Abstract:Much like visually impaired humans use a white-cane, nocturnal insects and mammals use antennae or whiskers for near-range orientation. Stick insects, for example, rely heavily on antennal tactile cues to find footholds and detect obstacles. Antennal contacts can even induce aimed reaching movements. Because tactile sensors are essentially one-dimensional, they must be moved to probe the surrounding space. Sensor movement is thus an essential cue for tactile sensing, which needs to be integrated by thoracic networks for generating appropriate adaptive leg movements. Based on single and double recordings, we describe a descending neural pathway comprising three identified ON- and OFF-type neurons that convey complementary, unambiguous, and short-latency information about antennal movement to thoracic networks in the stick insect. The neurons are sensitive to the velocity of antennal movements across the entire range covered by natural movements, regardless of movement direction and joint angle. Intriguingly, none of them originates from the brain. Instead, they descend from the gnathal ganglion and receive input from antennal mechanoreceptors in this lower region of the CNS. From there, they convey information about antennal movement to the thorax. One of the descending neurons, which is additionally sensitive to substrate vibration, feeds this information back to the brain via an ascending branch. We conclude that descending interneurons with complementary tuning characteristics, gains, input and output regions convey detailed information about antennal movement to thoracic networks. This pathway bypasses higher processing centers in the brain and thus constitutes a shortcut between tactile sensors on the head and the thorax.

Abstract:Force feedback could be valuable in adapting walking to diverse terrains but the effects of changes in substrate inclination on discharges of sensory receptors that encode forces have rarely been examined. In insects, force feedback is provided by campaniform sensilla, mechanoreceptors that monitor forces as cuticular strains. We neurographically recorded responses of stick insect tibial campaniform sensilla to 'naturalistic' forces (joint torques) that occur at the hind leg femur-tibia (FT) joint in uphill, downhill and level walking. The FT joint torques, obtained in a previous study that used inverse dynamics to analyze data from freely moving stick insects, are quite variable during level walking (including changes in sign) but are larger in magnitude and more consistent when traversing sloped surfaces. Similar to vertebrates, insects used predominantly extension torque in propulsion on uphill slopes and flexion torques to brake forward motion when going downhill. Sensory discharges to joint torques reflected the torque direction but, unexpectedly, often occurred as multiple bursts that encoded the rate of change of positive forces (dF/dt) even when force levels were high. All sensory discharges also showed hysteresis and substantial decrease or complete cessation during transient force decrements. These findings have been tested in simulation in a mathematical model of the sensilla (Szczecinski et al. 2021) which accurately reproduced the biological data. Our results suggest the hypothesis that sensory feedback from the femoro-tibial joint indicating force dynamics (dF/dt) can be used to counter the instability in traversing sloped surfaces in animals and, potentially, in walking machines.

Abstract:Abstract Legged locomotion through natural environments is very complex and variable. For example, leg kinematics may differ strongly among species, but even within the same species it is adaptive and context-dependent. Inter-species differences in locomotion are often difficult to interpret, because both morphological and ecological differences among species may be strong and, as a consequence, confound each other's effects. In order to understand better how body morphology affects legged locomotion, we compare unrestrained whole-body kinematics of three stick insect species with different body proportions, but similar feeding ecology: Carausius morosus, Aretaon asperrimus and Medauroidea extradentata (= Cuniculina impigra). In order to co-vary locomotory context, we introduced a gradually increasing demand for climbing by varying the height of stairs on the set-up. The species were similar in many aspects, for example in using distinct classes of steps, with minor differences concerning the spread of corrective short steps. Major differences were related to (1) antenna length, (2) segment lengths of thorax and head, and (3) the ratio of leg length over body length: (1) Whereas all species continuously moved their antennae, only Medauroidea executed high swing movements with its front legs to search for obstacles in the near-range environment. (2) Whereas all species adjusted their body inclination, the range in which body segments moved differed considerably, with longer thorax segments tending to be moved more. (3) Finally, leg posture, time courses of leg joint angles and intra-leg coordination differed most strongly in long-legged Medauroidea.

