Abstract:HPLC method is the standard method for the separation and quantification of impurities from quinolone antibiotics. However, due to the large differences in the UV absorption of the impurities in quinolone antibiotics, quantitative analysis without the availability of corresponding reference substances currently poses a challenge. A sensitive and direct method using high performance liquid chromatography coupled with diode array detector and charged aerosol detector (HPLC-DAD-CAD) was developed for the analysis of impurities in quinolone antibiotics. The chromatographic conditions were optimized for good separation and output signal of CAD detector by response surface method (RSM). The systematic variation of CAD parameter settings, such as nebulization temperature, filter constant and power function value (PFV), were used to study the effect of on the detector response of signal-to-noise ratios (S/N) and linearity for ofloxacin, ciprofloxacin and their impurities. In the method validation, good linearity of each component was obtained with coefficient of determination (r) greater than 0.999 in the range of 0.5-300 mu g mL-1. The average recoveries of each component were 99.02-102.39 % by DAD, were 98.22-101.91 % by CAD, RSDs were less than 2.5 % for intra-day and inter-day precision by DADCAD, with good precision and accuracy. The correction factor experimental results showed that the developed method provided a uniform response to the impurities with differences chromophores and could unbiasedly and directly detect the impurities in quinolone antibiotics. The method is first reported application of HPLC-DADCAD method for the analysis of impurities in quinolone antibiotics and it can be used for quality control of quinolone antibiotics.

Abstract:This study presents a simple and green dispersive micro-solid phase extraction method for preconcentration of acidic quinolones from honey prior to high performance liquid chromatography determination. A two-dimensional nanostructured zinc-aluminum layered double hydroxide was synthesized and used as the sorbent for dispersive micro-solid phase extraction. Its different characteristics from conventional sorbents is that it is dissolvable in acidic solution (pH < 4). After the extraction, the analyte elution step was omitted and thus the use of organic solvents was avoided. The key parameters influencing the extraction efficiency such as the amount of sorbent, pH of sample solution, vortex time, type and volume of acidic solution were investigated and optimized. The method exhibited low limits of detection (3.0-5.0 ng/g), good linearity (10-2000 ng/g) with coefficients of determinations higher than 0.9991, acceptable precision (RSD<9.1%) and accuracy (RE<5.8%). The proposed method is fast, efficient, eco-friendly, and suitable for the determination of acidic quinolones in honey samples.

Abstract:The extraction of quinolone antibiotics (QAs) is crucial for the environment and human health. In this work, polyacrylonitrile (PAN)/covalent organic framework TpPa–1 nanofiber was prepared by an electrospinning technique and used as an adsorbent for dispersive solid-phase extraction (dSPE) of five QAs in the honey and pork. The morphology and structure of the adsorbent were characterized, and the extraction and desorption conditions for the targeted analytes were optimized. Under the optimal conditions, a sensitive method was developed by using PAN/TpPa–1 nanofiber as an adsorbent coupled with high-performance liquid chromatography (HPLC) for five QAs detection. It offered good linearity in the ranges of 0.5–200 ng·mL−1 for pefloxacin, enrofloxacin, and orbifloxacin, and of 1–200 ng·mL−1 for norfloxacin and sarafloxacin with correlation coefficients above 0.9946. The limits of detection (S/N = 3) of five QAs ranged from 0.03 to 0.133 ng·mL−1. The intra-day and inter-day relative standard deviations of the five QAs with the spiked concentration of 50 ng·mL−1 were 2.8–4.0 and 3.0–8.8, respectively. The recoveries of five QAs in the honey and pork samples were 81.6–119.7%, which proved that the proposed method has great potential for the efficient extraction and determination of QAs in complex samples.

