Abstract:Copper-based nanoparticles (NPs) display a strong potential to replace copper salts (e.g., CuSO4) for application in agricultures as antimicrobial agents or nutritional amendments. Yet, their effects on crop quality are still not comprehensively understood. In this study, the Cu contents in soybeans grown in soils amended with Cu NPs and CuSO4 at 100-500 mg Cu/kg and the subsequent effects on the plant physiological markers were determined. The Cu NPs induced 29-89% at the flowering stage (on Day 40) and 100-165% at maturation stage (on Day 100) more Cu accumulation in soybeans than CuSO4. The presence of particle aggregates in the root cells with deformation upon the Cu NP exposure was observed by transmission electron microscopy. The Cu NPs at 100 and 200 mg/kg significantly improved the plant height and biomass, yet significantly inhibited at 500 mg/kg, compared to the control. In leaves chlorophyll-b was more sensitive than chlorophyll-a and carotenoids to the Cu NP effect. The Cu NPs significantly decreased the root nitrogen and phosphorus contents, while they significantly increased the leaf potassium content in comparison with control. Our results imply that cautious use of Cu NPs in agriculture is warranted due to relatively high uptake of Cu and altered nutrient quality in soybeans.

Abstract:Abstract Copper (Cu)-based fungicides have been used in viticulture to prevent downy mildew since the end of the 19th century, and are still used today to reduce fungal diseases. Consequently, Cu has built up in many vineyard soils, and it is still unclear how this affects soil functioning. The present study aimed to assess the short and medium-term effects of Cu contamination on the soil fungal community. Two contrasting agricultural soils, an acidic sandy loam and an alkaline silt loam, were used for an eco-toxicological greenhouse pot experiment. The soils were spiked with a Cu-based fungicide in seven concentrations (0–5000 mg Cu kg −1 soil) and alfalfa was grown in the pots for 3 months. Sampling was conducted at the beginning and at the end of the study period to test Cu toxicity effects on total microbial biomass, basal respiration and enzyme activities. Fungal abundance was analysed by ergosterol at both samplings, and for the second sampling, fungal community structure was evaluated via ITS amplicon sequences. Soil microbial biomass C as well as microbial respiration rate decreased with increasing Cu concentrations, with EC 50 ranging from 76 to 187 mg EDTA-extractable Cu kg −1 soil. Oxidative enzymes showed a trend of increasing activity at the first sampling, but a decline in peroxidase activity was observed for the second sampling. We found remarkable Cu-induced changes in fungal community abundance (EC 50 ranging from 9.2 to 94 mg EDTA-extractable Cu kg −1 soil) and composition, but not in diversity. A large number of diverse fungi were able to thrive under elevated Cu concentrations, though within the order of Hypocreales several species declined. A remarkable Cu-induced change in the community composition was found, which depended on the soil properties and, hence, on Cu availability.

Abstract:Copper has been successfully used in the sulfate form as a fungicide to control grapevine diseases since 150 years, yet high Cu accumulation in vineyards may alter soil life. Although actual Cu additions are about tenfold lower than 50 years ago, the use of Cu in the context of the agroecological transition is still debated. Indeed, copper is one of the rare pesticides allowed for organic farming. Therefore, we performed a meta-analysis on Cu ecotoxicity by selecting 19 articles out of 300 articles relevant to copper and soil biological quality. Results show that microbial activity decreased by 30% when more than 400 kg of Cu was applied yearly per ha. Nematodes abundance remained unchanged for copper application up to 3200 kg/ha/year. Collembola and enchytraeid reproduction declined by 50% after application of 400 and 1895 kg Cu/ha/year, respectively. Earthworm biomass was reduced by 15% after application of 200 kgCu/ha/year. For soil Cu levels higher than 200 kg Cu/ha, microbial respiration decreased by 50% and no effect was observed on collembola. Overall, while toxicity is observed, the corresponding literature investigations involved Cu levels that are at least 50 times higher than the dose of 4 kg Cu/ha/year currently authorized by the European Commission for viticulture. As a consequence, applying copper at 4 kg/ha/year should not modify substantially soil biological quality and functions.

