Abstract:Potential biomedical applications such as controlled delivery with sustained drug release profile demand for multifunctional polymeric particles of precise chemical composition and with welldefined physicochemical properties. The real challenge is to obtain the reproducible and homogeneous nanoparticles in a minimum number of preparation steps. Here, single-step nanoarchitectures of soft surface layered copolymer nanoparticles with a regular tuning in the size via micro flow-through assisted synthesis are reported. Interfacial copolymerization induces the controlled compartmentalization where a hydrophobic core adopts spherical shape in order to minimize the surface energy and simultaneously shelter in the hydrophilic shelllike surface layer. Surface layer can swell in the aqueous medium and allow controlled entrapping of functional hydrophobic nanoparticles in the hydrophilic interior via electrostatic interaction which can be particularly interesting for combined fluorescence activity. Furthermore, the nanoarchitecture of size and concentration controlled polymer-metal nanoassembly particles can be implemented as an ideal surface-enhanced Raman scattering substrate for detection of the trace amounts of various analytes.

Abstract:Microfluidic devices have recently emerged as promising tools for the synthesis of polymer particles. Over conventional processes, microfluidic‐assisted processes allow the production of polymer particles with an improved control over their sizes, size distributions, morphologies, and compositions. In this paper, the most common microfluidic devices are reviewed. Both projection photolithography and emulsification processes are reported for the continuous flow synthesis of polymer particles from a stream of polymerizable liquids. For the latter process, two distinct categories of microfluidic devices have been identified: microchannel‐based and capillary‐based microsystems. For each category, the existing geometries are described and the different emulsification methods including the co‐flowing, cross‐flowing, or flow‐focusing of the continuous and dispersed phases are commented upon. Finally, for each microsystem the various polymer particles achieved in such devices including, but not restricted to, janus, core–shell, or porous particles and capsules are reported.

Abstract:In this work, we report an approach to prepare segmented polymer nanofibers (SPNFs) composed of rodlike subunits by kinetically controlled self-assembly of polystyrene-b-poly(4-vinylpyridine)-based supramolecules in microfluidic chips. The length and morphology of the SPNFs could be effectively adjusted by changing the total flow rate (Vtotal) and the molar ratio (x) of 4-vinylpyridine (4VP) unit to a hydrogen-bonding molecule, 3-n-pentadecyphenol. Moreover, the subunits of SPNFs could transform from short rods to spheres when the interfacial tension between PS core and solvent increased. On the contrary, the SPNFs elongated along the major axis when the interfacial tension decreased. Abstract:Conventional batch syntheses of polymer-based nanoparticles show considerable shortcomings in terms of scarce control over nanomaterials morphology and limited lot-to-lot reproducibility. Droplet-based microfluidics represents a valuable strategy to overcome these constraints, exploiting the formation of nanoparticles within discrete microdroplets. In this work, we synthesized nanogels (NGs) composed of hyaluronic acid and polyethyleneimine using a microfluidic flow-focusing device endowed with a pressure-driven micro-actuator. The actuator achieves real-time modulation of the junction orifice width, thereby regulating the microdroplet diameter and, as a result, the NG size. Acting on process parameters, NG hydrodynamic diameter could be tuned in the range 92–190 nm while preserving an extremely low polydispersity (0.015); those values are hardly achievable in batch syntheses and underline the strength of our toolbox for the continuous in-flow synthesis of nanocarriers. Furthermore, NGs were validated in vitro as a drug delivery system in a representative case study still lacking an effective therapeutic treatment: ovarian cancer. Using doxorubicin as a chemotherapeutic agent, we show that NG-mediated release of the drug results in an enhanced antiblastic effect vs . the non-encapsulated administration route even at sublethal dosages, highlighting the wide applicability of our microfluidics-enabled nanomaterials in healthcare scenarios.

Abstract:This report describes a straightforward but robust tubing method for connecting polydimethylsiloxane (PDMS) microfluidic devices to external equipment. The interconnection is irreversible and can sustain a pressure of up to 4.5 MPa that is characterized experimentally and theoretically. To demonstrate applications of this high-pressure tubing technique, we fabricate a semicircular microfluidic channel to implement a high-throughput, size-controlled synthesis of poly(lactic-co-glycolic acid) (PLGA) nanoparticles ranging from 55 to 135 nm in diameter. This microfluidic device allows for a total flow rate of 410 mL h(-1), resulting in enhanced convective mixing which can be utilized to precipitate small size nanoparticles with a good dispersion. We expect that this tubing technique would be widely used in microfluidic chips for nanoparticle synthesis, cell manipulation, and potentially nanofluidic applications.

Abstract:We report one-step microfluidic synthesis and characterization of novel Janus microhydrogels composed entirely of the same base material, N-isopropylacrylamide (NIPAAm). The microhydrogels were fabricated by the microfluidic generation of Janus monomer microdroplets based on separation of a supersaturated aqueous NIPAAm solution into NIPAAm-rich and -poor phases followed by UV irradiation. The resulting Janus microhydrogels exhibited tunable anisotropic thermo-responsive behavior and organophilic/hydrophilic loading capability.

