Abstract:A Gram-stain-negative, aerobic, non-motile, short-rod-shaped bacterium, designated as strain TC11T, was isolated from rhizosphere soil of mangrove forest (Kandeliaobovata) in Fugong village, Zhangzhou, Fujian, China. Strain TC11T grew at 15-45 °C (optimum, 35 °C), 0-8 % (w/v) NaCl (optimum, 1 %, w/v) and pH 5.5-9.5 (optimum, pH 7.5). Phylogenetic analyses revealed that strain TC11T belonged to a clade of the genus Pseudomonas and showed the highest sequence similarity of 98.4 % to Pseudomonas fluvialis ASS-1T, followed by Pseudomonas oleovorans subsp.oleovorans DSM 1045T (97.9 %), Pseudomonas indoloxydans JCM 14246T (97.7 %), Pseudomonas guguanensis JCM 18416T(97.6 %) and Pseudomonas alcaliphila JCM 10630T (97.5 %) on the basis of their 16S rRNA gene sequences. The DNA G+C content was 64.3 mol%. In silico DNA-DNA hybridization and average nucleotide identity values between strain TC11T and the reference strains were 19-22 % and 72-78 %, respectively. Studies based on the three housekeeping genes, rpoB, gyrB and rpoD, further confirmed that strain TC11T is a novel member of the genus Pseudomonas. The major fatty acids of strain TC11Twere C16 : 0, summed feature 8 (C18 : 1ω6c/C18 : 1ω7c) and summed feature 3 (C16 : 1ω7c/C16 : 1ω6c). The sole isoprenoid quinone was Q-9. The major polar lipids were phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol. Based on the phenotypic, chemotaxonomic and phylogenetic properties, strain TC11T represents a novel species of the genus Pseudomonas, for which the name Pseudomonasmangrovi sp. nov., is proposed. The type strain is TC11T (=KCTC 62159=MCCC 1K03499).
Rhodococcus biphenylivorans sp. nov., a polychlorinated biphenyl-degrading bacterium

Abstract:A Gram-positive, aerobic, non-motile and rod–coccus shaped novel actinobacterial strain, designated as TG9 T , was isolated from a polychlorinated biphenyl (PCB)-contaminated sediment in Taizhou city, Zhejiang province, eastern China. The isolate was observed to grow at 10–45 °C (optimum 28–32 °C), pH 5.0–11.0 (optimum pH 7.0–8.0) and with 0–9.0 % (w/v) NaCl (optimum 0–3.0 %). Comparison of the 16S rRNA gene sequences of strain TG9 T and other members of the genus Rhodococcus showed that strain TG9 T shared highest similarities with Rhodococcus pyridinivorans DSM 44555 T (99.4 %), R. rhodochrous DSM 43241 T (99.2 %), R. gordoniae DSM 44689 T (99.2 %) and R. artemisiae DSM 45380 T (98.2 %). However, low levels of DNA–DNA relatedness (15–48 %), which are below the 70 % limit for prokaryotic species identification, were obtained by DNA–DNA hybridization. Strain TG9 T was found to contain meso-diaminopimelic acid as the diagnostic diamino acid and arabinose and galactose in the whole-cell hydrolysate. Mycolic acids were found to be present. The major fatty acids were identified as C 16:0 , C 16:1 ω 7 c and/or iso-C 15:0 2-OH, 10-methyl C 18:0 and C 18:1 ω 9 c . The only menaquinone detected was MK-8 (H 2 ). The major polar lipids detected were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, glycolipid and traces of some unknown lipids. The genomic DNA G+C content of strain TG9 T was determined to be 62.8 %. The combined phenotypic and genotypic data show that the strain represents a novel species of the genus Rhodococcus for which the name Rhodococcus biphenylivorans sp. nov. is proposed, with the type strain TG9 T (=CGMCC 1.12975 T = KCTC 29673 T = MCCC 1K00286 T ).
Pseudomonas Oryzae Sp. Nov. Isolated from a Paddy Soil in South China

