Abstract:Food supplements with microalgae are becoming increasingly abundant and can be easily found anywhere. The most popular products are based on cyanophytes, such as Aphanizomenon flos-aquae, Arthrospira platensis and Limnospira maxima, or on chlorophytes, such as Chlorella or Haematoccus. Although they are all advertised as being very beneficial for health, these products might be harmful because they may contain cyanotoxins and other contaminants, and no information on production methods or strain origins is usually provided. While legislation on the presence of microcystins in waters for different uses is clear, toxicological analyses are not compulsory for food supplements, nor for analyzing anatoxins. Given the potential risk of eating contaminated food, cyanotoxins, heavy metals and the presence of other contaminant organisms were analyzed in 10 microalgae food supplements. Microcystin-LR and anatoxin-a were detected in three analyzed products, and in both cyanophyte- and chlorophyte-based products. The light microscope study revealed the presence of different potentially harmful microbial contaminants. The ICP (OES) analyses detected high concentrations of some heavy metals, especially Pb. The results emphasize the need to promote the better control of food products containing microalgae, and to develop standard methodologies to analyze cyanotoxins and potential toxic compounds to protect consumer health.

Abstract:Dietary supplements based on algae, known for their nutritional value and bioactive properties, are popular products among consumers today. While commercial algal products are regarded safe by numerous studies, information about the production and origin of such products is scarce. In addition, dietary supplements are not as strictly regulated as food and medicinal drugs. We characterized different algal products (kelps: Laminariales, Spirulina spp., Chlorella spp., and Aphanizomenon flos-aquae ), obtained on Slovenian market, based on their elemental composition (X-ray fluorescence, inductively coupled plasma–mass spectrometry), antioxidative potential [DPPH (2,2-diphenyl-1-picrylhydrazyl) assay, total phenolic content], and stable isotope values [carbon (C), nitrogen (N), and sulfur (S); elemental analyzer isotope ratio mass spectrometry (EA-IRMS) method]. Antioxidative potential is consistent among products of the same type, with A. flos-aquae samples having 4.4 times higher antioxidative potential compared to Chlorella spp. and 2.7 times higher compared to Spirulina spp. Levels of toxic trace elements (arsenic, cadmium, mercury, and lead) are below the maximum allowed values and as such do not pose risk to consumers' health. Samples of Spirulina spp. have relatively high δ 15 N (7.4 ‰ ± 4.4‰) values, which indicate use of organic nitrogen sources in certain samples. Likewise, different elemental composition and isotopic ratios of stable elements (C, N, and S) for the samples with Spirulina spp. or Chlorella spp. are the consequence of using different nutrient sources and algae-growing techniques. Statistical analysis (principal component analysis) has confirmed that all tested A. flos-aquae samples originate from the same source, supposedly Klamath Lake (Oregon, USA). Hawaiian Spirulina pacifica can also be differentiated from all the other samples because of its characteristically high metal content (iron, manganese, zinc, cobalt, nickel, vanadium). Chlorella spp. and Spirulina spp. require further analyses with larger number of samples, as differentiation is not possible based on results of this study.

Abstract:In recent years, the total consumption of antibiotics to cure infections has widely increased. Consequently, the concentration of antibiotics in aquatic environments has remarkably heightened. Cefazolin sodium (CFZS) is a first-generation cephalosporin antibiotic with significant bactericidal activities. Immense consumption of CFZS for years along with its persistence in various environments led to its growing deposit in different ecosystems. In this study, the model microalga Chlorella vulgaris was chosen to evaluate the impacts of different concentrations of CFZS on aquatic organisms. Notably, the cell number of the microalgae has declined with the rising concentrations of CFZS. Prepared SEM images and flow cytometry charts confirmed the harmful effects of the antibiotic on the cells of C. vulgaris. Furthermore, CFZS caused an increase in the content of pigments (chlorophyll a, b, carotenoids and anthocyanins), level of non-enzymatic antioxidant compounds (total phenol and flavonoid) and activity of enzymatic antioxidants (CAT, SOD and APX) in C. vulgaris. However, C. vulgaris displayed remarkable potential to remove CFZS from the culture medium. The obtained data may shed light on the establishment of the possible risks of antibiotics to algal populations and also microalgae ability in antibiotics removal.
Cytotoxic and Genotoxic Effects of Cyanobacterial and Algal Extracts-Microcystin and Retinoic Acid Content