Abstract:In this work, we propose a novel method to fabricate polymer particles with controlled morphology in microchannel by solidification of polymer solutions and study the transition in the shape of these particles from spheres, to cups, and to donuts. The non-spherical geometries are achieved through nonuniform diffusion of the solvent, leading to non-uniform solidification. By adjusting the flow rates of both continuous and dispersed phases, the spatial heterogeneity in the solvent diffusion rate and thus the morphology of the final polymer particles can be tuned. This approach offers a one-step continuous process for the fabrication of non-spherical polymer particles. The technique is applicable to different polymers under appropriate polymer-solvent combinations. The ability to tune the shapes of polymer particles easily will create new opportunities for applications that require anisotropic functional particles, such as in biomedical engineering.

Abstract:Hydrophilic polymeric nanoparticles are synthesized through a Bergman cyclization- mediated intramolecular chain collapse of structurally well-defined linear polymers, and then used as size-tunable nanoreactors to fabricate and encapsulate quantum dots in a one-pot reaction. Crystalline quantum dots are formed in all of these nanoreactors and visualized by transmission electron microscopy. Smaller nanoreactors produce one quantum dot each while larger nanoreactors form a number, resulting in fluorescence quenching. By controlling the molecular weight of the linear polymer precursor, a variable number of nanocrystals are fabricated and assembled in a single nanoreactor.

Abstract:This work presents a one-step synthesis of monodisperse nanoflowers by aqueous polymerization-induced self-assembly (PISA), modulated by host-guest interactions. Owing to the low monomer swelling of nanoparticles restricted by host-guest complexation, hierarchical surficial micellar structures were generated at the outer surface of the vesicles, forming fractal nanoflowers with a diameter polydispersity as low as 1.01. Our method allows the straightforward synthesis of monodisperse hierarchical nanoparticles for a wide range of applications.

Abstract:We report the photo-induced, additive-free, continuous synthesis of polymeric particles using flow chemistry. Not only can these particles be formed under ambient conditions in a solely light induced precipitation polymerisation, they can be prepared via continuous flow techniques to up-scale the synthetic process. We carefully assess the flow chemical parameters and analyse the resulting particles quantitatively using Scanning Electron Microscopy (SEM). Particle formation is a direct result of the step growth polymerisation via a photochemically induced AA + BB Diels-Alder reaction, which we herein base on the dialdehyde monomer (AA) derived from the sustainable precursor, thymol. By employing a peroxyoxalate bismaleimide (BB), we introduce particles that can be selectively degraded on-demand, self-reported by light emission through chemiluminescence.

Abstract:This article provides an overview of our work in the area of the synthesis of poly in er particles in continuous microfluidic reactors. The method includes (a) the generation of highly monodisperse monomer droplets in a microfluidic flow-focusing device and (b) in-situ solidification of these droplets by means of photopolymerization. We discuss the effect of monomer properties on the emulsification process, the effect of the polymerization rate on the production of high-quality particles, the role of the material of the microfluidic device in droplet formation, and the synthesis of particles with different shapes and composition.,;, We also demonstrate the production of highly ordered arrays of polymer particles achieved by photopolymerization of the dynamic lattices of monomer droplets in microfluidic channels, The article is concluded with a summary of future research directions in the production of polymer colloids in microfluidic reactors.

Abstract:Macroporous copolymer particles have a broad range of applications such as ion exchange resins and sorbents, catalyst supports, and carriers of biologically active species. Many of these applications require precise control of the dimensions of microbeads in the range from 50 to 100 mu m and predetermined size of pores. IN This paper reports semicontinuous photoinitiated microfluidic synthesis of macroporous polymer particles with the designated dimensions and a range of internal structures. Comparison with microspheres synthesized by conventional suspension polymerization shows,that microfluidic synthesis provides better control over the porous structure of the microbeads.

Abstract:A microfluidic platform to synthesize monodispersed and spherical hydrogel beads (microspheres) of poly(acrylamide-co-acrylic acid) (p(AM-co-AA)) is reported. An aqueous phase consisting of monomer, cross-linker, functional moiety and initiator, all dissolved in water, is dispersed as uniform drops of desired size into a carrier phase (dodecane) at an opposed T-junction. The chemical and physical properties of the microspheres are systematically studied. Effects of operating parameters like velocity and O/A (oil to aqueous phase flow ratio) ratio on the average size, polydispersity and shape of microspheres are investigated. Microspheres within a size range of 960 to 502 microns can be synthesized. A comparison of two different mixing techniques namely in-situ and ex-situ mixing is also presented. A two dimensional computational fluid dynamic (2 D CFD) model is also developed to quantify and validate drop formation phenomena at the T junction at extreme O/A ratios. Abstract:Fluidic channels were employed to induce the self-assembly of poly(ethylene glycol)-b-polystyrene into polymeric vesicles and nanotubes. The laminar flow in the device enables controlled diffusion of two miscible liquids at the phase boundary, leading to the formation of homogeneous polymeric structures of different shapes. These structures could be easily loaded with small molecule cargoes and functionalized with nanometer sized catalytic platinum nanoparticles. This technique offers a facile methodology to rapidly and continuously produce well-defined polymeric structures for nanotechnology applications.

