Abstract:Polydopamine (PDA) is a biopolymer, which can form uniform thin films on almost all solid substrates as well as at the liquid/air interface. Carbonized polydopamine possesses graphite-like structure and exhibits high electrical conductivity, which makes it a potential carbon-based thin film conductor. However, studies on mechanical behavior of PDA and its derived materials are very limited. In this study, PDA samples were synthesized through self-assembly of dopamine in aqueous solution. Elastic modulus of thin films was measured using the nanoindentation technique. It is shown that the Young’s modulus of PDA thin film increased with increasing heat treatment temperature (up to 600°C). Doping with Cu ions also increased the Young’s modulus of PDA. Furthermore, all PDA thin films, with and without Cu, exhibited creep behavior.
Polydopamine Films with 2D-like Layered Structure and High Mechanical Resilience

Abstract:Highly oriented, layered, and mechanically resilient films of polydopamine (PDA) have been synthesized from the air/water interface. The films show a unique layered structure, as shown by scanning and transmission electron studies (SEM/TEM) and X-ray diffraction analysis (XRD), which resemble that of 2D layered materials. The films exhibit a composition typical of PDA-based materials, as evidenced by X-ray photoelectron spectroscopy (XPS); moreover, the samples present the distinctive resonance modes of PDA-based nanomaterials in Raman and infrared spectroscopy (FTIR) experiments. The presence of highly ordinated 3–4 protomolecule stacking, taking place at the air/water interface, with a unique eumelanin-like supramolecular arrangement is presented. Moreover, the films show superior mechanical resilience with E = 13 ± 4 GPa and H = 0.21 ± 0.03 GPa, as revealed by nanoindentation experiments, making them highly resilient and easily transferable. Finally, the ordering induced by the interface opens many possibilities for further studies, including those regarding the supramolecular structure on PDA due to their similarity to 2D layered materials.This article has not yet been cited by other publications.

Abstract:Surface-adherent polydopamine (PDA) films as multifunctional coatings can be easily deposited onto a wide range of materials through dopamine self-polymerization. However, a lack of in-depth understanding of PDA aggregation and deposition processes and definite structure elucidation of PDA make it challenging to tailor the surface characteristic and functionality of the PDA films. Herein, we demonstrate that the surface characteristics of the PDA films can be readily tuned by controlling the competitive interplay between PDA aggregation in solution and deposition on the substrate. Moreover, a structural investigation of the PDA films using analytical tools such as X-ray photoelectron spectroscopy (XPS), time-of-flight secondary ion mass spectrometry (ToF-SIMS), and matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) allows us to propose a new structure model for the PDA building block. The (DHI)2/PCA trimer complex, which consists of two 5,6-dihydroxyindole (DHI) units and one pyrrolecarboxylic acid (PCA) moiety, is definitely identified as a primary building block of PDA, and its formation is steered by covalent interactions in the initial stages of polymerization. In latter stages, the (DHI)2/PCA trimer complexes are further linked primarily through noncovalent interactions to build up the supramolecular structure of PDA. This study provides new insights into the mechanisms of PDA buildup.

Abstract:Modern infrared (IR) spectroscopic methods as in situ IR spectroscopic ellipsometry (in situ IRSE) and the photothermal atomic force microscopy (AFM)‐IR technique are introduced as novel analysis methods for characterization of polydopamine (PDA) films. The visible (VIS) and IR ellipsometric studies serve for determination of thicknesses, IR optical constants and discussion of specific vibrational bands. The in situ IR ellipsometric studies (IRSE) of the thin film growth of polydopamine (PDA), a versatile material for bioactive surfaces, in tris(hydroxymethyl)aminomethane (Tris) buffer solution are used to monitor the time‐dependent growth process of a PDA film. Complementary methods as X‐ray photoelectron spectroscopy (XPS) and the nanoscale photothermal AFM‐IR technique give access to study chemical homogeneity of the probed spots. All of the results are in qualitative agreement and show up new analysis possibilities, even further work is required to resolve the open questions of the chemical structure of PDA films. As proof of principle reaction for biosensor applications the binding of thiol‐terminated molecules to the PDA film via Michael‐addition was investigated by in situ IRSE. In this paper, infrared spectroscopic ellipsometry (IRSE) technique is used to in situ monitor and characterize the growth of polydopamine (PDA) film on solid/liquid (silicon/solution) interface. The structure of PDA is proposed based on the results from IRSE, X‐ray photoelectron spectroscopy (XPS), and the possible application toward biosensors is also validated.

