Abstract:This paper reviews aspects of NPY research that were emerging in 1985, shortly after the isolation and characterization of the peptide. NPY had become known for its widespread distribution especially in the central and peripheral nervous systems, but also in the gastro-intestinal and respiratory tracts and in fibers innervating smooth muscle around blood vessels. Consistent with its distribution, it was determined that NPY is a potent vasoconstrictor, affects neuroendocrine systems and is involved in appetite regulation--areas of research still relevant today. Through advances in technology knowledge about NPY's role in these and newly discovered physiological functions has deepened considerably. Successful cloning of a series of NPY receptors has opened up new and complex research vistas. Lately, the creation of mice genetically modified for NPY as well as for several receptor subtypes has brought many puzzling observations--followed by questions yet to be answered.
Enriched Expression of Serotonin 1B and 2A Receptor Genes in Macaque Visual Cortex and Their Bidirectional Modulatory Effects on Neuronal Responses

Abstract:To study the molecular mechanism how cortical areas are specialized in adult primates, we searched for area-specific genes in macaque monkeys and found striking enrichment of serotonin (5-hydroxytryptamine, 5-HT) 1B receptor mRNA, and to a lesser extent, of 5-HT2A receptor mRNA, in the primary visual area (V1). In situ hybridization analyses revealed that both mRNA species were highly concentrated in the geniculorecipient layers IVA and IVC, where they were coexpressed in the same neurons. Monocular inactivation by tetrodotoxin injection resulted in a strong and rapid (< 3 h) downregulation of these mRNAs, suggesting the retinal activity dependency of their expression. Consistent with the high expression level in V1, clear modulatory effects of 5-HT1B and 5-HT2A receptor agonists on the responses of V1 neurons were observed in in vivo electrophysiological experiments. The modulatory effect of the 5-HT1B agonist was dependent on the firing rate of the recorded neurons: The effect tended to be facilitative for neurons with a high firing rate, and suppressive for those with a low firing rate. The 5-HT2A agonist showed opposite effects. These results suggest that this serotonergic system controls the visual response in V1 for optimization of information processing toward the incoming visual inputs.

Abstract:Neuropeptide S (NPS) is a regulatory peptide with potent pharmacological effects. In rodents, NPS is expressed in a few pontine cell clusters. Its receptor (NPSR1) is, however, widely distributed in the brain. The anxiolytic and arousal-promoting effects of NPS make the NPS-NPSR1 system an interesting potential drug target in mood-related disorders. However, so far possible disease-related mechanisms involving NPS have only been studied in rodents. To validate the relevance of these animal studies for i.a. drug development, we have explored the distribution of NPS-expressing neurons in the human pons using in situ hybridization and stereological methods and we compared the distribution of NPS mRNA expressing neurons in the human and rat brain. The calculation revealed a total number of 22,317 ± 2411 NPS mRNA-positive neurons in human, bilaterally. The majority of cells (84%) were located in the parabrachial area in human: in the extension of the medial and lateral parabrachial nuclei, in the Kölliker-Fuse nucleus and around the adjacent lateral lemniscus. In human, in sharp contrast to the rodents, only very few NPS-positive cells (5%) were found close to the locus coeruleus. In addition, we identified a smaller cell cluster (11% of all NPS cells) in the pontine central gray matter both in human and rat, which has not been described previously even in rodents. We also examined the distribution of NPSR1 mRNA-expressing neurons in the human pons. These cells were mainly located in the rostral laterodorsal tegmental nucleus, the cuneiform nucleus, the microcellular tegmental nucleus region and in the periaqueductal gray. Our results show that both NPS and NPSR1 in the human pons are preferentially localized in regions of importance for integration of visceral autonomic information and emotional behavior. The reported interspecies differences must, however, be considered when looking for targets for new pharmacotherapeutical interventions.

Abstract:Oxygen isotope compositions of the interstitial waters have been measured for 21 samples taken from the depth intervals of 1.5 to 398.9 mbsf at Site 798 (Oki Ridge) and 16.5 to 435.6 mbsf at Site 799 (Kita-Yamato Trough) in Japan Sea. The δ 18 θ values decrease with depth from -0.49 to -3.38 o/oo (SMOW) at Site 798 and from -0.71 to -4.36 o/oo (SMOW) at Site 799 corresponding to an average depletion gradient of-0.8 o/ooper 100 m. Material balance calculations reveal that the δ18θ-variations at Sites 798 and 799 were principally controlled by low-temperature alteration of basement basalt and andesite, resulting in negative shifts in pore water δ 18 O values, and by the polymorphic transformations of biogenic opal-A to opal-CT and opal-CT to microquartz, which tend to increase δ 18 θ of interstitial waters. Carbonate diagenesis and ash alteration also caused weakly negative shifts in pore water δ 18 θ values.
Colocalization of somatostatin, neuropeptide Y, and NADPH-diaphorase in the caudate-putamen of the rat

