Abstract:Salinity and cadmium (Cd) contamination of soil are serious environmental issues threatening food security. This study investigated the role of salicylic acid (SA) and potassium (K) in enhancing the resilience of quinoa against the combined stress of salinity and Cd. Quinoa plants were grown under NaCl (0, 200 mM) and Cd (0, 100 μM) stress, with the addition of 0.1 mM SA and 10 mM K, separately or in combination. The joint stress of Cd and NaCl caused >50% decrease in plant growth, chlorophyll contents, and stomatal conductance compared to the control plants. The higher accumulation of Na and Cd reduced the uptake of K in quinoa tissues. The joint stress of salinity and Cd caused an 11-fold increase in hydrogen peroxide and 13-fold increase in thiobarbituric acid reactive substances contents, and caused a 61% decrease in membrane stability. An external supply of 0.1 mM SA and 10 mM K helped plants to better adapt to salinity and Cd stress with less of a reduction in plant biomass (shoot 19% and root 24%) and less accumulation of Na and Cd in plant tissues. The activities of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX) were enhanced by 11-fold, 10-fold, 7.7-fold, and 7-fold, respectively, when SA and K were applied together to the plants subjected to the joint stress of Cd and salinity. Based on the values of the bioconcentration factor (>1), the translocation factor (<1), and the higher tolerance index, it was clear that Cd-contaminated, salty soils could be stabilized with quinoa under the combined supply of SA and K.

Abstract:Cadmium (Cd) and lead (Pb) contaminated soils have increased recently, resulting in limited crop productivity. The ameliorative role of potassium (K) and silicon (Si) is well established in plants under heavy metals stress; however, their combined role under the co-contamination of Cd and Pb is not well understood. We hypothesized that the synergistic application of K and Si would be more effective than their sole treatment for increasing the Pb and Cd tolerance and phytostabilization potential of quinoa (Chenopodium quinoa Willd.). In the current study, quinoa genotype ‘Puno’ was exposed to different concentrations of Cd (0, 200 μM), Pb (0, 500 μM) and their combination with or without 10 mM K and 1.0 mM Si supplementation. The results revealed that the combined stress of Cd and Pb was more detrimental than their separate application to plant biomass (66% less than the control), chlorophyll content and stomatal conductance. Higher accumulation of Pb and Cd led to a limited uptake of K and Si in quinoa plants. The supplementation of metal-stressed plants with 10 mM K and 1.0 mM Si, particularly in combination, caused a significant increase in the growth, stomatal conductance and pigment content of plants. The combined stress of Cd and Pb resulted in an overproduction of H2O2 (11-fold) and TBARS (13-fold) and a decrease in membrane stability (59%). Oxidative stress induced by metals was lessened by 8-fold, 9-fold, 7-fold and 11-fold increases in SOD, CAT, APX and POD activities, respectively, under the combined application of K and Si. It is concluded that the exogenous supply of K and Si in combination is very promising for increasing Cd and Pb tolerance and the phytostabilization potential of quinoa.

Abstract:Cadmium (Cd) contamination and soil salinity are the main environmental issues reducing crop productivity. This study aimed to examine the combined effects of salinity (NaCl) and Cd on the physiological and biochemical attributes of quinoa (Chenopodium quinoa Willd.). For this purpose, 30-day-old plants of quinoa genotype "Puno" were transplanted in Hoagland's nutrient solution containing diverse concentrations of Cd: 0, 50, 100, 200 µM Cd, and salinity: 0, 150, and 300 mM NaCl. Results demonstrated that plant growth, stomatal conductance, and pigment contents were significantly lower at all Cd concentrations than the control plants. Quinoa plants exhibited improved growth and tolerance against Cd when grown at a lower level of salinity (150 mM NaCl) combined with Cd. In contrast, the elevated concentration of salinity (300 mM NaCl) combined with Cd reduced shoot and root growth of experimental plants more than 50%. Combined application of salinity and Cd increased Na (25-fold), while lessened the Cd (twofold) and K (1.5-fold) uptake. A blend of high concentrations of Na and Cd caused overproduction of H2O2 (eightfold higher than control) contents and triggered lipid peroxidation. The activities of antioxidant enzymes: ascorbate peroxidase (APX), catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD) were 13, 12, 7 and ninefold higher than control to mitigate the oxidative stress. Due to restricted root to shoot translocation, and greater tolerance potential against Cd, the quinoa genotype, Puno, is suitable for phytostabilization of Cd in saline soils.

