Abstract:Corn and wheat cultivated in hydroponic culture solution with supplies of cadmium (c(Cd)0, 4.0 μmol/L) and phosphorus (c(P)0, 0.12, 0.6, 3.0 mmol/L) under controlled growth conditions in relation to the chemical forms of Cd, growth and physiological parameters, and enzyme activities of 21-day-old plants were investigated. The three predominant forms of Cd were NaCl-extractable Cd (related to Cd-binding proteins or polypeptides), acetic acid-extractable Cd (CdHPO4, Cd3(PO4)2, or Cd-polyphosphates), and water-extractable Cd (organic acid complexes). Increasing application of P tended to reduce the percentage of Cd combined with proteins and increase the amount of Cd-phosphates. Decreased cell plasma membrane permeability and K+ leakage of Cd-treated plants were observed with the P supply of 0.6 mmol/L. The moderate supply of P (0.6 mmol/L) also improved the contents of chlorophyll pigments, the activities of root dehydrogenase and leaf nitrate reductase in Cd-treated plants, whereas supply with an excess of P (3.0 mmol/L) showed not to be more effective in the alleviation of Cd toxic to plants. Alleviating the toxic effects of Cd and Co on the seed germination and seedling biochemistry of wheat (Triticum aestivum L.) using Azolla pinnata

Abstract:One of the most significant environmental challenges in the twenty-first century is heavy metal pollution. The potential use of fresh Azolla pinnata to alleviate the toxic effects of Cd and Co on the germination measurements of wheat seeds (Triticum aestivum L.) and the biochemistry of seedlings was studied. Two concentrations (80 and 100 mg L-1 solutions) of CdNO3 and CoCl2 were used before and after treatment with A. pinnata. The highest removal efficiency (RE) by A. pinnata was obtained on the fifth day, with a Cd RE = 55.9 and 49.9% at 80 and 100 mg L-1, respectively. Cadmium and cobalt solutions reduced the germination percentage, and the measured variables of wheat seeds meanwhile increased the radicle phytotoxicity. In contrast, the presence of A. pinnata in the germination medium increased all the measured variables and decreased radicle phytotoxicity. At 80 and 100 mg L-1, Cd significantly reduced the fresh and dry biomass, and height of wheat seedlings after 21 days of cultivation compared to Co. Cadmium and high concentrations of cobalt increased the contents of H2O2, proline, MDA, phenolic, and flavonoid compounds. The application of treated Cd and Co solutions by A. pinnata showed a decrease in H2O2, proline, phenolic, and flavonoid compounds levels accompanied by a reduction in catalase and peroxidase activities compared to the control. This study showed the positive role of A. pinnata in alleviating the metal impacts, particularly Cd, on the seedling growth of wheat and its germination.

Abstract:Melatonin is a growth regulator that improves the growth and chlorophyll (chl) content in plants. This study aims to investigate the effect of melatonin pretreatment on chl synthesis and fluorescence parameters in Malva parviflora exposed to cadmium (Cd). The 42-day-old plants were transferred to nutrient solutions containing 50 μM melatonin. After two days, some plants were exposed to 50 μM Cd. Eight days after Cd treatment, some indicators related to chl fluorescence and some biochemical parameters were measured. In this study, melatonin increased chl content and chl a /pheophytin a (pheo a ) ratio, chlorophyllide a (chlide a ), porphyrin compounds, and 5-aminolevulinic acid (5-ALA) in the presence of Cd. However, it decreased chl a /chlide a ratio under these conditions. Whereas Cd treatment resulted in significant reductions in photochemical activity and electron transfer rate in PSII, melatonin improved photochemical efficiency of PSII by reducing the toxic effect of Cd on the activity of the oxygen evolving complex (OEC) on the electron donor site and reducing non-photochemical quenching (NPQ). Based on the results, it appears that melatonin can maintain the chl content of plants exposed to Cd by increasing the precursors of the chl biosynthesis pathway and reducing its degradation rate. These results may, at least in our experimental conditions, partly explain the reason for the improved yield and growth of Cd-exposed plants when pretreated with melatonin.

