Abstract:Immunobiosensors were developed for detection of Escherichia coli O157:H7 based on the surface immobilization of monoclone antibodies onto indium tin oxide (ITO) electrodes. The immobilization of antibodies onto ITO chips was carried out by silanization. The effects of epoxysilane monolayer, the antibody layer on the electrochemical properties of the electrode, and the combined target bacteria were analyzed through cyclic voltammetry and electrochemical impedance spectroscopy. By using Randles model as the equivalent circuit, the concentration of the target bacteria can be quantitatively analyzed in terms of the change of electron transfer resistance. The biosensor could detect the target bacteria with a detection limit of 4times103 CFU/mL. A linear response was found between 4times1034times106 CFU/mL. This biosensor was characterized with high sensitivity, excellent selectivity, short detection time and easy operation. It has a promising application in clinical laboratory diagnoses, environmental detection and food safety

Abstract:New biosensors based on electrochemical impedance spectroscopy (EIS) were developed for detection of Escherichia coli O157:H7 through the surface immobilization of monoclonal antibody onto indium tin oxide (ITO) electrode. The biosensor could detect the target bacteria with a detection limit of 4×10 3 CFU/mL. A linear response in the electron-transfer resistance for the concentration of E.coli cells was found between 4×10 3 and 4×10 6CFU/mL. The biosensor characterizes with high sensitivity, simple operation, enabling relatively rapid and reliable analysis. The result proved the prospect of EIS biosensors on application to biomedical and environmental detection of E.coli O157:H7.
EIS biosensor based on a novel Myoviridae bacteriophage SEP37 for rapid and specific detection of Salmonella in food matrixes

Abstract:Recently, using bacteriophages as new molecular probes in reliable platforms for the detection of bacterial pathogens has attracted more and more increasing attentions. In this paper, a novel isolated Myoviridae bacteriophage SEP37 was covalently immobilized onto gold nanoparticles (AuNPs) modified gold disk electrode (GDE) surfaces using cysteamine (Cys) as a crosslinker. Substrates of GDE-AuNPs-Cys-Phage SEP37 and specific capture of Salmonella cells had been characterized using scanning electron microscopy (SEM) separately. Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to study the electrochemical response of the biosensor interface manufacturing and bacterial capture process. Under the optimal experimental conditions, this phage-based EIS biosensor was able to detect Salmonella with a wide linear range from 2 × 101 to 2 × 106 colony forming unit (CFU)/mL within 30 min in spiked lake water and lettuce samples, with a limit of detection (LOD) of 17 CFU/mL. The detection linear range of spiked chicken samples was 2 × 102 to 2 × 105 CFU/mL, with a LOD of 1.3 × 102 CFU/mL. In combination with a pre-enrichment process for 3.5 h, this assay could reach a LOD of 1 CFU/mL in chicken breast meat samples. Besides, this phage-based EIS biosensor provided good reproducibility and stability. This phage-based EIS biosensor opens a new opportunity for the detection of pathogenic bacteria using the inherent selectivity of bacteriophage recognition.
Bacteriophage long tail fibre proteins as a biorecognition element in electrochemical biosensors for the detection of Salmonella in lake water

Abstract:We developed an electrochemical impedance spectroscopy (EIS) biosensor based on bacteriophage long tail fibre proteins (LTFPs) to capture and detect Salmonella . First, based on the genetic information of a Salmonella enterica bacteriophage (SEP37), we found that the genes ORF99, ORF100, and ORF256 encode proteins associated with host recognition. We used the pETdue1 plasmid to construct ORF99–ORF100 and ORF256 co-expression vectors to express (LTFPs) in Escherichia coli and used purified LTFPs as specific biorecognition elements. Then, we fixed gold nanoparticles (AuNPs), cysteamine (Cys), and LTFPs on a gold disk electrode surface via layer-by-layer assembly to construct an EIS biosensor. Salmonella in the sample can be captured by LTFPs on the electrode surface and reduce the electron transfer sites. We applied the EIS biosensor to detect Salmonella spiked in lake water. It showed a good linear relationship within the concentration range of 10 ∼ 1 × 10 6 colony-forming units (CFU)/mL. The detection limit was 9 CFU/mL. This method based on bacteriophage LTFPs does not require complex pretreatment steps and extends the application of tail fibre proteins as a biorecognition element in the detection field.

