Abstract:A Gram-stain-negative, coccoid, yellow, non-motile, aerobic bacterium, designated strain S36T, was isolated from soil of the Xixi wetland in Zhejiang province, PR China. Phylogenetic analysis, based on 16S rRNA gene sequences, revealed that strain S36T could represent a novel species of genus Altererythrobacter showing highest similarity to Altererythrobacter atlanticus 26DY36T (96.31 % 16S rRNA gene sequence similarity). The temperature, pH and NaCl concentration ranges for growth were 10-37 °C (optimum 32 °C), pH 5.0-10.0 (optimum pH 7.0) and 0.5-3 % (optimum 1 %, w/v), respectively. The predominant respiratory quinone of strain S36T was Q-10. The major fatty acids were C16 : 0, C17 : 1ω6c, C18 : 1ω7c and summed feature 3 (C16 : 1ω7c and/or iso-C15 : 0 2-OH). The G+C content of the genomic DNA was 62.7 mol%. These data all support the affiliation of strain S36T to the genus Altererythrobacter. The polar lipids profile of strain S36T comprised diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, two unidentified phospholipids and two unidentified glycolipids. The results of physiological and biochemical tests allowed differentiation of strain S36T from other members of the genus Altererythrobacter. Therefore, strain S36T represents a novel species of the genus Altererythrobacter, for which the name Altererythrobacter xixiisoli sp. nov. is proposed; the type strain is S36T (=CGMCC 1.12804T=NBRC 110413T).
Polyphasic Taxonomic Characterisation of a Novel Strain As Pararhizobium Haloflavum Sp. Nov., Isolated from Soil Samples Near a Sewage Treatment Tank

Abstract:A Gram-stain negative, aerobic, motile and ovoid- to rod-shaped bacteria strain, designated XC0140T, was isolated from soil samples near the sewage treatment tank of a chemical factory in Zhejiang Province, China, and subjected to polyphasic taxonomic investigation. Strain XC0140T grew at 10–37 °C and pH 6.0–9.0 (optimum, 35 °C and pH 7.5) and with 0–17% (w/v) NaCl (optimum, 1%). According to phylogenetic analysis based on 16S rRNA gene sequences, strain XC0140T was assigned to the genus Pararhizobium with high 16S rRNA gene sequence similarity of 95.97% to “Pararhizobium helanshanense CCNWQTX14T”, followed by Pararhizobium sphaerophysae CCNWGS0238T (95.95%). Chemotaxonomic analysis showed that strain XC0140T contains ubiquinone-10 as the predominant respiratory quinone and possessed summed feature 8 (comprising C18: 1 ω7c and/or ω6c), 11-methyl C18:1 ω7c, C18: 0 and C16: 0 as predominant forms of fatty acids. The polar lipids of strain XC0140T consisted of seven phospholipids (PL), two aminolipids (AL), one glycolipid (GL) and three unidentified lipids (L1, L2 and L3). The DNA G+C content was 62.7 mol%. Based on the polyphasic taxonomic characterization, strain XC0140T is considered to represent a novel species of the genus Pararhizobium, for which the name Pararhizobium haloflavum sp. nov. is proposed. Tepidibacter Mesophilus Sp Nov., A Mesophilic Fermentative Anaerobe Isolated from Soil Polluted by Crude Oil, and Emended Description of the Genus Tepidibacter

