Abstract:Cadmium (Cd) pollution is present in the world over especially in the industrialized parts of the world. To reduce Cd accumulation in various crops especially food crops, alleviating agents such as reduced glutathione (GSH) can be applied, which are capable either to exclude or to sequester Cd contamination. This study investigated the leaf-based spatial distribution of physiological, metabolic, and microstructural changes in two cotton cultivars (Coker 312 and TM-1) under GSH-mediated Cd stress using single levels of Cd (50 μM) and GSH (50 μM) both separately and in mix along with control. Results showed that GSH revived the morphology and physiology of both cotton cultivars alone or in mix with Cd. Cd uptake was enhanced in all segments of leaf and whole leaf upon the addition of GSH. GSH alleviated Cd-induced reduction in the photosynthetic pigment compositions and chlorophyll a fluorescence parameters. Mean data of biomarkers (2,3,5-triphenyltetrazolium (TTC), total soluble protein (TSP), malondialdehyde (MDA), hydrogen peroxide (H2O2)) revealed the adverse effects of Cd stress on leaf segments of both cultivars, which were revived by GSH. The oxidative metabolism induced by Cd stress was profoundly influenced by exogenous GSH application. The microstructural alterations were mainly confined to chloroplastic regions of leaves under Cd-stressed conditions, which were greatly revived upon the GSH addition. As a whole, Cd stress greatly affected TM-1 as compared to Coker 312. These results suggest a positive role of GSH in alleviating Cd-mediated changes in different leaf sections of cotton cultivars.

Abstract:Assessing the impact of Cadmium (Cd) on plant cells requires an understanding of the defensive mechanisms and adaptive responses employed by plants to counteract the deleterious effects of Cd toxicity. Wheat seedlings were grown under different concentrations (0 μM, 100 μM and 500 μM) of Cadmium salt (CdCl2). The growth of metal-treated seedlings was significantly affected. This study evaluated the ecotoxicological effects of two experimental concentrations of Cadmium (100 μM and 500 μM) against water control. It has been examined in recent studies that the toxic effects of Cadmium comprising the variables in fresh weight, dry weight and germination percentage of the wheat seedlings, photosynthesis and enzymatic activities in different stresses after seven days of cadmium exposure on roots and shoots of wheat seedling. Different antioxidant enzyme activities such as catalase (CAT), ascorbic acid oxidase (AOX), catechol peroxidase (CPX), superoxide dismutase (SOD), acid and alkaline phosphatase and inorganic pyrophosphatase have been measured. The various enzyme activities were increased in roots treated with high concentrations (500 μM) of Cadmium but decreased at low concentrations (100 μM) with respect to water control. This result indicated a complex defense mechanism in the root tissues induced by Cadmium. This work provides valuable insights for future research on plant responses to heavy metal stress and advances our understanding of the complex relationship between Cd toxicity and the antioxidant defense system in wheat seedlings.
Physio-Biochemical and Transcriptomic Features of Arbuscular Mycorrhizal Fungi Relieving Cadmium Stress in Wheat

Abstract:Arbuscular mycorrhizal fungi (AMF) can improve plant cadmium (Cd) tolerance, but the tolerance mechanism in wheat is not fully understood. This study aimed to examine the physiological properties and transcriptome changes in wheat inoculated with or without Glomus mosseae (GM) under Cd stress (0, 5, and 10 mg·kg-1 CdCl2) to understand its role in wheat Cd tolerance. The results showed that the Cd content in shoots decreased while the Cd accumulation in roots increased under AMF symbiosis compared to the non-inoculation group and that AMF significantly promoted the growth of wheat seedlings and reduced Cd-induced oxidative damage. This alleviative effect of AMF on wheat under Cd stress was mainly attributed to the fact that AMF accelerated the ascorbate-glutathione (AsA-GSH) cycle, promoted the production of GSH and metallothionein (MTs), improved the degradation of methylglyoxal (MG), and induced GRSP (glomalin-related soil protein) secretion. Furthermore, a comparative analysis of the transcriptomes of the symbiotic group and the non-symbiotic group revealed multiple differentially expressed genes (DEGs) in the 'metal ion transport', 'glutathione metabolism', 'cysteine and methionine metabolism', and 'plant hormone signal transduction' terms. The expression changes of these DEGs were basically consistent with the changes in physio-biochemical characteristics. Overall, AMF alleviated Cd stress in wheat mainly by promoting immobilization and sequestration of Cd, reducing ROS production and accelerating their scavenging, in which the rapid metabolism of GSH may play an important role.

