Abstract:For revealing the mechanism of allelopathic influence on phytoplankton by aquatic macrophytes, the growth and photosynthetic activities of cyanobacteria Microcystis aeruginosa and the chlorophyte Selenastrum capricornutum were investigated when they coexisted with submerged macrophyte Myriophyllum spicatum and were exposed to allelopathic polyphenols: pyrogallic acid (PA), gallic acid (GA), ellagic acid (EA) and (+)-catechin (CA). According to the results of coexistence assays, the non-photochemical quenching (NPQ) and effective quantum efficiency (YII) of M. aeruginosa were affected earlier and more rapidly than the cell density. However, the influence of M. spicatum on S. capricornutum was not found. When the Toxicity Index (TI) was applied to evaluate the combined effects of binary and multiple mixtures of polyphenols, it was found that the four tested polyphenols with the proportion identified in the M. spicatum-cultured solution were observed to present synergistic effect (0.36-0.49) according to the cell density, NPQ and YII of M. aeruginosa. With the combined effects of polyphenols on S. capricornutum, only additive action (0.52-1.62) was found. On the other hand, PA (2.97mgL(-1)), GA (2.65mgL(-1)) caused significant reductions of photosystem II (PSII) and whole electron transport chain activities of M. aeruginosa by 71.43 and 18.37%, 70.95 and 40.77% (P<0.05), respectively, after 24-h exposure, but no inhibition effect was found in S. capricornutum. The dark respiration and photosystem I (PSI) activities of M. aeruginosa were significantly increased by exposure to PA and GA (P<0.05). Nevertheless, EA and CA had no influence on the electron transport activities of the tested organisms. These results indicate that the reduction in photosynthetic activity of M. aeruginosa and the synergistic effect of allelochemicals may be two important causes for the inhibition of undesired phytoplankton by submersed macrophytes in natural aquatic ecosystems, and PSII in cyanobacteria is considered to be one of the target sites attacked by allelopathic polyphenols.

Abstract:Gramine, sanguinarine and berberine are potential algicides that can significantly inhibit the growth of cyanobacteria. Their effects on other phytoplankton have to be considered, because other phytoplankton species and cyanobacteria usually co-exist. In this study, we have established the relationships between phytoplankton biomasses and their marker pigments. Afterwards, we have assessed the allelopathic effects of three algicides (gramine, sanguinarine and berberine) on the mono-cultured and mixed-cultured cyanobacteria Microcystis and green algae Chlorella . Our results showed that zeaxanthine and lutein can be used to determine the cell densities of Microcystis and Chlorella in the mixed cultures, respectively. Our study also demonstrated that the inhibitory effects of these three allelopathic algicides on the growths of both Microcystis and Chlorella were sanguinarine > gramine > berberine. The concentrations for 50% of the maximal effect (EC 50 ) of sanguinarine against Microcystis and Chlorella were 0.065 and 0.09 mg L −1 , respectively; the EC 50 values of gramine against Microcystis and Chlorella were 1.687 and 0.870 mg L −1 , respectively; and the EC 50 values of berberine against Microcystis and Chlorella were 1.969 and 2.397, respectively. The inhibitory effect of allelopathic algicides on the multiple phytoplankton species in mixed cultures was significantly greater than that in monocultures. In mixed cultures without allelopathic addition or in cultures with gramine (0.5–3 mg L −1 ), the dominant species was Microcystis . However, Chlorella became the dominant species when the concentration of sanguinarine was 0.1 mg L −1 or that of berberine was greater than 0.5 mg L −1 . Both species of phytoplankton died completely when sanguinarine levels were greater than 0.1 mg L −1 . Our results suggested that the allelopathic algicides sanguinarine and berberine reduced the dominance of Microcystis in competition with Chlorella .

Taboo: working memory and mental control in an interactive task.

Abstract:Individual differences in working memory (WM) predict principled variation in tasks of reasoning, response time, memory, and other abilities. Theoretically, a central function of WM is keeping task-relevant information easily accessible while suppressing irrelevant information. The present experiment was a novel study of mental control, using performance in the game Taboo as a measure. We tested effects of WM capacity on several indices, including perseveration errors (repeating previous guesses or clues) and taboo errors (saying at least part of a taboo or target word). By most measures, high-span participants were superior to low-span participants: High-spans were better at guessing answers, better at encouraging correct guesses from teammates, and less likely to either repeat themselves or produce taboo clues. Differences in taboo errors occurred only in an easy control condition. The results suggest that WM capacity predicts behavior in tasks requiring mental control, extending this finding to an interactive group setting.

