Abstract:Photoacoustic mesoscopy is an emerging noninvasive imaging modality, which offers high resolution 3D images with optical absorption contrast at depths beyond the light diffusion limit. Ultrasound sensor based on a Fabry-Perot (FP) polymer cavity has the following advantages: broadband frequency response, wide angular coverage and small footprint. We present a photoacoustic mesoscope based on a tunable Fabry-Perot interferometer, which offers the potential for reducing system cost and making array of such sensor. A cw diode laser working at 650nm was used to heat the sensor, offering an active tune range of 5nm by elongating the cavity. Ex-vivo and in-vivo imaging experiments demonstrated the imaging capability of this PA mesoscope, showing great potential in biological and medical applications.

Abstract:Photoacoustic tomography (PAT) is an emerging technology for biomedical imaging that combines the superiorities of high optical contrast and acoustic penetration. In the PAT system, more photoacoustic (PA) signals are preferred to be detected from full field of view to reconstruct PA images with higher fidelity. However, the requirement for more PA signals detection leads to more time consumption for single-channel scanning based PAT system, or higher cost of data acquisition (DAQ) module for an array-based PAT system. To address this issue, we proposed a programmable acoustic delay line module to reduce DAQ cost and accelerate imaging speed for PAT system. The module is based on bidirectional conversion between acoustic signals and electrical signals, including ultrasound transmission in between to provide sufficient time delay. The acoustic delay line module achieves tens or hundreds of microseconds delay for each channel, and is controlled by a programmable control unit. In this work, it achieves to merge four inputs of PA signals into one output signal, which can be recovered into original four PA signals in the digital domain after DAQ. The imaging experiments of pencil leads embedded in agar phantom is conducted by the PAT system equipped with the proposed programmable acoustic delay-line module, which demonstrated its feasibility in biomedical imaging system.

Abstract:Photoacoustic (PA) measurements encode the information associated with both physical microstructures and chemical contents in biological tissues. A two-dimensional physio-chemical spectrogram (PCS) can be formulated by combining the power spectra of PA signals acquired at a series of optical wavelengths. The analysis of PCS, or namely PA physio-chemical analysis (PAPCA), enables the quantification of the relative concentrations and the spatial distributions of a variety of chemical components in the tissue. This study validated the feasibility of PAPCA in characterizing liver conditions during the progression of non-alcoholic fatty liver disease. A catheter based setup facilitating measurement in deep tissues was also tested.

Abstract:We have developed a photoacoustic (PA) sensor using a low-power, continuous- wave laser and a kHz-range microphone. The sensor is simple, flexible, cost-effective, and compatible with commercial optical microscopes. The sensor enables noncontact PA measurements through air, whereas most current existing PA techniques require an acoustic coupling liquid for detection. The PA sensor has three main components: one is the chamber that holds the sample, the second is a resonator column used to amplify the weak PA signals generated within the sample chamber, and the third is a microphone at the end of the resonator column to detect the amplified signals. The chamber size was designed to be 8 mm × 3 mm as the thermal diffusion length and viscous-thermal damping of air at room pressure and temperature are 2 and 1 mm, respectively. We numerically and experimentally examined the effect of the resonator column size on the frequency response of the PA sensor. The quality factor decreased significantly when the sample chamber size was reduced from 4 mm × 3 mm to 2 mm × 3 mm due to thermos-viscous damping of the air. The quality factor decreased by 27%, demonstrating the need for optimal design for the sample chamber and resonator column size. The system exhibited noise equivalent molecular sensitivity (NEM) per unit bandwidth (NEM / √ Δf) of ∼19,966 Hz - 1/2 or 33 × 10 - 21 mol or 33 zeptomol, which is an improvement of 2.2 times compared to the previous system design. This PA sensor has the potential for noncontact high-resolution PA imaging of materials without the need for coupling fluids. .

