Abstract:Ynamides consisting of an electron-withdrawing group on the nitrogen atom have proven to be powerful and versatile building blocks that are involved in a variety of useful chemical transformations. Nonetheless, their utilization in asymmetric catalysis is still in its infancy and has only recently attracted the attention of the synthetic organic community. The focus of this review is to present an overview on the exciting results achieved thus far in the field of asymmetric catalytic reactions of ynamides with an emphasis on the advantages, limitations, mechanisms, and applications associated with current approaches. In addition, a personal outlook on the challenges and limitations that need to be overcome in this area is also given at the end.This article has not yet been cited by other publications.

Abstract:Ynamides are electron‐rich heteroatom‐substituted alkynes with C−C triple bond directly tethered to the amide group. Over the past decades, ynamides have proven to be versatile reagents for organic synthesis and have received extensive attention. Compared with the well‐established ionic reactions of ynamides, radical‐based ynamide reactions have been exploited relatively seldom. Herein, radical reactions of ynamides, classified by radical attack at the α‐position and β‐position of ynamides, are reviewed by highlighting the reaction selectivity, scope, mechanism, and applicability. The aim of this review is to provide a comprehensive summarization of these advances, casting light on the further development of ynamide chemistry.

Transition Metal-Catalyzed Tandem Reactions of Ynamides for Divergent N-Heterocycle Synthesis

Abstract:ConspectusYnamides are electron-rich heteroatom-substituted alkynes with a C-C triple bond directly attached to the amide group. Importantly, this amide group is able to impose an electronic bias, thus resulting in the highly regioselective attack of this polarized alkyne by a large variety of nucleophiles. Over the past two decades, catalytic reactions of ynamides have experienced dramatic developments, especially those catalyzed by transition metals. As a result, ynamides have been widely applied to the rapid and efficient assembly of versatile structurally complex N-containing molecules, especially in an atom-economic and stereoselective way.On the basis of newly developed ynamide preparations and new alkyne transformations, we first developed oxidation-initiated tandem reactions of ynamides such as zinc-catalyzed ynamide oxidation/C-H functionalization and copper-catalyzed ynamide oxidation/carbene metathesis, leading to divergent synthesis of isoquinolones, β-carbolines, and pyrrolo[3,4-c]quinolin-1-ones. Importantly, this protocol represents the first non-noble-metal-catalyzed intermolecular oxidation of alkynes by N-oxide type oxidants, and the related overoxidation could be dramatically inhibited in this non-noble-metal catalysis. Then, we achieved gold-catalyzed amination-initiated tandem reactions of ynamides via α-imino gold carbenes for efficient construction of various 2-aminoindoles, 3-amino-β-carbolines, and 2-aminopyrroles, where two new types of nitrene transfer reagents (benzyl azides and isoxazoles) were discovered. In particular, the use of isoxazoles as nitrene transfer reagents for atom-economic generation of α-imino metal carbenes has also been elegantly exploited by Hashmi, Liu, and many other groups, providing ready access to a wide range of functionalized N-heterocycles. Moreover, we revealed that donor/donor copper carbenes could be generated via copper-catalyzed diyne cyclization under mild conditions. These novel copper carbenes could undergo asymmetric C-H insertion, cyclopropanation, and formal [3 + 2] cycloaddition to produce diverse chiral polycyclic pyrroles with good to excellent enantioselectivities. Thus, this strategy may open new avenues in catalytic asymmetric reaction of ynamides, which remain largely unexplored and deserve more attention. Meanwhile, we also accomplished the efficient and practical synthesis of medium-sized lactams by yttrium-catalyzed cascade cyclization of allyl alcohol-tethered ynamides and the combination of radical chemistry based on visible-light photoredox catalysis with ynamide chemistry for divergent synthesis of useful 2-benzhydrylindoles and 3-benzhydrylisoquinolines.In this Account, we describe a panoramic picture of our recent contributions since 2015 to the development and application of ynamide chemistry in organic synthesis via transition metal-catalyzed tandem reactions by focusing on the tetrafunctionalization of ynamides. These studies provide not only efficient and attractive methods for divergent synthesis of valuable N-heterocycles but also some new insights into the exploration of alkyne chemistry and metal carbene chemistry.

Abstract:Regioselective C-H alkenylation of N,N-dialkylanilines with ynamides was developed using AgNTf2 as a catalyst. This approach represents a facile hydroarylation of ynamides, allowing for the introduction of an alkenyl group exclusively at the para position of aniline derivatives. As a result, a series of 4-alkenyl N,N-dialkylanilines were synthesized with excellent regioselectivities.

