different doses in dairy cows with positive energy balance were evaluated on serum metabolic parameters. Twenty-four Simmental dairy cows in 60-190 days of lactation period with positive energy balance were included to this study. PG was drenched 3 h after morning feeding, at a doses of 300 mL (8 dairy cows) and 500 mL (8 dairy cows), once a day, for 3 days. Eight dairy cows were included for control group. Blood samples were collected once before PG drenching, daily after drenching of PG, and finally on day 4. Serum biochemical parameters were determined. With regard to energy metabolism, glucose concentrations from serum biochemical parameters significantly increased on day 3 in 500 mL PG group compared to control group, and there were not significant changes in BHBA and NEFA concentrations. Decreased urea, increased chloride concentrations were determined with within reference limits. In dairy cows with positive energy balance, 500 mL PG oral drenching had positive effect on energy balance as determined by serum glucose measurements, might not have negative effects on hepatic and renal function, and may cause serum electrolyte changes within reference limits.
Effect of propylene glycol supplementation around parturition on milk yield, reproduction performance and some hormonal and metabolic characteristics in dairy cows

Abstract:Thirty-nine multiparous Holstein cows were used to measure the effect of propylene glycol treatment around parturition on milk yield, reproductive efficiency and some hormone and metabolite concentrations. Cows were assigned randomly to control (n = 19) or propylene glycol treated (n = 20) groups. Propylene glycol (300 g) was administered directly mixed with the diet from day 10 prior to the expected calving date until parturition (day 0) and orally after dilution in 1 l water on days 3, 6, 9 and 12. Blood samples were collected on days -20, -5, 0, 3, 10, 25 and 50 while milk samples were taken weekly until 13 weeks post partum. Body condition scores, recorded on days -20, 15 and 50, were not affected by propylene glycol administration. Propylene glycol did not significantly affect milk yield or composition but linear somatic cell score measured from the first 13 weeks post partum was reduced by propylene glycol administration (P < 0.01). Moreover, propylene glycol reduced milk urea (-25 mg/l, P < 0.05), especially during the first 9 weeks post partum. Plasma insulin concentrations were similar in both groups during the experiment while insulin-like growth factor I (P < 0.05) and insulin-like growth factor-binding protein 3 (P < 0.001) levels were higher on days 10, 25 and 50 post partum in the propylene glycol group. Propylene glycol administration decreased plasma non-esterified fatty acid concentrations (P < 0.05 to P < 0.01) but increased total cholesterol levels (P < 0.01) after parturition while 3-hydroxybutyrate levels were unaffected by the treatment. Changes in the hormone and metabolic concentrations after propylene glycol administration in the last few days of gestation and the first week of lactation seem to indicate that energy balance in the treated group was probably more positive than in the control group. There was also evidence that propylene glycol administration prevented fatty liver syndrome and hastened the resumption of oestrous cycles (P < 0.001).

