Abstract:A new system for multiple simultaneous gas-liquid chromatographic analysis is described. A commercially available gas chromatograph was modified to accommodate three wall-coated glass-capillary columns. A novel design of both the injection port and sample splitter allows the admission of equal portions of the sample into each of the three columns from a single injection. This provides the analyst either with data for the sample in question on three different liquid phases, or with a single result in triplicate if the same liquid phase is used on all columns. This presentation briefly describes the geometry of the inlet system; details will be published shortly. The performance of this system is illustrated with a simple example. In an artificial mixture, the quantitative GLC-data of 6 compounds obtained on three different columns were unequivocally correlated. A correlation number (CN) is derived which gives the maximum possible number of compounds that can be theoretically correlated in a mixture by the system described.

Abstract:We report here the first coupling of fast GC to IRMS, in a system capable of 250 ms peak widths (fwhm) at 1 mL/min flow rates, one-fifth as narrow as any previously reported GCC-IRMS system. We developed an optimized postcolumn interface that results in minimal peak broadening, using a programmable temperature vaporization injector in place of a rotary valve or backflush system to divert solvent, a narrow capillary combustion reactor followed by a cryogenic water trap with narrow-bore (< 0.20 mm We transfer lines, and a narrow W. open split to the IRMS directly inserted into the column effluent. Quantitative combustion was demonstrated with CH4 injections. A comparison of CO2 injections with different fwhm peak widths (250, 2500, and 7500 ms) showed similar precisions, SD(delta C-13) = 0.2-0.3 parts per thousand, for injections of > 600 pmol C on column; precision for the narrow peaks (250 ms) was considerably better for injections < 150 pmol C on column. SD(delta C-13) < 1 parts per thousand was achievable for injections of 5-15 pmol on column for 250 ms wide peaks, 10-fold better precision than 2500 ms wide peaks, and within a factor of 3 of the counting statistics Emit For a mixture of 15 fatty acid methyl esters (FAME), 1.5 nmol C of each on column yielded typical SD(delta C-13(pdb)) = 0.4 parts per thousand for fast GC and 0.5%o for conventional GC. For 14 of the 15 FAME, delta C-13 values between the two systems were within 1.5 parts per thousand and not significantly different Fast GCC-IRMS required one-third the run time (450 s vs 1400 s) to achieve comparable resolution. Mean peak widths for fast GCC-IRMS of the FAME were 720 ins, compared to 650 ms by fast GC with flame ionization detection. At a 15-fold dilution (100 pmol C on column for each FAME), fast GCC-IRMS achieved -2-fold better precision and accuracy than similar injections on conventional GCC-ERMS. Finally, a mixture of 10 steroids (similar to 7 nmol C (100 ng) each on column) was analyzed with mean precision of SD)(delta C-13) = 0.2 parts per thousand in 620 s by fast GCC-IRMS, while conventional GCC-IRMS required 1200 s and achieved poorer resolution. delta C-13 values for the two system were similar (Delta delta C-13 <=:5 2 parts per thousand for all steroids), indicating that accuracy is not compromised. In summary, fast GCC-IRMS can achieve similar precision to conventional GC with considerable time savings for standard sample sizes (> 1 nmol C) and achieves modest precision (similar to 1 parts per thousand) near the counting statistics limit on low level components.

Abstract:This work illustrates how the Tandem micro-Reactor (TMR), when interfaced to a GC-MS, facilitates the rapid characterization of virtually any material: gases, liquids and solids. A TMR has two independently controlled micro-reactors in series. Each micro-reactor has three heating modes: isothermal (up to 900 degrees C), multi-linear (e.g., 100-400 degrees C at 20 degrees C/min) and distinct temperature steps (e.g., 100 degrees C, 200 degrees C, etc.). The TMR can be used for both Evolved Gas Analysis (EGA) and detailed component analysis. Three examples illustrate how the TMR-GC-MS can be configured to differentiate or identify virtually any material. (1) Lignin, oak, bagasse and cellulose are easily differentiated using EGA-MS - each material is analyzed directly by simply weighing the sample (<75 mesh) into the sample cup. (2,3) The temperature dependence of ethanol and methanol transformations to ethylene and propylene, respectively, are presented using an online, real-time configuration. The ethylene transformation is also performed using a series of ethanol injections at distinct catalyst temperatures. Lastly, regeneration of the catalyst's surface is performed in-situ, which improves laboratory productive as well as elevating the precision of the data. (C) 2014 Elsevier B.V. All rights reserved.