Abstract:Contamination of soils with cadmium (Cd) and lead (Pb) has emerged as a serious environmental issue that reduces crop productivity. However, the metals tolerance and accumulation potential of quinoa (Chenopodium Quinoa Willd) under the combined stress of Cd and Pb has not yet been explored. In the present hydroponic study, the physiological and biochemical characteristics of quinoa exposed to Cd and Pb were explored. Four-week-old plants of quinoa genotype ‘Puno’ were grown under different concentrations of Cd (0, 50 and 100 μM), Pb (0, 250 and 500 μM) alone as well as in combinations. The results showed that with increasing Cd and Pb levels in the nutrient solution, the plant biomass, stomatal conductance and chlorophyll contents were decreased. However, the concurrent application of higher concentrations of Cd (100 μM) and Pb (500 μM) caused even more reduction in the plant biomass (more than 50% than the control) and physiological attributes. The combined application of Pb and Cd caused oxidative stress through an overproduction of H2O2 (10-fold) and TBARS (12.5-fold), leading to decrease in membrane stability (52%). The oxidative stress was alleviated by a 7-fold, 10-fold and 9-fold overactivation of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), respectively. An excessive uptake of Cd resulted in a limited uptake of Pb and K in the roots and shoots of quinoa plants. The Cd and Pb tolerance and uptake potential of Puno showed its ability to stabilize Cd and Pb in co-contaminated soils.

Abstract:To assess the potential role of salicylic acid (SA) in plants under cadmium (Cd) stress, a study was conducted on three different oilseed rape (Brassica napus) genotypes. Gas exchange parameters, photosynthetic pigments, antioxidant enzymes activities, mineral nutrients concentration, and ultrastructural analysis were carried out for assessment. Interestingly, cadmium treatment reduced gas exchange parameters, photosynthetic pigments, mineral elements [calcium (Ca), magnesium (Mg), and iron] and the activity of catalase (CAT) enzyme. Whereas, a pronounced increase was observed in malondialdehyde content and antioxidant enzymes, cadmium and zinc accumulation. Impressively, SA played its role as an alleviatory agent by reducing the damage to all parameters caused by Cd except internal CO2 concentration which was further decreased by SA. Even more, Cd and SA showed synergistic effects by increasing superoxide dismutase, peroxidases, ascorbate peroxidase, guaiacol peroxidase, glutathione reductase and decreasing CAT. Apart from affecting the enzyme activities of oilseed rape, both Cd and SA had the same inhibitory effects on Ca and Mg concentrations. Damage caused by Cd to chloroplast and other internal organelles were almost rectified by SA. Effective role of SA, in alleviating Cd toxicity, could be attributed to the SA-induced improvement of photosynthetic activities, enhancement of antioxidant enzymes activities, reduction in lipid peroxidation and Cd uptake. These findings reflect the possible role of SA as a potential inhibitor of cadmium toxicity by strengthening the internal immunity in oilseed rape.
Selective synergistic effects of oxalic acid and salicylic acid in enhancing amino acid levels and alleviating lead stress in Zea mays L

Abstract:Lead is one of the major environmental pollutants which is highly toxic to plants and living beings. The current investigation thoroughly evaluated the synergistic effects of oxalic acid (OA) and salicylic acid (SA) on Zea mays L. plants subjected to varying durations (15, 30, 30, and 45 days) of lead (Pb) stress. Besides, the effects of oxalic acid (OA) combined with salicylic acid (SA) for different amino acids at various periods of Pb stress were also investigated on Zea mays L. The soil was treated with lead nitrate Pb (NO3)2 (0.5 mM) to induce Pb stress while the stressed plants were further treated using oxalic acid (25 mg/L), salicylic acid (25 mg/L), and their combination OA + SA (25 mg/L each). Measurements of protein content, malondialdehyde (MDA) levels, guaiacol peroxidase (GPOX) activity, catalase (CAT) activity, GSH content, and Pb concentration in maize leaves were done during this study. MDA levels increased by 71% under Pb stress, while protein content decreased by 56%, GSH content by 35%, and CAT activity by 46%. After treatment with SA, OA, and OA+SA, there was a significant reversal of these damages, with the OA+SA combination showing the highest improvement. Specifically, OA+SA treatment led to a 45% increase in protein content and a 39% reduction in MDA levels compared to Pb treatment alone. Moreover, amino acid concentrations increased by 68% under the Pb+OA+SA treatment, reflecting the most significant recovery (p < 0.0001).
Foliar application of salicylic acid improves growth and yield attributes by upregulating the antioxidant defense system in Brassica campestris plants grown in lead-amended soils

