Abstract:A rigorous preselection of identified compounds by in vitro cellular screening is necessary prior to using the drug candidates for the further time consuming and parallel monitoring stage. In this paper we introduced an integrated cell based chip which can detect changes of electrophysiological parameters by using microelectronic chips such as light-addressable potentiometric sensors(LAPS), micro-electrodes array sensors(MEAS) and interdigitated array sensor(IDA) etc. to record the extracellular metabolism chemical and biological substances, action potential and living characteristics of cells representing the different response. In this research we presented novel ideas including an integrating multifunctional cell sensors chip, obtaining the living cells as the source of cell sensors with cardiac myocytes. Further developments will be done by culturing living olfactory and taste cells on surface of the devices. Now, we have completed the fabrication of the sensor and obtained some promising experiments results in different sensor units. This research will be interesting and useful for parallel extracellular monitoring.
High Accuracy Determination of Multi Metabolite by an Origami-Based Coulometric Electrochemical Biosensor

Abstract:In this work, we present an origami-based coulometric electrochemical biosensor for high-accuracy determination of multi metabolite on one single electrode with only 0.5 mu L samples. The proposed biosensor was fabricated with a screen-printed electrode and an origami device that was equipped with three foldable tabs preloaded with diverse oxi-dases. The origami device with a hole in its center was pasted onto the screen-printed electrode to forma reaction cell for electrochemically electrolyzing the target analytes with the corresponding oxidases. As a demonstration, L-lactate, cho-lesterol, and glucose as the target analytes were successfully detected with this biosensor. The values of electric charge for the L-lactate, cholesterol, glucose oxidation by integrating the Faradaic current vs. time were in good agreement with the theoretical values. The linear ranges of detection were respectively 0-10 mM (L-lactate), 0-5 mM (cholesterol), 0-24 mM (glucose) with efficiency electrolysis 99% +/- 3%, 99% +/- 1%, 100% +/- 2% (n = 3, n: number of repetition). And the limit of detection of L-lactate, cholesterol, glucose was 0.25 mM, 0.23 mM, 0.03 mM respectively. The applica-bility of the device in POCT was performed in serum. And the above 95% recovery rate indicated it was suitable for the clinical application. Additionally, the proposed biosensor could be a candidate for more other metabolites detection, such as fructose, glutamate, bilirubin, and galactose. Abstract:MicroRNA-208a (miR208a) is a cardiac-specific miRNA and can provide an earlier prediction for AMI onset than currently used cardiac biomarkers. Graphene field-effect transistor (GFET) biosensor, as the promising candidate for next-generation biosensor, has been demonstrated for real-time detection of a variety of bioactive molecules with ultrasensitivity from nM to fM level. Currently, research on GFET sensors mostly focuses on the properties of the devices themselves, rather than aiming for ultra-sensitive detection based on biochemical substances. In this study, we developed a liquid-gated graphene field-effect transistor-based biosensor for detection of miR208a and designed a portable device to adapt with it for a fast readout of the electronic response, achieving a detection range of 0.01-1 pM, detection limit of 5.3 fM and detection time of 40 minutes. Our work provides a reliable strategy to rapidly and ultrasensitively quantifying miRNA-208a, which also holds great potential to be extended in a wide range of applications of other disease markers and lead to powerful tools for healthcare.

Abstract:Objective: Rising concerns over wellness and aging have heightened the demand for convenient and efficient on-site health monitoring and disease screening. Current research, focused on specific biomarker detection, often neglects the complexities of sample matrix interference and the absence of a comprehensive, automated platform. To address these issues, we have developed a universal, fully automated analyzer for multifaceted, on-site biochemical analysis of body fluids. Methods: This analyzer integrates automated sample pretreatment, automatic dilution, detection, and self-cleaning functionalities seamlessly. It is designed to detect a wide range of analytes, from small molecules to macromolecules, including ions and proteins, utilizing spectrophotometric sensing. After optimization, the analyzer achieves performance comparable to traditional Enzyme-Linked Immunosorbent Assay (ELISA), while significantly expanding its detection range through automated dilution. Results: Demonstrations of small molecule detection include the simultaneous assessment of citric acid (CA) and oxalic acid (OA) in urine, achieving recovery rates between 96.65%-106.42% and 93.13%-112.50%, respectively. For protein detection, the analyzer successfully identified Cyfra21-1 in saliva with a recovery rate of 104.93%-111.31%. The pre-treatment process requires only 8.8 minutes, showing enhanced recovery rates for CA and OA at 97.8% and 97.6% respectively, which are superior and more rapid than manual methods. Conclusion: The exemplary pretreatment and detection performance of the analyzer underlines its effectiveness in multifaceted, on-site biomarker detection, establishing it as a promising and versatile tool for disease screening and health monitoring. Significance: This analyzer offers a powerful technological solution for on-site fluid testing, advancing community health care by facilitating more reliable and rapid diagnostics.