Abstract:Continuous manufacturing is becoming increasingly important in the (bio-)pharmaceutical industry, as more product can be produced in less time and at lower costs. In this context, there is a need for powerful continuous analytical tools. Many established off-line analytical methods, such as mass spectrometry (MS), are hardly considered for process analytical technology (PAT) applications in biopharmaceutical processes, as they are limited to at-line analysis due to the required sample preparation and the associated complexity, although they would provide a suitable technique for the assessment of a wide range of quality attributes. In this study, we investigated the applicability of a recently developed micro simulated moving bed chromatography system (μSMB) for continuous on-line sample preparation for MS. As a test case, we demonstrate the continuous on-line MS measurement of a protein solution (myoglobin) containing Tris buffer, which interferes with ESI-MS measurements, by continuously exchanging this buffer with a volatile ammonium acetate buffer suitable for MS measurements. The integration of the μSMB significantly increases MS sensitivity by removing over 98% of the buffer substances. Thus, this study demonstrates the feasibility of on-line μSMB-MS, providing a versatile PAT tool by combining the detection power of MS for various product attributes with all the advantages of continuous on-line analytics.

Abstract:Although comprehensive two-dimensional (2D) gas chromatography (GC × GC) is a powerful technique for complex samples, component overlap remains likely. An intriguing route to address this challenge is to utilize the additional peak capacity and chemical selectivity provided by comprehensive three-dimensional (3D) gas chromatography (GC3), especially with time-of-flight mass spectrometry detection (GC3-TOFMS). However, the GC3-TOFMS instrumentation reported to date has employed one or both modulators with a duty cycle < 100%, making the potential gain in detection sensitivity over GC × GC modest, or perhaps even worse. Herein, we describe instrumentation for GC3-TOFMS in which both modulators provide total-transfer (100% duty cycle). Specifically, the instrument is based on the facile modification of a commercial thermally modulated comprehensive GC × GC-TOFMS platform for modulation from the 1D column to the 2D column, with recently described dynamic pressure gradient modulation (DPGM) as the second modulator from the 2D column to the 3D column, which is a total-transfer flow modulation technique. Area measurements of 1D peaks are compared to the sum of 3D peak areas to validate the assumption that total-transfer from 1D to 3D is accomplished. Additionally, peak heights were amplified by as high as a factor of 177 (x̅ = 130, s = 47) via comparison of 1D peak heights to the maximum 3D peak heights. Column selection is explored, with emphasis on the resulting peak width-at-base on each dimension and usage of 3D space as evaluation metrics. Using a nonpolar × polar × ionic liquid column combination, an effective peak capacity which considers modulation-induced broadening as high as 32,300 for select analytes was achieved (x̅ = 19,900, s = 10,700). The analytical benefits of employing three selective phases, mass spectrometry detection, and total-transfer modulation are explored with separations of a metabolomics-type sample, i.e., derivatized porcine serum, and a jet fuel spiked with various sulfur-containing compounds.

Abstract:By combining the high selectivity of a gas chromatograph (GC) with the high sensitivity and decent selectivity of an ion mobility spectrometer (IMS), GC-IMS have become increasingly popular in many applications. However, most GC suffer from long analysis times. In contrast, an hyper-fast GC allows for extremely fast analysis in the tens of seconds while reaching comparably high resolution. In turn, coupling such hyper-fast GC with IMS requires sufficiently high repetition rate of recording full IMS spectra to resolve the short GC peaks. Therefore, we present a drift tube IMS with 100 Hz repetition rate. Key is a small effective detector volume combined with short drift length. Therefore, the ion source of the IMS combines a small reaction region with an extended field-switching ion shutter and optimized gas flows. To resolve even the shortest GC peaks with a full width at half maximum of 100 ms, a short drift length of just 41 mm was used, achieving a measurement time of 10 ms per spectrum and hence ten data points across the shortest GC peak. To avoid condensation of the sample, the entire IMS was heated isothermally to 120 °C. Despite short drift times and high temperatures, the IMS still reaches high resolving power of Rp = 60. The hyper-fast GC-IMS reaches low detection limits in the low ppbV range. For demonstration, ketone mixes and three different hop varieties were analyzed in <30 s.

