Abstract:Nanoscale topography was constructed on a thermally responsive poly(N-isopropylacrylamide) (PNIPAAm) surface by grafting the polymer from silicon nanowire arrays via surface-initiated atom transfer radical polymerization. The as-prepared surface showed largely reduced platelet adhesion in vitro both below and above the lower critical solution temperature (LCST) of PNIPAAm ( approximately 32 degrees C), while a smooth PNIPAAm surface exhibited antiadhesion to platelets only below the LCST. Contact angle and adhesive force measurements on oil droplets (1,2-dichloroethane) in water demonstrated that the nanoscale topography kept a relatively high ratio of water content on the as-prepared surface and played a key role in largely reducing the adhesion of platelets; however, this effect did not exist on the smooth PNIPAAm surface. The results can be used to extend the applications of PNIPAAm in the fields of biomaterials and biomedicine under human physiological temperature and provide a new strategy for fabricating other blood-compatible materials.
Chemical Modification on Top of Nanotopography to Enhance Surface Properties of PDMS

Abstract:In the present study we report the surface modification of polydimethylsiloxane (PDMS) via combining both physical and chemical modification routes. Well defined nanostructures were created on PDMS surface through anodized aluminum oxide (AAO) template method. On top of the nanostructures, polyvinyl alcohol (PVA) was coated after the nano patterned PDMS was exposed to air plasma. The influence of the combined modification on surface properties of PDMS was examined by ATR-FTIR and water contact angle measurement. The impact of plasma pretreatment time and storage time in ambient environment were investigated, and property of films with or without nanostructures was compared. Experimental results show that long-term stable hydrophilic surfaces of PDMS can be achieved by PVA coating and be further improved by the surface nanostructures. The modified PDMS was also used for cell culture. The results indicate that both nanostructuring and chemically modifying the PDMS surface improves cell growth. Additionally, combination of these surface modifications has an additive effect on cell proliferation.

Abstract:In the preparation of a thermo-responsive, poly(N-isopropylacrylamide) (PNIPAAm)-grafted polydimethylsiloxane (PDMS) surface by means of benzophenone-initiated photopolymerization, we observed that thick (>1 mm) PDMS substrates were much more difficult to be grafted with PNIPAAm than thin ones. Investigations revealed that the shortage of diffused benzophenone molecules in the surface region of the thick substrate might be the reason. By prolonging the time spent for treating the substrate with a benzophenone solution, PNIPAAm could be successfully grafted onto thick PDMS substrates. The PNIPAAm-grafted PDMS surface was highly thermo-responsive. The contact angle on a grafted surface increased from 38 to 91° in response to the temperature increase from 20 to 38 °C. An electroosmotic flow (EOF) mobility of 5×10−4 cm2/Vs was supported by a PNIPAAm-grafted PDMS channel at 50 °C, whereas negligible EOF was observed at 20 °C. Doxorubicin (DX), an anticancer drug, was adsorbed by the grafted surface at 40 °C, and the majority of the adsorbed DX was quickly released from the surface to a stripping solution at 5 °C. Osteoblast cells adhered onto the PNIPAAm-grafted PDMS surface and proliferated therein at 37 °C, while the cell sheet detached from the surface by lowering the temperature to 25 °C without using any enzymatic agent.
Fabrication of Thermoresponsive Polymer Gradients for Study of Cell Adhesion and Detachment.

Abstract:A poly (N-isopropylacrylamide) (PNIPAAm) gradient covalently anchored on a silicon substrate with a linear variation of thickness was fabricated by continuous injection of the reaction mixture (NIPAAm, CuBr and its ligand, methanol, and water) into a glass chamber containing a silicon water, whose surface had been homogeneously immobilized with bromoisobutyryl bromide (BIBB). Because of the good control of the surface-initiated atom transfer radical polymerization (SI-ATRP) technique, the thickness of the PNIPAAm brushes was linearly proportional to the polymerization time. As a result, the gradient length and sharpness could be easily controlled by the experimental parameters such as the polymerization time and the injection rate. The as-prepared PNIPAAm gradients were characterized by ellipsometry, water contact angle, and atom force microscopy to detect their alteration of the thickness, surface wettability, and morphology, confirming the gradient structure. X-ray photoelectron spectroscopy confirmed the surface composition of the PNIPAAm. In vitro culture of HepG2 cells was implemented on the gradient surfaces, revealing that the cells could adhere at 37 degrees C and could be detached at 24 degrees C when the gradient thickness was in the range of 20-45 nm. The work thus develops a method to fabricate the stable gradient surface with better quality control, and clarifies in a facile manner the appropriate thickness of the PNIPAAm brushes in terms of cell adhesion and detachment.

