Abstract:Type I collagen is the main structural component of skin, tendons, and skin products, such as leather. Understanding the mechanical performance of collagen fibrils is important for understanding the mechanical performance of the tissues that they make up, while the mechanical properties of bulk tissue are well characterized, less is known about the mechanical behavior of individual collagen fibrils. In this study, bovine pericardium is subjected to strain while small angle X-ray scattering (SAXS) patterns are recorded using synchrotron radiation. The change in d-spacing, which is a measure of fibril extension, and the change in fibril diameter are determined from SAXS. The tissue is strained 0.25 (25%) with a corresponding strain in the collagen fibrils of 0.045 observed. The ratio of collagen fibril width contraction to length extension, or the Poisson's ratio, is 2.1 ± 0.7 for a tissue strain from 0 to 0.25. This Poisson's ratio indicates that the volume of individual collagen fibrils decreases with increasing strain, which is quite unlike most engineering materials. This high Poisson's ratio of individual fibrils may contribute to high Poisson's ratio observed for tissues, contributing to some of the remarkable properties of collagen-based materials.

Abstract:The elasmoid scales in teleost fish serve as exemplary models for natural fibre composites with integrated flexibility and protection. Yet, limited research has been focused on the potential structural, chemical, and mechanical heterogeneity within individual scales. This study presents systematic characterizations of the elasmoid scales from black drum fish ( Pogonias cromis ) at different zones within individual scales as a natural fibre composite, focusing on the microscopic structural heterogeneities and corresponding mechanical effects. The focus field at the centre of the scales exhibits a classical tri-layered collagen-based composite design, consisting of the mineralized outermost limiting layer, external elasmodine layer in the middle, and the unmineralized internal elasmodine layer. In comparison, the rostral field at the anterior end of the scales exhibits a two-layered design: the mineralized outermost limiting layer exhibits radii sections on the outer surface, and the inner elasmodine layer consists of collagen fibre-based sublayers with alternating mineralization levels. Chemical and nanoindentation analysis suggests a close correlation between the mineralization levels and the local nanomechanical properties. Comparative finite element modelling shows that the rostral-field scales achieve increased flexibility under both concave and convex bending. Moreover, the evolving geometries of isolated Mandle’s corpuscles in the internal elasmodine layer, transitioning from irregular shapes to faceted octahedrons, suggest the mechanisms of mineral growth and space-filling to thicken the mineralized layers in scales during growth, which enhances the bonding strength between the adjacent collagen fibre layers. This work offers new insights into the structural variations in individual elasmoid scales, providing strategies for bioinspired fibre composite designs with local-adapted functional requirements.

Abstract:The structure and mechanics of fish scales display unusual and attractive features which could inspire new protective materials and systems. This natural material is therefore attracting attention over the past few years, and recent work demonstrated the remarkable performance of individual fish scales. A puncture event as would occur from a predator's attack however involves more than one scale, and in this article we therefore investigate collective mechanisms occurring within the scaled skin of a fish in the event of a predator's attack. We first demonstrate that in striped bass (Morone saxatilis), the scales increase by four to five times the force required to puncture the skin. We show that individual scales from striped bass provide a remarkable barrier against sharp puncture, regardless of the stiffness of the substrate. The scalation pattern in striped bass is such that three scales overlap at any point on the surface of the fish, which we show effectively multiplies the puncture force by three. We determined that the friction between scales is negligible and therefore it does not contribute to increasing puncture force. Likewise, we found that the local arrangement of the scales had little effect on the puncture performance. Interestingly, because the scales are several orders of magnitude stiffer than the substrate, indenting a few isolated scales results in "sinking" of the scales into the substrate. The high local deflections and strain within the soft tissue may then result in blunt injury before the sharp indenter penetrates the scales. Stereo-imaging and image correlation performed around a puncture site in fish reveal that the surrounding scales collectively contribute to redistributing the puncture force over large volume, limiting local deflections and strains in the soft tissues. The structure and mechanisms of natural fish scales therefore offer an effective protection against several types of threat, and may inspire novel versatile protective systems with attractive flexural properties.

Abstract:Skin is the largest organ of many animals. Its protective function against hostile environments and predatorial attack makes high mechanical strength a vital characteristic. Here, we measured the mechanical properties of bass fish skins and found that fish skins are highly ductile with a rupture strain of up to 30–40% and a rupture strength of 10–15 MPa. The fish skins exhibit a strain-stiffening behavior. Stretching can effectively eliminate the stress concentrations near the pre-existing holes and edge notches, suggesting that the skins are highly damage tolerant. Our measurement determined a flaw-insensitivity length that exceeds those of most engineering materials. The strain-stiffening and damage tolerance of fish skins are explained by an agent-based model of a collagen network in which the load-bearing collagen microfibers assembled from nanofibrils undergo straightening and reorientation upon stretching. Our study inspires the development of artificial skins that are thin, flexible, but highly fracture-resistant and widely applicable in soft robots.