Abstract:A Gram-staining-negative, rod-shaped and motile with several polar flagellums bacterium, designated WM-3T, was isolated from a rice paddy soil in South China. Growth occurred with 0–3.0 % (w/v) NaCl (optimum 2.0 %), at pH 5.5–9.0 (optimum pH 7.0) and at 25–42 °C (optimum 30–37 °C) in liquid Reasoner’s 2A medium. Analysis of the 16S rRNA gene and gyrB gene sequences revealed that strain WM-3T was most closely related to the type strains of the species Pseudomonas linyingensis and Pseudomonas sagittaria. Its sequence similarities with P. linyingensis CGMCC 1.10701T and P. sagittaria JCM 18195T were 97.4 and 97.3 %, respectively, for 16S rRNA gene, and were 94.1 and 94.2 %, respectively, for gyrB gene. DNA–DNA hybridization between strain WM-3T and these two type strains showed relatedness of 35.6 and 30.9 %, respectively. G+C content of genomic DNA was 69.4 mol%. The whole-cell fatty acids mainly consisted of C16:0 (30.0 %), C16:1 ω6c and/or C16:1 ω7c (19.3 %) and C18:1 ω6c and/or C18:1 ω7c (16.3 %). The results of phenotypic, chemotaxonomic and genotypic analyses clearly indicated that strain WM-3T belongs to genus Pseudomonas but represents a novel species, for which the name Pseudomonas oryzae sp. nov. is proposed. The type strain is WM-3T (=KCTC 32247T =CGMCC 1.12417T).

Abstract:A Gram-stain-negative, motile, polyhydroxybutyrate-accumulating, aerobic, S-shaped bacterium, designated B3T, was isolated from the wastewater of a pickle-processing factory. 16S rRNA gene sequence similarity analysis showed that it was most closely related to the type strain, Terasakiella pusilla (96.6% similarity). Strain B3T was able to grow at 4-40 °C (optimum 32-37 °C), pH 5.5-9.0 (optimum 6.5-7.5) and with 0.5-8% (w/v) NaCl present (optimum 1-2%, w/v). Chemotaxonomic analysis showed that the respiratory quinone was ubiquinone Q-10, the major fatty acids included C16:0, C18:1ω7c and C16:1ω7c and/or iso-C15:2-OH. The major polar lipids included diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylserine, aminophospholipid and three uncharacterized phospholipids. The genomic DNA G+C content of strain B3T was 42.3 mol%. The DNA-DNA relatedness value between B3T and T. pusilla DSM 9263T was 23.9%. On the basis of the phenotypic, chemotaxonomic and genotypic characteristics of strain B3T, it represents a novel species of the genus Terasakiella, for which the name Terasakiella brassicae sp. nov. is proposed. The type strain is B3T (=KCTC 42652T=CGMCC 1.15254T). Emended descriptions of T. pusilla and the genus Terasakiella are also presented.
Pontibrevibacter Nitratireducens Gen. Nov., Sp. Nov., a Member of the Family Rhodobacteraceae Isolated from Seawater of the Indian Ocean and Intertidal Zone

Abstract:Two Gram-stain-negative, facultatively anaerobic, non-motile, rod-shaped bacteria, strains h42(T) and ALG8, were isolated individually from the Indian Ocean and intertidal zone of Zhoushan, China. The results of 16S rRNA gene sequence analysis showed that the sequence similarity between strains h42(T) and ALG8 was 99.7 %, and the closest related strains were Monaibacterium marinum C7(T) (97.77 and 97.62%) and Pontivivens insulae GYSW-23(T) (95.31 and 95.45%). Phylogenetic analysis based on 16S rRNA gene sequences shows that these two novel strains belong to a distinct new lineage of the family Rhodobacteraceae in the order Rhodobacterales. The average nucleotide identity and in silico DNA-DNA hybridization values between the two novel strains and M. marinum C7(T) and P. insulae GYSW-23(T) were 72.73-78.15 % and 19.70-20.80%, respectively. The DNA G+C content of strains h42(T) and ALG8 was 62.36 % and 62.17 mol%. The major fatty acids (>10%) in strain h42(T) were C-18:0, C-19:0 cyclo omega 8c and summed feature 8 (C-18:1 omega 6c and/or C-18:1 omega 7c), and in strain ALG8 were C-19:0 cyclo omega 8c and summed feature 8 (C-18:1 omega 6c and/or C-18:1 omega 7c). The predominant isoprenoid ubiquinone of the two novel strains was Q-10; their major polar lipids were identified as diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, two unidentified glycolipids, an unidentified aminoglycolipid, an unidentified phospholipid and an unidentified lipid. Based on the results of the morphological, physiological, chemotaxonomic and phylogenetic analysis of these two strains, a novel species of a new genus in the family Rhodobacteraceae is proposed, named as Pontibrevibacter nitratireducens gen. nov., sp. nov. The type strain and non-type strain of P. nitratireducens are h42(T) (=KCTC 72875(T)=CGMCC 1.17849(T)=MCCC 1K04735(T)) and ALG8 (=KCTC 82194=MCCC 1K04733).
Pseudomonas sihuiensis sp. nov., isolated from a forest soil in South China