Abstract:We present a process for fabricating nanostructured surfaces with feature sizes down to at least 50nm and aspect ratios of 1:1 by injection molding. We explored the effects of mold coatings and injection molding conditions on the final nanostructure quality. A plasma-polymerized fluorocarbon based antistiction coating was found to improve the replication fidelity (shape and depth) of nanoscale features substantially. Arrays of holes of 50nm diameter/35nm depth and 100nm/100nm diameter, respectively, were mass-produced in cyclic olefin copolymer (Topas 5013) by injection molding. Polymer microfluidic channel chip parts resulted from a separate injection molding process. The microfluidic chip part and the nanostructured chip part were successfully bonded to form a sealed microfluidic system using air plasma assisted thermal bonding.

Abstract:Self-assembly of block copolymers (BCPs) in microfluidic chips is a versatile yet effective route to produce micellar aggregates with various controllable sizes and morphologies. In this study, the morphological transformation of the BCP of polystyrene-b-poly(4-vinylpyridine) (PS-b-P4VP) assemblies from irregular aggregates to multicompartment micelles and ultimately to ordered spherical micelles is demonstrated in microfluidic chips. Our experimental and computational simulation results indicate that the transverse diffusion of solvents plays an important role in the morphological transformation of PS-b-P4VP assemblies in the confined flow condition. We find that the mixing time (tmix) between a BCP/tetrahydrofuran (THF) solution and water affects the morphological transformation. Micellar morphologies are intended to transform from aggregates to ordered spherical structures under a relatively long mixing time (tmix). In addition, it is observed that the size of the micelles decreases with the increase of the flow velocity ratio by tuning the hydrodynamic conditions of the flows. Moreover, by adjusting the initial polymer solution concentration, temperature, and weight fraction of the introduced homopolystyrene (hPS), which can affect the viscosity of the BCP solution, the flow diffusion in the microfluidic chip and the resulted micellar structures can also be readily adjusted. The current study provides a new flow-driven method to adjust the micellar ordered structural transformation under the nonequilibrium state.

Abstract:Soft-matter nanoparticles are of great interest for their applications in biotechnology, therapeutic delivery, and in vivo imaging. Underpinning this is their biocompatibility, potential for selective targeting, attractive pharmacokinetic properties, and amenability to downstream functionalisation. Morphological diversity inherent to soft-matter particles can give rise to enhanced functionality. However, this diversity remains untapped in clinical and industrial settings, and only the simplest of particle architectures [spherical lipid vesicles and lipid/polymer nanoparticles (LNPs)] have been routinely exploited. This is partially due to a lack of appropriate methods for their synthesis. To address this, we have designed a scalable microfluidic hydrodynamic focusing (MHF) technology for the controllable, rapid, and continuous production of lyotropic liquid crystalline (LLC) nanoparticles (both cubosomes and hexosomes), colloidal dispersions of higher-order lipid assemblies with intricate internal structures of 3-D and 2-D symmetry. These particles have been proposed as the next generation of soft-matter nano-carriers, with unique fusogenic and physical properties. Crucially, unlike alternative approaches, our microfluidic method gives control over LLC size, a feature we go on to exploit in a fusogenic study with model cell membranes, where a dependency of fusion on particle diameter is evident. We believe our platform has the potential to serve as a tool for future studies involving non-lamellar soft nanoparticles, and anticipate it allowing for the rapid prototyping of LLC particles of diverse functionality, paving the way toward their eventual wide uptake at an industrial level.

Abstract:In this study, we demonstrate the fabrication of polymersomes, protein-blended polymersomes, and polymeric microcapsules using droplet microfluidics. Polymersomes with uniform, single bilayers and controlled diameters are assembled from water-in-oil-in-water double-emulsion droplets. This technique relies on adjusting the interfacial energies of the droplet to completely separate the polymer-stabilized inner core from the oil shell. Protein-blended polymersomes are prepared by dissolving protein in the inner and outer phases of polymer-stabilized droplets. Cell-sized polymeric microcapsules are assembled by size reduction in the inner core through osmosis followed by evaporation of the middle phase. All methods are developed and validated using the same glass-capillary microfluidic apparatus. This integrative approach not only demonstrates the versatility of our setup, but also holds significant promise for standardizing and customizing the production of polymer-based artificial cells.
