Abstract:We demonstrate numerical refocusing in coherent confocal laser scanning microscopy based on synthetic optical holography. This physics-based approach implements a computational propagation on the complex signal recovered in synthetic holography consistent with the wave physics and the parameters of the microscope. An experimental demonstration is shown to restore an in-focus image of a test object from data acquired at several focal plane off-sets. Numerical refocusing can provide focused views on samples with large height variation with a potential application in confocal optical surface profiling.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to achieve numerical refocusing in phase - resolved confocal microscopy through synthetic optical holography (SOH). Specifically, the researchers aim to recover defocused images by numerical methods, enabling the confocal microscope to provide clear views at different focal planes and to perform accurate surface profile measurements on samples with large height variations. ### Analysis of the Core Problems in the Paper 1. **Background and Motivation** - Confocal microscopy is widely used in fluorescence and Raman imaging because of its high contrast and depth - resolving ability. - Traditional wide - field microscopy techniques cannot be directly applied to confocal systems because the defocus correction factors resulting from the confocal geometry have different forms. - Synthetic optical holography (SOH) is a new scanning quantitative phase - imaging mode that combines the advantages of technical simplicity and high - speed imaging. 2. **Specific Problems** - **Defocused Image Recovery**: How to recover clear images from defocused data by numerical methods. - **Utilization of Phase Information**: How to use phase information for numerical refocusing to obtain high - resolution images. - **Application Expansion**: How to apply this technique to confocal optical surface profile measurement and other scenarios that require accurate depth information. 3. **Solutions** - The researchers derived a forward model that describes the imaging process of the confocal microscope in the back - scattering geometry. - A back - propagation operator was found through pseudo - inverse operation to reconstruct the original object from the defocused scattering field. - An analytical solution was proposed, which can perform the refocusing operation in the Fourier domain. - The amplitude - and phase - resolved images of the United States Air Force test sample recorded using SOH were experimentally verified and compared with non - interferometric in - focus images. ### Representation of Mathematical Formulas The key formulas involved in the paper are as follows: - **Gaussian Beam Shape in Wave - number Space**: \[ \tilde{g}(k)=e^{-\alpha^{2}|k|^{2}/2k^{2}} \] where \( k \) is the transverse Fourier variable, \( k \) is the wave number, and \( \alpha \) controls the beam width. - **Imaging Operator \( P \)**: \[ P = \int dz d^{2}k' d^{2}k |k\rangle U_0 \tilde{g}(k') \tilde{g}(k' - k) e^{i(z - z_1)(k_z(k')+k_z(k' - k))}\frac{1}{k_z(k')}\langle k|\langle z| \] where \( k_z(k)=\sqrt{k^{2}-|k|^{2}} \). - **Imaging Operator \( P_1 \) under Far - Field Approximation**: \[ P_1\approx\int d^{2}k dz |k\rangle H_F\frac{1}{z - z_1}e^{i2k_z(k/2)(z - z_1)}\langle k|\langle z| \] - **Pseudo - Inverse Imaging Operator \( P_1^+ \)**: \[ P_1^+\approx\int d^{2}k dz |k\rangle |z\rangle\frac{1}{z - z_1}H_F e^{-i2k_z(k/2)(z - z_1)}\langle k| \] - **Near - Focus Imaging Operator \( P_2 \)**: \[ P_2\approx\int d^{2}k dz |k\rangle H_N\frac{1}{k_z(k/2)}e^{i2k_z(k/2)(z - z_2)}\langle k|\langle z| \] - **Refocusing Operator \( R_{12} \)**: \[ R_{12}=P_2P_1^+