Abstract:In the last decade, it has become evident that complex mixtures of cyanobacterial bioactive substances, simultaneously present in blooms, often exert adverse effects that are different from those of pure cyanotoxins, and awareness has been raised on the importance of studying complex mixtures and chemical interactions. We aimed to investigate cytotoxic and genotoxic effects of complex extracts from laboratory cultures of cyanobacterial species from different orders (Cylindrospermopsis raciborskii, Aphanizomenon gracile, Microcystis aeruginosa, M. viridis, M. ichtyoblabe, Planktothrix agardhii, Limnothrix redekei) and algae (Desmodesmus quadricauda), and examine possible relationships between the observed effects and toxin and retinoic acid (RA) content in the extracts. The cytotoxic and genotoxic effects of the extracts were studied in the human hepatocellular carcinoma HepG2 cell line, using the MTT assay, and the comet and cytokinesis-block micronucleus (cytome) assays, respectively. Liquid chromatography electrospray ionization mass spectrometry (LC/ESI-MS) was used to detect toxins (microcystins (MC-LR, MC-RR, MC-YR) and cylindrospermopsin) and RAs (ATRA and 9cis-RA) in the extracts. Six out of eight extracts were cytotoxic (0.04-2 mgDM/mL), and five induced DNA strand breaks at non-cytotoxic concentrations (0.2-2 mgDM/mL). The extracts with genotoxic activity also had the highest content of RAs and there was a linear association between RA content and genotoxicity, indicating their possible involvement; however further research is needed to identify and confirm the compounds involved and to elucidate possible genotoxic effects of RAs.
Microalgal food supplements from the perspective of Polish consumers: patterns of use, adverse events, and beneficial effects

Abstract:Microalgal food supplements are becoming increasingly popular due to their promising biological effects and high nutritional value, evidenced in in vitro, in vivo, and human studies. Some products of this kind have, however, raised controversies concerning their safety. At the same time, not much is known about the frequency of adverse events following the use of microalgal supplements, potential factors that may influence them, and general characteristics and behaviours of the consumer group. The present study aimed to fill this gap and surveyed a group of Polish consumers of microalgal products (n = 150) using an online questionnaire. As found, microalgal supplements (Spirulina, Chlorella, and Aphanizomenon) were popular in groups representing lacto-ovo-vegetarianism and veganism and were consumed predominantly for nutritional, immune-boosting, and detoxifying purposes. Their use was rarely discussed with specialists; the Internet constituted the most important source of information regarding these supplements. The most frequently self-reported health-beneficial effects of supplementation included the following: increased immunity, higher vitality, improved hair and skin quality, and better general well-being. Diarrhoea, nausea, abdominal pain, and skin rash were among the most often reported adverse events. Pre-existing medical conditions, namely renal failure and hypothyroidism, but not Hashimoto’s thyroiditis, were associated with increased occurrence of side effects. Those individuals who had consulted specialists as to the use of supplements reported adverse events significantly less often. A strikingly high frequency of side effects and very low consumer satisfaction were reported by a group of consumers supplementing Aphanizomenon-based products. In summary, the present study highlights that microalgal consumers may benefit from additional warnings of potential side effects and from consulting a qualified health specialist prior to use.

Abstract:Microcystins release from bloom-forming cyanobacteria is considered a way to gain competitive advantage in Microcystis populations, which threaten water resources security and aquatic ecological balance. However, the effects of microcystins on microalgae are still largely unclear. Through simulated culture experiments and the use of UHPLC-MS-based metabolomics, the effects of two microcystin-LR (MC-LR) concentrations (400 and 1,600 μg/L) on the growth and antioxidant properties of three algae species, the toxic Microcystis aeruginosa, a non-toxic Microcystis sp., and Chlorella vulgaris, were studied. The MC-LR caused damage to the photosynthetic system and activated the protective mechanism of the photosynthetic system by decreasing the chlorophyll-a and carotenoid concentrations. Microcystins triggered oxidative stress in C. vulgaris, which was the most sensitive algae species studied, and secreted more glycolipids into the extracellular compartment, thereby destroying its cell structure. However, C. vulgaris eliminated reactive oxygen species (ROS) by secreting terpenoids, thereby resisting oxidative stress. In addition, two metabolic pathways, the vitamin B6 and the sphingolipid pathways, of C. vulgaris were significantly disturbed by microcystins, contributing to cell membrane and mitochondrial damage. Thus, both the low (400 μg/L) and the high (1,600 μg/L) MC-LR concentration inhibited algae growth within 3 to 7 days, and the inhibition rates increased with the increase in the MC-LR concentration. The above results indicate that the toxin-producing Microcystis species have a stronger toxin tolerance under longer-term toxin exposure in natural water environments. Thus, microcystins participates in interspecific interaction and phytoplankton population regulation and creates suitable conditions for the toxin-producing M. aeruginosa to become the dominant species in algae blooms.
Improved method for the determination of anatoxin-a and two of its metabolites in blue-green algae using liquid chromatography with fluorescence detection