Core-shell magnetic porous organic polymer for magnetic solid-phase extraction of fluoroquinolone antibiotics in honey samples followed by high-performance liquid chromatography with fluorescence detection

Abstract:By monomer-mediated in-situ growth synthesis strategy, with hydroquinone and 1,3,5-tris(4-aminophenyl)benzene as monomers, a core-shell magnetic porous organic polymer was synthesized through a simple azo reaction. Based on this, a magnetic solid-phase extraction-high-performance liquid chromatography-fluorescence detection method was proposed for the analysis of fluoroquinolones in a honey sample. With ofloxacin, ciprofloxacin, enrofloxacin, lomefloxacin, and difloxacin as target analytes, factors affecting the extraction efficiency had been optimized. The LODs were 1.5-5.4 ng/L (corresponding to 0.23-0.81 μg/kg in honey). The linear range was 0.005-20 μg/L for difloxacin, 0.01-20 μg/L for ofloxacin, ciprofloxacin and lomefloxacin, and 0.02-20 μg/L for enrofloxacin. The enrichment factor was 84.4-91.7-fold with a high extraction efficiency of 84.4-91.7%. The method was assessed by the analysis of target fluoroquinolones in honey samples, and the recoveries for the spiked samples were 79.3-95.8%. The results indicated that the established magnetic solid-phase extraction-high-performance liquid chromatography-fluorescence detection method is efficient for the analysis of trace fluoroquinolones in honey.

Abstract:A novel Fe3O4@MDN magnetic nanomaterial was successfully developed as a magnetic dispersive solid-phase extraction adsorbent for the determination of triazole pesticide residuals in honey samples by high performance liquid chromatography coupled with triple quadrupole mass spectrometry (HPLC-MS/MS). The macromolecular structure named MDN was coated on the surface of a copolymer synthesized with silyl reagents methacrylic acid-3-(trimethoxysilyl) propyl ester (MPS) and tetraethyl orthosilicate (TEOS) based on ferriferrous oxide (Fe3O4), whose characteristics were confirmed by a scanning electron microscope (SEM). The proposed magnetic dispersive solid-phase extraction (MDSPE) method provided a rapid, environmentally friendly and magnetic stuff recyclable approach for the triazole pesticide analysis. By means of optimal conditions, the recoveries of 15 triazoles (spiked at 20 mu g kg(-1)) ranged from 90.5 to 105.9%. The method showed good linearity, sensitivity in the range of mu g per kilogram and repeatability (relative standard deviation below 6.8%, n = 5). The proposed method was confirmed to be a simple, rapid and efficient method for the analysis of triazole pesticide residuals in honey samples.

Abstract:In this study, a new metal-organic framework-monolith composite for in-tube solid phase microextraction phase (IT-SPME) of fluoroquinolones (FQs) was prepared. 4-Vinylbenzoic acid was copolymerized with ethylenedimethacrylate in a fused silica capillary to form porous monolith. After that, zeolitic imidazolate frameworks (ZIF-8) were synthesized in situ within the pores and the surface of the monolith by controlled layer-by-layer self-assembly of Zn2+ and imidazole. The introduction of ZIF-8 enhanced the surface area of monolith composite, and thus, improving the extraction performance of IT-SPME for FQs obviously. Under the optimized conditions, a highly sensitive method for the monitoring of FQs residue in water and honey samples was developed by the on-line combination of IT-SPME with high-performance liquid chromatography with fluorescence detection (HPLC-FLD). The limits of detection (S/N = 3) for the targeted FQs in water and honey samples were as low as 0.14-0.61 ng/L and 0.39-1.1 ng/L, respectively. The relative standard deviations (RSDs) for intra-day and inter-day assay variability were less than 10% in all samples. The established on-line IT-SPME-HPLC-FLD was successfully used to detect ultra-trace FQs in environmental water and honey samples. Recoveries at different spiked concentrations ranged from 80.1% to 119% and 80.2-117% for water and honey samples, respectively, with satisfactory reproducibility. Compared to up-to-date reported methods, the proposed approach exhibits some features such as high sensitivity, convenience, partial automation, low consumptions of sample and solvent.

