Abstract:This study aimed to investigate the fluoride release properties and the effect on bond strength of two experimental adhesive cements. Synthesized particles of nano-fluorapatite (nano-FA) or nano-fluorohydroxyapatite (nano-FHA) were incorporated into a resin-modified glass ionomer cement (Fuji Ortho LC) and characterized using X-ray diffraction and scanning electron microscopy. Blocks with six different concentrations of nano-FA or nano-FHA were manufactured and their fluoride release properties evaluated by ultraviolet spectrophotometry. The unaltered glass ionomer cement Fuji Ortho LC (GC, control) and the two experimental cements with the highest fluoride release capacities (nano-FA+Fuji Ortho LC (GFA) and nano-FHA+Fuji Ortho LC (GFHA)) were used to bond composite blocks and orthodontic brackets to human enamel. After 24h water storage all specimens were debonded, measuring the micro-tensile bond strength (μTBS) and the shear bond strength (SBS), respectively. The optimal concentration of added nano-FA and nano-FHA for maximum fluoride release was 25wt.%, which nearly tripled fluoride release after 70days compared with the control group. GC exhibited a significantly higher SBS than GFHA/GFA, with GFHA and GFA not differing significantly (P>0.05). The μTBS of GC and GFA were significantly higher than that of GFHA (P⩽0.05). The results seem to indicate that the fluoride release properties of Fuji Ortho LC are improved by incorporating nano-FA or nano-FHA, simultaneously maintaining a clinically sufficient bond strength when nano-FA was added.
Fluoridation of Synthetic Apatite: Effect on the Formation of Calcium-Deficient Hydroxyapatite and the Properties of Porous Scaffold

Abstract:Fluoridated apatite (FHA) has been considered as a promising bone substitute substance due to its promoting cell proliferation and osteoblastic activity. This paper reports on the synthesis of FHAs via chemical precipitation at room temperature with different KF additions. The prepared samples were characterized using various characterization methods. Both PO43- and HPO42- ions were found in Fourier transform infrared (FTIR) spectra. X-ray diffraction (XRD) results show that two kinds of CaHPO4•2H2O (DCPD1, DCPD2) mixed with FHA were present in the final products at the lower content of KF addition. However, the amount of DCPD decreased as the increment of KF added, which hints that fluoridation distinctly accelerates the reaction of transforming DCPD into FHA. Meanwhile, 31P magic angle spinning nuclear magnetic resonance (MAS NMR) spectra indicated that the electron density around P nuclei in FHAs changed with F content. The highly interconnected scaffold with small pores nesting inside large ones had been fabricated by using raw FHA powder and behaved in good bioactivity when examined in simulated body fluid (SBF).

Abstract:Bone supports animal bodies, is the place where blood is produced, and is essential for the immune system, among other important functions. The dominant inorganic component in bone is hydroxyapatite (Hap), the structure and dynamics of which still pose many unsolved puzzles. An updated understanding of HAp is of great significance to osteology, dentistry, and the development of artificial bone and other biomaterials. In this work, HAp nanoparticles were synthesized with the wet chemical precipitation method and their structure and morphologies were controlled by varying pH and adding fluoride ions by two different routes: (1) fluoride ions were added during synthesis, and (2) fluoride ions were introduced after the samples were synthesized by soaking the samples in solutions with fluoride ions. XRD and HRTEM were employed to confirm the composition and structure, while various multinuclear (1H, 19F, 31P) solid-state nuclear magnetic resonance (NMR) methods including 1D single pulse, cross-polarization under magic-angle spinning (CPMAS), and 2D heteronuclear correlation (HETCOR) were used to characterize the structure, morphology, and dynamics, validating the general core-shell morphology in these F-HAp samples. It was found that all hydroxide ions were substituted when the fluoride ion concentration was above 0.005 M. An NMR peak corresponding to water structure emerged and the bulk water peak was shifted upfield, indicating that fluoride substitution modifies both the crystalline core and the amorphous shell of F-HAp nanoparticles. With the second route of fluoride substitution, increases in soaking time or fluoride ion concentration could increase fluoride substitution in HAp, but could not achieve complete substitution. Finally, with 1H-31P CPMAS and HETCOR, it was established that there are two types of phosphorous, one in the crystalline core (PO43−) and the other in the amorphous shell (HPO42−). These results are valuable for clarifying the fluoride substitution mechanism in HAp in biomaterials or in organisms, and provide insights for developing next generation replacement materials for bone, tooth, or coating films, drug delivery systems, etc.