Abstract:Anatoxin-a, a neurotoxin produced by blue-green algae (BGA) species, can cause death to exposed organisms. In North America, BGA are harvested and sold as food supplements, some of which contain elevated levels of other algal toxins, such as microcystins. Concern that elevated levels of anatoxin-a also may be present in BGA food supplements has led to the development of a simple method to determine the presence of anatoxin-a in BGA. Some researchers have successfully analyzed this compound using liquid chromatography with fluorescence detection by forming a fluorescent derivative with 4-fluoro-7-nitrobenzofurazan (NBD-F) in water and phytoplankton extracts. With this method, the background noise is high in BGA extracts due to the presence of co-extractives. Addition of o-phthaldialdehyde (OPA) and mercaptoethanol to the extract before addition of the NBD-F resulted in the successful removal of primary amines from the background noise when the NBD-F derivatives were detected with fluorescence. Improved chromatograms were obtained when extracts were cleaned up in this manner, leading to a lower detection limit (approximately 50 microg/kg) for anatoxin-a. The detection limits obtained for the 2 degradation products dihydroanatoxin-a and epoxyanatoxin-a in BGA extracts were similarly low (55 and 65 microg/kg, respectively).

Abstract:Harmful algal blooms (HABs) formed by toxic microalgae have seriously threatened marine ecosystems and food safety and security in recent years. Among them, Karenia selliformis has attracted the attention of scientists and society due to its acute and rapid neurotoxicity in mice. Herein, the growth and gymnodimine A (GYM-A) production of K. selliformis were investigated in diverse culture systems with different surface-to-volume (S/V) ratios and nitrogen/phosphorus concentrations. The results showed that the specific growth rates (μ), maximal cell yields, and GYM-A production levels of K. selliformis increased with higher S/V, but no significant differences were observed under different culture volumes with the same S/V, which indicated that light penetration and gas exchange in the seawater culture systems actually influenced the growth and GYM-A production of K. selliformis. The maximum cell density and photosynthetic efficiency of K. selliformis decreased under nitrogen (N) and phosphorus (P) deficiency, suggesting that the growth of K. selliformis was significantly inhibited by the deficiency in N or P. Both N and P limitation conditions, especially P deficiency, promoted the cellular GYM-A quotas of K. selliformis. In this study, a scientific basis is provided for understanding the effects of culture systems and nutrient concentrations on the growth and toxin production of K. selliformis.

Abstract:Agriculture runoffs and discharge of wastewaters are the major causes of eutrophication. Although eutrophication could promote the thriving of any phytoplankter, frequently harmful algal blooms (HABs) are dominated by cyanobacteria. Currently, HABs dominated by the toxigenic cyanobacterium Microcystis aeruginosa in lakes and reservoirs are the main environmental concerns worldwide. This study was aimed to determine how M. aeruginosa (Ma) modifies the population growth of Pseudokirchneriella subcapitata (Ps) and Ankistrodesmus falcatus (Af). Growth kinetics were determined for each species and in the combinations: Ps‐Ma, Af‐Ma, Af‐Ps, and Ps‐Af‐Ma. At the end of experiments, photosynthetic pigments, phycobiliproteins, and microcystins were quantified. The Logistic equation significantly described the growth trend for all the tested species, enabling the identification of negative effects since the first stages in the population growth in co‐cultures with the cyanobacterium; in addition, the interaction effects in the growth rate and in the maximum attainable population density were determined. The biomass of A. falcatus, and P. subcapitata was significantly higher when cultured individually than in all the combinations with the cyanobacterium. The concentration of chlorophyll a and b, as well as carotenoids, was lower in combined cultures, but phycobiliproteins content in the cultures with M. aeruginosa was not significantly affected. M. aeruginosa negatively affected the growth of microalgae, but A. falcatus was significantly more inhibited than P. subcapitata; however, microcystins concentration was significantly reduced in the co‐cultures with microalgae. Results could help to explain the displacements of microalgae when cyanobacteria are present, giving place to cyanobacterial blooms in eutrophic freshwaters.

