Abstract:A series of novel calix[5]arenes 5 containing one 1,8-dimethoxytriptycene moiety were synthesized through an efficient fragment coupling strategy. The subsequent demethylation of 5 with BBr(3) in dry dichloromethane gave the calix[5]arenes 6. Debutylation of both 5a-b and 6a-b with AlCl(3) resulted in the same products 7a-7b. The structural studies revealed that all of the macrocyclic compounds have well-defined structures with fixed cone conformations in both solution and solid state. Moreover, it was also found that the triptycene-derived calix[5]arenes could encapsulate small neutral molecules in the solid state.

Abstract:Two pairs of novel triptycene-derived calix[6]arenes 4a,b and 5a,b have been efficiently synthesized through both one-pot and two-step fragment-coupling strategies starting from 2,7-bis(hydroxymethyl)-1,8-dimethoxytriptycene 1. Subsequent demethylation of 4a,b and 5a,b with BBr(3) in dry dichloromethane gave the macrocyclic compounds 6a,b and 7a,b. Treatment of either 4a or 6a with AlCl(3) resulted in the same debutylated product 8, while 9 was similarly obtained from either 5a or 7a. Structural studies revealed that all of the macrocycles have well-defined structures with fixed conformations both in solution and in the solid state owing to the introduction of the triptycene moiety with a rigid three-dimensional (3D) structure, making them very different from their classical calix[6]arene counterparts. As a consequence, it was found that all of these the triptycene-derived calix[6]arenes could encapsulate small neutral molecules in their cavities in the solid state. Moreover, it was also found that the macrocycles 4b and 5b showed highly efficient complexation abilities toward fullerenes C(60) and C(70), forming 1:1 complexes with association constants ranging from (5.22+/-0.20) x 10(4) to (8.68+/-0.30) x 10(4) M(-1).

Abstract:A novel deep-cavity calixarene, p-[1-(4-hydroxyphenyl)-1-methylethyl]calix[8]arene 1 was synthesized and its complexation with aromatic compounds was examined, Reaction of p-[1-(4-methoxyphenyl)-1-methylethyl]phenol 2 with paraformaldehyde under various conditions only resulted in p-[1-(4-methoxyphenyl)-1-methylethyl]calix[8]arene 3, and no corresponding calix[4]arene and calix[6]arene were detected, Removal of the methyl in the ether groups of 3 afforded 1 in 80% yield, Solubilization, fluorescence and photochemical probe studies demonstrated that 1 forms inclusion complexes with a variety of aromatic compounds in polar non-aqueous solutions.

Abstract:Presented in this paper are the practical synthesis and conformational structure of hydrazo-bridged homo calix[4] arenes. The step-wise fragment coupling method based on the nucleophilic aromatic substitution reaction of 2,6-dihydrazinylpyridine with 6-alkoxy-2,4-dichlorotriazine afforded effectively hydrazo-linked homo calix[2] pyridine[3] triazines. The exhaustive methylation of -NHNH-linkages led to lipophilic macrocycles, while the introduction of a THP-protected triglyme moiety on a triazine ring produced a water soluble hydrazo-bridged homo calix[2] pyridine[2] triazine. The acquired hydrazo-bridged homo calix[4] arenes adopted a cone conformational structure with two nitrogen atoms of the hydrazine linkage forming respectively a conjugation system with pyridine and triazine rings. The unique macrocyclic conformation and multidentate binding sites would render hydrazo-bridged homo calix[2] pyridine[2] triazines powerful hosts in molecular recognition and self-assembly.

Abstract:A novel calix[4]arene derivative, 26,28-bis(cyanomethoxy)-25,27-dihydroxycalix[4]arene (1), was synthesized and its crystal structure has been determined. The crystals, obtained by recrystallization from acetone, are triclinic, space group P-1, with a=10.1160(17)Å, b=11.996(2)Å, c=13.148(2)Å, and α=63.331(3)°, β=74.392(4)°, γ=85.077(4)°. As compared with a similar compound, 5,11,17,23-tetra-tert-butyl-26,28-bis(cyanomethoxy)-25,27-dihydroxycalix[4]arene (2), compound 1 shows a different configuration of the 1,3-bis(cyanomethoxy) groups attached to the lower rim.