Abstract:Coating layers consisting of a crystalline apatite matrix with immobilized basic fibroblast growth factor (bFGF) can release bFGF, thereby enhancing bone regeneration depending on their bFGF content. We hypothesized that the incorporation of fluoride ions into apatite crystals would enable the tailored release of bFGF from the coating layer depending on the layer's fluoride content. In the present study, coating layers consisting of fluoride-incorporated apatite (FAp) crystals with immobilized bFGF were coated on a porous collagen sponge by a precursor-assisted biomimetic process using supersaturated calcium phosphate solutions with various fluoride concentrations. The fluoride content in the coating layer increased with the increasing fluoride concentration of the supersaturated solution. The increased fluoride content in the coating layer reduced its solubility and suppressed the burst release of bFGF from the coated sponge into a physiological salt solution. The bFGF release was caused by the partial dissolution of the coating layer and, thus, accompanied by the fluoride release. The concentrations of released bFGF and fluoride were controlled within the estimated effective ranges in enhancing bone regeneration. These findings provide useful design guidelines for the construction of a mineralized, bFGF-releasing collagen scaffold that would be beneficial for bone tissue engineering, although further in vitro and in vivo studies are warranted.

The effect of fatigue on multifinger co‐ordination in force production tasks in humans

Abstract:1 This study investigated the effects of fatigue, induced by production of maximal isometric force for 60 s with four fingers, upon indices of multifinger co‐ordination. 2 Measurements of individual finger forces were performed during single‐ and multifinger maximal force production (maximal voluntary contraction, MVC) for two sites of force application, the middle of the distal or the middle of the proximal phalanxes. Two fatiguing exercises were used, involving force production at the distal phalanxes and at the proximal phalanxes. Fourteen subjects were tested. 3 The total force in four‐finger tasks dropped by about 43 % when it was produced at the site involved in the fatiguing exercise. During force production at the other site, MVC dropped by 23 %. During single‐finger MVC tests, force drop with fatigue was similar across all four fingers (about −25 % of their corresponding MVCs). 4 Force production by one finger was accompanied by involuntary force production by other fingers (enslaving). Enslaving remained unchanged by fatigue when measured during force generation at the site involved in the fatiguing exercise, but increased during force production at the other site. 5 The total MVC of four fingers acting in parallel was smaller than the sum of the MVCs of these fingers in single‐finger tasks (force deficit). The force deficit increased with fatigue. Force‐sharing patterns during four‐finger tasks showed only minor changes under fatigue. 6 These results indicate that the effects of fatigue were not limited to changes in the force‐generating capabilities of the muscles. In particular, fatigue could lead to a reorganisation at a neural level that defines commands to individual fingers.