Abstract:The development of polydopamine (PDA) coatings with a nanometer-scale thickness on surfaces is highly desirable for exploiting the novel features arising from the specific structure on the molecular level. Exploring the mechanisms of thin-film growth is helpful for attaining desirable control over the useful properties of materials. We present a systematic study demonstrating the growth of a PDA thin film on the surface of mica in consecutive short deposition time intervals. Film growth at each deposition time was monitored through instrumental techniques such as atomic force microscopy (AFM), water contact angle (WCA) analysis, and X-ray photoelectron spectroscopy (XPS). Film growth was initiated by adsorption of the PDA molecules on mica, with subsequent island-like aggregation, and finally, a complete molecular level PDA film was formed on the surface due to further molecular adsorption. A duration of 60−300 s was sufficient for complete formation of the PDA layer within the thickness range of 0.5−1.1 nm. An outstanding feature of PDA ultrathin films is their ability to act as a molecular adhesive, providing a foundation for constructing functional surfaces. We also explored antimicrobial applications by incorporating Ag nanoparticles into a PDA film. The Ag NPs/PDA film was formed on a surgical blade and then characterized and confirmed by SEM-EDS and XPS. The modified film inhibited bacterial growth by up to 42% on the blade after cutting through a pork meat sample.

One-Pot Preparation of Bioactive Surface by Electropolymerization of Dopamine

Abstract:An electrochemical technique involved facile method of cyclic voltammetric oxidation of dopamine to fabricate thickness controllable PDA films (ePDA) was reported. This process was preceded under a neutral environment and in the absence of oxygen. Most importantly, with precisely controlled electrochemical parameters, this dopamine electropolymerization technique can be further combined with bioactive molecules modification via a one-pot strategy to construct functionalized surfaces. By electropolymerization of dopamine in a one-pot mixture with REDV peptide, an endothelial cells (ECs) specific ligand, ECs adhesion enhanced bioactive surface incorporated with REDV was obtained. Ellipsometry and scanning electron microscopy (SEM) confirmed the homogeneous ePDA film fabrication. X-ray photoelectron spectroscopy (XPS) and fluorescence analysis results demonstrated the successful incorporation of REDV peptide into ePDA films (ePDA@REDV). Cells adhesion experiments suggested an excellent activity of incorporated REDV peptide was retained. Furthermore, ECs exhibited improved attachment, spreading and vinculin formation on ePDA@REDV functionalized surface. This dopamine electropolymerization assisted one-pot strategy for rapid construction of bioactive surfaces provides an innovative approach for multifunctional modification of complicated conductive biomaterials and devices.
Electrochemical deposition of conductive and adhesive polypyrrole-dopamine films

Abstract:Electrode surfaces have been widely modified with electrically conductive polymers, including polypyrrole (PPY), to improve the performance of electrodes. To utilize conductive polymers for electrode modification, strong adhesion between the polymer films and electrode substrates should be ensured with high electrical/electrochemical activities. In this study, PPY films were electrochemically polymerized on electrodes (e.g., indium tin oxide (ITO)) with dopamine as a bio-inspired adhesive molecule. Efficient and fast PPY electrodeposition with dopamine (PDA/PPY) was found; the resultant PDA/PPY films exhibited greatly increased adhesion strengths of up to 3.7 ± 0.8 MPa and the modified electrodes had electrochemical impedances two to three orders of magnitude lower than that of an unmodified electrode. This electrochemical deposition of adhesive and conductive PDA/PPY offers a facile and versatile electrode modification for various applications, such as biosensors and batteries.

Abstract:Polydopamine (PDA) coating provides a promising approach for immobilization of biomolecules onto almost all kinds of solid substrates. However, the deposition kinetics of PDA coating as a function of temperature and reaction method is not well elucidated. Since dopamine self-polymerization usually takes a long time, therefore, rapid-formation of PDA film becomes imperative for surface modification of biomaterials and medical devices. In the present study, a practical method for preparation of rapidly-deposited PDA coating was developed using a uniquely designed device, and the kinetics of dopamine self-polymerization was investigated by QCM sensor system. It was found that high temperature and vigorous stirring could dramatically speed up the formation of PDA film on QCM chip surface. Surface characterization, BSA binding study, cell viability assay and antibacterial test demonstrates that the polydopamine coating after polymerization for 30 min by our approach exhibits similar properties to those of 24 h counterpart. The method has a great potential for rapid-deposition of polydopamine films to modify biomaterial surfaces.