Abstract:A mesophilic, aerotolerant, endospore-forming, fermentative bacterium, designated strain B1(T), was isolated from soil polluted by crude oil in the Karamay Oil Field, China. Cells were Gram-positive, rod-shaped, 1.1-1.6 µm wide and 2.3-4.7 µm long, and were motile by means of peritrichous flagella. Growth occurred at 10-40 °C and pH 6.0-8.9; optimal growth occurred at 28-32 °C and pH 7.3. The optimal concentrations of NaCl and sea salts for growth were 0.5 and 1% (w/v), respectively. The strain was halotolerant and grew in the presence of NaCl or sea salts up to a concentration of 9% (w/v). Substrates utilized as sole carbon sources were beef extract, yeast extract, peptone, tryptone, casein, D-glucose, D-fructose, D-xylose, D-ribose, D-galactose, maltose, L-rhamnose, trehalose, L-valine, DL-alanine plus L-proline and DL-alanine plus L-glycine. The main products of glucose fermentation were ethanol and acetate. iso-C(15:0), iso-C(14:0), C(16:0) and iso-C(13:0) were the major fatty acids. 16S rRNA gene sequence analysis revealed that the isolate belongs to the genus Tepidibacter, showing 94.7 and 94.1% similarity to the type strains of Tepidibacter formicigenes and Tepidibacter thalassicus, respectively. The genomic DNA G+C content of strain B1(T) was 29.8 mol%. On the basis of its phenotypic and genotypic properties, strain B1(T) is suggested to represent a novel species of the genus Tepidibacter, for which the name Tepidibacter mesophilus sp. nov. is proposed. The type strain is B1(T) (=CGMCC 1.5148(T) =JCM 16806(T)).
Paenibacillus Tianmuensis Sp. Nov., Isolated from Soil

Abstract:Two closely related, Gram-stain-negative, rod-shaped, spore-forming strains, B27(T) and F6-B70, were isolated from soil samples of Tianmu Mountain National Natural Reserve in Zhejiang, China. Phylogenetic analysis based on 16S rRNA gene and rpoB sequences indicated that the isolates were members of the genus Paenibacillus. Both isolates were closely related to Paenibacillus ehimensis IFO 15659(T), Paenibacillus elgii SD17(T) and Paenibacillus koreensis YC300(T) (≥ 95.2 % 16S rRNA gene sequence similarity). DNA-DNA relatedness between strain B27(T) and P. ehimensis DSM 11029(T), P. elgii NBRC 100335(T) and P. koreensis KCTC 2393(T) was 21.2, 28.6 and 16.8 %, respectively. The major cellular fatty acids of strains B27(T) and F6-B70 were anteiso-C(15 : 0) and iso-C(15 : 0). The cell wall contained meso-diaminopimelic acid. The two isolates differed from their closest neighbours in terms of phenotypic characteristics and cellular fatty acid profiles (such as variable for oxidase, negative for methyl red test, unable to produce acid from d-fructose and glycogen and relatively higher amounts of iso-C(15 : 0) and lower amounts of C(16 : 0) and iso-C(16 : 0)). Strains B27(T) and F6-B70 represent a novel species of the genus Paenibacillus, for which the name Paenibacillus tianmuensis sp. nov. is proposed. The type strain is B27(T) ( = DSM 22342(T) = CGMCC 1.8946(T)).
Bacillus huizhouensis sp. nov., isolated from a paddy field soil

Abstract:A Gram-stain positive, facultative aerobic bacterium, designated as strain GSS03 T , was isolated from a paddy field soil. The cells were observed to be endospore forming, rod-shaped and motile with flagella. The organism was found to grow optimally at 35 °C at pH 7.0 and in the presence of 1 % NaCl. The strain was classified as a novel taxon within the genus Bacillus on the basis of phenotypic and phylogenetic analyses. The closest phylogenetic relatives were identified as Bacillus psychrosaccharolyticus DSM 6 T (97.61 %), Bacillus muralis DSM 16288 T (97.55 %), Bacillus asahii JCM 12112 T (97.48 %), Bacillus simplex DSM 1321 T (97.48 %) and “ Bacillus frigoritolerans ” DSM 8801 T (97.38 %). The menaquinone was identified as MK-7, the major cellular fatty acid was identified as anteiso-C 15:0 and the major cellular polar lipids as phosphatidylethanolamine, phosphatidylmonomethylethanolamine, diphosphatidylglycerol, phosphatidylglycerol and three unknown polar lipids. The DNA G+C content was determined to be 40.2 mol%. The DNA–DNA relatedness with the closest relatives was below 48 %. Therefore, on the basis of all the results, strain GSS03 T is considered to represent a novel species within the genus Bacillus , for which the name Bacillus huizhouensis sp. nov. is proposed. The type strain is GSS03 T (=KCTC 33172 T =CCTCC AB 2013237 T ).
Meiothermus Terrae Sp. Nov., Isolated from a Geothermally Heated Soil Sample.

