Abstract:Experiments are designed to assess whether free-flying honey bees have an aversion to an ethanol solution when given a choice between targets containing an ethanol solution in sucrose or sucrose only. Animals given a choice between a 1% ethanol solution and sucrose only show no aversion to the ethanol solution either in acquisition or extinction. Honey bees given a choice between a 5% ethanol solution and sucrose only show no differences in the initial choice of targets but some ees do switch over to the sucrose-only target. Performance during extinction indicates that bees landed on the previously reinforced sucrose-only target more than the target previously containing the 5% ethanol solution. An experiment in which bees were given a single 5%, ethanol target showed that of 20 bees, 11 returned for the entire 12 trials of the experiment. All bees returned at least 6 times to the 5% ethanol target. Additional experiments were run on harnessed foragers in a palatability study of alcoholic beverages consumed by humans. The results of the palatability experiment indicate that in general, bees prefer more sweet drinks with less alcohol.

Abstract:Experiments were designed to determine whether ethanol influenced aggression in honey bees. Two experiments are reported. In Exp. 1, harnessed honey bees were fed a 1%, 5%, 10%, or 20% ethanol solution. Two control groups received either a sucrose solution only or no pretreatment, respectively. The dependent variable was the number of sting extensions over 10 min. Analysis showed that aggression in harnessed bees was not influenced by prior ethanol consumption. Because there was some suspicion that the extension of the sting apparatus may be hindered by harnessing, and the authors wanted to use a design that increased ecological validity, Exp. 2 was conducted with free-flying bees. Sucrose or 20% ethanol solutions were placed in front of beehives, and the number of stings on a leather patch dangled in front of the hive served as the dependent variable. The experiment was terminated after 5 hr. because bees exposed to ethanol became dangerously aggressive. A unique aspect of the study was that Africanized honey bees were used.

Abstract:Background: This study examines the effect of ethanol (EtOH) on continuous reinforcement schedules in the free-flying honeybee (Apis mellifera L.). As fermented nectars may be encountered naturally in the environment, we designed an experiment combining the tools of laboratory research with minimal disturbance to the natural life of honeybees. Methods: Twenty-five honeybees were trained to fly from their colonies to a fully automated operant chamber with head poking as the operant response. Load size, intervisit interval, and interresponse times (IRTs) served as the dependent variables and were monitored over the course of a daily training session consisting of many visits. Experimental bees were tested using an ABA design in which sucrose only was administered during condition A and a 5% EtOH sucrose solution was administered during condition B. Control bees received sucrose solution only. Results: Most bees continued to forage after EtOH introduction. EtOH significantly reduced the load size and the intervisit interval with no significant effect on IRTs. However, a look on individual data shows large individual differences suggesting the existence of different kinds of behavioral phenotypes linked to EtOH consumption and effects. Conclusions: Our results contribute to the study of EtOH consumption as a normal phenomenon in an ecological context and open the door to schedule-controlled drug self-administration studies in honeybees.

Abstract:The ability of Antabuse (disulfiram) to influence ethanol consumption and learning in harnessed honey bees was investigated. In the first series of experiments a factorial design was used with 5 levels of ethanol concentration (0%, 1%, 5%, 10%, 20%), 4 doses of Antabuse (0, 37 microg/g, 3.7 microg/g, .37 microg/g), and 2 testing intervals (1 min., 10 min.). Animals were fed a single 1 microl dose of Antabuse and contact time with an ethanol solution measured. A second series of experiments investigated the influence of Antabuse on the formation of Pavlovian conditioning of the proboscis extension reflex. A factorial design was used with two levels of training (paired, unpaired), three levels of ethanol (0%, 1%, 5%), and 2 levels of pretreatment (distilled water, 3.7 microg/g). Analysis of the consumption experiments indicate that pretreatment with Antabuse reduces ethanol intake, although there was substantial variability. The findings of the Pavlovian experiments suggest that pretreatment with Antabuse significantly reduced responding to a CS signaling the availability of ethanol.

Abstract:Foraging behavior was evaluated in honeybees trained to fly to a feeder containing sucrose only, 1% ethanol, 5% ethanol, or 10% ethanol. The results indicated that exposure to ethanol disrupted several types of honeybee social behavior within the hive. Consumption of ethanol at the feeding site reduced waggle dance activity in foraging bees and increased occurrence of tremble dance, food exchange, and self-cleaning behavior. These ethanol-induced changes in behavior may reflect effects on the central nervous system similar to the previously observed effects of food poisoning with sublethal doses of insecticides.