Abstract:Motion plays an essential role in sensory acquisition. From changing the position in which information can be acquired to fine-scale probing and active sensing, animals actively control the way they interact with the environment. In olfaction, movement impacts the time and location of odour sampling as well as the flow of odour molecules around the olfactory organs. Employing a detailed spatiotemporal analysis, we investigated how insect antennae interact with the olfactory environment in a species with a well-studied olfactory system - the American cockroach. Cockroaches were tested in a wind-tunnel setup during the presentation of odours with different attractivity levels: colony extract, butanol and linalool. Our analysis revealed significant changes in antennal kinematics when odours were presented, including a shift towards the stream position, an increase in vertical movement and high-frequency local oscillations. Nevertheless, the antennal shifting occurred predominantly in a single antenna while the overall range covered by both antennae was maintained throughout. These findings hold true for both static and moving stimuli and were more pronounced for attractive odours. Furthermore, we found that upon odour encounter, there was an increase in the occurrence of high-frequency antennal sweeps and vertical strokes, which were shown to impact the olfactory environment's statistics directly. Our study lays out a tractable system for exploring the tight coupling between sensing and movement, in which antennal sweeps, in parallel to mammalian sniffing, are actively involved in facilitating odour capture and transport, generating odour intermittency in environments with low air movement where cockroaches dwell.
Dexterous Manoeuvre Through Touch in a Cluttered Scene

Abstract:Manipulation in a densely cluttered environment creates complex challenges in perception to close the control loop, many of which are due to the sophisticated physical interaction between the environment and the manipulator. Drawing from biological sensory-motor control, to handle the task in such a scenario, tactile sensing can be used to provide an additional dimension of the rich contact information from the interaction for decision making and action selection to manoeuvre towards a target. In this paper, a new tactile-based motion planning and control framework based on bioinspiration is proposed and developed for a robot manipulator to manoeuvre in a cluttered environment. An iterative two-stage machine learning approach is used in this framework: an autoencoder is used to extract important cues from tactile sensory readings while a reinforcement learning technique is used to generate optimal motion sequence to efficiently reach the given target. The framework is implemented on a KUKA LBR iiwa robot mounted with a SynTouch BioTac tactile sensor and tested with real-life experiments. The results show that the system is able to move the end-effector through the cluttered environment to reach the target effectively.

Abstract:Limb movements that are aimed toward tactile stimuli of the body provide a powerful paradigm with which to study the transformation of motor activity into context-dependent action. We relate the activity of excitatory motor neurons of the locust femoro-tibial joint to the consequent kinematics of hind leg movements made during aimed scratching. There is posture-dependence of motor neuron activity, which is stronger in large amplitude (putative fast) than in small (putative slow and intermediate) motor neurons. We relate this posture dependency to biomechanical aspects of the musculo-skeletal system and explain the occurrence of passive tibial movements that occur in the absence of agonistic motor activity. There is little recorded co-activation of antagonistic tibial extensor and flexor motor neurons, and there is differential recruitment of proximal and distal flexor motor neurons. Large-amplitude motor neurons are often recruited soon after a switch in joint movement direction. Motor bursts containing large-amplitude spikes exhibit high spike rates of small-amplitude motor neurons. The fast extensor tibiae neuron, when recruited, exhibits a pattern of activity quite different to that seen during kicking, jumping, or righting: there is no co-activation of flexor motor neurons and no full tibial flexion. Changes in femoro-tibial joint angle and angular velocity are most strongly dependent on variations in the number of motor neuron spikes and the duration of motor bursts rather than on firing frequency. Our data demonstrate how aimed scratching movements result from interactions between biomechanical features of the musculo-skeletal system and patterns of motor neuron recruitment.

Abstract:Tactile stimuli provide animals with important information about the environment, including physical features such as obstacles, and biologically relevant cues related to food, mates, hosts and predators. The antennae, the principal sensory organs of insects, house an array of sensory receptors for olfaction, gustation, audition, nociception, balance, stability, graviception, static electric fields, and thermo-, hygro- and mechanoreception. The antennae, being the anteriormost sensory appendages, play a prominent role in social interactions with conspecifics that involve primarily chemosensory and tactile stimuli. In the German cockroach ( Blattella germanica ) antennal contact during social interactions modulates brain-regulated juvenile hormone production, ultimately accelerating the reproductive rate in females. The primary sensory modality mediating this social facilitation of reproduction is antennal mechanoreception. We investigated the key elements, or stimulus features, of antennal contact that socially facilitate reproduction in B. germanica females. Using motor-driven antenna mimics, we assessed the physiological responses of females to artificial tactile stimulation. Our results indicate that tactile stimulation with artificial materials, some deviating significantly from the native antennal morphology, can facilitate female reproduction. However, none of the artificial stimuli matched the effects of social interactions with a conspecific female.
Intra- and intersegmental influences among central pattern generating networks in the walking system of the stick insect