Abstract:The multiplex and simultaneous determination of blood glucose, creatinine and uric acid is essential for the early screening of chronic diseases or regular disease monitoring. Here, we report for the first time a biosensing array for the multiplex and simultaneous determination of plasma glucose, creatinine and uric acid. The sensing electrodes are fabricated on a PET surface, including three working electrodes, one reference electrode, and one counter electrode. Each specific enzyme is immobilized on its corresponding working electrode. The biosensing array can exhibit high sensitivity and selectivity for the simultaneous determination of blood glucose, creatinine and uric acid in real blood samples, and the measurement results are accurate and consistent with those from the clinical biochemistry analyzer in the hospital. It is expected that this work could provide new avenues for the fundamental study of biosensing device construction, as well as practical applications of the detection of biomarkers in chronic diseases.

Abstract:Dysregulations in metabolic and physiological cellular processes are generally accompanied by changes in concentration profiles of multiple biomarkers. Measuring dynamic changes in concentrations of several affected biomarkers can enhance our comprehension of the underlying disease pathophysiology and/or mechanisms of cytotoxicity. Most analytical detection techniques require samples to be obtained at pre-defined times for offline analysis. However, the inadequate temporal resolution of these methods can lead to key biological events that occur at shorter timescales, being overlooked. Electrochemical biosensors are relatively simple, cost-effective detection techniques that have been used extensively for continuous analyte monitoring, and more recently, for sensing multiple analytes. We describe the design and functionality of a novel electrochemical detection platform, comprising a screen-printed Pt electrode array, a microfluidic chip, and an automated pump and valve system, capable of quantitatively detecting up to eight analytes. As a proof of concept, we show the simultaneous, sensitive, and selective amperometric detection of four essential biomarkers- glucose, lactate, glutamate, and acetylcholine, in real time. We also demonstrate the biological application of this platform by continuously monitoring exocytotic glutamate release from human induced pluripotent stem cell-derived neuronal cultures over time. Excessive accumulation of glutamate in the synaptic cleft leads to prolonged stimulation of glutamate receptors on neurons, triggering a cascade of neurotoxic events that ultimately results in loss of neuronal function and cell death. The results suggest that our multianalyte electrochemical sensor platform can be coupled with more complex biological systems such as in vitro organotypic models, for the accurate monitoring of biomarker changes during adverse events like acute neurotoxicity and microbial infection.

Abstract:As widely used commercial sensors, amperometric biosensors based on electrochemical (EC) reactions exhibit good linearity and reproducibility but suffer from a low dynamic range and poor limit of detection (LoD). In contrast, biosensors utilizing field-effect transistors (FETs) demonstrate a wide detection range and ultralow LoDs but are susceptible to interferences or ionic strength influences and have poor quantitative accuracy due to their nonlinear response mechanism. Here, EC and carbon nanotube (CNT) FET-based biosensors are integrated into one chip via a compatible fabrication process. The dual-mode biosensing chip (DMSC) provides the advantages from both the EC biosensors and FET biosensors; these advantages include high reliability and accuracy for quantitatively detecting glucose, a dynamic detection range spanning 6 orders of magnitude, and a detection limit of 1 fM. The introduction of a time-division multiplexing detection method enables the DMSC to carry out complementary detection of the same target, providing early warning of specific abnormal biomarkers and accurate determination of their concentration in real time. The DMSC approach is generally applicable to the detection of other biomarkers and provides exciting opportunities for more accurate self-monitoring diagnostics in real situations. Abstract:Significant progress has been made in recent decades; however, the development of an accurate, portability and high-throughput electrochemical biosensor for quantifying trace biomarker in complex biological samples remains challenging. In this study, we described a novel 3 × 3 multi-unit portable integrated biosensor array, specifically designed for parallel and precise detection of trace levels of carbohydrate antigen 125 (CA-125) using multiplex synergistic signal amplification. The sensor array substrate was constructed on a planar printed circuit board (PCB) using printing technology, making them integrated, portable, affordable and disposable. The exceptional sensitivity of the biosensor is attributed to the nitrogen-doped mesoporous carbon (NMC) nanomaterial, which possesses a large surface area, superior conductivity and tunable pore size, enabling high affinity of and enzyme catalytic activity. The integrated biosensing array platform consists of individually controlled sensors, allowing for high-throughput tests of the same sample simultaneously, ensuring the reliability and accuracy of assay. The developed biosensor array system demonstrated excellent sensitivity and rapid CA-125 assay, achieving an impressive detection limit as low as 0.027 U/mL (S/N = 3). Meanwhile, the high-throughput multiplex test results ( n = 9) were obtained within a short timeframe of 30 min. Ultimately, the biosensor array successfully quantified CA-125 in human serum, yielding results comparable to the enzyme–linked immunosorbent assay technique. This technology presents a highly promising strategy that can be potentially extended to detect other biomarkers for biological analysis and early disease diagnosis.

