Abstract:Acrylamide (AA) is a hazardous chemical that is widely used in industrial practices. Spirulina platensis (SP) is a blue green alga that is rich in bioactive compounds with many medicinal benefits. The aim of the present study was to evaluate the ameliorative effect of SP against AA toxicity in rats. Animals were divided into six groups: Group (1) was normal rats, groups (2) and (3) received SP at 500 and 1000 mg/kg BW orally respectively for 21 days, group (4) was administered 20 mg/kg BW AA daily for 14 days, while groups (5) and (6) were given orally SP at the same doses of groups (2) and (3), then AA at similar dose of group (4). Rats that received AA alone displayed markedly increased serum levels of liver enzymes (ALT, AST, and ALP), kidney function parameters (urea and creatinine), DNA damage marker (8-OHdG), and proinflammatory cytokines (IL-1β, IL-6, and TNF-α), compared to control rats. Furthermore, tissue analysis revealed marked increases in hepatic, renal, and brain MDA and NO, as well as marked reductions in the antioxidant biomarkers (GSH, GSH-Px, SOD, and CAT) in acrylamide-intoxicated rats. Spirulina ameliorated the alterations in serum biochemical parameters and reduced MDA and NO, as well as improved antioxidant biomarkers in AA-intoxicated rats in a dose-dependent manner. Our results show that SP has a powerful protective effect on serum biochemistry and liver, kidney, and brain antioxidant machinery in AA-intoxicated rats.

Abstract:The potential reprotoxicity of bifenthrin remains unclear if only the common clinical indicators of reproductive disease are examined. The present study aimed to investigate the efficacy of Spirulina platensis, a microalga rich in antioxidant compounds, against bifenthrin-induced testicular oxidative damage in male mice. At the first, we demonstrate that administration of bifenthrin resulted in a decline of testosterone level and in deterioration of sperm quality that was correlated with significant transcription changes of some specific mRNA and microRNA involved in cholesterol transport, testosterone synthesis, and spermatogenesis. At the biochemical level, we found that oxidative stress was obvious in the bifenthrin group, as evidenced by increase in malondialdehyde (MDA), protein carbonyls (PCO), reactive oxygen species (ROS), and nitrite oxide (NO) that was correlated with activation of genes related to mitochondrial apoptotic signal pathways. We then brought, for the first time to our knowledge, solid and complete experimental evidences that administration of mice with Spirulina extract was sufficient to protect against deleterious effects BF in testicular tissues by abrogating the change in antioxidant enzyme activities; the increase in MDA, PCO, and NO concentrations; and the altered expression level of miRNA and mRNA involved in spermatogenesis. We finally demonstrate that Spirulina restores the production of testosterone in mice as well as epididymal sperm viability and motility. These results suggest a potential antitoxic activity of Tunisian Spirulina deserving further attention.

Abstract:Allophylus edulis is used in folk medicine primarily for liver conditions such as hepatitis, liver cancer and liver cirrhosis. In vitro hepatoprotective activity was previously demonstrated. The aim of this work was to evaluate the antioxidant capacity and hepatoprotective effect of the ethanolic extract of A. edulis in mice. This was done by first determining the acute toxicity of the extract, evaluating the general behavior, and subsequently verifying the effect on paracetamol-induced toxic hepatitis in male mice. Additionally, the phytochemical profile was performed, and the content of total phenols and its total antioxidant capacity were quantified through the 2,2, azinobis-(3-ethylbenzothiazoline)-6 sulfonic acid radical cation (ABTS) method. The extract of A. edulis leaves did not demonstrate adverse effects up to 2000 mg/kg, p.o. Anthraquinones, flavonoids, triterpenoids, and tannins were detected. A high content of total phenolic compounds (TPC) was related to a high antioxidant capacity. Regarding the results of the biological tests, A. edulis did not affect the general behavior of the mice, and all doses tested decreased glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) activity, the main liver enzyme markers of hepatocellular damage. It is concluded that A. edulis has hepatoprotective activity, which could be related to its antioxidant activity. Protective and Therapeutic Effects of Lactic Acid Bacteria against Aflatoxin B1 Toxicity to Rat Organs

