Abstract:It is well established that the piezoelectric effect plays an important physiological role in bone growth, remodeling and fracture healing. Barium titanate, as a well-known piezoelectric ceramic, is especially an attractive material as a scaffold for bone tissue engineering applications. In this regard, we tried to fabricate a highly porous barium titanate based scaffolds by foam replication method and polarize them by applying an external electric field. In order to enhance the mechanical and biological properties, polarized/non-polarized scaffolds were coated with gelatin and nanostructured HA and characterized for their morphologies, porosities, piezoelectric and mechanical properties. The results showed that the compressive strength and piezoelectric coefficient of porous scaffolds increased with the increase of sintering temperature. After being coated with Gel/HA nanocomposite, the interconnected porous structure and pore size of the scaffolds almost remain unchanged while the Gel/nHA-coated scaffolds exhibited enhanced compressive strength and elastic modulus compared with the uncoated samples. Also, the effect of polarizing and coating of optimal scaffolds on adhesion, viability, and proliferation of the MG63 osteoblast-like cell line was evaluated by scanning electron microscope (SEM) and MTT assay. The cell culture experiments revealed that developed scaffolds had good biocompatibility and cells were able to adhere, proliferate and migrate into pores of the scaffolds. Furthermore, cell density was significantly higher in the coated scaffolds at all tested time-points. These results indicated that highly porous barium titanate scaffolds coated with Gel/HA nanocomposite has great potential in tissue engineering applications for bone tissue repair and regeneration.

Abstract:Calcium phosphate ceramics are rapidly degraded faster than the rate of tissue formation, which makes no Scaffolding enough strength and makes it unsuitable for applications that are bearing the load. Therefore, Bio-Ceramic a new generation of ceramics, with calcium silicate (Ca-Si) attention attracted to. Methodology: One way to improve the biochemical properties of these metal ions into their structure is Bio- Ceramic Add. Nowadays, along with approaching the normal structure of bones and making use of the properties of nanotechnology researchers to build Nano-composite scaffolds were driven. Results: The results of powder X-ray diffraction is shown in Figure 1. The pattern observed in this figure shows the corresponding Hardystonite reference card. Hardystonite powder pattern of tetragonal structure with a=7.8287 and c=5.0140 angstrom fixed network specifies. In addition, sharp peaks indicating the crystalline structure Hardystonite after heat treatment. Conclusion: Bioactive particles in the inner and outer composite is increased bioactivity. By placing the samples in a solution of calcium and phosphate ions in the SBF solution and those other elements on the surface sediment samples from the MAP images visible.

Abstract:The requirement for bone grafts needed for filling bone defect and fracture is increasing annually due to global ageing and a great amount of bone trauma cases, whereas the conventional bioceramic implants is suboptimal for bone regeneration and inconvenient for implantation operation. Here we describe the 3D printed granule-type bioceramics and evaluate the porous architectures and biological performances of two types of Mg-mediating silicate bioceramics. The 6 % and 9 % Mg doped wollastonite (CSi-6 and CSi-9) and pure or 9 % Mg-rich akermanite (AKT and AKT-9) granules had a gyroid-pore geometry and exhibited different sintering and mechanical properties. The granules were also evaluated on dissolution and osteogenic cell response in vitro and osteogenic capacity in rabbit femoral condylar defect in vivo for different time stages. It was indicated that Mg doping into CSi could contribute appreciable compressive resistance but low bio-dissolution in comparison with the AKT. These silicate bioceramics showed similar osteoblastic viability and the Mg-rich AKT promoted mesenchymal stem cell differentiation in vitro. The surface-curved granules exhibited high-filling networks in the bone defect and promoted bone growth during the early and mid-term stages (40 & 120 days), and especially the wollastonite groups were degraded quickly and akermanite groups stimulated faster bone formation. Taken together, this study demonstrated the potential of Mg-tuning silicate bioceramics in achieving high-efficient bone repair and important implications for optimizing the porous granule fillers design by advanced additive manufacture technique to match the clinical translation with high performance.