Abstract:Two new copper-thiacalix[4]arene compounds, [Cu-2(1)-Cl-2(H(4)TC4A)](CH3OH) (1) and [Cu(I)2Cl(2)(H(4)PTC4A)](CH3OH)(CHCl3)(0.5) (2) (where H(4)TC4A = p-tert-butylthiacalix[4]arene and H(4)PTC4A = p-phenylthiacalix[4]arene), were synthesized by the solvothermal method in the mixed CH3OH/CHCl3 (1: 1) solvent and reassembled in air at room temperature to two other structures, [(Cu4Cl3)-Cl-II(HCO2)(TC4A)(CH3-OH)(2)(H2O)](CHCl3)(CH3OH)(2.7) (3) and [(Cu4Cl4)-Cl-II(PTC4A)(CH3OH)(4)] (4), respectively. All these four compounds were characterized by TG analyses, FTIR spectroscopy, and singlecrystal X-ray diffraction analyses. Compounds 1 and 2 feature two-dimensional layered networks, while compounds 3 and 4 are assembled by some tetranuclear units. Abstract:Functionalised dialkoxy-substituted tetraoxacalix[2]arene[2]triazine macrocycles 6 have been readily synthesised by the fragment coupling approach using methyl 3,5-dihydroxy-4-alkoxybenzoates and cyanuric chloride as the starting materials under very mild conditions. AlCl3-mediated deallylation and debenzylation reactions afforded the lower-rim dihydroxy-substituted tetraoxacalix[2]arene[2]triazine derivatives 11 and 13 in good yields. Although dialkoxy-substituted tetraoxacalix[2]arene[2]triazine macrocycles are fluxional in solution on the NMR spectroscopy timescale, they adopt a symmetric or slightly distorted 1,3-alternate conformation with the bridging oxygen atoms conjugated with the triazine rings. The dihydroxylated tetraoxacalix[2]arene[2]triazine 13b, which gives a mixture of monomer and dimer in solution according to a diffusion NMR spectroscopy study, adopts a 1,3-alternate conformation and forms a cyclic tetrameric assembly in the solid state due to the formation of intermolecular hydrogen-bonding networks. This dihydroxylated macrocyclic host molecule, a hydrogen-bond donor macrocycle with a V-shaped cleft, interacts with 2,2'-bipyridine, 4,4'-bipyridine and 1,10-phenanthroline guests. Although in solution they form the corresponding 1:1 complexes with binding constants ranging from 37.7 to 213 M-1, 2:2 host-guest complexes were observed in the crystalline state. Hydrogen-bonding interactions, along with other non-covalent interactions, such as lone-pair-electron-pi and C-H center dot center dot center dot pi interactions, were found to be the driving force for the formation of host-guest complexes.

Abstract:A number of homo[2] and homo[4] heterocalix[2]arene[2]triazines were synthesized through a general and good-yielding fragment coupling approach starting from cyanuric halides, aromatic and aliphatic diols, and diamines under very mild reaction conditions. While homo[2] tetraazacalix[2]arene[2]triazine gave a twisted and pinched 1,2-alternate conformer, almost all homo[2] heterocalix[2]arene[2]triazines adopted different partial cone conformations in the solid state. Homo[41] heterocalix[2]arene[2]triazines yielded more diverse conformational structures including partial cone, pinched partial cone, 1,2-alternate and twisted 1.2-alternate, depending on the nature abridging moieties. On the basis of H-1 NMR spectra, homo[2] and homo[4] heterocalix[2]arene[2]triazines were fluxional macrocycles in solution, and they underwent rapid conformation interconversion at different temperatures. Efficient and straightforward nucleophilic aromatic substitution reaction and palladium-catalyzed cross-coupling reactions on chlorotriazine rings, and the nucleophilic alkylation reaction on the bridging nitrogen atoms led to the construction of various highly functionalized homo heterocalix[2]arene[2]triazine derivatives.