Abstract:A Gram-stain negative, motile, rod-shaped bacterium, designated strain WM-2 T , was isolated from a forest soil in Sihui City, South China, and characterized by means of a polyphasic approach. Growth occurred with 0–5 % (w/v) NaCl (optimum 0–1 %) and at pH 5.0–10.5 (optimum pH 8.5) and 4–40 °C (optimum 30 °C) in Luria–Bertani medium. Comparative 16S rRNA gene sequence analyses showed that strain WM-2 T is a member of the genus Pseudomonas and most closely related to P. guguanensis , P. oleovorans subsp. lubricantis , P. toyotomiensis , P. alcaliphila and P. mendocina with 97.1–96.6 % sequence similarities. In terms of gyr B and rpo B gene sequences, strain WM-2 T showed the highest similarity with the type strains of the species P. toyotomiensis and P. alcaliphila . The DNA–DNA relatedness values of strain WM-2 T with P. guguanensis and P. oleovorans subsp. lubricantis was 48.7 and 37.2 %, respectively. Chemotaxonomic characteristics (the main ubiquinone Q-9, major fatty acids C 18:1 ω 7 c /C 18:1 ω 6 c , C 16:0 and C 16:1 ω 7 c /C 16:1 ω 6 c and DNA G+C content 65.2 ± 0.7 mol%) were similar to those of members of the genus Pseudomonas . Polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, an unknown aminophospholipid, an unknown phospholipid and five unknown lipids. According to the results of polyphasic analyses, strain WM-2 T represents a novel species in the genus Pseudomonas, for which the name Pseudomonas sihuiensis sp. nov. is proposed. The type strain is WM-2 T (=KCTC 32246 T =CGMCC 1.12407 T ).
Roseiterribacter gracilis gen. nov., sp. nov., a novel filterable alphaproteobacterium isolated from soil using a gel-filled microwell array device

Abstract:Our previous studies indicate the abundant and diverse presence of yet-to-be-cultured microorganisms in the micropore-filtered fractions of various environmental samples. Here, we isolated a novel bacterium (designated as strain TMPK1 T ) from a 0.45-μm-filtered soil suspension by using a gel-filled microwell array device comprising 900 microwells and characterized its phylogenetic and physiological features. This strain showed low 16S rRNA gene sequence identities (<91%) and low average nucleotide identity values (<70%) to the closest validly described species, and belonged to a novel-family-level lineage within the order Rhodospirillales of Alphaproteobacteria . Strain TMPK1 T exhibited small cell sizes (0.08–0.23 μm 3 ) and had a high cyclopropane fatty acid content (>13%), and these characteristics were differentiated from other Rhodospirillales bacteria. A comprehensive habitability search using amplicon datasets suggested that TMPK1 T and its close relatives are mainly distributed in soil and plant-associated environments. Based on these results, we propose that strain TMPK1 T represents a novel genus and species named Roseiterribacter gracilis gen. nov., sp. nov. (JCM 34627 T = KCTC 82790 T ). We also propose Roseiterribacteraceae fam. nov. to accommodate the genus Roseiterribacter .