Abstract:Freshwater eutrophication is an increasing ecological and public health concern. One of the major consequences of eutrophication is the formation of harmful algal blooms (HABs), a rapid population increase and accumulation of microorganisms, namely algae and cyanobacteria. In this project, we aim to investigate a nature based solution to this environmental hazard. Our approach consisted in the evaluation of the potential of aquatic plants to control the growth and viability of Chrysosporum ovalisporum, a cyanobacteria species associated with HABs and that produces toxic metabolites such as cylindrospermopsin. First, we carried out a screening with different plant species – Iris pseudacoros , Typha latifolia , Sparganium erectum , Alisma plantago-aquatica , Alisma lanceolatum , Nasturtium officinale , Landoltia punctata , Lemna minor and Wolffia arrhiza – for the allelopathic effect of their extracts on this cyanobacteria. For this purpose, methanol/water (70:30, v:v) mixture extracts were prepared and tested, at various concentrations, in C. ovalisporum cultures. Exposure to L. punctata extracts at the highest concentration tested (2 mg L −1 ) showed significant cellular abundance inhibitory effects on C. ovalisporum . Secondly, a microcosm system was assembled to evaluate the effects of co-growth of C. ovalisporum and L. punctata . In this experiment, we tested the effect of the presence of this plant on C. ovalisporum cell abundance, under a nutritional surplus and equivalent cyanobacterial light exposure in between test and control groups. The results confirmed the allelopathic effect of L. punctata on C. ovalisporum . This work demonstrates that the allelopathic properties of aquatic plants could be exploited to control the growth of harmful algae, and could be used to develop novel, eco-friendly and practical solutions, for treatment of water contaminated with C. ovalisporum .

Abstract:Enhanced osmotic water permeability has been observed in Xenopus oocytes expressing cystic fibrosis transmembrane conductance regulator (CFTR) protein. Subsequent studies have shown that CFTR activates an endogenous water permeability in oocytes, but that CFTR itself is not the water channel. Here, we show CFTR-dependent activation of endogenous water permeability in normal but not in cystic fibrosis human airway epithelial cells. Cell volume was measured by novel confocal x-z laser scanning microscopy. Glycerol uptake and antisense studies suggest CFTR-dependent regulation of aquaporin 3 (AQP3) water channels in airway epithelial cells. Regulatory interaction was confirmed by coexpression of CFTR and AQP3 cloned from human airways inXenopus oocytes and of CFTR and rat AQP3 in Chinese hamster ovary cells. These findings indicate that CFTR is a regulator of AQP3 in airway epithelial cells.
Effects of Microcystin-Lr on the Metal Bioaccumulation and Toxicity in Chlamydomonas Reinhardtii.