Abstract:Background: Aflatoxin (AF), a metabolite of Aspergillus flavus, is injurious to vital body organs. The bacterial defense against such mycotoxins has attracted significant attention. Lactic acid bacteria (LAB) are known to ameliorate AF toxicity. Methods: Thirty adult male rats were divided into six groups (five each) to perform the experiments. The control (Co) group was fed a basal diet and water. Each of the following periods lasted 21 days: the milk (MK) group orally received milk (500 μL); LAB suspension (500 μL) containing 107 cfu/mL was orally provided to the LAB group; AF (0.5 mg/kg) was orally given to the AF group; and a combination of AF and LAB was administered to the AF + LAB group. The AF/LAB group was initially given AF for 21 days, followed by LAB for the same period. Finally, the rats were dissected to retrieve blood and tissue samples for hematological, biochemical, and histological studies. Results: The results revealed a significant decrease in RBCs, lymphocytes, total proteins, eosinophil count, albumin, and uric acid, whereas the levels of WBCs, monocytes, neutrophils, creatinine, urea, aspartate aminotransferase, alkaline phosphatase, alanine aminotransferase, lactate dehydrogenase, and creatinine kinase significantly increased in the AF group in comparison to the control group. The histological examination of the AF group revealed necrosis and apoptosis of the kidney’s glomeruli and renal tubules, nuclei vacuolization and apoptosis of hepatocytes, congestion of the liver’s dilated portal vein, lymphoid depletion in the white pulp, localized hemorrhages, hemosiderin pigment deposition in the spleen, and vacuolization of seminiferous tubules with a complete loss of testis spermatogenic cells. Meanwhile, protective and therapeutic LAB administration in AF-treated rats improved the hematological, biochemical, and histological changes. Conclusions: The study revealed LAB-based amelioration to AFB1-induced disruptions of the kidney, liver, spleen, and testis by inhibiting tissue damage. The therapeutic effects of LAB were comparatively more pronounced than the protective effects.

Abstract:[reaction: see text] A novel and efficient procedure for the generation of the reactive indole-2,3-quinodimethane intermediates from the allenylanilines is described. The indole-2,3-quinodimethane intermediates were captured by several dienophiles to afford the corresponding tetrahydro- and dihydrocarbazole derivatives. This method is significantly different from the previously reported ones, which involve the 1,4-elimination or its related reactions of the indole derivatives that possess suitable substituents at both the C-2 and C-3 positions.

Abstract:Recently, the use of medicinal plants including effective therapeutic molecules has become of the highest priority in treating various diseases and toxicities. The aim of the present study was to undertake the beneficial effect of Rhamnus alaternus L. aqueous extract (RAAE) against aluminum chloride-induced sub-chronic hematotoxicity and renal oxidative damage in rats. DPPH free radical scavenging, β-carotene bleaching, ferric-reducing antioxidant power (FRAP), phenolic, flavonoid, and tannin contents were measured in RAAE. Twenty-four male rats were divided into four groups. The first group was used as controls, and the other three groups received daily orally 50-mg AlCl 3 /kg b. wt, 250-mg RAAE/kg b. wt, and AlC 3 plus RAAE, respectively, for 4 weeks. The findings indicated the presence of an important amount of total phenolic, flavonoids, and tannins and high-capacity antioxidant activity. The administration of AlCl 3 caused induction of hematotoxicity evidenced by a significant decrease in hematocrit (Ht), hemoglobin concentration (Hb), red blood cell count (RBC), mean corpuscular volume (MCV), and mean corpuscular hemoglobin concentration (MCHC). In addition, AlCL 3 led to nephrotoxicity and oxidative stress occurrence, which were revealed by an increase of urea, creatinine, and uric acid, depletion of reduced glutathione concentration, superoxide dismutase, catalase, and glutathione peroxidase activities along with an increased level of the malondialdehyde level. However, the supplementation of RAAE significantly restored the previous mentioned parameters approximately to their normal values. These results were identical with the histological observations. In conclusion, the results showed that RAAE had efficient antioxidant properties due to its richness of antioxidant compounds, which played an important role against AlCl 3 -induced sub-chronic hematotoxicity and oxidative nephrotoxicity.
Effects of alfa lipoic acid and coenzyme Q10 treatment on AFB1-induced oxidative, inflammatory, and DNA damages in rats