Abstract:Due to the increasing cases of bone damage and bone graft demand, bone-repair technology has great social and economic benefits and the manufacturing of artificial bone implants has become a focus in the domain of regenerative therapy. Considering that the traditional manufacturing process cannot effectively control the overall size of the scaffold, the diameter and shape of micropores, and the interoperability of micropores, 3D printing technology has emerged as a focal point of research within the realm of bone tissue engineering. However, the printing accuracy of extrusion-based biological 3D printing techniques is low. In this research, we utilized three-dimensional printing technology to develop high-precision magnesium-containing silicate (CSi-Mg) scaffolds. The precision of this innovative method was scrutinized and the influence of pore size on scaffold strength was systematically analyzed. Furthermore, the influence of the pore architecture on the sidewalls of these 3D-printed scaffolds was evaluated in terms of mechanical properties. The CSi-Mg scaffold, post a 3-week immersion in a simulated body of fluid, demonstrated a high modulus of elasticity (exceeding 404 MPa) and significant compressive strength (beyond 47 MPa). Furthermore, it exhibited commendable bioactivity and biodegradability. These results suggest that the high-precision 3D-printed CSi-Mg scaffolds hold great promise for addressing challenging bone defect cases.

Abstract:Nano phase hydroxyapatite (HA) bioceramics have gained importance in the biomedical field due to their superior biological properties. In this study, nanostructured HA coating was used to increase the bioactivity of a piezoelectric bioceramic, barium titanate (BT). Early reports on the influence of collagen piezoelectricity in remodeling of bone have attracted many researchers to piezoelectric bioceramics such as BT. Hence; porous BT was used as the matrix of a new bone graft composite and then coated with nanostructured HA. BT ceramic was foamed via a direct foaming method with a spray of polyurethane foam. The surface of the foam voids was coated with HA via sol–gel and dip‐coating methods. X‐ray diffraction (XRD), transmission electron microscopy (TEM), and scanning electron microscopy (SEM) techniques were used to characterize the prepared coated foam. XRD and TEM analysis showed that the HA coating had a nanostructure with crystallite size of 20–30 nm. SEM images of the prepared samples showed that the HA coating has about 25 µm thickness. The bioactivity of the prepared composite was evaluated in an in vitro study. The variation of Ca2+ and PO43− ions versus time in simulated body fluid (SBF) solution were measured by inductively coupled plasma (ICP) analysis during 1 month and the results showed that the mineralization of calcium phosphate (Ca‐P) on HA coated porous samples was much more than that in non‐coated sample. The SEM micrographs and energy‐dispersive X‐ray spectroscopy (EDS or EDX) analysis of the samples after 1 month of immersing in SBF confirm that Ca‐P phase (bone‐like apatite) was significantly mineralized on HA coated porous BT samples. It was concluded that the nanostructured HA coating would improve the bioactivity of BT foam. In this study, a new bone graft composite, porous barium titanate (BT) with nanostructured hydroxyapatite (HA) coating is prepared. Analyses show that the HA coating has a nanostructure with crystallite size of 20–30 nm. The in vitro bioactivity tests of the prepared composites show that the nanostructured HA coating would improve the bioactivity of BT foam.

With the increase of sintering temperature, the piezoelectric coefficient d33 of the composite ceramics first increased and then decreased, reaching a maximum of 7.80 pC/N at 1070 °C. The doping of AK would lead to a significant decrease in the piezoelectric property, and the AK content should not exceed 10 wt% to achieve the piezoelectric coefficient of human bone. After immersion in Tris-HCl solution for 28 days, the weight loss rate of the composite ceramics doped with 15 wt% AK could reach 8.36%. The combination of KNN and AK promotes the application of piezoelectric bone scaffolds in the field of bone repair.

