Abstract:The genus Listeria includes foodborne pathogens that cause life-threatening infections in those at risk, and sensitive and specific methods for detection of these bacteria are needed. Based on their unrivaled host specificity and ability to discriminate viable cells, bacteriophages represent an ideal toolbox for the development of such methods. Here, the authors describe an ultrasensitive diagnostic protocol for Listeria by combining two phage-based strategies: (1) specific capture and concentration of target cells by magnetic separation, harnessing cell wall-binding domains from Listeria phage endolysins (CBD-MS); and (2) highly sensitive detection using an adaptation of the A511::luxAB bioluminescent reporter phage assay in a microwell plate format. The combined assay enabled direct detection of approximately 100 bacteria per ml of pure culture with genus-level specificity in less than 6 h. For contaminated foods, the procedure included a 16 h selective enrichment step, followed by CBD-MS separation and A511::luxAB detection. It was able to consistently detect extremely low numbers (0.1 to 1.0 cfu/g) of viable Listeria cells, in a total assay time of less than 22 h. These results demonstrate the superiority of this phage-based assay to standard culture-based diagnostic protocols for the detection of viable bacteria, with respect to both sensitivity and speed.
Detection and differentiation of Listeria species by a single reaction based on multiplex PCR

Abstract:The genus Listeria encompasses six bacterial species L. monocytogenes, L. ivanovii, L.seeligeri, L. innocua, L. welshimeri and L. grayi. Although these species resemble each other morphologically, ecologically, biochemically and genetically, they show varied pathogenic potential. L. monocytogenes is a significant human, animal and aquatic pathogen with clinical manifestation ranging from mild flu-like symptoms and gastroenteritis to septicemia, central nervous infection and feto-maternal infection. Through comparative genomics analysis, we used the iap gene encoding invasion-associated protein common to all members of the genus Listeria as the target because the comparison of all iap genes indicated there were conserved gene portions at the 5’and 3’ ends, while the internal portions were species specific. In addition, the liv22-228 gene region was specific in L. ivanovii. A novel multiplex PCR, based on liv22-228 in combination with optimized iap migration profiles, was developed for simultaneous identification of Listeria species, with a detection limit of down to 102 CFU/mL. Other 15 bacterial species did not show any amplicons via this PCR reaction. Furthermore, this assay was evaluated by 135 suspected Listeria isolates from aquatic products and corrected 4 and 5 misidentifications by Listeria selective agar plate screening and API system respectively. Therefore, this one-step molecular assay provides a rapid, accurate, reliable and inexpensive screening test to detect and differentiate Listeria species particularly the pathogenic species in surveillance programs concerning aquatic products safety and foodborne disease cases.

Abstract:Listeria monocytogenes (L. monocytogenes) is a food-borne pathogenic bacteria that frequently contaminates animal-derived food and low-temperature preserved food. Listeriosis caused by its infection has a high mortality rate and poses a serious threat to human health. Therefore, it is crucial to establish a sensitive, rapid and easy-to-operate technique. In this study, a Recombinase Aided Amplification (RAA) assisted CRISPR/Cas12a (RAA-CRISPR/Cas12a) fluorescence platform was established for highly sensitive nucleic acid detection of L. monocytogenes. The established RAA-CRISPR/Cas12a showed high sensitivity and high specificity, with the sensitivity of 350 CFU/mL and 5.4 × 10−3 ng/μL for pure bacterial solution and genomic DNA, and good specificity for 5 strains of Listeria spp. and 14 strains of other common pathogenic bacteria. L. monocytogenes could be detected at an initial concentration of 2.3 CFU/25g within 2 h of enriching the beef in the food matrix, and this method could be applied to food samples that were easily contaminated with L. monocytogenes The results of RAA-CRISPR/Cas12a could be observed in 5 min, while the amplification was completed in 20–30 min. The speed and sensitivity of RAA-CRISPR/Cas12a were significantly higher than that of the national standard method. In conclusion, the RAA-CRISPR/Cas12a system established in this study has new application potential in the diagnosis of food-borne pathogens.

