Abstract:Ethanol consumption has been shown to have many deleterious effects, including behavioral alterations, motor deficits, reduction in inhibition, and alteration of neurochemical expression. These effects occur in the wide variety of species that consume ethanol. Although studies have examined aversive conditioning in honey bees (Apis mellifera), few have examined the role of intoxication on the acquisition of learning in such paradigms. The current study continues a line of research using honey bees as a model to explore the behavioral effects of ethanol toxicity. A passive avoidance task is used to explore how increasing dosages of ethanol affects the ability of honey bees to perform this task. The results show that honey bees exposed to higher concentrations of ethanol have slower passive avoidance acquisition than bees exposed to lower concentrations under the same conditions. Abstract:Background: This study examines the effect of ethanol (EtOH) on continuous reinforcement schedules in the free-flying honeybee (Apis mellifera L.). As fermented nectars may be encountered naturally in the environment, we designed an experiment combining the tools of laboratory research with minimal disturbance to the natural life of honeybees. Methods: Twenty-five honeybees were trained to fly from their colonies to a fully automated operant chamber with head poking as the operant response. Load size, intervisit interval, and interresponse times (IRTs) served as the dependent variables and were monitored over the course of a daily training session consisting of many visits. Experimental bees were tested using an ABA design in which sucrose only was administered during condition A and a 5% EtOH sucrose solution was administered during condition B. Control bees received sucrose solution only. Results: Most bees continued to forage after EtOH introduction. EtOH significantly reduced the load size and the intervisit interval with no significant effect on IRTs. However, a look on individual data shows large individual differences suggesting the existence of different kinds of behavioral phenotypes linked to EtOH consumption and effects. Conclusions: Our results contribute to the study of EtOH consumption as a normal phenomenon in an ecological context and open the door to schedule-controlled drug self-administration studies in honeybees.

Abstract:Experiments are designed to assess whether free-flying honey bees have an aversion to an ethanol solution when given a choice between targets containing an ethanol solution in sucrose or sucrose only. Animals given a choice between a 1% ethanol solution and sucrose only show no aversion to the ethanol solution either in acquisition or extinction. Honey bees given a choice between a 5% ethanol solution and sucrose only show no differences in the initial choice of targets but some ees do switch over to the sucrose-only target. Performance during extinction indicates that bees landed on the previously reinforced sucrose-only target more than the target previously containing the 5% ethanol solution. An experiment in which bees were given a single 5%, ethanol target showed that of 20 bees, 11 returned for the entire 12 trials of the experiment. All bees returned at least 6 times to the 5% ethanol target. Additional experiments were run on harnessed foragers in a palatability study of alcoholic beverages consumed by humans. The results of the palatability experiment indicate that in general, bees prefer more sweet drinks with less alcohol.

Abstract:Conditioned taste aversion (CTA) learning induces the devaluation of a preferred food through its pairing with a stimulus inducing internal illness. In invertebrates, it is still unclear how this aversive learning impairs the memories of stimuli that had been associated with the appetitive food prior to its devaluation. Here we studied this phenomenon in the honey bee and characterized its neural underpinnings. We first trained bees to associate an odorant (conditioned stimulus, CS) with appetitive fructose solution (unconditioned stimulus, US) using a Pavlovian olfactory conditioning. We then subjected the bees that learned the association to a CTA training during which the antennal taste of fructose solution was contingent or not to the ingestion of quinine solution, which induces malaise a few hours after ingestion. Only the group experiencing contingent fructose stimulation and quinine-based malaise exhibited a decrease in responses to the fructose and a concomitant decrease in odor-specific retention in tests performed 23 h after the original odor conditioning. Furthermore, injection of dopamine- and serotonin-receptor antagonists after CTA learning revealed that this long-term decrease was mediated by serotonergic signaling as its blockade rescued both the responses to fructose and the odor-specific memory 23 h after conditioning. The impairment of a prior CS memory by subsequent CTA conditioning confirms that bees retrieve a devaluated US representation when presented with the CS. Our findings further highlight the importance of serotonergic signaling in aversive learning in the bee and uncover mechanisms underlying aversive memories induced by internal illness in invertebrates.

