Abstract:We proposed a new flow-focusing technique for generation of monodisperse femtoliter droplets, based on the capillary micro-cross. A funnel-shaped interface of two phase system is observed in a capillary cross for mass production of uniform drops, where a tapered exit orifice is extruded into the dispersed feeding capillary. The droplets, down to 2 μm in size at frequency of 20 kHz, are controllable in size when choosing orifice and capillary sizes, as well as flow rates of inner and outer fluids. For a specific diameter of exit orifice, there is a maximal flow rate of outer fluid, beyond which the interface will be penetrated. Until then, the interface is in steady state and all droplets are highly uniform (<3%), implicating an absolute instability in the whole process.

Abstract:Abstract Single-cell encapsulation in droplet microfluidics is commonly hindered by the tradeoff between cell suspension density and on-chip focusing performance. In this study, we introduce a novel droplet microfluidic chip to overcome this challenge. The chip comprises a double spiral focusing unit, a flow resistance-based sample enrichment module with fine-tunable outlets, and a crossflow droplet generation unit. Utilizing a low-density cell/bead suspension (2 × 10 6 objects/mL), cells/beads are focused into a near-equidistant linear arrangement within the double spiral microchannel. The excess water phase is diverted while cells/beads remain focused and sequentially encapsulated in individual droplets. Focusing performance was assessed through numerical simulations and experiments at three flow rates (40, 60, 80 μL/min), demonstrating successful focusing at 40 and 80 μL/min for beads and cells, respectively. In addition, both simulation and experimental results revealed that the flow resistance at the sample enrichment module is adjustable by punching different outlets, allowing over 50% of the aqueous phase to be removed. YOLOv8n-based droplet detection algorithms realized the counting of cells/beads in droplets, statistically demonstrating single-cell and bead encapsulation rates of 72.2% and 79.2%, respectively. All the results indicate that this on-chip sample enrichment approach can be further developed and employed as a critical component in single-cell encapsulation in water-in-oil droplets.

Abstract:Droplet microfluidic techniques have been rapidly developed as a powerful tool to perform high-throughput and low-cost analysis of single cells. Microscale droplets can be easily produced by a microfluidic manipulation to encapsulate and manipulate single cells for precise analysis. This offers a new approach to measure genetic and functional heterogeneity of cell division, growth, metabolism, and apoptosis. Functional characteristics of cellular molecules, such as DNA, RNA, and proteins, can be realized at a single-cell level. In this chapter, we will present a general introduction to single-cell analysis involving droplet-based microfluidic techniques. We will highlight the current state of droplet-based microfluidic single-cell analysis for deep insights understanding the biological process at the single-cell level.

Abstract:Single-cell analysis to investigate cellular heterogeneity and cell-to-cell interactions is a crucial compartment to answer key questions in important biological mechanisms. Droplet-based microfluidics appears to be the ideal platform for such a purpose because the compartmentalization of single cells into microdroplets offers unique advantages of enhancing assay sensitivity, protecting cells against external stresses, allowing versatile and precise manipulations over tested samples, and providing a stable microenvironment for long-term cell proliferation and observation. The present Review aims to give a preliminary guidance for researchers from different backgrounds to explore the field of single-cell encapsulation and analysis. A comprehensive and introductory overview of the droplet formation mechanism, fabrication methods of microchips, and a myriad of passive and active encapsulation techniques to enhance single-cell encapsulation efficiency were presented. Meanwhile, common methods for single-cell analysis, especially for long-term cell proliferation, differentiation, and observation inside microcapsules, are briefly introduced. Finally, the major challenges faced in the field are illustrated, and potential prospects for future work are discussed.