Abstract:Hydrogen sulfide (H2S) plays prominent multifunctional roles in the mediation of various physiological processes and stress responses to plants. In this study, hydroponic experiments were carried out to explore the effects of NaHS pretreatment on the growth of wheat (Triticum aestivum L.) under 50 μM cadmium (Cd). Compared with Cd treatment alone, 50 μM NaHS pretreatment increased the plant height, soluble sugar content of shoots and roots, and dry weight of shoots and roots under Cd stress, while the Cd concentration of shoots and roots was significantly reduced by 18.1% and 25.9%, respectively. Meanwhile, NaHS pretreatment protected the photosynthetic apparatus by increasing the net photosynthetic rate and PSII electron transportation rate of wheat leaves under Cd stress. NaHS pretreatment significantly increased the soluble sugar content to maintain the osmotic pressure balance of the leaf cells. The gene expression results associated with photosynthetic carbon assimilation and sucrose synthesis in wheat leaves suggested that the NaHS pretreatment significantly up-regulated the expression of TaRBCL, TaRBCS, and TaPRK, while it down-regulated the expression of TaFBA, TaSuSy, TaSAInv, and TaA/NInv. In summary, NaHS pretreatment improved the resistance of wheat seedlings under Cd stress by increasing the rate of photosynthesis and regulating the expression of genes related to sugar metabolism.

Abstract:Heavy metal contamination is one of the most important abiotic stresses affecting physiological activities of plants. We investigated the effects of cadmium (Cd) and lead (Pb) on chlorophyll fluorescence (Fv/Fm, Fo, and Fm), photosynthetic pigments (chlorophyll a and b), and proline in one-year-old seedlings of Robinia pseudoacacia. The seedlings were treated twice over a period of 10 days with Cd and Pb at concentrations of 0, 250, 500, 1000 and 2000 mg L−1. Saline solution containing Cd and Pb was sprayed on the leaves. Chlorophyll and proline contents were measured after 10 days. Chlorophyll fluorescence of R. pseudoacacia was affected slightly by high concentrations (1000, 2000 mg L−1) of Cd and Pb. Chlorophyll a and a/b increased at 1000 and 2000 mg L−1 of Cd and proline content of leaves was similar in all treatments of Cd and Pb. Our results indicated that photosynthetic sensitivity of R. pseudoacacia to Cd and Pb contamination was weak. Photosystem II chlorophyll pigments were not damaged by Pb and Cd stress. We conclude that chlorophyll fluorescence along with chlorophyll and proline contents are useful indicators of Cd and Pb stresses in R. pseudoacacia which widely planted in urban polluted regions in Iran.

Abstract:Cadmium (Cd) is toxic to plants and other living beings present in the next trophic level. It inhibits growth of the plant. Since Brassica juncea is a hyperaccumulator plant for Cd, its consumption is hazardous to living beings. Hence, it is important to reduce the accumulation of Cd inside the foliage of mustard plants to make it safe for consumption and reduce bio-magnification. 24-Epibrassinolide (EBL) was applied in various doses (10 –10 , 10 –8 or 10 –6 M) as a foliar spray at 25–30 days stage of growth on Brassica juncea var. Varuna, growing in Cd (25 or 50 mg Kg −1 of soil) amended soil. The sampling was done at 45 and 60 days after sowing (DAS) to determine photosynthesis, nutrients, antioxidants, cell death, enzymatic activities, reactive oxygen species, carbohydrate, and Cd content. EBL restricted the Cd uptake and translocation of Cd from root to shoot and induced defence responses to protect photosynthetic machinery. Out of the three doses, EBL (10 –8 M) proved most effective and the effect was more pronounced against lower concentration (25 mg Kg −1 soil) of Cd which persisted at both the stages of sampling. The study suggests that EBL alleviated the adverse effects generated by Cd on physiology of the plant.