Abstract:Because of the use of agricultural land for wine production and the frequent use of copper (Cu)‐based fungicides, Cu concentration has been increasing in soils in many parts of the world. In vineyards with acid parent materials, low soil pH may also encourage Cu toxicity, but it is common to use lime in order to increase the soil pH and improve crop yields. Thus, vineyard soils may have large variations in pH, which will affect the microbial communities, making it difficult to isolate possible toxic effects of Cu on soil microbes. Here, analyses of phospholipid fatty acid (PLFA) patterns and pollution‐induced community tolerance (PICT, using leucine incorporation) were used to study the effect of Cu accumulation at small to medium concentrations in acid soils (pH 4.0–5.8) in vineyards with different land‐use history (young, old and abandoned vineyards). The results suggested that soil pH played a dominant role in determining the composition of the microbial community of the soils. Only a small effect of soil Cu could be detected and the effect on microbial community composition was much less than the pH effect. PICT analysis suggested that bacterial communities in old vineyards were more tolerant of Cu than those in the abandoned vineyards. However, increased PICT (Cu tolerance of the bacterial community) was not directly correlated with Cu concentration in the soils. Instead, the estimated Cu tolerance was significantly correlated with soil pH.

Abstract:Abstract Copper (Cu) toxicity is a pressing concern for several soils, especially in organic viticulture. The objective of this work was to assess Cu toxicity on the non-target organism Eisenia fetida , employing both traditional and novel tools for early identification of Cu-induced damages. In addition to traditional tests like avoidance and reproductive toxicity experiments, other tests such as the single cell gel electrophoresis (SCGE) and gut microbiome analysis were evaluated to identify early and more sensitive pollution biomarkers. Four sub-lethal Cu concentrations were studied, and the results showed strong dose-dependent responses by the earthworm avoidance test and the exceeding of habitat threshold limit at the higher Cu doses. An inverse proportionality was observed between reproductive output and soil Cu concentration. Bioaccumulation was not detected in earthworms; soil concentrations of potentially bioavailable Cu were not affected by E. fetida presence or by time. On the contrary, the SCGE test revealed dose-dependent genotoxicity for the ‘tail length’ parameter already at the second day of Cu exposition. Gut microbiome analysis a modulation of microbial composition, with the most aboundant families being Pectobateriaceae , Comamonadaceae and Microscillaceae . Bacillaceae increased over time and showed adaptability to copper up to 165 mg/kg, while at the highest dose even the sensitive Acetobacteriaceae family was affected. The research provided new insights into the ecotoxicity of Cu sub-lethal doses highlighting both alterations at earthworms’ cellular level and changes in their gut microbiota.

Abstract:The chronic copper (Cu) bioavailability models have been successfully implemented in European risk assessment frameworks and compliance evaluations. However, they have been developed almost two decades ago, which calls for an update. Here, we present updated chronic Cu bioavailability models for invertebrates and algae. They consider recent ecotoxicity datasets, use the more recent speciation model WHAM VII, and an optimized model structure (i.e. a generalized bioavailability model; gBAM). Contrary to a classic Biotic Ligand Model, a gBAM models the effect of pH on Cu2+‐toxicity via a log‐linear relationship parametrized through the pH‐slope SpH. The recalibrated SpH‐parameters are ‐0.208 for invertebrates (Daphnia magna, two clones) and ‐0.975 for algae (Raphidocelis subcapitata and Chlorella vulgaris). The updated models predict 80‐100% of the observed effect levels for 8 different species within a factor of two. The only exception was one of the two datasets considering sub‐chronic 7d‐mortality to Hyalella azteca: prediction performance of the updated invertebrate model at pH≥8.3 was poor because the effect of pH on Cu2+ toxicity appeared to be dependent on the pH itself (with a steeper pH slope compared to the updated invertebrate model at pH≥8.1). The prediction performance of the updated Cu bioavailability models was similar to or better than that of the models used for regulatory application in Europe until today, with one exception (i.e. H. azteca). Together with the recently published fish bioavailability model, the models developed in the present study constitute a complete, updated, and consistent bioavailability model set. Overall, the updated chronic Cu bioavailability model set is robust and can be used in regulatory applications. The updated bioavailability model set is currently used under the European Union REACH (Registration, Evaluation, Authorisation, and Restriction of Chemicals‐framework) regulation to guide the safe use of Cu.