Abstract:Lead (Pb) toxicity causes a severe impact on plant growth and productivity. A protective role of salicylic acid (SA) is well known under different abiotic stress conditions. However, very little is known about the SA-induced Pb resistance mechanism. In this study, we investigated the effect of SA on mustard plants (Brassica campestris L.) under Pb-stress conditions. Plants were exposed to three levels of Pb amendment to the soil (0.25, 0.50, 1.00 mM), with or without SA (0.25 mM). Plant growth, yield attributes, and yield at harvest were reduced depending on the severity of the Pb stress. Exogenous application of SA improved plant growth and yield. Biochemical data revealed that Pb toxicity resulted in higher oxidative damage by reducing nonenzymatic antioxidants such as ascorbate and glutathione at the higher dose of Pb treatment. Antioxidant enzymes (ascorbate peroxidase – APX, monodehydroascorbate reductase – MDHAR, dehydroascorbate reductase – DHAR, glutathione reductase – GR, guaiacol peroxidase – POD, glutathione S-transferase – GST, and catalase – CAT) responses varied with the Pb doses. Both the nonenzymatic and enzymatic components of the antioxidant defense system were upregulated after application of SA, resulting in lower oxidative damage under Pb-stress conditions. Taken together, the results suggest that exogenous application of the SA mitigates Pb-induced oxidative damage and consequently results in better growth and yield in mustard plants.

Abstract:This study was undertaken to determine the effect of potassium (K) on alleviating cadmium (Cd) toxicity in soybean. Two genotypes of soybean, namely 'Liao 1' and 'Zhechun 3' were used in hydroponics experiment with the following treatments: control without Cd addition; 1M Cd addition; K supplementation at a rate of 380 mg L-1; and both Cd addition and K supplementation. Plant growth, chlorophyll content, and fluorescence, rate of photosynthesis and stomatal conductance reduced significantly in Cd-stressed plants. Meanwhile, Cd treatment increased malondialdehyde content, and activities of superoxide dismutase and peroxidase enzymes. Concentration of K, calcium (Ca), magnesium (Mg), and zinc (Zn) in shoot and root tissues also reduced in Cd treatment. 'Liao 1' had higher, antioxidant enzyme activity than 'Zhechun 3'. Potassium supplementation alleviated the reduction of growth, photosynthesis and nutrients uptake in Cd-treated plants. It was concluded that Cd toxicity could be alleviated through enhanced K nutrition in soybean.
Salicylic Acid Manipulates Ion Accumulation and Distribution in Favor of Salinity Tolerance in Chenopodium quinoa

Abstract:Although the effects of salicylic acid (SA) on increasing plant growth in saline conditions have been well known, the mechanisms of induction of salinity tolerance, especially in quinoa (Chenopodium quinoa Willd.), are not fully understood. In the present work, two quinoa genotypes (Titicaca and Giza1) were treated with different SA concentrations (0, 0.75, and 1.5 mM) under varied irrigation water salinities (0, 7, 14, and 21 dS m−1). Salinity decreased shoot and root growth, potassium (K+) concentration, and potassium to sodium ratio (K/Na) and increased sodium (Na+) and chlorine (Cl−) concentrations in both cultivars. Calcium (Ca2+) and magnesium (Mg2+) concentrations increased in 7 dS m−1 but decreased in higher salinities. The growth and salinity tolerance of Giza1 were higher, while the growth of Giza1 increased and of Titicaca decreased in high salinity. Salicylic acid at 0.75-mM concentration increased shoot and root growth and improved the ions concentration in favor of the plant, while the 1.5-mM concentration either had no significant effect or had a negative impact. The ions distribution estimated by K/Na selectivity and storage factor (SF) indicated quinoa accumulated more ions in roots under saline conditions. Salicylic acid increased NaSF, ClSF, and MgSF and decreased KSF and CaSF, meaning less Na+, Cl−, and Mg2+ and more K+ and Ca2+ transferred to shoots in SA-treated plants. Importantly, Giza1, as the more tolerant cultivar, had higher NaSF and ClSF and lower KSF, CaSF, and MgSF. In general, the concentrations of ions in roots were higher than in shoots. The results indicated more ions accumulation in the root could be one of the most important mechanisms of salinity tolerance in quinoa, and the more tolerant cultivar (Giza1) transferred less Na+ and Cl− and more K+ and Ca2+ and Mg2+ to the shoot.