Glycotyping and Specific Separation of Listeria monocytogenes with a Novel Bacteriophage Protein Tool Kit

Abstract:The Gram-positive pathogen Listeria monocytogenes can be subdivided into at least 12 different serovars, based on the differential expression of a set of somatic and flagellar antigens. Of note, strains belonging to serovars 1/2a, 1/2b, and 4b cause the vast majority of foodborne listeriosis cases and outbreaks. The standard protocol for serovar determination involves an agglutination method using a set of sera containing cell surface-recognizing antibodies. However, this procedure is imperfect in both precision and practicality, due to discrepancies resulting from subjective interpretation. Furthermore, the exact antigenic epitopes remain unclear, due to the preparation of the absorbed sera and the complex nature of polyvalent antibody binding. Here, we present a novel method for quantitative somatic antigen differentiation using a set of recombinant affinity proteins (cell wall-binding domains and receptor-binding proteins) derived from a collection of Listeria bacteriophages. These proteins enable rapid, objective, and precise identification of the different teichoic acid glycopolymer structures, which represent the O-antigens, and allow a near-complete differentiation. This glycotyping approach confirmed serovar designations of over 60 previously characterized Listeria strains. Using select phage receptor-binding proteins coupled to paramagnetic beads, we also demonstrate the ability to specifically isolate serovar 1/2 or 4b cells from a mixed culture. In addition, glycotyping led to the discovery that strains designated serovar 4e actually possess an intermediate 4b-4d teichoic acid glycosylation pattern, underpinning the high discerning power and precision of this novel technique.IMPORTANCEListeria monocytogenes is a ubiquitous opportunistic pathogen that presents a major concern to the food industry due to its propensity to cause foodborne illness. The Listeria genus contains 15 different serovars, with most of the variance depending on the wall-associated teichoic acid glycopolymers, which confer somatic antigenicity. Strains belonging to serovars 1/2 and 4b cause the vast majority of listeriosis cases and outbreaks, meaning that regulators, as well as the food industry itself, have an interest in rapidly identifying isolates of these particular serovars in food processing environments. Current methods for phenotypic serovar differentiation are slow and lack accuracy, and the food industry could benefit from new technologies allowing serovar-specific isolation. Therefore, the novel method described here for rapid glycotype determination could present a valuable asset to detect and control this bacterium.

Abstract:A simple, precise, and highly sensitive detection method is urgently needed for diagnosis and monitoring of foodborne pathogens. To meet these requirements, we designed a micro-propulsion reactor based on the trans-cleavage activity of Cas12a and recombinase polymerase amplification (RPA) for simultaneous amplification and detection and named it "Cas12a-MPR". This method can provide a contamination- and instrument-free nucleic acid rapid detection platform with precision and ultrasensitivity using CRISPR-Cas12a combined with RPA in the same vessel. For improved performance, the interaction between the CRISPR detection system and the RPA reagent was explored. Our data showed that the impact of CRISPR reagents on RPA was negligible, but magnesium ions played a vital role in the functioning of the CRISPR/Cas12a system. After exploring the interaction between the CRISPR detection system and the RPA reagent, the method was further optimized to specifically detect target DNA at the attomolar level and achieved a limit of detection of 10 CFU mL-1 at 37 °C. Compared to qPCR, our detection results exhibited 100 % accuracy for detection of Listeria monocytogenes contamination. All the results demonstrated that Cas12a-MPR has great promise for the detection of foodborne pathogens.

Abstract:Listeria monocytogenes ( L. monocytogenes ) is a prevalent food-borne pathogen that can cause listeriosis, which manifests as meningitis and other symptoms, potentially leading to fatal outcomes in severe cases. In this study, we developed an aptasensor utilizing carboxylated magnetic beads and Cas12a to detect L. monocytogenes . In the absence of L. monocytogenes , the aptamer maintains its spatial configuration, keeping the double-stranded DNA attached and preventing the release of a startup template and activation of Cas12a's trans-cleavage capability. Conversely, in the presence of L. monocytogenes , the aptamer undergoes a conformational change, releasing the double-stranded DNA to serve as a startup template, thereby activating the trans-cleavage capability of Cas12a. Consequently, as the concentration of L. monocytogenes increases, the observable brightness in a blue light gel cutter intensifies, leading to a rise in fluorescence intensity difference compared to the control. This Cas12a aptasensor demonstrates excellent sensitivity towards L. monocytogenes , with a lowest detection limit (LOD) of 57.15 CFU/mL and a linear range of 4×10 2 to 4×10 7 CFU/mL ( R 2 =0.9858). Notably, the proposed Cas12a aptasensor exhibited outstanding selectivity and recovery in beef samples, and could be employed for precise monitoring. This Cas12a aptasensor not only provides a novel fluorescent and visual rapid detection method for L. monocytogenes but also offers simplicity, speed, and stability compared to previous detection methods. Furthermore, it is suitable for on-site detection of beef samples.

