Abstract:Micro and nano technologies are of increasing importance in microfluidics devices used for electroporation (electroporation – the permeabilization of the cell membrane with brief high electric field pulses). Electrochemical reactions of electrolysis occur whenever an electric current flows between an electrode and an ionic solution. It can have substantial detrimental effects, both on the cells and solutions during the electroporation. As electrolysis is a surface phenomenon, between electrodes and solution, the extent of electrolysis is increased in micro and nano electroporation over macro-electroporation, because the surface area of the electrodes in micro and nano electroporation is much larger. A possible way to eliminate the electrolytic effect is to develop non-electrolytic microelectroporation by coating the microelectroporation devices with a dielectric insulating layer. In this study, we examine the effect of a dielectric insulating layer on the performance of a singularity microelectroporation device that we have recently designed. Using numerical analysis, we study the effects of various design parameters including, input sinusoidal voltage amplitude and frequency, geometrical configuration and material electrical properties on the electroporation performance of the non-electrolytic microelectroporation device. In the simulation, we used properties of four real dielectric materials and four solutions of interest for microelectroporation. We characterized the effect of various design parameters of relevance to singularity based microelectroporation, on non-electrolytic microelectroporation. Interestingly, we found that the system behaves in some aspects as a filter and in many circumstances saturation of performance is reached. After saturation is reached, changes in parameters will not affect the performance of the device.
Droplet-enabled controllable manipulation of tribo-charges from liquid-solid interface

Abstract:Efficient utilization of electrostatic charges is paramount for numerous applications, from printing to kinetic energy harvesting. However, existing technologies predominantly focus on the static qualities of these charges, neglecting their dynamic capabilities as carriers for energy conversion. Herein, we report a paradigm-shifting strategy that orchestrates the swift transit of surface charges, generated through contact electrification, via a freely moving droplet. This technique ingeniously creates a besptengngoke charged surface which, in tandem with a droplet acting as a transfer medium to the ground, facilitates targeted charge displacement and amplifies electrical energy collection. The spontaneously generated electric field between the charged surface and needle tip, along with the enhanced water ionization under the electric field, proves pivotal in facilitating controlled charge transfer. By coupling the effects of charge self-transfer, contact electrification, and electrostatic induction, a dual-electrode droplet-driven triboelectric nanogenerator (DD–TENG) is designed to harvest the water-related energy, exhibiting a two-order-of-magnitude improvement in electrical output compared to traditional single-electrode systems. Our strategy establishes a fundamental groundwork for efficient water drop energy acquisition, offering deep insights and substantial utility for future interdisciplinary research and applications in energy science.
Electrostatic layer-by-layer nano-assembly: films, cantilevers, micropatterns, and nanocapsules

Abstract:Layer-by-layer (LbL) nanoassembly in combination with traditional lithography and microfluidics was applied for the fabrication of ultrathin microcantilevers and for the modification of microchannel surface. Hundreds of cantilevers were fabricated on a silicon wafer simultaneously. The purpose is to develop chemical/biosensor arrays for parallel, massive data gathering. Microcantilever optical deflections were measured using a four-quadrant AFM head with integrated laser and position sensitive detector. In the second part, laminar flow fabrication of interpolyelectrolyte complexes was studied inside a microchannel. Polyelectrolyte micropatterns were studied using fluorescent confocal microscopy. Filament like, 15 mu m, interpolyelectrolyte microstripes were formed at flow rate higher than 0.01 mL(.)min(-1) and concentrations of the initial polyelectrolytes below 1 mg(.)mL(-1). New, soft micropatterning technique for the anisotropic modification of polyelectrolyte nanocapsules was also demonstrated. The microchannel surface was made sensitive to pH by coating the surface of the channel with a pH sensitive dye using LbL assembly to control the reaction.

