Abstract:Copper-based nanoparticles (NPs) display a strong potential to replace copper salts (e.g., CuSO4) for application in agricultures as antimicrobial agents or nutritional amendments. Yet, their effects on crop quality are still not comprehensively understood. In this study, the Cu contents in soybeans grown in soils amended with Cu NPs and CuSO4 at 100-500 mg Cu/kg and the subsequent effects on the plant physiological markers were determined. The Cu NPs induced 29-89% at the flowering stage (on Day 40) and 100-165% at maturation stage (on Day 100) more Cu accumulation in soybeans than CuSO4. The presence of particle aggregates in the root cells with deformation upon the Cu NP exposure was observed by transmission electron microscopy. The Cu NPs at 100 and 200 mg/kg significantly improved the plant height and biomass, yet significantly inhibited at 500 mg/kg, compared to the control. In leaves chlorophyll-b was more sensitive than chlorophyll-a and carotenoids to the Cu NP effect. The Cu NPs significantly decreased the root nitrogen and phosphorus contents, while they significantly increased the leaf potassium content in comparison with control. Our results imply that cautious use of Cu NPs in agriculture is warranted due to relatively high uptake of Cu and altered nutrient quality in soybeans.
Ecotoxicity of copper input and accumulation for soil biodiversity in vineyards

Abstract:Copper has been successfully used in the sulfate form as a fungicide to control grapevine diseases since 150 years, yet high Cu accumulation in vineyards may alter soil life. Although actual Cu additions are about tenfold lower than 50 years ago, the use of Cu in the context of the agroecological transition is still debated. Indeed, copper is one of the rare pesticides allowed for organic farming. Therefore, we performed a meta-analysis on Cu ecotoxicity by selecting 19 articles out of 300 articles relevant to copper and soil biological quality. Results show that microbial activity decreased by 30% when more than 400 kg of Cu was applied yearly per ha. Nematodes abundance remained unchanged for copper application up to 3200 kg/ha/year. Collembola and enchytraeid reproduction declined by 50% after application of 400 and 1895 kg Cu/ha/year, respectively. Earthworm biomass was reduced by 15% after application of 200 kgCu/ha/year. For soil Cu levels higher than 200 kg Cu/ha, microbial respiration decreased by 50% and no effect was observed on collembola. Overall, while toxicity is observed, the corresponding literature investigations involved Cu levels that are at least 50 times higher than the dose of 4 kg Cu/ha/year currently authorized by the European Commission for viticulture. As a consequence, applying copper at 4 kg/ha/year should not modify substantially soil biological quality and functions.

Iron fertilization to enhance tolerance mechanisms to copper toxicity of ryegrass plants used as cover crop in vineyards

Abstract:Ryegrass (Lolium perenne L.) is a plant species that can express mechanisms of tolerance to copper (Cu) toxicity. Therefore, the agronomical approach of intercropping system with ryegrass may represent a promising tool to limit the onset of Cu toxicity symptoms in the other intercropped plants species, particularly when an inadequate nutrient availability like iron (Fe) shortage is also concurrently present. This study aimed at assessing the mechanisms involved in the mitigation of Cu phytotoxicity and the stress effects on plant growth, root morphology and nutrition of ryegrass fertilized with two different Fe sources. To this purpose, seedlings of ryegrass were hydroponically grown for 14 days in controlled conditions with 4 different levels of Cu (0.2, 5.0, 25 and 50 μM) and with either 100 μM Fe-EDDHA or Fe-EDTA. Results show that high levels of Cu availability enhanced the root content of organic anions as well as the root exudation. Different Fe fertilizations at the condition of 50 μM Cu induced changes in root phenolic compounds, citrate and fumarate contents and the exudation pattern of phenolic compounds. Differences in plant growth were not observed between the two Fe sources, although Cu concentration in plant tissue fed with Fe-EDTA was lower in the condition of 50 μM Cu. The enhanced root exudation of Cu-complexing organic compounds (including phenolics) in ryegrass plants when exposed to excessive Cu availability could be at the basis of the ameliorated edaphic rhizosphere conditions (lower Cu availability). For this reason, from the agronomical point of view ryegrass plants used in intercropping systems with crops like vine plants could represent a promising strategy to control Cu toxicity in vineyard soils. Further studies under the field conditions must be taken to support present findings.
Phytoremediation of Cu-contaminated vineyard soils in Brazil: A compendium of Brazilian pot studies