Abstract:Food contamination with Aflatoxin B1 (AFB1) is a worldwide concern that adversely affects animal and human health. The study aimed to evaluate the protective effect of alpha lipoic acid (ALA) and/or co-enzyme Q10 (CQ10) against the harmful effects of AFB1 on the liver and kidneys. Fifty-six mature male Wistar Albino rats (180-200 g) were divided into seven groups, each with eight rats: (1) saline was given as a control, (2) ALA (100 mg/kg bw/day) was given by stomach gavage for fifteen days, and (3) CQ10 (10 mg/kg bw/day) was given by stomach gavage for fifteen days. Group (4) orally given AFB1 (2.5 mg/kg bw) on days 12th and 14th, (5) received AFB1 and ALA, (6) received AFB1 and CQ10, and (7) received AFB1, ALA, and CQ10, as previously described in the ALA, CQ10, and AFB1 groups. After the exposure to AFB1, a significant increase in liver markers (AST, ALT, ALP, and LDH) and renal function tests (BUN and creatinine) was observed compared with the control. ALA and/or CQ10 significantly reduced enzymes of liver and renal functions, as compared with AFB1. AFB1 exposure threw off the balance between oxidants and antioxidants. Still, ALA and/or CQ10 made oxidative stress (MDA, NO, and 8-OHdG) much lower and antioxidant activities (GSH, GSH-Px, SOD, and CAT) much higher. When we used the two together, the activities matched the control levels. Interestingly, this study shows that ALA and CQ10 significantly lowered IL-1β, IL-6, and TNF-α levels compared to the control values when used together after AFB1 exposure caused robust inflammation. Some CQ10 treatment parameters significantly outperformed those of ALA. ALA and CQ10 together worked better than either one alone to protect against AFB1-induced toxicity in the hepatic and renal parenchyma in terms of reducing inflammation, preventing DNA damage, and fighting free radicals.

Abstract:Mycotoxins are unavoidable contaminants of animal and human feed and food respectively. This study was aimed to investigate the effect of Vitamin E, Vitamin C and Selenium Supplementation to drink water against aflatoxins B1 on the various serum parameters , liver tissue antioxidant and the associated pathological changes. Thirty five male albino rats weighing 120-150 g were used in the present study. Group1 (G1): rats served as control , Group2 (G2) orally administrated aflatoxin B1 dissolved in propylene glycol per Os as a dose of 50µg/kg body weight /week, for 10 weeks . Group3-7 : administrated with aflatoxin B1 50µg/rat/week for 10 weeks . These groups were treated as Group3 (G3):was supplemented with selenium (as sodium selenite was used as 4mg/l). Group4 (G4):was supplemented with vitamin E as 1000mg/l. Group5 (G5):was supplemented with vitamin E plus selenium. Group6 (G6): was supplemented with vitamin C as 1000mg/l. Group7 (G7):was supplemented with vitamin C plus selenium. The results showed significant elevation in the liver and kidney functions, alpha globulin and beta globulin and decrease in concentrations of serum total protein, albumin, and gamma globulin together with A/G ratio. Lipid peroxidase as MDA levels were shown after AFB1 treatment. pathological changes of liver in aflatoxicated rats was discussed briefly. It can be concluded that the hepatotoxicity induced by aflatoxin B1 seemed to be modulated effectively by the simultaneous use of antioxidants: vitamin E or C alone or incombination, with selenium. Moreover, they are more efficient modulating the biochemical alteration , liver antioxidant enzymatic system and pathological changes.

