Abstract:The aim of this work was to investigate the efficacy of copper alternative products of natural origin against Plasmopara viticola to reduce or replace the use of copper in organic farming. The identification of new active and sustainable substances is in fact of priority importance because of the environmental impact of copper. The activity of the products was investigated in laboratory and greenhouse conditions. The laboratory tests included the evaluation of the effect of products on mycelial growth, using artificially inoculated leaf disks, and on sporangial germination. The greenhouse experiments were carried out on grape seedlings cv. Malvasia di Candia treated with products and inoculated, after 24 h, with sporangial suspension of P. viticola. The results both of laboratory and greenhouse tests showed inhibitory activity performed by several botanicals (licorice leaf extract, Yucca schidigera extract), by ALD1901 product, a derivative from Saccharomyces cerevisiae yeast, by Vacciplant formulation containing laminarin and by inorganic products as potassium hydrogen carbonate and MenoRame formulation based on micro-doses of copper and zeolites. The identification of sustainable approaches to manage grape downy mildew is very important to enhance the quality of organic production.

Abstract:Downy mildew caused by the obligate biotrophic oomycete Plasmopara viticola is one of the most important and devastating diseases of grapevine. Copper-based fungicides are used to control the disease in organic vineyards. However, since 2006, copper use has been limited by an European Commission regulation. In this study conducted from 2004 to 2007, we tested materials that might replace copper in organic viticulture, under controlled greenhouse and field conditions at two locations in Europe (northern Italy and Switzerland). We tested 112 different treatments, including biocontrol agents, materials of animal origin, homeopathic preparations, inorganic materials, microbial extracts, natural derivatives, plant extracts, physical methods and synthetic materials. Good control results were observed for two new copper formulations, but one of them induced phytotoxic effects. Two of the plant extracts provided good control on both leaves and clusters, and six of them controlled the infection on clusters only. One biocontrol agent reduced symptoms on leaves and bunches. Three of the other treatments provided good control on leaves, and four of them provided good control on bunches. Clay was as effective as copper on both leaves and bunches. Currently, even if there is no treatment that is as effective as copper for controlling grapevine downy mildew in organic vineyards, Yucca schidigera and Salvia officinalis extracts and Trichoderma harzianum, could be considered as promising candidates to be developed. This study may help the development of an integrated management program in which a less effective product is applied in combination with copper, to reduce the total quantities of copper applied.

Abstract:Grapevine downy mildew (GDM) is one of the most serious diseases of grapevines. With limitations in the use of copper-based products imposed for organic agriculture by the European Union, research for alternatives is encouraged. The aim of this research was to follow a 2-year trial to evaluate the control of GDM using some alternative compounds, and to determine their effects on shoot growth, plant photosynthesis, and grape quality and quantity. Under low disease pressure, Bordeaux mixture, copper hydroxide, laminarin combined with low copper, and 0.5 and 0.8% chitosan had the lowest GDM incidence, reduced on leaves by 96, 95, 75, 56, and 81%, respectively, compared with the untreated control in the last survey. With high disease pressure, Bordeaux mixture, laminarin combined with Saccharomyces extracts, and 0.5 and 0.8% chitosan had the lowest GDM incidence, reduced on grape by 86, 37, 66, and 75%, respectively, compared with the untreated control in the survey of mid-July. Chitosan at 0.8% lowered net photosynthesis, due to reduced stomatal conductance, leaf area, and dry weight, with no negative effects observed on the quantity of the grape berries and the quality parameters of their juice. Among the alternatives to copper, chitosan provided the best GDM protection and reduced the vigor of the vegetation, inducing physiological changes without negative effects on grape production.

