Abstract:UHFUS is a new technology with the range between 30 and 100 MHz. UHFUS frequency ultrasound represents a great opportunity for the diagnosis of numerous pathologies. UHFUS can identify the extent of the cutaneous lesion as accurately as the histopathologic examination. The ultra‐high frequency ultrasound (UHFUS) is characterized by the use of probes between 30 and 100 MHz. This technology has recently been introduced in clinical practice and represents an opportunity for the diagnosis of numerous pathologies. The high spatial resolution of UHFUS, up to 30 μ in pixel size, allows to study the pathological modifications and to guide microsurgery treatments in anatomical body structures not evaluable by conventional HFUS. The aim of this work is to provide a review of the literature on the current clinical applications of UHFUS and to discuss its added role in different clinical settings.

Abstract:Objectives: Ultra-high frequency ultrasonography (UHFUS) is a recently introduced diagnostic technique which finds several applications in diverse clinical fields. The range of frequencies between 30 and 100 MHz allows for high spatial resolution imaging of superficial structures, making this technique suitable for the imaging of skin, blood vessels, musculoskeletal anatomy, oral mucosa, and small parts. However, the current clinical applications of UHFUS have never been analyzed in a consistent multidisciplinary manner. The aim of this study is to revise and discuss the current applications of UHFUS in different aspects of research and clinical practice, as well as to provide some examples of the current work-in-progress carried out in our center. Materials and methods: A literature search was performed in order to retrieve articles reporting the applications of UHFUS both in research and in clinical settings. Inclusion criteria were the use of frequencies above 30 MHz and study design conducted in vivo on human subjects. Results: In total 66 articles were retrieved. The majority of the articles focused on dermatological and vascular applications, although musculoskeletal and intraoral applications are emerging fields of use. We also describe our experience in the use of UHFUS as a valuable diagnostic support in the fields of dermatology, rheumatology, oral medicine, and musculoskeletal anatomy. Conclusion: Ultra-high frequency ultrasonography application involves an increasing number of medical fields. The high spatial resolution and the superb image quality achievable allow to foresee a wider use of this novel technique, which has the potential to bring innovation in diagnostic imaging.

Abstract:High frequency ultrasonic imaging is considered by many to be the next frontier in ultrasonic imaging because higher frequencies yield much improved spatial resolution by sacrificing the depth of penetration. It has many clinical applications including visualizing blood vessel wall, anterior segments of the eye and skin. Another application is small animal imaging. Ultrasound is especially attractive in imaging the heart of a small animal like mouse which has a size in the mm range and a heart beat rate faster than 600 BPM. A majority of current commercial high frequency scanners often termed "ultrasonic backscatter microscope or UBM" acquire images by scanning single element transducers at frequencies between 50 to 80 MHz with a frame rate lower than 40 frames/s, making them less suitable for this application. High frequency linear arrays and linear array based ultrasonic imaging systems at frequencies higher than 30 MHz are being developed. The engineering of such arrays and development of high frequency imaging systems has been proven to be highly challenging. High frequency ultrasound may find other significant biomedical applications. The development of acoustic tweezers for manipulating microparticles is such an example.

Abstract:Ultrasound is a versatile and well-established technique using sound waves with frequencies higher than the upper limit of human hearing. Typically, therapeutic and diagnosis ultrasound operate in the frequency range of 500 kHz to 15 MHz with greater depth of penetration into the body. However, to achieve improved spatial resolution, high-frequency ultrasound (>15 MHz) was recently introduced and has shown promise in various fields such as high-resolution imaging for the morphological features of the eye and skin as well as small animal imaging for drug and gene therapy. In addition, high-frequency ultrasound microbeam stimulation has been demonstrated to manipulate single cells or microparticles for the elucidation of physical and functional characteristics of cells with minimal effect on normal cell physiology and activity. Furthermore, integrating machine learning with high-frequency ultrasound enhances diagnostics, including cell classification, cell deformability estimation, and the diagnosis of diabetes and dysnatremia using convolutional neural networks (CNNs). In this paper, current efforts in the use of high-frequency ultrasound from imaging to stimulation as well as the integration of deep learning are reviewed, and potential biomedical and cellular applications are discussed.