Abstract:Photoacoustic (PA) tomography (PAT), also referred to as optoacoustic tomography, is an emerging biomedical imaging method based on the PA eect (Xu and Wang 2006, Li and Wang 2009, Oraevsky and Karabutov 2003). e PA eect was reported by Alexander Graham Bell in 1880 (Bell 1880), and refers to the generation of acoustic waves by substances being illuminated by electromagnetic (EM) waves of varying power. e PA eect results from the variational thermal expansion within the material as it absorbs the EM radiative energy. Since the ability of EM absorption re¦ects a material’s characteristics, much physical and chemical information can be revealed by studying PA signals. Historically, the PA eect was of little research interest for a long time, but the invention of laser, an adequate PA illumination source, changed matters drastically. Abstract:Photoacoustic (PA) spectral response technique has shown good promise in efficient preclinical tissue diagnosis by depicting mechano-biological properties due to high spatial resolution and penetration depth. The conventional PA-based system is a pump–probe technique that utilizes neodymium-doped yttrium aluminum garnet pulsed laser as a pump and an ultrasound sensor as a probe. For biomedical studies, high-speed PA signals need to be acquired, requiring higher bandwidth ultrasound sensors. While the bandwidth increases, they exhibit a very low signal-to-noise ratio that inhibits acquiring PA signals of biomedical samples. An interferometer-based probe has recently been investigated as a potential ultrasound probe for obtaining PA signals as an alternative. This optical PA detection technique offers high sensitivity by combining low acoustic impedance with high electromechanical coupling. However, there is a lack of exploration of the same for real-time biomedical studies. This work shows the development of a homodyne Mach-Zehnder interferometer-based PA spectral response (PASR) followed by a correlation study between the conventional ultrasound sensor and the interferometer-based sensor. Further, this study demonstrates the capability of continuous monitoring of vascular growth and the effect of an antidrug (Cisplatin) on the vasculature tested on a chick-embryo chorioallantoic membrane model. PASR was able to monitor growth changes within one day, which was not possible with conventional methods. This opens up potential possibilities for using this technique in biomedical applications.

Abstract:Photoacoustic imaging (PAI) is a non-invasive, high-resolution hybrid imaging modality that combines optical excitation and ultrasound detection. PAI can image endogenous chromophores (melanin, hemoglobin, etc.) and exogenous contrast agents in different medical applications. However, most current equipment uses sophisticated and complicated OPO lasers with tuning and stability features inconsistent with broad clinical deployment. As the number of applications of PAI in medicine increases, there is an urgent need to make the imaging equipment more compact, portable, and affordable. Here, portable light emitting diode - based photoacoustic imaging (PLED-PAI) was introduced and characterized in terms of system specifications, light source characterizations, photoacoustic spatial/temporal resolution, and penetration. The system uses two LED arrays attached to the sides of a conventional ultrasound transducer. The LED pulse repetition rate is tunable between 1 K Hz, 2 K Hz, 3 K Hz, and 4 K Hz. The axial resolution was 0.268 mm, and the lateral resolution was between 0.55 and 0.59 mm. The system could detect optical absorber (pencil lead) at a depth of 3.2 cm and the detection limits of indocyanine green (ICG) and methylene blue (MB) were 9 μM and 0.78 mM. In vivo imaging of labeled human mesenchymal stem cells was achieved to confirm compatibility with small animal imaging. The characterization we report here may have value to other groups evaluating commercially available photoacoustic imaging equipment.
Induction chemotherapy with a new regimen alternating cisplatin, fluorouracil with mitomycin, hydroxyurea and bleomycin in carcinomas of nasopharynx or other sites of the head and neck region