Abstract:A total of 108 Holstein cows were used to evaluate the effect of drenching propylene glycol (PG) either individually or in combination with glycerol (G) on body condition score (BCS), production, reproductive performance, protection against subclinical ketosis and economic benefit of dairy cows during the transition period. The six groups (n = 18/group) were; Control group, cows received no treatment; PG300, cows were drenched 300 mL PG for 7 days pre-expected day of calving and 21 days post-calving; PG400, cows were drenched 400 mL PG for 7 days pre-and 7 days post-calving; PG500, cows were drenched 500 mL PG for 10 days pre-and 10 days post-calving; GPG150, cows were drenched a mixture of 150 mL G and 150 mL PG for 21 days pre-and 21 days post-calving; GPG250, cows were drenched a mixture of 250 mL G and 250 mL PG for 21 days pre-and 21 days post-calving. BCS at 30 days in milk (DIM) was significantly higher in all treated groups in comparison to the control one and the best values were observed in GPG250, GPG150 and PG500 groups. Lactation length (LL) was significantly (p < 0.001) shorter in GPG250, GPG150 and PG500 groups than control group. There was a significant increment in 305 milk yield (p < 0.001) and average daily milk yield (p < 0.001) in GPG250 and PG500 groups than other groups except for PG300 with the lowest values for control and PG400. Cows in all treatment groups were observed in heat and conceived earlier as well as had shorter open days and calving interval durations (p < 0.001) and low number of services per conception (p = 0.004) compared to control group with better results for PG500 and GPG250 groups. BHB level and percentage of cows suffered from subclinical ketosis at 7 DIM was significantly lower in GPG250, GPG150 and PG500 groups than control group. Cows in treatment groups had a significantly higher glucose level (p = 0.006) compared to control group. Regarding to economics, the control group showed the highest feed costs, insemination costs and total costs per animal. Additionally, control and PG400 groups had the highest cost per kilogram of milk from total and feed costs compared to other cows. PG300, PG500 and GPG250 groups recorded a greater net return and income over feed cost (IOFC). In conclusion, the continuous drenching of propylene glycol either individually or in combination with glycerol regimens for long duration (PG300, PG500, GPG150, GPG250) during the transition period of dairy cows may reduce the incidence of subclinical ketosis and consequently improve milk production, reproduction and economics compared to drenching for short duration (PG400).

Abstract:Forty-eight Holstein cows were used to determine the effects of short-term oral drenches of propylene glycol (PG) and Ca-soaps of palm oil fatty acids (fat) on plasma concentrations of key metabolites, liver composition, and milk production during the first 3 wk of lactation. Treatments (2 x 2 factorial arrangement) given orally once daily for the first 3 d postpartum were water (control), 500 ml of PG, 454 g of fat, or 500 ml of PG plus 454 g of fat. All treatments were administered as a total volume of 1.9 L. Administration of PG decreased concentrations of nonesterified fatty acids (NEFA) in plasma during the first 7 d and the first 21 d postpartum and tended to decrease concentrations of beta-hydroxybutyrate during the first 7 d postpartum. Concentrations of insulin in plasma were not affected by treatment. Administration of either PG or fat increased plasma glucose and liver glycogen concentrations compared to the control or PG plus fat treatments. Concentrations of triglycerides in liver were not affected by treatment. Administration of PG did not affect dry matter intake or milk yield and composition during the first 3 wk postpartum; however, cows drenched with fat tended to have lower dry matter intake and milk yield during the first 3 wk of lactation. Short-term drenching of PG effectively decreased NEFA concentrations in plasma during early lactation; however, data do not support administration of fat via drench to early lactation cows and concurrent administration of dietary fat appears to blunt the metabolic response of cows to PG.
Effect of peripartum dietary energy supplementation of dairy cows on metabolites, liver function and reproductive variables

Abstract:Multiparous Holstein cows (n=58) were used to study the effects of peripartum dietary supplementation on metabolic status, liver function and reproduction variables. Diets for cows were as follows: (a) no supplementation (CTL), (b) prilled fatty acids as 1.9% of DM (PrFA), (c) calcium salts of long chain n-6 fatty acids as 2.24% of DM (CaLFA) or (d) daily topdressing with 769 g of 65% propylene glycol (PGLY). Supplements were fed during the last 21 days before expected calving except for PGLY that continued until 21 days after parturition. Ovarian activity was monitored by transrectal ultrasonography and days to first ovulation were recorded. Liver biopsies were obtained on day 8 and 21 postpartum and analyzed for triglyceride content and mRNA expression of pyruvate carboxylase, cytosolic phosphoenolpyruvate carboxykinase, carnitine palmytoyltransferase 1A, and peroxisome proliferator-activated receptor-alpha. At 71 days following parturition, stage of ovarian cycles was synchronized and at day15 of the cycle oxytocin was injected i.v., blood samples were obtained at frequent intervals, and analyzed for 13,14 dihydro, 15-keto PGF(2alpha) (PGFM). Milk production and milk components were not different among treatment groups. Cows in PGLY gained body condition score (BCS) prepartum and net energy balance prepartum tended to be greater, but was not different postpartum from other groups. PGLY supplementation increased plasma insulin concentration prepartum, but not during the postpartum period. No significant differences were observed in plasma concentrations of glucose, NEFA, and insulin-like growth factor or hepatic triglyceride content, but all supplements tended to decrease beta hydroxybutyrate postpartum compared to CTL cows. Abundance of mRNA of gluconeogenic and lipid oxidation genes was not different among treatment groups. Days to first ovulation and uterine PGF(2alpha) production in response to an oxytocin treatment were not significantly different among treatment groups. Peripartum supplementation did not result in the substantial improvement of metabolic profile in early lactation nor significantly affect days to first ovulation and PGFM response to an oxytocin treatment.
Systemic blockade of transforming growth factor-beta signaling augments the efficacy of immunogene therapy.