Abstract:Because it drives the compromise between resolution and penetration, the diffraction limit has long represented an unreachable summit to conquer in ultrasound imaging. Within a few years after the introduction of optical localization microscopy, we proposed its acoustic alter ego that exploits the micrometric localization of microbubble contrast agents to reconstruct the finest vessels in the body in-depth. Various groups now working on the subject are optimizing the localization precision, microbubble separation, acquisition time, tracking, and velocimetry to improve the capacity of ultrasound localization microscopy (ULM) to detect and distinguish vessels much smaller than the wavelength. It has since been used in vivo in the brain, the kidney, and tumors. In the clinic, ULM is bound to improve drastically our vision of the microvasculature, which could revolutionize the diagnosis of cancer, arteriosclerosis, stroke, and diabetes.

[Super-resolution ultrasound imaging : Methods and applications]

Abstract:Clinical/methodical issue: The microvasculature plays an important role in many pathologic conditions but cannot be characterized in high resolution via conventional ultrasound methods. Standard radiological methods: Doppler-based techniques, contrast-enhanced sonography as well as dynamic contrast-enhanced computed tomography (CT) and magnetic resonance imaging (MRI) are commonly used to characterize tissue vascularization. However, these techniques cannot visualize the microvasculature adequately. Methodical innovation: Ultrasound localization microscopy (ULM) consists of contrast-enhanced ultrasound measurements in combination with a complex post-processing algorithm which detects microbubbles with high precision. The vasculature can then be visualized by accumulating the microbubble positions in a final image. Performance: Compared to conventional ultrasound techniques, ULM improves the image resolution by a factor of more than 10. This currently results in resolutions down to 10 µm and allows, therefore, the visualization of capillaries and the assessment of their perfusion. Also, this does not lead to a reduction of the penetration depth or the signal-to-noise ratio (SNR). Achievement: The method enables the visualization of vascular structures in unsurpassed detail and has the potential to offer new possibilities for the diagnosis of various diseases and for gaining insights into physiological processes. However, ULM is not commercially available yet but is intensely being tested in clinical studies. Practical recommendations: ULM could potentially be applied to all fields in which the vasculature is relevant. Current fields of application include oncology, nephrology, and neurological research.
Ultrafast 3-D Super Resolution Ultrasound using Row-Column Array specific Coherence-based Beamforming and Rolling Acoustic Sub-aperture Processing: In Vitro, In Vivo and Clinical Study

Abstract:The row-column addressed array is an emerging probe for ultrafast 3-D ultrasound imaging. It achieves this with far fewer independent electronic channels and a wider field of view than traditional 2-D matrix arrays, of the same channel count, making it a good candidate for clinical translation. However, the image quality of row-column arrays is generally poor, particularly when investigating tissue. Ultrasound localisation microscopy allows for the production of super-resolution images even when the initial image resolution is not high. Unfortunately, the row-column probe can suffer from imaging artefacts that can degrade the quality of super-resolution images as `secondary' lobes from bright microbubbles can be mistaken as microbubble events, particularly when operated using plane wave imaging. These false events move through the image in a physiologically realistic way so can be challenging to remove via tracking, leading to the production of 'false vessels'. Here, a new type of rolling window image reconstruction procedure was developed, which integrated a row-column array-specific coherence-based beamforming technique with acoustic sub-aperture processing for the purposes of reducing `secondary' lobe artefacts, noise and increasing the effective frame rate. Using an {\it{in vitro}} cross tube, it was found that the procedure reduced the percentage of `false' locations from $\sim$26\% to $\sim$15\% compared to traditional orthogonal plane wave compounding. Additionally, it was found that the noise could be reduced by $\sim$7 dB and that the effective frame rate could be increased to over 4000 fps. Subsequently, {\it{in vivo}} ultrasound localisation microscopy was used to produce images non-invasively of a rabbit kidney and a human thyroid.