Abstract:Vineyard soils can be contaminated by copper (Cu) due to successive applications of fungicides and organic fertilizers. Soil remediation can be addressed by altering soil properties or selecting efficient Cu-extracting cover crops tolerant to Cu toxicity. Our objectives were to synthesize the Cu-extracting efficiency by plant species tested in Brazil, classify them according to Cu resistance to toxicity, and assess the effect of soil properties on attenuating Cu toxicity. We retrieved results from 41 species and cultivars, totaling 565 observations. Freshly added Cu varied between 50 and 600 mg Cu kg-1 of soil across studies. The partition of Cu removal between the above- and below-ground portions was scaled as a logistic variable to facilitate data synthesis. The data were analyzed using the Adaboost machine learning model. Model accuracy (predicted vs. actual values) reached R2 = 0.862 after relating species, cultivar, Cu addition, clay, SOM, pH, soil test P, and Cu as features to predict the logistic target variable. Tissue Cu concentration varied between 7 and 105 mg Cu kg-1 in the shoot and between 73 and 1340 mg Cu kg-1 in the roots. Among soil properties, organic matter and soil test Cu most influenced the accuracy of the model. Phaseolus vulgaris, Brassica juncea, Ricinus communis, Hordeum vulgare, Sorghum vulgare, Cajanus cajan, Solanum lycopersicum, and Crotolaria spectabilis were the most efficient Cu-extracting cover crops, as shown by positive values of the logistic variable (shoot removal > root removal). Those Cu-tolerant plants showed differential capacity to extract Cu in the long run.

Abstract:Abstract Copper (Cu)-based fungicides have been used in viticulture to prevent downy mildew since the end of the 19th century, and are still used today to reduce fungal diseases. Consequently, Cu has built up in many vineyard soils, and it is still unclear how this affects soil functioning. The present study aimed to assess the short and medium-term effects of Cu contamination on the soil fungal community. Two contrasting agricultural soils, an acidic sandy loam and an alkaline silt loam, were used for an eco-toxicological greenhouse pot experiment. The soils were spiked with a Cu-based fungicide in seven concentrations (0–5000 mg Cu kg −1 soil) and alfalfa was grown in the pots for 3 months. Sampling was conducted at the beginning and at the end of the study period to test Cu toxicity effects on total microbial biomass, basal respiration and enzyme activities. Fungal abundance was analysed by ergosterol at both samplings, and for the second sampling, fungal community structure was evaluated via ITS amplicon sequences. Soil microbial biomass C as well as microbial respiration rate decreased with increasing Cu concentrations, with EC 50 ranging from 76 to 187 mg EDTA-extractable Cu kg −1 soil. Oxidative enzymes showed a trend of increasing activity at the first sampling, but a decline in peroxidase activity was observed for the second sampling. We found remarkable Cu-induced changes in fungal community abundance (EC 50 ranging from 9.2 to 94 mg EDTA-extractable Cu kg −1 soil) and composition, but not in diversity. A large number of diverse fungi were able to thrive under elevated Cu concentrations, though within the order of Hypocreales several species declined. A remarkable Cu-induced change in the community composition was found, which depended on the soil properties and, hence, on Cu availability.

Abstract:Because of the use of agricultural land for wine production and the frequent use of copper (Cu)‐based fungicides, Cu concentration has been increasing in soils in many parts of the world. In vineyards with acid parent materials, low soil pH may also encourage Cu toxicity, but it is common to use lime in order to increase the soil pH and improve crop yields. Thus, vineyard soils may have large variations in pH, which will affect the microbial communities, making it difficult to isolate possible toxic effects of Cu on soil microbes. Here, analyses of phospholipid fatty acid (PLFA) patterns and pollution‐induced community tolerance (PICT, using leucine incorporation) were used to study the effect of Cu accumulation at small to medium concentrations in acid soils (pH 4.0–5.8) in vineyards with different land‐use history (young, old and abandoned vineyards). The results suggested that soil pH played a dominant role in determining the composition of the microbial community of the soils. Only a small effect of soil Cu could be detected and the effect on microbial community composition was much less than the pH effect. PICT analysis suggested that bacterial communities in old vineyards were more tolerant of Cu than those in the abandoned vineyards. However, increased PICT (Cu tolerance of the bacterial community) was not directly correlated with Cu concentration in the soils. Instead, the estimated Cu tolerance was significantly correlated with soil pH.