Abstract:Ynamides are electron-rich alkynes with unique reactivities and act as flexible building blocks in organic synthesis. Therefore, the investigation for transformation of ynamides with exceptional selectivity and efficiency is attractive and interesting. Herein, we report an oxoarylation of ynamides with N-aryl hydroxamic acids. In the presence of catalytic Cu(OTf)2, both the terminal and internal ynamides could undergo an addition/[3,3] sigmatropic rearrangement cascade with N-aryl hydroxamic acids to achieve oxoarylation, along with providing selective entry to (ortho-amino)arylacetamides and oxindoles. Moreover, deuterium-labelling reaction and gram-scale reaction were conducted to probe the mechanism and showcase the scalability.

Abstract:Onium ylides are reactive intermediates that undergo versatile chemical transformations to give structurally interesting compounds. Rearrangement reactions of onium ylides are of great importance to synthetic organic chemists, as they provide efficient methods for C-C bond formations as well as installation of new stereogenic centers in molecules. Traditionally, onium ylides have been shown to undergo two types of rearrangements, namely, [2,3]- and [1,2]-rearrangements. In recent years, there have been tremendous developments in the field of metal-catalyzed onium ylide rearrangements through catalytic generation of ylide intermediates from diazocompounds. Several examples of selective catalytic onium ylide rearrangements involving sulfonium, oxonium, ammonium, as well as iodonium ylides have been developed over the years especially in allylic and propargylic systems. However, when the π-system that takes part in the rearrangement is part of an aromatic ring, the selectivity for rearrangements of reactive onium ylides is more challenging. In this review, we discuss recent advances in catalyst control of onium ylide rearrangements of aromatic systems.

Abstract:Amide, as a ubiquitous functional group, is essential in various aspects of chemistry and biology. Although the history of studying amide is rich and fruitful, the synthetic application of amide is very limited due to the inertness of amide C–N bond. Recently,significant advances have been achieved towards the nickel-mediated C–N activation of amides. This approach allows a facile generation of acyl-nickel intermediates, and a number of unique transformations have been designed and realized based on the amide C–N bond activation. Focused on the catalytic transformation, this review summarizes and categorizes the recent advances on the synthetic applications of Ni-mediated C–N bond activation of amides.

Abstract:Being important biological and pharmacological units, β-amino amides and β-acyloxy amides have a privileged position in both academia and industry. Developing a methods to prepare them has gained attention. Ynamides, which possess dual nucleophilic and electrophilic properties, are similar to isonitriles. Abstract:Recently, remarkable advances in the mechanistic understanding of Ni-catalyzed amide C-N bond activation have been achieved by computational chemists. These computational studies revealed the key mechanistic processes of Ni-catalyzed amide C-N bond cleavage, providing the molecular basis for the rationalization of reactivities and selectivities. This review summarizes the general mechanistic models of Ni-catalyzed amide C-N bond activation, and discusses their applications in the understanding and design of Ni-catalyzed cross coupling reactions involving amides.

Abstract:The first Ni(OTf)2-catalyzed hydroamination of ynamides 2 was developed by reacting with secondary amines (1 and 4). This protocol features excellent regioselectivity, a broad substrate scope of secondary aryl amines, and good functional group tolerance for ynamides. Using this method, a variety of substituted ethene-1,1-diamine compounds were prepared in moderate to excellent yields with high regioselectivities.

Abstract:Difunctionalization of ynamides, whether through an intermolecular approach or in an atom-economical manner, continues to pose a significant challenge. This work presents a simpler method for such unprecedented functionalization through a highly regio- and stereoselective bromoalkynylation. The developed strategy, which requires a Pd(II) catalyst and no additive, has a broad scope and high functional-group tolerance and provided access to fifty value-added -bromo ynenamides. In addition to late-stage functionalization, the synthetic potential of this method was demonstrated through rapid access to previously challenging -skeletons. A unique 1,3-alkynyl migration, which was enabled by carbopalladation, offers a platform for the development of atom-economical reactions. Experimental evidence, such as from Hammett plot analysis, XPS studies, and 13C kinetic isotope effect measurements, supported by density functional theory computations enabled a comprehensive understanding of the mechanism.