Abstract:The discovery of the piezoelectric properties of natural bone and their influence on bone growth within scaffolds has spurred researchers to investigate a range of bio-piezoelectric composite materials. By exploring these materials, researchers aim to better understand how piezoelectricity can be harnessed to promote bone growth and tissue regeneration, potentially leading to new breakthroughs in the field of regenerative medicine. In this study, the piezoelectric material, Bi 0.5 Na 0.5 TiO 3 -0.6BaTiO 3 /nano ZnO (BNT6BT/ZnO) and the biological hydroxyapatite (HA) were separately synthesized using the sol-gel method. The composite samples consisting of BNT6BT/ZnO and HA were fabricated using isostatic cold pressing . The composition and phase structure were studied using the X-ray diffraction (XRD) analysis, and the scanning electron microscopy (SEM) was used to study the microstructure and morphology. The presence of different phases and the distribution of elements were examined using EDAX elemental analysis and its corresponding maps. Among the composite samples the one containing 10 wt% HA (BNT6BT/ZnO-10HA) possessed the best dielectric properties . The Vickers hardness measurements showed that the addition of HA to the piezoelectric phase BNT6BT/ZnO results in higher hardness values, which expects to increase the fracture toughness of HA due to the higher elastic modulus to hardness ratio (E/H) of composite samples compared to pure HA. The in vitro bioactivity of the samples was tested in simulated body fluid (SBF) solution and the cell culture analysis was performed to examine the biocompatibility of the samples. Accordingly, apatite was deposited on the surface of the samples and all of the composite samples were bioactive. Furthermore, the BNT6BT/ZnO-10HA sample containing the largest amount of deposit showed the highest optical density (OD) in the MTT test.

Abstract:The utilization of piezoelectric materials in bone implants is appealing due to the inherent piezoelectric property of natural bone. The intrinsic electrical characteristics of piezoelectric biomaterials enhance antibacterial activities, biocompatibility, and bioactivity properties. This study delves into investigating the antibacterial properties, biocompatibility, and bioactivity of three-component biocomposites: sodium potassium niobate (KNN)-barium titanate (BT)-hydroxyapatite (HA). The combination of sodium potassium niobate and barium titanate, possessing suitable piezoelectric properties, with hydroxyapatite, known for its favorable biological properties, enhances the requisite properties for a bone implant. Among the various compositions studied, the combination comprising 70 wt% of the piezoelectric component (KNN-BT) and 30 wt% hydroxyapatite, labeled as 30HKB, emerged as the most optimal blend in terms of density, morphotropic phase boundary, and other biological tests conducted. Following polarization, the antibacterial efficacy of 30HKB against S. aureus bacteria cells increased by 61 %. Furthermore, the growth and adhesion of MC3T3-E1 osteoblast cells suggest enhanced biocompatibility of the 30HKB composite attributed to surface polarization. The surface charges generated by polarization facilitated the absorption of Ca2+ ions, as well as the interaction of HPO4 - and OH- ions with the precipitated Ca2+ ions, leading to the formation of the CaP layer. Hence, polarized piezoelectric ceramics exhibit heightened bioactivity compared to their non-polarized counterparts.

Fabrication of β-TCP/ Akermanite composite scaffold via DLP and in-situ modification of micro-nano surface morphology for bone repair

Abstract:Although β-TCP bioceramics possess favorable biocompatibility and bioactivity, their inherent brittleness restricts further applications. In this study, we aimed to enhance the mechanical performance and biocompatibility of β-TCP by incorporating 15% Akermanite (AK) into the structure. The fabrication process involved the use of digital light processing (DLP) technology to fabricate ceramic green bodies, followed by high-temperature debinding and sintering to obtain the ceramic scaffold. Subsequently, we employed an in-situ growth technique to create micro/nano surface topography on the scaffolds, facilitating synergistic regulation of cell behavior in conjunction with the controlled release of bioactive agents. Scanning electron microscopy (SEM) revealed that the addition of AK enlarged the grain size and reduced the number of micropores in the composite scaffold. Furthermore, after the in-situ growth treatment, a significant increase in micro/nano surface features was observed. Mechanical testing demonstrated that the incorporation of AK effectively improved the mechanical properties of the composite scaffold, resulting in an increase in strength of approximately 20%. Cell experiments indicated that AK supplementation further enhanced the material's cell compatibility, and the presence of micro/nano structures effectively promoted cell adhesion, proliferation, and expression of osteogenic genes. Consequently, we propose a strategy for fabricating porous bioceramics that stimulate bone growth, exhibit high strength, and ameliorate the degradation microenvironment. This approach opens up promising avenues for future applications in bone tissue engineering.