Abstract:Abstract Copper (Cu) toxicity is a pressing concern for several soils, especially in organic viticulture. The objective of this work was to assess Cu toxicity on the non-target organism Eisenia fetida , employing both traditional and novel tools for early identification of Cu-induced damages. In addition to traditional tests like avoidance and reproductive toxicity experiments, other tests such as the single cell gel electrophoresis (SCGE) and gut microbiome analysis were evaluated to identify early and more sensitive pollution biomarkers. Four sub-lethal Cu concentrations were studied, and the results showed strong dose-dependent responses by the earthworm avoidance test and the exceeding of habitat threshold limit at the higher Cu doses. An inverse proportionality was observed between reproductive output and soil Cu concentration. Bioaccumulation was not detected in earthworms; soil concentrations of potentially bioavailable Cu were not affected by E. fetida presence or by time. On the contrary, the SCGE test revealed dose-dependent genotoxicity for the ‘tail length’ parameter already at the second day of Cu exposition. Gut microbiome analysis a modulation of microbial composition, with the most aboundant families being Pectobateriaceae , Comamonadaceae and Microscillaceae . Bacillaceae increased over time and showed adaptability to copper up to 165 mg/kg, while at the highest dose even the sensitive Acetobacteriaceae family was affected. The research provided new insights into the ecotoxicity of Cu sub-lethal doses highlighting both alterations at earthworms’ cellular level and changes in their gut microbiota.

Abstract:The Cu accumulation in vineyard soils is a major ecological and toxicological burden. However, assessing the Cu in young vineyards with the intensive use of agrochemicals has received little attention. Therefore, for the first time, this study evaluates the accumulation and bioavailability of Cu in the soils of the five-year-old vineyard of an emerging grape-growing region in Phu Tho Province, NW Vietnam using different single extraction procedures (CH 3 COOH, CaCl 2 , and Na 2 -EDTA) and pseudo-total digestion. The ecological risk of Cu is also calculated by the contamination factor and the ecological risk factor. Results reveal that with the short-term use of Cu-fungicides, in the studied soil, Cu is present in an acceptable medium level compared to other reports. However, Cu enrichment can be noticed in the topsoil. A higher mean pseudo-total Cu content is found in the topsoil (0-20 cm) (42.7 mg/kg) compared to the subsoil (20-40-60 cm) (37.0 mg/kg). In addition, due to soil erosion, a downslope accumulation of Cu can be indicated. The extractants are effective comparing to the pseudo-total concentrations in the following order Na 2 -EDTA ˃ CH 3 COOH ˃ CaCl 2 . Based on the bioavailable contents, Cu shows an overall low mobility. The contamination factor implies moderate contamination of the soil. These indicate that Cu is slightly influenced by human activities, such as fungicide application. Although an overall low adverse ecological risk is observed, understanding of the potential enrichment of Cu in the soil of young vineyards can contribute to the development of environmentally friendly vineyard management practices.

Abstract:Inorganic copper (Cu) fungicides and bactericides are widely used to control disease in fruit and vegetable crops and has led to widespread accumulation of the metal in soil beyond regulatory thresholds. We aimed to elucidate the impacts of Cu on soil health within cherry orchard soils in New Zealand, focusing on three biological indicators: earthworm behaviour, soil respiration, and plant growth. We sampled soils from four blocks of different ages within a single orchard, varying in amounts of accumulated soil Cu (7–263 mg kg −1 ) but also in Soil Organic Matter (SOM) content (3–10 %). Experimental work was designed to isolate the impacts of both Cu and SOM on three critical biological descriptors: earthworm behaviour, soil respiration and root growth. Soils were amended to standardise both variables in laboratory and glasshouse experiments. The results demonstrated a pronounced inhibition of soil respiration and root development, as well as adverse effects on earthworm behaviour, with increasing Cu concentrations. SOM played a mitigating role, reducing the bioavailability and toxicity of Cu to soil organisms. However, the buffering capacity of SOM is limited and long-term reliance on SOM to mitigate Cu toxicity is not sustainable. Currently Cu continues to accumulate in most orchard soils. This study highlights the importance of assessing Cu bioavailability and soil health in the context of orchard management.