Abstract:To efficiently move around, animals need to coordinate their limbs. Proper, context-dependent coupling among the neural networks underlying leg movement is necessary for generating intersegmental coordination. In the slow-walking stick insect, local sensory information is very important for shaping coordination. However, central coupling mechanisms among segmental central pattern generators (CPGs) may also contribute to this. Here, we analyzed the interactions between contralateral networks that drive the depressor trochanteris muscle of the legs in both isolated and interconnected deafferented thoracic ganglia of the stick insect on application of pilocarpine, a muscarinic acetylcholine receptor agonist. Our results show that depressor CPG activity is only weakly coupled between all segments. Intrasegmental phase relationships differ between the three isolated ganglia, and they are modified and stabilized when ganglia are interconnected. However, the coordination patterns that emerge do not resemble those observed during walking. Our findings are in line with recent studies and highlight the influence of sensory input on coordination in slowly walking insects. Finally, as a direct interaction between depressor CPG networks and contralateral motoneurons could not be observed, we hypothesize that coupling is based on interactions at the level of CPG interneurons.NEW & NOTEWORTHY Maintaining functional interleg coordination is vitally important as animals locomote through changing environments. The relative importance of central mechanisms vs. sensory feedback in this process is not well understood. We analyzed coordination among the neural networks generating leg movements in stick insect preparations lacking phasic sensory feedback. Under these conditions, the networks governing different legs were only weakly coupled. In stick insect, central connections alone are thus insufficient to produce the leg coordination observed behaviorally.

Abstract:To navigate complex terrains, insects use diverse tarsal structures (adhesive pads, claws, spines) to reliably attach to and locomote across substrates. This includes surfaces of variable roughness and inclination, which often require reliable transitions from ambulatory to scansorial locomotion. Using bioinspired physical models as a means for comparative research, our study specifically focused on the diversity of tarsal spines, which facilitate locomotion via frictional engagement and shear force generation. For spine designs, we took inspiration from ground beetles (Family Carabidae), which is a largely terrestrial group known for their quick locomotion. Evaluating four different species, we found that the hind legs host linear rows of rigid spines along the entire tarsus. By taking morphometric measurements of the spines, we highlighted parameters of interest (e.g., spine angle and aspect ratio) in order to test their relationship to shear forces sustained during terrain interactions. We systematically evaluated these parameters using spines cut from stainless steel shim attached to a small acrylic sled loaded with various weights. The sled was placed on 3D-printed models of rough terrain, randomly generated using fractal Brownian motion, while a motorized pulley system applied force to the spines. A force sensor measured the reaction force on the terrain, recording shear force before failure occurred. Initial shear tests highlighted the importance of spine angle, with bioinspired anisotropic designs producing higher shear forces. Using this data, we placed the best (50○ angle) and worst (90○ angle) performing spines on the legs of our insect-scale ambulatory robot physical model. We then tested the robot on various surfaces at 0, 10 and 20○ inclines, seeing similar success with the more bioinspired spines.

Abstract:ABSTRACT To traverse complex terrain, animals often transition between locomotor modes. It is well known that locomotor transitions can be induced by switching in neural control circuits or driven by a need to minimize metabolic energetic cost. Recent work revealed that locomotor transitions in complex 3D terrain cluttered with large obstacles can emerge from physical interaction with the environment controlled by the nervous system. For example, to traverse cluttered, stiff grass-like beams, the discoid cockroach often transitions from using a strenuous pitch mode pushing across the beams to using a less strenuous roll mode rolling into and through the gaps. This transition can save mechanical energetic cost substantially (∼100–101 mJ) but requires overcoming a potential energy barrier (∼10−3–10−2 mJ). Previous robotic physical modeling demonstrated that kinetic energy fluctuation of body oscillation from self-propulsion can help overcome the barrier and facilitate this transition. However, the animal was observed to transition even when the barrier still exceeded kinetic energy fluctuation. Here, we further studied whether and how the cockroach makes active adjustments to facilitate this transition to traverse cluttered beams. The animal repeatedly flexed its head and abdomen, reduced hindleg sprawl, and depressed one hindleg and elevated the other during the pitch-to-roll transition, adjustments which were absent when running on a flat ground. Using a refined potential energy landscape with additional degrees of freedom to model these adjustments, we found that head flexion did not substantially reduce the transition barrier (by ∼10−3 mJ), whereas leg sprawl reduction did so dramatically (by ∼10−2 mJ). We speculate that head flexion is for sensing the terrain to guide the transition via sensory feedback control.

Abstract:Pest insects like Tribolium castaneum beetles cause 10.8% of global stored grain losses. However, how their nervous systems coordinate adaptive movements for successful infestation is unknown. Here, we assess how Tribolium larvae locomote over different substrates and analyze their gait kinematics across speeds. Unlike many hexapods, larvae employ a bilaterally symmetric, posterior-to-anterior wave gait during fast locomotion. During slow locomotion, thoracic intrasegmental coordination is disrupted, whilst intersegmental coordination is preserved. Additionally, terminal abdominal structures (pygopods) support challenging locomotion, such as climbing overhangs. The onset of pygopod engagement coincides with leg swing initiation, suggesting a stabilizing role. Surgically severing the connective between thoracic and abdominal ganglia impaired pygopod engagement and impeded flat-surface locomotion, climbing, and tunnelling without interrupting leg kinematics. These results suggest that thoracic-abdominal coordination underlies effective movement, and gait/limb recruitment is context-dependent. Our work provides the first kinematic analysis of Tribolium larval locomotion and insights into its neural control.