Abstract:The dispensing of tiny droplets is a basic and crucial process in a myriad of applications, such as DNA/protein microarray, cell cultures, chemical synthesis of microparticles and digital microfluidics. This work systematically demonstrates droplet dispensing into immiscible fluids through electric charge concentration (ECC) method. It exhibits three main modes (i.e., attaching, uniform and bursting modes) as a function of flow rates, applied voltages and gap distances between the nozzle and the oil surface. Through a conventional nozzle with diameter of a few millimeters, charged droplets with volumes ranging from a few μL to a few tens of nL can be uniformly dispensed into the oil chamber without reduction in nozzle size. Based on the features of the proposed method (e.g., formation of droplets with controllable polarity and amount of electric charge in water and oil system), a simple and straightforward method is developed for microparticle synthesis, including preparation of colloidosomes and fabrication of Janus microparticles with anisotropic internal structures. Finally, a combined system consisting of ECC-induced droplet dispensing and electrophoresis of charged droplet (ECD)-driven manipulation systems is constructed. This integrated platform will provide increased utility and flexibility in microfluidic applications because a charged droplet can be delivered toward the intended position by programmable electric control.

Accelerating Electrochemical Reactions in a Voltage‐Controlled Interfacial Microreactor

Abstract:AbstractMicrodroplet chemistry is attracting increasing attention for accelerated reactions at the solution–air interface. We report herein a voltage‐controlled interfacial microreactor that enables acceleration of electrochemical reactions which are not observed in bulk or conventional electrochemical cells. The microreactor is formed at the interface of the Taylor cone in an electrospray emitter with a large orifice, thus allowing continuous contact of the electrode and the reactants at/near the interface. As a proof‐of‐concept, electrooxidative C−H/N−H coupling and electrooxidation of benzyl alcohol were shown to be accelerated by more than an order of magnitude as compared to the corresponding bulk reactions. The new electrochemical microreactor has unique features that allow i) voltage‐controlled acceleration of electrochemical reactions by voltage‐dependent formation of the interfacial microreactor; ii) “reversible” electrochemical derivatization; and iii) in situ mechanistic study and capture of key radical intermediates when coupled with mass spectrometry.

Abstract:The combination of atomic force microscopy (AFM) with nanofluidics, also referred to as FluidFM, has facilitated new applications in scanning ion conductance microscopy, direct force measurements, lithography, or controlled nanoparticle deposition. An essential element of this new type of AFMs is its cantilever, which bears an internal micro-channel with a defined aperture at the end. Here, we present a new approach for in-situ characterization of the internal micro-channels, which is non-destructive and based on electrochemical methods. It allows for probing the internal environment of a micro-channeled cantilever and the corresponding aperture, respectively. Acquiring the streaming current in the microchannel allows to determine not only the state of the aperture over a wide range of ionic strengths but also the surface chemistry of the cantilever's internal channel. The high practical applicability of this method is demonstrated by detecting the aspiration of polymeric, inorganic and hydrogel particles with diameters ranging from several mu m down to 300 nm. By verifying in-situ the state of the aperture, i.e. open versus closed, electrophysiological or nano-deposition experiments will be significantly facilitated. Moreover, our approach is of high significance for direct force measurements by the FluidFM-technique and sub-micron colloidal probes.

Abstract:Electro-wetting-on-dielectric actuation enables droplets, the basic units of digital microfluidics, to be manipulated on a two-dimensional surface, providing a versatile platform for chemical assays or multi-step operations at micro- or nano-scale. In this paper, we carry out characterization experiments to demonstrate an electrode design to improve the reproducibility of on-chip droplet generation with no extra external apparatus. The overall reproducibility for a sequence of droplets dispensed consecutively from a non-refilling reservoir can be limited within ±0.5%. Results from the repetition of 1000 iterations offer the long-term reproducibility in the range of ±1%, indicating its validity in practical applications.

