Abstract:Anovel multi-parameter biosensor, which can measure blood glucose, lactic acid, cholesterol and beta-hydrobutyric acid simultaneously, is designed for monitoring diabetes. It combines the techniques of enzymatic analytical chemistry and micro electronic mechanical systems (MEMS). On a polymeric substrate, carbon electrode is screen-printed and an enzyme sensing film is coated. In order to obtain a good stability, some amylose are added into the enzyme film. The sensing ranges of the sensor for blood glucose(Glu), Total cholesterol (T-ch), lactic acid (LAC) and beta- hydrobutyric acid (HBA) are 1 similar to 30 mmol/L, 1.0 similar to 4.0 g/L, 1 similar to 15 mmol/L and 20 similar to 700 g/L, respectively while the corresponding response times are 20, 180, 50 and 50 seconds, respectively. Experimental results show that the correlation coefficient between the sensor and regular biochemical analyzer for hospital is greater than 0.92.
Mutual promotion of electrochemical-localized surface plasmon resonance on nanochip for sensitive sialic acid detection.

Abstract:Localized surface plasmon resonance (LSPR) induced charge separation were concentrated on the metal nanoparticles surface, which made it sensitive to the surface refractive index changes during optical sensing. Similarly, electrochemical detection was based on the electron transformation on the electrode surface. Herein, we fabricated a nanochip by decorating a nanocone-array substrate with gold nanoparticles and silver nanoparticles for dynamic electro-optical spectroscopy. Mercaptophenyl boronic acid (MPBA) was immobilized firmly on the nanochip by the metal-S bond for sensitive sialic acid sensing. Owing to the high stability of gold nanoparticles and the high sensitivity of silver nanoparticles, the nanochip showed good performance in LSPR detection with rich and high responses. Besides, the nanochip also showed sensitive electrical signals during electrochemical detection due to the excitation of the energetic charges from the nanoparticles surface to the reaction system. The dynamic electro-optical spectroscopy was based on a unique combination of LSPR and linear sweep voltammetry (LSV). On the one hand, electrochemical signals activated the electrons on the nanochip to promote the propagation and resonance of surface plasmon. On the other hand, LSPR concentrated the electrons on the nanochip surface, which made the electrons easily driven to enhance the current in electrochemical detection. Results showed that mutual promotion of electrochemical-LSPR on nanochip covered a linear dynamic range from 0.05 mM to 5 mM on selective sialic acid detection with a low detection limit of 17 μM. The synchronous amplification of the electro-optical response during electrochemical-LSPR, opened up a new perspective for efficient and sensitive biochemical detection.
A Polymeric Dual-Channel Amperometric Biosensor Chip Capable of Symmetrically Splitting Sample Bands for Parallel Micro Flow Injection Determination of Glucose and Lactate

Abstract:This paper presents a chip-based dual-channel micro flow injection biosensor system for parallel determination of two components that potentially interference with each other. With glucose and lactate being the model analytes, a hybrid polydimethylsiloxane (PDMS)-polycarbonate (PC) microfluidic chip with a Y-shaped main-to-branch channel network was designed. Glucose oxidase (GOD) and lactate oxidase (LOD) biosensors of amperometric type were separately prepared inside each of the two branch channels. A sample band injected into the main channel was split into two daughter bands at the bifurcated point of the Y-shaped channel. The daughter bands were delivered into branches where the two analytes in the daughter bands were independently detected by the GOD and LOD sensors, preventing the potential chemical crosstalk between the two sensors. Factors influencing the symmetry of splitting a sample band, in turn, the sensitivity and reproducibility of the system, were discussed. Symmetric splitting was realized by using back pressure regulators (BPR). Under optimized conditions, detection limits of 78 mu mol L-1 for glucose and 127 mu mol L-1 for lactate were observed with a sample volume of 20 nL and a sample throughput rate of 40 h(-1). RSDs of 0.8-0.9% for peak heights were observed for 11 runs of a standard solution containing glucose and lactate both at 2 mmol L-1. The designed dual-channel biosensor chip was applied to the simultaneous determination of glucose and lactate in human serum samples.

