Abstract:Abstract A Rh-catalyzed electrochemical vinylic C–H annulation of acrylamides with alkynes has been developed in an undivided cell, affording cyclic products in good to excellent yield. Divergent syntheses of α-pyridones and cyclic imidates are accomplished by employing N-phenyl acrylamides and N-tosyl acrylamides as substrates, respectively. Additionally, excellent regioselectivities are achieved when using unsymmetrical alkynes. This electrochemical process is environmentally benign compared to traditional transition metal-catalyzed C–H annulations because it avoids the use of stoichiometric metal oxidants. DFT calculations elucidated the reaction mechanism and origins of substituent-controlled chemoselectivity. The sequential C–H activation and alkyne insertion under rhodium catalysis leads to the seven-membered ring vinyl-rhodium intermediate. This intermediate undergoes either the classic neutral concerted reductive elimination to produce α-pyridones, or the ionic stepwise pathway to produce cyclic imidates.

Abstract:Novel divergent domino annulation reactions of sulfur ylides with aryldiazonium tetrafluoroborates have been developed, affording various tetra- and trisubstituted pyrazole derivatives in moderate to good yields. Three molecules of sulfur ylides were applied as C1 synthon to construct the complex products with five new chemical bonds formed in these one-pot reactions.

Abstract:An efficient approach for the reversal of regioselectivity in the nucleophilic introduction of difluorinated carbanion into α,β-enones has been developed via a silylium catalysis. The strong electron-withdrawing properties and bulky substituents of in situ-generated silyl triflic imide catalyst is the key for the 1,4-addition reaction to proceed smoothly. The synthetic utility is highlighted by the further use of this method for the synthesis of 2,4,6-triarylsubstituted 3-fluoropyridines in a one-pot manner.

Abstract:A catalyst‐based switchable regioselective C−H activation/annulation of acrylamides with propargyl carbonates has been developed, delivering C5 or C6 alkenyl substituted 2‐pyridones. This robust protocol proceeds with a broad substrate scope and good functional group tolerance under redox‐neutral reaction conditions. More significantly, this reaction is highly effective with previously challenging unsymmetrical alkynes, including unbiased alkyl‐alkyl substituted alkynes, with perfect and switchable regioselectivity. Additionally, mechanistic studies and DFT calculations were performed to shed light on the switchable regioselectivity.

Abstract:Lithium iodide enables regioconvergent C-F bond functionalization of isomeric Morita-Baylis-Hillman fluorides with carbon, sulfur, and nitrogen nucleophiles. The defluorinative carbon-carbon and carbon-heteroatom bond formations give multifunctional compounds in excellent yields and with good to high diastereoselectivities at room temperature. The possibility of catalytic enantioselective allylation is also discussed.

Abstract:The development of general catalytic methods for the regio- and stereoselective construction of chiral N-heterocycles in a diversity-oriented fashion remains a formidable challenge in organic synthesis. N-heterocyclic carbene (NHC) catalysis has been shown to produce a variety of outcomes, but control of the reactivity has rarely been demonstrated. Here we report a switchable catalytic activation of enals with aromatic azomethine imines that provides high selectivity using NHC organocatalysts. The original selectivity corresponds to the acidity of the base used in the reaction. The catalytically generated chiral homoenolate or enol intermediate undergoes enantioselective annulation with electrophiles such as N-iminoquinolinium ylides, N-iminoisoquinolinium ylides and β-N-iminocarboline ylides. The good-to-high overall yields, high regioselectivities and excellent enantioselectivities observed are controlled by the catalyst and reaction conditions.

Abstract:Presented herein is a palladium-catalyzed asymmetric (3 + 2) annulation reaction between 1,3-dienes and 2-formylarylboronic acids, proceeding in a cascade vinylogous addition and Suzuki coupling process. Both electron-neutral and electron-deficient 1,3-dienes are compatible under similar catalytic conditions, and distinct regioselectivity is observed via functional-group control of 1,3-diene substrates. A collection of 1-indanols with dense functionalities is constructed stereoselectively.

Abstract:Gold-catalyzed difunctionalizations of o-alkynylbenzenesulfonamides with aryldiazonium salts are reported herein. Upon irradiation with the blue LEDs, benzosultam products were formed via aminoarylation accompanied by the release of N2. Without irradiation, aryldiazonium salts were engaged as efficient electrophiles, facilitating electrophilic deaurations of the vinyl-Au(I) intermediates, followed by tautomerization to give the N-aryl-substituted α-imino (E)-hydrazones. This protocol offers a streamlined synthesis of a wide range of stereospecific and densely functionalized difluoromethyl and trifluoromethyl cyclopropane-1,1-dicarbonitriles. Further functional group interconversions and/or skeletal elaborations afford structurally distinct cyclopropyl variants.