Abstract:In the defense against downy mildew, copper is still widely used, particularly in organic management. At any rate, specific investigations are needed to significantly reduce the use of copper and minimize its environmental impact. This study, conducted in several farms in Friuli-Venezia Giulia (Northeast Italy), aimed to evaluate the concentration of copper on leaves in various climatic conditions, in order to create an important database for the development of a rapid control system through image analysis. The tests involved sampling leaves and grapes from bud break until the last treatment with copper-based products. After copper recovery using a nitric acid solution, the copper values were analyzed via ICP-AES analysis. Cuprotesmo imprints were also made on the same leaves to develop an application (APP) capable of quickly and non-destructively detecting copper on the leaf surface through image analysis. The analytical data revealed significant correlations between image analysis and copper values only in certain situations involving adult leaves. However, due to the variability in treatment situations, the use of the APP with image analysis for all copper defense scenarios is not yet feasible. Nevertheless, the results are encouraging and will serve for further extensive investigations to develop an image analysis system capable of detecting truly active copper and optimizing pesticide treatments for a precision defense strategy. This strategy aims to ensure low environmental impact, production sustainability, and grape quality.
Effects of fungicide application on physiological and molecular responses of grapevine (Vitis vinifera L.): a comparison between copper and sulfur fungicides applied alone and in combination with novel fungicides

Abstract:Background: Chemical products against fungi and oomycetes pose serious environmental issues. In the last decade, the use of less impacting active ingredients was encouraged to reduce chemical inputs in viticulture. In this study, the effect of different antifungal compounds on grapevine agronomic, physiological, and molecular responses in the vineyard was evaluated in addition to protection against powdery and downy mildews. Results: In 2 years and in two Vitis vinifera cultivars (Nebbiolo and Arneis), a conventional crop protection approach, based on traditional fungicides (sulfur and copper), was compared to combined strategies. A well-known resistance inducer (potassium phosphonate), Bacillus pumilus strain QST 2808 and calcium oxide, both active ingredients whose biological interaction with grapevine is poorly characterized, were applied in the combined strategies in association with chemical fungicides. Despite a genotype effect occurred, all treatments optimally controlled powdery and downy mildews, with minimal variations in physiological and molecular responses. Gas exchange, chlorophyll content and photosystem II efficiency increased in treated plants at the end of season, along with a slight improvement in the agronomic performances, and an activation of molecular defense processes linked to stilbene and jasmonate pathways. Conclusion: The disease control strategies based on potassium phosphonate, Bacillus pumilus strain QST 2808 or calcium oxide combined with traditional chemical compounds did not cause severe limitations in plant ecophysiology, grape quality, and productive yields. The combination of potassium phosphonate and calcium oxide with traditional fungicides can represent a valuable strategy for reducing copper and sulfur inputs in the vineyards, including those organically managed. © 2023 The Authors. Abstract:The persistent use of copper-based fungicides to control grapevine downy mildew (GDM) still represents risks to both human health and the environment, especially in organic production. This heavy metal can affect wine quality due to residues' presence on grape bunches. Implementing sustainable viticultural practices to control diseases is crucial for the wine industry. This work aimed to evaluate chitosan effectiveness toward GDM on a large scale, over a period of three years (2019–2021), and in three distinct vineyards with different environmental conditions. Three GDM management strategies were tested: application of 0.5% chitosan individually; in alternation with copper; and during the last year, in combination with low rate of copper, using conventional strategies and untreated plants as controls. Over three consecutive seasons, chitosan treatments effectively reduced GDM McKinney Index on leaves and bunches under different environmental conditions. On bunches, chitosan alone was less effective than copper, when applied after copper had the same effectiveness, and combined with copper was more effective than copper standard. In 2020 and 2021, the analysis of cooper amount detected in berries of two cultivars showed that all strategies allowed to decrease the amount of copper residue in berries. The application of chitosan alone or in alternation with copper was able to reduce this amount as untreated control. This large scale investigation suggests chitosan as alternative that can integrate and eventually replace copper application against GDM. Further studies to validate these strategies in different environments, disease pressure conditions and cultivar are fundamental towards achieving green and sustainable solutions for GDM management strategies.

Abstract:Control of downy mildew on grapevines in organic viticulture in Europe is mainly based on using copper-based fungicides. However, the effect of these agents is often insufficient to prevent losses when infestation pressure is high. The addition of sulfur, which is primarily used to control powdery mildew, improves the effect of copper on downy mildew. In three out of five trial years, sulfur had a clear positive effect when it was added during spray treatments against Plasmopara viticola, the causal agent of downy mildew on grapevines. This was evident mainly in the fruits; no further positive effect on the leaves was evident. In addition, the present study shows that copper-based fungicides are effective on leaves but have a rather poor effect on grapevine fruit. Furthermore, the study demonstrates that the effect of copper is significantly worse than that of the chemical synthetic contact fungicide folpet.