Abstract:The microalgae Arthrospira platensis (AP), commonly known as Spirulina, has gained widespread popularity as a food supplement in recent years. AP is particularly abundant in protein, B vitamins, iron, magnesium, potassium, and various antioxidants. In this study we aimed to evaluate the effect of nitrate limitation in the AP culture medium on AP growth and composition. In addition, the cytotoxicity of the respective aqueous AP extracts on three different mammalian cell-lines (HepG2, Caco2, L929) was tested. AP was cultivated over a 10-day period under nitrogen-rich (Nrich: 1.8 g/L) and nitrogen-deficient (Nlimited: 0.2-0.4 g/L) conditions in two separate experiments, each with three biological replicates (three bioreactors). Throughout the cultivation, the kinetic progress of dry biomass, pH, pigment content, the levels of essential elements (sulphur, phosphate, and nitrate) and the composition of elements in the harvested biomass was determined. While the biomass slightly but significantly differed, the phycocyanin concentration differed considerably (around 10-fold higher in the Nrich medium, p < 0.05). Aqueous extracts of the Nrich medium had significantly stronger effects on the cell membrane integrity and the metabolic activity of the cells than extracts of the Nlimited medium. Particularly was the finding that AP had a significantly stronger toxic effect on the two tumour cell types (HepG2, Caco2) than on the non-tumour cells (L929). This study underscores the significance of nitrate content in the cultivation media of AP.

Abstract:Macrophytic allelochemicals are considered an environment-friendly and promising alternative to control algal bloom. However, studies examining the potential mechanisms of inhibitory allelochemicals on algae are few. The allelochemical ethyl 2-methyl acetoacetate (EMA), isolated from reed (Phragmites communis), was a strong allelopathic inhibitor on the growth of Microcystis aeruginosa. EMA-induced antioxidant responses were investigated in the cyanobacterium M. aeruginosa to understand the mechanism of EMA inhibition on algal growth. The activities of enzymatic antioxidants superoxide dismutase (SOD) and catalase (CAT), and the contents of non-enzymatic antioxidants reduced glutathione (GSH) and ascorbic acid (AsA) of M. aeruginosa cells were analyzed after treatments with different concentrations of EMA. Exposure of M. aeruginosa to EMA caused changes in enzyme activities and contents of non-enzymatic antioxidants in different manners. The decrease in SOD activity occurred first after 4h of EMA exposure, and more markedly after 40h. CAT activity did not change after 4h of EMA exposure, but increased obviously after 40h. The contents of AsA and GSH were increased greatly by EMA after 4h. After 60h, low EMA concentrations still increased the CAT activity and the contents of AsA and GSH, but high EMA concentrations started to impose a marked suppression on them. EMA increased dehydroascorbate (DHAsA) and oxidized glutathione (GSSG) contents during all exposure times. After 60h, the regeneration rates of AsA and GSH (represented by the AsA/DHAsA ratio and GSH/GSSG ratio, respectively) were reduced by high EMA concentrations. These results suggest that the activation of CAT and the availability of AsA and GSH at early exposure are important to counteract the oxidative stress induced by EMA, and the inactivation of SOD may be crucial to the growth inhibition of M. aeruginosa by EMA.

Abstract:Some well-known hazards of blooming cyanobacteria are caused by toxic metabolites such as microcystins (MCs), though many other bioactive chemicals of unknown toxicity are present in their exudates. It is also unclear whether toxicity of cyanobacterial cells depends on growth phases in the life cycle. In this study, we compared toxicity to Daphnia magna of Microcystis aeruginosa - a common cyanobacterial species - exudates (MaE) from two MC-producing strains over both exponential growth and stationary phases in acute and chronic experiments. Specifically, we assessed mitochondrial dysfunction, oxidative stress and lipid peroxidation, and filtering activity and heartbeat rate of Daphnia exposed to MaE. All MaE treatments induced common characteristics of Microcystis toxicity including disorder in the mitochondrial membrane and aberrant heart rate. MaE from cells at stationary growth phase were more toxic than those at exponential phase. Surprisingly, the MC-lower strain had higher toxicity than MC-higher one. Microcystis at different stage of blooms may differentially affect waterfleas owing to variable MaE-induced physiological dysfunction, abundance and grazing rate. Our study suggested that Microcystis strains with lower microcystin-producing ability might release other detrimental chemicals and should not be ignored in harmful bloom monitoring.