Abstract:Somatostatin, neuropeptide Y, and nicotinamide adenine dinucleotide phosphate-diaphorase are colocalized within a small population of medium aspiny neurons in the caudate-putamen of the rat. The extent of colocalization, however, appears to be in dispute. In order to examine the question of colocalization between these three neuroactive substances, a series of double-labelling experiments was performed. This was accomplished by combining immunocytochemistry for somatostatin or neuropeptide Y or enzyme histochemistry for nicotinamide adenine dinucleotide phosphate-diaphorase with in situ hybridization for somatostatin and/or neuropeptide Y mRNA. The results of such analysis indicate that nicotinamide adenine dinucleotide phosphate-diaphorase and somatostatin mRNA are 100% colocalized throughout the caudate-putamen, except for the area bordering the globus pallidus. All neurons that contain neuropeptide Y contain somatostatin message. Only 84% of the neurons that contain somatostatin mRNA, however, also contain neuropeptide Y. Neurons that contain somatostatin 28 but not neuropeptide Y are found throughout the caudate-putamen. These results indicate that the somatostatin neuron population in the rat caudate-putamen is not homogeneous. Instead, the medium aspiny neuron population is actually composed of several subpopulations based on the content of neuroactive substances.

Abstract:Previous double-label studies have demonstrated that progesterone receptors (PR) may coexist with nitric oxide synthase (NOS) and also with neuroactive peptides such as somatostatin (SOM), neurotensin (NT) and substance P (SP) in neurons of the ventrolateral hypothalamic nucleus (VL) of the guinea pig. In the present study, triple-label immunofluorescence histochemistry was used to determine whether neurons co-expressing both PR and NOS also contain one neuropeptide (SOM, NT or SP) and to quantify these colocalization relationships. Ovariectomized guinea pigs were primed with estradiol to induce PR immunoreactivity and treated with colchicine to visualize immunoreactive (IR) peptidergic neurons. Using three primary antibodies raised in different species and labeled with three different markers, it is possible to recognize which type(s) of immunoreactivity each cell contains on the same sections. We observed that PR, NOS and SOM co-occurred extensively, whereas PR, NOS and NT or PR, NOS and SP were colocalized infrequently in neurons of the VL. Combining our various quantitative observations, we found that SOM immunoreactivity appeared in 33% of the PR/NOS-IR neurons in the caudal aspect of the nucleus and that nearly all neurons containing SOM and NOS contained PR. The occasional localization between PR, NOS and SP seen in the rostral subdivision of the VL represented a very low proportion of the PR/NOS-IR neurons but 48% of NOS/SP-IR neurons. The relatively high proportion of PR/NOS-IR cells expressing SOM in the VL suggests that these neurons are part of a neural circuitry involved in a variety of steroid-dependent functions.
Mapping neuropeptide sigaling in the human brain

Abstract:Neuropeptides are functionally diverse signaling molecules in the brain, regulating a wide range of basal bodily and cognitive processes. Despite their importance, the distribution and function of neuropeptides in the human brain remains underexplored. Here we comprehensively map the organization of human whole-brain neuropeptide receptors across multiple levels of description; from molecular and cellular embedding to mesoscale connectivity and macroscale cognitive specialization. Using gene transcription as a proxy, we reconstruct a topographic cortical and subcortical atlas of neuropeptide receptors for 38 neuropeptide receptors, across 14 different neuropeptide families. We find that most neuropeptide receptors are highly expressed either in cortex or subcortex, delineating an anatomical cortical-subcortical gradient. Neuropeptides preferentially co-localize with metabotropic neurotransmitters, suggesting a system-wide correspondence between slow-acting molecular signaling mechanisms. Mapping neuropeptide receptors and their cognate ligands onto white-matter connectomes, we demonstrate that specific neuropeptides families shape electrophysiological and haemodynamic inter-regional connectivity. To investigate the behavioural consequences of distributed neuropeptide systems, we apply meta-analytic decoding to neuropeptide maps and show a gradient of functions, from sensory-cognitive to reward and bodily functions. Finally, evolutionary analysis indicates extended positive selection for neuropeptides in early mammals, suggesting that refinement of neuropeptides coincides with the emergence of neocortex and higher cognitive function. Collectively, these results show that the neuropeptide receptors are highly organized across the human brain and closely intertwined with multiple features of brain structure and function.