Abstract:Citrus production is worldwide threatened by Colletotrichum spp., causal agents of pre- and postharvest anthracnose. The recent limitation on the use of copper-based antimicrobials, due to its demonstrated noxious effect on the environment, makes the control of this pathogen difficult. Thus, alternative products able to reduce/phase out copper in organic citrus farming are needed. In this study, the efficacy of 11 commercial alternative products were evaluated in vitro, in growth chamber, in open field and in postharvest environments. In vitro, mineral fertilizers, basic substances, essential oils, plant defense stimulators and biological control agents were able to inhibit the mycelial growth with variable efficacy. On artificially infected citrus fruit, almost all tested products significantly reduced disease incidence and severity, but with lower efficacy than copper. The efficacy of mineral fertilizers-based Kiram and Vitibiosap 458 Plus, citrus essential oil-based Prev-Am Plus and chitosan-based Biorend was confirmed in open field trials, in naturally infected citrus fruits. In these trials Biorend was the best alternative product, significantly reducing disease incidence (71% DI reduction) with better results than copper (47.5%). Field treatments reduced the incidence and severity of the disease in postharvest conditions, especially in fruits field-treated three times. Overall, selected products tested in open field can represent a good alternative to copper compounds in the view of future limitation of its use.

Abstract:The European Green Deal aims to reduce the pesticide use, notably by developing biocontrol products to protect crops from diseases. Indeed, the use of significant amounts of chemicals negatively impact the environment such as soil microbial biodiversity or groundwater quality, and human health. Grapevine ( Vitis vinifera ) was selected as one of the first targeted crop due to its economic importance and its dependence on fungicides to control the main damaging diseases worldwide: grey mold, downy and powdery mildews. Chitosan, a biopolymer extracted from crustacean exoskeletons, has been used as a biocontrol agent in many plant species, including grapevine, against a variety of cryptogamic diseases such as downy mildew ( Plasmopara viticola ), powdery mildew ( Erysiphe necator ) and grey mold ( Botrytis cinerea ). However, the precise molecular mechanisms underlying its mode of action remain unclear: is it a direct biopesticide effect or an indirect elicitation activity, or both? In this study, we investigated six chitosans with diverse degrees of polymerization (DP) ranging from low to high DP (12, 25, 33, 44, 100, and 470). We scrutinized their biological activities by evaluating both their antifungal properties and their abilities to induce grapevine immune responses. To investigate their elicitor activity, we analyzed their ability to induce MAPKs phosphorylation, the activation of defense genes and metabolite changes in grapevine. Our results indicate that the chitosans with a low DP are more effective in inducing grapevine defenses and possess the strongest biopesticide effect against B. cinerea and P. viticola . We identified chitosan with DP12 as the most efficient resistance inducer. Then, chitosan DP12 has been tested against downy and powdery mildews in the vineyard trials performed during the last three years. Results obtained indicated that a chitosan-based biocontrol product could be sufficiently efficient when the amount of pathogen inoculum is quite low and could be combined with only two fungicide treatments during whole season programs to obtain a good protection efficiency. On the whole, a chitosan-based biocontrol product could become an interesting alternative to meet the chemicals reduction targeted in sustainable viticulture.

Abstract:Improper application of copper-based fungicides has made copper stress critical in viticulture, necessitating the need to identify substances that can mitigate it. In this study, leaves of 'Shine Muscat' ('SM') grapevine seedlings were treated with CuSO4 solution (10 mM/L), CuSO4 + 5-aminolevulinic acid (ALA) (50 mg/L), and distilled water to explore the mitigation effect of ALA. Physiological assays demonstrated that ALA effectively reduced malondialdehyde accumulation and increased peroxidase and superoxide dismutase activities in grapevine leaves under copper stress. Copper ion absorption, transport pathways, chlorophyll metabolism pathways, photosynthetic system, and antioxidant pathways play key roles in ALA alleviated-copper stress. Moreover, expression changes in genes, such as CHLH, ALAD, RCA, and DHAR, play vital roles in these processes. Furthermore, abscisic acid reduction caused by NCED down-regulation and decreased naringenin, leucopelargonidin, and betaine contents confirmed the alleviating effect of ALA. Taken together, these results reveal how grapevine responds to copper stress and the alleviating effects of ALA, thus providing a novel means of alleviating copper stress in viticulture.