Abstract:A Gram-stain negative, aerobic, non-motile and ovoid- to rod-shaped bacterial strain, designated JB3 T , was isolated from a wastewater sample collected from the biochemical reaction basin of Haiyan fine chemical factory in Zhejiang, China. Strain JB3 T was found to grow optimally at pH 7.0–8.0, at 28 °C and in the presence of 1.0–2.0 % (w/v) NaCl. Chemotaxonomic analysis showed that strain JB3 T contains ubiquinone-10 (>99 %) as the predominant respiratory quinone and C 18:1 ω 7 c (70.9 %) as the most abundant fatty acid. The polar lipids of strain JB3 T were identified as phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, an aminophospholipid, an unidentified aminolipid, four unidentified phospholipids and three unidentified lipids. The DNA G+C content of strain JB3 T was determined to be 68.1 mol%. The 16S rRNA gene sequence similarities between the isolate and Roseovarius crassostreae DSM 16950 T , Roseovarius sediminilitoris KCTC 23959 T and Roseovarius halocynthiae MA1-10 T were found to be 97.1, 96.8 and 96.2 %, respectively. Morevoer, the similarity between strain JB3 T and the type strain of the genus Roseovarius ( Roseovarius tolerans DSM 11457 T ) was found to be 93.8 %. The phylogenetic trees reconstructed with all three treeing methods showed that strain JB3 T constituted a different taxon, which was separate from other taxa with validly published names, and formed a cluster with R. crassostreae DSM 16950 T , R. sediminilitoris KCTC 23959 T and R. halocynthiae MA1-10 T . These three species were not placed within the phylogenetic cluster formed by R. tolerans DSM 11457 T . Differential phenotypic properties, together with the phylogenetic distinctiveness, demonstrated that strain JB3 T is clearly distinct from species of the genus Roseovarius . On the basis of these features, we propose strain JB3 T represents a novel species of a novel genus with the name Pseudoroseovarius zhejiangensis gen. nov., sp. nov. The type strain is JB3 T (=MCCC 1K00457 T = KCTC 42443 T ). We also propose that R. crassostreae , R. sediminilitoris and R. halocynthiae should be transferred to this new genus as Pseudoroseovarius crassostreae comb. nov., Pseudoroseovarius sediminilitoris comb. nov. and Pseudoroseovarius halocynthiae comb. nov., respectively.
Rhodohalobacter Barkolensis Sp Nov., Isolated from a Saline Lake and Emended Description of the Genus Rhodohalobacter

Abstract:A Gram-stain-negative, non-motile, aerobic, rod-shaped bacterium, designated 15182T, was isolated from a saline lake in China. The novel strain 15182T was able to grow at 10-40 °C (optimum, 37 °C), pH 7.0-8.0 (optimum, 7.5) and with 0.5-4 % NaCl (optimum, 2-3 %, w/v). The phylogenetic analysis based on 16S rRNA gene sequences revealed that strain 15182T was most closely related to the genus Rhodohalobacter by sharing the highest sequence similarity of 97.0 % with Rhodohalobacter halophilus JZ3C29T. Chemotaxonomic analysis showed that the sole respiratory quinone was menaquinone 7, the major fatty acids included C16 : 0 N alcohol and C16 : 1ω11c. The major polar lipids included diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, four uncharacterized glycolipids, one uncharacterized phospholipid and two uncharacterized lipids. The genomic DNA G+C content of the strain 15182T was 42.4 mol%. The average nucleotide identity value between 15182T and R. halophilus JZ3C29T was 75.4 %, and the in silico DNA-DNA hybridization value of the two strains was 19.1 %. On the basis of its phenotypic, chemotaxonomic, genotypic and genomic characteristics presented in this study, strain 15182T is suggested to represent a novel species in the genus Rhodohalobacter, for which the name Rhodohalobacter barkolensis sp. nov. is proposed. The type strain is 15182T (=KCTC 62172T=MCCC 1K03442T). An emended description of the genus Rhodohalobacter is also presented.
Anaeromyxobacter oryzae sp. nov., Anaeromyxobacter diazotrophicus sp. nov. and Anaeromyxobacter paludicola sp. nov., isolated from paddy soils