Abstract:The simultaneous detection of multiple analytes using electrochemical biosensor arrays has demonstrated significant potential in various fields, including medical diagnostics. These platforms are designed to be low‐cost, easy to use, and offer fast detection of a range of molecules. This concept article discusses the components necessary to achieve multiplexing with electrochemical biosensor arrays. The various methods used to fabricate electrode arrays of different types are discussed. Furthermore, the methods for the selective immobilization of multiple different bioreceptors onto individual electrodes within the array, a crucial step essential for conferring specificity to the analytical processes, are presented with relevant examples. We have focused on enzymes, antibodies and aptamers as examples of bioreceptors that have been deployed in various multianalyte electrochemical detection platforms. Finally, we discuss the key challenges associated with their application for the analysis of real samples and provide a future outlook on possible strategies that can be used to overcome these challenges.

Abstract:Current advances in non-invasive fluid diagnostics highlight unique benefits for monitoring metabolic diseases. However, the low concentrations and complex compositions of biomarkers in fluids such as sweat, urine, and saliva impose stringent demands on the sensitivity and stability of detection technologies. Here, we developed a high-sensitivity, low-cost instantaneous electrochemical sensor based on the superadditive effect mechanism of Cu-TCPP(Fe)/Mxene (MMs Paper-ECL Sensor), which has been successfully applied for the simultaneous real-time detection of glucose and uric acid. Strong interfacial interactions between Mxene and Cu-TCPP(Fe) were revealed through precise simulation calculations and multi-dimensional characterization analysis, significantly enhancing the sensor's electrocatalytic performance and reaction kinetics. Experimentally, this exceptional electrocatalytic activity was demonstrated in its unprecedented high sensitivity and wide linear detection range for glucose and uric acid, with a non-invasive linear range from 0.001 nM to 5 mM, 0.025 nM-5 mM, detection limits as low as 1.88 aM and 5.80 pM, and stability extending up to 100 days. This represents not only a breakthrough in sensitivity and stability but also provides an effective, low-cost solution that overcomes the limitations of existing electronic devices, enabling multi-channel simultaneous detection. The universality of this sensor holds vast potential for application in the field of non-invasive fluid diagnostics.

Abstract:Simultaneous detection of multiple biomarkers is benefit for reducing the detection cycles, avoiding the false-positive signals, and providing the cross validation, which provide the opportunity to understand the pathogenic mechanisms and achieve precise early diagnosis. Here, we demonstrate the mini-pillar-based individual electrochemical array for simultaneous detection of multiple biomarkers. On such platform, the mini-pillar could confine the microdroplet as individual and open-channel microreactor, which is extremely helpful for reducing reagent consumption and extracting internal information, and the electrodes array embedded in mini-pillar are integrated on one side to achieve multiple and simultaneous electrochemical sensing. The introduction of gold nanodendrites by electrodeposition has greatly enhanced sensitivity via improving probe-binding capacity and response signals. Sensitive and selective detection of multiple Alzheimer's biomarkers including Tau, ApoE4, Amyloid-β and miRNA-101 on such mini-pillar-based biosensor is also achieved. Such biosensor platform with the advantages of high-yield, high sensitivity, low-waste and multiple signals output shows great promise in sensing multiple biomolecules for disease diagnosis and health monitoring.
Multidimensional Interactive Cascading Nanochips for Detection of Multiple Liver Diseases via Precise Metabolite Profiling