Abstract:A Gram-negative, aerobic bacterium, designated strain YIM 77755T, was isolated from a geothermally heated soil sample collected at Rehai National Park, Tengchong, Yunnan province, south-west China. Cells of the strain were rod-shaped and colonies were yellow and circular. Growth occurred in 0–1 % (w/v) NaCl, at pH 6.0–8.0 (optimum, pH 7.0) and at 35–55 °C (optimum, 50 °C). The predominant menaquinone was MK-8 and the DNA G+C content was 68.9 mol%. Major fatty acids (>10 %) were anteiso-C15 : 0 and iso-C15 : 0. The polar lipids consisted of an uncharacterized phospholipid and four glycolipids. Based on 16S rRNA gene sequence analysis, strain YIM 77755T formed a cluster with Meiothermus chliarophilus ALT-8T and showed the highest 16S rRNA gene sequence similarity to M. chliarophilus ALT-8T (98.23 %). DNA–DNA relatedness between YIM 77755T and M. chliarophilus DSM 9957T was 54.9±4.1 %. On the basis of the morphological and chemotaxonomic characteristics as well as genotypic data, it is proposed that strain YIM 77755T represents a novel species of the genus Meiothermus , named Meiothermus terrae sp. nov. The type strain is YIM 77755T ( = DSM 26712T = CCTCC AB 2012942T).

Abstract:Two aerobic, non-motile and non-sporulating bacteria, strains LY1( T) and L462, were recovered from a hot spring located in the Qianshan area of north-eastern China. The novel strains had Gram-negative cell walls and grew at 30-66 degrees C (optimum 55-60 degrees C) and pH 5.5-10.0 (optimum pH 8.0-8.5). 16S rRNA gene sequence similarity analysis revealed that strains LY1( T) and L462 were most closely related to Meiothermus ruber ATCC 35948(T), M. taiwanensis WR-30(T) and M. cerbereus GY-1(T), with 97.1-98.4 % sequence similarity. Phylogenetic analysis indicated that the new isolates represented a novel species by forming a distinctive lineage within genus Meiothermus, which was confirmed by DNA-DNA reassociation values. Strain LY1( T) was obviously different from its closest relatives in a number of phenotypic characteristics, such as the inability to hydrolyse casein or to assimilate melibiose, trehalose, sucrose and lactose. Strain L462 was distinct in its ability to reduce nitrate. In addition, the novel strains were distinct in having larger amounts of anteiso-C(17 : 0) than their closest phylogenetic neighbours. On the basis of this polyphasic taxonomic characterization, the name Meiothermus cateniformans sp. nov. is proposed for the novel species, currently represented by the type strain LY1( T) (=CGMCC 1.6990(T) =JCM 15151(T)) and strain L462 (=CGMCC 1.6989 =JCM 15150).

Abstract:A Gram-stain-negative, motile, polyhydroxybutyrate-accumulating, aerobic, S-shaped bacterium, designated B3T, was isolated from the wastewater of a pickle-processing factory. 16S rRNA gene sequence similarity analysis showed that it was most closely related to the type strain, Terasakiella pusilla (96.6% similarity). Strain B3T was able to grow at 4-40 °C (optimum 32-37 °C), pH 5.5-9.0 (optimum 6.5-7.5) and with 0.5-8% (w/v) NaCl present (optimum 1-2%, w/v). Chemotaxonomic analysis showed that the respiratory quinone was ubiquinone Q-10, the major fatty acids included C16:0, C18:1ω7c and C16:1ω7c and/or iso-C15:2-OH. The major polar lipids included diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylserine, aminophospholipid and three uncharacterized phospholipids. The genomic DNA G+C content of strain B3T was 42.3 mol%. The DNA-DNA relatedness value between B3T and T. pusilla DSM 9263T was 23.9%. On the basis of the phenotypic, chemotaxonomic and genotypic characteristics of strain B3T, it represents a novel species of the genus Terasakiella, for which the name Terasakiella brassicae sp. nov. is proposed. The type strain is B3T (=KCTC 42652T=CGMCC 1.15254T). Emended descriptions of T. pusilla and the genus Terasakiella are also presented.