Abstract:In this study, the hydroxypropyl β‐cyclodextrin hybrid monolithic material was fabricated and firstly applied as adsorbent for dispersive solid‐phase extraction coupled with high‐performance liquid chromatography to detect trace‐level seven fluoroquinolones in water samples. The prepared hydroxypropyl β‐cyclodextrin hybrid monolithic material was characterized by fourier transform infrared spectroscopy, scanning electron microscopy and adsorption experiments, which showed excellent specific adsorption to the target fluoroquinolones. Under the optimized conditions, the extraction methodology showed satisfactory precision with relative standard deviations between 2.6 and 5.6%, good linearity (R2≥0.9990) and satisfactory recoveries (82.5∼91.8%). The limits of detection and limits of quantification of the method were in the range of 0.4∼1.2 ng mL–1 and 1.4∼4.0 ng mL–1 respectively, which confirmed the possibility of quantifying trace levels. Furthermore, the material could be reused at least five times. These results demonstrated that the hydroxypropyl β‐cyclodextrin hybrid monolithic material was a promising adsorbent for fluoroquinolones, and the established method combined dispersive solid‐phase extraction with high‐performance liquid chromatography was suitable for the determination of fluoroquinolones in aqueous samples.This article is protected by copyright. Abstract:A novel type of three-dimensional network structure, covalent organic frameworks (COFs) aerogel, was fabricated and applied to dispersive solid-phase extraction (dSPE) of quinolone antibiotics (QAs). Density functional theory (DFT) was applied to investigate the possible interaction mechanism and results confirmed that the strong adsorption affinity is attributed to the intralayer hydrogen bonds and π-π interaction. Furthermore, a sensitive analytical method based on COFs-aerogel for determining quinolone antibiotics residues in water and honey samples was developed and HPLC-MS/MS was used for sample detection and quantification. Under the optimal conditions, COFs-aerogel exhibited a wide linearity (0.1-500 ng∙L-1), low limits of detection (0.02-0.06 ng∙L-1), and good precision (RSD ˂ 10%) for selected QAs. A preliminary practical application of the developed method was proved by the efficient detection of quinolone antibiotics in water and food samples with good recoveries (68.2-104% and 64.0-100% for water and honey samples, respectively). Combining the experimental data with theoretical calculation, results illustrated that COFs-aerogel holds a great potential to capture contaminants and address environmental and food safety issues.

Abstract:The sample was diluted with pH 4.0 of Na2EDTA-Mcllvaine buffer solution,and pesticides and antibiotics in the sample were extracted with a mixture of acetic acid-acetonitrile (1 + 19),and the extract was stratified by salting-out agents.The supernatant was cleaned up with purification pipe filled with magnetic C18 and PSA.Luna C18 column was used as stationary phase,and a mixture of 0.1％ (φ) formic acid solution and acetonitrile in various ratios as mobile phase in gradient elution.ESI+ and MRM were adopted in MS.Linear relationships between values of peak area and mass concentration of 30 compounds were found in definite ranges,with detection limits (3S/N) in the range of 0.3-4.1 μg · kg 1.Test for recovery was made by standard addition method,giving results in the range of 71.1％-116％,with RSDs (n=5) ranged from 0.80％ to 21％.

Abstract:In this study, Zeolitic Imidazole Framework-8/cellulose aerogel (ZIF-8/CA) hybrid was successfully fabricated through a simple doping method and ZIF-8 acted as the major component for adsorption. In order to elucidate the adsorption mechanism deeply, molecular simulation was adopted to the expound the interaction modes between ZIF-8 and the fluoroquinolones (FQs). ZIF-8/CA was used as the adsorbent for semi-automated pipette tip solid phase extraction (PT-SPE). In combination with high performance liquid chromatography tandem fluorescence detector (HPLC-FLD), the established method was successfully employed to determine trace amount of FQs in water samples. Extraction parameters such as the content of ZIF-8, pH of sample solution, volume of sample, flow rate of sampling, type and volume of elution solvent were investigated. Under the optimized conditions, satisfactory linearity was achieved with the correlation coefficient (R2) ranging from 0.9954 to 0.9992. The limits of detection were in the range of 0.337–1.707 ng L-1. And the recoveries varied from 75.9% to 96.8% with RSD less than 8.0%. The established method was demonstrated to be sensitive, efficient and convenient.