Abstract:Thermoresponsive surface was prepared from commercial poly(vinylidene fluoride) (PVDF) films via surface-initiated atom transfer radical polymerization. The direct initiation of the secondary fluorinated site of PVDF facilitated grafting of the N-isopropylacrylamide (NIPAAm) monomer. The PVDF surfaces grafted with poly(N-isopropylacrylamide) [P(NIPAAm)] were characterized by X-ray photoelectron spectroscopy. Kinetics study revealed that the P(NIPAAm) chain growth from the PVDF surface was consistent with a "controlled" process. The temperature-dependent swelling behavior of the surfaces in aqueous solution was studied by atomic force microscope. At 37 degrees C [above the lower critical solution temperature (LCST, about 32 degrees C) of NIPAAm], the seeded cells adhered and spread on the NIPAAm grafted PVDF surface. Below the LCST, the cells detached from the P(NIPAAm)-grafted PVDF surface spontaneously. A thermo-responsive PNIPAAm-grafted-PDMS surface used for cell culture in microfluidic channels

Abstract:A thermo-responsive PNIPAAm-grafted-PDMS (PNIPAAm-g-PDMS) surface was used for cell culture. Cos7 cells could adhere to the PNIPAAm-g-PDMS surface and proliferated therein at 37°C. The cultured cells could detach from the surface at room temperature without trypsin digestion. The viability of cells harvested from the PNIPAAm-g-PDMS surface via lowering temperature was higher than that of cells harvested from native PDMS surface via trypsin digestion. A PNIPAAm-g-PDMS microchannel was designed for the in-channel thermo-modulated cell culture/harvest. Cells could be long-term cultivated and be harvested without enzymatic treatment. Successful thermo-modulated cell passage operation was demonstrated in the microfluidic chips with the PNIPAAm-gPDMS channels.
Tuning the Thermoresponsive Behavior of Surface-Attached PNIPAM Networks: Varying the Crosslinker Content in SI-ATRP

Abstract:The synthesis and thermoresponsive properties of surface-attached poly(N-isopropylacrylamide)-co-N,N′-methylene bisacrylamide (PNIPAM-co-MBAM) networks are investigated. The networks are formed via SI-ARGET-ATRP ("grafting-from") on thiol-based initiator-functionalized gold films. This method is reliable, well controlled, fast, and applicable to patterned surfaces (e.g., nanopores) for networks with dry thicknesses >20 nm. Surface-attached PNIPAM-co-MBAM gels are swollen below their volume phase transition temperature but above collapse without complete expulsion of water (retain ∼50 vol %). The swelling/collapse transition is studied using complementary SPR and QCMD techniques. The ratio between swollen and collapsed heights characterizes the thermoresponsive behavior and is shown to not depend on network height but to vary with MBAM content. The higher the proportion of the crosslinker, the lower the magnitude of the phase transition, until all responsiveness is lost at 5 mol % MBAM. The temperature range of the transition is broadened for more crosslinked PNIPAM-co-MBAM gels but remains centered around 32 °C. Upon reswelling, less crosslinked networks display sharp transitions, while for those containing ≥3 mol % MBAM, transitions remain broad. This tunable behavior persists for gels on nanostructured gold surfaces. Investigating PNIPAM-co-MBAM networks on gold plasmonic nanowell arrays is a starting point for expanding their scope as thermo-controlled nanoactuators.This article has not yet been cited by other publications.