Effects of fungicide application on physiological and molecular responses of grapevine (Vitis vinifera L.): a comparison between copper and sulfur fungicides applied alone and in combination with novel fungicides

Abstract:Background: Chemical products against fungi and oomycetes pose serious environmental issues. In the last decade, the use of less impacting active ingredients was encouraged to reduce chemical inputs in viticulture. In this study, the effect of different antifungal compounds on grapevine agronomic, physiological, and molecular responses in the vineyard was evaluated in addition to protection against powdery and downy mildews. Results: In 2 years and in two Vitis vinifera cultivars (Nebbiolo and Arneis), a conventional crop protection approach, based on traditional fungicides (sulfur and copper), was compared to combined strategies. A well-known resistance inducer (potassium phosphonate), Bacillus pumilus strain QST 2808 and calcium oxide, both active ingredients whose biological interaction with grapevine is poorly characterized, were applied in the combined strategies in association with chemical fungicides. Despite a genotype effect occurred, all treatments optimally controlled powdery and downy mildews, with minimal variations in physiological and molecular responses. Gas exchange, chlorophyll content and photosystem II efficiency increased in treated plants at the end of season, along with a slight improvement in the agronomic performances, and an activation of molecular defense processes linked to stilbene and jasmonate pathways. Conclusion: The disease control strategies based on potassium phosphonate, Bacillus pumilus strain QST 2808 or calcium oxide combined with traditional chemical compounds did not cause severe limitations in plant ecophysiology, grape quality, and productive yields. The combination of potassium phosphonate and calcium oxide with traditional fungicides can represent a valuable strategy for reducing copper and sulfur inputs in the vineyards, including those organically managed. © 2023 The Authors. Abstract:In the defense against downy mildew, copper is still widely used, particularly in organic management. At any rate, specific investigations are needed to significantly reduce the use of copper and minimize its environmental impact. This study, conducted in several farms in Friuli-Venezia Giulia (Northeast Italy), aimed to evaluate the concentration of copper on leaves in various climatic conditions, in order to create an important database for the development of a rapid control system through image analysis. The tests involved sampling leaves and grapes from bud break until the last treatment with copper-based products. After copper recovery using a nitric acid solution, the copper values were analyzed via ICP-AES analysis. Cuprotesmo imprints were also made on the same leaves to develop an application (APP) capable of quickly and non-destructively detecting copper on the leaf surface through image analysis. The analytical data revealed significant correlations between image analysis and copper values only in certain situations involving adult leaves. However, due to the variability in treatment situations, the use of the APP with image analysis for all copper defense scenarios is not yet feasible. Nevertheless, the results are encouraging and will serve for further extensive investigations to develop an image analysis system capable of detecting truly active copper and optimizing pesticide treatments for a precision defense strategy. This strategy aims to ensure low environmental impact, production sustainability, and grape quality.
Europe-wide spatial trends in copper and imidacloprid sensitivity of macroinvertebrate assemblages