Abstract:Pharmacological support for the appropriate use of drugs is important. To promote such support, it is necessary to be involved in drug therapy from viewpoints different from those of physicians and nurses, using tools unique to pharmacists, pharmacologically discuss individual cases, and investigate the validity of prescriptions by accumulating data. In Chapter 1, digoxin is necessary to monitor therapy very closely. In addition, patients with an impaired renal dysfunction have a predisposition for developing digitalis toxicity. In clinical cases, digoxin and verapamil are often co-administered for heart rate control, and we have observed the serum trough level of beta-methyldigoxin to be elevated due to drug-interaction. We build upon our previous findings and generated a simple index for the adequate administration dosage of beta-methyldigoxin based on variable degrees of renal function and the serum trough level of beta-methyldigoxin. In Chapter 2, to investigate risk factors of postoperative infection following cardiac surgery, we conducted a retrospective analysis of two surgical procedures, off-pump coronary artery bypass grafting (OPCAB) and surgery for valvular heart disease (valve operation). After discussing the analysis results with the respective physicians, the dosing guidelines for cefazolin (CEZ) were changed. We also analyzed the rate of CEZ replacement with other antibiotics after surgery finding that it decreased in both groups for OPCAB and valve operations. From these results, we conclude that, if CEZ is also administered intra-operatively when surgery is prolonged, its administration for two days following surgery is adequate for prophylaxis against postoperative infection.
Fucoidan supplementation modulates hepato-renal oxidative stress and DNA damage induced by aflatoxin B1 intoxication in rats

Abstract:Aflatoxins are a common food contaminant of global concern. Aflatoxin B1 (AFB1) intoxication is associated with serious health hazards. Recently, fucoidan (FUC) has gained much attention from pharmaceutical industry due to its promising therapeutic effects. The impacts of FUC on AFB1-induced liver and kidney injures have not been sufficiently addressed. This research was conducted to evaluate the ameliorative effect of FUC in AFB1-induced hepatorenal toxicity model in rats over 14 days. Five groups were assigned; control, FUC (200 mg/kg/day, orally), AFB1 (50 μg/kg, i.p.), and AFB1 plus a low or high dose of FUC. AFB1 induced marked hepatorenal injury elucidated by substantial alterations in biochemical tests and histological pictures. The oxidative distress instigated by AFB1 enhanced production of malondialdehyde (MDA) and nitric oxide (NO) along with reduction in the reduced-glutathione (GSH), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and catalase (CAT) activities. DNA damage in the liver and kidney tissues has been demonstrated by overexpression of proliferating cell nuclear antigen (PCNA). Unambiguously, FUC consumption alleviates the AFB1-induced mitochondrial dysfunction, oxidative harm, and apoptosis. These ameliorated effects are proposed to be attributed to fucoidan's antioxidant and anti-apoptotic activities. Our results recommend FUC supplementation to food because it exerts both preventive and therapeutic effects against AFB1-induced toxicity.
Phytochemical screening, antioxidant activity of selected methanolic plant extracts and their detoxification capabilities against AFB1 toxicity

Abstract:Aflatoxin B1 (AFB1) is a secondary metabolite produced principally by Aspergillus parasiticus and A. flavus. It is one of the most potent and commonly occurring dietary carcinogen with its carcinogenic potential being linked to the formation of DNA adducts and reactive oxygen species (ROS). Plant extracts contain a plethora of biologically active phytochemicals that act against ROS. This study aimed to assess the phytochemical content and antioxidant activity of methanolic extracts of some medicinal plants and investigate their detoxification potentials against AFB1. Phytochemical screening together with total phenolic content (TPC), total flavonoid content (TFC), and antioxidant (2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS+)) assays) were performed on nine methanolic plant extracts. Extracts were incubated with AFB1 for 24 and 48 h and liquid chromatography mass spectrometry (LC-MS) analysis done to assess their AFB1 detoxification activities. The TPC of the extracts ranged from 88.92 ± 6.54 to 210.19 ± 7.90 mg GAE/g, while TFC ranged between 4.01 ± 0.94 and 32.48 ± 1.02 mg QE/g. Radical scavenging activities of extracts varied from 4.18 ± 1.37 to 251.53 ± 9.30 μg/mL and 8.36 ± 1.65 to 279.22 ± 8.33 μg/mL based on DPPH and ABTS+ assays, respectively. Six of the plant extracts showed a time-dependent detoxification activity against AFB1 after 48 h ranging from 20.17 to 38.13 %. C. dentata bark extract showed the highest percentage of AFB1 reduction, with mean percentages of 43.57 and 70.96 % at 24 and 48 h, respectively. This was followed by C. asiatica leaves and A. melegueta seeds with a maximum of 40.81 and 38.13 %, respectively after 48 h. These extracts also possessed high TPC, TFC, and antioxidant activities compared to all the other extracts. Findings from this study demonstrate the abundance of bioactive compounds with antioxidant activity playing a role in potent AFB1 detoxification activity.
Chemoprevention of rat liver toxicity and carcinogenesis by Spirulina