Abstract:Using artificial biomaterials in bone regenerative medicine for highly efficient osteoconduction into the bone defect to decrease the bone healing time is still a challenge. In this research, magnesium (Mg)-doped wollastonite (similar to 10% Mg was substituted for calcium (Ca) in beta-CaSiO3) (CSi-Mg10) bioceramic scaffolds with ultrahigh mechanical strength were fabricated using ceramic ink writing three dimensional (3D) printing. To evaluate the potential of other additives on the new bone regeneration efficiency, beta-tricalcium phosphate (beta-TCP) was introduced to the CSi-Mg10 ceramic ink at a concentration of 15% and the biphasic bioceramic scaffolds (CSi-Mg10/TCP15) were also fabricated using 3D printing. The mechanical characterization indicated that introduction of beta-TCP led to nearly 50% mechanical decay, although the effect of the two heating schedules (one- and two-step sintering) on the compressive and flexural strengths of the scaffolds was significantly different. The bone regeneration results in critical sized calvarial defect of rabbits showed that the CSi-Mg10/TCP15 scaffolds displayed a markedly higher osteogenic capability than those on the CSi-Mg10 and beta-TCP scaffolds after eight weeks, and reached similar to 35% new bone tissue regeneration at 12 weeks postoperatively. These findings demonstrate that the CSi-Mg10/TCP15 bioceramic scaffolds can be well suited for stimulating in situ bone regeneration and for use in tissue engineering applications.

Abstract:Piezoelectric materials are the group of smart materials which have been recently developed for biomedical applications, such as bone tissue engineering. These materials could provide electrical signals with no external source power making them effective for bone remodeling. Between various types of materials, BaTiO3 and CaTiO3 are nontoxic piezoelectric ceramics, which recently have been introduced for bone tissue engineering. It is expected that, the combination of these two ceramics could provide suitable piezoelectricity, bioactivity and biocompatibility for bone tissue engineering applications. The aim of this research is to synthesize (BaxCa1-x)TiO3 (x= 0, 0.6, 0.8, 0.9 and 1) nanopowder using sol-gel method. Moreover, the incorporation of Ca+2 ions in the structure of (BaxCa1-x)TiO3 nanoparticles was chemically, structurally and biologically studied. X-ray diffraction (XRD) and scanning electron microscopy (SEM) studies confirmed the role of substituted Ca content on the chemical properties and morphology of particles. Indeed, increasing the amounts of Ca+2 ions resulted in the reduced crystallite size. While incorporation of more than 20 at.% Ca resulted in the formation of a biphasic structure, monophasic solid solution without any secondary phase was detected at less Ca content. Moreover, SEM images revealed that Ca substitution reduced particle size from 70.5 ±12 nm to 52.4 ±9 nm, while the morphology of synthesized powders did not significacntly change. Furthermore, incorporation of upon 10 at.% Ca content within (BaxCa1-x)TiO3 significantly promoted MG63 proliferation compared to pure CaTiO3.

Abstract:Herein we report the synthesis of a piezopolymer composed of chitosan (CS) / barium titanate (BT) nanoparticles (NPs) with enhanced biocompatibility, non-toxicity, and piezoelectric behavior that can be advantageously used in biomedical applications. FTIR and SEM measurements were performed to assess the mechanism of interaction between CS matrix and BT NPs and their correlation with the biological responses. The dielectric measurements of the as prepared composites reveals that incorporation of 50% BT NPs in the chitosan matrix lead to a steady increase of the dielectric constant as compared with neat chitosan films. The ferroelectric behavior of the sample was confirmed by the values of the loss factor (0.21 – 0.003) in the analyzed frequency range (10-1 – 106 Hz). This behavior suggests that ferroelectric CS/BT nanocomposites can act as active material that promotes accelerate bone regeneration.