Abstract:It is challenging to detect and differentiate multiple diseases with high complexity/similarity from the same organ. Metabolic analysis based on nanomatrix-assisted laser desorption/ionization mass spectrometry (NMALDI-MS) is a promising platform for disease diagnosis, while the enhanced property of its core nanomatrix materials has plenty of room for improvement. Herein, a multidimensional interactive cascade nanochip composed of iron oxide nanoparticles (FeNPs)/MXene/gold nanoparticles (AuNPs), IMG, is reported for serum metabolic profiling to achieve high-throughput detection of multiple liver diseases. MXene serves as a multi-binding site and an electron-hole source for ionization during NMALDI-MS analysis. Introduction of AuNPs with surface plasmon resonance (SPR) properties facilitates surface charge accumulation and rapid energy conversion. FeNPs are integrated into the MXene/Au nanocomposite to sharply reduce the thermal conductivity of the nanochip with negligible heat loss for strong thermally-driven desorption, and construct a multi-interaction proton transport pathway with MXene and AuNPs for strong ionization. Analysis of these enhanced serum fingerprint signals detected from the IMG nanochip through a neural network model results in differentiation of multiple liver diseases via a single pass and revelation of potential metabolic biomarkers. The promising method can rapidly and accurately screen various liver diseases, thus allowing timely treatment of liver diseases.

Abstract:Here a novel electrochemical microarray platform towards high-throughput detection is presented. This microarray owns 8×5 well-organized oval cells and 8×5 pairs of ITO discs (two ITO discs for each pair in one cell). 8 mobile and aligned Pt–Ag/AgCl coaxial electrodes with compact configuration, where Pt column is used for counter electrode and Ag/AgCl for reference electrode, are inserted in 8 cells separately and made up 8 groups of three-electrode detection system together with 8 pairs of ITO discs. To demonstrate the applicability of such design, two kinds of bioconjugates, carboxyl graphene nanosheets–methylene blue (CGS–MB) and carboxyl graphene nanosheets–Prussian blue (CGS–PB) nanocomposites selected as model redox indicators and crosslinked with two kinds of antibodies were coated on the pair of ITO discs to realize a high-throughput detection of two tumor markers based on the increasing spatial blocking and impedance from the formed immunocomplex.

Abstract:The review gives a systematic account of published data devoted to multisensor technologies for manufacturing effective miniature devices capable of simultaneous accurate determination of several analytes and/or characteristics of biological fluids. The use of electrolyte-gated field-effect transistors as a biosensing platform is considered in detail. The devices based on these transistors demonstrate record-low limits of detection and are easy for mass production. The use of electrolyte-gated field-effect transistors in combination with aptamers capable of specifically binding to various analytes forms the grounds for innovations in early medical diagnosis (label-free biomarker detection). The most successful examples of multisensor devices using these transistors are presented, and the prospects of using aptamers as a recognition element in these devices are demonstrated. Abstract:We present a highly efficient multichannel microfluidic electrochemical sensor integrated with an electroactive nanocarbon microelectrode for sensitive and selective detection of multiple biomarkers in different biological samples. Our results have shown that ionic liquid-assisted wet spinning followed by tailored growth of metal-organic frameworks and pyrolysis treatment led to structural and molecular engineering of mechanically robust all-carbon microfibers for excellent electrochemical activities. The flexible bottlebrush-like nanocarbon microelectrode features a "stem" of freestanding N, B-codoped graphene fiber and high-density "bristles" of Co, N-codoped carbon nanotube arrays, leading to promoted electrocatalytic mechanism that has been substantiated by density functional theory calculations. The structural characteristics, high catalytic activities, and favorable biocompatibility of the bottlebrush nanocarbon electrodes provide opportunities for multichannel, microfluidic detection of redox-active biomolecules, including hydrogen sulfide (H<sub>2</sub>S), dopamine (DA), uric acid (UA), and ascorbic acid (AA), and have been applied to on-chip monitoring of H<sub>2</sub>S and DA released from live cancer cells or neuroblastoma cells and DA, UA, and AA in trace amounts of body fluids such as sweat, finger blood, tears, saliva, and urine, which is of great significance for clinical diagnosis and prognosis in point-of-care testing.