Abstract:A novel electrochemical biosensor is reported for simultaneous detection of two of the most common food-borne pathogens: Listeria monocytogenes and Staphylococcus aureus. The biosensor is composed of an array of gold nanoparticles-modified screen-printed carbon electrodes on which magnetic nanoparticles coupled to specific peptides were immobilized via streptavidin-biotin interaction. Taking advantage of the proteolytic activities of the protease enzymes produced from the two bacteria on the specific peptides, the detection was achieved in 1 min. The detection was realized by measuring the percentage increase of the square wave voltammetric peak current at 0.1 V versus a Ag/AgCl reference electrode in ferro/ferricyanide redox couple after incubation with the bacteria protease. The integration of the specificity of the bacterial enzymes towards their peptide substrates with the sensitivity of the electrochemical detection on the sensor array allows the rapid, sensitive and selective quantification of the two bacteria. Outstanding sensitivities were achieved using this biosensor array platform with limit of detection of 9 CFU mL−1 for Listeria monocytogenes and 3 CFU mL−1 for Staphylococcus aureus. The multiplexing capability and selectivity of the array voltammetric biosensor were demonstrated by analysing samples of Staphylococcus aureus, Listeria monocytogenes or E. coli and also containing a mixture of two or three bacteria. Using this biosensor, the two bacteria were successfully quantified simultaneously in one step without the need for DNA extraction or amplification techniques. This platform offers promise for rapid, simple and cost-effective simultaneous detection of various bacteria.

Abstract:Listeria monocytogenes (LM) is a foodborne pathogen, and it can pose a risk of serious diseases to the human health. Hence, the development of an effective method for the detection of LM is very important. In this study, by selecting LM as the template and 3-thiopheneacetic acid as the functional monomer, an LM-imprinted polymer (LIP)-based sensor was proposed for the first time to detect LM by electropolymerizing TPA on the glassy carbon electrode (GCE) surface in the presence of LM. After the removal of the LM template from the electrode surface, the obtained sensor was denoted as LIP/GCE, which could effectively recognize and capture LM cells. By using [Fe(CN)6]4-/3- as the probe, its peak current at LIP/GCE could be restricted when the LM cells were captured into the imprinted cavity of LIP/GCE, and the current value decreased with an increase in the LM concentration. Serious conditions were optimized for achieving highly sensitive detection, and a low detection limit (6 CFU mL-1) coupled with a wide linear range (10 to 106 CFU mL-1) was obtained for LM. Finally, the inter-electrode reproducibility, stability, selectivity, and applicability of LIP/GCE were also investigated, and the obtained results were acceptable.

Abstract:Listeria monocytogenes (L.M.) is a gram-positive bacillus with wide distribution in the environment. This bacterium contaminates water sources and food products and can be transmitted to the human population. The infection caused by L.M. is called listeriosis and is common in pregnant women, immune-deficient patients, and older adults. Based on the released statistics, listeriosis has a high rate of hospitalization and mortality; thus, rapid and timely detection of food contamination and listeriosis cases is necessary. During the last few decades, biosensors have been used for the detection and monitoring of varied bacteria species. These devices are detection platforms with great sensitivity and low detection limits. Among different types of biosensors, electrochemical biosensors have a high capability to circumvent several drawbacks associated with the application of conventional laboratory techniques. In this review article, different electrochemical biosensor types used for the detection of listeriosis were discussed in terms of actuators, bioreceptors, specific working electrodes, and signal amplification. We hope that this review will facilitate researchers to access a complete and comprehensive template for pathogen detection based on the different formats of electrochemical biosensors.