Abstract:The biosphere pollution with the heavy metals (HM) has increased significantly in recent decades due to human activity. Plants can accumulate and concentrate HM, which negatively affects their growth, productivity and quality of agricultural products. Some HM, such as copper, belong to the group of biogenic elements that, in low concentrations, are essential for the normal functioning of plant organisms. Other HM such as cadmium are toxic even in low concentrations. The toxicity of HM is related to oxidative damage. In the plant cell, the antioxidant system provides protection against this kind of stress. However, data on changes in antioxidant enzyme activities in the early stage of the cellular response to HM-induced stress remain scarce. Therefore, we focused our research on studying peroxidase (POD) activity changes in Arabidopsis thaliana under conditions of rapid uptake of copper and cadmium ions into leaf tissue. For the experiments, 4.5–5-week-old A. thaliana plants were used. The plants were incubated on 0.5x MS liquid medium containing copper or cadmium chloride at concentrations of 0.1, 0.5 and 5 mM. The HM salt treatment was carried out in the dark at 20 °C for 2 (short-term stress) and 12 (long-term stress) hours. After that, the leaves were frozen and the POD activity was measured. Evaluation of the effects of Cd2+ and Cu2+ ions shows that these HM cause a decrease in POD activity after 2 hours and its increase after 12 hours of treatment. Therefore, modulation of POD activity is a component of the HM stress response in A. thaliana. Analysis of the available data revealed that the enzymes POD and CAT, which eliminate hydrogen peroxide, can partially replace each other and thus provide cellular protection in different phases of the stress response.

Abstract:Assessing the impact of Cadmium (Cd) on plant cells requires an understanding of the defensive mechanisms and adaptive responses employed by plants to counteract the deleterious effects of Cd toxicity. Wheat seedlings were grown under different concentrations (0 μM, 100 μM and 500 μM) of Cadmium salt (CdCl2). The growth of metal-treated seedlings was significantly affected. This study evaluated the ecotoxicological effects of two experimental concentrations of Cadmium (100 μM and 500 μM) against water control. It has been examined in recent studies that the toxic effects of Cadmium comprising the variables in fresh weight, dry weight and germination percentage of the wheat seedlings, photosynthesis and enzymatic activities in different stresses after seven days of cadmium exposure on roots and shoots of wheat seedling. Different antioxidant enzyme activities such as catalase (CAT), ascorbic acid oxidase (AOX), catechol peroxidase (CPX), superoxide dismutase (SOD), acid and alkaline phosphatase and inorganic pyrophosphatase have been measured. The various enzyme activities were increased in roots treated with high concentrations (500 μM) of Cadmium but decreased at low concentrations (100 μM) with respect to water control. This result indicated a complex defense mechanism in the root tissues induced by Cadmium. This work provides valuable insights for future research on plant responses to heavy metal stress and advances our understanding of the complex relationship between Cd toxicity and the antioxidant defense system in wheat seedlings.
Comparative study of alleviating effects of GSH, Se and Zn under combined contamination of cadmium and chromium in rice (Oryza sativa)

Abstract:A hydroponic experiment was conducted to study the ameliorative effects of separate or combined application of exogenous glutathione (GSH), selenium (Se) and zinc (Zn) upon 20 μM cadmium (Cd) plus 20 μM chromium (Cr) heavy metal stress (HM) in rice seedlings. The results showed that HM caused a marked reduction in seedling height, chlorophyll content (SPAD) and biomass, and activities of catalase (CAT) and ascorbate peroxidase (APX) in leaves and H+-ATPase in roots/leaves, but elevated superoxide dismutase (SOD) and guaiacol peroxidase (POD) activities in leaves with elevated malondialdehyde (MDA) accumulation both in leaves and roots over the control. The best mitigation effect was recorded in HM+GSH+Zn and HM+GSH (addition of GSH+Zn and GSH to HM solution), which greatly alleviated HM-induced growth inhibition and oxidative stress. Compared with HM alone, HM+GSH and HM+GSH+Zn markedly reduced Cr uptake and translocation but not affected Cd concentration; improved H+-ATPase activity and Fe, Zn, Mn uptake and translocation, and repressed MDA accumulation. Meanwhile exogenous GSH and GSH+Zn counteracted HM-induced response of antioxidant enzymes, via suppressing HM-induced dramatic increase of root/leaf SOD and leaf POD activities, and elevating stress-depressed leaf APX and leaf/root CAT activities.