Abstract:During locomotion, soft-bodied terrestrial animals solve complex control problems at substrate interfaces, but our understanding of how they achieve this without rigid components remains incomplete. Here, we develop new all-optical methods based on optical interference in a deformable substrate to measure ground reaction forces (GRFs) with micrometre and nanonewton precision in behaving larvae. Combining this with a kinematic analysis of substrate interfacing features, we shed new light onto the biomechanical control of larval locomotion. Crawling in larvae measuring ∼1 mm in length involves an intricate pattern of cuticle sequestration and planting, producing GRFs of 1-7 µN. We show that larvae insert and expand denticulated, feet-like structures into substrates as they move, a process not previously observed in soft bodied animals. These ‘protopodia’ form dynamic anchors to compensate counteracting forces. Our work provides a framework for future biomechanics research in soft-bodied animals and promises to inspire improved soft-robot design.

Abstract:Locomotion in animals provides a model for adaptive behavior as it is able to deal with various kinds of perturbations. Work in insects suggests that this evolved flexibility results from a modular architecture, which can be characterized by a recurrent neural network allowing for various emerging attractor states. Whereas a lower control-level coordinates joint movements on a short timescale, a higher-level handles action selection on longer timescales. Implementation of such a control system on a walking hexapod robot was able to deal with various walking patterns including disturbances such as uneven terrain or loss of a leg. Here, we propose a cognitive expansion to the adaptive control system that allows dealing with novel challenging situations. This approach makes use of an internal simulation-based planner that is triggered when the model-free controller fails to recover from an unstable pose. Using a grounded internal body model, the planner then tries, in internal simulation, different solutions out of context, and thus, proposes a new plan to be executed on the real robot. We demonstrate the feasibility of this control approach for walking over terrain with uncertain footholds in three scenarios.

Capillary adhesion of stick insects

Abstract:Scientific progress within the last few decades has revealed the functional morphology of an insect's sticky footpads -- a soft, sponge-like pad that secretes a thin liquid film. However, the physico-chemical mechanisms underlying their adhesion remain elusive. Here, we explore these underlying mechanisms by simultaneously measuring adhesive force and contact geometry of the adhesive footpads of live, tethered Indian stick insects, \textit{Carausius morosus}, spanning more than two orders of magnitude in body mass. We find that the adhesive force we measure is similar to previous measurements that use a centrifuge. Our measurements afford use the opportunity to directly probe the adhesive stress \textit{in vivo}, and use existing theory on capillary adhesion to predict the surface tension of the secreted liquid and compare it to previous assumptions. From our predictions, we find that the surface tension required to generate the adhesive stresses we observed ranges between 0.68 mN/m and 12 mN/m. The low surface tension of the liquid would enhance the wetting of the stick insect's footpads and promote their ability to conform to various substrates. Our insights may inform the biomimetic design of capillary-based, reversible adhesives and motivate future studies on the capillary properties of the secreted liquid.

Abstract:To navigate in the world, an animal's brain must produce commands to move, change direction, and negotiate obstacles. In the insect brain, the central complex integrates multiple forms of sensory information and guides locomotion during behaviors such as foraging, climbing over barriers, and navigating to memorized locations. These roles suggest that the central complex influences motor commands, directing the appropriate movement within the current context. Such commands are ultimately carried out by the limbs and must therefore interact with pattern generators and reflex circuits that coordinate them. Recent studies have described how neurons of the central complex encode sensory information: neurons subdivide the space around the animal, encoding the direction or orientation of stimuli used in navigation. Does a similar central-complex code directing movement exist, and if so, how does it effect changes in the control of limbs? Recording from central-complex neurons in freely walking cockroaches (Blaberus discoidalis), we identified classes of movement-predictive cells selective for slow or fast forward walking, left or right turns, or combinations of forward and turning speeds. Stimulation through recording wires produced consistent trajectories of forward walking or turning in these animals, and those that elicited turns also altered an inter-joint reflex to a pattern resembling spontaneous turning. When an animal transitioned to climbing over an obstacle, the encoding of movement in this new context changed for a subset of cells. These results indicate that encoding of movement in the central complex participates in motor control by a distributed, flexible code targeting limb reflex circuits.