Biochar increases salt tolerance and grain yield of quinoa on saline-sodic soil: multivariate comparison of physiological and oxidative stress attributes

Abstract:PurposePlant growth and grain yield of most of the crops are reduced on saline-sodic soils. Different organic and inorganic amendments have been used to increase plant growth and productivity on salt-affected soils. However, the role of cotton shell biochar (CSBC) for increasing salt tolerance of plants grown on saline-sodic soils has not been investigated yet. Therefore, the current study was intended to estimate the impact of cotton shell biochar (CSBC) addition @1 and 2% for ameliorating Na-induced phytotoxicity in quinoa grown on saline-sodic soil.Materials and methodsQuinoa plants were grown on saline-sodic soil with and without CSBC addition. Different physiological attributes were measured at vegetative stage. Plants were harvested at maturity when growth and grain yield were recorded.Results and discussionResults revealed that plant biomass, grain yield, stomatal conductance, relative water contents, and chlorophyll contents of quinoa were declined by > 20%, 18%, 23%, 27%, and 16%, respectively under salt stress. Excessive uptake of Na decreased the uptake of K and K:Na, and caused the overproduction of H2O2 (62%) and lipid peroxidation (58%) in quinoa. The CSBC addition @2% was more effective than 1% in ameliorating the negative impacts of Na toxicity on plant growth and physiology. Amendment of saline-sodic soil with 2% CSBC increased plant growth, water contents, stomatal conductance, and chlorophyll contents by 60%, 36%, 41%, and 43%, respectively as compared to saline-sodic soil without CSBC. There was a slight increase in soil EC after the addition of CSBC. Additionally, 2% CSBC limited the uptake of Na and caused a 43%, 48%, and 75% increase in superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) activities to ameliorate the oxidative stress.ConclusionsIt was concluded through multivariate analysis that 2% CSBC was very promising in reducing Na toxicity and increasing plant biomass and grain yield of quinoa in saline-sodic soils.

Abstract:Heavy metal (HM) pollution has become a serious issue globally, endangering both human health and agricultural production, particularly in acidic soils with high HM bioavailability. Although the exact mechanism of either rice husk biochar (RH) or salicylic acid (SA) in lowering HM levels in soil and plants has been discovered, their combined effect remains unknown. Therefore, this study mitigated HM stress and reduced their content in the Chinese mustard plants by adding different levels of RH alone or with foliar application of SA to produce safe foods. RH was added to the soil at 0%, 2.5%, and 5% either alone or with foliar application of SA, which was applied to the plant leaves at concentrations of 0, 0.5, and 1 mM in triplicate for each. The highest reductions of the DTPA-extractable metals were recorded in a high level of RH (5%), which were 32.16%, 43.22%, 49.72%, and 29.54% for Pb, Cd, Cu, and Zn, respectively, compared with the control. Additionally, the combined effect of RH and SA at a high rate demonstrated the highest increase in fresh (19-fold) and dry (threefold) shoot, and the greatest relative reductions in shoot Pb, Cu, Cd, and Zn concentrations were 74.72%, 69.19%, 54.16%, and 53.86%, respectively, compared with the control. Furthermore, the combination significantly improved all studied antioxidant enzymes and increased chlorophyll a and b and carotenoid contents by approximately 300%, 400%, and 250%, respectively, compared with the untreated soil. H 2 O 2 , malondialdehyde, and proline contents were significantly stimulated to the highest levels in the HM-contaminated soil, whereas that amended with RH alone or with sprayed SA significantly reduced their content. Adding RH with sprayed SA can remediate HM-contaminated soil and reduce the concentrations of potentially toxic metals in plants.

