Abstract:A one-pot procedure has been developed to prepare the derivatives of 2-arylimidazo(2, 1-a)-isoquinolines (2a similar to 2b), 2-arylimidazo(1, 2-a) quinolines (4a similar to 4b) and 2-arylimidazo(1, 2-a) phthalazines (6a similar to 6c) in moderate yields (51% similar to 62%) by the reaction of isoquinolinium salts, quinolinium salts or phdialazinium salts with ammonium acetate in the presence of tetrakispyridine cobalt(II) dichromate (TPCD).

Abstract:A new method for synthesis of synthesis 6-hydroxy-3, 4-dihydro-2(lH) quinolinone was investigated with higher yield and simpler operation. The title compound was prepared starting from 3-chloropropionyl chloride, via cyclizing, nitration, reduction, diazotization and hydrolysis. The total yield is 67.7%, and the yield of each step is between 86.5% -99.6%. 6-Hydroxy-3, 4-dihydro-2(1H)quinolinone was characterized by MS and 1H NMR.

Abstract:In this work, a conceptually novel approach to diversely substituted 2‐(alkylsulfanyl)quinolines is described. The approach includes the assembly of the target molecules from allenic or acetylenic carbanions, aromatic isothiocyanates, and alkylating agents, The process proceeds in a single synthetic operation via the formation and 6π‐electrocyclization of N‐aryl‐1‐aza‐1,3,4‐trienes, R2R3C=C=C(R1)C(SAlk)=NAr, with the participation of the carbon‐carbon double bond of the phenyl group and the azadiene fragment of the azatrienic system.

Abstract:A method was proposed for the regioselective synthesis of new 2,3-disubstituted quinolines based on the reaction of anilines with 4-chloro-1,1-diethoxybutane and N -(4,4-diethoxybutyl)tosylamide. Structure of the resulting compounds was proven by NMR spectroscopy techniques ( 1 H, 13 C, 15 N, COSY, HSQC 1 H– 13 C, HMBC 1 H– 13 C, HMBC 1 H– 15 N, NOESY), the composition was confirmed by elemental analysis data. The proposed method is distinguished by the possibility of obtaining a wide range of substituted quinolines in one step and the absence of the need to use expensive metal catalysts.

Abstract:An oxidant-free and highly efficient synthesis of phenolic quinazolin-4(3H)-ones was achieved by simply stirring a mixture of 2-aminobenzamides, sulfonyl azides, and terminal alkynes. The intermediate N-sulfonylketenimine underwent two nucleophilic additions and the sulfonyl group eliminated through the power of aromatization. The natural product 2-(4-hydroxybenzyl)quinazolin-4(3H)-one can be synthesized on a large scale under mild conditions with this method.

Abstract:3-Diazoindolin-2-imines reacted with nitrones to furnish 2-iminoindolin-3-ones through a Au(I)-catalyzed cascade oxygen transfer/imine exchange process. The prepared 2-iminoindolin-3-ones could be further transformed into 2-alknyl-2,3-dihydroquinazolin-4(1H)-ones through a Ag(I)-catalyzed reaction with terminal alkynes. A MeOH-triggered ring expansion mechanism involving cyclic iminium formation and nucleophilic addition is proposed for this novel alkynylation reaction. This two-step procedure provides a general and convenient approach to 2-alknyl-2,3-dihydroquinazolin-4(1H)-ones, which are privileged structures in medicinal chemistry.

Abstract:An iodine-mediated one-pot synthesis of pyrrolo/indolo [1,2-a]quinoxalines and quinazolin-4-one via utilizing epoxides as alkyl precursors under metal-free conditions has been described. Both 1-(2-aminophenyl)-pyrrole and 2-aminobenzamide could be applied to this protocol. A total of 33 desired products were obtained with moderate to good yields. This methodology was suitable for wide-scale preparation and the obtained products could be further modified into promising pharmaceutically active reagents.

Abstract:We have developed a versatile and efficient method for copper-catalyzed synthesis of both 2-amino-4(3H)-quinazolinone and 2-aminoquinazoline derivatives. The protocol uses readily available substituted 2-halobenzoic acids, 2-bromobenzaldehyde, 2-bromophenyl ketones and guanidines as the starting materials, inexpensive copper(I) iodide as the catalyst, and the method has important application values for construction of N-heterocycles in organic chemistry and medicinal chemistry.