Abstract:Cadmium, a high toxicity element, is a potential threat to plant and human health, and a dangerous pollutant in the environment. Uptake and accumulation by crops represent the main entry pathway for potentially health-threatening toxic metals into human and animal food. Crops and other plants take up Cd from the soil or water and may distribute it in their roots and shoots. Soil and/or water are usually contaminated with Cd through natural sources, industrial effluent, and anthropogenic activities. In this review, the sources of Cd contamination, evaluation of the phytotoxic effects on plants, and mode of action of Cd toxicity, were summarized. Plant defensive strategies upon excess Cd are also considered in this review. Cd-induced effects include oxidative stress, disintegration of the photosynthetic apparatus, reduction in gas exchange parameters, nutrient imbalance, and subcellular organelle degradation. In addition, Cd severely impairs biomolecules such as DNA, protein, and lipids. Although plants are sessile in nature, they are equipped with certain mechanisms to cope with unfavorable conditions. These mechanisms include synthesis of metal-chelating proteins, expression of enzymatic and non-enzymatic antioxidants, organic acids, and plant root-mycorrhiza association. The built-in system of plant tolerance to Cd can be further enhanced by the application of exogenous organic and inorganic metal sources. This review will broaden the knowledge about the Cd accumulation in plants and the responses to metal exposure, as well as our understanding of metal tolerance and overcoming this serious issue for sustainable agriculture and human health worldwide.
Effects of nickel, lead, and copper stress on the growth and biochemical responses of Aegilops tauschii seedlings

Abstract:Heavy metal pollution causes severe abiotic stress in cereal crops around the world. This study investigated the effects of different concentrations (0, 100, 200, and 300 mg·kg-1) of nickel, lead, and copper stress on the growth and biochemical responses of Aegilops tauschii seedlings, to provide a reference for research on the mechanism of invasion and screening potential sources of wheat tolerance genes. The results showed that nickel, lead, and copper stress caused a significant decrease in the contents of chlorophyll a, chlorophyll b, and chlorophyll (a + b) in A. tauschii, thereby inhibiting photosynthesis to different degrees and hindering seedling growth, which was reflected in significant reductions in plant height and root length, with the most notable effect observed under stress by 300 mg·kg-1 lead. As the concentration of heavy metals increased, the activities of antioxidant enzymes (SOD, POD, and APX), non-enzymatic antioxidants (GSH and AsA), and the contents of osmotic regulatory substances (proline and soluble proteins) in A. tauschii significantly increased. Additionally, heavy metal stress increased H2O2 and TBARS levels. However, when the nickel, lead, and copper concentrations reached 300 mg·kg-1, no significant differences were found in H2O2 or TBARS levels compared to those in the CK group. To summarize, A. tauschii can mitigate the accumulation of ROS and membrane lipid peroxidation caused by heavy metal stress through self-regulation, thus exhibiting a certain degree of tolerance to stress caused by different concentrations of nickel, lead, and copper. Finally, the evaluation using the membership function method revealed that among the three heavy metals, A. tauschii exhibited the strongest adaptation to Cu, followed by Ni and Pb.