Abstract:A high-throughput method using a new ZIF-8@GO thin-film microextraction coating was established for determining macrolides and lincosamides in honey. The coating preparation parameters (ZIF-8@GO synthesis conditions, coating material proportions, dipping time) and analysis parameters (sample diluent solvent, adsorption and desorption conditions using the ZIF-8@GO coating) were optimized. The optimized parameters were: diluent solvent sodium carbonate/sodium bicarbonate buffer solution (pH 9), adsorption time 45 min, desorption time 5 min, desorption solvent 45:40:15 v/v/v methanol/acetonitrile/water. The extracted targets were determined by ultra-high performance liquid chromatography tandem mass spectrometry. The recoveries of 10 analytes were 67.5-107.2% and the detection and quantification limits were 0.1-0.4 and 0.4-1.4 mu g/kg, respectively. The method could analyze 96 samples per run. The minimal manual time and effort is required since the bulk of the sample processing is fully automated. It was a useful and efficient method for monitoring drug residues and was successfully used to analyze real samples.

Abstract:A simple and efficient dispersive solid-phase extraction (D-SPE) method combined with gas chromatography tandem mass spectrometry (GC-MS/MS) was developed to determine organochlorine pesticides (OCP) in honey. Firstly, a type of hybrid nanocomposite (CD-MOF/TiO2) was prepared by grafting a metal-organic framework material synthesized with cyclodextrin as an organic ligand onto titanium dioxide. Then, the CD-MOF/TiO2 was used as a D-SPE adsorbent to extract the OCP, and the effects of the amount of adsorbent, ultrasonic time, vortex time, pH, and salinity on the extraction were investigated using Plackett-Burman design and Box-Behnken Design. Under the optimized adsorption and desorption conditions, an analysis method that combined D-SPE with GC-MS/MS was established. The linear ranges of 14 OCP are 1-500 μg kg-1 and the correlation coefficients are between 0.9991 and 1.000. The limits of detection and quantification vary from 0.01 to 0.04 μg kg-1 and 0.04 to 0.12 μg kg-1, respectively. The intra-day and inter-day precision of this method are suitable (RSDs% less than 11.3%). The established CD-MOF/TiO2 / D-SPE method was used for the extraction of OCP in honey samples with recovery in the range of 76.4 to 114.3%. The results demonstrate that the CD-MOF/TiO2 has a good selective enrichment ability for OCP and is suitable for the D-SPE pretreat of honey sample analysis.

Abstract:Metal-organic frameworks (MOFs), a sort of dispersive solid-phase extraction (d-SPE) material, has shown considerable prospects in the pretreatment of food, biological and other complex samples. Herein, we developed a method for compounding MOFs for d-SPE and trace determination of tetracyclines (TCs) in honey. When the compounding ratio of MIL-101 (Cr), MIL-100 (Fe) and MIL-53 (Al) was 7:1:2, adsorption-extraction was effective. Followed by high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), the limits of detection were 0.073-0.435 ng/g and the limits of quantitation ranged from 0.239 to 1.449 ng/g for oxytetracycline, tetracycline, chlortetracycline and doxycycline. The method was applied to four kinds of honey samples with recoveries from 88.1% to 126.2%. The compounding of MOFs provides a strategy for purification and multi-target extraction from complex food matrices by d-SPE.
Highly-sensitive detection of eight typical fluoroquinolone antibiotics by capillary electrophoresis-mass spectroscopy coupled with immunoaffinity extraction