Abstract:Locally produced transforming growth factor-beta (TGF-beta) promotes tumor-induced immunosuppression and contributes to resistance to immunotherapy. This article explores the potential for increased efficacy when combining immunotherapies with TGF-beta suppression using the TGF-beta type I receptor kinase inhibitor SM16. Adenovirus expressing IFN-beta (Ad.IFN-beta) was injected intratumorally once in established s.c. AB12 (mesothelioma) and LKR (lung cancer) tumors or intratracheally in a Kras orthotopic lung tumor model. Mice bearing TC1 (lung cancer) tumors were vaccinated with two injections of adenovirus expressing human papillomavirus-E7 (HPV-E7; Ad.E7). SM16 was administered orally in formulated chow. Tumor growth was assessed and cytokine expression and cell populations were measured in tumors and spleens by real-time PCR and flow cytometry. SM16 potentiated the efficacy of both immunotherapies in each of the models and caused changes in the tumor microenvironment. The combination of SM16 and Ad.IFN-beta increased the number of intratumoral leukocytes (including macrophages, natural killer cells, and CD8(+) cells) and increased the percentage of T cells expressing the activation marker CD25. SM16 also augmented the antitumor effects of Ad.E7 in the TC1 flank tumor model. The combination did not increase HPV-E7 tetramer-positive CD8(+) T cells in the spleens but did induce a marked increase in the tumors. Tumors from SM16-treated mice showed increased mRNA and protein for immunostimulatory cytokines and chemokines, as well as endothelial adhesion molecules, suggesting a mechanism for the increased intratumoral leukocyte trafficking. Blockade of the TGF-beta signaling pathway augments the antitumor effects of Ad.IFN-beta immune-activating or Ad.E7 vaccination therapy. The addition of TGF-beta blocking agents in clinical trials of immunotherapies may increase efficacy.
The effect of propylene glycol delivery method on blood metabolites in dairy cows

Abstract:Four dairy cows in a positive energy balance were used to study the effect of propylene glycol supplementation variants on blood indices. The treatments were: without glycol, glycol mixed with the total mixed ration (TMR), glycol delivered as top dressing (TD), and glycol administered as an oral drench (OD). Oral drenching had the positive effect of higher blood insulin concentration 1.5 and 2.5 h after supplementation compared to the TD method ( P ≤ 0.05). Glycol supplemented as TD had the negative effect of higher concentration of non-esterified fatty acids (NEFA) 3.5 h after supplementation ( P ≤ 0.05) with a trend towards a higher mean concentration of this marker ( P ≤ 0.1). A decreasing tendency was recorded for the NEFA concentration 1.5 h after glycol supplementation with TMR or OD ( P ≤ 0.1). Glycol supplemented as OD had the positive effect of lowering the β-hydroxybutyric acid concentration 1.5 and 2.5 h after feeding ( P ≤ 0.05). A downward trend in the mean β-hydroxybutyric acid level after glycol delivery as OD was also observed ( P ≤ 0.1). Glycol supplemented as TD had the negative effect of decreasing dry matter intake of the diet ( P ≤ 0.05). Glycol supplemented as OD had a more glucogenic effect than as part of the TMR. Due to reduction of dry matter intake and the lowest glucogenic effect, using glycol as TD is unjustified. The results confirm that the best method of propylene glycol administration is OD, which can be useful in the prevention of ketosis, however, it should be confirmed in a long-term experiment covering the transition and early lactation periods.