Abstract:Spirulina platensis (SP) is a filamentous cyanobacterium microalgae with potent dietary phyto-antioxidant, anti-inflammatory and anti-cancerous properties. The present study aimed to investigate the chemopreventive effect of SP against rat liver toxicity and carcinogenesis induced by dibutyl nitrosamine (DBN) precursors, and further characterized its underlying mechanisms of action in HepG2 cell line. Investigation by light and electron microscopy showed that DBN treatment induced severe liver injury and histopathological abnormalities, which were prevented by SP supplementation. The incidence of liver tumors was significantly reduced from 80 to 20% by SP. Immunohistochemical results indicated that both PCNA and p53 were highly expressed in the liver of DBN-treated rats, but were significantly reduced by SP supplementation. Molecular analysis indicated that SP treatment inhibited cell proliferation, which was accompanied by increased p21 and decreased Rb expression levels at 48hrs post-treatment. In addition, SP increased Bax and decreased Bcl-2 expression, indicating induction of apoptosis by 48hrs. This is the first report of the in vivo chemopreventive effect of SP against DBN-induced rat liver cytotoxicity and carcinogenesis, suggesting its potential use in chemoprevention of cancer.

Bacillus amyloliquefaciens B10 can alleviate liver apoptosis and oxidative stress induced by aflatoxin B1

Abstract:Aflatoxin B1 (AFB1) is a mycotoxin often found in food and livestock feed. It can affect human and animal health and is especially damaging to the liver. This study aims to evaluate whether Bacillus amyloliquefaciens (hereafter referred to as B. amyloliquefaciens) B10 can alleviate the toxic effects of AFB1 and, if so, what mechanism is responsible for its action. Specific pathogen-free (SPF) Kunming mice (5-6 weeks old) were divided into four groups (Control, AFB1, B10 strain, and AFB1 + B10 strain) and conducted continuously via gavage for 28 days. Oxidation indices (MDA, T-AOC, SOD, GSH-Px, and CAT) were then measured using their liver tissues and liver coefficient were calculated. Apoptosis was determined using the TUNEL method. Gene expression was determined for Bax, Bcl-2, BIP, CHOP, JNK, Caspase-12, Caspase-9, and Caspase-3, and protein expression was detected for Bax, Bcl-2, and Caspase-3. Our results showed that AFB1 induced the oxidative damage and apoptosis in the livers of mice. However, for mice given B. amyloliquefaciens B10, the biochemical indices, pathological changes, the expressions of genes and proteins related to oxidative stress and apoptosis were significantly reversed. The results indicate that B. amyloliquefaciens B10 antagonizes oxidative damage and apoptosis induced by AFB1 in the livers of mice. The results of this study are of significance for the future use of this strain to reduce the harm of AFB1 to human health and animal reproductive performance.
Acute aflatoxin B1-induced hepatic and cardiac oxidative damage in rats: Ameliorative effects of morin

Abstract:Aflatoxins (AFs) are secondary metabolites produced by the fungus Aspergillus flavus, of which Aflatoxin-B1 (AFB1) appears to be the most cancerogenic and of the highest toxicity. AFB1 causes serious effects on several organs including the liver. Morin is a flavonol that exists in many fruits and plants and has diverse biological properties including anticancer, anti-atherosclerotic, antioxidant, anti-inflammatory, immunomodulatory, and multi-organ protective activities. The present study aims to evaluate the potential protective effects of morin against acute AFB1-induced hepatic and cardiac toxicity in rats. Forty rats were divided into five groups (n = 8) as follows: control received the vehicle, morin was orally administered 30/mg/kg body weight (MRN30), the AFB1 was administered orally at a dose of 2.5 mg/kg, twice on days 12 and 14 of the experiment for the 3rd, 4th, and 5th groups., AFB1-MRN15 was orally given morin at a dose of 15 mg/kg body weight, and AFB1-MRN30 orally received morin at 30 mg/kg body weight. The results indicated a significant decrease in serum AST, ALP, LDH, GGT, CK, CK-MB, 8-OHdG, IL-1β, IL-6, TNF-a levels in MRN30 compared to AFB1, and AFB1-MRN15 groups. However, the results indicated non-significant differences in the serum levels between MRN30, control, and AFB1-MRN30 groups. Meanwhile, regarding the hepatic and cardiac parameters, there were significant differences in the levels of MDA, NO, GSH, GSH-Px, SOD, and CAT in MRN30 compared to AFB1, and AFB1-MRN15 groups, overall implying the protective effects of morin. To conclude, morin at a dose of 30 mg/kg b. wt. showed significant enhancements in acute AFB1-induced hepatic and cardiac toxicity in rats, which could play a role in limiting the public health hazards of AFs.