Abstract:An amine-catalyzed [4 + 2] annulation of Morita-Baylis-Hillman allylic acetates 2 with electron-deficient alkenes or diazenes has been developed for efficient syntheses of highly functionalized cyclohexenes, tetrahydropyridazines, and important spirocycles. This reaction unveils a new reactivity pattern of the intensely studied allylic compounds 2 acting as a C(4) synthon in Lewis base catalyzed annulation reactions and also showcases divergent catalysis between tertiary amines and phosphines.

Abstract:An approach for reliable and predictable carbon-carbon or carbon-heteroatom bond formation that produces either regioisomer starting from the same raw materials, also known as a regiodivergent methodology, is highly desirable. Altering the chemical reaction variables, including catalysts, ligands, solvents, or other additives, were the predominant strategy for the implementation of regiodivergent features in metal-catalysed organic synthesis. The achievement of switchable selectivity using quickly and conveniently controlled physical variables constitutes a desirable goal with an intriguing method of attainment. Herein, we report our discovery that temperature-regulated switchable site-selectivity can be achieved in alkene (migratory) functionalization. Judicious selection of reaction temperatures, one of the most easily changed variables, led to protocols that provide the regiodivergent alkylation products starting from a single alkene substrate. This protocol allows for the convenient synthesis of α- and β-branched protected amines, both of which are substantially important to the pharmaceutical chemistry and biochemistry fields. In addition, enantioenriched β-branched alkyl amines could be accessed in a catalytic asymmetric variant manner. This work may inspire more research interests in metal-catalysed regiodivergent reaction discovery using easily changed physical variables.

Abstract:Chemoselective annulations of phosphoryl carbenes generated from diazo(aryl)methyl(diaryl)phosphine oxides with pyridinium 1,4-zwitterionic thiolates were performed under microwave irradiation, affording 1-diarylphosphoryl-1H-benzo[c]thiopyran derivatives via [3+3] annulation and indolizine derivatives via ([1+5]-1) annulation with P-Cope elimination as the key step. The annuloselectivity was controlled by the steric hindrance of pyridiniums in pyridinium 1,4-zwitterionic thiolates.

Abstract:The control of linear/branched selectivity is one of the major focuses in transition-metal catalyzed allyl-allyl cross-coupling reactions, in which bond connection occurs at the terminal site of both the allyl fragments forming different types of 1,5-dienes. Herein, terminal/internal regioselectivity is investigated and found to be switchable in allyl-allyl cross-coupling reactions between gem-difluorinated cyclopropanes and allylboronates. The controlled terminal/internal regioselectivity arises from the fine-tuning of the rhodium catalytic system. Fluorinated 1,3-dienes, 1,4-dienes and 1,5-dienes are therefore produced in good yields with respectively isomerized terminal, internal, and terminal regioselectivity.

Abstract:The transition-metal-catalyzed alkenylation strategy of propargylic alcohol derivatives provides an efficient protocol to access multifunctional products in a double-nucleophilic attack pattern. While limited relevant asymmetric examples have been reported via palladium catalysis, here we first demonstrate that a nonprecious Ni(0)-based chiral complex can efficiently promote the tandem substitution process between propargylic carbonates and N-trifluoroethyl ketimines via consecutive aza-vinylogous activations, finally accomplishing a (3 + 2) annulation reaction to afford products embedding a 4-methylene-3,4-dihydro-2H-pyrrole framework with high regio-, diastereo-, and enantiocontrol. Their assemblies with a few all-carbon-based vinylogous precursors are also successful, and enantioenriched adducts containing a 3-methylenecyclopentene scaffold are furnished effectively. The substitution patterns for both types of substrates are substantial, and an array of synthetic elaborations is conducted to deliver more versatile architectures with high application potential. In addition, density functional theory calculations and control experiments have been conducted to rationalize the catalytic pathways and regio- and enantioselectivity control.

Abstract:Herein, an efficient, substrate-directed divergent [2+3]/[2+1] annulation of tetra-substituted oxa-dienes and allylic sulfur ylides has been successfully developed. Under precise annulation regulation, a series of functionalized bispirocyclopentane and bispirocyclopropane derivatives were synthesized in a highly stereoselective and economical manner. This protocol offers the advantages of mild conditions, high chemo-, regio- and diastereoselectivity and broad substrate compatibility.

Abstract:A diastereoselective and enantioselective formal [4+1] ylide annulation of chiral sulfonium salts with various substituted Morita-Baylis-Hillman (MBH) adducts leading to optically active isoxazoline N-oxides with three chiral carbon centers has been explored. The salient features of this methodology include a high diastereo- and enantioselective transformation, easily accessible starting materials, and mild reaction conditions. In addition, the isoxazoline N-oxides products can be conveniently transformed into functionalized compounds.