Artificial Neural Network Based Node Location Prediction for Applications in Mobile Communication

Abstract:- Present generation cellular networks provide different services to the mobile users. The movement of the users is highly dependent on individual characteristics. To offer an uninterrupted service to the mobile users, continuous tracking of its location is very important. This can be achieved by the proper location management schemes. In this paper, a prediction-based location management scheme for locating a mobile station is proposed. A multilayer neural network model for mobile movement prediction is designed to predict the future movement of a mobile host. The model is trained with the data obtained from the past movement pattern of a mobile host. The prediction method is found to give satisfactory results.
Corticotropin-releasing Factor Receptor Type 1 Colocalizes with Type 2 in Corticotropin-Releasing Factor-Containing Cellular Profiles in Rat Brain.

Abstract:OBJECTIVES:To investigate whether CRHR1 and CRHR2 are colocalized in CRH-specific neurons in rat brain.METHODS:Double/triple immunofluorescence, and combined in situ hybridization were performed in the PVN, amygdala and hippocampus, and triple immunofluorescence was applied to the median eminence (ME), dorsal raphe (DR) and locus coeruleus (LC).RESULTS:Both CRHR1 and CRHR2 immunoreactivity were highly coexpressed in the PVN, central nucleus of the amygdala (CeA) and hippocampus. Triple immunofluorescence under confocal microscopy confirmed that CRHR1 and CRHR2 are coexpressed in CRH-producing neurons in these regions. The results of in situ hybridization combined with double immunofluorescence further strengthened the finding that CRHR1 and CRHR2 were coexpressed in CRH-specific neurons in the PVN, CeA and hippocampus. In addition, CRH immunoreactivity signals were evidently distributed in the ME, DR and LC, and were coexpressed with both receptors.CONCLUSION:CRH receptors colocalize in CRH-containing neurons in the PVN, CeA and hippocampus, and CRH, CRHR1, and CRHR2 coexist in the DR and LC. Our results implicate CRHR1 and CRHR2 in coordinating the regulation of CRH neuronal activity in stress and behavioral responses.
Physiological and Therapeutic Roles of Neuropeptide Y on Biological Functions

Abstract:Neuropeptide Y (NPY), an amino acid, used for various physiological processes for management and treatment of various ailments related to central nervous system, cardiovascular system, respiratory system, gastro-intestinal system and endocrinal system. In nasal mucosa, high concentrations of NPY are stored with noradrenaline in sympathetic nerve fibers. NPY Y1 receptor mediates nitric oxide levels and reduction in blood flow in nasal mucosa of the human. NPY plays a role in dietary consumption via various factors like signaling the CNS for a prerequisite of energy in hypothalamus by mediating appetite and shows orexigenic effect. NPY emerges as a natural ligand of G-protein coupled receptors which activates the Y-receptors (Y1-Y6). But applications of NPY are limited due to shows the cost inefficiency and stability issues in the formulation design and development. In this review, authors present the findings on various therapeutic applications of NPY on different organ systems. Moreover, its role in food intake, sexual behavior, blood pressure, etc. by inhibiting calcium and activating potassium channels. The combination therapies of drugs with neuropeptide Y and its receptors will show new targets for treating diseases. Further evaluation and detection of NPY needs to be investigated for animal models of various diseases like retinal degeneration and immune mechanisms.
Immunohistochemical Identification of Interneuronal Subpopulations in the Basolateral Amygdala of the Rhesus Monkey (Macaca mulatta)