Abstract:This research aimed to clarify the effects of exogenously applied chitosan on the physiological characteristics, antioxidant activities, and Cd accumulation of wheat (Triticum aestivum L.) seedlings under cadmium (Cd) stress and to identify the key indicators based on the partial least squares model. The wheat variety studied was Bainong207 (BN207), and Cd-stress was achieved by growing seedlings in a hydroponic culture experiment with 10 and 25 μmol·L-1 Cd2+ added to the culture solution. It was found that both Cd-stress at 10 and 25 μmol·L-1 significantly inhibited the chlorophyll content, photosynthesis, and biomass accumulation of wheat seedlings. Seedling roots became shorter and thicker, and the lateral roots decreased under Cd-stress. The Cd-stress also increased H2O2 and MDA accumulation and the degree of cell membrane lipid peroxidation and affected the activities of antioxidant enzymes such as superoxide dismutase (SOD) and peroxidase (POD). Under Cd stress, exogenous chitosan decreased the Cd content in the aboveground and underground parts of wheat by 13.22 %-21.63 % and 7.92 %-28.32 % and reduced Cd accumulation in the aboveground and underground parts by 5.37 %-6.71 % and 1.91 %-4.09 %, respectively. Whereas exogenous chitosan application significantly reduced the content of H2O2 in roots and aboveground parts of wheat by 38.21 %-47.46 % and 45.81 %-55.73 % and MDA content by 37.65 %-48.12 % and 29.87 %-32.51 %, it increased the activities of SOD and POD in roots by 2.78 %-5.61 % and 13.81 %-18.33 %, respectively. In summary, exogenous chitosan can improve the photosynthetic characteristics and antioxidant enzyme activities of wheat seedlings under Cd stress, reduce the content and accumulation of Cd in the root and aboveground parts of wheat, and alleviate the damage of lipid peroxidation to the cell membrane. All of these results provide the basal data for the application of exogenous chitosan to alleviate Cd toxicity to wheat seedlings.

Abstract:Selenium (Se), iron (Fe), and humic acid (HA) are beneficial fertilizers that inhibit cadmium (Cd) uptake in crops and are crucial for agricultural yields as well as human health. However, the joined effect of Se, Fe, and HA on Cd uptake in rice are still poorly understood. Therefore, a hydroponic culture experiment was established to evaluate the combined effect of Se (Se4+ or Se6+), Fe, and HA on the biomass, Cd uptake, and Cd translocation of/in rice seedlings. Compared to Se6+ application, Se4+ application in most treatments resulted in lower Cd translocations from roots to shoots, leading to a significant decrease in shoot Cd concentrations. Compared to the treatments with Se4+ or Fe2+ application, joined application of Se4+ and Fe2+ inhibited Cd uptake in shoots by decreasing Cd adsorption onto (iron plaque) and uptake by roots, and alleviating Cd translocation from root to shoot. Compared to the treatments with Se6+ or Fe2+ application, joined application of Se6+ and Fe2+ inhibited Cd uptake in shoots by sequestering (retaining) Cd onto root surface (iron plaque). HA inhibited Cd uptake in all treatments by decreasing the bioavailability of Cd in the nutrient solution through complexation. The simultaneous application of Se, Fe, and HA decreased the shoot Cd concentrations the most, followed by the combined application of two fertilizers and their individual application; the mean shoot Cd concentration in the Fe-SeIV-HA2 treatment was the lowest among all the treatments, at only 11.39 % of those in the control treatments. The 3-way ANOVA results indicated that the Cd concentrations in shoots were significantly affected by Se, Fe, HA, and certain of their interactions (Fe×Se and Se×HA) (p< 0.05). The above findings suggest that the joined application of Se, Fe, and HA ameliorated Cd uptake mainly by inhibiting Cd adsorption onto (iron plaque) and uptake by roots and the translocation from roots to shoots (Fe×Se4+), retaining (sequestering) Cd in iron plaque (Fe×Se6+), and decreasing Cd availability in nutrient solution (HA).Graphical abstract