Abstract:The assembly of colloidal particles into micro-patterns is essential in optics, informatics, and microelectronics. However, it is still a challenge to achieve quick, reversible, and precise assembly patterns within micro-scale spaces like droplets. Hereby, a method is presented that utilizes in-plane dielectrophoresis to precisely manipulate particle assemblies within microscale droplets. The electro-microfluidic particle assembly platform, equipped with ingenious electrode designs, enables the formation of diverse micro-patterns within a droplet array. The tunability, similarity, stability, and reversibility of this platform are demonstrated. The ability to assemble letters, numbers, and Morse code patterns within the droplet array underscores its potential for information encoding. Furthermore, using an example with four addressing electrodes beneath a droplet, 16 distinct pieces of information through electrical stimuli is successfully encoded. This unique capability facilitates the construction of a dynamic electronic token, indicating promising applications in anti-counterfeiting technologies.

Abstract:Droplet manipulation on a substrate by electrical signals is instrumental to the automation and miniaturization of labor-intensive assays in life science and chemistry. Current techniques are primarily based on either electrowetting or a more recent ionic-surfactant-mediated electro-dewetting effect. Here, we report that the two effects can occur simultaneously on the same substrate. Using a dope silicon substrate and an aqueous droplet with a cationic surfactant, the surface exhibits dewetting at positive biases and wetting at negative. Such a polarity-dependent wetting–dewetting transition enables a more significant wettability change (>60° contact angle change between ±3 V), which preserves after multiple wetting–dewetting cycles. We also find that the transition does not experience contact angle hysteresis that sole electrowetting commonly suffers from. Benefitting from these features, we experimentally show that droplet manipulation on a digital microfluidic device is more efficient and robust using this joint mechanism.

Abstract:We develop an electrokinetic technique that continuously manipulates colloidal particles to concentrate into patterned particulate groups in an energy efficient way, by exclusive harnessing of the intrinsic Joule heating effects. Our technique exploits the alternating current electrothermal flow phenomenon which is generated due to the interaction between non-uniform electric and thermal fields. Highly non-uniform electric field generates sharp temperature gradients by generating spatially-varying Joule heat that varies along radial direction from a concentrated point hotspot. Sharp temperature gradients induce local variation in electric properties which, in turn, generate strong electrothermal vortex. The imposed fluid flow brings the colloidal particles at the centre of the hotspot and enables particle aggregation. Further, manoeuvering structures of the Joule heating spots, different patterns of particle clustering may be formed in a low power budget, thus, opening up a new realm of on-chip particle manipulation process without necessitating highly focused laser beam which is much complicated and demands higher power budget. This technique can find its use in Lab-on-a-chip devices to manipulate particle groups, including biological cells.

Abstract:Ionic and molecular selectivity is considered unique for the nanoscale and not realizable in microfluidics. This is due to the scale-matching problem -- a difficulty to match the dimensions of ions and electrostatic potential screening lengths with the micron-sized confinements. Here, we demonstrate a microscale realization of the ionic transport processes closely resembling those specific to ionic channels or in nanofluidic junctions, including selectivity, guidance through complex geometries and flow focusing. As a model system, we explore electrokinetic spherical Janus micromotors moving over charged surfaces with a complex spatial charge distribution and without any topographical wall. We discuss peculiarities of the long-range electrostatic interaction on the behavior of the system including interface crossing and reflection of positively charged particles from negatively charged interfaces. These results are crucial for understanding the electrokinetic transport of biochemical species under confinement, have the potential to increase the precision of lab-on-chip-based assays, as well as broadening use cases and control strategies of nano-/micromachinery.

Abstract:Insulator-based electrokinetically driven microfluidic devices stimulated with direct current (DC) voltages are an attractive solution for particle separation, concentration, or isolation. However, to design successful particle manipulation protocols, it is mandatory to know the mobilities of electroosmosis, and linear and nonlinear electrophoresis of the microchannel/liquid/particle system. Several techniques exist to characterize the mobilities of electroosmosis and linear electrophoresis. However, only one method to characterize the mobility of nonlinear electrophoresis has been thoroughly assessed, which generally requires DC voltages larger than 1000 V and measuring particle velocity in a straight microchannel. Under such conditions, Joule heating, electrolysis, and the DC power source cost become a concern. Also, measuring particle velocity at high voltages is noisy, limiting characterization quality. Here we present a protocol—tested on 2 μm polystyrene particles—for characterizing the mobility of nonlinear electrophoresis of the liquid/particle system using a DC voltage of only 30 V and visual inspection of particle dynamics in a microchannel featuring insulating obstacles. Multiphysics numerical modelling was used to guide microchannel design and to correlate particle location during an experiment with electric field intensity. The method was validated against the conventional characterization protocol, exhibiting excellent agreement while significantly reducing measurement noise and experimental complexity.