Abstract:Water-in-oil droplet generation for object encapsulation is of increasing importance in biochemical reaction, drug diagnostics, and environmental science. Optofluidic technique is a powerful tool for manipulating mesoscopic particles and droplets within a continuous liquid phase. Here, we report a flexible and miniaturized droplet generator based on a microvortex chip by light-heat energy conversion to fuse, trap, and release picoliter aqueous droplets in the immiscible carrier oil. The droplet sizes in the generators can be tuned by the flow rate of the vortices. Further, it is demonstrated that the generated droplet can realize size-selective encapsulation of micro-matters with tunable rotating trajectories. The results showed that the microvortex-based droplet generator propose a method for the stable fusion and encapsulation of droplet with controllable positions and volumes. This system will provide a controllable microfluidic environment for droplet tweezers, micro-reactors, micro-mixers, and object encapsulation in practical applications.

Abstract:Microfluidic droplet-based technology enables encapsulation of cells in the isolated aqueous chambers surrounded by immiscible fluid but single-cell encapsulation efficiency is usually less than 30%. In this letter, we introduce a simple microgroove structure to break droplets into random sizes which further allows collecting of single-cell [Escherichia coli (E. coli)] containing droplets by their size differences. Pinched-flow separation method is integrated to sort out droplets of certain sizes which have high probability of containing one cell. Consequently, we were able to obtain more than 50% of droplets having single E. coli inside, keeping the proportion of multiple-cell containing droplets less than 16%.

Abstract:We conducted experiments on specially designed microfluidic chips that generate droplets through a microfluidic flow-focusing approach. The fluid flow in the microfluidic channel produced a shear flow field at low Reynolds numbers. The droplets in the microfluidic system exhibited special droplet pattern formations similar to periodic crystal-like lattices because of the competition between shear forces and surface tension. By adjusting the flow rate ratio of the water (droplet phase) to oil (continuous phase) phases and changing the outlet channel widths, the droplets formed monolayer dispersion to double-layer formation to monolayer squeezing when the outlet channel widths were 250 or 300 mu m. We also obtained droplets with monolayer dispersion, three-layer arrangements, double-layer squeezing, and monolayer squeezing when the outlet channel width was 350 mu m. The outlet channel width was increased to 400 mu m, and four-layer arrangements were observed. We also studied the translation of droplet formation, which resulted in a detailed strategy to control drop size and droplet pattern formation for emulsification in microfluidic devices. We expect that our strategy can provide theoretical guidance to synthesize dispersion or polydisperse colloid particles.

Abstract:Cells are the most basic structural and functional units of living organisms. Studies of cell growth, differentiation, apoptosis, and cell-cell interactions can help scientists understand the mysteries of living systems. However, there is considerable heterogeneity among cells. Great differences between individuals can be found even within the same cell cluster. Cell heterogeneity can only be clearly expressed and distinguished at the level of single cells. The development of droplet microfluidics technology opens up a new chapter for single-cell analysis. Microfluidic chips can produce many nanoscale monodisperse droplets, which can be used as small isolated micro-laboratories for various high-throughput, precise single-cell analyses. Moreover, gel droplets with good biocompatibility can be used in single-cell cultures and coupled with biomolecules for various downstream analyses of cellular metabolites. The droplets are also maneuverable; through physical and chemical forces, droplets can be divided, fused, and sorted to realize single-cell screening and other related studies. This review describes the channel design, droplet generation, and control technology of droplet microfluidics and gives a detailed overview of the application of droplet microfluidics in single-cell culture, single-cell screening, single-cell detection, and other aspects. Moreover, we provide a recent review of the application of droplet microfluidics in tumor single-cell immunoassays, describe in detail the advantages of microfluidics in tumor research, and predict the development of droplet microfluidics at the single-cell level.

Abstract:Negative pressures were used to generate droplets for single-cell encapsulations.Iodixanol was used to change the suspension solution densities in order to reduce the issue of cellular sedimentations.The effects of negative pressures and geometries of microfluidic T-channels on volumes and frequencies of generated droplets and the effects of iodixanol on the percentages of single-cell encapsulations were investigated and compared.This microfluidic platform may enable stable single-cell encapsulations,paving ways for single-cell analysis.