Abstract:Sixty‐six patients with locally advanced (Stages III and IV) carcinoma of the head and neck were treated with three cycles of induction chemotherapy, consisting of cisplatin, fluorouracil (FU) infusion, bleomycin, mitomycin, and hydroxyurea, followed by radiotherapy and/or surgery. There were 48 men and 18 women with a median age of 55 years (range, 18 to 75 years) and Karnofsky performance status of 80 (range, 40 to 90). Primary site was nasopharynx (28 patients), followed by larynx (12) and others (26). Forty‐one (62%) patients were presented with Stage IV disease. The response rate to induction chemotherapy was 27% complete response, 50% partial response, 20% stable disease, and 3% progressive disease. There was no significant difference in response rate between patients with cancer of nasopharynx or other sites (P> 0.1). Survival was 61% at 24 months. Patients with cancer of nasopharynx had a better survival than those with other primaries (P = 0.033). Toxicities from chemotherapy included alopecia (73%), nausea/vomiting (66%), leukopenia (54%), stomatitis (36%), anemia (32%), thrombocytopenia (16%), and diarrhea (9%). Grade IV toxicity was not observed. Induction chemotherapy with this new regimen resulted in a high response rate but may not be superior to cisplatin and FU alone. It can be safely combined with radiotherapy as a potentially curative therapy in squamous cell carcinoma of the head and neck. Chemotherapy followed by radiation therapy may yield survival similar to radical surgery in laryngeal and other head and neck cancers.

Abstract:SIGNIFICANCE: Photoacoustic (PA) tomography has demonstrated versatile biomedical applications. However, an array-based PA computed tomography (PACT) system is complex and expensive, whereas a single-element detector-based scanning PA system is too slow to detect some fast biological dynamics in vivo. New PA imaging methods are sought after.AIM: To overcome these limitations, we developed photoacoustic topography through an ergodic relay (PATER), a novel high-speed imaging system with a single-element detector.APPROACH: PATER images widefield PA signals encoded by the acoustic ergodic relay with a single-laser shot.RESULTS: We applied PATER in vivo to monitor changes in oxygen saturation in a mouse brain and also to demonstrate high-speed matching of vascular patterns for biometric authentication.CONCLUSIONS: PATER has achieved a high-speed temporal resolution over a large field of view. Our results suggest that PATER is a promising and economical alternative to PACT for fast imaging.

Disposable Opto-Acoustic Window Enabled Cost-effective Photoacoustic-Ultrasound Dual-modal Imaging

Abstract:Photoacoustic imaging (PAI) and ultrasound imaging (USI) are important biomedical imaging techniques, due to their unique and complementary advantages in tissue's structure and function visualization. In this Letter, we proposed a coaxial photoacoustic-ultrasound dual-modal imaging system (coPAUS) with disposable opto-acoustic window. This opto-acoustic window allows part of light to go through it, and another part of light to be converted to ultrasound transmission signal by photoacoustic effect. By single laser pulse illumination, both PA signals and reflected US signals can be generated. Then, a linear array probe receives both PA and US signals, enabling simultaneous dual-modal PA and US imaging. Ex vivo experiments were conducted involving pencil lead, hair, and plastic tube with black spot, as well as in vivo experiment on human finger. The system's resolutions for PA and US imaging are 215 um and 91.125 um, with signal-to-noise ratios for PA and US signals reached up to 37.48 dB and 29.75 dB, respectively, proving the feasibility of the coPAUS dual-modal imaging. The proposed coPAUS system with disposable opto-acoustic window provides an immediate and cost-effective approach to enable US imaging capability based on an existing PA imaging system.