Abstract:For dental/orthopedic implants to achieve better bone apposition and bone-implant bonding, various approaches to improve titanium surfaces have been developed. Recently, a fluoridated hydroxyapatite (FHA) coating on titanium (Ti) implants was made by sol–gel method and shown to be a possible applicative bone implant. The purpose of the current study was to evaluate biological responses and biomechanical bonding strength of FHA coated Ti implants as compared with that of the conventional Ti alloys and hydroxyapatite (HA) coated Ti implants. In vitro assays were made using human osteoblast-like cell (MG63) culture on different implants with cell attachment, morphology and differentiation evaluations. The implant plates were also implanted into the proximal metaphysis of New Zealand White rabbit tibiae. After 8 and 16 weeks implantation, mechanical and histological assessments were performed to evaluate biomechanical and biological behavior in vivo. The results showed that the cell adhesion and cell growth rate on the FHA and HA surface was higher than that on cp Ti surface (p<0.01), and insignificant difference was observed between two coated groups. Mechanical test demonstrated that the FHA implants had a higher interface shear strength than the both controls at 8 and 16 wks, with no significant difference with HA-Ti. Histologically, the coated implants revealed a significantly greater percentage of bone-implant contact when compared with the uncoated implants. Results demonstrated that the new FHA surface improved cell adhesion and proliferation. The coating exhibited a bioactive mechanical and histological behavior at bone-implant interface, suggesting that a useful approach by combined coating processes could optimize implant surfaces for bone deposition and early implant fixation.
Effect of borate, fluoride and strontium ions on biomimetic nucleation of calcium phosphate studied using solid-state nuclear magnetic resonance and X-ray diffraction

Abstract:Objectives: Apatite minerals can have various anions and cations in their crystal structure in addition to phosphate ion (PO₄³⁻) and calcium ion (Ca2+). The aim of this study is to investigate effects of the borate, fluoride and strontium ions on biomimetic nucleation of calcium phosphate. Methods: Nano-crystalline hydroxyapatite (H-Ap) was obtained from a supersaturated buffered solution containing 4.12 mM HPO42- and 5.88 mM Ca2+ (H-Ap). Four additives were used in solid solution methods: (i) 0.588 mM F- (F-Ap), (ii) 5.88 mM Sr2+ (Sr-Ap), (iii) 4.12 mM BO33- (BO3-Ap), and (iv) a surface pre-reacted glass ionomer (S-PRG) filler eluate that contained 0.17 mM Sr2+, 0.588 mM F-, 11.1 mM BO33-- (SPRG-Ap). Apatite crystallization was investigated using a solid-state magic-angle spinning NMR spectroscopy and X-ray diffraction (XRD) with the Rietveld analysis. Results: A 2D 1H-31P heteronuclear-correlation NMR showed F- ion incorporation in the apatite structure of the F-Ap and SPRG-Ap. The peaks on the 31P axis of the F-Ap, Sr-Ap, and BO3-Ap were different from that of the H-Ap, and the full width at half maximum increased in the following order: H-Ap∼F-Ap∼BO3-Ap< SPRG-Ap< Sr-Ap, suggesting the incorporation of the F-, Sr2+ and BO33-. The incorporation of F and BO3 was further confirmed by 19F and 11B NMR. The XRD revealed that Sr2+ was preferentially incorporated into the CaII site. Significance: The F-, Sr2+ and BO33-ions might be involved in modifying the crystallization of apatite precipitation, producing a variety of apatite. S-PRG filler that release these ions may have an effect on remineralization, i.e., the reformation of apatite lost due to caries.

Abstract:Hydroxyapatite (HA) is a non-bioceramic commonly used in human implants in the form of coatings, which are limited in their application by mechanical and wear resistance properties, as well as biodegradability. In this study, fluorine substituted hydroxyapatite (FHA) coatings were prepared on Ti–6Al–4V surfaces by plasma spraying method using a mixture of calcium fluoride and hydroxyapatite powders. The prepared coatings were characterized by X-ray diffraction and fourier transform infrared (FTIR) spectroscopy at different levels of calcium fluoride (3 wt.%, 6 wt.%, 9 wt.%, and 12 wt.%). The biocompatibility of the coatings was evaluated by in vitro mineralization experiments. Experimental results showed that at 9 wt.% of calcium fluoride, the prepared FHA coatings had better mechanical properties, with improved bond strength (28.2 MPa). The X-ray diffraction patterns of the coatings reflect the fluorine substitution during the spraying process and the 9FHA has the highest crystallinity according to the XRD analysis, which is closely related to the biological activity of the coating. In addition, Potentiodynamic polarisation showed that the sample coated with the 9FHA coating had the highest Ecorr and lowest Icorr, indicating the best corrosion resistance. The FHA coating exhibits faster apatite deposition in simulated body fluid, and the efficiency of apatite deposition increases with the increase of CaF 2 .