Abstract:Listeria monocytogenes is a Gram-positive facultative intracellular pathogen that is widely used as a model organism for the analysis of infection biology. In this context, there is a current need to develop improved reporters for enhanced bioluminescence imaging (BLI) of the pathogen in infection models. We have developed a click beetle red luciferase (CBR-luc) based vector (pPL2CBRopt) expressing codon optimized CBR-luc under the control of a highly expressed Listerial promoter (PHELP) for L. monocytogenes and have compared this to a lux-based system expressing bacterial luciferase for BLI of the pathogen using in vitro growth experiments and in vivo models. The CBR-luc plasmid stably integrates into the L. monocytogenes chromosome and can be used to label field isolates and laboratory strains of the pathogen. Growth experiments revealed that CBR-luc labeled L. monocytogenes emits a bright signal in exponential phase that is maintained during stationary phase. In contrast, lux-labeled bacteria produced a light signal that peaked during exponential phase and was significantly reduced during stationary phase. Light from CBR-luc labeled bacteria was more efficient than the signal from lux-labeled bacteria in penetrating an artificial tissue depth assay system. A cell invasion assay using C2Bbe1 cells and a systemic murine infection model revealed that CBR-luc is suited to BLI approaches and demonstrated enhanced sensitivity relative to lux in the context of Listeria infection models. Overall, we demonstrate that this novel CBR reporter system provides efficient, red-shifted light production relative to lux and may have significant applications in the analysis of L. monocytogenes pathogenesis.

Abstract:The simultaneous and sensitive detection of live/dead bacteria in food samples is crucial for the control and prevention of foodborne pathogen contamination. Herein, a novel three-signal biosensor for simultaneous and ultrasensitive detection of live/dead Listeria monocytogenes ( L.m) was developed. Concretely, a phage/prussian blue nanoparticles (PBNPs) and phage/polyethyleneimine@magnetic nanoparticles (PEI@MNPs) multifunctional probes were prepared. In the presence of L.m , a sandwich complex of phage/PBNPs- L.m -phage/PEI@MNPs was formed and separated by magnetic field. With the lytic activity of phages, the bioluminescent signal generated by adenosine triphosphate (ATP) was utilized for the quantification of live L.m . Photothermal and colorimetric signals triggered by the PBNPs could quantitative determination of total L.m . The combined results of total/live bacteria detection facilitate the calculation of dead bacteria. Bioluminescent signal allows for the precise detection of live L.m with a limit of detection (LOD) of 1 CFU/mL, while the photothermal and colorimetric signals enable on-site measurement of total bacteria with a LOD of 5 CFU/mL. This biosensor exhibits the multiple signals, realizing the diversification of bacterial information output. Meanwhile, the three signals can realize the mutually support, thereby improving the accuracy of detection results. This work opens a new avenue for the simple, ultrasensitive and simultaneous quantification of both live and dead bacteria.

The Development and Application of Lyophilized LAMP Detection Reagent for Listeria monocytogenes

Abstract:Listeria monocytogenes , a zoonotic foodborne pathogen, presents a significant threat to global public health. Therefore, rapid and sensitive detection methods are crucial in mitigating the spread of L. monocytogenes induced diseases. This study introduced a loop-mediated isothermal amplification (LAMP) lyophilized powder detection reagent specifically designed for identifying Listeria monocytogenes . The reagent is user-friendly, quick, and can be easily transported and stored at room temperature. It exhibits no cross-reactivity with eight other types of bacteria and boasts a sensitivity of 10 1 CFU/mL. In a comparative study of 30 samples, the LAMP lyophilized powder detection reagent demonstrated higher sensitivity than the commercial Listeria monocytogenes qPCR detection kit. Additionally, the experimental time was reduced by approximately 30 min, making it highly suitable for rapid diagnosis. Preparation of lyophilized LAMP reagents may facilitate large-scale deployment, particularly in endemic areas or regions facing rapid outbreaks. This could greatly aid in controlling the transmission of pathogens, especially those transmitted through food.