Abstract:By forming lipid bilayers within SU8 patterns, between interdigitated electrodes, we have demonstrated that it is possible to manipulate charged membrane components using low applied voltages over relatively short time scales. Two distinct patterns were studied: a nested "fish trap" which served as a molecular trap, and a diffusion aided Brownian ratchet which operated as a molecular pump. By reducing the size of the structures we have demonstrated that large applied fields (>200 V/cm) can be achieved on-chip, using low applied potentials (<13 V). By using ac fields applied orthogonal to the direction of desired motion, the molecular pumps provide a voltage independent method for moving charged components within lipid membranes over large distances. The reduced scale of the trap structures compared to those previously used in our laboratory has led to over a 10-fold decrease in the operational time require for charge build-up, from 16 h down to 1.5 h. The observed benefits of scaling means that these systems should be suitable for the on-chip separation and manipulation of charged species within supported lipid membranes.

Abstract:The authors have analyzed the medico-legal records of 70 workers from an asbestos factory, who developed pneumoconiosis, sometimes after prolonged exposure. X-ray films were interpreted (though not without some difficulty) according to the I.L.O. classification criteria. Respiratory function tests included spirometry, gas exchange determination and blood gas measurements. There was a correlation between the duration of occupational exposure and the onset of chest lesions visible on X-ray films (pleural thickening, pulmonary fibrosis and later, pleural calcifications) and a closer correlation between these and the results of the two main respiratory function tests: vital capacity and fractional global and arterio-alveolar CO uptake coefficient (DuaCO). It appeared, however, that lung function impairment sometimes preceded radiological abnormalities, and this should be taken into consideration when assessing the financial compensation due to workers exposed to asbestos.

Abstract:Transport of fluids and particles at the microscale is an important theme both in fundamental and applied science. One of the most successful approaches is to use an electric field, which requires the system to carry or induce electric charges. We describe a versatile approach to generate electrokinetic flows by using a liquid crystal (LC) with surface-patterned molecular orientation as an electrolyte. The surface patterning is produced by photo-alignment. In the presence of an electric field, the spatially varying orientation induces space charges that trigger flows of the LC. The active patterned LC electrolyte converts the electric energy into the LC flows and transport of embedded particles of any type (fluid, solid, gaseous) along a predesigned trajectory, posing no limitation on the electric nature (charge, polarizability) of these particles and interfaces. The patterned LC electrolyte exhibits a quadratic field dependence of the flow velocities; it induces persistent vortices of controllable rotation speed and direction that are quintessential for micro- and nanoscale mixing applications.

A soft-lithographed chaotic electrokinetic micromixer for efficient chemical reactions in lab-on-chips

Abstract:Mixing is one of the basic functions which automated lab-on-chips require for the effective management of liquid samples. In this paper we report on the working principle, design, fabrication and experimental characterization of a soft-lithographed micromixer for microfluidic applications. The device effectively mixes two liquids by means of chaotic advection obtained as an implementation of a Linked Twisted Map (LTM). In this sense it is chaotic. The liquids are electrokinetically displaced by generating rolls through AC electroosmosis on co-planar electrodes. The device performance has been tested on dyed DI-water for several voltages, frequencies and flow-rates, displaying good mixing properties in the range of $10 ÷100$kHz, at low peak-to-peak voltages ($\sim15 ÷20$ volts). Low voltage supply, small dimensions and possibility of fabrication via standard lithographic techniques make the device highly integrable in lab-on-a-chip platforms.