Abstract:Rapid detection of bacterial pathogens is a critical unmet need for both food and environmental samples such as irrigation water. As a part of the Food safety Modernization Act (FSMA), The Produce Safety rule has established several requirements for testing for the presence of generic Escherichia coli in water, but the current method available for testing (EPA M1603) demands specified multiple colony verification and highly trained personnel to perform these tests. The purpose of the study was to assess a phage induced bacterial lysis using quantitative image analysis to achieve rapid detection of E. coli at low concentrations within 8 hours. This study aimed to develop a simple yet highly sensitive and specific approach to detect target bacteria in complex matrices. In the study, E. coli cells were first enriched in tryptic soy broth (TSB), followed by T7 phage induced lysis, concentration, staining and fluorescent imaging. Image analysis was conducted including image pre-processing, image segmentation and quantitatively analysis of cellular morphological features (area, eccentricity and full width at half maximum). Challenge experiments using realistic matrices, including simulated fresh produce wash water, coconut water and spinach wash water, demonstrated the method can be applied for use in situations that occur in food processing facilities. The results indicated E. coli cells that are lysed by T7 phages demonstrated significantly (P < 0.05) higher extracellular DNA release, altered cellular shape (from rod to circular) and diffused fluorescent signal intensity. Using this biosensing strategy, a sensitivity to detect Escherichia coli at 10 CFU/ml within 8 hours was achieved, both in laboratory medium and in complex matrices. The proposed phage based biosensing strategy enables rapid detection of bacteria and is applicable to analysis of food systems. Furthermore, the steps involved in this assay can be automated to enable detection of target bacteria in food facilities without extensive resources.

Abstract:The isolation and identification of pathogenic bacteria from a variety of samples are critical for controlling bacterial infection-related health problems. The conventional methods, such as plate counting and polymerase chain reaction-based approaches, tend to be time-consuming and reliant on specific instruments, severely limiting the effective identification of these pathogens. In this study, we employed the specificity of the cell wall-binding (CBD) domain of the Staphylococcus aureus bacteriophage 80 alpha (80α) endolysin towards the host bacteria for isolation. Amidase 3-CBD conjugated magnetic beads successfully isolated as few as 1 × 102 CFU/mL of S. aureus cells from milk, blood, and saliva. The cell wall hydrolyzing activity of 80α endolysin promoted the genomic DNA extraction efficiency by 12.7 folds on average, compared to the commercial bacterial genomic DNA extraction kit. Then, recombinase polymerase amplification (RPA) was exploited to amplify the nuc gene of S. aureus from the extracted DNA at 37 °C for 30 min. The RPA product activated Cas12a endonuclease activity to cleave fluorescently labeled ssDNA probes. We then converted the generated signal into a fluorescent readout, detectable by either the naked eye or a portable, self-assembled instrument with ultrasensitivity. The entire procedure, from isolation to identification, can be completed within 2 h. The simplicity and sensitivity of the method developed in this study make it of great application value in S. aureus detection, especially in areas with limited resource supply.

Abstract:Early diagnosis and appropriate treatment of Escherichia coli (E. coli) O157:H7 and methicillin-resistant Staphylococcus aureus (MRSA) are key elements in preventing resultant life-threatening illnesses, such as hemorrhagic colitis, hemolytic uremic syndrome, and septicemia. In this report, we describe the use of surface plasmon resonance (SPR) for the biodetection of pathogenic bacteria, using bacteriophages as the recognition elements. T4 bacteriophages were used to detect E. coli, while a novel, highly specific phage was used to detect MRSA. We found that the system permits label-free, real-time, specific, rapid and cost-effective detection of pathogens, for concentrations of 10(3) colony forming units/milliliter, in less than 20 min. This system promises to become a diagnostic tool for bacteria that cause major public concern for food safety, bioterrorism, and nosocomial infections.

Abstract:A novel bacteriophage-targeted electrochemical biosensor designed for accurate and quantitative detection of live Salmonella in food samples is presented. The biosensor is simply constructed by electrostatic immobilizing bacteriophages on MXene-nanostructured electrodes. MXene, renowned for its high surface area, biocompatibility, and conductivity, serves as an ideal platform for bacteriophage immobilization. This allows for a high-density immobilization of bacteriophage particles, achieving approximately 71 pcs μm-2. Remarkably, the bacteriophages immobilized MXene nanostructured electrodes still maintain their viability and functionality, ensuring their effectiveness in pathogen detection. Therefore, the proposed biosensor exhibited enhanced sensitivity with a low limit of detection (LOD) of 5 CFU mL-1. Notably, the biosensor shows excellent specificity in the presence of other bacteria that commonly contaminate food and can distinguish live Salmonella from a mixed population. Furthermore, it is applicable in detecting live Salmonella in food samples, which highlights its potential in food safety monitoring. This biosensor offers simplicity, convenience, and suitability for resource-limited environments, making it a promising tool for on-site monitoring of foodborne pathogenic bacteria.