Abstract:Oxidative stress caused by heavy metals (HMs) has been considered as a big threat for the production of crop plants. Several emerging studies recommend that some signaling molecules could help mitigate the adverse effects of HMs. This study tested the interactive effects of H 2 S, and melatonin MT, on lead (Pb) homeostasis and Pb- induced oxidative injury in safflower seedlings. Seedlings from 21-day-old safflower plants were subjected to 50 μM PbNO 3 individually and/or mixed with 100 μM MT and 100 μM NaHS, as H 2 S donor. The data showed that MT, NaHS, and especially MT+NaHS treatments were able to significantly reduce MDA and H 2 O 2 content and lipoxygenase activity, indicating the important role of MT and H 2 S in alleviating oxidative stress generated by Pb toxicity. By prompting the activity of enzymes responsible for the ascorbate-glutathione cycle, MT- and NaHS ameliorated the oxidative stress caused by Pb on safflower seedlings. This indicates that H 2 S and MT can prevent oxidative stress by assisting the ascorbate-glutathione cycle. Based on phytochelatin (PC) analysis of safflower plants exposed to Pb stress, MT and NaHS seem to be able to reduce oxidative stress caused by Pb by stimulating metal-binding ligands metabolism. The results of our study suggest that even though H 2 S and MT alone might ameliorate Pb-induced toxicity in seedlings of safflower plants, their combined action could substantially ameliorate Pb toxicity by reducing Pb absorption and transmission, and reducing oxidative stress.

Abstract:Heavy metals like cadmium (Cd), mercury (Hg), bismuth (Bi), and arsenic (As) are potent and harmful poisonous sources that cause havoc on health conditions for the population of the world. However, the response of our crop species to these potent heavy metals-related toxicity is still left to be fully understood. It is a matter of great concern, as we are heavily dependent on crop species like rice, wheat, peas, etc. Our study here aims to learn about the defensive mechanism of Pisum sativum L. aided with putrescine and mycorrhiza against the stress created by Cd-related toxicity. We quantified physiological parameters such as the membrane-related injury and stability index. We further measured the total free proline content, lipid peroxidation content, and SOD activity. We executed our quantitative experiments on the stressed pea plants due to the exogenously applied Cd-toxicity in the presence and absence of mycorrhiza and putrescine. Insights of our significant results will improve the understanding of readers of the role of mycorrhiza and putrescine in improvising the tolerance level of a pea plant over Cd-related toxicity.
Melatonin alleviates cadmium phytotoxicity through regulation of growth, photosynthesis, and antioxidant potential in two pepper genotypes

Abstract:The global agricultural productivity has been significantly impaired due to the extensive use of heavy metal. Cadmium (Cd) is now recognized as a significant soil and environmental contaminant that is primarily spread by human activity. This study investigates the possible impact of melatonin (ME) in mitigating the toxicity caused by Cd in pepper ( Capsicum annuum L.) seedlings. There were three groups of plants used in the experiment: control (CK) plants, Cd-stressed plants and ME-pretreated + Cd-stressed plants. The concentration of ME and Cd was 1 μM and 0.1 mM, respectively, and applied as root application. The results described that Cd treatment remarkably reduced growth parameters, impaired pigment concentration, hindered gas exchange traits. In contrast, ME supplementation significantly recovered these parameters by increase in growth and biomass production of pepper seedlings under Cd toxicity. In addition, ME application considerably increased osmolyte production and protein level in pepper leaves and roots. Furthermore, ME positively upregulated the antioxidant enzymes activity and effectively decreased the oxidative damage in pepper leaves and roots. The enhanced antioxidant enzymes activity performed a significant role in the reduction of H 2 O 2 and MDA concentration in plants under Cd stress. The findings indicated that the application of ME to plants effectively alleviates the stress caused by Cd exposure. Moreover, ME demonstrates significant efficacy in mitigating the adverse impacts of Cd on pepper plants.