Abstract:Three bacterial strains (Red232T, Red267T and Red630T) were isolated from paddy soils sampled in Japan. Cells of these strains were Gram-stain-negative, facultative anaerobic, long rod-shaped with monotrichous flagella or pilus-like structures for motility, and formed red colonies on agar plates. Phylogenetic trees based on 16S rRNA gene and multiple single-copy gene sequences showed that the three strains formed a cluster with the type strains of Anaeromyxobacter species, independent from any other strain genera. Similarity values of the 16S rRNA gene sequences and genomes among the three isolated strains and the type strain of Anaeromyxobacter, Anaeromyxobacter dehalogenans 2CP-1T, were 95.4-97.4% for 16S rRNA gene sequence, 75.3-79.5% for average nucleotide identity, 19.6-21.7% for digital DNA-DNA hybridization and 64.1-72.6% for average amino acid identity, all of which are below the species delineation thresholds. Nitrogenase genes were observed in the genomes of the three novel strains, but not in A. dehalogenans 2CP-1T. Moreover, multiple genomic, physiological and chemotaxonomic features supported the discrimination between these three strains. Based on the evidence in this study, the three isolates represent three novel independent species for which the following names are proposed: Anaeromyxobacter oryzae sp. nov., Anaeromyxobacter diazotrophicus sp. nov. and Anaeromyxobacter paludicola sp. nov. The type strains are Red232T (=NBRC 114074T=MCCC 1K03954T), Red267T (=NBRC 114075T=MCCC 1K04211T), and Red630T (=NBRC 114076T=MCCC 1K03957T), respectively.
Bacillus huizhouensis sp. nov., isolated from a paddy field soil

Abstract:A Gram-stain positive, facultative aerobic bacterium, designated as strain GSS03 T , was isolated from a paddy field soil. The cells were observed to be endospore forming, rod-shaped and motile with flagella. The organism was found to grow optimally at 35 °C at pH 7.0 and in the presence of 1 % NaCl. The strain was classified as a novel taxon within the genus Bacillus on the basis of phenotypic and phylogenetic analyses. The closest phylogenetic relatives were identified as Bacillus psychrosaccharolyticus DSM 6 T (97.61 %), Bacillus muralis DSM 16288 T (97.55 %), Bacillus asahii JCM 12112 T (97.48 %), Bacillus simplex DSM 1321 T (97.48 %) and “ Bacillus frigoritolerans ” DSM 8801 T (97.38 %). The menaquinone was identified as MK-7, the major cellular fatty acid was identified as anteiso-C 15:0 and the major cellular polar lipids as phosphatidylethanolamine, phosphatidylmonomethylethanolamine, diphosphatidylglycerol, phosphatidylglycerol and three unknown polar lipids. The DNA G+C content was determined to be 40.2 mol%. The DNA–DNA relatedness with the closest relatives was below 48 %. Therefore, on the basis of all the results, strain GSS03 T is considered to represent a novel species within the genus Bacillus , for which the name Bacillus huizhouensis sp. nov. is proposed. The type strain is GSS03 T (=KCTC 33172 T =CCTCC AB 2013237 T ).

Abstract:A Gram-stain-positive, neutrophilic, rod-shaped bacterium, strain A1g(T), was isolated from activated sludge of a bioreactor and was subjected to a polyphasic taxonomic characterization. The isolate grew in the presence of 0-17.0% (w/v) NaCl and at pH 6.0-9.0; optimum growth was observed in the presence of 3.0-5.0% (w/v) NaCl and at pH 7.0. Strain A1g(T) was motile, formed cream-coloured colonies, was catalase- and oxidase-positive and was able to hydrolyse aesculin, Tween 40 and Tween 60. Chemotaxonomic analysis revealed menaquinone-7 as the predominant respiratory quinone and anteiso-C(15:0), anteiso-C(17:0), iso-C(16:0) and iso-C (5:0) as. major fatty acids. The genomic DNA G+C content of strain A1g(T) was 36.3 mol%. Comparative 16S rRNA gene sequence analysis revealed that the new isolate belonged to the genus Oceanobacillus and exhibited closest phylogenetic affinity to the type strains of Oceanobacillus oncorhynchi subsp. incaldanensis (97.9% similarity) and O. oncorhynchi subsp. oncorhynchi (97.5%), but less than 97% sequence similarity with respect to the type strains of other recognized Oceanobacillus species. Levels of DNA DNA relatedness between strain AlgT and reference strains O. oncorhynchi subsp. incaldanensis DSM 16557(T), O. oncorhynchi subsp. oncorhynchi JCM 12661(T) and Oceanobacillus iheyensis DSM 14371(T) were 29,45 and 38%, respectively. On the basis of phenotypic and genotypic data, strain A1g(T) is considered to represent a novel species of the genus Oceanobacillus, for which the name Oceanobacillus neutriphilus sp. nov. is proposed. The type strain is A1g(T) (=CGMCC 1.7693(T) =JCM 15776(T)).
Polyphasic Taxonomic Characterisation of a Novel Strain As Pararhizobium Haloflavum Sp. Nov., Isolated from Soil Samples Near a Sewage Treatment Tank