Abstract:Campylobacter jejuni is a common foodborne pathogen found in poultry that can cause severe life-threatening illnesses in humans. It is important to detect this pathogen in food to manage foodborne outbreaks. This study reports a novel impedimetric phage protein-based biosensor to detect C. jejuni NCTC 11168 at 100 CFU/mL concentrations using a genetically engineered receptor-binding phage protein, FlaGrab, as a bioreceptor. The electrochemical impedance spectroscopy (EIS) technique was employed to measure changes in resistance upon interaction with C. jejuni. The sensitivity of the phage protein-immobilized electrode was assessed using the various concentrations of C. jejuni NCTC 11168 ranging from 102–109 colony forming units (CFU)/mL). The change transfer resistance of the biosensor increased with increasing numbers of C. jejuni NCTC 11168 cells. The detection limit was determined to be approximately 103 CFU/mL in the buffer and 102 CFU/mL in the ex vivo samples. Salmonella enterica subsp. enterica serotype Typhimurium-291RH and Listeria monocytogenes Scott A were used as nontarget bacterial cells to assess the specificity of the developed biosensor. Results showed that the developed biosensor was highly specific toward the target C. jejuni NCTC 11168, as no signal was observed for the nontarget bacterial cells.

Ultrasensitive and Fast Diagnostics of Viable Listeria Cells by CBD Magnetic Separation Combined with A511:: luxAB Detection

Abstract:The genus Listeria includes foodborne pathogens that cause life-threatening infections in those at risk, and sensitive and specific methods for detection of these bacteria are needed. Based on their unrivaled host specificity and ability to discriminate viable cells, bacteriophages represent an ideal toolbox for the development of such methods. Here, the authors describe an ultrasensitive diagnostic protocol for Listeria by combining two phage-based strategies: (1) specific capture and concentration of target cells by magnetic separation, harnessing cell wall-binding domains from Listeria phage endolysins (CBD-MS); and (2) highly sensitive detection using an adaptation of the A511::luxAB bioluminescent reporter phage assay in a microwell plate format. The combined assay enabled direct detection of approximately 100 bacteria per ml of pure culture with genus-level specificity in less than 6 h. For contaminated foods, the procedure included a 16 h selective enrichment step, followed by CBD-MS separation and A511::luxAB detection. It was able to consistently detect extremely low numbers (0.1 to 1.0 cfu/g) of viable Listeria cells, in a total assay time of less than 22 h. These results demonstrate the superiority of this phage-based assay to standard culture-based diagnostic protocols for the detection of viable bacteria, with respect to both sensitivity and speed.
Phage M13KO7 detection with biosensor based on imaging ellipsometry and AFM microscopic confirmation

Abstract:A rapid detection and identification of pathogens is important for minimizing transfer and spread of disease. A label-free and multiplex biosensor based on imaging ellipsometry (BIE) had been developed for the detection of phage M13KO7. The surface of silicon wafer is modified with aldehyde, and proteins can be patterned homogeneously and simultaneously on the surface of silicon wafer in an array format by a microfluidic system. Avidin is immobilized on the surface for biotin-anti-M13 immobilization by means of interaction between avidin and biotin, which will serve as ligand against phage M13KO7. Phages M13KO7 are specifically captured by the ligand when phage M13KO7 solution passes over the surface, resulting in a significant increase of mass surface concentration of the anti-M13 binding phage M13KO7 layer, which could be detected by imaging ellipsometry with a sensitivity of 109pfu/ml. Moreover, atomic force microscopy is also used to confirm the fact that phage M13KO7 has been directly captured by ligands on the surface. It indicates that BIE is competent for direct detection of phage M13KO7 and has potential in the field of virus detection.
Expression of the active caspase-3 in children and adolescents with classical Hodgkin lymphoma.