Abstract:Separation and direct detection of amino acids, glucose and peptide in a 3.1 cm separation channel made of poly(dimethylsiloxane) (PDMS) with end-column amperometric detection at a copper microdisk electrode was developed. This system is the integration of a normal sized working electrode with electrochemical detection on a PDMS microfabricated device. The PDMS channels dynamically modified by 2-morpholinoethanesulfonic acid (MES) show less adsorption and more enhanced efficiency than that of unmodified ones when applied to separations of these biological molecules. The migration time is less than 100 s and the reproducibility of migration time is satisfactory with relative standard deviation (RSD) of 2.8% in 19 successive injections. The limits of detection of arginine (Arg), glucose, and methionine-glycine (Met-Gly) are estimated to be 2.0, 8.5, and 64.0 muM at S/N = 3, approximately 0.5-16.0 fmol, respectively. Variances influencing the separation efficiency and amperometric response, including injection, separation voltage, detection potential, or concentration of buffer and additive, are assessed and optimized.

Abstract:As widely used commercial sensors, amperometric biosensors based on electrochemical (EC) reactions exhibit good linearity and reproducibility but suffer from a low dynamic range and poor limit of detection (LoD). In contrast, biosensors utilizing field-effect transistors (FETs) demonstrate a wide detection range and ultralow LoDs but are susceptible to interferences or ionic strength influences and have poor quantitative accuracy due to their nonlinear response mechanism. Here, EC and carbon nanotube (CNT) FET-based biosensors are integrated into one chip via a compatible fabrication process. The dual-mode biosensing chip (DMSC) provides the advantages from both the EC biosensors and FET biosensors; these advantages include high reliability and accuracy for quantitatively detecting glucose, a dynamic detection range spanning 6 orders of magnitude, and a detection limit of 1 fM. The introduction of a time-division multiplexing detection method enables the DMSC to carry out complementary detection of the same target, providing early warning of specific abnormal biomarkers and accurate determination of their concentration in real time. The DMSC approach is generally applicable to the detection of other biomarkers and provides exciting opportunities for more accurate self-monitoring diagnostics in real situations. In-Channel Modification of Biosensor Electrodes Integrated on A Polycarbonate Microfluidic Chip for Micro Flow-Injection Amperometric Determination of Glucose

Abstract:A microfluidic electrochemical biosensor chip made of full polycarbonate (PC) sheets was fabricated for micro flow-injection amperometric determination of glucose. Microchannel network was embossed onto a PC sheet, and micro gold film electrode bases were prepared on a PC cover sheet with a photo-directed electroless plating technique. After thermal bonding, the integrated gold bases were modified to a glucose oxidase working electrode, an Ag/AgCl reference electrode and a platinum counter electrode, respectively. The sequence of the in-channel modification was studied to avoid the possible contamination of one modified electrode by the reagents used for another electrode. Cooperated with a gravity pump and an automatic sample dispenser, the fabricated micro flow-injection biosensor chip was used for determination of glucose. A detection limit of 4.1μmolL−1 was achieved at the sampling volume of about 130nL, and sample throughput rate (or sampling frequency) reached 116h−1. Inter- and intra-day precisions (RSD) for the determination of a 100μmolL−1 glucose solution were 0.7% and 1.8%, respectively. Chip-to-chip reproducibility, expressed by the precision of analytical results obtained with individual chips, were less than 10% (RSD). The developed micro flow-injection amperometric biosensor system was successfully applied to the determination of glucose content in pharmaceutical injections.