Abstract:In the case of a large bone defect, the human endogenous electrical field is no longer sufficient. Therefore, it is necessary to support structural electrical bone scaffolds. Barium titanate (BT) has received much attention in bone tissue engineering applications due to its biocompatibility and ability to maintain charged surfaces. However, its processability is poor and it does not have the biological activity to promote mineralization, which limits its use in bone repair. In this paper, a composite bone scaffold with excellent piezoelectric properties was prepared by combining 20 wt% calcium silicate. The influence of the light curing process on the properties of the piezoelectric biological scaffold was investigated by comparing it with the traditional piezoelectric ceramic molding method (dry pressing). Despite the structural features of 3D printing (layered structure, pore features), the piezoelectric and mechanical properties of the scaffold are weakened. However, 3D-printed scaffolds can combine structural and piezoelectric properties, which makes the 3D-printed scaffold more effective in terms of degradation and antibacterial performance. In terms of cell activity, piezoelectric properties attract proteins and nutrients into the scaffold, promoting cell growth and differentiation. Besides, it is undeniable that the pore structure of the scaffolds plays an important role in the biological properties. Finally, the 3D printed scaffolds have excellent antimicrobial properties due to the redox reaction under piezoelectric effect as well as structural characterization.

Although many studies have attempted to develop piezoelectric biomaterials for bone regeneration, post-treatment steps, such as sintering, are always needed. In this study, we prepared an injectable and piezoelectric bone substitute based on nanosized BaTiO3 (nBT)-added calcium phosphate silicate (CPS). The impacts of nBT on the CPS handleability and mechanical strength were characterized, and show that adding nBT could improve the CPS handleability but affect the CPS mechanical strength in a concentration-dependent manner (from 25.3 ± 1.0 MPa for 10BC to 13.5 ± 1.0 MPa for 40BC). In addition, our approach could fabricate a piezoelectric bone substitute with comparable piezoelectricity to the native bone without any post-treatment. The in vitro analyses demonstrated that nBT/CPS was biocompatible and could promote osteoblast differentiation. In conclusion, our results strongly indicate that the injectable formulation based on nBT/CPS can be a promising candidate in bone tissue engineering, and further research is needed to investigate the biomaterial's performance in bone defect animal models.

Abstract:Nanomaterials with piezoelectric properties can significantly improve the applicability of polymers used in tissue engineering applications. In this study, we report the one-step synthesis of a novel hybrid piezoelectric composite comprising barium titanates and boron nitride nanotubes. This composite is distinguished by its unique microstructures, including nanoflakes, triangular boron nitride structures, and fiber-like boron nitride nanotube configurations, which contribute to its enhanced piezoelectric properties. The composite was incorporated into a chitosan-based tissue scaffold and evaluated in vitro. Electric-responsive Human Osteoblast cells cultured on the scaffolds are exposed to low-frequency ultrasound stimulation during cell growth. The biocompatibility, cell adhesion, alkaline phosphatase activities, and mineralization of osteoblast cells on the piezo-composite scaffolds were evaluated. The results show that the hybrid piezoelectric composite significantly enhances the properties of chitosan-based scaffold.