Abstract:Cadmium is known to hamper plant growth through disruption in structure, nutrition, water relations and metabolism in addition to oxidative stress induction. While, potassium has crucial role in the energy status of plants, translocation/storage of assimilates and tissue water relations. Further, it activates over 60 enzyme systems, aids in photosynthesis, regulates stomata] opening and nutrients translocation and enhances synthesis of starch and proteins. These physiological and biochemical aspects of potassium could also be having a remedial role in counteracting the detrimental effects of cadmium. However, such relationship has rarely been studied in detail. Therefore, the study in view was envisaged for exploiting potassium to counter-balance the cadmium toxicity in soybean. Emphasis was laid on growth, physiology and biochemistry of two soybean genotypes differing in cadmium response. Following treatments were employed under factorial design in hydroponics: Check (no cadmium or potassium applied); cadmium only; potassium only; cadmium + potassium. Cadmium reduced growth, chlorophyll content and fluorescence, photosynthesis and stomatal conductance markedly in both genotypes. Enhanced malondialdehyde (MDA) content, super oxide dismutase (SOD) and peroxidase (POD) activities were also detected in plants exposed to cadmium than those with potassium alone or combined with cadmium. The activities of SOD and POD were found higher in cv. Liao-1 than in cv. Zhechun3. Apparently, potassium and cadmium behaved antagonistically; suggesting that potassium could be a candidate for cadmium detoxification in crops cultivated under polluted environments.
Calcium and Potassium Supplementation Enhanced Growth, Osmolyte Secondary Metabolite Production, and Enzymatic Antioxidant Machinery in Cadmium-Exposed Chickpea (Cicer arietinum L.)

Abstract:This work examined the role of exogenously applied calcium (Ca; 50 mM) and potassium (K; 10 mM) (alone and in combination) in alleviating the negative effects of cadmium (Cd; 200 μM) on growth, biochemical attributes, secondary metabolites and yield of chickpea (Cicer arietinum L.). Cd stress significantly decreased the length and weight (fresh and dry) of shoot and root and yield attributes in terms of number of pods and seed yield (vs. control). Exhibition of decreases in chlorophyll (Chl) a, Chl b, and total Chl was also observed with Cd-exposure when compared to control. However, Cd-exposure led to an increase in the content of carotenoids. In contrast, the exogenous application of Ca and K individually as well as in combination minimized the extent of Cd-impact on previous traits. C. arietinum seedlings subjected to Cd treatment exhibited increased contents of organic solute (proline, Pro) and total protein; whereas, Ca and K-supplementation further enhanced the Pro and total protein content. Additionally, compared to control, Cd-exposure also caused elevation in the contents of oxidative stress markers (hydrogen peroxidase, H2O2; malondialdehyde, MDA) and in the activity of antioxidant defense enzymes (superoxide dismutase, SOD; catalase, CAT; ascorbate peroxidase, APX; glutathione reductase, GR). Ca, K, and Ca + K supplementation caused further enhancements in the activity of these enzymes but significantly decreased contents of H2O2 and MDA, also that of Cd accumulation in shoot and root. The contents of total phenol, flavonoid and mineral elements (S, Mn, Mg, Ca and K) that were also suppressed in Cd stressed plants in both shoot and root were restored to appreciable levels with Ca- and K-supplementation. However, the combination of Ca + K supplementation was more effective in bringing the positive response as compared to individual effect of Ca and K on Cd-exposed C. arietinum. Overall, this investigation suggests that application of Ca and/or K can efficiently minimize Cd-toxicity and eventually improve health and yield in C. arietinum by the cumulative outcome of the enhanced contents of organic solute, secondary metabolites, mineral elements, and activity of antioxidant defense enzymes.

Abstract:Due to many uses of zinc oxide nanoparticles (ZnO NPs) in various industries, the release of these particles in the environment and their effects on living organisms is inevitable. In this study, the role of salicylic acid (SA) pretreatments in modulating the toxicity of ZnO NPs was investigated using a hydroponic system. After pretreatment with different concentrations of SA (0, 25, 75, and 150 μM), Chenopodium murale plants were exposed to ZnO NPs (50 mg L−1). The results showed that exogenous SA increased the length, weight, chlorophyll, proline, starch, and soluble sugars in the plants. Besides, SA pretreatments improved water status in the plants treated with ZnO NPs. In SA-pretreated plants, increased activity of catalase (CAT), guaiacol peroxidase (GPX), and superoxide dismutase (SOD) was associated with a decline in electrolyte leakage (EL %) and membrane peroxidation. Under NPs stress, SA pretreatments increased the content of phenolic compounds by increasing the activity of phenylalanine ammonia-lyase (PAL). Exogenous SA reduced the translocation of larger amounts of Zn to the shoots, with more accumulation in the roots. This result can be used to produce healthy food from plants grown in environments contaminated with nanoparticles. It seems that all concentrations of SA reduced the symptoms of ZnO NPs toxicity in the plant by strengthening the function of the antioxidant system and increasing the content of some metabolites. Findings also suggest that SA pretreatment can compensate for the growth reduction caused by ZnO NPs.