Abstract:Purpose Copper is a trace element of environmental concern. Repeated applications of Cu-containing fungicides have resulted in a large scale of Cu contamination in agricultural soils. However, limited information is available regarding Cu accumulation and availability in soils under citrus production in the Indian River Area, South Florida, which has received increasing amounts of Cu fungicides to control canker and other diseases. The purpose of this study was to investigate Cu transformation, availability, and mobility in soils as affected by external Cu loading and soil properties. Materials and methods Soil samples ( n = 18) were collected from representative commercial citrus groves in the Indian River area. These samples were characterized to understand the accumulation, availability to plant, and leaching loss, as influenced by soil properties, agricultural practices, and Cu fungicide inputs. Results and discussion Significant accumulation and increased availability and mobility of Cu occurred in the soils with increasing citrus production period. Soil acidification enhanced Cu leaching in soil profiles. Available Cu estimated by Mehlich-3 or NH 4 OAc extraction was highly correlated with total recoverable Cu, whereas the relationship between CaCl 2 -extractable Cu and total recoverable Cu was affected by soil pH, with a greater slope of the relationship curve at soil pH < 6.5. Vertical movement of Cu in a soil profile was more pronounced at pH < 6.5. A large portion of soil Cu was present as organically bound, ranging from 43% to 62%, which was significantly correlated with both recoverable Cu and Mehlich-3-extractable Cu ( P < 0.001). Conclusions Significant accumulation of Cu occurred in citrus grove soils due to anthropogenic inputs, and the availability and mobility of Cu in the soils were related to Cu accumulation and soil pH. Copper migration from Cu-contaminated soils to the environment was reduced when soil pH was 6.5 or higher.
Prevention values for copper (low tier approach) in subtropical acidic soils

Abstract:Copper is a trace element in plants and animals whose importance can be understood due to its role in different essential metabolic processes. Anthropogenic activities such as agriculture and mining are potential sources of pollution due to the emission of copper into the environment. Brazilian legislation ties soil quality to guideline values, among which the Prevention Value indicates the critical environmental limit for trace elements. The aim of this study was to obtain PVs for copper for two subtropical soils (Cambisol and Nitisol), given that the pedological richness was not considered when deriving the PVs contained in the federal normative. Reproduction assays followed ISO guidelines with the earthworm species Eisenia andrei and Perionyx excavatus , the enchytraeids Enchytraeus crypticus and Enchytraeus bigeminus and the springtails Folsomia candida and Proisotoma minuta . Results showed that the sensitivity of the organisms was greater in Cambisol. The most sensitive species were the earthworms, especially Perionyx excavatus (EC 50 = 67.83 in Cambisol; EC 50 = 264.96 in Nitisol). The springtails, on the other hand, were the least sensitive to contamination. These findings reinforce the need to include organisms from different ecological groups in ecotoxicological assessments. It was also observed that the PV adopted in federal legislation (= 60 mg kg −1 ) is in fact protective for the species and soils we evaluated, since the PVs we obtained based on the EC 50 were 346.74 mg kg −1 in Nitisol and 134.05 mg kg −1 in Cambisol. It is important to note that our results do not exclude the need for evaluations with other subtropical soils, given the influence of their properties on the toxicity and bioavailability of copper to soil organisms.