Abstract:Bacterial strain Y-6T, isolated from a landfill site in Yiwu, PR China, was characterized using a polyphasic taxonomy approach. Cells were Gram- stain-negative, aerobic, rod-shaped, motile by means of a single polar flagellum and formed pale beige colo-nies. Strain Y-6T grew at 4-40 degrees C (optimal at 30-37 degrees C), pH 6.5-9.5 (optimal at pH 7.2-8.5) and in the presence of 0.5-10.0 % (w/v) NaCl (optimal at 1.0-3.0 %). Phylogenetic analysis revealed that strain Y -6T was a member of the genus Aliidiomarina and closely related to Aliidiomarina taiwanensis MCCC 1A06493T with a 16S rRNA sequence similarity of 98.2 %. The major cellular fatty acids of the isolate were iso-C15:0, C16:0, iso-C17: 0 and summed feature 9 (iso-C17: 1 omega 9c and/or 10-methyl-C16:0). Q-8 was the predominant ubiquinone. The major polar lipids comprised diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethan-olamine, aminoglycophospholipid, aminophospholipid, phospholipid, three glycolipids and two unknown lipids. The genomic DNA G+C content was 46.6mol%. The digital DNA-DNA hybridization value between Y -6T and A. taiwanensis MCCC 1A06493T was 18.3 %. Strain Y -6T had an average nucleotide identity value of 74.09 % with A. taiwanensis MCCC 1A06493T. Results from the polyphasic taxonomy study support the conclusion that strain Y -6T represents a novel Aliidiomarina species, for which the name Aliidiomarina quisquiliarum sp.nov. is proposed. The type strain is Y -6T (=MCCC 1K06228T=KCTC 82676T).
Maribacter Thermophilus Sp Nov., Isolated From An Algal Bloom In An Intertidal Zone, And Emended Description Of The Genus Maribacter

Abstract:A novel facultatively anaerobic, Gram-stain-negative bacterium, designated strain HT7-2(T), was isolated from Ulva prolifera collected from the intertidal zone of Qingdao sea area, China, during its bloom. Cells were rod-shaped (1.9-3.5x0.4-0.6 mu m), non-sporulating and motile by gliding. Strain HT7-2(T) was able to grow at 4-50 degrees C (optimum 40-42 degrees C), pH 5.5-8.5 (optimum pH 7.0), 0-8 % (w/v) NaCl (optimum 2-3 %) and 0.5-10% (w/v) sea salts (optimum 2.5 %). The genomic DNA G+C content was 38.8 mol%. The phylogenetic analysis based on 16S rRNA gene sequences revealed that strain HT7-2(T) belonged to the genus Maribacter with sequence similarity values of 94.5-96.6 %, and was most closely related to Maribacter aestuarii GY20(T) (96.6 %). Chemotaxonomic analysis showed that the main isoprenoid quinone was MK-6 and the major fatty acids were iso-C-15:0 and unknown equivalent chain-length 13.565. The polar lipids of strain HT7-2(T) consisted of one phosphatidylethanolamine, four unidentified lipids and one unidentified aminolipid. On the basis of the phenotypic, phylogenetic and chemotaxonomic characteristics, strain HT7-2(T) (=CGMCC 1.12207(T)=JCM 18466(T)) is concluded to represent a novel species of the genus Maribacter, for which the name Maribacter thermophilus sp. nov. is proposed. An emended description of the genus Maribacter is also proposed.