Abstract:Stimuli-responsive polymer interfaces with tethered poly (N-isopropylacrylamide) (PNIPAm) chains and its cross-linked porous hydrogel as sensitive phases were fabricated by cycling an electronic potential in aqueous solution. Surface morphological analysis and electrochemical measurements revealed that, owing to their different novel structures, the PNIPAm chain-modified interface showed ON/OFF switch behavior, whereas the PNIPAm gel-modified interface exhibited a 'breathing in' process. The results suggest that the interfacial physicochemical properties, which are greatly affected by the graft conformations and topologies of PNIPAm on the substrates, could be effectively modulated by easily varying the synthesis conditions and are investigated by simple electrochemical methods. The results also demonstrate that the ON/OFF switch behavior of the PNIPAm chain-modified interface has potential applications in controlled ion/molecule permeation, and the 'breathing in' mechanism of the gel-modified interface might be applied to immobilize ions or nanoparticles. These thermo-sensitive interfaces might be used to design adaptive/responsive biocompatible surfaces in a variety of areas.
In Situ Preparation of Tannic Acid-Modified Poly(N-isopropylacrylamide) Hydrogel Coatings for Boosting Cell Response

Abstract:The improvement of the capability of poly(N-isopropylacrylamide) (PNIPAAm) hydrogel coating in cell adhesion and detachment is critical to efficiently prepare cell sheets applied in cellular therapies and tissue engineering. To enhance cell response on the surface, the amine group-modified PNIPAAm (PNIPAAm-APTES) nanohydrogels were synthesized and deposited spontaneously on tannic acid (TA)-modified polyethylene (PE) plates. Subsequently, TA was introduced onto PNIPAAm-APTES nanohydrogels to fabricate coatings composed of TA-modified PNIPAAm-APTES (PNIPAAm-APTES-TA). Characterization techniques, including TEM, SEM, XPS, and UV-Vis spectroscopy, confirmed the effective deposition of hydrogels of PNIPAAm as well as the morphologies, content of chemical bonding-TA, and stability of various coatings. Importantly, the porous hydrogel coatings exhibited superhydrophilicity at 20 °C and thermo-responsive behavior. The fluorescence measurement demonstrated that the coating's stability effectively regulated protein behavior, influencing cell response. Notably, cell response tests revealed that even without precise control over the chain length/thickness of PNIPAAm during synthesis, the coatings enhanced cell adhesion and detachment, facilitating efficient cell culture. This work represented a novel and facile approach to preparing bioactive PNIPAAm for cell culture.

Proliferation and Multi-Differentiation Potentials of Human Mesenchymal Stem Cells on Thermoresponsive PDMS Surfaces Grafted with PNIPAAm.

Abstract:The thermo-responsivity of PNIPAAm [poly(N-isopropylcarylamide)]-grafted PDMS [poly(dimethylsiloxane)] surface is a property that could be feasibly used for detaching cells adhered on the surface. We used benzophenone-initiated photopolymerization to graft PNIPAAm on PDMS substrates to construct the PNIPAAm-grafted PDMS surface and this PDMS surface was highly thermo-responsive. hMSCs (human mesenchymal stem cells) were used to analyse the proliferation and multi-differentiation of stem cells on the PNIPAAm-grafted PDMS surface. The results showed that hMSCs could adhere on the PNIPAAm-grafted PDMS surface at 37 degrees C and form cell colonies, and then become fibroblastic. The proliferation potential of hMSCs on the PNIPAAm-grafted PDMS surface was not significantly different from that on a plate surface coated with gelatin. However, as it proved easier to detach cells from the surface, by changing temperature, a higher viability of detached cells could be obtained with the PNIPAAm-grafted PDMS surface, using a temperature shift, compared with a gelatin-coated surface, where cells are detached by treatment with trypsin. hMSCs on the PNIPAAm-grafted PDMS surface were induced into osteoblasts, adipocytes and neurocytes under osteogenic medium, adipogenic medium and neurogenic medium respectively. The PNIPAAm-grafted PDMS surface was favourable for osteogenesis of hMSCs, although the potentials of adipogenesis and neurogenesis of hMSCs on the PNIPAAm-grafted PDMS surface were similar to those on the plate surface coated with gelatin. The above results demonstrate that the PNIPAAm-grafted PDMS surface not only kept the potentials of proliferation and multi-differentiation of hMSCs, but also increased the viability of hMSCs.
Thermomodulated Cell Culture/harvest in Polydimethylsiloxane Microchannels with Poly(n-Isopropylacrylamide)-grafted Surface