Abstract:Protecting vineyards from cryptogamic diseases such as downy mildew, caused by Plasmopara viticola, generally requires a massive use of phytochemicals. However, the issues on unintentional secondary effects on environment and human health, and the occurrence of P. viticola resistant strains, are leading to the development of alternative strategies, such as the use of biocontrol products. In this paper, we evidenced the ability of a plant extract to protect grapevine from P. viticola. Further experiments carried out both on cell suspensions and on plants revealed that plant extract activates typical defense-related responses such as the production of H2O2, the up-regulation of genes encoding pathogenesis-related proteins and stilbene synthase, as well as the accumulation of resveratrol or its derivative piceid. We also brought to light a strong direct effect of PE on the release and motility of P. viticola zoospores. Furthermore, we found out that PE application left dried residues on leaf surface, impairing zoospores to reach stomata. Altogether, our results highlight the different modes of action of a new biocontrol product able to protect grapevine against downy mildew.

Abstract:This study focuses on countering Fusarium graminearum, a harmful fungal pathogen impacting cereal crops and human health through mycotoxin production. These mycotoxins, categorized as type B trichothecenes, pose significant health risks. Research explores natural alternatives to synthetic fungicides, particularly investigating phenolics in grapevine byproducts. Thirteen eco-extracts from five French grape varieties (Merlot, Cabernet Sauvignon, Sauvignon blanc, Tannat, and Artaban) exhibited substantial antifungal properties, with ten extracts displaying remarkable effects. Extracts from grapevine stems and roots notably reduced fungal growth by over 91% after five days. Through UHPLC-HRMS/MS analysis and metabolomics, the study identified potent antifungal compounds such as ampelopsin A and cyphostemmin B, among other oligomeric stilbenes. Interestingly, this approach showed that flavan-3-ols have been identified as markers for extracts that induce fungal growth. Root extracts from rootstocks, rich in oligostilbenes, demonstrated the highest antifungal activity. This research underscores grapevine byproducts' potential both as a sustainable approach to control F. graminearum and mycotoxin contamination in cereal crops and the presence of different metabolites from the cultivars of grapevine, suggesting different activities.

Abstract:The efficacy of soil conditioner (vermicompost tea), fertiliser (potassium silicate), and biological control agents (BCAs) as practical agroecological copper alternatives against olive leaf spot (Spilocaea oleaginea (Cast.) Hughe.) disease was investigated between 2018 and 2021 under organic management in a Mediterranean climate. In total, 9 agroecological alternatives to copper oxychloride (vermicompost tea, potassium silicate, Bacillus subtilis EU 007 WP, Platanus orientalis leaf extract, Mycorrhiza mix, seaweed commercial product, Trichoderma citrinoviride TR1, vermicompost tea+Platanus orientalis mix, Penicillium (Mouldy bread pieces)) were applied to olive trees in a randomised block design with 4 replicationsTotal water soluble phenol compounds (TWSP) were found to be the main bioindicator to assess the alternatives and their potential to phase-out copper application. Results related to TWSP indicated that copper oxychloride (control), potassium silicate and vermicompost tea showed significantly higher content of TWSP as we compared zero application of copper and other treatments. These stimulate the antioxidant capacity in olive fruits and reduce the olive leaf spot disease incidence. The pollution effect of copper was monitored during the trial to identify soil pollution in the organic in-conversion experimental land. The total annual ‘active copper’ application was 4.7 kg.ha−1.year−1 and this is in accordance with the legal organic legislation of Turkey. During the conversion period from conventional to organic management, we determined approximately 50% reduced copper content in the soil 0–30 cm depth samples in 2020 (3.70 mg.kg−1) as it is compared to those initial samples (6.43 mg.kg−1) in 2018. We conclude that alternatives to copper that are easily accessible, e.g., vermicompost tea, have a potential for use in organic olive production to replace copper in mitigating olive leaf spots. Furthermore, we find that reduced copper application in organic management with the aim to decrease copper accumulation in soil, fruits and leaves was not yet enough to reduce copper to satisfactory levels. We conclude that further research with the aim of a total replacement of copper fungicide treatments in organic and non-organic systems is needed.