Abstract:This review summarized the methodology development on the cycloaddition of ynol ethers and thioynol ethers in every sort of catalytic cycloaddition. Comprehensive Summary Electron‐rich alkynes, such as ynol and thioynol ethers, have proven to be versatile and appealing partners in catalytic cycloaddition reactions, and thus have raised considerable attentions owing to the practical application in the modular assembly of valuable carbo‐ and heterocycles. The past decades have witnessed inspiring advances in this emerging field, and an increasing number of related discoveries have been exploited. Divided into two main sections on the basis of substrate type, in each section this comprehensive review will initially summarize their synthetic preparations and subsequently examine their reactivity in every sort of catalytic cycloaddition with emphasis on the methodology development, aimed at providing an access to this burgeoning area and encouraging further innovations in the near future. Key Scientists For the cycloaddition of ynol ethers, in 2004, Kozmin et al. firstly developed a silver‐catalyzed [2 + 2] cycloaddition of siloxy alkynes with electron‐poor olefins. In 2012, Hiyama et al. realized a palladium‐catalyzed formal [4 + 2] annulation of alkynyl aryl ethers with internal alkynes. In the same year, Sun et al. discovered an efficient [6 + 2] cyclization between siloxy alkynes and 2‐(oxetan‐3‐yl)benzaldehydes by applying HNTf2 as catalyst. In 2017, Wender et al. first utilized vinylcyclopropanes (VCPs) as coupling partners in the [5 + 2] annulation of ynol ethers. In 2018 and 2020, Ye et al. reported zinc‐catalyzed formal [3 + 2] and [4 + 3] cycloaddition, respectively. For the cycloaddition of thioynol ethers, in 2004, Hilt et al. realized a [4 + 2] cycloaddition by employing the alkynyl sulfides and acyclic 1,3‐dienes. In 2006, a ruthenium‐catalyzed [2 + 2] cycloaddition of thioynol ethers with bicyclic alkenes was accomplished by Tam. In 2014, Sun et al. reported an elegant iridium‐catalyzed click reaction of thioalkynes with azides.

Abstract:The mechanism, regioselectivity, chemoselectivity and the effects of substituents were explored by the density functional theory (DFT) in the oxidative functionalization of yndiamides catalysed by the gold(I) [IPrAuNTf2] or Brønsted acid (HNTf2) systems. The calculations revealed that both catalysts entail a similar type of nucleophilic attack on the catalyst-ligated yndiamide, forming a vinyl intermediate that can isomerize to gold carbene or carbocation upon the cleavage of the N−O bond. Next, the gold carbene or carbocation are easily captured by external nucleophilic reagents and finally desaturated via a proton shift, assisted by a substrate or NTf2− to yield the final product. The unique features of regio- and chemoselectivity were explored in detail through the distortion/interaction and global reactivity index (GRI) analyses. The effects of substituents in yndiamides were also studied. Apart from completely rationalizing the experimental data and intricate selectivity patterns, the obtained theoretical evidence can provide a significant contribution toward understanding the resembling types of reaction mechanisms

Abstract:AbstractA palladium‐catalyzed oxygenative cross‐coupling of ynamides and benzyl bromides has been developed. After subsequent hydrogenation, α,α‐disubstituted amide derivatives were obtained in good yields. Migratory insertion of α‐oxo palladium carbene species, generated by intermolecular oxidation, is proposed as the key step in this reaction. The study demonstrates the potential of ynamides to serve as carbene precursors in palladium‐catalyzed C−C bond‐forming cross‐coupling reactions.

Abstract:Herein, we describe experimental and computational studies to understand the features of oxycyanopyridinium ylides generated in situ from oxy-cyanopyridines and rhodium carbene. This chemistry has enabled the concomitant formation of both C–N and C–C bonds, providing a complementary approach for cyanation reactions. Density functional theory calculations indicate the sequential metal-bound ylide formation, rhodium-associated five-membered transition state, and 1,4-cyano group relocation. Moreover, the enantioselective rearrangement has been realized by using chiral dirhodium complexes as the catalysts.

Abstract:In this review, we focus on the summry of the development of NHC‐activation of carbonyl carbon (or imine carbon) in situ, α‐, β‐, γ‐, and beyond, and the cycloaddition reaction of these species. Over the recent decades, due to the special electronic characteristics and diverse reactivities, N‐heterocyclic carbene (NHC) has received significant interest in organocatalyzed reactions. The formation of Breslow intermediates by NHC can convert into acyl anion equivalent, enolates, homoenolate, acyl azolium, and vinyl enolate etc., and the cycloaddition reactions of these species has attracted lots of attention. In this review, we focus on the summry of the development of NHC‐activation of carbonyl carbon (or imine carbon) in situ, α‐, β‐, γ‐, and beyond, and the cycloaddition reaction of these species.