Abstract:SignificanceThe photoacoustic (PA) technique, a noninvasive pump–probe technique, has found interesting applications in biomedical tissue diagnosis over the last decade. To take it a step further to clinical applications, the PA technique needs to be designed as an instrument focusing on a compact design, reducing the cost, and quickly providing a quantitative diagnosis.AimThis work presents a design and characterization of a cost-effective, compact PA sensing instrument for biomedical tissue diagnosis.ApproachA compact laser diode case design is developed to house several laser diodes for PA excitation, and a pulsed current supply unit is also developed in-house to power the laser diodes to generate a 25 ns current pulse at a frequency of 20 kHz. After PA experimental data acquisition, the signal's frequency spectra were calculated to characterize the tissue quantitatively and correlated with their mechanobiological properties.ResultsThe corresponding dominant frequency peak in the PA spectral response (PASR) study was low in the fibrofatty normal breast tissue 0.26±0.03 MHz, compared to the dominant frequency peak of 1.60±0.016 MHz in the fibrocystic disease tissue, which had increased glandular and stromal elements, thereby increased tissue density. The histopathological findings correlated with the PASR results, and the fibrocystic breast disease tissue exhibited a higher dominant frequency peak and energy compared to the normal breast tissue.ConclusionsWe experimented with an in vitro PASR study of fibrocystic human breast tissues and successfully differentiated different tissue types using quantitative spectral parameters peak frequency, mean frequency, and spectral energy. This gives the potential to take this technique further for cost-effective and quick clinical applications.

Abstract:D-type cyclin-dependent kinases (Cdk4 and Cdk6) regulate the G1 to S phase progression of the mammalian cell cycle. It has been suggested that Cdk4 and Cdk6 may have distinct functions in vivo, even though they are indistinguishable biochemically. Here we show that although these Cdks phosphorylate multiple residues in pRB, they do so with different residue selectivities in vitro; Thr821 and Thr826 are preferentially phosphorylated by Cdk6 and Cdk4, respectively. This raises the possibility different substrate specificities lead to their different roles in the regulation of cellular events. Furthermore, our results indicate the new concept that Cdk itself contributes to substrate recognition.

Abstract:Photoacoustic imaging (PAI)is a state of art biomedicine imaging technique in the 21 st centu-ry,for it inherits the high resolution of ultrasonography in deep tissue and the ability of optical imaging in biochemical information detection simultaneously.The recent progresses of PAI in biomedicine are re-viewed.The basic principles and two major implementations of PAI,photoacoustic tomography and pho-toacoustic microscopy are introduced.Then the capability of multi-wavelength PAI in evaluating chemical components in tissues,and the feasibility of PA spectral analyses in evaluating histological microstruc-tures in biological tissue are demonstrated,at the same time,several analysis methods and clinical appli-cations of PAI in biomedical imaging are discussed.Finally,the advantages and potential applications of PAI in biology and medicine are sunmarized.

Abstract:Photoacoustic tomography (PAT) of biological tissue offers potential advantages in distinguishing different structures according to their chemical composition. However, as medical photoacoustic (PA) signals are broad band, which usually cover a 0.2-50 MHz range, current band-limited ultrasound transducers can only receive and present a limited fraction of that range. Besides, the received PA signals are usually of low signal-to-noise ratio (SNR), which further deteriorates the image quality. Therefore, the goal of this work is to recover the out-of-band frequency components in the received PA signals based on a best estimate of the frequency response of the ultrasound transducers, and to improve the PA image quality. Hereby, we came up with a method including signal recovery based on inverse filtering and multi-sampled signal de-noising, with which we were capable of rebuilding wider band PA signals and reconstructing PA images with higher SNR. In this way, detailed information that used to be buried in the background noise was revealed, and the boundaries of the imaging object were enhanced. Computer simulations as well as phantom and in vivo experiments were carried out and reconstructed images were given for illustration. In the future, our work can be applied to clinical and preclinical biomedical imaging fields, such as multi-spectrum PA imaging, angiography and other related fields.
Advances in Photoacoustic Imaging Reconstruction and Quantitative Analysis for Biomedical Applications