Abstract:When a GC/MS system is coupled with a pressurized reactor, the separation efficiency and the retention time are directly affected by the reactor pressure. To keep the GC column flow rate constant irrespective of the reaction pressure, a restrictor capillary tube and an open split interface are attached between the GC injection port and the head of a GC separation column. The capability of the attached modules is demonstrated for the on-line GC/MS analysis of catalytic reaction products of a bio-oil model sample (guaiacol), produced under a pressure of 1 to 3 MPa.

Abstract:Multiplex sampling, so far mainly used as a tool for S/N ratio improvement in spectroscopic applications and separation techniques, has been investigated here for its potential suitability for time-resolved monitoring where chromatograms of transient signals are recorded at intervals much shorter than the chromatographic runtime. Different designs of multiplex sample introduction were developed and utilized to analyze lithium-ion battery degradation products under normal or abuse conditions to achieve fast and efficient sample introduction. After comprehensive optimization, measurements were performed on two different GC systems, with either barrier discharge ionization detection (BID) or mass spectrometric detection (MS). Three different injector designs were examined, and modifications in the pertinent hardware components and operational conditions used. The shortest achievable sample introduction time was 50 ms with an interval of 6 s. Relative standard deviations were lower than 4% and 10% for the intra- and inter-day repeatability, respectively. The sample introduction system and column head pressure had to be carefully controlled, as this parameter most critically affects the amount of sample introduced and, thus, detector response. The newly developed sample introduction system was successfully used to monitor volatile degradation products of lithium-ion batteries and demonstrated concentration changes over the course of time of the degradation products (e.g., fluoroethane, acetaldehyde and ethane), as well as for solvents from the battery electrolyte like ethyl carbonate.

Abstract:Gas chromatography is widely used to identify and quantify volatile organic compounds for applications ranging from environmental monitoring to homeland security. We investigate a new architecture for microfabricated gas chromatography systems that can significantly improve the range, speed, and efficiency of such systems. By using a cellular approach, it performs a partial separation of analytes even as the sampling is being performed. The subsequent separation step is then rapidly performed within each cell. The cells, each of which contains a preconcentrator and separation column, are arranged in progression of retentiveness. While accommodating a wide range of analytes, this progressive cellular architecture (PCA) also provides a pathway to improving energy efficiency and lifetime by reducing the need for heating the separation columns. As a proof of concept, a three-cell subsystem (PCA3mv) has been built; it incorporates a number of microfabricated components, including preconcentrators, separation columns, valves, connectors, and a carrier gas filter. The preconcentrator and separation column of each cell are monolithically implemented as a single chip that has a footprint of 1.8 × 5.2 cm2. This subsystem also incorporates two manifold arrays of microfabricated valves, each of which has a footprint of 1.3 × 1.4 cm2. Operated together with a commercial flame ionization detector, the subsystem has been tested against polar and nonpolar analytes (including alkanes, alcohols, aromatics, and phosphonate esters) over a molecular weight range of 32–212 g/mol and a vapor pressure range of 0.005–231 mmHg. The separations require an average column temperature of 63–68 °C within a duration of 12 min, and provide separation resolutions >2 for any two homologues that differ by one methyl group.

Abstract:A wide variety of analytical techniques have been employed for monitoring chemical reactions, with online instrumentation providing additional benefits compared to offline analysis. A challenge in the past for online monitoring has been placement of the monitoring instrumentation as close as possible to the reaction vessel to maximize sampling temporal resolution and preserve sample composition integrity. Furthermore, the ability to sample very small volumes from bench-scale reactions allows the use of small reaction vessels and conservation of expensive reagents. In this study, a compact capillary LC instrument was used for online monitoring of as small as 1 mL total volume of a chemical reaction mixture, with automated sampling of nL-scale volumes directly from the reaction vessel used for analysis. Analyses to demonstrate short term (~2 h) and long term (~ 50 h) reactions were conducted using tandem on-capillary ultraviolet absorbance followed by in-line MS detection or ultraviolet absorbance detection alone, respectively. For both short term and long term reactions (10 and 250 injections, respectively), sampling approaches using syringe pumps minimized the overall sample loss to ~0.2% of the total reaction volume.