Abstract:Microcystin-LR (MC-LR) is one of the most notorious toxins liberated from cyanobacteria in eutrophicated freshwater ecosystems. Its effects on the bioaccumulation and toxicity of Cd2+, CrO42-, Cu2+, and Zn2+ in a green alga Chlamydomonas reinhardtii were investigated in the present study. The metal bioaccumulation in the alga was unaffected by MC-LR. The surface-adsorbed and intracellular metal concentrations in the treatments with and without the addition of MC-LR could be well simulated by a single Freundlich isotherm for each metal with their accumulation ability following the order of Cu2+ > Cd2+ > Zn2+ > CrO42-. The bioavailable metal concentrations measured by diffusion gradients in thin-films remained unchanged when MC-LR was applied. Accordingly, the growth of C. reinhardtii was similarly inhibited at the same metal concentration regardless of the addition of MC-LR. The metal toxicity could also be well delineated with the classic free ion activity and biotic ligand models. However, the intracellular metal concentration was found to have the best predictability suggesting its more direct relationship with metal toxicity. Metal exposure induced the accumulation of MC-LR in the alga, which was leveled off at high metal levels. The underlying uptake mechanisms need to be further examined. (C) 2011 Elsevier Ltd. Abstract:Agriculture runoffs and discharge of wastewaters are the major causes of eutrophication. Although eutrophication could promote the thriving of any phytoplankter, frequently harmful algal blooms (HABs) are dominated by cyanobacteria. Currently, HABs dominated by the toxigenic cyanobacterium Microcystis aeruginosa in lakes and reservoirs are the main environmental concerns worldwide. This study was aimed to determine how M. aeruginosa (Ma) modifies the population growth of Pseudokirchneriella subcapitata (Ps) and Ankistrodesmus falcatus (Af). Growth kinetics were determined for each species and in the combinations: Ps‐Ma, Af‐Ma, Af‐Ps, and Ps‐Af‐Ma. At the end of experiments, photosynthetic pigments, phycobiliproteins, and microcystins were quantified. The Logistic equation significantly described the growth trend for all the tested species, enabling the identification of negative effects since the first stages in the population growth in co‐cultures with the cyanobacterium; in addition, the interaction effects in the growth rate and in the maximum attainable population density were determined. The biomass of A. falcatus, and P. subcapitata was significantly higher when cultured individually than in all the combinations with the cyanobacterium. The concentration of chlorophyll a and b, as well as carotenoids, was lower in combined cultures, but phycobiliproteins content in the cultures with M. aeruginosa was not significantly affected. M. aeruginosa negatively affected the growth of microalgae, but A. falcatus was significantly more inhibited than P. subcapitata; however, microcystins concentration was significantly reduced in the co‐cultures with microalgae. Results could help to explain the displacements of microalgae when cyanobacteria are present, giving place to cyanobacterial blooms in eutrophic freshwaters.

Abstract:Ubiquitous distribution of pharmaceutical contaminants in environment has caused unexpected adverse effects on ecological organisms; however, how microorganisms recover from their toxicities remains largely unknown. In this study, we comprehensively investigated the effect of a representative pollutant, doxylamine (DOX) on a freshwater microalgal species, Chlorella sp. by analyzing the growth patterns, biochemical changes (total chlorophyll, carotenoid, carbohydrate, protein, and antioxidant enzymes), and transcriptomics. We found toxicity of DOX on Chlorella sp. was mainly caused by disrupting synthesis of ribosomes in nucleolus, and r/t RNA binding and processing. Intriguingly, additional bicarbonate enhanced the toxicity of DOX with decreasing the half-maximum effective concentrations from 15.34 mg L-1 to 4.63 mg L-1, which can be caused by inhibiting fatty acid oxidation and amino acid metabolism. Microalgal cells can recover from this stress via upregulating antioxidant enzymatic activities to neutralize oxidative stresses, and photosynthetic pathways and nitrogen metabolism to supply more energies and cellular signaling molecules. This study extended our understanding on how microalgae can recover from chemical toxicity, and also emphasized the effect of environmental factors on the toxicity of these contaminants on aquatic microorganisms.

Abstract:Increasing toxic cyanobacterial blooms in freshwater demand environmentally friendly solutions to control their growth and toxicity, especially in arid countries, where most drinking water is produced from surface reservoirs. We tested the effects of macrophyte allelochemicals on Microcystis aeruginosa and on the fundamental role of bacteria in nutrient recycling. The effects of Ranunculus aquatilis aqueous extract, the most bioactive of four Moroccan macrophyte extracts, were tested in batch systems on M. aeruginosa growth, toxin production and oxidative stress response and on the ectoenzymatic activity associated with the bacterial community. M. aeruginosa density was reduced by 82.18%, and a significant increase in oxidative stress markers was evidenced in cyanobacterial cells. Microcystin concentration significantly decreased, and they were detected only intracellularly, an important aspect in managing toxic blooms. R. aquatilis extract had no negative effects on associated bacteria. These results confirm a promising use of macrophyte extracts, but they cannot be generalized. The use of the extract on other toxic strains, such as Planktothrix rubescens, Raphidiopsis raciborskii and Chrysosporum ovalisporum, caused a reduction in growth rate but not in cyanotoxin content, increasing toxicity. The need to assess species-specific cyanobacteria responses to verify the efficacy and safety of the extracts for human health and the environment is highlighted.