Abstract:Significant progress has been made in recent decades; however, the development of an accurate, portability and high-throughput electrochemical biosensor for quantifying trace biomarker in complex biological samples remains challenging. In this study, we described a novel 3 × 3 multi-unit portable integrated biosensor array, specifically designed for parallel and precise detection of trace levels of carbohydrate antigen 125 (CA-125) using multiplex synergistic signal amplification. The sensor array substrate was constructed on a planar printed circuit board (PCB) using printing technology, making them integrated, portable, affordable and disposable. The exceptional sensitivity of the biosensor is attributed to the nitrogen-doped mesoporous carbon (NMC) nanomaterial, which possesses a large surface area, superior conductivity and tunable pore size, enabling high affinity of and enzyme catalytic activity. The integrated biosensing array platform consists of individually controlled sensors, allowing for high-throughput tests of the same sample simultaneously, ensuring the reliability and accuracy of assay. The developed biosensor array system demonstrated excellent sensitivity and rapid CA-125 assay, achieving an impressive detection limit as low as 0.027 U/mL (S/N = 3). Meanwhile, the high-throughput multiplex test results ( n = 9) were obtained within a short timeframe of 30 min. Ultimately, the biosensor array successfully quantified CA-125 in human serum, yielding results comparable to the enzyme–linked immunosorbent assay technique. This technology presents a highly promising strategy that can be potentially extended to detect other biomarkers for biological analysis and early disease diagnosis.

Abstract:A fully integrated polycarbonate (PC) microchip for CE with end-channel electrochemical detection operated in an amperometric mode (CE-ED) has been developed. The on-chip integrated three-electrode system consisted of a gold working electrode, an Ag/AgCl reference electrode and a platinum counter electrode, which was fabricated by photo-directed electroless plating combined with electroplating. The working electrode was positioned against the separation channel exit to reduce post-channel band broadening. The electrophoresis high-voltage (HV) interference with the amperometric detection was assessed with respect to detection noise and potential shifts at various working-to-reference electrode spacing. It was observed that the electrophoresis HV interference caused by positioning the working electrode against the channel exit could be diminished by using an on-chip integrated reference electrode that was positioned in close proximity (100 pm) to the working electrode. The CE-ED microchip was demonstrated for the separation of model analytes, including dopamine (DA) and catechol (CA). Detection limits of 132 and 164 nM were achieved for DA and CA, respectively, and a theoretical plate number of 2.5 x 10(4)/m was obtained for DA. Relative standard deviations in peak heights observed for five runs of a standard solution containing the two analytes (0.1 mM for each) were 1.2 and 3.1% for DA and CA, respectively. The chip could be continuously used for more than 8 h without significant deterioration in analytical performance.

Abstract:We proposed herein a novel approach for fabricating nanoband microelectrodes for electrochemical detection on an electrophoresis microchip. The metal films were first obtained via region-selective electroless deposition of gold or copper films on PDMS substrates by selective region plasma oxidation through shadow masking. Both metal films show uniform surfaces with the thickness at the level of 100nm. By casting another PDMS layer on the metal films, the cross section of the sandwich structures can be used as nanoband microelectrodes, which can be renewed just by cutting. These nanoband microelectrodes are successfully used as electrochemical detectors in microchip electrophoresis for the detection of amino acids, proteins and neurotransmitter molecules. Moreover, integrating an Au-Cu double-metal detector with a double-channel electrophoresis system, we can easily distinguish electroactive amino acids from that of non-electroactive amino acids.

Abstract:A novel, simple and label-free multianalyte immunoassay system is presented here by integrating arrayed electrodes on a silicon chip via MEMS. The chip is consisted of six Au disk electrodes, an Au counter electrode and an Ag/AgCl reference electrode. Semi-insulating poly(o-phenylenediamine) (PoPD) was utilized to co-polymerize and immobilize antibodies at the arrayed Au electrodes, and wider linear detection range was obtained than those prepared with completely insulating PoPD. Electrochemical cyclic voltammogram (CV), AC impedance spectroscopy, AFM and fluorescence microscopy were employed to characterize the system. The arrayed electrodes offered exact control of deposition position via electrochemical operation, allowing selectively immobilization of different antibodies at desired positions on a single chip. Specific recognition of antibody (Ab) to corresponding antigen (An) was quantitatively monitored by cyclic voltammograms in the presence of electrochemical redox probe, ferrocene methanol. The proposed immunoassay chips showed sensitive response to three liver fibrosis markers, hyaluronic acid (HA), collagen type IV (IV-C) and lamin (LN) at ng/mL level simultaneously and specifically in a tiny amount of volume, usually 50 microL. The results obtained via chips were well consistent with those obtained by commercial radio immunoassays (RIA).