Abstract:Fluoroquinolones (FQs) are a class of synthetic antibiotics with a broad active spectrum towards mycoplasma and bacteria (both Gram positive and negative). The contamination of environmental water samples with FQs has been a concern due to the extensive use of them in the veterinary and pharmaceutical industries. Reliable and simple analytical methods are needed for the monitoring of these FQs. An off-line procedure of immunoaffinity extraction (IAE) followed by capillary electrophoresis-mass spectroscopy (CE-MS) has been developed for simultaneous determination of eight typical FQs in environment water. A broad-specific polyclonal antibody was home-prepared and immobilized on a Sepharose 4B stationary phase for sample affinity extraction and the enrichment factor of our columns was about 100. CE-MS was subsequently applied to FQs separation and quantification. The immunoaffinity column was evaluated in the parameters of selectivity, binding capacity, elution protocol, and recovery. Under the optimized conditions, the developed method was successfully applied to analyze FQs spiked with 2.5 nM to 30 nM of environmental water from a local lake, providing satisfactory recoveries from 72% to 112%, and LODs and LOQs ranging from 1.2 nM to 5.0 nM and 4.0 nM to 17 nM, respectively.
Multiresidue analysis of 15 antibiotics in honey using modified QuEChERS and high performance liquid chromatography-tandem mass spectrometry

Abstract:Sugars, organic acids, and lipids generally cause strong matrix effects, posing a challenge in the analysis of trace substances in honey. In this study, method involving a modified QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe) protocol coupled with high–performance liquid chromatography–tandem mass spectrometry (HPLC–MS/MS) was developed for the analysis of multiresidue antibiotic in honey samples. Several adsorbents were selected and their abilities to reduce matrix interference and improve detection sensitivity were tested. Zinc oxide (ZnO) proved to be the most effective clean-up adsorbent for the purification of honey samples. Good linearity was found in the concentration range from limits of quantification (LOQs) to 20.0 ng mL–1 for all antibiotics, and for concentration up to 40.0 ng mL–1 for some types of antibiotics, with correlation coefficients exceeding 0.9991. The limits of detection (LODs) ranged from 0.24 to 0.68 μg kg–1, and the LOQs were in the range of 0.80–2.27 μg kg–1. The average recoveries ranged from 81.3 % to 99.0 % at three fortified levels with RSDs below 9.18 %. Twenty-eight honey samples obtained from local markets were analyzed using our optimized method, and lincomycin was detected in one sample.

Abstract:A novel analytical method based on stir‐bar sorptive extraction proposed for the determination of three trace quinolones in fish and shrimp samples. UiO‐66‐(OH)2, a hydroxyl‐functionalized zirconium metal–organic framework, has been coated on frosted glass rods by an in situ growth method. The product, UiO‐66‐(OH)2modified frosted glass rods, has been characterized and key parameters have been optimized in combination with ultrahigh‐performance liquid chromatography. The detection limits of enoxacin, norfloxacin, and ciprofloxacin were 0.48–0.8 ng mL−1, and the detection concentrations were in the range 10–300 ng mL−1, showing a good linear relationship. This method was used for the determination of three quinolones in aquatic organisms, and the recoveries in spiked fish and shrimp muscle tissue samples were 74.8–105.4% and 82.5–115.8%, respectively. The relative standard deviations were less than 6.9%. The established method combined stir‐bar sorptive extraction based on UiO‐66‐(OH)2 modified frosted glass rods with ultrahigh‐performance liquid chromatography, has good application prospects for the detection of quinolone residues in fish and shrimp muscle samples. Abstract:A method of solid phase extraction-liquid chromatography-tandem mass spectrometry (SPE-LC-MS/MS) for simultaneous determination of 54 drug residues (sulfonamides, nitroimidazoles, quinolones, macrolides, lincosamides and praziquantel) in honey was developed. The honey samples were diluted by phosphate buffer solution (pH 8) followed by a further cleanup procedure with an Oasis (HLB) SPE column. The detection was carried out by LC-MS/ MS, using positive-ion electrospray ionization in multiple reaction monitoring (MRM) mode, with one precursor/two product ion transitions for each compound. Isotope dilution internal standard method or external standard method was used to determine the residue contents with a good linear relationship (r > 0.992). The limits of quantification (LOQs, S/N > 10) were 1.0 microg/kg for sulfonamides and nitroimidazoles, 2.0 microg/kg for quinolones and lincosamides, 3.0 microg/kg for macrolides, and 0.3 kg/kg for praziquantel. The recovery ranges were from 32.6% to 114% with the relative standard deviations (RSDs) between 1.3% and 28.9%. As a screening method, the developed procedure is suitable for the determination and confirmation of the drug residues in honey samples.