Abstract:Heavy metal stress adversely affects plant growth and productivity worldwide. Alleviating the stress effect through the exogenous use of various chemical substances has become an interesting area of study in the field of plant stress tolerance. As a thiol compound, the cysteine derivative N-acetylcysteine (N-acetyl- L-cysteine, NAC) is the precursor of glutathione synthesis and a potent ROS scavenger with powerful antioxidant and free radical scavenging capabilities. This study investigated the effects of heavy metals (Cd, Hg and Pb, 100 ?M) on accumulation of soluble sugars and polyamine content in roots and shoots of wheat seedlings, the water potential and proline content in shoots and the role of NAC in protection against heavy metal toxicity. The addition of 1 mM NAC significantly increased root content of glucose, fructose and sucrose in varying degrees (avg. 1.34-, 1.20- and 1.51-fold, respectively) in comparison with heavy metals alone. The treatments led to a significant reduction of sugar content in shoots. Water potential values were highly correlated with proline and sugar content in wheat seedling shoots. Heavy metal stress significantly reduced polyamine content in both plant parts. The addition of NAC increased polyamine content in seedlings in comparison with heavy metals alone in both roots and shoots. These results suggest that NAC may protect plants from oxidative stress damage in heavy metal stress, and this enhancement of stress tolerance seems to involve soluble sugar and polyamine biosynthesis.

Abstract:Corn (Zea Mays L.) and wheat (Triticum aestivum L. ) seedlings were grown in four cadmium (Cd) concentration levels (0 - 1 mg/l) in a hydroponic system to analyze the antioxidant enzyme system, Cd concentration in the shoots and roots of plants, proline contents, growth responses and chlorophyll contents in the leaves of corn and wheat. The results showed that there was a significant increase in malondialdehyde (MDA) concentration from 0 to 1 mg/l, and peroxidase (POD) andcatalase (CAT) activities in the corn and wheat subjected to 0 – 1 mg/l of Cd. However, there was a significant decrease in the superoxide dismutase (SOD) activities in the corn and wheat subjected to 0 - 1 mg/l of Cd. This indicated that Cd stress induced an oxidative stress response in corn and wheat seedlings, characterized by an accumulation of MDA, decrease in the activities of SOD and increase in the activities of POD and CAT. Root and leaf Cd concentrations of corn and wheat increased with their exposure to the Cd level,s and the highest Cd concentration occurred in roots, followed by the leaves. An increase in proline in the leaves of corn and wheat seedlings exposed to Cd occurred as well as a decrease in chlorophyll (CHL) contents and shoots and roots biomass. Thus, Cd levels negatively affected the corn and wheat seedlings growth. Key words: Cadmium, corn (Zea mays L.), wheat (Triticum aestivum L.).

Abstract:The current state of heavy metal (HM) environmental pollution problems was considered in the review: the effects of HMs on the vital activity of plants and the functioning of their antioxidant system, including phenolic antioxidants. The latter performs an important function in the distribution and binding of metals, as well as HM detoxification in the plant organism. Much attention was focused on cadmium (Cd) ions as one of the most toxic elements for plants. The data on the accumulation of HMs, including Cd in the soil, the entry into plants, and the effect on their various physiological and biochemical processes (photosynthesis, respiration, transpiration, and water regime) were analyzed. Some aspects of HMs, including Cd, inactivation in plant tissues, and cell compartments, are considered, as well as the functioning of various metabolic pathways at the stage of the stress reaction of plant cells under the action of pollutants. The data on the effect of HMs on the antioxidant system of plants, the accumulation of low molecular weight phenolic bioantioxidants, and their role as ligand inactivators were summarized. The issues of polyphenol biosynthesis regulation under cadmium stress were considered. Understanding the physiological and biochemical role of low molecular antioxidants of phenolic nature under metal-induced stress is important in assessing the effect/aftereffect of Cd on various plant objects—the producers of these secondary metabolites are widely used for the health saving of the world's population. This review reflects the latest achievements in the field of studying the influence of HMs, including Cd, on various physiological and biochemical processes of the plant organism and enriches our knowledge about the multifunctional role of polyphenols, as one of the most common secondary metabolites, in the formation of plant resistance and adaptation.