Abstract:Inhibitory circuits in the basolateral nuclear complex of the amygdala (BNC) critical for controlling the acquisition, expression, and extinction of emotional responses are mediated by GABAergic interneurons (INs). Studies in rodents have demonstrated that separate IN subpopulations, identified by their expression of calcium-binding proteins and neuropeptides, play discrete roles in the intrinsic circuitry of the BNC. Far less is known about IN subpopulations in primates. In order to fill in this gap in our understanding of primate INs, the present investigation used dual-labeling immunohistochemistry for IN markers to identify subpopulations expressing cholecystokinin (CCK), calbindin (CB), calretinin (CR), and somatostatin (SOM) in somata and axon terminals in the monkey BNC. In general, colocalization patterns seen in somata and axon terminals were similar. It was found that there was virtually no colocalization of CB and CR, the two calcium-binding proteins investigated. Three subtypes of CCK-immunoreactive (CCK+) INs were identified on the basis of their expression of CR or CB: (1) CCK+/CR+; (2) CCK+/CB+); and (3) CCK+/CR-/CB-. Almost no colocalization of CCK with SOM was observed, but there was extensive colocalization of SOM and CB. CCK+, CR+, and CCK+/CR+ double-labeled axon terminals were seen surrounding pyramidal cell somata in basket-like plexuses, as well as in the neuropil. CB+, SOM+, and CB+/SOM+ terminals did not form baskets, suggesting that these IN subpopulations are mainly dendrite-targeting neurons. In general, the IN subpopulations in the monkey are not dissimilar to those seen in rodents but, unlike rodents, CB+ INs in the monkey are not basket cells.
Neuropeptide FF (NPFF)-positive nerve cells of the human cerebral cortex and white matter in controls, selected neurodegenerative diseases, and schizophrenia

Abstract:We quantified and determined for the first time the distribution pattern of the neuropeptide NPFF in the human cerebral cortex and subjacent white matter. To do so, we studied n = 9 cases without neurological disorders and n = 22 cases with neurodegenerative diseases, including sporadic amyotrophic lateral sclerosis (ALS, n = 8), Alzheimer's disease (AD, n = 8), Pick's disease (PiD, n = 3), and schizophrenia (n = 3). NPFF-immunopositive cells were located chiefly, but not exclusively, in the superficial white matter and constituted there a subpopulation of white matter interstitial cells (WMIC): Pyramidal-like and multipolar somata predominated in the gyral crowns, whereas bipolar and ovoid somata predominated in the cortex surrounding the sulci. Their sparsely ramified axons were unmyelinated and exhibited NPFF-positive bead-like varicosities. We found significantly fewer NPFF-immunopositive cells in the gray matter of the frontal, cingulate, and superior temporal gyri of both sporadic ALS and late-stage AD patients than in controls, and significantly fewer NPFF-positive cells in the subjacent as well as deep white matter of the frontal gyrus of these patients compared to controls. Notably, the number of NPFF-positive cells was also significantly lower in the hippocampal formation in AD compared to controls. In PiD, NPFF-positive cells were present in significantly lower numbers in the gray and white matter of the cingulate and frontal gyrii in comparison to controls. In schizophrenic patients, lower wNPFF cell counts in the neocortex were significant and global (cingulate, frontal, superior temporal gyrus, medial, and inferior gyri). The precise functions of NPFF-positive cells and their relationship to the superficial corticocortical white matter U-fibers are currently unknown. Here, NPFF immunohistochemistry and expression characterize a previously unrecognized population of cells in the human brain, thereby providing a new entry-point for investigating their physiological and pathophysiological roles.
Different subcellular distributions of the vesicular monoamine transporter, VMAT2, in subclasses of sympathetic neurons

Abstract:A subpopulation of neurons in the rat superior cervical ganglion (SCG) was found to lack immunostaining for VMAT2, an isoform of the vesicular monoamine transporter that loads catecholamines into vesicles for release at the synapse. Double labeling with neuropeptide Y (NPY), a marker for vasomotor neurons, revealed selective cellular colocalization of NPY together with intense perinuclear staining for VMAT2. This implied that VMAT2-negative neurons were likely to have secretomotor and pilomotor phenotypes. We tested this by identifying peripheral noradrenergic axons by their expression of immunoreactivity for tyrosine hydroxylase (TH) and determining whether they also expressed NPY and VMAT2. This analysis revealed the presence of VMAT2-positive, non-vasomotor sympathetic axons in the submandibular gland and at the base of piloerector hairs. Together the results confirm earlier indications that virtually all sympathetic neurons in the rat SCG express VMAT2 and they show for the first time that functional subclasses of cells can be distinguished by different somatic levels of immunoreactivity for VMAT2.

Abstract:In response to three highly conserved neuropeptides, neuropeptide Y (NPY), peptide YY, and pancreatic polypeptide (PP), four G protein–coupled receptors mediate multiple essential physiological processes, such as food intake, vasoconstriction, sedation, and memory retention. Here, we report the structures of the human Y 1 , Y 2 , and Y 4 receptors in complex with NPY or PP, and the G i1 protein. These structures reveal distinct binding poses of the peptide upon coupling to different receptors, reflecting the importance of the conformational plasticity of the peptide in recognizing the NPY receptors. The N terminus of the peptide forms extensive interactions with the Y 1 receptor, but not with the Y 2 and Y 4 receptors. Supported by mutagenesis and functional studies, subtype-specific interactions between the receptors and peptides were further observed. These findings provide insight into key factors that govern NPY signal recognition and transduction, and would enable development of selective drugs.