Abstract:Dysregulations in metabolic and physiological cellular processes are generally accompanied by changes in concentration profiles of multiple biomarkers. Measuring dynamic changes in concentrations of several affected biomarkers can enhance our comprehension of the underlying disease pathophysiology and/or mechanisms of cytotoxicity. Most analytical detection techniques require samples to be obtained at pre-defined times for offline analysis. However, the inadequate temporal resolution of these methods can lead to key biological events that occur at shorter timescales, being overlooked. Electrochemical biosensors are relatively simple, cost-effective detection techniques that have been used extensively for continuous analyte monitoring, and more recently, for sensing multiple analytes. We describe the design and functionality of a novel electrochemical detection platform, comprising a screen-printed Pt electrode array, a microfluidic chip, and an automated pump and valve system, capable of quantitatively detecting up to eight analytes. As a proof of concept, we show the simultaneous, sensitive, and selective amperometric detection of four essential biomarkers- glucose, lactate, glutamate, and acetylcholine, in real time. We also demonstrate the biological application of this platform by continuously monitoring exocytotic glutamate release from human induced pluripotent stem cell-derived neuronal cultures over time. Excessive accumulation of glutamate in the synaptic cleft leads to prolonged stimulation of glutamate receptors on neurons, triggering a cascade of neurotoxic events that ultimately results in loss of neuronal function and cell death. The results suggest that our multianalyte electrochemical sensor platform can be coupled with more complex biological systems such as in vitro organotypic models, for the accurate monitoring of biomarker changes during adverse events like acute neurotoxicity and microbial infection.

Abstract:This paper presents an electrochemical detector for poly(dimethylsiloxane) (PDMS) microchip CE with a three-dimensional adjustor which makes it possible to accurately align a separate working electrode that can be easily fabricated in laboratory to the uncertain PDMS microchannel outlet. The substantial influence of the electrode—PDMS microchannel distance was investigated. The optimal electrode-outlet distance was found to be 10 μm for the PDMS microchannel with the width of 50 μm due to its relatively slow electroosmotic flow. Adrenaline and catechol were well separated, with a linear response range from 20 μM to 1 mM, and a detection limit of 2 μM for catechol, using carbon disk electrode (diameter of 300 μm). Furthermore, arginine and histidine can be well separated and detected directly in the PDMS microchannel using a Cu disk electrode (diameter of 150 μm).
Chronoampermetric detection of enzymatic glucose sensor based on doped polyindole/MWCNT composites modified onto screen-printed carbon electrode as portable sensing device for diabetes

Abstract:Doped-polyindole (dPIn) mixed with multi-walled carbon nanotubes (MWCNTs) were coated on a screen-printed electrode to improve the electroactive surface area and current response of the chronoamperometric enzymatic glucose sensor. Glucose oxidase mixed with chitosan (CHI-GOx) was immobilized on the electrode. (3-Aminopropyl) triethoxysilane (APTES) was used as a linker between the CHI-GOx and the dPIn. The current response of the glucose sensor increased with increasing glucose concentration according to a power law relation. The sensitivity of the CHI-GOx/APTES/dPIn was 55.7 μA mM −1 cm −2 with an LOD (limit of detection) of 0.01 mM, where the detectable glucose concentration range was 0.01–50 mM. The sensitivity of the CHI-GOx/APTES/1.5%MWCNT-dPIn was 182.9 μA mM −1 cm −2 with an LOD of 0.01 mM, where the detectable glucose concentration range was 0.01–100 mM. The detectable concentration ranges of glucose well cover the glucose concentrations in urine and blood. The fabricated enzymatic glucose sensors showed high stability during a storage period of four weeks and high selectivity relative to other interferences. Moreover, the sensor was successfully demonstrated as a continuous or step-wise glucose monitoring device. The preparation method employed here was facile and suitable for large quantity production. The glucose sensor fabricated here, consisting of the three-electrode cell of SPCE, were simple to use for glucose detection. Thus, it is promising to use as a prototype for real glucose monitoring for diabetic patients in the future.