Abstract:A number of unprecedented homo heteracalix[2]arene[2]triazines were synthesized by means of a fragment coupling approach. Two directional nucleophilic substitution reactions of N-Boc-protected 1,3-dihydrazobenzene with cyanuric acid chloride and 2-butoxy-4,6-dichloro-1,3,5-triazine led to hydrazo-linked trimers, which underwent an efficient macrocyclic condensation reaction with functionalized resorcinol derivatives to afford (NHNBoc)(2),O-2-calix[2]arene[2]triazine macrocycles, which contain a functional group either on the upper rim or the lower rim. The use of 1,3-phenylenediamines instead of resorcinol in the reaction produced (NR)(2),(NHNBoc)(2)-calix[2]arene[2]triazines. Postmacrocyclization modifications such as a nucleophilic substitution reaction of chloro on triazine by amines and the removal of Boc from hydrazo moieties produced homo calix[2]arene[2]triazine derivatives. In the solid state, (NHNR)(2),O-2-bridged calix[2]arene[2]triazines with and without a substituent on the upper rim position and (NMe)(2),(NHNBoc)(2)-calix[2]arene[2]triazine adopted a typical partial cone conformation while the heavily twisted 1,3-alternate conformational structures were observed for both (NHNBoc)(2),O-2-calix[2]arene[2]triazines bearing a functional group on the lower rim position and (NH)(2),(NHNBoc)(2)-calix[2]arene[2]triazine. In solution, all synthesized homo heteracalix[2]arene[2]triazines existed as the mixture of different macrocyclic conformers, which underwent slow interconversions at room temperature relative to the NMR time scale.

Abstract:Construction of macrocyclic hosts with a novel structure and excellent property has emerged as an intriguing undertaking for the past few years. Here, we reported the synthesis of shape-persistent triptycene-derived pillar[6]arene (TP[6]). The single crystal structure analysis revealed that the macrocyclic molecule adopts a hexagonal structure, featuring a helical and electron-rich cavity capable of encapsulating electron-deficient guests. In order to obtain chiral TP[6] from an enantiomerically pure triptycene building block, an efficient resolution of chiral triptycene was successfully developed through introducing chiral auxiliaries into triptycene skeletons. The 1H NMR and isothermal titration calorimetry investigations demonstrated that chiral TP[6] exhibited enantioselectivity toward four pairs of chiral guests containing a trimethylamino group, implying a significant promising application in area of enantioselective recognition.

Abstract:This paper describes our attempts to synthesize inherently chiral heteroatom-bridged calixaromatics. Based on a stepwise fragment-coupling approach using a chiral 3,5-dihydroxybenzamide or benzoate, 2,4-dihydroxyacetophenone, and cyanuric chloride as reactants, chiral tetraoxacalix[2]arene[2]triazine derivatives 16 and 17 were synthesized in good yields. Subsequent macrocyclic condensation with a diamine 6 furnished efficiently the pairs of diastereomers of inherently chiral tetraoxacalix[2]arene[2]triazine azacrowns 18 and 19.

Abstract:The syntheses and conformational structures of various functionalized tetraoxacalix[2]arene[2]triazines were studied. Applying the fragment coupling approach and the post-macrocyclization chemical manipulations, a number of tetraoxacalix[2]arene[2]triazines that contain, on the lower rim, one or two aldehyde, ester, carboxylic acid, hydroxymethyl, and aminomethyl functional groups were prepared in moderate to high chemical yields from cheap and commercially available materials. On the basis of X-ray crystallography and NMR spectroscopy, all tetraoxacalix[2]arene[2]triazines containing electron-withdrawing group(s) adopted 1,3-alternate conformation both in solution and in the solid state, while tetraoxacalix[2]arene[2]triazines bearing hydroxymethyl and aminomethyl substituent(s) existed as pinched or distorted partial cone conformers due to the formation of intramolecular hydrogen bond between hydroxyl or amino group and triazine ring. Abstract:Triphenylamine[3]arenes (TPA[3]s), featuring [16]paracyclophane backbone with alternating carbon and nitrogen bridging atoms, were synthesized through a BF3·Et2O-catalyzed cyclization reaction using triphenylamine derivatized monomers and paraformaldehyde. This molecular design yielded a series of TPA[3] macrocycles with high efficiency, with their facile derivatizations also successfully demonstrated. On account of the strong electron-donating properties of the TPA moieties, these TPA[3]s exhibit remarkable delayed fluorescence, and possess a significant affinity for iodine. Furthermore, their inherent three-fold symmetry rendered TPA[3]s as novel building blocks for the construction of extended frameworks and molecular cages. This advancement expands the versatility of discrete macrocycles into complex architectures, enhancing their applicability across a broad spectrum of applications.

