Abstract:The pathosystem of tomato and leaf mold fungal pathogen Cladosporium fulvum is a model system to study molecular mechanism of interaction between plant and pathogen.Four C.fulvum avirulence(Avr) genes have been cloned.All the Avr genes encode small molecular weight proteins with even numbers of cysteines,and their existence and variance are different in the virulent strains.Avr4 is of chitin-binding activity,while Avr9 might be involved in nitrogen regulation in the fungus.Seven functional tomato resistance(R) genes to C.fulvum have been cloned.They are clustered with nonfunctional homologues in complex loci and named as Hcr(homologues of C.fulvum resistance gene Cf) genes.The Cf proteins,which are anchored at plasma membrane with the main body outside,mainly consist of a leucine-rich repeat(LRR) and a transmembrane domain.Recognition of an Avr by the complementary Cf complies with the "guard" hypothesis.In re-(cognition) complex of Avr2 by Cf-2,the guarded protein is protease Rcr3.Avr recognition by a Cf rapidly eli-(cits) downstream signaling and defense responses,including occurrence of hypersensitive response(HR) and oxidative burst,activation of K~+ ion channel,a variety of protein kinases,SGT1,thioredoxin and phosphati-(dic) acid signaling pathway.Environmental conditions,including temperature,humidity and light,significantly affect Cf-dependent HR and resistance.Different Cf/Avr interactions might be distinct in Avr recognition and downstream signal transduction pathways.

Abstract:Tomato Cf genes confer resistance to the leaf mold pathogen Cladosporium fulvum. The Avr4/Cf-4- and Avr9/Cf-9-dependent hypersensitive responses (HRs) are distinct in cell death pattern, intensity, and sensitivity to environmental conditions. To understand the mechanism resulting in these differences, comparative transcript profiling for Avr4/Cf-4- and Avr9/Cf-9-dependent defence gene expression by cDNA-AFLP was performed previously. 367 ACE (Avr/Cf-elicited) transcript-derived fragments (TDFs) were identified, among which 189 were cloned and sequenced. In this study, we report another 89 ACE fragments. These ACE genes were associated with: defence, signal transduction, HR and cell death, transcriptional regulation, metabolism, protein synthesis, photosynthesis, membrane fusion, secretion and trafficking, miscellaneous biological processes, and genes with unknown function or with no significant similarity to known sequences. Among these sequences 43 (potentially encoding 36 types of proteins) were identified for the first time as genes differentially expressed during the development of Avr/Cf-dependent HR. Sequence and expression data from this study further support that transcription is reprogrammed to promote defence response and HR and repress photosynthesis in the Avr/Cf HR+ seedlings.

Abstract:Fusarium asiaticum is a critical pathogen of Fusarium head blight (FHB) in the southern part of China. The fungicide phenamacril has been extensively used for controlling FHB in recent years, which reduced both FHB severity and mycotoxin production. Our previous report indicated that resistance of F. asiaticum to phenamacril was related to mutations in myosin5. A recent article revealed that the resistance level of phenamacril-resistant mutants was associated with the genotypes of myosin5 in these mutants. In total, we obtained 239 resistant isolates by fungicide domestication, and 82 resistant mutants were randomly selected for further study. Of these mutants, 25.6, 7.3, and 67.1% showed low resistance (LR), moderate resistance (MR), and high resistance (HR), respectively, to phenamacril determined by 50% effective concentration values. Point mutations A135T, V151M, P204S, I434M, A577T, R580G/H, or I581F led to LR. Point mutations S418R, I424R, and A577G were responsible for MR and point mutations K216R/E, S217P/L, or E420K/G/D conferred HR. Interestingly, all of the mutations concentrated in the myosin5 motor domain and mutations conferring HR occurred at codon 217 and 420, which we called the core region. Homology modeling revealed that mutations far from the core region led to a lower resistance degree. Phenotype assays revealed that the most highly resistant mutants did not significantly change pathogenicity but decreased conidia production compared with the wild type, which may slow down the formation of the resistant pathogen population in the fields.
A Novel Strain of Fusarium oxysporum Alternavirus 1 Isolated from Fusarium oxysporum f. sp. melonis Strain T-BJ17 Confers Hypovirulence and Increases the Sensitivity of Its Host Fungus to Difenoconazole and Pydiflumetofen