Abstract:Prothymosin alpha (ProTα) is an acidic nuclear protein implicated in several cellular functions including cell survival. ProTα is found in the central nervous system, but the regional and cell type-specific expression patterns are not known. In this study, our immunohistochemical analysis demonstrated that ProTα is expressed ubiquitously throughout adult brain with difference in the intensity of region-specific protein reactivity. Interestingly, the highest ProTα signals were observed in the brain regions relevant to neurogenesis, such as sub-ventricular zone, granular cell layer of dentate gyrus, as well as granule cell layer of olfactory bulb. Strong immunoreactivity was also found in habenula, ependymal cells lining the dorsal third and fourth ventricle, and in neurons in the Purkinje cell layer of cerebellum. We showed that ProTα was strictly localized in the nuclei of neurons, while it was found in the cytosolic space of astroglial and microglial processes and cell body in the adult brain. To clarify the phenomenon underlying cytosolic localization of ProTα in non-neuronal cells, ZVAD-fmk, a caspase-3 inhibitor, was delivered intracerebroventricularly in the brain. At the follow-up 24 h after ZVAD-fmk injection, we found that nuclear intensity of ProTα was significantly increased in astrocytes, whereas the ProTα expression was not affected in microglia. The present study would contribute toward better understanding of physiological and pathophysiological roles of ProTα in the brain.

Abstract:OBJECTIVE To describe the diagnostic work-up and treatment of patients with a pancreatic carcinoma in the Amsterdam area, the Netherlands, particularly in general hospitals. DESIGN Retrospective, descriptive. METHOD During 1997, 286 patients with a pancreatic carcinoma were diagnosed in 20 hospitals in the Amsterdam area. Diagnostic work-up and treatment data were collected from the medical records and analysed. RESULTS Ninety percent of the patients presented in one of the 17 general hospitals (n = 252; 132 men and 154 women; mean age: 70 years). Thirty-five percent of them underwent diagnostic investigations which did not focus directly on pancreatic pathology. Ultrasound was performed in 97% of patients (4% in combination with Doppler) and CT in 60% (4% spiral CT). Endoscopic retrograde cholangiopancreatography (ERCP) was performed in 39% of these patients and endoprostheses were only inserted in half the cases. Thirty-five percent of the patients who underwent both CT and ERCP underwent ERCP first. Ninety-nine patients (39%) were referred to a reference hospital for further investigation or treatment. The period between the first investigation and the histological diagnosis was 4 weeks. CONCLUSION In the diagnostic work-up of patients with a pancreatic carcinoma, invasive diagnostic procedures were often performed before the non-invasive tests. Spiral CT was used minimally and ERCP was frequently performed without subsequent biliary drainage. The mean duration of diagnostic work-up was relatively long.
Distribution of Seven Major Neurotransmitter Receptors in the Striate Cortex of the New World Monkey Callithrix Jacchus.

Abstract:The distribution of seven different binding sites for the transmitters L-glutamate (L-glutamate binding sites and N-methyl-D-aspartate receptor), GABA (GABAA receptor), noradrenaline (alpha 1 receptor), acetylcholine (muscarinic M1 and M2 receptors) and serotonin (5-hydroxytryptamine1 receptor) are analysed in the primary visual cortex (area 17) of the common marmoset, Callithrix jacchus, using quantitative autoradiography. All binding sites show a well-defined laminar pattern, which changes sharply at the cytoarchitectonic border to area 18. The quantitative data show that the distribution of different receptors is relatively invariant across the cortical layers. Almost all receptors show a maximum in supragranular layers, low densities in layers IVA/IVB and a second maximum in layer IVC. Statistical analysis of these similarities in laminar distribution patterns of different receptors (co-distribution) reveals, as in other brain regions and species, that L-glutamate binding sites are co-distributed with N-methyl-D-aspartate, GABAA, and muscarinic M1 and M2 receptors. This may reflect the structural basis of a possible interaction between these receptors and their respective transmitters on the level of single cortical layers. Further co-distributions are found between N-methyl-D-aspartate, GABAA and M1, as well as between alpha 1 and M1 and finally between M1 and M2 receptors. Since not all receptors are co-distributed, the similarities in laminar patterns reveal specific aspects of the neurochemical organization of the cortex when receptors of different transmitter systems are analysed in the same brain.(ABSTRACT TRUNCATED AT 250 WORDS)