Chitosan as a Coagulant to Remove Cyanobacteria Can Cause Microcystin Release

Abstract:Chitosan has been tested as a coagulant to remove cyanobacterial nuisance. While its coagulation efficiency is well studied, little is known about its effect on the viability of the cyanobacterial cells. This study aimed to test eight strains of the most frequent bloom-forming cyanobacterium, Microcystis aeruginosa, exposed to a realistic concentration range of chitosan used in lake restoration management (0 to 8 mg chitosan L-1). We found that after 1 h of contact with chitosan, in seven of the eight strains tested, photosystem II efficiency was decreased, and after 24 h, all the strains tested were affected. EC50 values varied from 0.47 to > 8 mg chitosan L-1 between the strains, which might be related to the amount of extracellular polymeric substances. Nucleic acid staining (Sytox-Green®) illustrated the loss of membrane integrity in all the strains tested, and subsequent leakage of pigments was observed, as well as the release of intracellular microcystin. Our results indicate that strain variability hampers generalization about species response to chitosan exposure. Hence, when used as a coagulant to manage cyanobacterial nuisance, chitosan should be first tested on the natural site-specific biota on cyanobacteria removal efficiency, as well as on cell integrity aspects.
Microcystin-LR (MC-LR) inhibits green algae growth by regulating antioxidant and photosynthetic systems

Abstract:Microcystins release from bloom-forming cyanobacteria is considered a way to gain competitive advantage in Microcystis populations, which threaten water resources security and aquatic ecological balance. However, the effects of microcystins on microalgae are still largely unclear. Through simulated culture experiments and the use of UHPLC-MS-based metabolomics, the effects of two microcystin-LR (MC-LR) concentrations (400 and 1,600 μg/L) on the growth and antioxidant properties of three algae species, the toxic Microcystis aeruginosa, a non-toxic Microcystis sp., and Chlorella vulgaris, were studied. The MC-LR caused damage to the photosynthetic system and activated the protective mechanism of the photosynthetic system by decreasing the chlorophyll-a and carotenoid concentrations. Microcystins triggered oxidative stress in C. vulgaris, which was the most sensitive algae species studied, and secreted more glycolipids into the extracellular compartment, thereby destroying its cell structure. However, C. vulgaris eliminated reactive oxygen species (ROS) by secreting terpenoids, thereby resisting oxidative stress. In addition, two metabolic pathways, the vitamin B6 and the sphingolipid pathways, of C. vulgaris were significantly disturbed by microcystins, contributing to cell membrane and mitochondrial damage. Thus, both the low (400 μg/L) and the high (1,600 μg/L) MC-LR concentration inhibited algae growth within 3 to 7 days, and the inhibition rates increased with the increase in the MC-LR concentration. The above results indicate that the toxin-producing Microcystis species have a stronger toxin tolerance under longer-term toxin exposure in natural water environments. Thus, microcystins participates in interspecific interaction and phytoplankton population regulation and creates suitable conditions for the toxin-producing M. aeruginosa to become the dominant species in algae blooms.
The Detrimental Effect of High Salinity on the Growth and Microcystins Contamination of Microcystis Aeruginosa

Abstract:The occurrence of cyanobacterial species, especially toxic ones, poses a great threat to coastal and estuary areas. In this study, the toxigenic Microcystis aeruginosa (M. aeruginosa) FACHB-905 was exposed to BG-11 medium with different salinities (1, 4, 7, 10 and 15 ppt) to investigate the physiological responses of this species in terms of oxidative stress, chl a fluorescence and microcystins (MCs) contamination. The results showed that low salinity (≤7 ppt) favored the electron transfer of photosystem II, which promoted the growth and photosynthesis of M. aeruginosa and induced MCs production. However, increased salinity (≥10 ppt) suppressed the growth and photosynthesis of M. aeruginosa and aggravated the oxidative stress of the strain. Salinity of 15 ppt reduced MCs contamination and caused irreversible damage to the photosynthetic system of M. aeruginosa, leading to the lysis and death of algal cells. These results indicated that changes in salinity exerted important regulations on the growth and MCs contamination of the toxic M. aeruginosa, which may provide a reference for the risk assessment of the harmful cyanobacterial species in the coastal and estuary areas.