Abstract:Cover crops (CCs) are increasingly used in viticulture because they benefit the soil and the environment in many ways. This study investigated the extent to which the incorporation of CC residues altered organic matter (OM) and Cu dynamics in a Cu-contaminated vineyard topsoil . A 92-day incubation period was used to monitor changes over time in carbon mineralization, carbon hydrolytic enzyme activity , concentration and optical properties of dissolved organic matter (DOM), and Cu solubility after the addition (or not) of two CC residues, oat or faba bean. The results revealed that adding CCs transitorily increased the concentration of DOM in soil solution, as well as the activity of C hydrolytic enzymes and C mineralization rates. DOM content was approximately two orders of magnitude higher in CC-amended soils than in the control soil on day 0, after which it gradually decreased to reach concentrations similar to those measured in the control soil on day 92. Analyses of DOM optical properties showed that its molecular weight and degree of humification increased over time with a decrease in its concentration. The close relationship between DOM and Cu concentrations in the soil solution suggests that degradation of CCs releases soluble forms of C capable of complexing and solubilizing Cu, and hence that incorporating CC residues can transitorily increase the solubility of Cu in vineyard topsoils. Despite their different C:N ratios, oat and faba bean had almost the same effect on Cu dynamics, implying that C inputs played a prominent role in explaining the interactions between OM and Cu within the timeframe of our experiment. In conclusion, this study enabled recommendations on how to mitigate the risk of Cu ecotoxicity associated with incorporating CCs in Cu-contaminated vineyard soils.

Effect of Manuring Practices and Increased Copper Concentrations on Soil Microbial-Populations

Abstract:Addition of piggery manure to soils over 5 yr (manured soils) increased the amount of Cu extracted by EDTA and diethylenetriaminepentaacetic acid (DTPA) compared to selected reference soils. DTPA-extractable Cu ranged from about 3 to 10 μg Cug−1 soil in the manured soils and from about 1 to 2 μg Cu g−1 soil in the reference soils. Although the soil Cu concentrations in the manured soils were many times smaller than currently permitted, the increase in DTPA- or EDTA-extractable Cu was correlated with a 10- to a 1000-fold increase in the number of aerobic Cu-resistant bacteria. Although the Cu was mainly concentrated in the plough layer (15 cm) of the manured soils, Cu-resistant bacteria were also detected down to 110cm soil depth. In contrast, no such differences were found between the total number of colony forming units, total microbial biomass or the degree of Cu-resistance of fungi and anaerobic bacteria between manured and reference soils. It is suggested that the degree of Cu-resistance of the aerobic soil bacteria may provide a sensitive measure of Cu bioavailability in soil. In general, bacteria were more sensitive to Cu than fungi. All of the 42 Cu-resistant bacterial strains investigated were oxidase-positive and 50% of the strains were pigmented. In contrast, only 20% of the 37 Cu-sensitive bacterial strains investigated were oxidase-positive and none were pigmented. Cu-resistant bacteria exhibited more resistance to several antibiotics and heavy metals than Cu-sensitive bacteria.

In order to better-understand the underlying incentives, we construct and test an agency-theoretic model of broiler production contracts. We discuss the analysis' implications for industry participants and government policy.

Abstract:This review focuses on the issue of metal antagonism in soils contaminated by multiple metals as a result of industrial emissions. Building upon previous findings in aquatic ecosystems, the potential of zinc to mitigate copper toxicity in more complex soil systems is explored. A range of studies investigating the role of zinc in reducing copper toxicity to plants and microorganisms in soils contaminated by copper mining in central Chile are examined. The mechanisms underlying metal interactions in soils, including the terrestrial biotic ligand model and the intensity/capacity/quantity concept, are thoroughly discussed. Furthermore, the review underscores the pressing need for future studies to enhance our understanding and develop effective strategies for mitigating copper toxicity in industrially contaminated soils.