Abstract:Exposure to synthetic chemicals, such as pesticides and pharmaceuticals, affects freshwater communities at broad spatial scales. This risk is commonly managed in a prospective environmental risk assessment (ERA). Relying on generic methods, a few standard test organisms, and safety factors to account for uncertainty, ERA determines concentrations that are assumed to pose low risks to ecosystems. Currently, this procedure neglects potential variation in assemblage sensitivity among ecosystem types and recommends a single low-risk concentration for each compound. Whether systematic differences in assemblage sensitivity among ecosystem types exist or their size, are currently unknown. Elucidating spatial patterns in sensitivity to chemicals could therefore enhance ERA precision and narrow a fundamental knowledge gap in ecology, the Hutchinsonian shortfall. We analyzed whether taxonomic turnover between field-sampled macroinvertebrate assemblages of different broad river types across Europe results in systematic differences in assemblage sensitivity to copper and imidacloprid. We used an extensive database of macroinvertebrate assemblage compositions throughout Europe and employed a hierarchical species sensitivity distribution model to predict the concentration that would be harmful to 5% of taxa (HC 5 ) in each assemblage. Predicted values varied over several orders of magnitude. However, variation within the 95% highest density intervals remained within one order of magnitude. Differences between the river types were minor for imidacloprid and only slightly higher for copper. The largest difference between river-type-specific median values was a factor of 3.1. This level of variation is below the assessment factors recommended by the European Food Safety Authority and therefore would be captured in the current ERA for plant protection products. We conclude that the differences in taxonomic composition between broad river types translate into relatively small differences in macroinvertebrate assemblage sensitivity toward the evaluated chemicals at the European scale. However, systematic differences in bioavailability and multi-stressor context were not evaluated and might exacerbate the differences in the ecological effects of chemicals among broad river types in real-world ecosystems.

Abstract:The effect of fungicides, commonly used in vine cultures, on the health of terrestrial and aquatic ecosystems has been poorly studied. The objective of this study was to evaluate the toxicity of three viticulture fungicides (myclobutanil, cymoxanil, and azoxystrobin) on non-target organisms, the bacteria Rhodopirellula rubra, Escherichia coli, Pseudomonas putida, and Arthrobacter sp., the microalgae Raphidocelis subcapitata, and the macrophyte Lemna minor. Fungicide toxicity was performed in acute cell viability assay for bacteria; 72-h and 7-day growth inhibition tests for R. subcapitata and L. minor, respectively. Contents of photosynthetic pigments and lipid peroxidation in L. minor were evaluated. Arthrobacter sp. and P. putida showed resistance to these fungicides. Even though azoxystrobin affected R. rubra and E. coli cell viability, this effect was due to the solvent used, acetone. Cell viability decrease was obtained for R. rubra exposed to cymoxanil and E. coli exposed to myclobutanil (30 min of exposure at 10 mg/L and 240 min of exposure at 46 mg/L, respectively). R. subcapitata showed about 10-fold higher sensitivity to azoxystrobin (EC50-72h = 0.25 mg/L) and cymoxanil (EC50-72h = 0.36 mg/L) than L. minor to azoxystrobin and myclobutanil (EC50-72h = 1.53 mg/L and EC50-72h = 1.89 mg/L, respectively). No lipid peroxidation was observed in L. minor after fungicide exposure, while changes of total chlorophyll were induced by azoxystrobin and myclobutanil. Our results showed that non-target aquatic organisms of different trophic levels are affected by fungicides used in viticulture.

Abstract:To evaluate the environmental implications of nanopesticides (NPs) relative to conventional pesticides, we selected CuO NPs and conventional CuSO4 for tomato disease control as a case study and used life cycle assessment (LCA) to evaluate their environmental impacts. Data obtained from the Ecoinvent database and laboratory experiments (i.e., synthesis and characterization of CuO NPs) were processed by a theoretical scale-up framework to determine the collateral environmental impacts at an industrial production scale. The results were compared on the basis of mass (per unit mass) and antimicrobial performance (per unit efficacy), showing that CuO NPs exhibited over a 10-fold efficiency improvement than conventional CuSO4. Particularly, CuO NPs had obvious advantages in renewable energy (reduced 64.7%), human health (reduced 69.6%), ecosystem quality (reduced 53.2%), and overall environmental performance (reduced 67.8%) under per unit efficacy, compared to conventional CuSO4 pesticides. Additionally, ethanol, ethylene, and copper anode were found to be significant contributors to environmental impacts in the process of producing CuO NPs. Our work provides a holistic comparative assessment between nano- and conventional pesticides, which sheds light on the promising environmental potential of nanopesticides and contributes to agricultural productivity and environmental sustainability.