Abstract:Cyanobacterial blooms have become more frequent and serious in recent years. Not only do massive blooms cause environmental pollution and nutrient eutrophication, but they also produce microcystins (MCs), a group of toxic cycloheptapeptides, which threaten aquatic ecosystem and human health. As such, clarifying the allelopathic interactions between cyanobacteria and other algae is critical to better understand the driving factors of blooms. To date, however, such studies remain largely insufficient. Here, we treated model alga Chlamydomonas reinhardtii with microcystin-LR (MC-LR) to determine its allelopathic effects. Results showed that MC-LR markedly suppressed C. reinhardtii cell viability. Comparative proteomic and physiological analyses revealed that MC-LR significantly up-regulated protein abundance of antioxidants ascorbate peroxidase (APX) and catalase (CAT) at the beginning stage of exposure. This was accompanied by an over-accumulation of hydrogen peroxide (H2O2), suggesting that MC-LR suppresses cell viability via oxidative damage. Furthermore, we found that MCs induced desulfhydrase (DES) activity for hydrogen sulfide (H2S) generation at the beginning stage. Additional H2S donors reactivated antioxidant enzyme activity, which reduced H2O2 accumulation and ultimately enhanced C. reinhardtii tolerance to MC-LR damage. This effect could be reserved by inhibiting H2S biosynthesis. Simultaneously, we found that H2S also suppressed MC-LR-induced cell autophagy, and thus attenuated the toxic effects of MC-LR. Our findings suggest that oxidative bursts may be the main reason for the allelopathic effects of MC-LR on C. reinhardtii viability and that H2S signaling may enhance C. reinhardtii tolerance to MC-LR through the activation of antioxidant enzyme activity and suppression of cell autophagy.

Abstract:Food supplements with microalgae are becoming increasingly abundant and can be easily found anywhere. The most popular products are based on cyanophytes, such as Aphanizomenon flos-aquae, Arthrospira platensis and Limnospira maxima, or on chlorophytes, such as Chlorella or Haematoccus. Although they are all advertised as being very beneficial for health, these products might be harmful because they may contain cyanotoxins and other contaminants, and no information on production methods or strain origins is usually provided. While legislation on the presence of microcystins in waters for different uses is clear, toxicological analyses are not compulsory for food supplements, nor for analyzing anatoxins. Given the potential risk of eating contaminated food, cyanotoxins, heavy metals and the presence of other contaminant organisms were analyzed in 10 microalgae food supplements. Microcystin-LR and anatoxin-a were detected in three analyzed products, and in both cyanophyte- and chlorophyte-based products. The light microscope study revealed the presence of different potentially harmful microbial contaminants. The ICP (OES) analyses detected high concentrations of some heavy metals, especially Pb. The results emphasize the need to promote the better control of food products containing microalgae, and to develop standard methodologies to analyze cyanotoxins and potential toxic compounds to protect consumer health.

Abstract:Copper (Cu) nanomaterials have been increasingly researched and produced for many different consumer products. They have high reactivity and bactericidal properties, making them important in antifouling paints, which are thus directly introduced into aquatic ecosystems. However, studies are scarce on the behavior of Cu nanoparticles (Cu-NPs) in natural aquatic systems and their interactions with primary producers such as microalgae. We investigated the effects of NPs on some physiological responses of the freshwater phytoplankton Chlorella sorokiniana. The cells were exposed to nominal concentrations ranging from 2.50 to 635.00 μg L-1 Cu-NPs for 96 h under laboratory-controlled conditions. The cultures were monitored daily for population growth and maximum photosynthetic quantum yield. Total lipids, proteins, and carbohydrates were quantified at 72 h of Cu-NP exposure. The results showed a positive correlation between nominal Cu-NPs and Cu in the biomass (0.97 correlation coefficient) and that this was inversely proportional to total carbohydrates, with a -0.64 correlation coefficient. At the higher end of the Cu-NP concentrations tested, higher total proteins and reduced growth rates were obtained in comparison with controls; we suggest that metal-binding proteins/antioxidants and nonstructural proteins were preferentially produced under these conditions. Our results contribute to an understanding of the interaction between Cu-NPs and a cosmopolitan phytoplankton, C. sorokiniana, and we emphasize that the disposal and use of Cu-NPs requires monitoring because even at environmentally relevant concentrations, the composition of the algae was affected. Abstract:The effects on the green alga Chlorella vulgaris of 40 herbicides in 19 different chemical structure classes and with 11 dissimilar modes of action were studied through 96-h acute toxicity tests. Experimental results indicated that the average acute toxicity of acetolactate synthase (ALS)-inhibiting herbicides to C. vulgaris was close to those of the acetyl-CoA carboxylase (ACCase)-inhibiting herbicides and the lipid synthesis-inhibiting protoporphyrinogen oxidase (Protox) inhibitor. Their acute toxicities were higher than those of the microtubule process inhibitor, mitotic process inhibitor, the glutamine synthase inhibitors, and 5-enolpyruvyl-shikimate-3-phosphate synthase (EPSP synthase) inhibitors. The acute toxicities of auxin herbicides to C. vulgaris were the lowest among all herbicides tested and that of the photosynthesis-inhibiting herbicides was the highest.