Abstract:A facile method is presented for achieving comprehensive two-dimensional droplet manipulations in closed microstructures consisting of microwell arrays and a straight microchannel. In this method, picoliter/nanoliter droplets with controllable sizes and numbers are sampled from nanoliter samples/reagents with almost 100% efficiency. Droplet motions are precisely controlled in the ±X-direction and ±Y-direction by managing hydrostatic pressure and magnetic repulsion, respectively. As a demonstration, a fluorescein-labeled droplet and a deionized droplet are successively generated and trapped in adjacent microwells. Then their positions are quickly exchanged without cross-contamination and fusion is implemented on-demand. After operations, hydrophobic ferrofluid can be completely replaced by mineral oil and droplets still remain in microwells safely. A typical fluorescence intensity-based assay is demonstrated: droplet arrays containing copper ion are diluted disproportionately first and then detected by addition of droplet arrays containing Calcein. With the ability of comprehensive two-dimensional droplet manipulations, this method could be used in various miniaturized biochemical analyses including requirements of multistep procedures and in situ monitoring.

Abstract:Droplet microfluidics has become an indispensable technology to encapsulate cells of interest in a monodispersed aqueous compartment for single-cell analysis. In addition, the confinement of cells in picoliter droplets offers high-throughput, single-cell resolution, and high signal-to-noise ratio for various cellular assays that unmasks cellular heterogeneity from a bulk population. Particularly, co-encapsulation of two distinct cells in a droplet is critically important for studying cell–cell interaction, transcriptomics, genomics, antibody, and drug screening. However, the co-encapsulation of one type A cell and one type B cell per single droplet, termed 1–1–1 encapsulation, has been dictated by double Poisson distribution due to the intrinsic random dispersion of cells, which yields mostly empty droplets and only up to 13.5% of droplets under optimal conditions. Such low 1–1–1 encapsulation efficiency makes it impractical for biological analyses at scale involving low cell concentrations or a large number of variables. Here, we demonstrate a passive co-encapsulation microfluidic device that leverages close packing of cells by hydrodynamic draining to overcome the double Poisson limitation. The results suggest a significant improvement of the 1–1–1 encapsulation efficiency by over two-fold compared to the double Poisson model. The enhanced encapsulation efficiency of this platform demonstrates great potential for a high-throughput, versatile, and simple platform for cell–cell interaction studies within a confined microenvironment.

Abstract:Well-defined microdroplet generation has attracted great interest, which is important for the high-resolution patterning and matrix distribution for chemical reactions and biological assays. By sliding a droplet on a patterned superhydrophilic/superhydrophobic substrate, tiny microdroplet arrays low to femtoliter were achieved with uniform volume and composition. Using this method, cells were successfully isolated, resulting in a single cell array. The droplet-splitting method is facile, sample-effective, and low-cost, which will be of great potential for the development of microdroplet arrays for biological analysis as well as patterning system and devices.

Abstract:Droplets generated through microfluidics serve as a common platform for assembling artificial cells, which are feasibly tailored using microfluidic methodology. The ability of natural cells to undergo shape changes, such as phagocytosis, is a typical characteristic that researchers aim to mimic in artificial cells. However, simulating the deformation behavior of natural cells within droplets is exceptionally challenging. Here, this study reports a pinocytosis-like phenomenon observed in droplets, termed "droplet drinking". When droplets traverse a capillary with constrictions, the shear force from the continuous-phase fluid induces relative motion within the droplets, creating concave regions at the rear. These regions facilitate engulfing of the continuous-phase fluid, resulting in the formation of multiple emulsions. This behavior is influenced by the capillary number, and the size of the ingested droplets is governed by the interfacial tension between the two phases. The production of multicore or multi-shell emulsions can be easily accomplished by making slight adjustments to the constrictions. Furthermore, this method demonstrates the integration of reactants into pre-existing droplets, facilitating biochemical reactions. This study presents a convenient approach for generating complex emulsions and an innovative strategy for studying deformation behavior in droplet-based artificial cells.