Effects of fungicide application on physiological and molecular responses of grapevine (Vitis vinifera L.): a comparison between copper and sulfur fungicides applied alone and in combination with novel fungicides

Abstract:Background: Chemical products against fungi and oomycetes pose serious environmental issues. In the last decade, the use of less impacting active ingredients was encouraged to reduce chemical inputs in viticulture. In this study, the effect of different antifungal compounds on grapevine agronomic, physiological, and molecular responses in the vineyard was evaluated in addition to protection against powdery and downy mildews. Results: In 2 years and in two Vitis vinifera cultivars (Nebbiolo and Arneis), a conventional crop protection approach, based on traditional fungicides (sulfur and copper), was compared to combined strategies. A well-known resistance inducer (potassium phosphonate), Bacillus pumilus strain QST 2808 and calcium oxide, both active ingredients whose biological interaction with grapevine is poorly characterized, were applied in the combined strategies in association with chemical fungicides. Despite a genotype effect occurred, all treatments optimally controlled powdery and downy mildews, with minimal variations in physiological and molecular responses. Gas exchange, chlorophyll content and photosystem II efficiency increased in treated plants at the end of season, along with a slight improvement in the agronomic performances, and an activation of molecular defense processes linked to stilbene and jasmonate pathways. Conclusion: The disease control strategies based on potassium phosphonate, Bacillus pumilus strain QST 2808 or calcium oxide combined with traditional chemical compounds did not cause severe limitations in plant ecophysiology, grape quality, and productive yields. The combination of potassium phosphonate and calcium oxide with traditional fungicides can represent a valuable strategy for reducing copper and sulfur inputs in the vineyards, including those organically managed. © 2023 The Authors. Abstract:Summary The relative rates of absorption of 7 short chain fatty acids from jejunum and colon of man have been compared with their partition into chloroform from an aqueous solution. The longer the chain length the faster the acids were absorbed. This probably reflected their greater solubility in lipid as shown by their partition into chloroform.

Abstract:In the defense against downy mildew, copper is still widely used, particularly in organic management. At any rate, specific investigations are needed to significantly reduce the use of copper and minimize its environmental impact. This study, conducted in several farms in Friuli-Venezia Giulia (Northeast Italy), aimed to evaluate the concentration of copper on leaves in various climatic conditions, in order to create an important database for the development of a rapid control system through image analysis. The tests involved sampling leaves and grapes from bud break until the last treatment with copper-based products. After copper recovery using a nitric acid solution, the copper values were analyzed via ICP-AES analysis. Cuprotesmo imprints were also made on the same leaves to develop an application (APP) capable of quickly and non-destructively detecting copper on the leaf surface through image analysis. The analytical data revealed significant correlations between image analysis and copper values only in certain situations involving adult leaves. However, due to the variability in treatment situations, the use of the APP with image analysis for all copper defense scenarios is not yet feasible. Nevertheless, the results are encouraging and will serve for further extensive investigations to develop an image analysis system capable of detecting truly active copper and optimizing pesticide treatments for a precision defense strategy. This strategy aims to ensure low environmental impact, production sustainability, and grape quality.
Mobility of copper, chromium and arsenic from treated timber into grapevines

Abstract:The use of chromated copper arsenate (CCA) treated timber posts as support structures in New Zealand vineyards has raised concerns regarding the release of heavy metal(loid)s from the treated timber into the environment. A laboratory experiment was set up to evaluate if post sealing by painting reduces the release rate of CCA metal(loid)s from timber posts. Three posts were painted (Gripset 38, Multi Purpose Bitumen Rubber) on the bottom part, submerged in freshwater, and the concentrations of copper (Cu), chromium (Cr) and arsenic (As) in the water were monitored over a period of 8 months. Three additional, unpainted posts were also used. The CCA contents in the water showed a clear difference between the painted and the unpainted posts, and painting the bottom of the posts reduced the release rate by 50-75%. To monitor the possible mobility of CCA from treated posts into grapevines further, an experiment with four-year-old grapevines planted into sixteen lysimeters was set up in a greenhouse. To half the lysimeters Cu, Cr and As were added to the soil surface at rates of 16, 20, and 12.5 mg/month from 15 November 2005 to 5 May 2006. The other lysimeters acted as a control. Soil solutions were collected at 50, 150 and 300 mm depths using suction cups after seven application of the CCA solution. The results showed that all the elements moved to a depth of 50 mm. Grape fruit, leaves and rachis were analysed for CCA metal(loid)s, but did not show any differences between the CCA-treated and control lysimeters. This indicated either that these metals were not taken up by grapevines or that their translocation from roots to the upper part of the vine was negligible. Further monitoring of CCA metal(loid)s in various parts of the grapevines, including roots, needs to be undertaken.