Abstract:Introduction: Cyanobacterial blooms in tropical water bodies are increasingly common, because of eutrophication and rising temperatures. Consequently, many freshwater systems are affected, by reducing water quality, biodiversity, and ecosystem services. With the increased frequency of harmful algal blooms, the development of biological tools to improve water quality is an urgent issue. Objective: To evaluate the effects of a submerged macrophyte and a cladoceran on the microcystin-producing cyanobacteria Microcystis aeruginosa (NPLJ-4) and the chlorophyte Raphidocelis subcapitata (BMIUFRPE-02) in mixed cultures. Methods: Two parallel experiments were carried out for ten days to evaluate the effects of the submerged macrophyte Ceratophyllum demersum and the cladoceran Moina micrura on microalgal interactions. Microalgal strains were cultivated in the ASM1 culture medium, under controlled laboratory conditions. The first experiment presented four treatments: M (C. demersum), Z (M. micrura), MZ (C. demersum and M. micrura), and C (control). Meanwhile, the second experiment consisted of five treatments, in which the microalgae were cultivated together at different Microcystis:Raphidocelis ratios: 1:0, 3:1, 1:1, 1:3, and 0:1. Biomass and growth rates of the strains were evaluated every two days, which were statistically treated with three-way or two-way repeated-measures ANOVA. Results: In the first experiment, M. aeruginosa was significantly inhibited in M and MZ treatments from the second day, and Z from the fourth, while R. subcapitata showed no reduction in its biomass in any treatment. On the other hand, R. subcapitata was stimulated from the eighth and tenth days in M treatment and only on the eighth day in Z treatment. In the second experiment, M. aeruginosa was significantly inhibited when cultivated with R. subcapitata in low ratios (Microcystis:Raphidocelis ratio of 1:3) throughout the experiment, while the chlorophyte was stimulated in that treatment. Conclusions: The coexistence of a cyanobacterium with a green alga did not alter the main negative response of M. aeruginosa to the submerged macrophyte and zooplankton but stimulated the green alga. Accordingly, the introduction of submerged macrophytes and cladocerans already adapted to eutrophic conditions, both isolated and combined, proved to be a good method to control cyanobacterial blooms without negatively affecting other coexisting phytoplankton species.

Abstract:Low bone mass is the main risk factors for osteoporosis. The role of the rate of bone loss following cessation of menses is more difficult to evaluate. We prospectively studied vertebral bone loss in 92 women six months to three years after cessation of menses in order to determine the clinical and biological characteristics of the subgroup of subjects with accelerated bone loss. Clinical characteristics, hormone assay results, and values of the main biochemical markers of bone turnover were recorded at the initial evaluation and correlations between these parameters and the rate of bone loss were investigated. Serial measurements of vertebral bone density were performed using dual photon absorptiometry. Mean duration of the observation period was 31 +/- 13 months. Mean rate of bone loss was -1.66% per year (range 1.6% to 7.7%). In women with accelerated vertebral bone loss i.e., a greater than 2.5% decrease in bone density per year (25th centile), there were trends towards lower values for body mass index and weight (non-significant) and a significantly higher urinary calcium/urinary creatinine ratio (p < 0.05) as compared with the rest of the study group. Rate of bone loss was weakly correlated (p < 0.05) with body mass index (r = 0.22) and with the urinary calcium/urinary creatinine ratio (r = 0.23) but demonstrated no correlations with osteocalcin levels or serum alkaline phosphatase activity. In conclusion, none of the clinical features or laboratory parameters studied proved capable of reliably predicting the rate of vertebral bone loss in individual women shortly after cessation of menses.