Acoustic Molecular Imaging Beyond the Diffraction Limit In Vivo

Abstract:Ultrasound molecular imaging (USMI) is a technique used to noninvasively estimate the distribution of molecular markers in vivo by imaging microbubble contrast agents (MCAs) that have been modified to target receptors of interest on the vascular endothelium. USMI is especially relevant for preclinical and clinical cancer research and has been used to predict tumor malignancy and response to treatment. In the last decade, methods that improve the resolution of contrast-enhanced ultrasound by an order of magnitude and allow researchers to noninvasively image individual capillaries have emerged. However, these approaches do not translate directly to molecular imaging. In this work, we demonstrate super-resolution visualization of biomarker expression in vivo using superharmonic ultrasound imaging (SpHI) with dual-frequency transducers, targeted contrast agents, and localization microscopy processing. We validate and optimize the proposed method in vitro using concurrent optical and ultrasound microscopy and a microvessel phantom. With the same technique, we perform a proof-of-concept experiment in vivo in a rat fibrosarcoma model and create maps of biomarker expression co-registered with images of microvasculature. From these images, we measure a resolution of 23 μm, a nearly fivefold improvement in resolution compared to previous diffraction-limited molecular imaging studies.

Abstract:Abstract Background Ultrasound was first introduced in clinical dermatology in 1979. Since that time, ultrasound technology has continued to develop along with its popularity and utility. Main text summary Today, high-frequency ultrasound (HFUS), or ultrasound using a frequency of at least 10 megahertz (MHz), allows for high-resolution imaging of the skin from the stratum corneum to the deep fascia. This non-invasive and easy-to-interpret tool allows physicians to assess skin findings in real-time, enabling enhanced diagnostic, management, and surgical capabilities. In this review, we discuss how HFUS fits into the landscape of skin imaging. We provide a brief history of its introduction to dermatology, explain key principles of ultrasonography, and review its use in characterizing normal skin, common neoplasms of the skin, dermatologic diseases and cosmetic dermatology. Conclusion As frequency advancements in ultrasonography continue, the broad applications of this imaging modality will continue to grow. HFUS is a fast, safe and readily available tool that can aid in diagnosing, monitoring and treating dermatologic conditions by providing more objective assessment measures.

Abstract:Point-of-care ultrasound demonstrates significant potential in biomedical research due to its noninvasive, real-time visualization, cost-effectiveness, and other biological benefits. Ultrasound irradiation can precisely control the mechanical and physicochemical effects on pathogenic lesions, enabling real-time visualization, tunable tissue penetration depth, and therapeutic applications. This review summarizes recent advancements in ultrasound-enabled diagnostics and therapeutics, focusing on mechanochemical effects that can be directly integrated into biomedical applications. Additionally, the structure-functionality relationships of sonotheranostic nanoplatforms are systematically discussed, providing insights into the underlying biological effects. Finally, the limitations of current ultrasonic medicine are discussed, along with potential expansions to facilitate patient-centered translations.

Abstract:Ultrasonic technologies show great promise for diagnostic imaging and drug delivery in theranostic applications. The development of functional and molecular ultrasound imaging is based on the technical breakthrough of high frame–rate ultrasound. The evolution of shear wave elastography, high-frequency ultrasound imaging, ultrasound contrast imaging, and super-resolution blood flow imaging are described in this review. Recently, the therapeutic potential of the interaction of ultrasound with microbubble cavitation or droplet vaporization has become recognized. Microbubbles and phase-change droplets not only provide effective contrast media, but also show great therapeutic potential. Interaction with ultrasound induces unique and distinguishable biophysical features in microbubbles and droplets that promote drug loading and delivery. In particular, this approach demonstrates potential for central nervous system applications. Here, we systemically review the technological developments of theranostic ultrasound including novel ultrasound imaging techniques, the synergetic use of ultrasound with microbubbles and droplets, and microbubble/droplet drug-loading strategies for anticancer applications and disease modulation. These advancements have transformed ultrasound from a purely diagnostic utility into a promising theranostic tool.

Abstract:At present, medical ultrasound technology is gradually developing from the traditional imagingof tissue and organ structure based on wave effect of ultrasound, and the treatment of diseased tissue based on thermal and cavitation effects, to the direction of multi-functional integration of diagnosis and treatment. Therefore, asone of the most important biological effects, the mechanical effect of ultrasound (i.e. acoustic radiation force) hasattracted more and more attention from scientific researchers and medical staffs. Benefiting from the in-depthunderstanding of the theory and mechanism of ultrasonic mechanical effect, as well as the rapid development ofinformation and electronic technology, a variety of new ultrasonic diagnosis and treatment technologies based onacoustic radiation force have emerged in recent years, which have greatly expanded the application of ultrasoundin clinics. For example, ultrasound elastography that can quantitatively measure the tissue stiffness and image itsdistribution, which can provide early screening for liver cirrhosis and breast cancer; as well as ultrasound neuromodulation that can non-invasively regulate the state of nerve cells in the brain, which can provide unique toolsfor brain disease treatment and neuroscience research. This article will systematically introduce the representativeachievements of these new medical ultrasound technologies based on the mechanical effect of ultrasound in recentyears, and analyze their future development directions.