Abstract:Based on advantages of capillary electrophoresis (CE), a new solid-phase extraction (SPE) coupled with CE has been developed for preconcentration, enrichment and determination of anthraquinones and flavonoids (rutin, emodin, quercetin, 1,8-dihydroxyanthraquinone) in honey. The environmental-friendly chitin activated after an easy processing is selected as the adsorbent to enrich analytes. Then, chitin was filled into the filter as the solid phase. To improve the extraction effect, some key parameters of extraction were optimized. Under the optional extraction conditions, the chitin showed excellent adsorption capacity and selectivity over rutin, emodin, quercetin, and 1,8-dihydroxyanthraquinone, with enrichment factors reaching 5 folds. The CE coupled with fluorescence detection was used for the detection. Results prove the method is simple, fast, and highly sensitive, with the limit of detection (LOD) is 3.00-200.0 ng/mL; the recovery is 90.0-107.0%, and relative standard deviation of (RSD) is 1.8-8.3%.
Porous electrospun microfibers for low flow-resistant solid phase extraction of fluoroquinolones in tap water, egg and milk samples

Abstract:In this work, porous electrospun microfibers (PEMFs) were prepared using a polyimide/polyvinylpyrrolidone/polyethylene glycol (PI/PVP/PEG) solution mixture with coaxial ultrasonic water vapor spraying. After removing PVP and PEG by ultrasonic water washing, the PEMFs were successfully demonstrated as adsorbents for solid phase extraction (SPE). Most non-porous electrospun nanofibers are hundreds of nanometers in diameter, with a specific surface area of dozens of square meters per gram. In contrast, the diameter of the as-prepared PEMFs was tuned between 3 and 8 μm, the specific surface area was 76 m2g-1 and the pore size was ca 25 nm. Therefore, the flow resistance of the PEMF-SPE cartridges was similar to those of conventional commercial SPE cartridges, and much lower than those of SPE cartridges packed with electrospun nanofibers. Using the PEMF-SPE cartridges with ultra-performance liquid chromatography-fluorescence detector (UPLC-FLD), five fluoroquinolones (FQs) in tap water, egg and milk samples were extracted and quantified successfully. After optimizing the extraction conditions, FQs in water samples were extracted and eluted with high recoveries of 84.8-114.8%. The inter-batch and intra-batch relative standard deviation (RSD) values for the FQs were in the range of 1.9-9.5% (n=3), and the limits of detection were between 0.0024-0.014 μg L-1. The method was linear in the concentration range of 0.005-10 μg L-1. The reliability of the developed method was validated by analyzing tap water, egg and milk samples, and the recovery values were found to be in the range of 74.8-116.6% under the optimized conditions.

Abstract:Due to the ecosystem and public health concerns, contaminants must be removed from the water, and water purification methods aim to find superior materials with superior chemistry and unique structures. We developed a new 3D flower-like double-oxide hydrotalcite composite as an extraction phase for the enrichment of quinolone antibiotics (QAs). In this work, NiAl double-oxide composite microspheres (Al2O3) with 3D flower-like structure were prepared by calcining pollen to prepare Al2O3-C carbon templates and homogeneously coating LDO nanosheets on Al2O3-C microspheres. Compared with the pristine NiAl-LDH (82.70 m2/g), the Al2O3 microspheres exhibited a higher specific surface area (139.70 m2/g), making them rich in active adsorption sites. In addition, a large number of anions that can be exchanged between the layers can effectively enhance adsorption capacity through multiple interactions. The adsorption performance of Al2O3 microspheres on QAs was investigated in detail. The prepared composites proved to be suitable for the adsorption of QAs in complex samples with a maximum adsorption capacity of 64.08 mg g−1. The developed method was successfully applied to the analysis of QAs in real samples with a satisfactory recovery.