Abstract:Cadmium (Cd) is a common agricultural soil pollutant, which does serious harm to the environment and the human body. In this study, Brassica juncea was treated with different concentrations of CdCl2 and Na2SeO3. Then, physiological indexes and transcriptome were measured to reveal the mechanisms by which Se reduces the inhibition and toxicity of Cd in B. juncea. The results showed that Se alleviated the inhibitive Cd effects on seedling biomass, root length, and chlorophyll, and promoted the adsorption of Cd by pectin and lignin in the root cell wall (CW). Se also alleviated the oxidative stress induced by Cd, and reduced the content of MDA in cells. As a result, SeCys and SeMet alleviated the transport of Cd to the shoots. Transcriptome data showed that the bivalent cation transporter MPP and ABCC subfamily participated in the separation of Cd in vacuoles, CAL1 was related to the chelation of Cd in the cytoplasm of cells, and ZIP transporter 4 reduced the transport of Cd to the shoots. These results indicated that Se alleviated the damage of Cd in plants and decreased its transport to the shoots by improving the antioxidant system, enhancing the ability of the CW to adsorb Cd, reducing the activity of Cd transporters, and chelating Cd.

Abstract:In the present study, role of hydrogen sulfide (H2S) in alleviating cadmium (Cd) induced stress in oilseed rape (Brassica napus L.) was studied under greenhouse conditions. Plants were grown hydroponically under three levels (0, 100, and 500µM) of Cd and three levels (0, 100 and 200µM) of H2S donor, sodium hydrosulfide (NaHS). Results showed that application of H2S significantly improved the plant growth, root morphology, chlorophyll contents, elements uptake and photosynthetic activity in B. napus plants under Cd stress. Moreover, addition of H2S reduced the Cd concentration in the leaves and roots of B. napus plants under Cd-toxicity. Exogenously applied H2S decreased the production of malondialdehyde and reactive oxygen species in the leaves and roots by improving the enzymatic antioxidant activities under Cd stress conditions. The microscopic examination indicated that application of exogenous H2S improved the cell structures and enabled a clean mesophyll cell having a well developed chloroplast with thylakoid membranes, and a number of mitochondria could be observed in the micrographs. A number of modifications could be found in root tip cell i.e. mature mitochondria, long endoplasmic reticulum and golgibodies under combined application of H2S and Cd. On the basis of these findings, it can be concluded that application of exogenous H2S has a protective role on plant growth, photosynthetic parameters, elements uptake, antioxidants enzyme activities and ultrastructural changes in B. napus under high Cd stress conditions.

Abstract:In view of damaging impacts of cadmium (Cd) toxicity on various vital processes of plants and strategies for alleviating these effects, selenium (Se) application has been recently achieved great attention. In addition, chitosan (CS) and its nano-form, besides many positive effects on plants, could be considered as an excellent adsorption matrix and a carrier for a wide range of materials like Se with various applications in agricultural sector. For that point, the combination nano-form of Se and CS (CS-Se NPs), using CS as a carrier and control releaser for Se, could enhance Se efficiency particularly at lower doses under stress conditions. Therefore, Se (10 mg L-1), CS (0.1%) and CS-Se NPs (in two concentrations; 5 and 10 mg L-1) were applied on Moldavian balm plant under 0, 2.5 and 5 mg kg-1 Cd-stress conditions. The results demonstrated that mostly Se and CS-Se NPs treatments could lessen negative effects of Cd-stress conditions through enhancing agronomic traits, photosynthetic pigments, chlorophyll fluorescence parameters and SPAD, proline, phenols, antioxidant enzymes activities and some dominant constituents of essential oils and decreasing MDA and H2O2. These encouraging impacts were more significant at lower dose of CS-Se NPs (5 mg L-1) introducing it as the best treatment to ameliorate Moldavian balm performance under Cd-stress conditions. In conclusion, CS-Se NPs could be considered as a supportive approach in plant production mainly under different heavy metal stressful conditions and probably a potential plant growth promoting and stress protecting agent with new outlooks for applying in agricultural sector.