Abstract:In the current study, a novel strain of Fusarium oxysporum alternavirus 1 (FoAV1) was identified from the Fusarium oxysporum f. sp. melonis (FOM) strain T-BJ17 and was designated as Fusarium oxysporum alternavirus 1-FOM (FoAV1-FOM). Its genome consists of four dsRNA segments of 3515 bp (dsRNA1), 2663 bp (dsRNA2), 2368 bp (dsRNA3), and 1776 bp (dsRNA4) in length. Open reading frame 1 (ORF1) in dsRNA1 was found to encode a putative RNA-dependent RNA polymerase (RdRp), whose amino acid sequence was 99.02% identical to that of its counterpart in FoAV1; while ORF2 in dsRNA2, ORF3 in dsRNA3, and ORF4 in dsRNA4 were all found to encode hypothetical proteins. Strain T-BJ17-VF, which was verified to FoAV1-FOM-free, was obtained using single-hyphal-tip culture combined with high-temperature treatment to eliminate FoAV1-FOM from strain T-BJ17. The colony growth rate, ability to produce spores, and virulence of strain T-BJ17 were significantly lower than those of T-BJ17-VF, while the dry weight of the mycelial biomass and the sensitivity to difenoconazole and pydiflumetofen of strain T-BJ17 were greater than those of T-BJ17-VF. FoAV1-FOM was capable of 100% vertical transmission via spores. To our knowledge, this is the first time that an alternavirus has infected FOM, and this is the first report of hypovirulence and increased sensitivity to difenoconazole and pydiflumetofen induced by FoAV1-FOM infection in FOM.

Abstract:The effects of polymer structure and water-macromolecule interactions on proton relaxation in an aqueous model polymer have been investigated using quantitative measurements of magnetization transfer. Polyacrylamide gels composed of 95% water, 5% comonomers acrylamide and N,N'-methylene-bis-acrylamide were studied. The structure and rigidity were varied by changing the cross-linking density of the polymer. The polymer showed a biphasic change in transverse relaxation with increasing cross-linking density which was accompanied by a sudden increase in magnetization transfer above 40% cross linking. This change may be attributed to the formation of rigid domains in the polymer which exhibit solid-like behavior with a short T2 (11 microseconds) and a Gaussian lineshape. Water-macromolecule interactions were controlled by varying the pH of the gel. At high pH (> 8), there was an increase in magnetization transfer and transverse relaxivity consistent with a chemical-exchange-mediated interaction between water protons and the polymer. By analyzing the system as two proton reservoirs coupled by magnetization exchange, the proton populations, intrinsic relaxation rates, and exchange rates were estimated, for different degrees of cross linking and pH. This model affords useful insights into the relevance of both supramolecular structure and chemical exchange on relaxation in tissues.
Suppression of plant resistance gene-based immunity by a fungal effector

Abstract:The innate immune system of plants consists of two layers. The first layer, called basal resistance, governs recognition of conserved microbial molecules and fends off most attempted invasions. The second layer is based on Resistance (R) genes that mediate recognition of effectors, proteins secreted by pathogens to suppress or evade basal resistance. Here, we show that a plant-pathogenic fungus secretes an effector that can both trigger and suppress R gene-based immunity. This effector, Avr1, is secreted by the xylem-invading fungus Fusarium oxysporum f.sp. lycopersici (Fol) and triggers disease resistance when the host plant, tomato, carries a matching R gene (I or I-1). At the same time, Avr1 suppresses the protective effect of two other R genes, I-2 and I-3. Based on these observations, we tentatively reconstruct the evolutionary arms race that has taken place between tomato R genes and effectors of Fol. This molecular analysis has revealed a hitherto unpredicted strategy for durable disease control based on resistance gene combinations.

Abstract:E‐selectin (CD62E, formerly termed ELAM‐1) is a cytokine‐inducible adhesion molecule which mediates the binding of neutrophils, monocytes, and skin homing T‐cells. The murine homologue of E‐selectin has been cloned. A monoclonal antibody (21KC10) was used here to study immunohistochemically the expression and regulation of murine E‐selectin in vitro and in vivo. As described for the human system, there was no staining of normal endothelium in skin and other tissues. LPS and tumour necrosis factor‐alpha (TNF‐α), but not interleukin‐4 (IL‐4) or interferon‐gamma (IFN‐γ), induced a transient expression of E‐selectin, both when injected in vivo and when added to endothelial cell lines in vitro. To analyse temporal expression of E‐selectin under pathophysiological conditions in vivo, we chose two murine models of inflammation: allergic (ACD) and irritant contact dermatitis (ICD). Expression of E‐selectin was found to be induced on vascular endothelium of post‐capillary venules in both ACD and ICD. In ICD, maximal staining of endothelial cells occurred earlier than in ACD. Expression of E‐selectin during ICD and ACD was then compared between strains of mice which differ with regard to the intensity of their inflammatory reaction. BALB/c mice, which in contrast to C57BI/6 mice show a denser infiltrate and prolonged influx of granulocytes and monocytes, revealed a more pronounced and more prolonged expression of E‐selectin than C57BI/6 mice. This held true for both ACD and ICD, and in each case, peak expression of E‐selectin was associated with the highest density of the leukocytic infiltrate. This study thus reveals regulatory mechanisms involved in the expression of murine E‐selectin in vivo and in vitro. It also demonstrates a correlation between endothelial expression of E‐selectin and the genetically determined intensity of the inflammatory response.