Abstract:Listeria monocytogenes (L. monocytogenes) is a Gram-positive microbe and is reported to be potentially pathogenic for both humans and animals, exhibiting a high mortality rate of up to 30%. The invention of a fast and sensitive method of point-of-care testing (POCT) is of great significance for the field of food safety of the rapid detection of harmful microbes and prevention of food contaminated by such microbes from reaching the markets. A novel isothermal molecular amplification technique, recombinase polymerase amplification (RPA), has been used to detect L. monocytogenes in food combined with a highly sensitive end determining method, lateral flow dipstick (LFD) technology. RPA-LFD was implemented at 37 °C, and amplification only took 20 min. By optimizing the primers and probes, the system had no cross-reaction with seven other pathogens, and it was proven to have a 100 times higher sensitivity (1360 cfu/mL) than a real-time PCR for L. monocytogenes testing. Also, RPA-LFD was applied without complex or expensive equipment, only a thermostatic heater such as the Thermomixer Comfort was required. Thus, RPA-LFD was found to be a solution for POCT for L. monocytogenes, especially for low-resource locations with little available laboratory equipment.

Abstract:Bacteriophages (“phages”) infect and multiply within specific bacterial strains, causing lysis of their target. Due to the specific nature of these interactions, phages allow a high-precision approach for therapy which can also be exploited for the detection of phage-sensitive pathogens associated with chronic diseases due to gut microbiome imbalance. As rapid phage-mediated detection assays becoming standard-of-care diagnostic tools, they will advance the more widespread application of phage therapy in a precision approach. Using a conventional method and a new cloning approach to develop luminescent phages, we engineered two phages that specifically detect a disease-associated microbial strain. We performed phage sensitivity assays in liquid culture and in fecal matrices and tested the stability of spiked fecal samples stored under different conditions. Different reporter gene structures and genome insertion sites were required to successfully develop the two nluc-reporter phages. The reporter phages detected spiked bacteria in five fecal samples with high specificity. Fecal samples stored under different conditions for up to 30 days did not display major losses in reporter-phage-based detection. Luminescent phage-based diagnostics can provide a rapid co-diagnostic tool to guide the growing field of phage therapy, particularly for a precision-based approach to chronic diseases treatment.

Abstract:Listeria monocytogenes (Listeria) is a facultative pathogenic bacterium, and a sensitive method for specific detection of Listeria is considered of clinically significant. However, current approaches for identifying Listeria are time consuming or tentative, and especially, cannot identify bacterial viability and antibiotic efficacy, which are critical in establishing a treatment recipe. Herein, we have developed a nitroreductase (NTR) responsive fluorescent probe (NRFP) with a fluorescence off-on feature, which could rapidly detect Listeria in vitro and in vivo with high specificity and sensitivity. NRFP showed a selective response to NTRs over other biological reductants, and could detect NTRs as low as 12.5 ng/mL. Furthermore, NRFP responded rapidly to NTRs within 10 min, enabling it real-time monitoring NTR production. Most importantly, NRFP could not only distinguish Listeria from other bacteria in vitro and in vivo for the first time, but could provide valuable information about Listeria desired for healthcare professionals, such as the presence and viability. Particularly, NRFP could real-time monitor antimicrobial effects in vivo, thereby identifying effective antibiotics for Listeria infections. Overall, NRFP appears to be an ideal imaging probe for Listeria, and possesses great potentials in diagnostic and therapeutic applications.
Rapid and Specific Detection of Listeria monocytogenes With an Isothermal Amplification and Lateral Flow Strip Combined Method That Eliminates False-Positive Signals From Primer–Dimers