Abstract:The multiplex and simultaneous determination of blood glucose, creatinine and uric acid is essential for the early screening of chronic diseases or regular disease monitoring. Here, we report for the first time a biosensing array for the multiplex and simultaneous determination of plasma glucose, creatinine and uric acid. The sensing electrodes are fabricated on a PET surface, including three working electrodes, one reference electrode, and one counter electrode. Each specific enzyme is immobilized on its corresponding working electrode. The biosensing array can exhibit high sensitivity and selectivity for the simultaneous determination of blood glucose, creatinine and uric acid in real blood samples, and the measurement results are accurate and consistent with those from the clinical biochemistry analyzer in the hospital. It is expected that this work could provide new avenues for the fundamental study of biosensing device construction, as well as practical applications of the detection of biomarkers in chronic diseases.

Abstract:As an important carbon and energy source, glucose is a biomarker of many diseases, such as diabetes, peritoneal carcinomatosis and peritonitis. Nevertheless, real-time glucose detection strategy with single portable device to monitor different diseases was seldom established. In this study, a Pt-Ni nanocomposite and silica nanoporous membrane modified electrode with a miniaturized electrochemical circuit board was developed for glucose detection to monitor different diseases. Through construction of Pt-Ni nanocomposite, the sensitivity of glucose detection was improved obviously compared with Pt electrode and Ni electrode. Silica nanoporous membrane eliminated the negatively charged interferences, such as uric acid and ascorbic acid, simultaneously resisting the interference of albumin. Modified with these nanomaterials, the glucose electrode realized 1-40 mM detection in artificial blood and 0.2-15 mM detection in artificial ascites respectively, displaying good selectivity and stability. Miniaturized electrochemical circuit board was established to execute electrochemical test and realize data interaction with a Bluetooth-enabled smartphone. Furthermore, the accuracy of this portable system was validated with biochemical analyzer in clinical samples, including diabetic whole blood and pathological ascites. In this way, the system could offer real-time and rapid opportunities towards dynamic evaluation of diabetes, peritoneal carcinomatosis and peritonitis.

Abstract:Poly(dimethylsiloxane) (PDMS) microfluidic channels modified by citrate-stabilized gold nanoparticles after coating a layer of linear polyethylenimine (LPEI) were successfully used to separate dopamine and epinephrine, which were difficult to be separated from baseline in native and hybrid PDMS microchannels. In-channel amperometric detection with a single carbon fibre cylindrical electrode was employed. Experimental parameters of separation and detection processes were optimized in detail. The analytes were well separated within 100s in a 3.7cm long separation channel at a separation voltage of +800V using a 30mM phosphate buffer solution (PBS, pH 7.0). Linear responses of them were obtained both from 25 to 600μM with detection limits of 2μM for dopamine and 5μM for epinephrine, respectively. The modified PDMS channels have a long-term stability and an excellent reproducibility within 2 weeks.
Electrochemical Microfluidic Multiplexed Bioanalysis by a Highly Active Bottlebrush-like Nanocarbon Microelectrode

Abstract:We present a highly efficient multichannel microfluidic electrochemical sensor integrated with an electroactive nanocarbon microelectrode for sensitive and selective detection of multiple biomarkers in different biological samples. Our results have shown that ionic liquid-assisted wet spinning followed by tailored growth of metal-organic frameworks and pyrolysis treatment led to structural and molecular engineering of mechanically robust all-carbon microfibers for excellent electrochemical activities. The flexible bottlebrush-like nanocarbon microelectrode features a "stem" of freestanding N, B-codoped graphene fiber and high-density "bristles" of Co, N-codoped carbon nanotube arrays, leading to promoted electrocatalytic mechanism that has been substantiated by density functional theory calculations. The structural characteristics, high catalytic activities, and favorable biocompatibility of the bottlebrush nanocarbon electrodes provide opportunities for multichannel, microfluidic detection of redox-active biomolecules, including hydrogen sulfide (H<sub>2</sub>S), dopamine (DA), uric acid (UA), and ascorbic acid (AA), and have been applied to on-chip monitoring of H<sub>2</sub>S and DA released from live cancer cells or neuroblastoma cells and DA, UA, and AA in trace amounts of body fluids such as sweat, finger blood, tears, saliva, and urine, which is of great significance for clinical diagnosis and prognosis in point-of-care testing.