Abstract:Multi-column periodic counter-current chromatography (PCC) has been developed for continuous antibody capture, but the complexity of continuous processes makes experimental optimization time consuming and costly. In this work, with twin-column continuous system as a typical case, mathematical models were established and used to evaluate the impacts of operating parameters for process development. The model fitted well with the experimental breakthrough curves and process performance under varying protein concentrations and residence times. Three important operating parameters, residence time for interconnected feeding (RTC), breakthrough percentage control for interconnected feeding (s) and disconnected feeding time (tDC), were evaluated systematically. The profiles of productivity and resin capacity utilization showed three phases as a function of RTC, which resulted in different optimization strategies towards s and tDC. Based on the model prediction, a working window of RTC and s can be determined for process development. Finally, a model-based design approach was proposed to determine the optimum operating conditions and to design a suitable continuous process for high productivity and capacity utilization. With the model-based design approach developed, the best performance of 12.8 g/L/h productivity and 91.9% capacity utilization was found for MabSelect SuRe resin under 1 mg/mL feeding IgG concentration at RTC = 2 min, s = 65% and tDC = 26 min.
A Portable Micro-Gas Chromatography with Integrated Photonic Crystal Slab Sensors on Chip

Abstract:The miniaturization of gas chromatography (GC) systems has made it possible to utilize the analytical technique in various on-site applications to rapidly analyze complex gas samples. Various types of miniaturized sensors have been developed for micro-gas chromatography (µGC). However, the integration of an appropriate detector in µGC systems still faces a significant challenge. We present a solution to the problem through integration of µGC with photonic crystal slab (PCS) sensors using transfer printing technology. This integration offers an opportunity to utilize the advantages of optical sensors, such as high sensitivity and rapid response time, and at the same time, compensate for the lack of detection specificity from which label-free optical sensors suffer. We transfer printed a 2D defect free PCS on a borofloat glass, bonded it to a silicon microfluidic gas cell or directly to a microfabricated GC column, and then coated it with a gas responsive polymer. Realtime spectral shift in Fano resonance of the PCS sensor was used to quantitatively detect analytes over a mass range of three orders. The integrated µGC-PCS system was used to demonstrate separation and detection of a complex mixture of 10 chemicals. Fast separation and detection (4 min) and a low detection limit (ng) was demonstrated.

Abstract:After its introduction in the early 1990's, comprehensive two-dimensional gas chromatography (GC×GC) has evolved from a separation science research tool to the central component of many industries. Despite the maturity of the technique, some fields remain reluctant to its use in routine applications. In the case of forensic science, some constraints are the strict requirements enforced in forensic laboratories and the time and effort that must be invested for intra-laboratory method validation. Concerns may also arise about whether information could be lost when transitioning to a new technique. This study reports on a method translation from conventional one-dimensional (1D) GC to GC×GC, ensuring the integrity of data as conversion is made. The GC was retrofitted with a reverse fill/flush (RFF) flow modulator and equipped with dual channel detection using a quadrupole mass spectrometer (qMS) and a flame ionization detector (FID). The parallel use of two detectors, where qMS was applied for qualitative identification and FID for quantification, allowed higher flows and slightly wider peaks to be exploited for the analysis of a volatile organic compound (VOC) reference mixture relevant to forensic VOC profiling. Peak quality assessment and calibration curves using GC-qMS and GC×GC-qMS/FID document the transfer and adaptation of the original method without a loss in data quality. Furthermore, the pre-processing and the data analysis pro-cessing steps, including calibration and peak quality assessment for each of the three datasets, are explained in detail. This information provides benchmark data for routine laboratories that want to implement a GC×GC approach into routine workflows.