Abstract:Copper-based formulations of wood preservatives are widely used in industry to protect wood materials from degradation caused by fungi. Wood treated with preservatives generate toxic waste that currently cannot be properly recycled. Despite copper being very efficient as an antifungal agent against most fungi, some species are able to cope with these high metal concentrations. This is the case for the brown-rot fungus Rhodonia placenta and the white-rot fungus Phanerochaete chrysosporium, which are able to grow efficiently in pine wood treated with Tanalith E3474. Here, we aimed to test the abilities of the two fungi to cope with copper in this toxic environment and to decontaminate Tanalith E-treated wood. A microcosm allowing the growth of the fungi on industrially treated pine wood was designed, and the distribution of copper between mycelium and wood was analysed within the embedded hyphae and wood particles using coupled X-ray fluorescence spectroscopy and Scanning Electron Microscopy (SEM)/Electron Dispersive Spectroscopy (EDS). The results demonstrate the copper biosorption capacities of P. chrysosporium and the production of copper-oxalate crystals by R. placenta. These data coupled to genomic analysis suggest the involvement of additional mechanisms for copper tolerance in these rot fungi that are likely related to copper transport (import, export, or vacuolar sequestration).

Using ontogenetic resistance of grapevine for fungicide reduction strategies

Abstract:Resistance of grapevine against powdery mildew ( Erysiphe necator ) and downy mildew ( Plasmopara viticola ) increases during ontogenetic development of leaves and berries. The aim of this study was to determine whether the use of fungicides against both pathogens could be reduced during the last treatment before harvest (LTBH) by treating only the susceptible leaves of the vines in the upper half of the canopy. For this purpose, field trials were carried out under experimental or practical conditions in vineyards in southern Germany in eight different years. All plants were sprayed after a standard regimen until LTBH, performed at BBCH 77–79. LTBH was either performed on the whole canopy or limited to the upper half of the canopy excluding the fruit zone. For both pathogens, no differences were observed between the two treatments with regard to the disease severity on the grape clusters. Only in one out of eight years, an increased fruit disease incidence could be observed. As far as the disease severity of leaves is concerned, no differences were observed for downy mildew, and in the case of powdery mildew, significantly higher infestation was observed only in two out of eight years. Furthermore, the efficacy of different fungicides for LTBH was investigated in a 3-year experiment. Sole use of a copper fungicide for LTBH had no disadvantages against E. necator , compared to a treatment with synthetic fungicides. Taken together, LTBH at BBCH 77–79 can be limited to the upper canopy without risk to the health of the fruit.

Abstract:Abstract Copper (Cu)-based fungicides have been used in viticulture to prevent downy mildew since the end of the 19th century, and are still used today to reduce fungal diseases. Consequently, Cu has built up in many vineyard soils, and it is still unclear how this affects soil functioning. The present study aimed to assess the short and medium-term effects of Cu contamination on the soil fungal community. Two contrasting agricultural soils, an acidic sandy loam and an alkaline silt loam, were used for an eco-toxicological greenhouse pot experiment. The soils were spiked with a Cu-based fungicide in seven concentrations (0–5000 mg Cu kg −1 soil) and alfalfa was grown in the pots for 3 months. Sampling was conducted at the beginning and at the end of the study period to test Cu toxicity effects on total microbial biomass, basal respiration and enzyme activities. Fungal abundance was analysed by ergosterol at both samplings, and for the second sampling, fungal community structure was evaluated via ITS amplicon sequences. Soil microbial biomass C as well as microbial respiration rate decreased with increasing Cu concentrations, with EC 50 ranging from 76 to 187 mg EDTA-extractable Cu kg −1 soil. Oxidative enzymes showed a trend of increasing activity at the first sampling, but a decline in peroxidase activity was observed for the second sampling. We found remarkable Cu-induced changes in fungal community abundance (EC 50 ranging from 9.2 to 94 mg EDTA-extractable Cu kg −1 soil) and composition, but not in diversity. A large number of diverse fungi were able to thrive under elevated Cu concentrations, though within the order of Hypocreales several species declined. A remarkable Cu-induced change in the community composition was found, which depended on the soil properties and, hence, on Cu availability.