Abstract:Polydopamine coatings have garnered significant attention due to their versatility and multifunctional properties, rendering them suitable for a wide array of applications, including medical devices and the creation of coatings with varying degrees of amphiphilicity. Nevertheless, the fabricating of durable polydopamine coatings with substantial thickness remains challenging, primarily due to difficulties in controlling the deposition process and the potential for coating delamination or detachment under mechanical stress. Here, we reported the development of easily scalable polydopamine (PDA) coatings with remarkable thickness (approximately 1.7 μm) and stability, adaptable to diverse materials, by incorporating an amine-containing dendrimer. Through a systematic screening process, we identified an optimal dopamine-dendrimer combination that effectively modified the synthesis of polycatecholamine, facilitated nanoparticle formation, and enhanced stability. This resulted in the controlled deposition of composite PDA nanoparticles formed in situ. Using this optimal binary composition, we achieved the eco-friendly creation of a superhydrophobic coating with exceptional stability via a one-step dip post-modification process involving polydimethylsiloxane (PDMS). This innovative approach yielded a remarkable water contact angle exceeding 150°. Furthermore, we measured advancing and receding contact angles of 155.88° and 150.90°, respectively, resulting in a hysteresis of only 4.98°.

Abstract:This study aims to explore the fundamental surface characteristics of polydopamine (pDA)-coated hydrophobic polymer films. A poly(vinylidene fluoride) (PVDF) film was surface modified by dip coating in an aqueous solution of dopamine on the basis of its self-polymerization and strong adhesion feature. The self-polymerization and deposition rates of dopamine on film surfaces increased with increasing temperature as evaluated by both spectroscopic ellipsometry and scanning electronic microscopy (SEM). Changes in the surface morphologies of pDA-coated films as well as the size and shape of pDA particles in the solution were also investigated by SEM, atomic force microscopy (AFM), and transmission electron microscopy (TEM). The surface roughness and surface free energy of pDA-modified films were mainly affected by the reaction temperature and showed only a slight dependence on the reaction time and concentration of the dopamine solution. Additionally, three other typical hydrophobic polymer films of polytetrafluoroethylene (PTFE), poly(ethylene terephthalate) (PET), and polyimide (PI) were also modified by the same procedure. The lyophilicity (liquid affinity) and surface free energy of these polymer films were enhanced significantly after being coated with pDA, as were those of PVDF films. It is indicated that the deposition behavior of pDA is not strongly dependent on the nature of the substrates. This information provides us with not only a better understanding of biologically inspired surface chemistry for pDA coatings but also effective strategies for exploiting the properties of dopamine to create novel functional polymer materials.

Abstract:Polydopamine (PDA), the final oxidation product of dopamine or other catecholamines, attracted much attention as versatile coatings that can be used to cover the surface of almost all materials with a conformal layer of adjustable thickness ranging from a few to about 100 nm. These PDA layers can be subsequently modified with molecules carrying nucleophilic groups or with metallic nanoparticles from solutions containing metallic cations. However, during the deposition of PDA film on the surfaces, the reaction products that are simultaneously obtained from the oxidation of catecholamines in solution precipitate. Hence, some recent effort has been devoted to produce PDA in the form of nanoparticles. The aim of this short review is to give a comprehensive description of the synthesis methods yielding of PDA nanoparticles in the absence or in the presence of templating agents (polymers, polyelectrolytes, surfactants, proteins, and even some small organic molecules). We will also describe the use of thin PDA layers to coat already synthesized nanoparticles or nanotubes. Finally, several first applications of the obtained PDA nanoparticles will be described.

Abstract:Mussel-inspired polydopamine (PDA) deposition provides a prominent approach for constructing functional coatings, which has received much research interest over the past decade. However, large PDA aggregates often formed and precipitated from the solution during the deposition process, significantly lowering the utilization efficiency of dopamine for surface modification. It is of both fundamental and practical importance to "reactivate" and reuse the precipitated aggregates to achieve higher usage efficiency of PDA in surface modifications. In this work, we report a facile, substrate-independent, and cost-effective coating strategy, by disassembling the precipitated PDA aggregates, to achieve the coating deposition through the complexation of disassembled polydopamine (d-PDA) species with Fe(III) ions on various substrates. Adsorption tests determined by a quartz crystal microbalance with dissipation (QCM-D) monitoring technique indicated that the pH of the solution and the ratio of d-PDA to Fe(III) significantly influence the deposition behavior of d-PDA/Fe(III). Force measurements using a surface force apparatus demonstrated that the coordination interaction between d-PDA and Fe(III) was the major force leading to the formation of coatings. The deposited d-PDA/Fe(III) coatings featured controllable nanoscale thickness, uniform surface morphologies, and light color. Furthermore, the d-PDA/Fe(III) coating could act as an intermediate layer in the preparation of hydrophobic polyurethane sponge for highly efficient oil/water separation. This work provides a useful strategy to realize the reusability of PDA aggregates for versatile surface functionalization, with implications for the fundamental understanding of the formation mechanism in the metal-phenolic complexation systems and development of new coating approaches in various engineering applications.