Abstract:The synthesis and crystal structure of a novel calix[8)arene ester are reported herein. The calix[8]arene ester derivative has been characterized by IR, NMR and X-ray crystal analysis. The X-ray structure analysis revealed that the 8 phenolic hydroxy groups of the calix(8]arene have been substituted by 4 diethyl dibromomalonate molecules with each two adjacent hydroxy oxygen atoms attached to a bridge diethyl malonate.
Triptycene-Derived Macrocyclic Arenes: From Calixarenes to Helicarenes

Abstract:The development of new types of synthetic macrocyclic hosts is always one of the most important and attractive topics in macrocyclic and supramolecular chemistry. Calixarenes, resorcinarenes, cyclotriveratrylenes, pillararenes, and their analogues are all composed of hydroxy-substituted aromatic rings bridged by methylene or methenyl groups and thus can be considered a type of macrocyclic arenes. Because of their unique structural features, easy functionalization, and wide applications in many research areas, such macrocyclic arenes have become some of the most important and studied synthetic macrocyclic hosts during the last decades. Triptycene and its derivatives are a class of organic molecules having unique three-dimensional rigid structures, and they have proved to be useful building blocks for constructing new synthetic macrocyclic hosts with specific structures and properties. Dihydroxy-substituted triptycene derivatives are readily available compounds, which encouraged us to conduct studies of triptycene-derived macrocyclic arenes about 10 years ago. Consequently, a series of triptycene-derived calixarenes and analogues containing 1,8-dihydroxy-substituted triptycene subunits were conveniently synthesized. With 2,7-dihydroxy-substituted triptycene as a precursor, new types of calixarene, oxacalixarene, and homooxacalixarene analogues were also obtained. These triptycene-derived macrocyclic hosts all showed fixed conformations in solution and exhibited expanded cavities compared with the corresponding typical calixarenes and analogues. The special structural features also make these triptycene-derived macrocycles show wide potential applications in molecular recognition and self-assembly. In particular, it was found that the threading direction and the orientation based on macrocycles with nonsymmetric structures could be finely controlled by adjusting the electrostatic and steric effects of the guests, which could form the oriented [2]rotaxane by unidirectional threading. We recently developed a new kind of chiral macrocyclic arenes named helicarenes that are composed of chiral 2,6-dihydroxy-substituted triptycene subunits bridged by methylene groups. It was found that the helicarenes not only exhibited convenient synthesis, high stability, good solubility, fixed conformations, and easy functionalization but also showed complexation abilities with various chiral and achiral organic guests. In particular, the switchable complexation based on these macrocycles could be efficiently controlled by multiple stimuli, including acid/base, redox, anion, or light stimuli under a photoacid. Moreover, the helicarenes have also found applications in the construction of interlocked molecules and molecular machines. This Account summarizes our recent research results on the synthesis and structures of the triptycene-derived macrocyclic arenes and analogues and their applications in host-guest chemistry and molecular assembly. We believe that these macrocyclic arenes, especially helicarenes, could be utilized as new synthetic hosts and find wide potential applications in macrocyclic and supramolecular chemistry.

Abstract:Heteracalixaromatics are an emerging generation of macrocyclic host molecules in supramolecular chemistry. As a typical example of heteracalixaromatics, oxacalix[2]arene[2]triazine adopts a shape-persistent 1,3-alternate conformation and can be easily functionalized. Taking it as a platform, a series of oxacalix[2]arene[2]triazine-based amphiphiles bearing long alkyl chains were synthesized through post-macrocyclization functionalization or 3+1 fragment coupling protocols. The self-assembly behavior of these amphiphiles in a mixture of tetrahydrofuran (THF) and water was investigated. Dynamic light scattering (DLS) measurements revealed that the size of the self-assembled aggregates is dependent on the structure of the amphiphiles. The long alkyl chain substituents and/or intermolecular hydrogen bonds were found to promote the self-assembly.