Abstract:The Cf-2 gene of tomato confers resistance to strains of the biotrophic pathogenic fungus Cladosporium fulvum carrying avirulence gene Avr2. To allow dissection of the biochemical mechanism of perception of AVR2 by Cf-2, we set out to clone the Avr2 gene. Here, we report the functional cloning of Avr2 cDNA, based on the induction of a hypersensitive response (HR) by the encoded AVR2 protein in Cf2 tomato plants. Analysis of strains of C. fulvum that are virulent on Cf2 tomato lines revealed various independent frameshift mutations in the Avr2 open reading frame (ORF) and a point mutation resulting in a premature stop codon. All modifications result in the production of truncated AVR2 proteins. Interestingly, an additional modification involves the insertion of a LINE-like element, Cfl1, in the Avr2 ORF. Cfl1 is the first LINE-like element identified in C. fulvum and provides the first example of loss of avirulence of a plant pathogen caused by insertion of a retrotransposable element in an Avr gene. Rcr3 represents an additional plant protein that is specifically required for Cf-2-mediated resistance. Analysis of two different rcr3 mutant Cf2 tomato plants revealed that their ability to respond to AVR2 with a HR correlates with their degree of resistance to AVR2-producing strains of C. fulvum. These data support a role for Rcr3 in the perception of AVR2 by Cf-2.

Abstract:Biofilm formation is important for virulence of a large number of plant pathogenic bacteria. Indeed, some virulence genes have been found to be involved in the formation of biofilm in bacterial fruit blotch pathogen Acidovorax citrulli. However, some virulent strains of A. citrulli were unable to format biofilm, indicating the complexity between biofilm formation and virulence. In this study, virulence-related genes were identified in the biofilm-defective strain A1 of A. citrulli by using Tn5 insertion, pathogenicity test, and high-efficiency thermal asymmetric interlaced PCR (hiTAIL-PCR). Results from this study indicated that 22 out of the obtained 301 mutants significantly decreased the virulence of strain A1 compared to the wild-type. Furthermore, sequence analysis indicated that the obtained 22 mutants were due to the insertion of Tn5 into eight genes, including Aave 4244 (cation diffusion facilitator family transporter), Aave 4286 (hypothetical protein), Aave 4189 (alpha/beta hydrolase fold), Aave 1911 (IMP dehydrogenase/GMP reductase domain), Aave 4383 (bacterial export proteins, family 1), Aave 4256 (Hsp70 protein), Aave 0003 (histidine kinase, DNA gyrase B, and HSP90-like ATPase), and Aave 2428 (pyridoxal-phosphate dependent enzyme). Furthermore, the growth of mutant Aave 2428 was unaffected and even increased by the change in incubation temperature, NaCl concentration and the pH of the LB broth, indicating that this gene may be directly involved in the bacterial virulence. Overall, the determination of the eight pathogenicity-related genes in strain A1 will be helpful to elucidate the pathogenesis of biofilm-defective A. citrulli.

Abstract:Fourteen isolates of Fusarium were isolated from wilt affected tomato samples collected from 10 different states of India. Characterization of the fungal cultures based on morphology and sequencing of ITS rDNA revealed that they belonged to Fusarium oxysporum f.sp. lycopersici (Fol). Pathogenicity assay on two susceptible tomato cultivars showed all the 14 isolates were pathogenic and categorized in high-, moderate- and low-virulent groups. Differential host assay on Bonny Best (no resistant gene), UC82-L (harboring I-1), Fla.MH1 (harboring I-1 and I-2) and I3R-1 (harboring I-1, I-2 and I-3) tomato genotypes and PCR amplification with race-specific primers indicated that all the Fusarium isolates infecting tomato in India were belonging to race 1. Molecular diversity analysis based on ISSR markers revealed the presence of 3 distinct groups of Fol isolates. Abundant diversity was observed among the Fol isolates in harboring the virulence-related genes (endo-polygalacturonase gene pg1 and tomatinases) and toxin production (fumonisin). However, presence of pg1 does not correlate with virulence and the isolates carrying tomatinase 4 (tom-4) in combination with other tomatinase genes were of virulent group. Detection of fumonisin gene in six isolates of Fusarium infecting tomato indicated their toxigenic nature.

Abstract:The interaction between tomato and Fusarium oxysporum f. sp. lycopersici has become a model system for the study of the molecular basis of disease resistance and susceptibility. Gene-for-gene interactions in this system have provided the basis for the development of tomato cultivars resistant to Fusarium wilt disease. Over the last 6 years, new insights into the molecular basis of these gene-for-gene interactions have been obtained. Highlights are the identification of three avirulence genes in F. oxysporum f. sp. lycopersici and the development of a molecular switch model for I-2, a nucleotide-binding and leucine-rich repeat-type resistance protein which mediates the recognition of the Avr2 protein. We summarize these findings here and present possible scenarios for the ongoing molecular arms race between tomato and F. oxysporum f. sp. lycopersici in both nature and agriculture.