Abstract:Listeria monocytogenes is an important foodborne pathogenic bacterium that is explicitly threatening public health and food safety. Rapid, simple, and sensitive detection methods for this pathogen are of urgent need for the increasing on-site testing demands. Application of the isothermal recombinase polymerase amplification (RPA) and the lateral flow strip (LFS) in the detection is promising for fast speed, high sensitivity, and little dependency on equipment and trained personnel. However, the simplicity comes with an intrinsic and non-negligible risk, the false-positive signals from primer–dimers. In this study, an improved RPA–LFS system was established for detection of L. monocytogenes that eliminated false-positive signals from primer–dimers. Primer candidates were carefully selected from the entire L. monocytogenes genome sequence and rigorously screened for specific amplifications in PCR and RPA reactions. For the optimal primer pairs, probes that matched the targeted fragment sequences, although had the smallest chance to form cross-dimers with the primers, were designed and screened. The intelligent use of the probe successfully linked the positive signal to the actual amplification product. This RPA–LFS system was highly specific to L. monocytogenes and was able to detect as low as 1 colony-forming unit of the bacterium per reaction (50 μl) without DNA purification, or 100 fg of the genomic DNA/50 μl. The amplification could be conducted under the temperature between 37 and 42°C, and the whole detection finished within 25 min. Test of artificially contaminated milk gave 100% accuracy of detection without purification of the samples. Various food samples spiked with 10 colony-forming unit of L. monocytogenes per 25 g or 25 ml were successfully detected after an enrichment time period of 6 h. The RPA–LFS system established in this study is a rapid, simple, and specific detection method for L. monocytogenes that has eliminated false-positive results from primer–dimers. In addition, this study has set a good example of eliminating the false-positive risk from primer–dimers in isothermal amplification-based detection methods, which is applicable to the development of detection technologies for other pathogens.

Abstract:Conventional molecular detection methods cannot distinguish between viable and dead Escherichia coli O157 cells. In this study, the loop-mediated isothermal amplification (LAMP) method combined with propidium monoazide (PMA) treatment was developed to selectively detect viable E. coli O157 cells. Four primers, including outer primers and inner primers, were specially designed for the recognition of six distinct sequences on the serogroups (O157) of the specific rfbE gene of the E. coli O157 genome. PMA selectively penetrated through the compromised cell membranes and intercalated into DNA. Amplification of DNA from dead cells was completely inhibited by 3.0 μg/ml PMA, whereas the DNA derived from viable cells was amplified remarkably within 1 h by PMA-LAMP. Exhibiting high sensitivity and specificity, PMA-LAMP is a suitable method for evaluating the inactivation efficacy of slightly acidic electrolyzed water in broth. PMA-LAMP can selectively detect viable E. coli O157 cells. This study offers a novel molecular detection method to distinguish between viable and dead E. coli O157 cells.

Abstract:Listeria monocytogenes (L. monocytogenes) is one of the most common food-borne pathogens. The authors describe a sensitive and reliable multicolorimetric assay for L. monocytogenes using a sensing system based on TMB2+ etching of gold nanorods. Apt-MNP was used as the capture probe, and IgY-BSA-MnO2 NPs was chosen as an oxidase-like nano-artificial enzyme to oxidize TMB to generate TMB2+. Under the optimized conditions, the longitudinal shift of localized surface plasmon resonances had a linear correlation with the L. monocytogenes concentration in the range between 10 to 10(6) cfu mL(-1). Meanwhile, the sensing system can generate vivid color responses as colorful as a rainbow, and the limit of detection is as low as 10 cfu mL(-1) at a glance. Recoveries ranging from 97.4 to 101.3% are found when analyzing spiked food samples without pre-enrichment. In our perception, it shows promise in rapid instrumental and on-site visual detection of L. monocytogenes. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:Listeria monocytogenes ( L. monocytogenes ) is a ubiquitous foodborne pathogens responsible for listeriosis, which is associated with high morbidity and mortality. Development of simple and fast methods for L. monocytogenes detection is of great needed. In this study, we present a simple method for L. monocytogenes detection combined aptamers decorated magnetic beads for selective capture and gold nanoparticles (AuNPs) mediated signal amplication. A NH 2 labeled aptamer specific to L. monocytogenes was decorated on magnetic beads for selective capture and isolation of L. monocytogenes from food sample. AuNPs was functionalized with sulfhydryl modified primers specific to hly gene of L. monocytogenes in acidic environments. The AuNP@primers could catalyze TMB-H 2 O 2 to TMB 2+ and generate a significant blue color, this reaction was found to be enhanced by DNA. AuNP@primers was used as a signal indicator for the presence of L. monocytogenes DNA by enhancing the catalytic reaction of TMB-H 2 O 2 . The colour change was observed by naked eye and the optical density of TMB 2 + was analyzed to quantify the result. The developed assay was highly specific and the limit of detection was 10 1 CFU/mL of L. monocytogenes . For the detection of L. monocytogenes in spiked food samples, the recoveries were ranging from 73.63 to 84.21% with the coefficient of variation less than 2.41% in meat, and ranged from 94.13 to 100.71% with the coefficient of variation less than 2.82% in milk, respectively. Therefore, the developed colorimetric assay is suitable for food sample testing, the detection takes less than 1.5 h, which enables on-site screening. This study offers a promising approach for rapid, sensitive and accurate detection of L. monocytogenes in food.