Abstract:Copper-based plant protection products (PPPs) are widely used in both conventional and organic farming, and to a lesser extent for non-agricultural maintenance of gardens, greenspaces, and infrastructures. The use of copper PPPs adds to environmental contamination by this trace element. This paper aims to review the contribution of these PPPs to the contamination of soils and waters by copper in the context of France (which can be extrapolated to most of the European countries), and the resulting impacts on terrestrial and aquatic biodiversity, as well as on ecosystem functions. It was produced in the framework of a collective scientific assessment on the impacts of PPPs on biodiversity and ecosystem services in France. Current science shows that copper, which persists in soils, can partially transfer to adjacent aquatic environments (surface water and sediment) and ultimately to the marine environment. This widespread contamination impacts biodiversity and ecosystem functions, chiefly through its effects on phototrophic and heterotrophic microbial communities, and terrestrial and aquatic invertebrates. Its effects on other biological groups and biotic interactions remain relatively under-documented.

Abstract:The Cu accumulation in vineyard soils is a major ecological and toxicological burden. However, assessing the Cu in young vineyards with the intensive use of agrochemicals has received little attention. Therefore, for the first time, this study evaluates the accumulation and bioavailability of Cu in the soils of the five-year-old vineyard of an emerging grape-growing region in Phu Tho Province, NW Vietnam using different single extraction procedures (CH 3 COOH, CaCl 2 , and Na 2 -EDTA) and pseudo-total digestion. The ecological risk of Cu is also calculated by the contamination factor and the ecological risk factor. Results reveal that with the short-term use of Cu-fungicides, in the studied soil, Cu is present in an acceptable medium level compared to other reports. However, Cu enrichment can be noticed in the topsoil. A higher mean pseudo-total Cu content is found in the topsoil (0-20 cm) (42.7 mg/kg) compared to the subsoil (20-40-60 cm) (37.0 mg/kg). In addition, due to soil erosion, a downslope accumulation of Cu can be indicated. The extractants are effective comparing to the pseudo-total concentrations in the following order Na 2 -EDTA ˃ CH 3 COOH ˃ CaCl 2 . Based on the bioavailable contents, Cu shows an overall low mobility. The contamination factor implies moderate contamination of the soil. These indicate that Cu is slightly influenced by human activities, such as fungicide application. Abstract:The widespread use of copper-based pesticides, while effective in controlling plant diseases, has been identified as a major source of copper contamination in soils. This raises concerns about potential adverse effects on earthworms, key players in soil health and ecosystem function. To inform sustainable pesticide practices, this study aimed to establish copper toxicity thresholds for earthworm avoidance in agricultural soils impacted by copper-based pesticides. We collected 40 topsoil samples (0–5 cm) from orchards and vineyards in the O'Higgins Region of central Chile, and 10 additional soils under native vegetation as background references. A standardized avoidance bioassay using Eisenia fetida assessed the impact of copper-based pesticides on the soils. Total copper concentrations ranged between 23 and 566 mg kg −1 , with observed toxic effects on earthworms in certain soils. The effective concentration at 50% (EC 50 ) for total soil copper, determined by Eisenia fetida's avoidance response, was 240 mg kg −1 , with a 95% confidence interval of 193–341 mg kg −1 . We further compared our EC 50 values with existing data from agricultural soils impacted by mining activities. Interestingly, the results revealed a remarkable similarity between the thresholds for earthworm avoidance, regardless of the source of copper contamination. This observation underscores the universality of copper toxicity in agricultural ecosystems and its potential impact on soil biota. This study provides novel insights into copper toxicity thresholds for earthworms in real-world, pesticide-contaminated soils.