Abstract:UNLABELLED The aim of the study was to determine whether the expression of active caspase-3 in neoplastic Hodgkin and Reed-Sternberg (H/RS) cells correlates with the treatment response and provides prognostic information on treatment outcome. In this retrospective study, we included 56 patients with classical Hodgkin lymphoma treated at the Department of Paediatric Haematology and Oncology between January 2000 and June 2005. Active caspase-3 was detected by immunohistochemistry in primary biopsy specimens. Seventeen patients (29.3%) were evaluated as caspase-3 positive and remained alive in the first complete remission. This stood in contrast to patients with less than 5% caspase-3 positive cells, five of whom experienced relapse and three patients died. Adequate treatment response was achieved in 11 patients (19.6%). Comparison of event-free survival with regard to the percentage of caspase-3 positive tumour cells showed a tendency for a better clinical outcome in patients with 5% or more active caspase-3 positive cells. Abstract:An increasing proportion of pituitary adenomas are recognized in the elderly, raising the question of their optimal diagnosis and management. Age-related endocrine changes and associated diseases may significantly modify the clinical presentation and hormonal evaluation of these patients. About 80% of pituitary adenomas in this age group are non-secreting, requiring careful differential diagnosis with non-adenomatous sellar lesions. In this group, visual deterioration and hypopituitarism remain the leading symptoms. Recognized secreting tumours are mainly GH-secreting, most of them intrasellar, followed by prolactinomas, which present as clinically non-secreting and are usually invasive. Cushing's disease appears as a very rare eventuality in the elderly. Optimal therapeutic management should aim to control the disease while preserving or improving patient's quality of life. Transsphenoidal surgery has proved to be an efficient and well-tolerated option for non-secreting adenomas with visual defects and intrasellar GH-secreting adenomas, being able to improve metabolic and cardiovascular complications of acromegaly even in this age group. In contrast, dopamine-agonist drugs can be proposed as a primary therapy for prolactinomas even in the presence of severe neurological complications. Because the use of radiotherapy is hampered by its delay of action and potential neurological side effects, its indications should be better defined in this age group. The clinical importance of hypopituitarism should not be underestimated, and thyroid- and adrenal-replacement therapy are mandatory in the presence of documented hormone deficiency, carefully avoiding overtreatment in order to limit possible side effects on the cardiovascular system and bone mineralization. Indications for GH- and sex steroid-replacement therapy still await age-specific guidelines.

Abstract:A 59-year-old woman was referred to our laryngology clinic for a 2-month history of fluctuating vocal quality and poor projection after a prodromal upper respiratory infection. Her initial vocal quality was severely breathy. It had partially improved, but she still complained of vocal fatigue and lack of vocal volume. Physical exam demonstrated no significant abnormalities aside from severe breathy dysphonia with a maximum phonation time of just 3 seconds and abnormal vocal fold motion. Videostroboscopy revealed sluggish adduction of the left vocal fold and arytenoid complex with a significant glottic gap throughout the phonatory cycle and decreased mucosal wave propagation. The patient was diagnosed with left true vocal fold paresis, and vocal fold augmentation was recommended to improve her glottic insufficiency. For topical nasal anesthesia, 4% lidocaine mixed equally with oxymetazoline was sprayed into the nasal cavity, and a total of 5 ml of 4% lidocaine was delivered directly to the endolarynx using the operative port on a flexible laryngoscope. Injection laryngoplasty was performed using a 23-gauge flexible sclerotherapy needle directed through the laryngoscope (figure, A). The paretic left vocal fold was augmented with 0.7 ml of hyaluronic acid delivered via a single posterior lateral injection (figure, B). The patient had immediate improvement in glottic closure with increased vocal strength and projection. Percutaneous or transoral vocal fold augmentation is a common in-office technique performed under local anesthesia, avoiding the high cost associated with direct laryngoscopy and general anesthesia in a hospital setting. Both approaches involve endoscopic imaging to visualize the injection site.1,2 Transnasal injection using a flexible needle via the operative port of a flexible laryngoscope allows for intraoperative assessment of vocal quality during injection laryngoplasty, with decreased gag reflex compared to transoral techniques.3 Transnasal vocal fold augmentation is tolerated well by patients, easy to perform, and can be done without an assistant. It allows for direct laryngeal delivery of numerous products including micronized Alloderm, hyaluronic acid, botulinum toxin, and steroid suspensions, among others.4 Viscous injection materials such as fat or calcium hydroxyapatite may be challenging to deliver using the 23-gauge flexible needle. Flexible laryngoscopy is a standard skill of the trained otolaryngologist, and injection by this technique is learned easily compared to more challenging percutaneous approaches.