Effect of prepartum propylene glycol administration on periparturient fatty liver in dairy cows

Abstract:Plasma glucose concentration during late gestation was thought to be important for the development of fatty liver near parturition. Thirteen multiparous cows were given a 1-L oral drench of propylene glycol once daily beginning 10 +/- 3.6 d prepartum until parturition. Eleven control cows received a 1-L water drench. Plasma glucose increased following propylene glycol administration. Plasma NEFA concentration was 403 and 234 microM, and plasma insulin concentrations were .354 and .679 ng/ml, for control cows and cows treated with propylene glycol measured from 1 to 7 d prepartum. Plasma NEFA tended to be lower in cows treated with propylene glycol from 1 to 21 d postpartum. Prepartum propylene glycol administration reduced hepatic triglyceride accumulation by 32 and 42% at 1 and 21 d postpartum, respectively. Prepartum plasma BHBA was reduced during propylene glycol administration. Prepartum plasma glucose, NEFA, BHBA, and insulin were strongly correlated with liver triglyceride at 1 d postpartum (r = -.49, .45, .36, and -.49, respectively). Pre- and postpartum DMI were not affected by treatment. Milk production and composition measured through 21 d postpartum were not different between groups.

Abstract:Seventeen nonlactating Holstein cows were superovulated in a Latin-square designed experiment to determine the effects of increased propylene glycol (PROP) and luteinizing hormone (LH) during antral follicle development on ovarian function, fertilization, and early embryo quality. PROP was orally drenched every 4 h for 7 days to induce hyperinsulinemia and associated metabolic changes. LH concentrations were altered by increasing LH (3-fold) during last 2 days of superovulation. Treatment groups were as follows: (1) control-oral drenching with water plus low-LH preparation; (2) high LH(HLH)-water plus HLH preparation; (3) PROP-drenching with PROP plus low LH; (4) PROP/HLH-PROP plus HLH. PROP increased glucose (P < 0.05) and insulin (P < 0.02) concentrations at all time points analyzed. Neither PROP nor LH affected numbers of follicles > 9 mm at time of gonadotropin-releasing hormone-induced LH surge, although percentage of these follicles that ovulated was decreased by both PROP (P = 0.002) and LH (P = 0.048). In addition, PROP tended (P = 0.056) to decrease total number of ovulations. PROP reduced (P = 0.028) fertilization rate, while LH tended (P = 0.092) to increase fertilization rate. There was no effect of either PROP or LH on any measure of embryo quality including percentage of embryos that were degenerate, quality 1, or quality 1 and 2 of total structures collected or fertilized structures. These results indicate that acute elevation in insulin during the preovulatory follicular wave can decrease percentage of large follicles that ovulate, particularly when combined with increased LH, and reduce fertilization of ovulated oocytes.

Abstract:The aim of the study was to determine the effect of propylene glycol and maize grain content by-pass starch supplementation during the transition period and the first 56 days of lactation on blood metabolic indices, milk production and fertility parameters in dairy cows. Seventy-five Polish Holstein-Friesian dairy cows were assigned to treatment 21 days before calving. The treatments included: TG-2.5 kg triticale grain/cow per day supplemented from 14 days prepartum to day 56 postpartum, PG-2.5 kg triticale grain/cow per day supplemented from day 14 before parturition to day 56 postpartum, and 400 g propylene glycol/cow per day from 14 days prepartum to 14 days of lactation and MG-2.5 kg maize grain/cow per day supplemented from day 14 before parturition to day 56 postpartum. PG and MG had an effect resulting in the highest glucose concentration at 28 d of lactation. Cows assigned to the PG and MG groups had significantly higher cholesterol levels confronted with TG group at day 14 of lactation, while at days 28 and 56 the same difference was observed only between the MG and TG groups. PG had an effect on shortening the period to first ovulation. PG and MG improved the first service conception rate and decreased the number of services per conception in cows. In conclusion, both treatments of dairy cows with PG and MG improved their fertility parameters, while they had a slight effect on their metabolic profile and no effect on their milking performance.