Abstract:Application of bioactive materials as synthetic bone graft substitutes in regenerative medicine has seen great evolution over the past decades in treating challengeable bone defects. However, balancing the preparation conditions and biological performances of inorganic biomaterials remain a great challenge, especially when there is lack of biomaterial design on how to control component distribution and how pathological bone responds to the biomaterial stimulations and osteogenesis. Here, our objective is to develop yolk-shell Ca-silicate microspheres and to investigate the potential biological performances to overcome the limitations in repair of osteoporotic bone defects. The introduction of beta-calcium silicate (CaSiO3) or mesoporous bioactive glass (MBG) into self-curing beta-dicalcium silicate (Ca2SiO4) cement shell to form spherical granules (CaSiO3@Ca2SiO4, MBG@Ca2SiO4) was to retain the physicochemical property and/or microstructure of each component for optimizing bioactive ion release that could maximize osteostimulation in osteoporosis. We report a scalable shape-controlled mild fabrication protocol to yield the yolk-shell granules, endowing to different phases in yolk layer and interconnected macropore networks in the closely packed granule scaffolds. This unique heterostructure preparation is governed by coaxially aligned bilayer nozzle, inorganic powders and biocompatible binders. Extensive in vitro and in vivo evaluation showed that the CaSiO3@Ca2SiO4 and MBG@Ca2SiO4 granules exhibited many superior properties such as controllable ion release, improved biodegradation and enhanced osteogenic capability in comparison with the pure Ca2SiO4@Ca2SiO4, thereby opening new mild-condition approach in fabricating osteogenesis-tailored silicate biomaterials for bone regenerative medicine, especially for efficient reconstruction of challenging pathological bone defects.
Three-dimensional printing akermanite porous scaffolds for load-bearing bone defect repair: An investigation of osteogenic capability and mechanical evolution

Abstract:Some Ca–Mg-silicate ceramics have been widely investigated to be highly bioactive and biodegradable, whereas their osteogenic potential and especially biomechanical response in the early stage in vivo are scarcely demonstrated. Herein, the osteogenesis capacity and mechanical evolution of the akermanite (Ca 2 MgSi 2 O 7 ) porous materials manufactured by ceramic ink writing three-dimensional printing technique were investigated systematically in a critical size femur defect model, in comparison with the clinically available β-tricalcium phosphate porous bioceramic. Such three-dimensional printed akermanite scaffolds possess fully interconnected pores of ∼280 × 280 μm in size and over 50% porosity with appreciable compressive strength (∼71 MPa), that is 7-fold higher than that of the β-tricalcium phosphate porous bioceramics (∼10 MPa). After 6 weeks and 12 weeks of implantation, the percentage of newly formed bone and more new bone was observed in the akermanite group as compared with the β-tricalcium phosphate group ( p < 0.01). Moreover, significant higher mRNA expressions of osteogenic genes were detected in the akermanite group by PCR analysis ( p < 0.01). The in vivo mechanical strength decreased during the process of implantation, but maintained a relative high level (∼14 MPa) which was still higher than that of the host cancellous bone (5–10 MPa) at 12 weeks post-implantation. On the contrary, the β-tricalcium phosphate scaffold always exhibited a very low mechanical strength (∼8 MPa). These results suggest that the three-dimensional printed akermanite scaffolds are promising for the bone tissue regeneration and repair of load-bearing bone defects.

Abstract:In recent years, porous titanium (Ti) scaffolds with BaTiO3 coatings have been designed to promote bone regeneration. However, the phase transitions of BaTiO3 have been understudied, and their coatings have yielded low effective piezoelectric coefficients (EPCs < 1 pm/V). In addition, piezoelectric nanomaterials bring many advantages in eliciting cell-specific responses. However, no study has attempted to design a nanostructured BaTiO3 coating with high EPCs. Herein, nanoparticulate tetragonal phase BaTiO3 coatings with cube-like nanoparticles but different effective piezoelectric coefficients were fabricated via anodization combining two hydrothermal processes. The effects of nanostructure-mediated piezoelectricity on the spreading, proliferation, and osteogenic differentiation of human jaw bone marrow mesenchymal stem cells (hJBMSCs) were explored. We found that the nanostructured tetragonal BaTiO3 coatings exhibited good biocompatibility and an EPC-dependent inhibitory effect on hJBMSC proliferation. The nanostructured tetragonal BaTiO3 coatings of relatively smaller EPCs (<10 pm/V) exhibited hJBMSC elongation and reorientation, broad lamellipodia extension, strong intercellular connection and osteogenic differentiation enhancement. Overall, the improved hJBMSC characteristics make the nanostructured tetragonal BaTiO3 coatings promising for application on implant surfaces to promote osseointegration.