Abstract:Fish skin is a biological material with high flexibility and compliance and can provide good mechanical protection against sharp punctures. This unusual structural function makes fish skin a potential biomimetic design model for flexible, protective, and locomotory systems. In this work, tensile fracture tests, bending tests, and calculation analyses were conducted to study the toughening mechanism of sturgeon fish skin, the bending response of the whole Chinese sturgeon, and the effect of bony plates on the flexural stiffness of the fish body. Morphological observations showed some placoid scales with drag-reduction functions on the skin surface of the Chinese sturgeon. The mechanical tests revealed that the sturgeon fish skin displayed good fracture toughness. Moreover, flexural stiffness decreased gradually from the anterior region to the posterior region of the fish body, which means that the posterior region (near the tail) had higher flexibility. Under large bending deformation, the bony plates had a specific inhibition effect on the bending deformation of the fish body, especially in the posterior region of the fish body. Furthermore, the test results of the dermis-cut samples showed that the sturgeon fish skin had a significant impact on flexural stiffness, and the fish skin could act as an external tendon to promote effective swimming motion.
Toughening mechanism of coelacanth-fish-inspired double-helicoidal composites

Abstract:The scales of coelacanth fish feature a rare double-helicoidal structure of collagenous fibrils, exhibiting great toughness. In this study, inspired by such biomaterials, engineering composite materials are designed and fabricated. Quasi-static three-point bending and Charpy impact tests are performed to evaluate their mechanical performance compared with the single-helicoidal counterparts (emulated from the Bouligand structure in Odontodactylus scyllarus). Multiple delaminations are observed in the double-helicoidal composites, which preclude tensile crack propagation and then prolong the failure displacement; meanwhile, for single-helicoidal composites, the fiber architecture at the mid-plane serves as a vital role for hindering translaminar cracks propagation across the mid-plane. The dynamic energy absorption of double-helicoidal composites can reach 163.44 kJ·m-2, 26.5% greater than that of the corresponding single-helicoidal samples, which is attributed to the more complex failure modes including fiber breakage and matrix cracking that both contribute to energy absorption. Double-helicoidal composites are not sensitive to fiber orientations on failure mode and such attribute enables more design freedoms. Additionally, the interlaminar stress is analyzed, and a greater value of the interlaminar stresses σ13 is discovered to be responsible for the delamination of double-helicoidal composites. Results reveal the underlying toughening mechanism of coelacanth-fish-inspired double-helicoidal composites and promote the next-generation impact-resistant composites design.

Abstract:To study the structure and the cytocompatibility of fish scales from Carassius auratus, scanning electron microscopy (SEM) was used to observe the morphology of fish scales treated with different processing methods. Based on varying morphologies and components, the fish scales can be divided into three regions on the surface and three layers in vertical. The functions of these three individual layers were analyzed. SEM results show that the primary inorganic components are spherical or cubic hydroxyapatite (HA) nanoparticles. The fish scales have an ~60° overlapped plywood structure of lamellas in the fibrillary plate. The plywood structure consists of co-aligned type I collagen fibers, which are parallel to the HA lamellas. X-ray diffraction (XRD), thermogravimetric analysis/differential scanning calorimetry (TGA/DSC) and Fourier transform infrared (FTIR) analysis indicate that the main components are HA and type I collagen fibers. MC3T3-E1 cell culture results show a high cytocompatibility and the ability to guide cell proliferation and migration along the scale ridge channels of the fish scales. This plywood structure provides inspiration for a structure-enhanced composite material.

Abstract:Abstract Biological structures integrate morphometry (shape-based rules) with materials design to maximize organism survival. The exoskeleton of the armored fish, Polypterus senegalus , balances flexibility with protection from predatory and territorial threats. Material properties of the exoskeleton are known; however, the geometric design rules underlying its anisotropic flexibility are uncharacterized. Here, we show how scale shape, articulation, and composite architecture produce anisotropic mechanics using bio-inspired, multi-material 3D-printed prototypes. Passive loading (draping) shows that compliant connections between the scales contribute to mechanical anisotropy. Simulated and experimental active loading (bending) show orientation-dependent stiffness ranging over orders of magnitude, including ‘mechanical invisibility’ of the scales where they do not add stiffness to the exoskeleton. The results illustrate how morphometry provides a powerful tool to tune flexibility in composite architectures independent of varying constituent materials composition. We anticipate that introducing morphometric design strategies will enable flexible, protective systems tuned to complex shapes and functions.
Collagen Membrane Derived from Fish Scales for Application in Bone Tissue Engineering