Abstract:Stretchable electronics represent a new generation of electronics that utilize soft, deformable elastomers as the substrate or matrix instead of the traditional rigid printed circuit boards. As the most essential component of stretchable electronics, the conductors should meet the requirements for both high conductivity and the capability to maintain conductive under large deformations such as bending, twisting, stretching, and compressing. This review summarizes recent progresses in various aspects of this fascinating and challenging area, including materials for supporting elastomers and electrical conductors, unique designs and stretching mechanics, and the subtractive and additive patterning techniques. The applications are discussed along with functional devices based on these conductors. Finally, the review is concluded with the current limitations, challenges, and future directions of stretchable conductors.

Abstract:The purpose of this study was to determine the effect of oral propylene glycol (PG) administration on ketosis resolution and milk yield in cows diagnosed with subclinical ketosis (SCK). Cows from 4 freestall dairy herds (2 in New York and 2 in Wisconsin) were each tested 6 times for SCK from 3 to 16 d in milk on Mon-days, Wednesdays, and Fridays. Subclinical ketosis was defined as a β-hydroxybutyrate (BHBA) concentration of 1.2 to 2.9 m M /L, and clinical ketosis was defined as ≥3.0 m M /L. Cows with SCK were randomized to the treatment group (oral PG) or control group (no PG); treatment cows were drenched with 300 mL of PG once daily from the day they tested 1.2 to 2.9 m M /L until the day they tested <1.2 m M /L. Outcomes evaluated for all farms included time from SCK until BHBA test <1.2 m M /L or until BHBA test ≥3.0 m M /L. Individual milk weights for the first 30 d of lactation were evaluated for the 3 farms monitoring daily milk. Semiparametric proportional hazards models were used to evaluate time to event outcomes; repeated-measures ANOVA was used to assess milk weights. A total of 741 of 1,717 (43.2%) eligible enrolled cows had at least one BHBA test of 1.2 to 2.9 m M /L. Of these, 372 were assigned to the treatment group and 369 to the control group. Based on hazard ratios, PG-treated cows were 1.50 times more likely (95% confidence interval = 1.26 to 1.79) to resolve their SCK and 0.54 times less likely (95% confidence interval = 0.34 to 0.86) to develop clinical ketosis than control cows. Across the 3 herds measuring individual milk weights, treated cows produced 0.23 kg more milk per milking in the first 30 d of lactation than control cows, for a total difference of 0.69 kg/cow per day. After identification of a treatment by herd interaction, stratification by herd showed that treated cows produced more milk per milking on farm A (0.44 kg) and farm B (0.53 kg) in the first 30 d of lactation than control cows, for a total difference of 1.34 and 1.59 kg/d, respectively; milk production did not differ (0.02 kg per milking) between the 2 groups on farm D. These results show the positive effects of oral PG administration in fresh cows with SCK by helping to resolve SCK and preventing clinical ketosis. In addition, oral PG improves milk yield during early lactation in cows diagnosed with SCK.