Analytical screening of marine algal toxins for seafood safety assessment in a protected Mediterranean shallow water environment

Abstract:Microalgal species growing in marine and aquaculture environments can be responsible for harmful events because of their ability to produce potent natural toxins that can accumulate in edible mollusc species. Their consumption can cause severe illness and even be lethal. The European Union provides comprehensive regulations covering various general food safety aspects to manage the risk of contamination in shellfish farms. Many analytical methods have been proposed to evaluate algal toxins presence in the environment and in food products, for conducting surveillance studies of the main molluscs production sites and, where necessary, immediate monitoring of possible contamination of shellfish. In this work, a one-year analytical surveillance study was carried out to verify the possible presence of algal biotoxins in molluscs from a Mediterranean breeding area. Water and molluscs were sampled from a district of the North-East coast of Sicily, consisting of a unique brackish ecosystem of two lakes connected to each other and to the sea by narrow canals. Water samples were collected to investigate phytoplankton i by microscope analysis to assess the presence of potentially toxin-producing species, such as Pseudo-nitzschia spp, Alexandrium spp and Gonyaulax spinifera, although the presence of toxic phytoplankton has never reached alert levels. Mussels and clams samples were submitted to analysis of paralytic shellfish poisoning toxins, amnesic shellfish poisoning toxins and lipophilic toxins by liquid chromatography-based methods Only a few yessotoxins were detected, having concentrations always below the regulation limits. An existing liquid chromatography-tandem mass spectrometry-based multiresidue method for lipophilic biotoxins was adopted and extended to cover emerging biotoxins such as cyclic imines. The performance of the analytical method for Gymnodimine A and Spirolide 13-desMeC was assessed, obtaining respective quantitation limits of 20 and 10 µg kg−1, a precision always lower than 13% and trueness in the 81–120% range. Method applicability was confirmed using certified materials and a naturally contaminated sample.

Abstract:It has been found that algae have a variety of health benefits, although investigations showed that they contain radiotoxic elements, including 226Ra, 232Th, 40K, and 137Cs, which may affect human health. This study is connected to activity concentration measurements of the above radionuclides in the algae supplements available in the Middle East markets. The annual effective radiation doses of measured radionuclides in analyzed algal supplements have been calculated. The highest values of annual effective doses have been estimated for 226Ra in Ecklonia (13.39 μSv/y) and for 232Th in Red Marine Algae (11.80 μSv/y), both from South Korea. In algal "superfoods", the effective dose of 137Cs is not significantly affected by the Fukushima Nuclear Power Plant. Based on these results, it can be concluded that the activity levels of radionuclides are low, the naturally occurring radionuclides provide the most effective doses, and algae supplements can be considered safe.

Abstract:Eutrophication leads a mass of algae, and algae bloom directly influence the ecological function of water bodies and the safety of drinking water. It is very important to control the growth and reproduction of blooming algae for fishery, industrial and agricultural production as well as for the health of humans. Although some methods have been proposed to control harmful algal growth, only a few are applicable due to high cost, secondary pollution, or impracticability. The commonly used algicides, such as copper (Cu2+) and organic herbicide, can inhibit the entire phytoplankton community and cause subsequent water quality deterioration that may stress or kill aquatic animals. Furthermore, copper might accumulate in the bodies of aquatic predators, and herbicides might be toxic to all the living species in the water body. Therefore, it is necessary to explore an effective and secure new method to control the growth of harmful algae. Submerged macrophytes can reduce harmful algae blooms to maintain clear water in natural and experimental aquatic ecosystems. Using allelochemicals, which were produced by some macrophytes, to inhibit the growth of some harmful algae has been reported, but the allelopathic effects of alkaloids from submerged macrophytes were not found. In this study, the alkaloids of Elodea nuttallii were extracted by ethanol and then fractionated with some chemical reagent. The alkaloids compounds were analyzed by gas chromatography-mass spectrometry (GC-MS). Mass fragments of the components were compared to the mass fragmentation data contained in the NIST 02. 9 alkaloids were identified from Elodea nuttallii by GC-MS, and the allelopathic effects of the alkaloids were also explored by bioassay of Microcystin aeruginosa. The alkaloids from Elodea nuttallii inhibited obviously to the growth of M. aeruginosa with the inhibition rate of 44.0% for three days, when the concentration of alkaloids were 62.0 mg/L. The alkaloids were a complicated mixture, which further purification was a challenging job. The detailed components of alkaloids were not clear now, but the result of bioassay showed that there were active constituents in alkaloids which provide a useful information for further study of the allelochemical in submerged macrophytes.