Abstract:A colorimetric hydrogel sensing platform for the detection of pathogens in water and clinical samples is described. The polyacrylamide hydrogel is held together with pathogen‐responsive DNAzyme‐substrate cross‐linking and has demonstrated high sensitivity for detecting E. coli in lake water and urine. The bacteriophage‐mediated signal amplification effect outlined in the design has potential applications for future biosensor development. Developing cost‐effective, consumer‐accessible platforms for point‐of‐use environmental and clinical pathogen testing is a priority, to reduce reliance on laborious, time‐consuming culturing approaches. Unfortunately, a system offering ultrasensitive detection capabilities in a form that requires little auxiliary equipment or training has remained elusive. Here, a colorimetric DNAzyme‐crosslinked hydrogel sensor is presented. In the presence of a target pathogen, DNAzyme cleavage results in hydrogel dissolution, yielding the release of entrapped gold nanoparticles in a manner visible to the naked eye. Recognizing that Escherichia coli holds high relevance within both environmental and clinical environments, an E. coli‐responsive DNAzyme is incorporated into this platform. Through the optimization of the hydrogel polymerization process and the discovery of bacteriophage‐induced DNAzyme signal amplification, 101 CFU mL−1 E. coli is detected within real‐world lake water samples. Subsequent pairing with an artificial intelligence model removed ambiguity in sensor readout, offering 96% true positive and 100% true negative accuracy. Finally, high sensor specificity and stability results supported clinical use, where 100% of urine samples collected from patients with E. coli urinary tract infections are accurately identified. No false positives are observed when testing healthy samples. Ultimately, this platform stands to significantly improve population health by substantially increasing pathogen testing accessibility.

Abstract:Graphical representation for chemical immobilization of E. coli‐specific bacteriophage with NH2‐MIL‐101(Fe) MOF a high photoluminescence rod‐shaped microcrystals for low‐level bacteria detection. As concern raised by the World Health Organization (WHO) of antibiotic‐resistant and bio‐defensive bacteria, a metal–organic framework (MOF) based optical biosensor came into consideration for precise, quick, and sensitive detection of Escherichia coli (E. coli) (ATCC 10799) using bacteriophage as a bio‐recognition element. In the present study, amine‐functionalized Fe‐based MOF, i.e., NH2‐MIL‐101(Fe), was synthesized by the solvothermal method (approx. 531–1106 nm in size and 20 mV zeta potentials by DLS) and further characterized by SEM, XRD, ATR‐FTIR, UV–VIS, and photoluminescent (PL) spectroscopy. The lytic bacteriophage was isolated from a sewage sample, purified, and concentrated using the ultra‐centrifugation method and achieved a high titer of 7.3 × 1012 PFU/ml. The concentration, stability, and accessible receptor binding domains (RBDs) of the biorecognition element for binding with their analytes play an important role in developing sensitive and specific biosensor systems. To fulfill the mentioned criteria, optimized glutaraldehyde concentration was estimated at 0.25%, at 30 °C for conjugating maximum bacteriophage titer of 8.6 × 105 PFU/ml for each 1 mg amine functionalized iron‐based MOF. The synthesized detection probe has shown excellent photoluminescence and antibacterial activity and achieved a detection limit of 652 CFU/ml over a bacterial detection concentration range from 5.78 × 101 to 5.78 × 106 CFU/ml for E. coli with 10–12 min of response time, high specificity, and long‐term stability even at room temperature. Therefore, it can be inferred that this MOF‐based strategy can be helpful in the specific and sensitive detection of various bacterial pathogens using bacteriophage as a bio‐recognition element.