Abstract:The purpose was to determine the effect of oral propylene glycol (PG) administration in fresh cows diagnosed with subclinical ketosis (SCK). Measured outcomes were development of displaced abomasum (DA) and removal from herd in the first 30 d in milk (DIM), conception to first service, and time to conception within 150 DIM. Cows from 4 freestall dairy herds (2 in New York and 2 in Wisconsin) were each tested 6 times for SCK from 3 to 16 DIM on Mondays, Wednesdays, and Fridays using the Precision Xtra meter (Abbott Laboratories, Abbott Park, IL). Subclinical ketosis was defined as a blood β-hydroxybutyrate concentration of 1.2 to 2.9 mmol/L. Cows with SCK were randomized to treatment group (oral PG) or untreated control group (no PG); treatment cows were orally drenched with 300 mL of PG once daily from the day they tested 1.2 to 2.9 mmol/L until the day they tested <1.2 mmol/L. Mixed effects multivariable Poisson regression was used to assess the effect of PG on DA, removal from herd, and conception to first service; a semiparametric proportional hazards model was used to evaluate the days-to-conception outcome. A total of 741 of 1,717 (43.2%) eligible enrolled cows had at least 1 β-hydroxybutyrate test of 1.2 to 2.9 mmol/L. Of these, 372 were assigned to the PG treatment group and 369 to the control group. Thirty-nine cows (5.3%) developed a DA after testing positive for SCK and 30 cows (4.0%) died or were sold within the first 30 DIM. Based on risk ratios, control cows were 1.6 times more likely [95% confidence interval (CI)=1.3 to 2.0] to develop a DA and 2.1 times more likely (95% CI=1.2 to 3.6) to die or be sold than cows treated with PG. In addition, PG-treated cows were 1.3 times more likely (risk ratio 95% CI=1.1 to 1.5) to conceive at first insemination than control cows in 3 of the herds. No difference was observed in days to conception within 150 DIM between treatment groups (hazard ratio for PG cows=1.1, 95% CI=0.8 to 1.4), with a median time to conception of 100 d (95% CI=93 to 111) and 104 d (95% CI=95 to 114) for PG-treated and control cows, respectively. These results show that intensive detection of SCK, followed by treatment of positive cows with oral PG decreased the risk of developing a DA or leaving the herd within the first 30 DIM and increased the risk of conception to first service.

Abstract:We determined the effect of ketosis treatment with propylene glycol (PG) or PG plus l-carnitine and methionine (Metabolase®, Fatro, Bologna, Italy) on the resolution, postpartum health, milk yield, and reproductive performances of dairy cows. Blood from 475 Holstein cows was collected weekly until 4 weeks after calving to measure blood β-hydroxybutyrate (BHBA) concentrations. Cows with blood BHBA concentration ≥1.2 mmol/L were diagnosed with ketosis and were enrolled. One hundred and fifty cows diagnosed with ketosis were randomly assigned to three treatment groups (Day 0): (1) PG (300 g, PO) for 3 days (PG group, n = 50), (2) PG (300 g, PO) plus l-carnitine (1.25 g) plus methionine (5 g, IV) for 3 days (PG + CM group, n = 50), and (3) no treatment (control group, n = 50). On Day 3, blood was collected to evaluate whether the ketosis had resolved. Cows in the PG and PG + CM groups with blood BHBA ≥1.2 mmol/L were retreated for an additional 2 days, and then blood BHBA concentration was evaluated on Days 5 and 10. Blood glucose and haptoglobin concentrations, rumen fill score (RFS), and body condition score (BCS) were measured on Days 0, 3, 5, and 10. Postpartum complications, milk yield during the first 2 months, and reproductive outcomes were evaluated. The probability of resolution from ketosis was higher (P < 0.05) in the PG + CM group than in the control group on Days 3, 5, and 10 (odds ratio: 2.6-6.3). Blood BHBA in the PG + CM group was lower (P < 0.05) than that of the control group on Days 3 and 5, whereas blood glucose in the PG + CM group was higher (P < 0.05) than that of the control group on Days 3 and 5. RFS in the PG and PG + CM groups was higher than that of the control group on Day 10 (P < 0.01), while BCS loss from Day 0-10 in the control group was higher than those of the PG and PG + CM groups (P < 0.05). Milk yields on the 30th and 60th days postpartum were higher in the PG + CM group than the control and PG groups (P < 0.05). Postpartum complications and intervals between calving and first postpartum insemination or pregnancy did not differ among the groups (P > 0.05). In conclusion, treatment of dairy cows with PG plus l-carnitine and methionine improved the chances of resolution of ketosis and increased milk yield, while affecting neither the incidence of postpartum complications nor reproductive performance.