Abstract:A simple, area-specific coating technique for fluoridated apatite (FAp) on teeth would be useful in dental applications. Recently, we achieved area-specific FAp coating on a human dentin substrate within 30 min by a laser-assisted biomimetic (LAB) process; pulsed Nd:YAG laser irradiation in a fluoride-containing supersaturated calcium phosphate solution (FCP solution). The LAB-processed, FAp-coated dentin substrate exhibited antibacterial activity against a major oral bacterium, Streptococcus mutans. In the present study, we refined the LAB process with a combination of a dental diode laser and a clinically approved light-absorbing molecule, indocyanine green (ICG). A micron-thick FAp layer was successfully formed on the dentin surface within only 3 min by the refined LAB process, i.e., dental diode laser irradiation in the FCP solution following ICG treatment. The ICG layer precoated on the dentin substrate played a crucial role in inducing rapid pseudo-biomineralization (FAp layer formation) on the dentin surface by absorbing laser light at the solid-liquid interface. In the refined LAB process, the precoated ICG layer was eliminated and replaced with the newly formed FAp layer composed of vertically oriented pillar-like nanocrystals. Cross-sectional ultrastructural analysis revealed a smooth interface between the FAp layer and the dentin substrate. The refined LAB process has potential as a tool for the tooth surface functionalization and hence, is worth further process refinement and in vitro and in vivo studies.

Abstract:Abstract Hydroxyapatite (HA)‐based materials are widely used as bone substitutes due to their inherent biocompatibility, osteoconductivity, and bio‐absorption properties. However, HA scaffolds lack compressive strength when compared to autograft bone. It has been shown that the fluoridated form of HA, fluorapatite (FA), can be sintered to obtain this desired strength as well as slower degradation properties. Also, FA surfaces have been previously shown to promote stem cell differentiation toward an osteogenic lineage. Thus, it was hypothesized that FA, with and without stromal vascular fraction (SVF), would guide bone healing to an equal or better extent than the clinical gold standard. The regenerative potentials of these scaffolds were tested in 32 Lewis rats in a femoral condylar defect model with untreated (negative), isograft (positive), and commercial HA as controls. Animals were survived for 12 weeks post‐implantation. A semi‐quantitative micro‐CT analysis was developed to quantify the percent new bone formation within the defects. Our model showed significantly higher ( p < .05) new bone depositions in all apatite groups compared to the autograft group. Overall, the FA group had the most significant new bone deposition, while the differences between HA, FA, and FA + SVF were insignificant ( p > .05). Histological observations supported the micro‐CT findings and highlighted the presence of healthy bone tissues without interposing capsules or intense immune responses for FA groups. Most importantly, the regenerating bone tissue within the FA + SVF scaffolds resembled the architecture of the surrounding trabecular bone, showing intertrabecular spaces, while the FA group presented a denser cortical bone‐like architecture. Also, a lower density of cells was observed near FA granules compared to HA surfaces, suggesting a reduced immune response. This first in vivo rat study supported the tested hypothesis, illustrating the utility of FA as a bone scaffold material.

In Vivo Comparison of Bone Formation on Titanium Implant Surfaces Coated with Biomimetically Deposited Calcium Phosphate or Electrochemically Deposited Hydroxyapatite