Abstract:Cell culture and harvest are the most upstream operation for a completely integrated cell assay chip. In our previous work, thermoresponsive poly(N-isopropylacrylamide) (PNIPAAm) was successfully grafted onto polydimethylsiloxane (PDMS) surface via benzophenone-initiated photopolymerization. In the present work, the PNIPAAm-grafted-PDMS (PNIPAAm-g-PDMS) surface was explored for thermomodulated cell culture and noninvasive harvest in microfluidic channels. Using COS 7 fibroblast from African green monkey kidney as the model cells, the thermomodulated adhering and detaching behaviors of the cells on the PNIPAAm-g-PDMS surfaces were optimized with respect to PNIPAAm-grafting yields and gelatin modification. The viability of the cells cultured on and harvested from the PNIPAAm-g-PDMS surface with the thermomodulated noninvasive protocol was estimated against the traditional cell culture∕harvest method involving trypsin digestion. The configuration of the microchannel on the PNIPAAm-g-PDMS chip was evaluated for static cell culture. Using a pipette-shaped PNIPAAm-g-PDMS microchannel, long-term cell culture could be achieved at 37 °C with periodic change of the culture medium every 12 h. After moving the microchip from the incubator set at 37 °C to the room temperature, the proliferated cells could be spontaneously detached from the PNIPAAm-g-PDMS surface of the upstream chamber and transferred by a gentle fluid flow to the downstream chamber, wherein the transferred cells could be subcultured. The thermomodulated cell culture, harvest, and passage operations on the PNIPAAm-g-PDMS microfluidic channels were demonstrated.
Preparation of thermosensitive PNIPAm‐based copolymer coated cytodex 3 microcarriers for efficient nonenzymatic cell harvesting during 3D culturing

Abstract:Enzymatic detachment of cells might damage important features and functions of cells and could affect subsequent cell-based applications. Therefore, non-enzymatic cell detachment using thermosensitive polymer matrix is necessary for maintaining cell quality after harvesting. In this study, we prepared thermosensitive PNIPAm-co-AAc-b-PS and PNIPAm-co-AAm-b-PS copolymers and LCST was tuned near to body temperature. Then, spin coated polymer films were prepared for cell adhesion and thermal-induced cell detachment. The alpha-step analysis and SEM image of the films suggested that the thickness of the films depends on the molecular weight and concentration which ranged from 206 nm to 1330 nm for PNIPAm-co-AAc-b-PS and 97.5 nm to 497 nm for PNIPAm-co-AAm-b-PS. The contact angles of the films verified that the polymer surface was moderately hydrophilic at 37°C. Importantly, RAW264.7 cells were convincingly proliferated on the films to a confluent of >80 % within 48 days and abled to detach by reducing the temperature. However, relatively more cells were grown on PNIPAm-co-AAm-b-PS (5%w/v) films and thermal-induced cell detachment was more abundant in this formulation. As a result, PNIPAm-co-AAm-b-PS (5%w/v) was further used to coat commercial cytodex 3 microcarriers for 3D cell culturing and interestingly enhanced cell detachment with preserved potential of recovery was observed at a temperature of below LCST. Thus, surface modification of microcarriers with thermosensitive PNIPAm-co-AAm-b-PS could be vital strategy for non-enzymatic cell detachment and to achieve adequate number of cells with maximum cell viability and functionality.This article is protected by copyright. Gradient immobilization of a cell adhesion RGD peptide on thermal responsive surface for regulating cell adhesion and detachment.