Abstract:A Gram-stain negative, aerobic, motile and ovoid- to rod-shaped bacteria strain, designated XC0140T, was isolated from soil samples near the sewage treatment tank of a chemical factory in Zhejiang Province, China, and subjected to polyphasic taxonomic investigation. Strain XC0140T grew at 10–37 °C and pH 6.0–9.0 (optimum, 35 °C and pH 7.5) and with 0–17% (w/v) NaCl (optimum, 1%). According to phylogenetic analysis based on 16S rRNA gene sequences, strain XC0140T was assigned to the genus Pararhizobium with high 16S rRNA gene sequence similarity of 95.97% to “Pararhizobium helanshanense CCNWQTX14T”, followed by Pararhizobium sphaerophysae CCNWGS0238T (95.95%). Chemotaxonomic analysis showed that strain XC0140T contains ubiquinone-10 as the predominant respiratory quinone and possessed summed feature 8 (comprising C18: 1 ω7c and/or ω6c), 11-methyl C18:1 ω7c, C18: 0 and C16: 0 as predominant forms of fatty acids. The polar lipids of strain XC0140T consisted of seven phospholipids (PL), two aminolipids (AL), one glycolipid (GL) and three unidentified lipids (L1, L2 and L3). The DNA G+C content was 62.7 mol%. Based on the polyphasic taxonomic characterization, strain XC0140T is considered to represent a novel species of the genus Pararhizobium, for which the name Pararhizobium haloflavum sp. nov. is proposed. Abstract:A Gram-stain-negative, strictly aerobic and moderately halophilic bacterium, designated strain MA-7-27(T), was isolated from a marine red alga, Porphyridium marinum, in the Republic of Korea. The cells of strain MA-7-27(T) were non-motile rods showing oxidase- and catalase-positive activities. Growth of strain MA-7-27(T) was observed at 15-45 degrees C (optimum, 30 degrees C), pH 5.0-9.0 (pH 7.0) and in the presence of 0.0-5.0 % (w/v) NaCl (2.0 %). Strain MA-7-27(T) contained C-10:0, summed feature 1 (comprising iso-C-15:1 h and/or C-13:1 3-OH) and summed feature 8 (comprising C-18:1 omega 7c and/or C-18:1 omega 6c) as the major fatty acids. The only isoprenoid quinone detected was ubiquinone-10. The major polar lipids of strain MA-7-27(T) were phosphatidylglycerol, two unidentified phospholipids and two unidentified aminolipids. The G+C content of the genomic DNA was approximately 63.6 mol%. Strain MA-7-27(T) was most closely related to the type strains of Boseongicola aestuarii BS-W15(T) and Nioella nitratireducens SSW136(T) with 96.98 % and 96.12 % 16S rRNA gene sequence similarities, respectively, but phylogenetic analyses showed that strain MA-7-27(T) formed a clearly distinct phylogenic lineage from the closely related strains. The phenotypic, chemotaxonomic and molecular properties support that strain MA-7-27(T) represents a novel genus of the family Rhodobacteraceae, for which the name Rhodophyticola porphyridii gen. nov., sp. nov. is proposed. The type strain is MA-7-27(T) (=KACC 18805(T) =JCM 31537(T)).
Photorhabdus africana sp. nov. isolated from Heterorhabditis entomopathogenic nematodes