Abstract:Guided tissue/bone regeneration (GTR/GBR) is currently the main treatment for alveolar bone regeneration. The commonly used barrier membranes in GTR/GBR are collagen membranes from mammals such as porcine or cattle. Fish collagen is being explored as a potential substitute for mammalian collagen due to its low cost, no zoonotic risk, and lack of religious constraints. Fish scale is a multi-layer natural collagen composite with high mechanical strength, but its biomedical application is limited due to the low denaturation temperature of fish collagen. In this study, a fish scale collagen membrane with a high denaturation temperature of 79.5 °C was prepared using an improved method based on preserving the basic shape of fish scales. The fish scale collagen membrane was mainly composed of type I collagen and hydroxyapatite, in which the weight ratios of water, organic matter, and inorganic matter were 20.7%, 56.9%, and 22.4%, respectively. Compared to the Bio-Gide® membrane (BG) commonly used in the GTR/GBR, fish scale collagen membrane showed good cytocompatibility and could promote late osteogenic differentiation of cells. In conclusion, the collagen membrane prepared from fish scales had good thermal stability, cytocompatibility, and osteogenic activity, which showed potential for bone tissue engineering applications.

Abstract:Abstract Natural hard composites like human bone possess a combination of strength and toughness that exceeds that of their constituents and of many engineered composites. This augmentation is attributed to their complex hierarchical structure, spanning multiple length scales; in bone, characteristic dimensions range from nanoscale fibrils to microscale lamellae to mesoscale osteons and macroscale organs. The mechanical properties of bone have been studied, with the understanding that the isolated microstructure at micro- and nano-scales gives rise to superior strength compared to that of whole tissue, and the tissue possesses an amplified toughness relative to that of its nanoscale constituents. Nanoscale toughening mechanisms of bone are not adequately understood at sample dimensions that allow for isolating salient microstructural features, because of the challenge of performing fracture experiments on small-sized samples. We developed an in situ three-point bend experimental methodology that probes site-specific fracture behavior of micron-sized specimens of hard material. Using this, we quantify crack initiation and growth toughness of human trabecular bone with sharp fatigue pre-cracks and blunt notches. Our findings indicate that bone with fatigue cracks is two times tougher than that with blunt cracks. In situ data-correlated electron microscopy videos reveal this behavior arises from crack-bridging by nanoscale fibril structure. The results reveal a transition between fibril-bridging (∼1 μ m) and crack deflection/twist (∼500 μ m) as a function of length-scale, and quantitatively demonstrate hierarchy-induced toughening in a complex material. This versatile approach enables quantifying the relationship between toughness and microstructure in various complex material systems and provides direct insight for designing biomimetic composites.

Abstract:A series of fish collagen proteins were prepared for wound healing application. The physicochemical properties such as molecular weight, solubility, thermal stability of the products was studied. Fourier transform infrared spectroscopy (FTIR) was employed to identify the chemical bonds of fish collagen and its resemblance to type 1 collagen. The surface morphology of the prepared collagen was also studied by scanning electron microscopy (SEM). Results obtained showed that Tilapia and Grey mullet collagen consists of identical four subunits including β chains, γ chains and two α chains (α1 and α2). Due to strong fibrillar nature of collagen, it greatly enhances the cell adhesion capacities of the prepared products. Moreover, the antibacterial activity of collagen was investigated. It was found that all the extracted collagen products have inhibitory activity against all of the tested bacteria. Wound healing efficiency of the prepared collagen was also investigated through assessing the rate of wound contraction, histological evaluations demonstrated that collagen treatment resulted in better resolution and closure of the wound. The above analysis suggested that the isolated fish collagen can be a potential candidate for wound healing application.