Abstract:Using surface initiated atomic transfer radical polymerization (ATRP) and an injection method, a poly(N-isopropylacrylamide)-b-poly(acrylic acid)-g-RGD (PNIPAAm-b-PAA-g-RGD) gradient surface was prepared. First, a thermoresponsive surface with a constant thickness of PNIPAAm was fabricated, onto which the AA monomers were block copolymerized using the PNIPAAm macromolecules as initiators. During this process, a continuous injection method was employed to yield a molecular weight gradient of PAA on the underlying uniform PNIPAAm layer. RGD peptide was finally covalently immobilized onto the PAA gradient by carbodiimide chemistry. In vitro culture of HepG2 cells showed that immobilization of the RGD peptide could accelerate cell attachment, while the thermoresponsive layer beneath could effectively release the cells by simply lowering temperature. Thus, the PNIPAAm-b-PAA-g-RGD gradient surface, combining the thermal response with cell affinity properties, can well regulate the cell adhesion and detachment, which may thus be useful for investigation of cell–substrate interactions with a smaller number of samples.
3D Temperature-Controlled Interchangeable Pattern for Size-Selective Nanoparticle Capture

Abstract:Patterned surfaces with distinct regularity and structured arrangements have attracted great interest due to their extensive promising applications. Although colloidal patterning has conventionally been used to create such surfaces, herein, we introduce a novel 3D patterned poly(N-isopropylacrylamide) (PNIPAM) surface, synthesized by using a combination of colloidal templating and surface-initiated photoinduced electron transfer-reversible addition-fragmentation chain transfer (SI-PET-RAFT) polymerization. In order to investigate the temperature-driven 3D morphological variations at a lower critical solution temperature (LCST) of ∼32 °C, multifaceted characterization techniques were employed. Atomic force microscopy confirmed the morphological transformations at 20 and 40 °C, while water contact angle measurements, upon heating, revealed distinct trends, offering insights into the correlation between surface wettability and topography adaptations. Moreover, quartz crystal microbalance with dissipation monitoring and electrochemical measurements were employed to detect the topographical adjustments of the unique hollow capsule structure within the LCST. Tests using different sizes of PSNPs shed light on the size-selective capture-release potential of the patterned PNIPAM, accentuating its biomimetic open-close behavior. Notably, our approach negates the necessity for expensive proteins, harnessing temperature adjustments to facilitate the noninvasive and efficient reversible capture and release of nanostructures. This advancement hopes to pave the way for future innovative cellular analysis platforms.

Abstract:Meticillin-resistant Staphylococcus aureus (MRSA) strains producing the potent tissue necrotizing toxin Panton–Valentine leukocidin (PVL) encoded by the pvl gene, and harbouring SCCmec type IV or V elements, have been implicated as being associated with MRSA infection acquired in the community setting (Vandenesch et al., 2003; Boyle-Vavra & Daum, 2007). Since the 1990s, community-associated (CA)-MRSA infections have shown a steady increase, and are becoming a significant public-health concern. PVLpositive MRSA isolates have been shown to have distinct genetic backgrounds in different areas, for instance, multilocus sequence typing (MLST) types ST80, ST8 and ST59 were found in the major clones in Europe, USA and Taiwan, respectively (Vandenesch et al., 2003; Tristan et al., 2007; Chen et al., 2005).
Surface Modification of PTLG Terpolymer Using PNIPAAm and Its Cell Adhesion/Detachment Behavior

Abstract:Novel biodegradable terpolymer poly (1,3-trimethylene carbonate-b-(l-lactide-r-glycolide)) grafted with poly(N-isopropylacrylamide) (PTLG-PNIPAAm) is prepared by plasma-induced polymerization. The experimental results show that PNIPAAm segments of samples have a dramatic effect on surface layer structure and cell behavior of PTLG-PNIPAAm. The thickness of PNIPAAm layer on PTLG surface increases rapidly from 50 nm to 1 mu m with the molecular weight of samples increasing. Cell toxicity analysis indicates that PTLG-PNIPAAm possesses good cytocompatibility to human adipose-derived stem cells (hADSCs). For cell adhesion and detachment process on modified surface, the "hook and loop" process is proposed to understand the cell adhesion on modified surface with coiled conformation PNIPAAm segments at 37 degrees C, and then the cell sheet detached from surface spontaneously due to the conformation of PNIPAAm segments extends below its lower critical solution temperature. The research demonstrates that with increasing thickness of the PNIPAAm layer on PTLG surface, the hADSCs with well cell viability presents rapider thermal-responsive detachment behavior. Therefore, the surface modification of biodegradable PTLG with stimuli-sensitive functional PNIPAAm is an efficacious route for further application in stem cell culture and tissue engineering stent.
Submicrometric Films of Surface-Attached Polymer Network with Temperature-Responsive Properties