Abstract:A novel Fe(III)-reducing bacterium, designated GSS01(T), was isolated from a forest soil sample using a liquid medium containing acetate and ferrihydrite as electron donor and electron acceptor, respectively. Cells of strain GSS01(T) were strictly anaerobic, Gram-stain-negative, motile, non-spore-forming and slightly curved rod-shaped. Growth occurred at 16-40 °C and optimally at 30 °C. The DNA G+C content was 60.9 mol%. The major respiratory quinone was MK-8. The major fatty acids were C(16:0), C(18:0) and C(16:1)ω7c/C(16:1)ω6c. Strain GSS01(T) was able to grow with ferrihydrite, Fe(III) citrate, Mn(IV), sulfur, nitrate or anthraquinone-2,6-disulfonate, but not with fumarate, as sole electron acceptor when acetate was the sole electron donor. The isolate was able to utilize acetate, ethanol, glucose, lactate, butyrate, pyruvate, benzoate, benzaldehyde, m-cresol and phenol but not toluene, p-cresol, propionate, malate or succinate as sole electron donor when ferrihydrite was the sole electron acceptor. Phylogenetic analyses based on 16S rRNA gene sequences revealed that strain GSS01(T) was most closely related to Geobacter sulfurreducens PCA(T) (98.3% sequence similarity) and exhibited low similarities (94.9-91.8%) to the type strains of other species of the genus Geobacter. The DNA-DNA relatedness between strain GSS01(T) and G. sulfurreducens PCA(T) was 41.4 ± 1.1%. On the basis of phylogenetic analysis, phenotypic characterization and physiological tests, strain GSS01(T) is believed to represent a novel species of the genus Geobacter, and the name Geobacter soli sp. nov. is proposed. The type strain is GSS01(T) ( =KCTC 4545(T) =MCCC 1K00269(T)).
Description of Aliirhizobium terrae sp. nov., A Plant Growth-Promoting Bacterium Isolated from a Maize-Rice Rotation Agriculture Field

Abstract:A polyphasic taxonomic approach was used to characterize a novel bacterium, designated strain CC-CFT758T, isolated from a maize-rice rotation agriculture field in Taiwan. The cells are aerobic, Gram-stain-negative, rod-shaped, positive for catalase and oxidase, and grow at 20-30 °C (optimal 30 ℃), at pH 6.0-8.0 (optimal 8.0), and with 0-4% (w/v) NaCl (optimum, 2-3%). Phylogenetic analysis based on 16S rRNA gene sequencing, the strain CC-CFT758T belongs to the genus "Aliirhizobium" of the family Rhizobiaceae. The closest known relatives of this strain are "Aliirhizobium wenxiniae" 166T (with 98.7% similarity), "Aliirhizobium cellulosilyticum" SEMIA 448T (with 97.9% similarity), and "Aliirhizobium smilacinae" PTYR-5T (with 97.0% similarity). The genome size was 5.9 Mbp, with a G + C content of 60.6%. Values of digital DNA-DNA hybridization between the strain and closely related species were 29.5% for "Ali. cellulosilyticum", and 23.9% for "Ali. wenxiniae" and "Ali. smilacinae". Strain CC-CFT758T exhibited the highest orthologous average nucleotide identity (OrthoANI) values with members of the genus "Aliirhizobium", ranging from 80.4 to 81.6% (n = 3). Chemotaxonomical analysis indicated that strain CC-CFT758T contained C16:0, C16:0 3OH, C19:0 cyclo ω8c, C14:0 3OH/iso-C16:1 I, and C18:2 ω6,9c/ante C18:0 as dominant fatty acids, and the major polyamines were putrescine and spermidine. The polar lipids comprised diphosphatidylglycerol, phosphatidylcholin, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylmonomethylethanolamine, seven unidentified aminolipids, three unidentified phospholipids, and two unidentified polar lipids. Strain CC-CFT758T exhibited distinct phylogenetic, phenotypic, and chemotaxonomic characteristics, as well as unique results in comparative analysis of 16S rRNA gene sequence, OrthoANI, AAI, dDDH, and phylogenomic placement. Therefore, this strain represents a new species of the genus "Aliirhizobium", for which the name Aliirhizobium terrae sp. nov. is proposed, with the type strain is CC-CFT758T (= BCRC 81364T = JCM 35482T).