Abstract:The aim of this work was to investigate the efficacy of copper alternative products of natural origin against Plasmopara viticola to reduce or replace the use of copper in organic farming. The identification of new active and sustainable substances is in fact of priority importance because of the environmental impact of copper. The activity of the products was investigated in laboratory and greenhouse conditions. The laboratory tests included the evaluation of the effect of products on mycelial growth, using artificially inoculated leaf disks, and on sporangial germination. The greenhouse experiments were carried out on grape seedlings cv. Malvasia di Candia treated with products and inoculated, after 24 h, with sporangial suspension of P. viticola. The results both of laboratory and greenhouse tests showed inhibitory activity performed by several botanicals (licorice leaf extract, Yucca schidigera extract), by ALD1901 product, a derivative from Saccharomyces cerevisiae yeast, by Vacciplant formulation containing laminarin and by inorganic products as potassium hydrogen carbonate and MenoRame formulation based on micro-doses of copper and zeolites. The identification of sustainable approaches to manage grape downy mildew is very important to enhance the quality of organic production.

Abstract:Cover crops (CCs) are increasingly used in viticulture because they benefit the soil and the environment in many ways. This study investigated the extent to which the incorporation of CC residues altered organic matter (OM) and Cu dynamics in a Cu-contaminated vineyard topsoil . A 92-day incubation period was used to monitor changes over time in carbon mineralization, carbon hydrolytic enzyme activity , concentration and optical properties of dissolved organic matter (DOM), and Cu solubility after the addition (or not) of two CC residues, oat or faba bean. The results revealed that adding CCs transitorily increased the concentration of DOM in soil solution, as well as the activity of C hydrolytic enzymes and C mineralization rates. DOM content was approximately two orders of magnitude higher in CC-amended soils than in the control soil on day 0, after which it gradually decreased to reach concentrations similar to those measured in the control soil on day 92. Analyses of DOM optical properties showed that its molecular weight and degree of humification increased over time with a decrease in its concentration. The close relationship between DOM and Cu concentrations in the soil solution suggests that degradation of CCs releases soluble forms of C capable of complexing and solubilizing Cu, and hence that incorporating CC residues can transitorily increase the solubility of Cu in vineyard topsoils. Despite their different C:N ratios, oat and faba bean had almost the same effect on Cu dynamics, implying that C inputs played a prominent role in explaining the interactions between OM and Cu within the timeframe of our experiment. In conclusion, this study enabled recommendations on how to mitigate the risk of Cu ecotoxicity associated with incorporating CCs in Cu-contaminated vineyard soils.

Abstract:The persistent use of copper-based fungicides to control grapevine downy mildew (GDM) still represents risks to both human health and the environment, especially in organic production. This heavy metal can affect wine quality due to residues' presence on grape bunches. Implementing sustainable viticultural practices to control diseases is crucial for the wine industry. This work aimed to evaluate chitosan effectiveness toward GDM on a large scale, over a period of three years (2019–2021), and in three distinct vineyards with different environmental conditions. Three GDM management strategies were tested: application of 0.5% chitosan individually; in alternation with copper; and during the last year, in combination with low rate of copper, using conventional strategies and untreated plants as controls. Over three consecutive seasons, chitosan treatments effectively reduced GDM McKinney Index on leaves and bunches under different environmental conditions. On bunches, chitosan alone was less effective than copper, when applied after copper had the same effectiveness, and combined with copper was more effective than copper standard. In 2020 and 2021, the analysis of cooper amount detected in berries of two cultivars showed that all strategies allowed to decrease the amount of copper residue in berries. The application of chitosan alone or in alternation with copper was able to reduce this amount as untreated control. This large scale investigation suggests chitosan as alternative that can integrate and eventually replace copper application against GDM. Further studies to validate these strategies in different environments, disease pressure conditions and cultivar are fundamental towards achieving green and sustainable solutions for GDM management strategies.