Abstract:The objective of this study was to detect the effects of acute aflatoxin B 1 (AFB 1 ) exposure in Nile tilapia ( Oreochromis niloticus ) and the effectiveness of Saccharomyces cerevisiae and silicate in reducing these effects. Two hundred and forty Nile tilapia fingerlings (16 ± 0.5 g) were randomly assigned to four experimental groups, each with 60 fish and three replicates. Control basal diet (Diet 1) and three test diets were formulated, where Diet 2 was supplemented with 200 ppb AFB 1 . Diets 3 and 4 were intoxicated with AFB 1 (200 ppb) and supplemented with 0.5% S. cerevisiae or 0.5%, respectively. After 60 days, Diet 1 had considerably greater growth characteristics than the other groups ( p < 0.05). Diet 2 revealed a reduced ( p < 0.05) survival rate after 1 month of exposure. In addition, Diet 1 showed higher ( p < 0.05) total protein and albumin levels than Diets 3 and 4. AFB 1 residues were detected in the liver in fish-fed Diet 2, Diet 4, and Diet 3. Alanine aminotransferase, aspartate aminotransferase, creatinine, and urea levels increased ( p < 0.05) in fish-fed Diet 2. The glutathione peroxidase, lysozyme, and catalase activity were decreased ( p < 0.05) in the fish-fed Diet 2. The malondialdehyde level was significantly higher in fish given Diet 2 ( p < 0.05) than in fish-fed Diets 3 and 4. Histopathological investigation of fish-fed Diet 2 revealed impaired liver and spleen; however, both treatments (Diets 3 and 4) successfully lowered inflammation and preserved liver and spleen integrities. In conclusion, AFB 1 impaired growth performance and posed a severe health risk to Nile tilapia. Furthermore, S. cerevisiae alleviated the contamination of AFB 1 effects more efficiently than silicate employed for toxin adsorption.

Abstract:* Abstract　Our primary concern in this review is to give possible explanations and pose new questions on the phenomena concerning eﬀ ects of neonatal estrogen exposure on the development and functions of reproductive organs and immune mechanisms particularly in male animals in the light of new ﬁ ndings and cencepts. At ﬁ rst our own experimental results on the pathological changes in the epididymis and testis in neonatally estrogenized mice have been presented. The results of our ﬁ rst experiment showed that neonatal single injection of 10 μg of estradiol benzoate induced marked inﬂ ammatory changes in the epididymis and testis in which the initial pathological change occurred concomitantly with the appearance of sperms in the epididymal duct. The second experiment showed that such epididymo‑testicular changes did not occur when the testis was removed or neonatal estrogen was followed by testosterone. From these results a possibility was suggested that a hormone‑related autoimmune mechanism might participate in such changes. Relating to these various reports concerning eﬀ ects of neonatal estrogen treatment either alone or with concommitent treatment with androgen on the development and functions of reproductive organs and immune mechanisms in the adolescent or adult have been reviewed. In addition advances in the related areas such as estrogen‑related mechanisms in the development and functions of reproductive organs and immune mechanisms including autoimmune disease mechanisms gender diﬀ erences and the role of CD25 + CD4 + T lymphocytes have also been reviewed brieﬂ y.