Abstract:One Gram-negative, rod-shaped bacterial strain, isolated from an undescribed Heterorhabditis entomopathogenic nematode species was characterized to determine its taxonomic position. The 16S rRNA gene sequences indicate that it belongs to the class Gammaproteobacteria , to the family Morganellaceae , to the genus Photorhabdus , and likely represents a novel bacterial species. This strain, designated here as CRI-LC T , was therefore molecularly, biochemically, and morphologically characterized to describe the novel bacterial species. Phylogenetic reconstructions using 16S rRNA gene sequences show that CRI-LC T is closely related to P. laumondii subsp . laumondii TT01 T and to P. laumondii subsp . clarkei BOJ-47 T . The 16rRNA gene sequences between CRI-LC T and P. laumondii subsp . laumondii TT01 T are 99.1% identical, and between CRI-LC T and P. laumondii subsp . clarkei BOJ-47 T are 99.2% identical. Phylogenetic reconstructions using whole genome sequences show that CRI-LC T is closely related to P. laumondii subsp . laumondii TT01 T and to P. laumondii subsp . clarkei BOJ-47 T . Moreover, digital DNA-DNA hybridization (dDDH) values between CRI-LC T and its two relative species P. laumondii subsp . laumondii TT01 T and P. laumondii subsp . clarkei BOJ-47 T are 65% and 63%, respectively. In addition, we observed that average nucleotide identity (ANI) values between CRI-LC T and its two relative species P. laumondii subsp . laumondii TT01 T and P. laumondii subsp . clarkei BOJ-47 T are 95.8% and 95.5%, respectively. These values are below the 70% dDDH and the 95–96% ANI divergence thresholds that delimits prokaryotic species. Based on these genomic divergence values, and the phylogenomic separation, we conclude that CRI-LC T represents a novel bacterial species, for which we propose the name Photorhabdus africana sp. nov. with CRI-LC T (= CCM 9390 T = CCOS 2112 T ) as the type strain. The following biochemical tests allow to differentiate P. africana sp. nov. CRI-LC T from other species of the genus, including its more closely related taxa: β -Galactosidase, citrate utilization, urease and tryptophan deaminase activities, indole and acetoin production, and glucose and inositol oxidation. Our study contributes to a better understanding of the taxonomy and biodiversity of this important bacterial group with great biotechnological and agricultural potential.

Abstract:A novel bacterial species, Photorhabdus viridis sp. nov., represented by strain Green T , isolated from Heterorhabditis zealandica MJ2C entomopathogenic nematodes, is described. Phylogenetic reconstructions using 16S rRNA gene sequences show that strain Green T is closely related to P. thracensis DSM 15199 T . The 16rRNA gene sequences of these two strains are 98.8% identical. Phylogenetic reconstructions using whole-genome sequences show that strain Green T is closely related to P. tasmaniensis DSM 22387 T , P. thracensis DSM 15199 T , and P. temperata DSM 14550 T . Digital DNA-DNA hybridization (dDDH) values between strain Green T and its three more close relative species, P. tasmaniensis DSM 22387 T , P. thracensis DSM 15199 T , and P. temperata DSM 14550 T , are 49%, 59%, and 59%, respectively. In addition, average nucleotide identity (ANI) values between Green T and P. tasmaniensis DSM 22387 T , P. thracensis DSM 15199 T , and P. temperata DSM 14550 T are 92.4%, 94.4%, and 94.6%, respectively. The novel species also differs in their biochemical capacities from the biochemical capacities of their more closely related taxa. The following biochemical tests may be particularly useful in this context: Arginine dihydrolase, gelatinase, and glucose and mannitol oxidation. Given the clear phylogenetic separation, the sequence divergence values, and the phenotypic differences, we conclude that strain Green T represents a novel bacterial species, for which we propose the name Photorhabdus viridis sp. nov. with Green T (= CCM 9407 T = CCOS 2117 T = MJ2C T ) as the type strain. Our study contributes to a better understanding of the taxonomy and biodiversity of an important bacterial group with great biotechnological and agricultural potential.