Abstract:Photoacoustic imaging (PAI) represents an innovative biomedical imaging modality that harnesses the advantages of optical resolution and acoustic penetration depth while ensuring enhanced safety. Despite its promising potential across a diverse array of preclinical and clinical applications, the clinical implementation of PAI faces significant challenges, including the trade-off between penetration depth and spatial resolution, as well as the demand for faster imaging speeds. This paper explores the fundamental principles underlying PAI, with a particular emphasis on three primary implementations: photoacoustic computed tomography (PACT), photoacoustic microscopy (PAM), and photoacoustic endoscopy (PAE). We undertake a critical assessment of their respective strengths and practical limitations. Furthermore, recent developments in utilizing conventional or deep learning (DL) methodologies for image reconstruction and artefact mitigation across PACT, PAM, and PAE are outlined, demonstrating considerable potential to enhance image quality and accelerate imaging processes. Furthermore, this paper examines the recent developments in quantitative analysis within PAI, including the quantification of haemoglobin concentration, oxygen saturation, and other physiological parameters within tissues. Finally, our discussion encompasses current trends and future directions in PAI research while emphasizing the transformative impact of deep learning on advancing PAI.

Abstract:We established a photoacoustic imaging (PAI) system that can provide variable gain at different depths. The PAI system consists of a pulsed laser with an optical parametric oscillator working at a 728[Formula: see text]nm wavelength and an imaging-acquisition-and-processing unit with an ultrasound transducer. A voltage-controlled attenuator was used to realize variable gain at different depths when acquiring PAI signals. The proof-of-concept imaging results for variable gain at different depths were achieved using specific phantoms. Both resolution and optical contrast obtained through the results of variable gain for a targeted depth range are better than those of constant gain for all depths. To further testify the function, we imaged the sagittal section of the body of in vivo nude mice. In addition, we imaged an absorption sample embedded in a chicken breast tissue, reaching a maximum imaging depth of [Formula: see text]4.6[Formula: see text]cm. The results obtained using the proposed method showed better resolution and contrast than when using 50[Formula: see text]dB gain for all depths. The depth range resolution was [Formula: see text]1[Formula: see text]mm, and the maximum imaging depth of our system reached [Formula: see text]4.6[Formula: see text]cm. Furthermore, blood vessels can be revealed and targeted depth range can be selected in nude mice imaging.

Abstract:OBJECTIVE: Photoacoustic (PA) imaging emerges as a unique tool to study biological samples based on optical absorption contrast. In PA imaging, piezoelectric transducers are commonly used to detect laser-induced ultrasonic waves. However, they typically lack adequate broadband sensitivity at ultrasonic frequency higher than 100 MHz, whereas their bulky size and optically opaque nature cause technical difficulties in integrating PA imaging with conventional optical imaging modalities. To overcome these limitations, optical methods of ultrasound detection were developed and shown their unique applications in PA imaging.METHODS: We provide an overview of recent technological advances in optical methods of ultrasound detection and their applications in PA imaging. A general theoretical framework describing sensitivity, bandwidth, and angular responses of optical ultrasound detection is also introduced.RESULTS: Optical methods of ultrasound detection can provide improved detection angle and sensitivity over significantly extended bandwidth. In addition, its versatile variants also offer additional advantages, such as device miniaturization, optical transparency, mechanical flexibility, minimal electrical/mechanical crosstalk, and potential noncontact PA imaging.CONCLUSION: The optical ultrasound detection methods discussed in this review and their future evolution may play an important role in PA imaging for biomedical study and clinical diagnosis.

Abstract:Photoacoustic (PA) imaging is a rapidly emerging biomedical imaging modality that is capable of visualizing cellular and molecular functions with high detection sensitivity and spatial resolution in deep tissue. Great efforts and progress have been made on the development of various PA imaging technologies with improved resolution and sensitivity over the past two decades. Various PA probes with high contrast have also been extensively developed, with many important biomedical applications. In comparison with chemical dyes and nanoparticles, genetically encoded probes offer easier labeling of defined cells within tissues or proteins of interest within a cell, have higher stability in vivo, and eliminate the need for delivery of exogenous substances. Genetically encoded probes have thus attracted increasing attention from researchers in engineering and biomedicine. In this review, we aim to provide an overview of the existing PA imaging technologies and genetically encoded PA probes, and describe further improvements in PA imaging techniques and the near-infrared photochromic protein BphP1, the most sensitive genetically encoded probe thus far, as well as the potential biomedical applications of BphP1-based PA imaging in vivo.