Abstract:We developed an automated and multifunctional microfluidic platform based on Drop Lab to perform flexible generation and complex manipulations of picoliter-scale droplets. Multiple manipulations including precise droplet generation, sequential reagent merging, and multistep solid-phase extraction for picoliter-scale droplets could be achieved in the present platform. The system precision in generating picoliter-scale droplets was significantly improved by minimizing the thermo-induced fluctuation of flow rate. A novel droplet fusion technique based on the difference of droplet interfacial tensions was developed without the need of special microchannel networks or external devices. It enabled sequential addition of reagents to droplets on demand for multistep reactions. We also developed an effective picoliter-scale droplet splitting technique with magnetic actuation. The difficulty in phase separation of magnetic beads from picoliter-scale droplets due to the high interfacial tension was overcome using ferromagnetic particles to carry the magnetic beads to pass through the phase interface. With this technique, multistep solid-phase extraction was achieved among picoliter-scale droplets. The present platform had the ability to perform complex multistep manipulations to picoliter-scale droplets, which is particularly required for single cell analysis. Its utility and potentials in single cell analysis were preliminarily demonstrated in achieving high-efficiency single-cell encapsulation, enzyme activity assay at the single cell level, and especially, single cell DNA purification based on solid-phase extraction.

Abstract:In the context of microfluidic technology, investigating the encapsulation of single cells is of great importance, providing valuable insight into cellular behavior and contributing to advancements in single-cell analysis. This paper presents a computational investigation into the dynamics of single-cell encapsulation within a flow-focusing microfluidic system, with a specific emphasis on addressing the challenges associated with high-efficiency encapsulation. This study utilizes a combined lattice Boltzmann and immersed boundary method to provide an accurate simulation of a three-phase system. This allowed for an in-depth exploration of various critical parameters, including cell injection frequency, cell size, and inlet position. This study identifies optimal conditions for maximizing single-cell encapsulation efficiency, emphasizing the impact of the ratio between cell injection and droplet generation frequencies on encapsulation outcomes. This study investigates the effects of cell-induced changes on droplet formation characteristics. It explains the generation of larger droplets and the occurrence of additional satellite droplets. These findings provide insight into the microfluidic platforms designed for single-cell assays, which have potential applications in various fields such as drug development and personalized therapies.

Abstract:Manipulating droplets by light in microscale allows precise control of microfluidics, liquid delivery, micromachines, and so on. Among these applications, microfluidic technology is of particular interest for miniaturization of the portable analysis systems, which require the integration of various liquid operations in one device. Here, a photodeformable microfluidic platform is constructed by combining Laplace pressure and capillary condensation to integrate the transportation, fusion, separation, and mixing of liquid slugs in one chip. The Laplace pressure, attributed to the photodeformation of the liquid crystal polymers, is generated to propel the slug. The capillary condensation is introduced by the delicate design of the fluid channels, allowing the fusion and separation of slugs without any connected microvalves. Catalytic oxidation reaction and protein detection processes are realized in the platform, which are amenable to a variety of miniaturized bio-medical applications, such as portable analysis and point of care testing.

Abstract:Microfluidic devices based on droplet manipulation have been applied in drug screening, chemical manufacturing, DNA library preparation and protein modification. A droplet manipulation system is generally composed of modules such as droplet generation, droplet fusion and droplet split. However, it remains challenging to make these devices commercially and industrially available. Here, an integrated PDMS device was designed for high-throughput generation of droplets, droplet fusion and split on a single chip. Compared with the existing devices, our device needs lower voltage for fusion and the cross-like structure enables dynamic adjustment of droplet split size, which makes this device potentially useful for quantitative analysis. In addition, the potential of this integrated device in multiplex sequencing library preparation is also discussed.