Abstract:Purpose: The purpose of this study was to investigate and compare bone formation on titanium implant surfaces coated with biomimetically deposited calcium phosphate (BDCaP) or electrochemically deposited hydroxyapatite (EDHA). Materials and Methods: The implants were separated into three groups: a control group, a BDCaP group, and an EDHA group. Surface analysis was performed by field-emission scanning electron microscopy, x-ray diffractometry, and Fourier transform infrared spectroscopy. Implants were inserted in a randomized arrangement into rabbit tibiae. After 2, 4, and 8 weeks, the tibiae were retrieved and prepared for histomorphometric evaluation. Results: Field-emission scanning electron microscopy showed that the BDCaP crystals were flakelike and the EDHA crystals were rodlike with a hexagonal cross section. X-ray diffractometric patterns and Fourier transform infrared spectroscopy spectra showed that the BDCaP coating consisted of HA and octacalcium phosphate, whereas the EDHA coating consisted of HA. Histologic observation showed that new bone on the EDHA-coated implant became mature after 4 weeks, while new bone on the control and BDCaP-coated implants was mature after 8 weeks. The EDHA implant showed significantly greater BIC and bone area compared to the control and BDCaP implants during 4 to 8 weeks. The BDCaP coating failed to show increased bone formation during the test period. Fluoride-doped amorphous calcium phosphate nanoparticles as a promising biomimetic material for dental remineralization

Abstract:Demineralization of dental hard tissue is a widespread problem and the main responsible for dental caries and dentin hypersensitivity. The most promising strategies to induce the precipitation of new mineral phase are the application of materials releasing gradually Ca2+ and PO43- ions or mimicking the mineral phase of the host tissue. However, the design of formulations covering both processes is so far a challenge in preventive dentistry. In this work, we have synthesized innovative biomimetic amorphous calcium phosphate (ACP), which has been, for the first time, doped with fluoride ions (FACP) to obtain materials with enhanced anti-caries and remineralizing properties. Significantly, the doping with fluoride (F) did not vary the physico-chemical features of ACP but resulted in a faster conversion to the crystalline apatite phase in water, as observed by in-situ time-dependent Raman experiments. The efficacy of the as synthesized ACP and FACP samples to occlude dentinal tubules and induce enamel remineralization has been tested in vitro in human molar teeth. The samples showed good ability to partially occlude the tubules of acid-etched dentin and to restore demineralized enamel into its native structure. Results demonstrate that ACP and FACP are promising biomimetic materials in preventive dentistry to hinder demineralization of dental hard tissues.
Efficacy of sintered Zinc-doped fluorapatite scaffold as an antimicrobial regenerative bone filler for dental applications

Abstract:Objectives The objective of this study was to assess whether zinc-doped fluorapatite (ZnFA) could serve as an effective antimicrobial dental bone filler for bone regeneration compared to autografts. Methods FA and 2% zinc-doped FA (2ZnFA) were synthesized and characterized in-house. Compressed and sintered FA and 2ZnFA disks were incubated with bacteria to assess antimicrobial properties. Adipose-derived stem cells were cultured on these discs to evaluate the surfaces' ability to support cell growth and promote osteogenic differentiation. Surfaces exhibiting the highest expressions of the bone markers osteopontin and osteocalcin were selected for an in vivo study in a rat mandibular defect model. Twenty rats were divided into 5 groups, equally, and a 5mm surgical defect of the jaw was left untreated or filled with 2ZnFA, FA, autograft, or demineralized bone matrix (DBM). At 12 weeks, the defects and surrounding tissues were harvested and subjected to microCT and histological evaluations. Results Standard techniques such as FTIR, ICP-MS, fluoride probe, and XRD revealed the sintered FA and ZnFA's chemical compositions and structures. Bacterial studies revealed no significant differences in surface bacterial adhesion properties between FA and 2ZnFA, but significantly fewer bacterial loads than control titanium discs (p<0.05). Cell culture data confirmed that both surfaces could support cell growth and promote the osteogenic differentiation of stem cells. MicroCT analysis confirmed statistical similarities in bone regeneration within FA, 2ZnFA, and autograft groups. Conclusion The data suggests that both FA and 2ZnFA could serve as alternatives to autograft materials, which are the current gold standard. Moreover, these bone fillers outperformed DBM, an allograft material commonly used as a dental bone void filler. Clinical Significance The use of FA or 2ZnFA for treating mandibular defects led to bone regeneration statistically similar to autograft repair and significantly outperformed the widely used dental bone filler, DBM. Additional translational research may confirm FA-based materials as superior substitutes for existing synthetic bone fillers, ultimately enhancing patient outcomes.