Abstract:This paper assumes that a neo‐Hookean matrix with neo‐Hookean fibres is representative of soft tissue. Under this assumption, a unit cell model is proposed to investigate the fibre–matrix interfacial stress field for biological soft tissue under biaxial loadings. In this unit cell model, the soft tissue is treated as a composite where the matrix is unidirectionally reinforced with a single family of aligned fibres. The results are compared with the model of Guo et al., which accounts for the fibre–matrix interfacial stress field, and Qiu and Pence's model, which does not proceed from the assumption that the fibres are themselves neo‐Hookean. It is found that the stress representative of the fibre–matrix interface plays an important role in the deformation of the composite, and the model of Guo et al. underestimates this stress under large biaxial deformation. Abstract:Ethanol consumption has been shown to have many deleterious effects, including behavioral alterations, motor deficits, reduction in inhibition, and alteration of neurochemical expression. These effects occur in the wide variety of species that consume ethanol. Although studies have examined aversive conditioning in honey bees (Apis mellifera), few have examined the role of intoxication on the acquisition of learning in such paradigms. The current study continues a line of research using honey bees as a model to explore the behavioral effects of ethanol toxicity. A passive avoidance task is used to explore how increasing dosages of ethanol affects the ability of honey bees to perform this task. The results show that honey bees exposed to higher concentrations of ethanol have slower passive avoidance acquisition than bees exposed to lower concentrations under the same conditions. Abstract:Ethanol consumption produces characteristic behavioral states in animals that include sedation, disorientation, and disruption of motor function. Using individual honey bees, we assessed the effects of ethanol ingestion on motor function via continuous observations of their behavior. Consumption of 1 M sucrose solutions containing a range of ethanol doses led to hemolymph ethanol levels of approximately 40-100 mM. Using ethanol doses in this range, we observed time and dose-dependent effects of ethanol on the percent of time our subjects spent walking, stopped, or upside down, and on the duration and frequency of bouts of behavior. The effects on grooming and flying behavior were more complex. Behavioral recovery from ethanol treatment was both time and ethanol dose dependent, occurring between 12 and 24 h post-ingestion for low doses and at 24-48 h for higher doses. Furthermore, the amount of ethanol measured in honey bee hemolymph appeared to correlate with recovery. We predict that the honey bee will prove to be an excellent model system for studying the influence of ethanol on the neural mechanisms underlying behavior.
Tolerance and efficient metabolization of extremely high ethanol concentrations by a social wasp

Abstract:Ethanol, a natural by-product of sugar fermentation, can be found in various fruits and nectar. Although many animals routinely consume ethanol in low concentrations as part of their natural diets, its inherent toxicity can cause severe damage. Even species particularly well adapted to ethanol consumption face detrimental effects when exposed to concentrations above 4%. Here, we investigated the metabolism of ethanol and its impact on survival and behavior in the Oriental hornet (Vespa orientalis), a social wasp that naturally consumes ethanol. We show that chronic ethanol consumption, even at concentrations as high as 80%, had no impact on hornet mortality, construction behavior, or agonistic behavior. Using 13C1 labeled ethanol, we show that hornets efficiently metabolized ingested ethanol and at a much higher rate than honey bees. The presence of multiple copies of the alcohol dehydrogenase (NADP+) gene in the Vespa genera suggests a potential mechanism for ethanol tolerance. These findings support the hypothesis that the mutualistic relationship between ethanol-producing organisms and vespid hosts may be at the origin of their remarkable capacity to utilize and metabolize ethanol.

Abstract:Animal models have been essential for advancing research of fetal alcohol spectrum disorder (FASD) in humans, but few animal species effectively replicate the behavioural and clinical signs of FASD. The honey bee (Apis mellifera) is a previously unexplored research model for FASD that offers the distinct benefit of highly social behaviour. In this study, we chronically exposed honey bee larvae to incremental concentrations of 0, 3, 6, and 10% ethanol in the larval diet using an in vitro rearing protocol and measured developmental time and survival to adult eclosion, as well as body weight and motor activity of newly emerged adult bees. Larvae reared on 6 and 10% dietary ethanol demonstrated significant, dose-responsive delays to pupation and decreased survival and adult body weight. All ethanol-reared adults showed significantly decreased motor activity. These results suggest that honey bees may be a suitable social animal model for future FASD research.

Abstract:Physiological state profoundly influences the expression of the behaviour of individuals and can affect social interactions between animals. How physiological state influences food sharing and social behaviour in social insects is poorly understood. Here, we examined the social interactions and food sharing behaviour of honeybees with the aim of developing the honeybee as a model for understanding how an individual's state influences its social interactions. The state of individual honeybees was manipulated by either starving donor bees or feeding them sucrose or low doses of ethanol to examine how a change in hunger or inebriation state affected the social behaviours exhibited by two closely-related nestmates. Using a lab-based assay for measuring individual motor behaviour and social behaviour, we found that behaviours such as antennation, willingness to engage in trophallaxis, and mandible opening were affected by both hunger and ethanol intoxication. Inebriated bees were more likely to exhibit mandible opening, which may represent a form of aggression, than bees fed sucrose alone. However, intoxicated bees were as willing to engage in trophallaxis as the sucrose-fed bees. The effects of ethanol on social behaviors were dose-dependent, with higher doses of ethanol producing larger effects on behaviour. Hungry donor bees, on the other hand, were more likely to engage in begging for food and less likely to antennate and to display mandible opening. We also found that when nestmates received food from donors previously fed ethanol, they began to display evidence of inebriation, indicating that ethanol can be retained in the crop for several hours and that it can be transferred between honeybee nestmates during trophallaxis.