Abstract:Two Gram-negative, aerobic, rod-shaped bacterial strains, 7MK25 T and 6Y81 T , were isolated from forest soil of Dinghushan Biosphere Reserve, Guangdong Province, PR China. Based on the results of 16S rRNA gene sequence analysis, strain 7MK25 T showed the highest similarity (93.6 %) to Methyloferula stellata AR4 T , followed by Bosea thiooxidans DSM 9653 T (93.3 %). Strain 6Y81 T had the highest similarity of 97.9 % to Lichenibacterium minor RmlP026 T , followed by Lichenibacterium ramalinae RmlP001 T (97.2 %). Phylogenomic analysis using the UBCG and PhyloPhlAn methods consistently showed that strain 7MK25 T formed a sister clade to Boseaceae , while strain 6Y81 T formed an independent clade within the genus Lichenibacterium , both in the order Hyphomicrobiales . The digital DNA–DNA hybridization and average nucleotide identity values between strains 7MK25 T , 6Y81 T and their close relatives were in the ranges of 19.1–29.9 % and 72.5–85.5 %, respectively. The major fatty acids of 7MK25 T were summed feature 8 (C 18 : 1 ω7 c /C 18 : 1 ω6 c ), C 19 : 0 cyclo ω8 c , C 16 : 0 and C 17 : 0 cyclo, while those of 6Y81 T were summed feature 8 (C 18 : 1 ω7 c /C 18 : 1 ω6 c ), C 16 : 0 and C 16 : 0 3-OH. Strains 7MK25 T and 6Y81 T took diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol and phosphatidylcholine as their dominant polar lipids, and Q-10 as their major respiratory quinone. On the basis of phenotypic and phylogenetic data, strain 7MK25 T is proposed to represent a novel species of a novel genus with name Terrirubrum flagellatum gen. nov., sp. nov., within a novel family Terrirubraceae fam. nov., with 7MK25 T (=KCTC 62738 T =GDMCC 1.1452 T ) as its type strain. Strain 6Y81 T represents a novel species in the genus Lichenibacterium , for which the name Lichenibacterium dinghuense sp. nov. (type strain 6Y81 T =KACC 21 727 T =GDMCC 1.2176 T ) is proposed. Rhodoblastaceae fam. nov. with Rhodoblastus as the type genus is also proposed to solve the non-monophylectic problem of the family Roseiarcaceae .

Abstract:A novel Gram-staining-negative, oval-shaped (0.4-0.6×0.8-1.0 µm), non-motile strain without flagella, designated B7T, was isolated from deep seawater in the South China Sea. Strain B7T was able to grow at 25-40 °C (optimum 35 °C), at pH 5.5-9.0 (optimum pH 7.0) and with 0-8 % (w/v) NaCl (optimum 3 %). Chemotaxonomic analysis showed that the predominant isoprenoid quinone was Q-10 and the dominant fatty acids were C19 : 0 cyclo 8c and summed feature 8 (C18 : 1ω7c/C18 : 1ω6c). The polar lipids of strain B7T were diphosphatidyglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, one unknown aminophospholipid, one unknown glycolipid and three unknown lipids. The DNA G+C content of the genomic DNA was 65.1 mol%. Phylogenetic analysis of 16S rRNA gene sequences showed that strain B7T belongs to the genus Mesorhizobium with similarities ranging from 96.2 to 97.5 %. Phylogenetic analyses of housekeeping genes recA, atpD and glnII indicated that strain B7T represented a distinct evolutionary lineage with the genus Mesorhizobium. OrthoANI values between strain B7T and related strains of the genus Mesorhizobium (<80 %) were lower than the threshold value of 95 % ANI relatedness for species demarcation. Therefore, strain B7T is concluded to represent a novel species of the genus Mesorhizobium, for which the name Mesorhizobium oceanicumsp. nov. is proposed. The type strain is B7T (=KCTC 42783T=MCCC 1K02305T).