In Vitro Behavior of Osteoblast-Like Cells on Fluoridated Hydroxyapatite Coatings.

Abstract:In this work, fluoridated hydroxyapatite (Ca10(PO4)6Fx(OH)2−x or FHA) coatings are prepared by sol–gel method for study of the influence of F content on the behavior of osteoblast-like cells. The results show that the cells well attach and proliferate on the FHA coatings studied (Ca10(PO4)6F0.67–2.00(OH)0–1.33). With increasing F content in the FHA coatings, percentage of cell in S period increases, indicating F in the coating favors the proliferation process of the cells. On the other hand, the proliferation rate increases inversely with zeta potential of the coating surface. As tested from the MTT of the cells cultured in the leaching out solution, increase of F content in the FHA coatings results in a slight decrease in cell proliferation rate, which is most probably due to reduction in release of Ca2+ ions. As a compromise among cell attachment, cell proliferation, apatite deposition and ability to resist dissolution, it is suggested that FHA coatings (Ca10(PO4)6Fx(OH)2−x) with x in the range of 0.67–1.48, from the results of this study, may be most suitable for real case implantation.
A Porous Fluoride-Substituted Bovine-Derived Hydroxyapatite Scaffold Constructed for Applications in Bone Tissue Regeneration

Abstract:Hydroxyapatite is widely used in bone implantation because of its similar mineral composition to natural bone, allowing it to serve as a biocompatible osteoconductive support. A bovine-derived hydroxyapatite (BHA) scaffold was developed through an array of defatting and deproteinization procedures. The BHA scaffold was substituted with fluoride ions using a modified sol-gel method to produce a bovine-derived fluorapatite (BFA) scaffold. Fourier-transform infrared spectroscopy and X-ray diffraction analysis showed that fluoride ions were successfully substituted into the BHA lattice. According to energy dispersive X-ray analysis, the main inorganic phases contained calcium and phosphorus with a fluoride ratio of ~1–2 wt%. Scanning electron microscopy presented a natural microporous architecture for the BFA scaffold with pore sizes ranging from ~200–600 μm. The BHA scaffold was chemically stable and showed sustained degradation in simulated-body fluid. Young's modulus and yield strength were superior in the BFA scaffold to BHA. In vitro cell culture studies showed that the BFA was biocompatible, supporting the proliferative growth of Saos-2 osteoblast cells and exhibiting osteoinductive features. This unique technique of producing hydroxyapatite from bovine bone with the intent of producing high performance biomedically targeted materials could be used to improve bone repair.

Abstract:Standard zirconia implants used in restoration still present problems related to inertness and long-term stability. Various physicochemical approaches have been used to modify the implant surfaces to improve early and late bone-to-implant integration; however, no ideal surface modification has been reported. This study used pulsed laser deposition to deposit a fluorinated hydroxyapatite (FHA) film on a zirconia implant to create a biologically active surface. The film prepared was uniform, dense, and crack-free, and exhibited granular surface droplets; it also presented excellent mechanical strength and favorable biological behavior. The FHA-coated implant was implanted on the femur of Sprague–Dawley rats, and various tests and analyses were performed. Results show that the in vitro initial cell activity on the FHA-coated samples was enhanced. In addition, higher alkaline phosphatase activity and cell mineralization were detected in cells cultured on the FHA-coated groups. Further, the newly formed bone volume of the FHA-coated group was higher than that of the bare micro-adjusted composite nano-zirconia (NANOZR) group. Therefore, the FHA film facilitated osseointegration and may improve the long-term survival rates of dental implants, and could become part of a new treatment technology for implant surfaces, promoting further optimization of NANOZR implant materials.