Abstract:Cadmium (Cd) and lead (Pb) are among the most toxic heavy metals affecting human health and crop yield. Suaeda maritima (L.) Dumort is an obligate halophyte that is well adapted to saline soil. The inbuilt salinity tolerance mechanisms of halophytes help them to survive in heavy metal-contaminated rhizospheric soil. In the present study, growth and ionomic responses, reactive oxygen species (ROS) accumulation, modulations of phytochelatins, antioxidative defense, and metabolomic responses were studied in S. maritima imposed to Cd and Pb stresses with an aim to elucidate Cd and Pb tolerance mechanisms and phytoremediation potential of this halophyte. Our results showed a reduction of biomass in S. maritima, which may serve as an energy conservation strategy for survival under heavy metal stress. The increased accumulation of ROS with concomitant higher expression of various antioxidative enzymes suggests the efficient scavenging of ROS. The metabolite profiling revealed significant up-regulation of sugars, sugar alcohols, amino acids, polyphenols, and organic acids under Cd and Pb stresses suggesting their possible role in osmotic balance, ionic homeostasis, ROS scavenging, and signal transduction for stress tolerance. In S. maritima, the translocation factors (Tf) are <1 in both Cd and Pb treatments, which indicates that this halophyte has high phytostabilization potential for Cd and Pb in roots and through restricted translocation of heavy metal ions to the aboveground part. The findings of this study offer comprehensive information on Cd and Pb tolerance mechanisms in S. maritima and suggest that this halophyte can detoxify the HMs through physiological, ionic, antioxidative, and metabolic regulations.
Exploring the combined effect of heavy metals on accumulation efficiency of Salix alba raised on lead and cadmium contaminated soils

Abstract:The present study illustrated that Salix alba can accumulate high level of Pb and Cd in different plant parts, with maximum accumulation in roots followed by stem and leaves in the order Cd > Pb > Cd + Pb. The phytoremediation evaluation factors such as bioconcentration factor (BCF) and translocation factor (TF) was higher for Cd over Pb in all plant parts, further the BCF for both Pb and Cd was maximum in root (BCF > 1) followed by stem and leaves. Higher accumulation of Cd over the Pb was observed inside the plant tissues due to Cd mimics with other elements and gets transported through respective transporters. The combined treatment of Pb and Cd affected the bioaccumulation at every treatment level suggesting the negative effect among both elements. Higher survival rate (>85%) was recorded up to 200mgPb/kg and 15mgCd/kg, while further increase in metal concentration reduced the plant efficiency to remediate contaminated soils, hence results in declined survival rate. The FTIR analysis revealed that Pb and Cd accumulation in plants induced changes in carboxy, amino, hydroxyl and phosphate groups that ultimately caused alteration in physiological and biochemical processes of plant and thus provided an insight to the interaction, binding and accumulation of heavy metals. The present study conferred that Salix alba is a heavy metal (Pb and Cd) excluder plant on the basis of phytoremediation efficiency evaluation factors such BCF >1 (root) and TF <1. The correlation studies suggested the negative correlation among Pb and Cd accumulation and morphological traits. Physiological studies indicated that Pb and Cd accumulation negatively affect chlorophyll concentration and the antioxidant mechanism of plants gets activated, further these results are confirmed with FTIR studies, which reported the alteration in functional groups and associated compounds in plant tissues under Pb and Cd stress.