Abstract:The ability of Antabuse (disulfiram) to influence ethanol consumption and learning in harnessed honey bees was investigated. In the first series of experiments a factorial design was used with 5 levels of ethanol concentration (0%, 1%, 5%, 10%, 20%), 4 doses of Antabuse (0, 37 microg/g, 3.7 microg/g, .37 microg/g), and 2 testing intervals (1 min., 10 min.). Animals were fed a single 1 microl dose of Antabuse and contact time with an ethanol solution measured. A second series of experiments investigated the influence of Antabuse on the formation of Pavlovian conditioning of the proboscis extension reflex. A factorial design was used with two levels of training (paired, unpaired), three levels of ethanol (0%, 1%, 5%), and 2 levels of pretreatment (distilled water, 3.7 microg/g). Analysis of the consumption experiments indicate that pretreatment with Antabuse reduces ethanol intake, although there was substantial variability. The findings of the Pavlovian experiments suggest that pretreatment with Antabuse significantly reduced responding to a CS signaling the availability of ethanol.

Abstract:This paper assumes that a neo‐Hookean matrix with neo‐Hookean fibres is representative of soft tissue. Under this assumption, a unit cell model is proposed to investigate the fibre–matrix interfacial stress field for biological soft tissue under biaxial loadings. In this unit cell model, the soft tissue is treated as a composite where the matrix is unidirectionally reinforced with a single family of aligned fibres. The results are compared with the model of Guo et al., which accounts for the fibre–matrix interfacial stress field, and Qiu and Pence's model, which does not proceed from the assumption that the fibres are themselves neo‐Hookean. It is found that the stress representative of the fibre–matrix interface plays an important role in the deformation of the composite, and the model of Guo et al. underestimates this stress under large biaxial deformation. Abstract:Background A strong relationship exists between individual sensitivity to the aversive properties of ethanol and risk for alcohol use disorder (AUD). Despite this, our understanding of the neurobiological mechanisms underlying the subjective response to ethanol is limited. A major contributor to this lack of knowledge is the absence of preclinical models that enable exploration of this individual variability such as is possible in studies of humans. Methods Adult male and female Long‐Evans rats were trained to associate a novel tastant (saccharin) with acute exposure to either saline or ethanol (1.5 g/kg or 2.0 g/kg i.p.) over three conditioning days using a standard conditioned taste aversion (CTA) procedure. Variability in sensitivity to ethanol‐induced CTA was phenotypically characterized using a median split across the populations studied. Results When examining group averages, both male and female rats exposed to saccharin paired with either dose of ethanol exhibited lower saccharin intake relative to saline controls indicative of ethanol‐induced CTA. Examination of individual data revealed a bimodal distribution of responses uncovering two distinct phenotypes present in both sexes. CTA‐sensitive rats exhibited a rapid and progressive reduction in saccharin intake with each successive ethanol pairing. In contrast, saccharin intake was unchanged or maintained after an initial decrease from baseline levels in CTA‐resistant rats. While CTA magnitude was similar between male and female CTA‐sensitive rats, among CTA‐resistant animals females were more resistant to the development of ethanol‐induced CTA than males. Phenotypic differences were not driven by differences in baseline saccharin intake. Conclusions These data parallel work in humans by revealing individual differences in sensitivity to the aversive properties of ethanol that emerge immediately after initial exposure to ethanol in both sexes. This model can be used in future studies to investigate the neurobiological mechanisms that confer risk for AUD.

Abstract:Foraging behavior was evaluated in honeybees trained to fly to a feeder containing sucrose only, 1% ethanol, 5% ethanol, or 10% ethanol. The results indicated that exposure to ethanol disrupted several types of honeybee social behavior within the hive. Consumption of ethanol at the feeding site reduced waggle dance activity in foraging bees and increased occurrence of tremble dance, food exchange, and self-cleaning behavior. These ethanol-induced changes in behavior may reflect effects on the central nervous system similar to the previously observed effects of food poisoning with sublethal doses of insecticides.