Abstract:Background: Alveolar bone augmentation is a complex process influenced by a multitude of factors. The materials applied in augmentation procedures must be confirmed as non-toxic, and their physicochemical properties should allow proper bone reconstruction. The specifics of oral surgical procedures require the use of regenerative biomaterials with antimicrobial properties. This study focuses on the physicochemical characteristics of chosen nanosized biomaterials, as well as their cytotoxicity and antimicrobial properties. Methods: nanosized hydroxyapatite and fluorapatite (abbreviated as nHAp and nFAp) pellets were manufactured using a microwave hydrothermal synthesis method. The impact on Candida albicans, Streptococcus mutans, and Lactobacillus rhamnosus strains activity and adherence to apatites was tested. Cytotoxic evaluation was performed based on the differentiation process of MC3T3 cells. The effectiveness of MC3T3 differentiation was confirmed by Alizarin Red staining. Results: Contact with both biomaterials caused a reduction in the mean microbial count of S. mutans and C. albicans strains, as observed. Studied biomaterials demonstrated enhanced proliferation of MC3T3 cells, with the exception of the 1:1 nFAp concentration. Conclusions: Both biomaterials enhance the proliferation of fibroblasts and limit the activity of specific oral pathogens in vitro. The research clearly demonstrates the advantage of nFAp over nHAp, with a notable reduction in microbial count of Candida albicans and Streptococcus mutans over time. The lowest microbial count reduction was observed in the case of L. rhamnosus. Further research is required in order to fully understand the specifics of nHAp and nFAp antimicrobial action. However, the results were found to be more favourable for nFAp biomaterial.

Abstract:Rapid bone regeneration is crucial for restoring alveolar bone and oral functions following periodontal diseases. However, the development of effective biomedical materials for this purpose remains insufficient. While bone autografts can enhance bone regeneration, they are invasive to healthy areas. Specifically, for alveolar bone regeneration, the implanted material must possess adequate mechanical strength. Moreover, local administration is preferred for older adults, who are a primary target population, to maintain their quality of life. We developed a silica-substituted carbonate apatite (CO 3 Ap–silica) block as newly bone substitute with a bone growth factor, featuring the major inorganic component of mature bone to enhance bone regeneration. CO 3 Ap–silica block stimulated the bone remodeling process at the implantation site and demonstrated significantly better bone regeneration compared to currently used carbonate apatite substitutes. Therefore, this new material is expected to advance technologies for restoring occlusal function after periodontal disease. On the osteogenic differentiation of dental pulp stem cells by a fabricated porous nano-hydroxyapatite substrate loaded with sodium fluoride

Abstract:In the present study, nano-hydroxyapatite (n-HA) powder was extracted from carp bone waste to fabricate porous n-HA substrates by a molding and sintering process. Subsequently, the substrates were loaded with different amounts of sodium fluoride (NaF) through immersion in NaF suspensions for 10, 7.5, and 5 min. The NaF-loaded n-HA substrates were then examined for their structural and physical properties, chemical bonds, loading and release profile, pH changes, cytotoxicity, and osteogenic effect on dental pulp stem cells (DPSCs) at the level of RNA and protein expression. The results showed that the n-HA substrates were porous (> 40% porosity) and had rough surfaces. The NaF could be successfully loaded on the substrates, which was 6.43, 4.50, and 1.47 mg, respectively for n-HA substrates with immersion times of 10, 7.5, and 5 min in the NaF suspensions. It was observed that the NaF release rate was rather fast during the first 24 h in all groups (39.06%, 36.43%, and 39.57% for 10, 7.5, and 5 min, respectively), and decreased dramatically after that, indicating a slow detachment of NaF. Furthermore, the pH of the medium related to all materials was changed during the first 4 days of immersion (from 7.38 to pH of about 7.85, 7.84, 7.63, and 7.66 for C0, C5, C7.5, and C10, respectively). The pH of media associated with the C7.5, and C10 increased up to 4 days and remained relatively constant until day 14 (pH = 7.6). The results of the cytotoxicity assay rejected any toxicity of the fabricated NaF-loaded n-HA substrates on DPSCs, and the cells could adhere to their surfaces with enlarged morphology. The results showed no effect on the osteogenic differentiation at the protein level. Nevertheless, this effect was observed at the gene level.