Abstract:Agricultural soil quality degradation by potentially toxic elements, specifically cadmium (Cd), poses a significant threat to plant growth and the health of humans. However, the supplementation of various salts of silicon (Si) to mitigate the adverse effect of Cd on the productivity of peas (Pisum sativum L.) is less known. Therefore, the present investigation was designed to evaluate the exogenous application at various levels (0, 0.50, 1.00 and 1.50 mM) of silicate compounds (sodium and potassium silicates) on pea growth, gaseous exchange, antioxidant enzyme activities and the potential health risk of Cd stress (20 mg kg−1 of soil) using CdCl2. The findings of the study showed that Cd stress significantly reduced growth, the fresh and dry biomass of roots and shoots and chlorophyll content. In addition, electrolyte leakage, antioxidant enzymes and the content of Cd in plant tissues were enhanced in Cd-induced stressed plants. An application of Si enhanced the development of stressed plants by modulating the growth of fresh and dry biomass, improving the chlorophyll contents and decreasing leakage from the plasma membrane. Furthermore, Si addition performed a vital function in relieving the effects of Cd stress by stimulating antioxidant potential. Hence, a significant level of metal protection was achieved by 1.00 mM of potassium silicate application under the Cd levels related to stress conditions, pointing to the fact that the Si concentration required for plant growth under Cd stress surpassed that which was required for general growth, enzymatic antioxidants regulation and limiting toxic metal uptake in plant tissues under normal conditions. The findings of this research work provide a feasible approach to reduce Cd toxicity in peas and to manage the entry and accumulation of Cd in food crops.

Abstract:The present attempt was made to study the role of exogenously applied salicylic acid (SA) and putrescine (Put) on the phytoremediation of heavy metals and on the growth parameters of chickpea grown in sandy soil. The SA and Put were applied alone as well as in combination with plant growth promoting rhizobacteria (PGPR). The PGPRs, isolated from the rhizosphere of chickpea, were characterized on the basis of colony morphology and biochemical traits through gram staining, catalase and oxidase tests, and identified by 16S-rRNA gene sequencing as Bacillus subtilis, Bacillus thuringiensis and Bacillus megaterium. The chickpea seeds were soaked in 24 h old fresh cultures of isolates for 2-3 h prior to sowing. The growth regulators (PGRs), SA and Put (150 mg/L), were applied to the seedlings as foliar spray at three-leaf stage. The result revealed that plants treated with SA and Put alone or in combination with PGPRs, significantly enhanced the accumulation of heavy metals in plant shoot. PGPR induces Ni accumulation in sensitive variety and Pb in both the varieties, the PGR in combination augment the bioremediation effects of PGPR and both sensitive and tolerant variety showed significant accumulation of Ni, Cd, and Pb. SA was more effective in accumulating Ni and Cd whereas, significant accumulation of Pb was recorded in Put. PGPRs further augmented the PGRs induced accumulation of heavy metals and macronutrients in chickpea shoot and in rhizosphere. SA increased the proline content of tolerant variety while decreasing the lipid peroxidation and proline content of the sensitive variety but decreased the stimulating effect of PGPR in proline production. Interactive effects of PGPR and PGRs are recommended for inducing phytoremediation in chickpea plants under drought stress.

Abstract:Cadmium (Cd), a pervasive noxious heavy metal, is a key threat to agricultural system. It is rapidly translocated and has detrimental effects on plant growth and development. Hydrogen sulphide (H2 S) is emerging as a potential messenger molecule for modulating plant tolerance to Cd. Salicylic acid (SA), a phenolic signalling molecule, can alleviate Cd toxicity in plants. The present study investigated the mediatory role of H2 S (100 µM) and SA (0.5 mM), individually and in combination, in modulating antioxidant defence machinery and nutrient balance to impart Cd (50 µM) resistance to mustard. Accumulation of Cd resulted in oxidative stress (TBARS and H2 O2 ), mineral nutrient imbalance (N, P, K, Ca), decreased leaf gas exchange and PSII efficiency, ultimately reducing plant growth. Both H2 S and SA independently attenuated phytotoxic effects of Cd by triggering antioxidant systems, enhancing the nutrient pool, eventually leading to improved photosynthesis and biomass of mustard plants. The positive effects were more pronounced under combined application of H2 S and SA, indicating a synergistic relationship between these two signalling molecules in mitigating the detrimental effects of Cd on nutrient homeostasis and overall health of mustard, primarily by boosting antioxidant pathway. Our findings provide new insights into H2 S- and SA-induced protective mechanisms in mustard plants subjected to Cd stress and suggest their combined use as a feasible strategy to confer Cd tolerance.