Abstract:Temperature-responsive properties of surface-attached poly(N-isopropylacrylamide) (PNIPAM) network films with well-controlled chemistry are investigated. The synthesis consists of cross-linking and grafting preformed ene-reactive polymer chains through thiol--ene click chemistry. The formation of surface-attached and cross-linked polymer films has the advantage of being wellcontrolled without any caution of no-oxygen atmosphere or addition of initiators. PNIPAM hydrogel films with same cross-link density are synthesized on a wide range of thickness, from nanometers to micrometers. The swelling-collapse transition with temperature is studied by using ellipsometry, neutron reflectivity, and atomic force microscopy as complementary surface-probing techniques. Sharp and high amplitude temperature-induced phase transition is observed for all submicrometric PNIPAM hydrogel films. For temperature above LCST, surface-attached PNIPAM hydrogels collapse similarly but without complete expulsion of water. For temperature below LCST, the swelling of PNIPAM hydrogels depends on the film thickness. It is shown that the swelling is strongly affected by the surface attachment for ultrathin films below $\sim$150 nm. For thicker films above 150 nm (to micrometers), surface-attached polymer networks with the same cross-link density swell equally. The density profile of the hydrogel films in the direction normal to the substrate is confronted with in-plane topography of the free surface. It results that the free interface width is much larger than the roughness of the hydrogel film, suggesting pendant chains at the free surface.

Abstract:Designing cell-compatible, bio-degradable, and stimuli-responsive hydrogels is very important for biomedical applications in cellular delivery and micro-scale tissue engineering. Here, we report achieving flexible control of cellular microencapsulation, permeability, and release by rationally designing a diblock copolymer, alginate-conjugated poly(N-isopropylacrylamide) (Alg-co-PNiPAM). We use the microfluidic technique to fabricate the bifunctional copolymers into thousands of mono-disperse droplet-templated hydrogel microparticles for controlled encapsulation and triggered release of mammalian cells. In particular, the grafting PNiPAM groups in the synthetic cell-laden microgels produce lots of nano-aggregates into hydrogel networks at elevated temperature, thereafter enhancing the permeability of microparticle scaffolds. Importantly, the hydrogel scaffolds are readily fabricated via on-chip quick gelation by triggered release of Ca2+ from the Ca-EDTA complex; it is also quite exciting that very mild release of microencapsulated cells is achieved via controlled degradation of hydrogel scaffolds through a simple strategy of competitive affinity of Ca2+ from the Ca-Alginate complex. This finding suggests that we are able to control cellular encapsulation and release through ion-induced gelation and degradation of the hydrogel scaffolds. Subsequently, we demonstrate a high viability of microencapsulated cells in the microgel scaffolds.

Abstract:Thermal responsive polymers were introduced for effectively cell detachment from the extracellular matrix (ECM), where the poor cell adhesive capacity was improved by the immobilized biodhesion molecules (most are proteins) at the termini of grafted polymer. However, this protein-mediated adhesion makes it hard to be precisely controlled and takes longer time to start initial cell attachment. Here we describe a choline phosphate(CP) modified thermal responsive surface with enhanced initial cell adhesion based on multivalent CP-PC (phosphatidyl choline, the head-group of phospholipids in all the eukaryotic cell membranes) interaction. The well-defined functional surfaces were constructed using surface-initiated ATRP and click reaction, where the initial cell attachment reach saturation in 10 min and can be regulated by CP density on the surface. Consequently, the fast cell adhesion significantly accelerates cell spreading and proliferation. The facile cell harvest, as opposed to the original mechanism of thermally-induced cell detachment, is to take advantage of the thermal contraction of the polymer above the LCST, which release the adherent cell by reducing the accessibility of the CP groups to cell membrane PC lipids. Si-CP is expected to provide a promising universal structural model for modulating cell attachment and detachment in tissue engineering and regenerative medicine field.