Abstract:BACKGROUND:Automated and convenient multianalyte detection with high throughput is increasingly needed in clinical diagnosis. We developed a disposable 4-by-2 array for programmed simultaneous amperometric immunoassay of 4 tumor markers.METHODS:We used a screen-printed technique, 1-step immobilization method, and flow injection technique. We immobilized carcinoembryonic antigen, alpha-fetoprotein, beta-human choriogonadotropin, and carcinoma antigen 125 as model analytes in a redox mediator-grafted, biopolymer-modified, screen-printed carbon electrode array to capture corresponding horseradish peroxidase-labeled antibodies in competitive immunoreactions. The simultaneous multianalyte immunoassay was automatically carried out to amperometrically monitor the mediator-catalyzed enzymatic response to hydrogen peroxide, which decreased in proportion to the concentrations of analytes in samples.RESULTS:The multianalyte immunosensor array had a throughput of 60 samples/h and allowed simultaneous detection of carcinoembryonic antigen, alpha-fetoprotein, beta-human choriogonadotropin, and carcinoma antigen 125 in clinical serum samples with concentrations up to 188 microg/L, 250 microg/L, 266 IU/L, and 334 kIU/L, respectively. The detection limits (limits of the blank, mean of blank plus 3 SD) were 1.1 micro/L, 1.7 microg/L, 1.2 IU/L, and 1.7 kIU/L. The inter- and intraassay imprecision (CVs) of the immunosensor arrays were <7.8% and <9.0%, respectively. The immunosensor arrays were stable for 28 days.CONCLUSIONS:This newly constructed immunosensor array provides a simple, automated, simultaneous multianalyte immunoassay with high throughput, short analytical time, and sufficiently low detection limits for clinical application. This method offers the capability of miniaturizing the multianalyte detection device.

Abstract:The reliability of analysis is becoming increasingly important as point-of-care diagnostics are transitioning from single analyte detection towards multiplexed multianalyte detection. Multianalyte detection benefits greatly from complementary metal-oxide semiconductor (CMOS) integrated sensing solutions, offering miniaturized multiplexed sensing arrays with integrated readout electronics and extremely large sensor counts. The development of CMOS back end of line integration compatible graphene field-effect transistor (GFET) based biosensing has been rapid during the last few years, both in terms of the fabrication scale-up and functionalization towards biorecognition from real sample matrices. The next steps in industrialization relate to improving reliability and require increased statistics. Regarding functionalization towards truly quantitative sensors and on-chip bioassays with improved statistics require sensor arrays with reduced variability in functionalization. Such multiplexed bioassays, whether based on graphene or on other sensitive nanomaterials, are among the most promising technologies for label-free electrical biosensing. As an important step towards that, we report wafer-scale fabrication of CMOS integrated GFET arrays with high yield and uniformity, designed especially for biosensing applications. We demonstrate the operation of the sensing platform array with 512 GFETs in simultaneous detection for sodium chloride concentration series. This platform offers a truly statistical approach on GFET based biosensing and further to quantitative and multi-analyte sensing. The reported techniques can also be applied to other fields relying on functionalized GFETs, such as gas or chemical sensing or infrared imaging.

Abstract:Continuous efforts to produce functional nanomaterials and flexible/stretchable devices have promoted cumbersome, laboratorial, detection processes toward wearable and portable intelligent sensing approaches. Responding to the challenges of the multiple analytes, mixtures, and complex components of practical samples, sensing array and multivariate analysis techniques have a significant advantage in terms of superior analytical capabilities, i.e., they are convenient, rapid, sensitive and have high-throughput for multi-analyte identification in food safety, clinical diagnoses, and environmental monitoring. Besides traditional molecular design and recognition mechanisms, materials with micro/nano structures also contribute to strong signals, sensitive responses, and novel properties. In this review, through a new perspective of signal amplification for responsive discrimination, we summarize progress in developing sensing arrays based on diverse micro/nanomaterials and their integrated devices for multi-analyte discrimination. An overview of strategies for constructing sensing arrays through various micro or nano building blocks, including 0D nanoparticle assembly and modification, 1D nanowires and fibers, 2D graphene and textiles, is schematized. Then, portable and wearable devices integrating colorimetric sensors or flexible electrochemical electrodes with the newest microelectronic units and circuit boards are presented. Meanwhile, the latest artificial intelligence (AI) algorithms are introduced for massive data analysis in complex biological and environmental systems. With future developments in facile and accurate discrimination for multi-analyte research, extended applications will gear up in various fields.