This material emulates the adhesive properties of mussel foot proteins, allowing the formation of robust and stable organic coatings on various surfaces. Researchers have attempted to identify and improve the surface‐coating properties of PDA by incorporating additional organic functional groups. This review explores the structural modifications of dopamine molecules at the organic chemistry level, with a focus on changes in the catechol moiety and aliphatic amines, and examines their impact on the applications of PDA. The flexibility of these modifications underscores their importance for optimizing PDA coatings for a wide range of applications.

Thin Films of Porphyrin-Perylene Molecular Array Fabricated by Electrophoresis Methodology

Abstract:Thin solid films of organic conjugated molecules are at the center of organic electronics. Low solubility and high sublimation temperature of porphyrin-perylene arrays make it impossible to fabricate uniform solid films with spin-coating and vacuum deposition methodology, though these arrays have important applications in the area of opto-electronics. Here we show that high quality thin films of a porphyrin-perylene array can be prepared by electrochemical deposition, a facile and widely used film-forming technique. The electrophoretic species are protonated porphyrin-perylene molecules, which allow us to grow molecular array films on electrodes. By annealing in ammonia atmosphere or in vacuum at elevated temperature, the protons coordinated with molecular arrays on the deposited films can be eliminated and the porphyrin-perylene arrays recovered to their pristine state.

Abstract:Currently available patterning methods, such as lithographic techniques and inkjet printing approaches, heavily rely on material properties and specific applications. This motivates us to explore and develop patterning strategies tailored to the unique characteristics of materials. In this context, we discuss the advancements in site-specific fabrication and patterning of polydopamine (pDA), a versatile coating material widely utilized in biomedical devices. First, we provide a brief Review of the current understanding of the chemical structure of pDA, addressing the ongoing challenges related to its uncontrollability due to its heterogeneous nature. Following this, we examine the control of the shape and morphology during the growth of pDA in the solution phase. Lastly, we comprehensively cover various patterning strategies specifically developed and optimized for the pDA coating. Our goal is to offer insights into the controllable synthesis and fabrication of pDA. We anticipate that this Review will contribute to the advancement of precise and tailored pDA coatings, opening up possibilities for applications in diverse fields, particularly in integrated devices and molecular architecting.

Abstract:Self-assembling polydopamine has shown great versatility as a multifunctional surface coating. This study investigates the adhesive properties of polydopamine used to bind electroless copper films to several polymer substrates widely used for commercial applications, including polyethylene (PE), polycarbonate (PC), poly(methyl methacrylate) (PMMA), and nylon 6,6. Each substrate was coated in a similar manner resulting in continuous copper films of 52±12nm in thickness. Transmission electron microscopy revealed a non-preferential polycrystalline microstructure of copper grains averaging 53±17nm in diameter with a range of 21nm to 123nm, indicating there is some overlap of grains in the film. Peel testing, tensile tests, and tribological oscillating wear tests were used to characterize the mechanical performance of the adhered copper films. The nylon substrate had the highest peel strength, 49±5mN/mm, while the remaining substrates ranged from 5 to 13mN/mm on average. Coated substrates were strained up to 20% tensile strain to investigate film delamination, buckling, and rupture. Transverse buckling of the films relative to the direction of strain was observed due to Poisson's effect during tension. Films on nylon substrates resisted buckling and rupture up to 20% strain, whereas buckling, delamination, and rupture was observed in the other substrates below 20% strain. Tribological testing revealed that the coefficient of friction was governed by the polymer substrate and was not dependent upon the polydopamine or copper films. As a demonstration of the robust nature of this method, a nylon suture was coated with polydopamine-adhered copper film and tied in a knot to show a large complex deformation state without showing signs of delamination.