Abstract:Flying social insects can provide model systems for in-flight interactions in computationally-constrained aerial robot swarms. The social interactions in flying insects may be chemically modulated and quantified via recent measurement advancements able to simultaneously make precise measurements of insect wing and body motions. This paper presents the first in-flight quantitative measurements of ethanol-exposed honey bee body and wing kinematics in archival literature. Four high-speed cameras (9000 frames/sec) were used to record the wing and body motions of flying insects (Apis mellifera) and automated analysis was used to extract 9000 frame/sec measurements of honey bees' wing and body motions through data association, hull reconstruction, and segmentation. The kinematic changes induced by exposure to incremental ethanol concentrations from 0% to 5% were studied using statistical analysis tools. Analysis considered trial-wise mean and maximum values and gross wingstroke parameters, and tested deviations for statistical significance using Welch's t-test and Cohen's d test. The results indicate a decrease in maximal heading and pitch rates of the body, and that roll rate is affected at high concentrations (5%). The wingstroke effects include a stroke frequency decrease and stroke amplitude increase for 2.5% or greater concentrations, gradual stroke inclination angle increase up to 2.5% concentration, and a more planar wingstroke with increasing concentration according to bulk wingstroke analysis. These ethanol-exposure effects provide a basis to separate ethanol exposure and neighbor effects in chemically mediated interaction studies.

Abstract:Previous studies on various organisms have suggested that low doses of ethanol can have stimulatory effects, while higher doses may lead to toxicity, a response known as hormesis. Low ethanol concentrations occur naturally in the environment, particularly in fermenting fruits and flower nectar, where pollinators such as honey bees may encounter it. This study aimed to investigate the potential hormetic effects of low-level ethanol consumption on honey bees. Bees were divided into three groups: one provided with only sucrose solution, one both with sucrose and 0.5% ethanol in sucrose, and one with only 1% ethanol in sucrose. The bees were exposed to these diets for 14 days, and their performance was assessed through survivorship, flight endurance, body mass, lipid content, and trehalose and ethanol levels in the haemolymph. The results showed no significant differences in most parameters between the groups. However, bees constantly exposed to 1% ethanol had slightly higher trehalose levels compared to the control group, suggesting a possible adaptive response to ethanol exposure. Ethanol levels in the haemolymph differed significantly between groups, with bees exposed to ethanol showing its detectable levels in their system. While no clear hormetic effects were observed in terms of improved performance, the elevated trehalose levels in bees constantly exposed to 1% ethanol may indicate adaptations protecting from ethanol-induced damage. The study provides insights into how honey bees tolerate low-level ethanol exposure and highlights the need for further research on the ecological implications of ethanol consumption in pollinators.

Abstract:The honeybee continues to be developed as a model species in many research areas, including studies related to the effects of alcohol. Here, we investigate whether workers display one of the key features of alcoholism, namely withdrawal symptoms. We show that workers fed for a prolonged time on food spiked with ethanol, after discontinuation of access to such food, exhibited a marked increase in the consumption of ethanol and a slight increase in mortality. We additionally show that withdrawal symptoms do not include an increase in appetitiveness of ethanol diluted in water. Our results demonstrate that workers can develop alcohol dependence, which might be especially important in the natural setting of repeated exposure to ethanol in floral nectar and for their potential as a model of alcohol addiction.

Abstract:Honey bees (Apis mellifera) are one of the most crucial pollinators, providing vital ecosystem services. Their development and functioning depend on essential nutrients and substances found in the environment. While collecting nectar as a vital carbohydrate source, bees routinely encounter low doses of ethanol from yeast fermentation. Yet, the effects of repeated ethanol exposure on bees' survival and physiology remain poorly understood. Here, we investigate the impacts of constant and occasional consumption of food spiked with 1% ethanol on honey bee mortality and alcohol dehydrogenase (ADH) activity. This ethanol concentration might be tentatively judged close to that in natural conditions. We conducted an experiment in which bees were exposed to three types of long-term diets: constant sugar solution (control group that simulated conditions of no access to ethanol), sugar solution spiked with ethanol every third day (that simulated occasional, infrequent exposure to ethanol) and daily ethanol consumption (simulating constant, routine exposure to ethanol). The results revealed that both constant and occasional ethanol consumption increased the mortality of bees, but only after several days. These mortality rates rose with the frequency of ethanol intake. The ADH activity remained similar in bees from all groups. Our findings indicate that exposure of bees to ethanol carries harmful effects that accumulate over time. Further research is needed to pinpoint the exact ethanol doses ingested with food and exposure frequency in bees in natural conditions.