Abstract:Cadmium is commonly recognized as toxic to plant growth, low-level Cd has promoting effects on growth performance, which is so-called hormesis. Although Cd toxicity in wheat has been widely investigated, knowledge of growth response to a broad range of Cd concentrations, especially extremely low concentrations, is still unknown. In this study, the morphological, physiological, and biochemical performance of wheat seedlings to a wide range of Cd concentrations (0–100 μΜ) were explored. Low Cd treatment (0.1–0.5 μM) improved wheat biomass and root development by enhancing the photosynthetic system and antioxidant system ability. Photosynthetic rate ( P n ) was improved by 5.72% under lower Cd treatment (1 μΜ), but inhibited by 6.05–49.85% from 5 to 100 μΜ. Excessive Cd accumulation induced oxidative injury manifesting higher MDA content, resulting in lower photosynthetic efficiency, stunted growth, and reduction of biomass. Further, the contents of ascorbate, glutathione, non-protein thiols, and phytochelatins were improved under 5–100 μΜ Cd treatment. The ascorbate peroxidase activity in the leaf showed a hormetic dose–response characteristic. Correlation analysis and partial least squares (PLS) results indicated that antioxidant enzymes and metabolites were closely correlated with Cd tolerance and accumulation. The results of the element network, correlation analysis, and PLS showed a crucial role for exogenous Cd levels in K, Fe, Cu, and Mn uptake and accumulation. These results provided a deeper understanding of the hormetic effect of Cd in wheat, which would be beneficial for improving the quality of hazard and risk assessments.

Abstract:The study examined the effects of jasmonic acid (100 nmol, JA) priming and foliar application of spermidine (1 mM, Spd), both individually and combined, on mitigating cadmium (100 μM, Cd) stress-induced oxidative damage in wheat. Cadmium stress reduced plant height and dry mass, but JA priming and/or Spd treatment increased resistance. Cd stress significantly decreased carotenoids, total chlorophylls, glutamate 1-semialdehyde (GSA), and δ-aminolevulinic acid (ALA), but JA and Spd treatments counteracted these reductions. Photosynthetic parameters improved under JA and Spd treatments, with combined treatment showing greater alleviation. Cd exposure increased lipid peroxidation, hydrogen peroxide, electrolyte leakage, and superoxide, but these oxidative stress indicators were significantly reduced after JA and Spd treatment. Antioxidant enzyme activity was upregulated by JA priming and Spd application, both under unstressed and Cd-stressed conditions. JA and/or Spd treatments also increased ascorbic acid, lowered glutathione concentration, and upregulated glyoxylase activity, reducing methylglyoxal accumulation. Additionally, secondary compounds (phenols and flavonoids) and osmolytes (proline and glycine betaine) levels improved. Proline oxidase activity decreased, indicating controlled proline buildup, while γ-glutamyl kinase activity increased. JA and/or Spd treatments significantly reduced Cd accumulation in seedlings. The study concluded that JA and Spd treatments enhance the plant's defensive mechanisms against oxidative stress by boosting antioxidant enzymes and secondary metabolism.

Abstract:Effects of different concentrations of soil cadmium (0–33mgkg−1) on growth, oxidative stress, and antioxidant response of wheat seedlings (Triticum aestivum L.) were investigated using pot experiments. A slight stimulatory effect on seedling growth was observed, especially at low Cd concentrations (less than 3.3mgkg−1). Results of the electron paramagnetic resonance (EPR) determination showed a decrease in unstable free radical level in the leaves, followed by a significant increase with increasing Cd concentrations. Malondialdehyde (MDA) contents were significantly enhanced by a high Cd concentration. Activity levels of some antioxidant enzymes in the leaves, including superoxide dismutase (SOD, EC1.12.1.1), catalase (CAT, EC1.11.1.6), guaiacol peroxidase (GPX, EC1.11.1.7), ascorbate peroxidase (APX, EC1.11.1.11) and glutathione reductase (GR, EC1.6.4.2), did not change much at low Cd concentrations (less than 3.3mgkg−1), but fluctuated drastically at high Cd concentrations. GSH contents and GSH/GSSG ratios decreased at low Cd concentrations, then increased at high Cd concentrations. Wheat seedlings might overcompensate at low Cd concentrations, resulting in a low oxidative stress and a positive effect on growth. Changes in biochemical parameters would occur before any visible symptom of toxicity appeared, and the endpoint based on these parameters might be more sensitive or indicative than morphological observations in revealing the eco-toxicity of Cd. Based on the results of this study, we propose that the toxic critical value of soil Cd in inducing oxidative stress to wheat seedlings is between 3.3mgkg−1 and 10mgkg−1.
Cadmium-induced ethylene production and responses in Arabidopsis thaliana rely on ACS2 and ACS6 gene expression