Abstract:<section class="article-section article-section__abstract" lang="en" data-lang="en" id="section-1-en"> <h3 class="article-section__header main main">Abstract</h3> <div class="article-section__content en main"> <p>Inspired by the adhesive proteins of mussels, polydopamine (pDA) has emerged as one of the most widely employed methods for functionalizing material surfaces, fueled in part by the versatility, simplicity, and spontaneity of pDA film deposition on most materials upon immersion in an alkaline aqueous solution of dopamine. However, the rapid adoption of pDA for surface modification over the last decade stands in stark contrast to the slow pace in understanding the composition of pDA. Numerous attempts to elucidate the formation mechanism and structure of this fascinating material have resulted in little consensus mainly due to the insoluble nature of pDA; which renders most conventional methods of polymer molecular weight characterization ineffective.[1] Here, we employed the non‐traditional approach of single molecule force spectroscopy (SMFS) to characterize pDA films. Retraction of a pDA coated cantilever from an oxide surface shows the characteristic features of a polymer with contour lengths up to 200nm. pDA polymers are generally weakly bound to the surface through much of their contour length, with occasional “sticky” points. Our findings represent the first direct evidence for the polymeric nature of pDA and provide a foundation upon which to understand and tailor its physicochemical properties.</p>

Nano-FTIR spectroscopy of surface confluent polydopamine films – What is the role of deposition time and substrate material?

Abstract:Polydopamine (PDA) is a widely used anchoring layer for multiple purposes. While simple to prepare, PDA is characterized by high chemical and topological diversity, which can limit its versatility. Unraveling the formation mechanism and physicochemical properties of continuous confluent layer and adherent nanoparticles on the nanoscale is crucial to further extend the prospective applications of PDA. Utilizing nano-FTIR spectroscopy, we investigate layers of PDA on three different substrates (silicon/silicon dioxide, nitrogen-doped titanium oxide, and gold substrates) at varying times of deposition (ToD). We observed a good correlation between the nano-FTIR and macroscopic FTIR spectra that reflected the changes in the relative abundance of PDA and polymerization intermediates as ToD increased. To gain analytical power, we utilized the principal component analysis (PCA) and extracted additional information from the resulting loadings spectral curves and data distribution in the score plots. We revealed a higher variability of the spectra of ultrathin surface confluent layers compared to the adherent nanoparticles. While the spectra of nanoparticles showed no apparent dependency on either ToD or the substrate material, the spectra of layers were highly affected by the increasing ToD and exhibited a rise in the absorption of PDA. Concomitantly, the spectra of layers grouped according to the substrate material at the lowest ToD point to the fact that the substrate material affects the PDA's initial physicochemical structure. The observed separation gradually diminished with the increasing ToD as the PDA physicochemical structure became less influenced by the substrate material.

Ultrasmooth Polydopamine Modified Surfaces for Block Copolymer Nanopatterning on Flexible Substrates

Abstract:Nature has engineered universal, catechol-containing adhesives which can be synthetically mimicked in the form of polydopamine (PDA). In this study, PDA was exploited to enable the formation of block copolymer (BCP) nanopatterns on a variety of soft material surfaces. While conventional PDA coating times (1 h) produce a layer too rough for most applications of BCP nanopatterning, we found that these substrates could be polished by bath sonication in a weakly basic solution to form a conformal, smooth (root-mean-square roughness ∼0.4 nm), and thin (3 nm) layer free of large prominent granules. This chemically functionalized, biomimetic layer served as a reactive platform for subsequently grafting a surface neutral layer of poly(styrene-random-methyl methacrylate-random-glycidyl methacrylate) to perpendicularly orient lamellae-forming poly(styrene-block-methyl methacrylate) BCP. Moreover, scanning electron microscopy observations confirmed that a BCP nanopattern on a poly(ethylene terephthalate) substrate was not affected by bending with a radius of ∼0.5 cm. This procedure enables nondestructive, plasma-free surface modification of chemically inert, low-surface energy soft materials, thus overcoming many current chemical and physical limitations that may impede high-throughput, roll-to-roll nanomanufacturing.

Abstract:It has been demonstrated that dopamine can undergo oxidation and complicated self‐reaction in aqueous solution to form so‐called polydopamine (PDA). In this paper, for the first time, the hierarchical self‐assembly of dopamine into surface patterned structures is described. PDA nanoaggregates are first prepared assisted by ethylenediamine, based on the covalent self‐polymerization and noncovalent self‐assembly. The surface fractal patterns are further created and modulated by the dynamic self‐assembly of PDA nanoaggregates to develop more macroscopic ordered structure of PDA. Various surface patterns of PDA aggregates are successfully obtained by evaporative dewetting method. The effects of solution pH, type of inorganic salts, and temperature on the morphology of surface pattern are investigated. Mineralization or metal deposition is used to keep the pattern fixed. The surface patterning strategy reported here is applicable to a broad range of materials, which allows the developments of materials with controllable patterned structure.