Abstract:Experiments were designed to determine whether ethanol influenced aggression in honey bees. Two experiments are reported. In Exp. 1, harnessed honey bees were fed a 1%, 5%, 10%, or 20% ethanol solution. Two control groups received either a sucrose solution only or no pretreatment, respectively. The dependent variable was the number of sting extensions over 10 min. Analysis showed that aggression in harnessed bees was not influenced by prior ethanol consumption. Because there was some suspicion that the extension of the sting apparatus may be hindered by harnessing, and the authors wanted to use a design that increased ecological validity, Exp. 2 was conducted with free-flying bees. Sucrose or 20% ethanol solutions were placed in front of beehives, and the number of stings on a leather patch dangled in front of the hive served as the dependent variable. The experiment was terminated after 5 hr. because bees exposed to ethanol became dangerously aggressive. A unique aspect of the study was that Africanized honey bees were used.
Discontinued alcohol consumption elicits withdrawal symptoms in honeybees

Abstract:The honeybee continues to be developed as a model species in many research areas, including studies related to the effects of alcohol. Here, we investigate whether workers display one of the key features of alcoholism, namely withdrawal symptoms. We show that workers fed for a prolonged time on food spiked with ethanol, after discontinuation of access to such food, exhibited a marked increase in the consumption of ethanol and a slight increase in mortality. We additionally show that withdrawal symptoms do not include an increase in appetitiveness of ethanol diluted in water. Our results demonstrate that workers can develop alcohol dependence, which might be especially important in the natural setting of repeated exposure to ethanol in floral nectar and for their potential as a model of alcohol addiction.

Abstract:Blunted sensitivity to ethanol’s aversive effects can increase motivation to consume ethanol; yet, the neurobiological circuits responsible for encoding these aversive properties are not fully understood. Plasticity in cells projecting from the insular cortex (IC) to the basolateral amygdala (BLA) is critical for taste aversion learning and retrieval, suggesting this circuit’s potential involvement in modulating the aversive properties of ethanol. Here, we tested the hypothesis that GABAergic activity onto IC-BLA projections would be facilitated following the retrieval of an ethanol-conditioned taste aversion (CTA). Consistent with this hypothesis, frequency of mIPSCs was increased following retrieval of an ethanol-CTA across cell layers in IC-BLA projection neurons. This increase in GABAergic plasticity occurred in both a circuit-specific and learning-dependent manner. Additionally, local inhibitory inputs onto layer 2/3 IC-BLA projection neurons were greater in number and strength following ethanol-CTA. Finally, DREADD-mediated inhibition of IC parvalbumin-expressing cells blunted the retrieval of ethanol-CTA in male, but not female, mice. Collectively, this work implicates a circuit-specific and learning-dependent increase in GABAergic tone following retrieval of an ethanol-CTA, thereby advancing our understanding of how the aversive effects of ethanol are encoded in the brain.

Abstract:Conditioned taste aversion (CTA) is a well established learning and memory paradigm in rats and mice that is considered to be a special form of classical conditioning. Rodents--as well as many other species including man--learn to associate a novel taste (CS) with nausea (US), and as a consequence avoid drinking fluid with this specific taste. In contrast to other types of classical conditioning, even CS-US intervals lasting several hours lead to an aversion to the gustatory CS. With increasing CS-US delay duration, however, the aversion against the CS gradually decreases. Mice differ from rats in their reaction to the CS as well as the US. They tolerate a much higher concentration of saccharin and they do not show any clear signs of nausea when injected with the US. Advantages of this task are its relative independence of motor behavior, well described pathways for the CS and partly the US, and the wealth of available anatomical and pharmacological data implying several brain structures (e.g. parabrachial nucleus, amygdala, insular cortex), neurotransmitters and their receptors (e.g. cholinergic system, NMDA-receptors), and cellular processes (e.g. expression of immediate early genes, Ras-MAP kinase signaling pathway, CREB phosphorilation, protein tyrosine phosphorilation, protein synthesis) in CTA. The CTA paradigm has also been successfully used to phenotype mouse mutants.

Nevertheless, although conditioned inhibition has been reported, it has yet to be thoroughly investigated in invertebrates. Here, we evaluate the role of the US in producing conditioned inhibition by using proboscis extension response conditioning of the honeybee (Apis mellifera). Specifically, using variations of a "feature-negative" experimental design, we use downshifts in US intensity relative to US intensity used during initial excitatory conditioning to show that an odorant in an odor–odor mixture can become a conditioned inhibitor. We argue that some alternative interpretations to conditioned inhibition are unlikely. However, we show variation across individuals in how strongly they show conditioned inhibition, with some individuals possibly revealing a different means of learning about changes in reinforcement. We discuss how the resolution of these differences is needed to fully understand whether and how conditioned inhibition is manifested in the honeybee, and whether it can be extended to investigate how it is encoded in the CNS. It is also important for extension to other insect models. In particular, work like this will be important as more is revealed of the complexity of the insect brain from connectome projects.