Abstract:BackgroundAnthropogenic activities cause metal pollution worldwide. Plants can absorb and accumulate these metals through their root system, inducing stress as a result of excess metal concentrations inside the plant. Ethylene is a regulator of multiple plant processes, and is affected by many biotic and abiotic stresses. Increased ethylene levels have been observed after exposure to excess metals but it remains unclear how the increased ethylene levels are achieved at the molecular level. In this study, the effects of cadmium (Cd) exposure on the production of ethylene and its precursor 1-aminocyclopropane-1-carboxylic acid (ACC), and on the expression of the ACC Synthase (ACS) and ACC Oxidase (ACO) multigene families were investigated in Arabidopsis thaliana.ResultsIncreased ethylene release after Cd exposure was directly measurable in a system using rockwool-cultivated plants; enhanced levels of the ethylene precursor ACC together with higher mRNA levels of ethylene responsive genes: ACO2, ETR2 and ERF1 also indicated increased ethylene production in hydroponic culture. Regarding underlying mechanisms, it was found that the transcript levels of ACO2 and ACO4, the most abundantly expressed members of the ACO multigene family, were increased upon Cd exposure. ACC synthesis is the rate-limiting step in ethylene biosynthesis, and transcript levels of both ACS2 and ACS6 showed the highest increase and became the most abundant isoforms after Cd exposure, suggesting their importance in the Cd-induced increase of ethylene production.ConclusionsCadmium induced the biosynthesis of ACC and ethylene in Arabidopsis thaliana plants mainly via the increased expression of ACS2 and ACS6. This was confirmed in the acs2-1acs6-1 double knockout mutants, which showed a decreased ethylene production, positively affecting leaf biomass and resulting in a delayed induction of ethylene responsive gene expressions without significant differences in Cd contents between wild-type and mutant plants.

Abstract:This study investigates the modulation of antioxidant defence system of Typha angustifolia after 30 days exposure of 1 mM chromium (Cr), cadmium (Cd), or lead (Pb). T. angustifolia showed high tolerance to heavy metal toxicity with no visual toxic symptom when exposed to metal stress, and Cd/Pb addition also increased plant height and biomass especially in Pb treatment. Along with increased Cr, Cd, and Pb uptake in metal treatments, there was enhanced uptake of plant nutrients including Ca and Fe, and Zn in Pb treatment. A significant increase in malondialdehyde (MDA) content and superoxide dismutase (SOD) and peroxidase (POD) activities were recorded in plants subjected to Cr, Cd, or Pb stress. Furthermore, Pb stress also improved catalase (CAT), ascorbate peroxidase (APX), and glutathione peroxidase (GPX) activities; whereas Cr stress depressed APX and GPX. The results indicate that enzymatic antioxidants and Ca/Fe uptake were important for heavy metal detoxification in T. angustifolia , stimulated antioxidative enzymes, and Ca, Fe, and Zn uptake could partially explain its hyper-Pb tolerance.