Abstract:A Gram-stain-negative, aerobic, non-motile, short-rod-shaped bacterium, designated as strain TC11T, was isolated from rhizosphere soil of mangrove forest (Kandeliaobovata) in Fugong village, Zhangzhou, Fujian, China. Strain TC11T grew at 15-45 °C (optimum, 35 °C), 0-8 % (w/v) NaCl (optimum, 1 %, w/v) and pH 5.5-9.5 (optimum, pH 7.5). Phylogenetic analyses revealed that strain TC11T belonged to a clade of the genus Pseudomonas and showed the highest sequence similarity of 98.4 % to Pseudomonas fluvialis ASS-1T, followed by Pseudomonas oleovorans subsp.oleovorans DSM 1045T (97.9 %), Pseudomonas indoloxydans JCM 14246T (97.7 %), Pseudomonas guguanensis JCM 18416T(97.6 %) and Pseudomonas alcaliphila JCM 10630T (97.5 %) on the basis of their 16S rRNA gene sequences. The DNA G+C content was 64.3 mol%. In silico DNA-DNA hybridization and average nucleotide identity values between strain TC11T and the reference strains were 19-22 % and 72-78 %, respectively. Studies based on the three housekeeping genes, rpoB, gyrB and rpoD, further confirmed that strain TC11T is a novel member of the genus Pseudomonas. The major fatty acids of strain TC11Twere C16 : 0, summed feature 8 (C18 : 1ω6c/C18 : 1ω7c) and summed feature 3 (C16 : 1ω7c/C16 : 1ω6c). The sole isoprenoid quinone was Q-9. The major polar lipids were phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol. Based on the phenotypic, chemotaxonomic and phylogenetic properties, strain TC11T represents a novel species of the genus Pseudomonas, for which the name Pseudomonasmangrovi sp. nov., is proposed. The type strain is TC11T (=KCTC 62159=MCCC 1K03499).
Psychromicrobium Xiongbiense sp. nov., an Actinobacterium Isolated from Forest Soil

Abstract:A novel Gram-stain-positive, oval-shaped, and non-flagellated bacterial strain YIM S02556 T was isolated from forest soil in Xiongbi Town, Shizong County, Qujing City, Yunnan Province, southwestern China. The strain exhibited high pairwise 16 S rRNA gene sequence similarity with Psychromicrobium lacuslunae (97.3%) and Psychromicrobium silvestre (96.3%). Strain YIM S02556 T exhibited an average nucleotide identity (ANI) of 72.5% with P . lacuslunae IHBB 11,108 T and 72.8% ANI with P . silvestre AK 20-18 T . The digital DNA-DNA hybridization (dDDH) value between strain YIM S02556 T and P . lacuslunae IHBB 11,108 T was 20.2%, while with P . silvestre AK 20-18 T , the dDDH value was 20.8%. Strain YIM S02556 T exhibited optimal growth at 28 °C, pH 7.0, without NaCl. Growth occurred within 10–37 °C, pH 5.0–8.0, and in the presence of up to 5% w/v NaCl concentration. The genome size was 3.1 Mbp with 64.2% G + C content. The predominant menaquinone was MK-8(H 4 ). The major cellular fatty acid was anteiso-C 15:0 . Based on the polyphasic analysis, strain YIM S02556 T (= KCTC 49,805 T = CCTCC AB2020166 T ) represents a novel Psychromicrobium species in which the name Psychromicrobium xiongbiense sp.nov. was proposed.

Noncanonical Wnt Signaling Promotes Obesity-Induced Adipose Tissue Inflammation and Metabolic Dysfunction Independent of Adipose Tissue Expansion

Abstract:Adipose tissue dysfunction plays a pivotal role in the development of insulin resistance in obese individuals. Cell culture studies and gain-of-function mouse models suggest that canonical Wnt proteins modulate adipose tissue expansion. However, no genetic evidence supports a role for endogenous Wnt proteins in adipose tissue dysfunction, and the role of noncanonical Wnt signaling remains largely unexplored. Here we provide evidence from human, mouse, and cell culture studies showing that Wnt5a-mediated, noncanonical Wnt signaling contributes to obesity-associated metabolic dysfunction by increasing adipose tissue inflammation. Wnt5a expression is significantly upregulated in human visceral fat compared with subcutaneous fat in obese individuals. In obese mice, Wnt5a ablation ameliorates insulin resistance, in parallel with reductions in adipose tissue inflammation. Conversely, Wnt5a overexpression in myeloid cells augments adipose tissue inflammation and leads to greater impairments in glucose homeostasis. Wnt5a ablation or overexpression did not affect fat mass or adipocyte size. Mechanistically, Wnt5a promotes the expression of proinflammatory cytokines by macrophages in a Jun NH2-terminal kinase–dependent manner, leading to defective insulin signaling in adipocytes. Exogenous interleukin-6 administration restores insulin resistance in obese Wnt5a-deficient mice, suggesting a central role for this cytokine in Wnt5a-mediated metabolic dysfunction. Taken together, these results demonstrate that noncanonical Wnt signaling contributes to obesity-induced insulin resistance independent of adipose tissue expansion.
Puerhibacterium puerhi gen. nov., sp. nov., a novel member of the family Promicromonosporaceae, isolated from Pu-erh tea pile-fermentation