Abstract:Recent advances in electronics and digital processing techniques have significantly improved conventional ultrasonic imaging systems and allowed the development of new and sophisticated scanning methods. As a result, ultrasonic imaging devices have become an important modality for the clinical radiologist, complimenting the images obtained from X-ray and nuclear cameras. A particular advantage of ultrasonic waves is that they are nonionizing, thus presenting less risk to both patient and examiner. This paper presents some of the basic principles of ultrasonic propagation in tissue and how those principles impact the design of imaging devices. The characteristics of both the B- scan and C-scan techniques are described along with a summary of various scan formats that are currently available. Examples of B-scan and C-scan instruments are presented along with their relative advantages, limitations, and current usage;representative images are presented whenever possible. This paper concludes with a description of some new research developments in this rapidly emerging technology.

Abstract:Objective: This study aimed to realise 3-D super-resolution ultrasound imaging transcutaneously with a row-column array which has far fewer independent electronic channels and a wider field of view than typical fully addressed 2-D matrix arrays. The in vivo image quality of the row-column array is generally poor, particularly when imaging non-invasively. This study aimed to develop a suite of image formation and post-processing methods to improve image quality and demonstrate the feasibility of ultrasound localisation microscopy using a row-column array, transcutaneously on a rabbit model and in a human. Methods: To achieve this, a processing pipeline was developed which included a new type of rolling window image reconstruction, which integrated a row-column array specific coherence-based beamforming technique with acoustic sub-aperture processing. This and other processing steps reduced the 'secondary' lobe artefacts, and noise and increased the effective frame rate, thereby enabling ultrasound localisation images to be produced. Results: Using an in vitro cross tube, it was found that the procedure reduced the percentage of 'false' locations from ∼26% to ∼15% compared to orthogonal plane wave compounding. Additionally, it was found that the noise could be reduced by ∼7 dB and the effective frame rate was increased to over 4000 fps. In vivo, ultrasound localisation microscopy was used to produce images non-invasively of a rabbit kidney and a human thyroid. Conclusion: It has been demonstrated that the proposed methods using a row-column array can produce large field of view super-resolution microvascular images in vivo and in a human non-invasively.

Abstract:The present study highlights the advances in ultrasound, especially regarding its clinical applications to critically ill patients. Artificial intelligence (AI) is crucial in automating image interpretation, improving accuracy and efficiency. Software has been developed to make it easier to perform accurate bedside ultrasound examinations, even by professionals lacking prior experience, with automatic image optimization. In addition, some applications identify cardiac structures, perform planimetry of the Doppler wave, and measure the size of vessels, which is especially useful in hemodynamic monitoring and continuous recording. The "strain" and "strain rate" parameters evaluate ventricular function, while "auto strain" automates its calculation from bedside images. These advances, and the automatic determination of ventricular volume, make ultrasound monitoring more precise and faster. The next step is continuous monitoring using gel devices attached to the skin.

Abstract:Medical imaging specialists continue to explore better ways of demonstrating pathology and anatomy of the musculo-skeletal system. The continuous quest is fuelled by the desire to improve diagnostic yield, perform procedures more quickly and accurately, reduce risks to patient or operator, achieve better cost efficiency and utilize less complex methodologies. In many instances, musculoskeletal ultrasound acts as a screening, diagnostic tool but also guide and monitor therapeutic interventions. The paper outlines the use of ultrasound in the imaging of peripheral nerve disorders, traumatic and atraumatic joint disorders, Doppler techniques such as super micro vascular Imaging and sono-elastography. Refinements in probe technology and application of digital and novel proprietary software, have continued to improve the resolution of ultrasound images and with finer details on a scale not previously possible. With increasing experience and standardization of protocols, Musculoskeletal ultrasound will continue to play a great role in the diagnostic work-up and treatment of related disorders.