Abstract:<p>The electrostatic spray method is a promising nonvacuum technique for efficient deposition of thin films from solutions or dispersions. The multitude of electrostatic spray process parameters, including surface tension, viscosity, and conductivity of the liquid, applied voltage, nozzle size, and flow rate, make electrostatic spray deposition very versatile for the morphological engineering of nanostructured films. The current state‐of‐the‐art in electrostatic spraying can produce exceptional morphologies. Such tailoring of morphologies is notably useful in electrochemical applications where high electrolyte‐accessible surface area often improves performance. Interesting morphologies of metal oxides and their composites are highlighted, including nanopillars, nanoferns, and porous microspheres produced by electrostatic spraying to enhance energy conversion and storage performance. The physics associated with the electrostatic spray process and morphology control using it are also presented. The manuscript highlights the potential of electrospray processing for producing thin films of controlled microstructure, from ultrasmooth layers in organic photovoltaics and perovskite photovoltaics to hierarchical nanostructured films for anodes and photoanodes. It aims to help researchers appreciate essential aspects of electrostatic spray deposition efficiency, process control, and morphology engineering for energy conversion (e.g., solar cell, fuel cell, and photoelectrochemical cell) and energy storage (e.g., lithium‐ion battery and supercapacitor) electrodes.</p>

Abstract:Electrochemistry takes a key position in products and manufacturing processes of microtechnology, which has established a multi-billion dollar market with applications in information, entertainment, medical, automotive, telecom and many other technologies. In combination with microlithographic pattern generation electrochemical deposition processes are applied in manufacturing CDs and DVDs, nozzle plates for ink jet printers, read–write heads for hard magnetic disks and solder bumps for flip-chip technology. Three-dimensional microstructures with extremely high precision and aspect ratio are manufactured by means of LIGA technology, which combines deep lithography, electroforming and moulding process steps. Field assisted etching processes with ultra-short voltage pulses, anodization of silicon and field assisted ion exchange in glass have been successfully used for generating microstructures. Meanwhile, there are a huge number of microdevices, which utilize electrochemical phenomena. ISFETs and ion sensitive electrodes are standard components in the field of microsensors, microfuel cells will become high-performance power supply systems for mobile IT and entertainment products. In communication technology electrophoresis-based on electronic ink displays will become more and more important products. The most challenging development is dealing with so-called labs-on-a-chip which utilize electrochemical phenomena like electrophoresis, isoelectric focussing and electroosmosis to create a novel platform for ultra-miniaturized analytics in life sciences, medical diagnosis, environmental technologies and many other areas of modern development.

Detecting haplotype effects in genomewide association studies

Abstract:The analysis of genomewide association studies requires methods that are both computationally feasible and statistically powerful. Given the large‐scale collection of single nucleotide polymorphisms (SNPs), it is desirable to explore the information contained in their interrelationships. In particular, utilizing haplotypes rather than individual SNPs and accounting for correlations of polymorphisms in adjustment for multiple testing can lead to increased power. We present a statistically powerful and numerically efficient method based on sliding windows of adjacent SNPs to detect haplotype‐disease association in genomewide studies. This method consists of an efficient algorithm to calculate a proper likelihood‐ratio statistic for any given window of SNPs, along with an accurate and efficient Monte Carlo procedure to adjust for multiple testing. Simulation studies using the HapMap data showed that the proposed method performs well in realistic situations. Abstract:Abstract Electric fields have been highlighted as a smart reagent in nature’s enzymatic machinery, as they can directly trigger or accelerate chemical processes with stereo- and regio-specificity. In enzymatic catalysis, controlled mass transport of chemical species is also key in facilitating the availability of reactants in the active reaction site. However, recent progress in developing a clean catalysis that profits from oriented electric fields is limited to theoretical and experimental studies at the single molecule level, where both the control over mass transport and scalability cannot be tested. Here, we quantify the electrostatic catalysis of a prototypical Huisgen cycloaddition in a large-area electrode surface and directly compare its performance to the conventional Cu(I) catalysis. Our custom-built microfluidic cell enhances reagent transport towards the electrified reactive interface. This continuous-flow microfluidic electrostatic reactor is an example of an electric-field driven platform where clean large-scale electrostatic catalytic processes can be efficiently implemented and regulated.