Abstract:A Gram-staining positive aerobic bacterium, designated TLY-12T, was isolated from the Pu-erh tea pile-fermentation process in Pu'er city, Yunnan, China. Strain TLY-12T grew at 15-37 °C (optimum, 30 °C), pH 6.0-11.0 (optimum, pH 9.0) and 0-9.0% (w/v) NaCl (optimum, 3.0%). The major cellular fatty acids were anteiso-C15:0, C16:0 and iso-C16:0. The respiratory quinone were menaquinones MK-9 (H2) and MK-9 (H4). The polar lipids were phosphatidylglycerol (PG), diphosphatidylglycerol (DPG), phosphatidylinositol (PI), phosphoglycolipid (PGL), glycolipid (GL) and an unidentified phospholipid (PL). The peptidoglycan contained glutamic acid, aspartic acid, alanine and lysine, with the last named being the diagnostic diamino acid. Whole-cell sugars of the isolate were ribose, galactose and glucose. Phylogenetic analyses of 16S rRNA gene showed that this strain belonged to the family Promicromonosporaceae, and was most closely related to Isoptericola cucumis DSM 101603 T, which gave sequence similarity of 97.9%. Genome sequencing revealed a genome size of 3.91 Mbp and a G + C content of 75.0%. Average nucleotide identity and digital DNA-DNA hybridization values were all below the species threshold of described Promicromonosporaceae species. Genome phylogenetic analysis showed that strain TLY-12T formed a separate evolutionary branch, and was parallel to other related genera of Promicromonosporaceae. Based on the phylogenetic, phenotypic, chemotaxonomic and genome pairwise data, strain TLY-12T is considered to represent a novel species in a new genus in the family Promicromonosporaceae, for which the name Puerhibacterium puerhi gen. nov, sp. nov. is proposed. The type strain is TLY-12T (= CGMCC 1.17157T = KCTC 49467T).

Abstract:Three microaerophilic bacterial strains, designated SG22 T , SG63 T and SG29 T were isolated from paddy soils in PR China. Cells of these strains were Gram-staining-negative and long rod-shaped. SG22 T , SG63 T and SG29 T showed the highest 16S rRNA gene sequence similarities with the members of the genus Anaeromyxobacter . The results of phylogenetic and phylogenomic analysis also indicated that these strains clustered with members of the genus Anaeromyxobacter . The main respiratory menaquinone of SG22 T , SG63 T and SG29 T was MK-8 and the major fatty acids were iso-C 15 : 0 , iso-C 17 : 0 and C 16 : 0 . SG22 T , SG29 T and SG63 T not only possessed iron reduction ability but also harboured genes ( nifHDK ) encoding nitrogenase. The genomic DNA G+C contents of SG22 T , SG63 T and SG29 T ranged from 73.3 to 73.5 %. The average nucleotide identity (ANI) and digital DNA–DNA hybridisation (dDDH) values between SG22 T , SG63 T and SG29 T and the closely related species of the genus Anaeromyxobacter were lower than the cut-off values (dDDH 70 % and ANI 95–96 %) for prokaryotic species delineation. On the basis of these results, strains SG22 T , SG63 T and SG29 T represent three novel species within the genus Anaeromyxobacter , for which the names Anaeromyxobacter terrae sp. nov., Anaeromyxobacter oryzisoli sp. nov. and Anaeromyxobacter soli sp. nov., are proposed. The type strains are SG22 T (= GDMCC 1.3185 T = JCM 35581 T ), SG63 T (= GDMCC 1.2914 T = JCM 35124 T ) and SG29 T (= GDMCC 1.2911 T = JCM 35123 T ).