Abstract:An integrated system having the combined capability to perform gas chromatography (GC), comprehensive two-dimensional GC (GC × GC), and target heart-cut multidimensional GC (MDGC) using olfactometry (O), flame ionization (FID), and/or mass spectrometry (MS) detection is described. This combines a number of contemporary GC methods into a single instrument to provide very high resolution profiling of a sample. This provides initial assessment of volatile compound composition through GC × GC analysis with FID, which can be correlated with GC analysis using parallel O and FID detection. Subsequent microfluidic (Deans) switching selects regions (heart-cuts) of the chromatographic elution from the first dimension ((1)D) column for further resolution on a long second dimension ((2)D(L)) column for parallel detection of O and MS. Various (2)D(L) operational conditions, as well as the effect of different heart-cut (H/C) duration, were compared. The favored mode involves cryotrapping of heart-cuts, cooling the oven, and reducing carrier flow to offer greater efficiency. An analytical strategy that incorporates GC-FID/O, GC × GC-FID, and MDGC-MS/O analyses with cumulative solid phase microextraction (SPME) sampling for volatile sample enrichment is presented in this work. Excellent qualitative and quantitative performance was demonstrated with a Shiraz wine sample and an allergens mixture, with tentative identification of acetic acid, octen-3-ol, and ethyl octanoate as aroma contributors in Shiraz wine and determination of β-damascenone (floral odor) well separated from hexanoic acid (sweaty odor). A novel approach to obtain (2)D retention indices is reported, allowing matching of mass spectral, (1)I (retention index in (1)D) and (2)I (retention index in (2)D) data. The method employs the same olfactory detector at the end of the (1)D and (2)D(L) columns.

Abstract:The request for novel hyphenated instruments and techniques, capable of affording exhaustive information and results, is a focus continuously watched out. In this context, the present work aimed at the development of an integrated system combining gas chromatographic (GC) separation with mass spectrometry (MS) and (solid deposition) Fourier transform infrared spectroscopy (FTIR) detection. An external transfer line was designed in the lab for the parallel coupling of the two detectors, in such a way to obtain complementary analytical information consisting of an MS spectrum, an IR spectrum and linear retention indices (LRI), within a single analysis. The instrument performance was demonstrated for the analysis of a commercial mixture consisting of 139 hydrocarbons, comprising linear, branched, unsaturated and aromatic compounds. A 100-m poly(dimethylsiloxane) column was employed for the separation, and the outlet flow was split 95:5 between the IR and MS detectors using two uncoated capillaries. The IR spectra were acquired from solid deposits on a zinc selenide disc (-90 °C), over a spot (detector area) of about 0.1 mm2, in the range of 4000-700 cm-1 and at a resolution of 4 cm-1. Final identification of the separated compounds by a library search was achieved by excluding incorrect results, sequentially using a three-filter approach (85% similarity against reference MS and IR library spectra and ±10 LRI unit tolerance). Based on these preliminary results, the GC-MS/sd-FTIR system is a promising tool for the characterization of complex matrix constituents, for which identification is cumbersome, by using only one detection technique.

Abstract:Advances in chemical sampling using miniature mass spectrometer technology are used to monitor slow reactions at a frequency of ca. 180 h-1 (on the Mini 12) with no sample carryover and with inline derivatization in the case of poorly ionizing compounds. Moreover, we demonstrate high reproducibility with a relative error of less than 10% for major components. Monitoring is enabled using a continuous-flow nanoelectrospray (CF-nESI) probe contained in a custom-built 3D-printed rotary holder. The holder position is automatically set using a stepper motor controlled by a microcontroller. Reaction progress of up to six reactions, including hydrazone formation and Katritzky transamination, can be monitored simultaneously without carryover for several hours.

Abstract:Gas chromatography (GC) is one of the most widespread techniques used in laboratories as a way to separate and analyze complex mixtures of volatile and semi-volatile compounds. The main advantage of miniaturization of these systems is the increased performance due to the reduced time for analysis and reduced fabrication cost of the complex pneumatic flow system. In this paper advanced design ideas and fabrication processes to facilitate integration of the sample concentrator and the column will be presented. Using the LIGA process, 0.5- and 2-m-long, 50-μm-wide, and up to 600-μm-high aspect ratio metal GC separation columns with on-chip integrated sample injection and detection were fabricated. Initial experiments of coating these high aspect ratio columns show promising results when compared to simple tubular columns.