Abstract:Ferroelectric memories are the most promising alternative to traditional embedded nonvolatile memories, such as flash and EEPROMs, because of their fast read/write cycle time, non-volatile data retention, low voltage/low power operation and low number of additional masks for fabrication (+2). An embedded ferroelectric memory (FRAM) has been developed using a 1.5 V, 130 nm 5 metal layer Cu/FSG logic process. The only modification to the logic process was the addition of a ferroelectric process module consisting of two additional masks (FECAP, VIAO) immediately before MET1. The ferroelectric was 70 nm Pb(Zr,Ti)O3 (PZT) deposited by metalorganic chemical vapor deposition (MOCVD). The electrical properties of a 8 Mb 1T-1C embedded FRAM were characterized. This eFRAM process has been used to simultaneously fabricate a digital signal processor (DSP) using the eFRAM process flow and the operating frequency is nearly the same relative to the CMOS baseline. This eFRAM process flow creates a technology platform that enables ultra-low-power devices.
Skin stiffness in ray-finned fishes: Contrasting material properties between species and body regions

Abstract:The skin of aquatic vertebrates surrounds all the mechanical lineages of the body and must, therefore, play an important role in locomotion. A cross-woven collagenous dermal design has converged across several clades of vertebrates. Despite this intriguing pattern, the biomechanical role of skin in swimming fishes remains largely unknown. A direct force transmission role for fish skin has been proposed, a hypothesis that is supported by the arrangement of the connective tissues linking the skin to the axial musculature. To evaluate this direct force-transmission hypothesis, we undertook hundreds of uniaxial tensile tests on skin samples from coho salmon (Oncorhynchus kisutch), Florida pompano (Trachinotus carolinus), and red snapper (Lutjanus campechanus). To do this, we developed highly precise, low-cost, custom-built material testing units. To augment our data, we also assembled a data set of skin stiffness of four additional species of actinopterygians fishes from previously published studies. We found that stiffness varies significantly between species and that the skin of our study species was increasingly stiff along a rostrocaudal gradient. Placing our results in the context of the limited body of previous work, we found that species with lower skin stiffness exhibit shorter propulsive wavelengths and low thrust production at the caudal fin and species with higher skin stiffness possess longer propulsive wavelengths and high thrust production at the caudal fin. In addition, we found that mean collagen fiber angle was close to 50° and that fiber angle was lower in posterior samples than in anterior and midlateral samples. Taken as a whole, our mechanical and morphological results support the hypothesis that the skin functions as an important direct force-transmission device in actinopterygians whereby muscular force generated in anterior myotomes is transmitted to the posterior of the body through the increasingly stiff skin.

Abstract:Despite the fact that various reports have been discussing bone tissue regeneration, precise bone tissue manipulation, such as controlling the physical properties of the regenerated bone tissue, still remains a big challenge. Here, we focused on the teleost fish ribs showing flexible and tough mechanical properties to obtain a deeper insight into the structural and functional features of bone tissue from different species, which would be valuable for the superior design of bone-mimicking materials. Herein, we examined their compositions, microstructure, histology, and mechanical properties. The first rib of Carassius langsdorfii showed a higher Young’s modulus with a small region of chondrocyte clusters compared with other smaller ribs. In addition, highly oriented collagen fibers and osteocytes were observed in the first rib, indicating that the longest first rib would be more mature. Moreover, the layer-by-layer structure of the oriented bone collagen was observed in each rib. These microarchitectural and compositional findings of fish rib bone would give one the useful idea to reproduce such a highly flexible rib bone-like material.

Large extensibility and strong backbone of protein are essential for the toughness of natural materials

Abstract:Protein-based natural materials and fibers display superior strength and toughness, which have been attributed to the parallel staggered arrangement of microstructure, as well as to the unique force-extension behavior of protein-rich organic matrix. It has been suggested in recent years that the sawtooth behavior in the force-extension curve of protein may play a major role in the toughness of natural materials. To examine this hypothesis, here we conduct rigorous fracture mechanics simulations and theoretical analysis of crack propagation in a protein-like material to show that the sawtooth behavior actually only has a minimal effect on the fracture toughness. Instead, we show that it is the combination of extraordinary extensibility due to domain unfolding and significant force increase at the stage of backbone stretching that gives the high steady-state fracture toughness of a protein-like material. In fact, we show that an infinite steady-state fracture toughness, indicative of a transition of failure mode from crack propagation to uniform rupture near the theoretical strength of the material, can be achieved for protein-like materials with proper designs of domain unfolding and backbone stretching.

Abstract:Marine collagen derived from fish scales, skin, and bone has been widely investigated for application as a scaffold and carrier due to its bioactive properties, including excellent biocompatibility, low antigenicity, and high biodegradability and cell growth potential. Fish type I collagen is an effective material as a biodegradable scaffold or spacer replicating the natural extracellular matrix, which serves to spatially organize cells, providing them with environmental signals and directing site-specific cellular regulation. This study was conducted to confirm the safety of fish (tilapia) atelocollagen for use in clinical application. We performed in vitro and in vivo biological studies of medical materials to investigate the safety of fish collagen. The extract of fish collagen gel was examined to clarify its sterility.