Abstract:Anthropogenic activities such as mining, manufacturing, and application of fertilizers release substantial quantities of cadmium (Cd) into the environment. In the natural environment, varying pH may play an important role in the absorption and accumulation of Cd in plants, which can cause toxicity and increase the risk to humans. We conducted a hydroponic experiment to examine the impact of pH on cadmium (Cd) solubility and bioavailability in winter wheat (Triticum aestivum L.) under controlled environmental conditions. The results showed that Cd concentration was significantly reduced in wheat with an increase in pH from 5 to 7, while it was dramatically increased at pH ranging from 7 to 9. However, in both cases, a significant reduction in physiological traits was observed. The addition of Cd (20, 50, and 200 μmol L-1) at all pH levels caused a substantial decline in wheat growth, chlorophyll and carotenoids contents, nutrient availability, while elevated cell membrane damage was observed in terms of electrolytic leakage (EL), osmoprotectants, and antioxidants activity. In our findings, the negative effects of acidic pH (5) on wheat growth and development were more pronounced in the presence of Cd toxicities. For instance, Cd concentration with 20, 50, and 200 μmol L-1 at acidic pH (5) reduced shoot dry biomass by 45%, 53%, and 79%, total chlorophyll contents by 26%, 41%, 56% while increased CAT activity in shoot by 109%, 175%, and 221%, SOD activity in shoot by 122%, 135%, and 167%, POD activity in shoot by 137%, 250%, and 265%, MDA contents in shoot by 51%, 83%, and 150%, H2O2 contents in shoot by 175%, 219%, and 292%, EL in shoot by 108%, 165%, and 230%, proline contents in shoot by 235%, 280%, and 393%, respectively as compared to neutral pH without Cd toxicities. On the other hand, neutral pH with Cd toxicities alleviated the negative effects of Cd toxicity on wheat plants by limiting Cd uptake, reduced reactive oxygen species (ROS) formation, and increased nutrient availability. In conclusion, neutral pH minimized the adverse effects of Cd stress by minimizing its uptake and accumulation in wheat plants.
Effects of cadmium on plant growth and physiological traits in contrast wheat recombinant inbred lines differing in cadmium tolerance

Abstract:Four wheat (Triticum aestivum L.) lines differing in cadmium (Cd) tolerance previously identified from a recombinant inbred line population were subjected to 50 microM CdCl2 from the three-leaf stage for 24 d, to investigate the responses of wheat seedlings to Cd toxicity. Under Cd stress, most growth parameters and root morphological traits were reduced, except for secondary root numbers and average root diameter. Cd enhanced leaf cell peroxidation due to increased malondialdehyde (MDA) content and reduced activities of superoxide dismutase (SOD) and catalase (CAT) in leaves. In addition, CAT activity decreased in the Cd-sensitive lines while increased in the tolerant lines. Leaf photosystem II (PSII) was damaged, since the maximum efficiency of PSII photochemistry (Fv/Fm) and potential efficiency of PSII photochemistry under dark-adapted (Fv/Fo) decreased, while the initial fluorescence (Fo) increased in all lines under Cd stress. Then, total soluble sugar concentration decreased while free amino acids concentration increased in both shoot and root. We concluded that Cd-tolerant lines accumulated less Cd in plant and contained low Cd concentration in shoot (less translocation of Cd to shoot), maintained higher CAT activity in leaf and higher PS II function than the Cd-sensitive lines under Cd toxicity, thus could be related to their tolerant capacity to Cd in the present study.
Interactive effects of temperature and heavy metal stress on the growth and some biochemical compounds in wheat seedlings

Abstract:In this study, the interaction between temperature and heavy metal stress was investigated in two wheat varieties (Triticum aestivum L. cv. Gerek-79 and Bolal-2973). Effects of different concentrations of lead and cadmium (0, 50, 100, 250, 500 mg l(-1)) simultaneously applied at various temperatures (8/4, 25/18, 35/26 degrees C day/night) to the seedlings were detected by measuring changes in the plant length, percentage of dry weight, chlorophyll (a, b, a/b), total soluble phenolics, and free proline. At the high cadmium concentrations, significant changes occurred in plant length and in the amounts of dry weight, chlorophyll, total soluble phenolics, and free proline. Significant differences were not observed in the parameters examined for the lead treatments. These data indicated that heavy metal toxicity increases in parallel with temperature. In addition, lead did not have any toxic effects on plants in sand-perlite mixture at 500 mg l(-1), whereas cadmium showed toxic effect even at such low concentrations as 50 mg l(-1).