Abstract:Rapid detection of bacterial pathogens is a critical unmet need for both food and environmental samples such as irrigation water. As a part of the Food safety Modernization Act (FSMA), The Produce Safety rule has established several requirements for testing for the presence of generic Escherichia coli in water, but the current method available for testing (EPA M1603) demands specified multiple colony verification and highly trained personnel to perform these tests. The purpose of the study was to assess a phage induced bacterial lysis using quantitative image analysis to achieve rapid detection of E. coli at low concentrations within 8 hours. This study aimed to develop a simple yet highly sensitive and specific approach to detect target bacteria in complex matrices. In the study, E. coli cells were first enriched in tryptic soy broth (TSB), followed by T7 phage induced lysis, concentration, staining and fluorescent imaging. Image analysis was conducted including image pre-processing, image segmentation and quantitatively analysis of cellular morphological features (area, eccentricity and full width at half maximum). Challenge experiments using realistic matrices, including simulated fresh produce wash water, coconut water and spinach wash water, demonstrated the method can be applied for use in situations that occur in food processing facilities. The results indicated E. coli cells that are lysed by T7 phages demonstrated significantly (P < 0.05) higher extracellular DNA release, altered cellular shape (from rod to circular) and diffused fluorescent signal intensity. Using this biosensing strategy, a sensitivity to detect Escherichia coli at 10 CFU/ml within 8 hours was achieved, both in laboratory medium and in complex matrices. The proposed phage based biosensing strategy enables rapid detection of bacteria and is applicable to analysis of food systems. Furthermore, the steps involved in this assay can be automated to enable detection of target bacteria in food facilities without extensive resources.

Abstract:Background: Listeria monocytogenes is a pathogenic bacterium that can lead to severe illnesses, especially among vulnerable populations. Therefore, the development of rapid and sensitive detection methods is vital to prevent and manage foodborne diseases. In this study, we used tetraethylenepentamine (TEPA)-functionalized magnetic nanoparticles (MNPs) and a loop-mediated isothermal amplification (LAMP)-based CRISPR/Cas12a-based biosensor to concentrate and detect, respectively, L. monocytogenes. LAMP enables DNA amplification at a constant temperature, providing a highly suitable approach for point-of-care testing (POCT). The ability of CRISPR/Cas12a to cleave ssDNA reporter, coupled with TEPA-functionalized MNPs effective attachment to negatively charged bacteria, forms a promising biosensor. Results: The LAMP assay was meticulously developed by selecting specific primers and designing crRNA sequences targeting a specific region within the hly gene of L. monocytogenes. We selected primer and refined the amplification conditions by systematically exploring a temperature range from 59 °C to 69 °C, ensuring the attainment of optimal performance. This process was complemented by systematic optimization of LAMP-CRISPR/Cas12a system parameters. In particular, we successfully established the optimal ssDNA reporter concentrations (0-1.2 μM) and Cas12a-mediated trans-cleavage times (0-20 min), crucial components that underpin the effectiveness of the LAMP-CRISPR/Cas12a-based biosensor. For optimizing parameters in capturing L. monocytogenes using TEPA-functionalized MNPs, capture efficiency was significantly enhanced through adjustments in TEPA-functionalized MNPs concentration, incubation times, and magnetic separation duration. Large-volume (20 mL) magnetic separation exhibited a 10-fold sensitivity improvement over conventional methods. Utilizing TEPA-functionalized MNPs, the LAMP-CRISPR/Cas12a-based biosensor achieved detection limits of 100 CFU mL-1 in pure cultures and 100 CFU g-1 in enoki mushrooms. Significance: The integration of this novel technique with the LAMP-CRISPR/Cas12a-based biosensor enhances the accuracy and sensitivity of L. monocytogenes detection in foods, and it can be a promising biosensor for POCT. The 10-fold increase in sensitivity compared to conventional methods makes this approach a groundbreaking advancement in pathogenic bacteria detection for food safety and public health.