Abstract:ABSTRACT A total number of 35 Male albino rats were obtained from El Osman Farm, Cairo, Egypt. Animals were housed in stainless steel cages and provided with food and water ad libitum. All animals were healthy and clinically free of diseases. Rats were randomly divided into 5 equal groups, each group contained 7 rats and fed on one of the following diets: Group 1: Control diet, Group 2: Control diet + 1000ppm Aluminum, Group 3: Control diet+ 1000ppm+Garlic 5%, Group 4: Control diet+ 1000ppm + Nigella sativa 5%, Group 5: Control diet+ 1000ppm + Garlic 2.5 %+ Nigella sativa 2.5%. Blood samples were collected of each group after 30 and 60days from the beginning of the experiment. Blood samples were centrifuged at 3000 rpm for 15 min. and serum was separated and kept in frozen until subsequent analyses. And at the end of the experiment blood samples were collected too in EDTA tubes for Determination the hematological parameters. Finally, at the end of the experiment, rats were sacrificed to obtain the livers, and kidneys for examination it. The results indicated that Aluminum has adverse effects on animal and human health. The present study demonstrated that medicinal plants (garlic and nigella) in combination with aluminum minimized its hazards. Consequently, the exposure to aluminum should be reduced and attention paid to sources of aluminum in foods, water and personal care products. Furthermore, using diets rich in medicinal plants (garlic and nigella) could be beneficial in alleviating aluminum toxicity.

Abstract:Abstract The aim of this investigation was to evaluate the efficacy of Alisma orientalis methanolic extract (AOME) on the experimental nonalcoholic fatty liver disease (NAFLD) induced by high-fat diet. Rats were fed with high-fat diet for six weeks and then gavaged the AOME for another six weeks. Typical pathological symptoms of NAFLD occurred in the high-fat diet rats. Administration with the AOME (150,300 and 600 mg kg−1) markedly decreased the serum and liver lipids; the high level of fasting serum glucose was reduced and insulin resistance was improved. The AOME treatment was also helpful in preventing the oxidative stress by lessening lipid peroxidation and activating antioxidant enzymes. Markers of the liver injury, aminotransferase abnormalities and hepatomegaly were improved and morphological changes, such as liver steatosis, mixed inflammation and collagen deposition, were lessened in rats treated with the AOME. These results suggested that the AOME showed hepatoprotective effects on NAFLD and may be a potential clinical application for treatment of this chronic liver disease.

Abstract:Forty-two cases of pre-invasive and invasive cancer of the cervix diagnosed by histology were studied cytologically and cytogenetically. Preinvasive lesions showed a good correlation between cytologic impression and the histologic diagnosis. However, among 17 patients with invasive cancer, there were 2 completely negative cytologic impressions. These were in cases having modal chromosome counts of 47 and 48, respectively. This study demonstrates that some invasive cancers of the cervix are resistant to cytologic diagnosis because of their peridiploid chromosome counts and lack of anisokaryosis.

Abstract:The study aimed to conduct chemical profiling, acute in-vivo toxicity evaluation, and the potential anti-diabetic effect of standardized Aloe sabaea flowers ethanolic extracts (ASFEE) on alloxan-induced diabetic rats. The chemical composition was analyzed using GC-MS and TLC techniques. The oral acute toxicity study was performed according to the WHO 2000 and the OECD 420 guidelines. Furthermore, anti-diabetic activity was investigated using two doses of ASFEE (0.2 and 0.5 g/kg/day BW, p.o.) compared with glibenclamide (5 mg/kg/day, p.o.). A molecular docking investigation of the identified components with the PTPN9 enzyme was performed to figure out the proposed anti-diabetic mechanism. GC-MS analysis displayed the existence of 18 compounds; most of the compounds were fatty acids and their esters, and phytosterols. Total phenolic and flavonoid contents were 42.00±1.26 mg GAE/g DW and 22.21±1.55 mg QE/g DW, respectively. The results of the in-vivo toxicity study revealed the absence of noticeable signs of toxicity or mortality at various doses establishing the safety of the tested extract. The estimated LD50 value was higher than 10 g/kg. Antidiabetic action exhibited a noticeable decline in fasting blood glucose (FBG) levels comparable to glibenclamide with no inducing intense hypoglycemia and considerable excess weight.