Description of Aliirhizobium terrae sp. nov., A Plant Growth-Promoting Bacterium Isolated from a Maize-Rice Rotation Agriculture Field

Abstract:A polyphasic taxonomic approach was used to characterize a novel bacterium, designated strain CC-CFT758T, isolated from a maize-rice rotation agriculture field in Taiwan. The cells are aerobic, Gram-stain-negative, rod-shaped, positive for catalase and oxidase, and grow at 20-30 °C (optimal 30 ℃), at pH 6.0-8.0 (optimal 8.0), and with 0-4% (w/v) NaCl (optimum, 2-3%). Phylogenetic analysis based on 16S rRNA gene sequencing, the strain CC-CFT758T belongs to the genus "Aliirhizobium" of the family Rhizobiaceae. The closest known relatives of this strain are "Aliirhizobium wenxiniae" 166T (with 98.7% similarity), "Aliirhizobium cellulosilyticum" SEMIA 448T (with 97.9% similarity), and "Aliirhizobium smilacinae" PTYR-5T (with 97.0% similarity). The genome size was 5.9 Mbp, with a G + C content of 60.6%. Values of digital DNA-DNA hybridization between the strain and closely related species were 29.5% for "Ali. cellulosilyticum", and 23.9% for "Ali. wenxiniae" and "Ali. smilacinae". Strain CC-CFT758T exhibited the highest orthologous average nucleotide identity (OrthoANI) values with members of the genus "Aliirhizobium", ranging from 80.4 to 81.6% (n = 3). Chemotaxonomical analysis indicated that strain CC-CFT758T contained C16:0, C16:0 3OH, C19:0 cyclo ω8c, C14:0 3OH/iso-C16:1 I, and C18:2 ω6,9c/ante C18:0 as dominant fatty acids, and the major polyamines were putrescine and spermidine. The polar lipids comprised diphosphatidylglycerol, phosphatidylcholin, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylmonomethylethanolamine, seven unidentified aminolipids, three unidentified phospholipids, and two unidentified polar lipids. Strain CC-CFT758T exhibited distinct phylogenetic, phenotypic, and chemotaxonomic characteristics, as well as unique results in comparative analysis of 16S rRNA gene sequence, OrthoANI, AAI, dDDH, and phylogenomic placement. Therefore, this strain represents a new species of the genus "Aliirhizobium", for which the name Aliirhizobium terrae sp. nov. is proposed, with the type strain is CC-CFT758T (= BCRC 81364T = JCM 35482T).
Arthrobacter pokkalii sp nov, a Novel Plant Associated Actinobacterium with Plant Beneficial Properties, Isolated from Saline Tolerant Pokkali Rice, Kerala, India

Abstract:A novel yellow colony-forming bacterium, strain P3B162T was isolated from the pokkali rice rhizosphere from Kerala, India, as part of a project study aimed at isolating plant growth beneficial rhizobacteria from saline tolerant pokkali rice and functionally evaluate their abilities to promote plant growth under saline conditions. The novel strain P3B162T possesses plant growth beneficial traits such as positive growth on 1-aminocyclopropane-1-carboxylic acid (ACC), production of indole acetic acid (IAA) and siderophore. In addition, it also showed important phenotypic characters such as ability to form biofilm and utilization of various components of plant root exudates (sugars, amino acids and organic acids), clearly indicating its lifestyle as a plant rhizosphere associated bacterium. Taxonomically, the novel strain P3B162T was affiliated to the genus Arthrobacter based on the collective results of phenotypic, genotypic and chemotaxonomic analyses. Moreover, molecular analysis using 16S rRNA gene showed Arthrobacter globiformis NBRC 12137T, Arthrobacter pascens DSM 20545T and Arthrobacter liuii DSXY973T as the closely related phylogenetic neighbours, showing more than 98% 16S rRNA similarity values, whereas the recA gene analysis displayed Arthrobacter liuii JCM 19864T as the nearest neighbour with 94.7% sequence similarity and only 91.7% to Arthrobacter globiformis LMG 3813T and 88.7% to Arthrobacter pascens LMG 16255T. However, the DNA-DNA hybridization values between strain P3B162T, Arthrobacter globiformis LMG 3813T, Arthrobacter pascens LMG 16255T and Arthrobacter liuii JCM 19864T was below 50%. In addition, the novel strain P3B162T can be distinguished from its closely related type strains by several phenotypic characters such as colony pigment, tolerance to NaCl, motility, reduction of nitrate, hydrolysis of DNA, acid from sucrose, cell wall sugars and cell wall peptidoglycan structure. In conclusion, the combined results of this study support the classification of strain P3B162T as a novel Arthrobacter species and we propose Arthrobacter pokkalii sp.nov.as its name. The type strain is P3B162T (= KCTC 29498T = MTCC 12358T).