Abstract:Heavy metal (HM) pollution, specifically cadmium (Cd) contamination, is a worldwide concern for its consequences for plant health and ecosystem stability. This review sheds light on the intricate mechanisms underlying Cd toxicity in plants and the various strategies employed by these organisms to mitigate its adverse effects. From molecular responses to physiological adaptations, plants have evolved sophisticated defense mechanisms to counteract Cd stress. We highlighted the role of phytochelatins (PCn) in plant detoxification, which chelate and sequester Cd ions to prevent their accumulation and minimize toxicity. Additionally, we explored the involvement of glutathione (GSH) in mitigating oxidative damage caused by Cd exposure and discussed the regulatory mechanisms governing GSH biosynthesis. We highlighted the role of transporter proteins, such as ATP-binding cassette transporters (ABCs) and heavy metal ATPases (HMAs), in mediating the uptake, sequestration, and detoxification of Cd in plants. Overall, this work offered valuable insights into the physiological, molecular, and biochemical mechanisms underlying plant responses to Cd stress, providing a basis for strategies to alleviate the unfavorable effects of HM pollution on plant health and ecosystem resilience.

Abstract:Anthropogenic activities pose a more significant threat to the environment than natural phenomena by contaminating the environment with heavy metals. Cadmium (Cd), a highly poisonous heavy metal, has a protracted biological half-life and threatens food safety. Plant roots absorb Cd due to their high bioavailability through apoplastic and symplastic pathways and translocate it to shoots through the xylem with the help of transporters and then to the edible parts via the phloem. The uptake and accumulation of Cd in plants pose deleterious effects on plant physiological and biochemical processes, which alter the morphology of vegetative and reproductive parts. In vegetative parts, Cd stunts root and shoot growth, photosynthetic activities, stomatal conductance, and overall plant biomass. Plants' male reproductive parts are more prone to Cd toxicity than female reproductive parts, ultimately affecting grain/fruit production and survival. To alleviate/avoid/tolerate Cd toxicity, plants activate several defense mechanisms, including enzymatic and non-enzymatic antioxidants, Cd-tolerant genes up-regulations, and phytohormonal secretion. Additionally, plants tolerate Cd through chelating and sequestering as part of the intracellular defensive mechanism with the help of phytochelatins and metallothionein proteins, which help mitigate the harmful effects of Cd. The knowledge on the impact of Cd on plant vegetative and reproductive parts and the plants' physiological and biochemical responses can help selection of the most effective Cd-mitigating/avoiding/tolerating strategy to manage Cd toxicity in plants.

Abstract:Cadmium (Cd) is an inauspicious abiotic traction that not only influences crop productivity and its growth parameters, but also has adverse effects on human health if these crops are consumed. Among crops, leafy vegetables which are the good source of mineral and vitamins accumulate more Cd than other vegetables. It is thus important to study photosynthetic variables, amino acid composition, and ultrastructural localization of Cd differences in response to Cd accumulation between two low and high Cd accumulating Brassica rapa ssp. chinensis L. (pak choi) cultivars, differing in Cd accumulation ability. Elevated Cd concentrations significantly lowered plant growth rate, biomass, leaf gas exchange and concentrations of amino acids collated to respective controls of both cultivars. Electron microscopy indicated that the impact of high Cd level on ultrastructure of leaf cells was associated to affecting cell functionalities, i.e. irregular cell wall, withdrawal of cell membrane, and chloroplast structure which has negative impact on photosynthetic activities, thus causing considerable plant growth suppression. Damage in root cells were observed in the form of enlargement of vacuole. The energy dispersive micro X-ray spectroscopy of both cultivars leaves indicated that cellular structure exhibited exudates of Cd-dense material. Ultrastructural damages and phytotoxicity were more pronounced in high accumulator cultivar as compared to the low accumulator cultivar. These findings are useful in determining the mechanisms of differential Cd-tolerance among cultivars with different Cd tolerance abilities at cellular level.
